Abstract
Description
i)ポリマーもしくはコポリマーとNSAIDとを共通溶媒中に溶解させるステップ;および
ii)ステップi)の結果として生じた溶液をエレクトロスピンするステップを含む方法を、
提供する。
1.スカフォールドの合成および特性解析
A.ポリマー
本試験のために、様々な比率のラクチド対グリコチドの75:25(Mw(分子量):66〜107k)、50:50(Mw:40〜75k)およびこれら2種の1:1ブレンドを有する3種のポリ(D,L−ラクチド−コ−グリコリド)(PLGA)コポリマーを使用した。全PLGAコポリマーは、Sigma Aldrich社から購入した。これらのポリマーは、エレクトロスピニングのために適切な粘度の溶液を形成するためにジクロロメタン(DCM)中に溶解させた:75:25および50:50を用いてそれぞれ20%および25%(重量/重量)溶液を形成し、等量のこれらの溶液を使用してブレンドを形成した。イブプロフェン粉末(Sigma Aldrich社)をDCM中のポリマー溶液に加えて均質混合液を形成した(ポリマーの重量に比較して1〜10重量%のイブプロフェンを加えた)。
ポリマー溶液は、内径が0.8mmの鈍端のステンレススチール製ニードルを装備した2mLシリンジ(I&J Fisnar社)内に装填した。この溶液は3.5mL/時の一定供給速度で、プログラム可能なシリンジポンプ(Aladdin 1000)を使用して送達され、高電圧電源装置(Brandenburg、AlphaシリーズIII)によって供給される15kVの加速電圧を用いて水平にエレクトロスピンされた。繊維マットは、ニードル先端から、アースされたアルミニウム製回転コレクター(300rpmで回転する)20cmの周囲で被覆されたアルミニウム製ホイルシート(18cm×16cm)上で収集した。ジェット形成/安定性は、ニードル先端の5mm後方で、15kVの電圧でアルミニウム製焦点リングによって支援された。スカフォールドは、21℃、相対湿度約30%に空調した室内で製造され、その後室温で少なくとも24時間にわたり真空下で乾燥された。スカフォールドの再現性に環境が及ぼす効果(湿度および温度)を分析するために、スカフォールドは高湿度条件(>85%の相対湿度)で時々スピンされ、真空下で乾燥されなかった。
3種のスカフォールドタイプ(各々10%のイブプロフェンを装填)からのイブプロフェンの放出は、UV/Vis分光計を用いてモニターされた。スカフォールドの25mg切片は、製造した各スカフォールドから切断し、3mLの純水中に浸けられた。222nmでの強度を1週間までの様々な時点に測定し(IBUのλmax)、これらの数値を事前に計算したイブプロフェン標準曲線と比較した。
イブプロフェンを含む、または含まないスカフォールドは、ティッシューパンチを用いて18mm径円板に切断され、それらを70%EtOHで緩徐にスプレーすることによって殺菌し、無菌フローキャビネット内で放置して乾燥させた。次にスカフォールドは、100IU/mLのペニシリンおよび100IU/mLのストレプトマイシンを有する1mLのリンガー液中に浸漬され、37℃でインキュベートされた。pHがスカフォールドの分解に及ぼす効果を試験するために、それらは3つの相違するpH(6、7.4および8.5)にあるリンガー液中に配置された。この溶液のサンプルは、pHの変化をモニターするために1日1回採取された。スカフォールドマットにおける物理的変化も、スカフォールドの光学顕微鏡写真を1日1回撮影することによって記録された。
A.正常ヒトケラチン生成細胞および線維芽細胞の培養
正常ヒト線維芽細胞は、以前に報告されたように単離して培養した(Ralston et al.,1997)。第3〜7継代の線維芽細胞を使用した。ケラチン生成細胞は、10%(容積/容積)のウシ胎児血清(FCS)、2mMのグルタミン、0.625μg/mLのアムホテリシンB、100IU/mLのペニシリンおよび100μg/mLのストレプトマイシンが補給されたダルベッコ(Dulbecco)の改変イーグル培地(DMEM)中で培養された。Azowipe(登録商標)スカフォールド(スチレンブタジエン・コポリマーと接着させた不織布ビスコースレーヨン・マイクロファイバースカフォールド、Vernon−Carus社、英国チョーリー(Chorley,UK))または75:25 PLGAスカフォールドいずれかの中での3D組織遺伝子組換え構築体の調製を、以前に記載されたように実施した(Blackwood et al.,Biomaterials.2008;29(21):3091−104)。
イブプロフェン(ナトリウム塩または異性体混合物;Sigma−Aldrich社、英国(UK))が細胞代謝活性に及ぼす効果は、イブプロフェンを細胞に直接加えることによって、または単層(2D)および3Dスカフォールドの両方においてスカフォールドからの放出を試験することによって調査された。2D分析のためには、3×104個のHDF細胞/ウエルを80%コンフルエントとなるまで24ウエルプレート内で培養した。3D分析のためには、3×105個のHDF細胞/ウエルを48時間にわたり2層のビスコースレーヨン・スカフォールド内の12ウエルプレート内で培養した。スカフォールド(イブプロフェンを含む、および含まない)は、スカフォールドが24時間は細胞と同一培地中に浸漬されるように、しかし線維芽細胞からは物理的に分離されるように、共培養インサートを使用して培養細胞の上方に配置された。細胞代謝活性(2Dまたは3Dで)を、MTT(3−(4,5−ジメチルチアゾール−2−イル)−2,5ジフェニルテトラゾリウムブロミド)(Sigma−Aldrich社、ミズーリ州セントルイス)を使用して評価した。簡潔には、細胞をPBS中で3回洗浄し、次にMTT溶液(PBS中の0.5mg/mLのMTT、各々24および12ウエルプレートの1ウエルに付き1および2mL)とともに37℃の加湿インキュベータ(5% CO2/95%空気)内で1時間にわたりインキュベートした。健常生存細胞中では、MTTは、ミトコンドリア酵素スクシニルデヒドロゲナーゼの活性によって紫色のホルマザン塩に還元される。60分後、溶液を吸引し、不溶性細胞内ホルマザン生成物を可溶化し、イソプロパノール(24ウエルプレートの1ウエルまたは1mL/cm2の培養組織に付き0.5mL)を加えることによって細胞から放出させ、10分間にわたりインキュベートした。次に540nmでの光学密度は、プレート読取り分光計を使用して測定した。
環境上のスピニング条件(湿度および温度)がスカフォールドの物理的構造および細胞成長に及ぼす効果を分析するために、様々な条件下でエレクトロスピンされたPLGAスカフォールド上の3D培養物をコラーゲン沈着について試験した。スカフォールド繊維上へのコラーゲン沈着を、Blackwood et al.,2008に記載されたように分析した。簡潔には、構築体を7日間にわたり成長させ、次にPBS中で3回洗浄し、4%ホルマリン中で固定した。PBSを用いた3回の洗浄後、構築体は18時間にわたり飽和ピクリン酸中のシリウスレッドF3B(C.I.35780、Direct Red 80、Sigma−Aldrich社)の0.1%溶液中でコラーゲン沈着について染色した。その後、構築体を、もはや赤色が溶出しなくなるまで水で洗浄した。次にデジタルカメラを用いてピクロシリウスレッドで染色したスカフォールドを撮影した。
100ng/mLの大腸菌(E.coli)血清型O111:B4(Sigma−Aldrich社)または1,000U/mLのTNF−α(Sigma−Aldrich社)由来のLPSを線維芽細胞内でのNF−κBの活性化についての陽性コントロールとして使用した。予備実験において、イブプロフェン(10−3および−4M、ナトリウム塩および異性体混合物;Sigma−Aldrich社、英国(UK))を使用してNF−κB活性化をダウンレギュレートさせた。次に10%イブプロフェン装填PLGAスカフォールド(75/25、50/50およびこれら2つの1:1ブレンド)を試験した。陰性コントロールを、正常DMEM中で細胞をインキュベートすることによって生成した。全実験は3回ずつ実施した。
結果間の有意差を評価するためにはStudent’sの対応のないt検定(unpaired t-test)を使用した。
A.材料
アセチルサリチル酸(アスピリン)およびポリ(D,L−ラクチド−コ−グリコリド)(75/25)は、Sigma Aldrich社から購入し、受領したままで使用した。
ジクロロメタン中のPLGA(75/25)の20重量%溶液に、10重量%のアセチルサリチル酸(ポリマーに対して)を加え、完全に溶解するまで撹拌すると、均質な溶液が得られた。エレクトロスピニング条件は、上述したようにイブプロフェン装填75/25 PLGA繊維性スカフォールドのための条件と同様であった。
繊維からのアスピリンの放出を、1週間の期間にわたり様々な時点にUV/Vis分光計(強度:240nm)を使用してモニターし、検量線値と比較した。
1.スカフォールドの合成および特性解析
A.イブプロフェンを用いたエレクトロスパン材料を製造するための再現性のスピニング条件の開発
PLGAスカフォールドを、イブプロフェンが均一に分散されている繊維を製造するために、ポリマーに対して1〜10重量%のイブプロフェンが装填されたDCM溶液からエレクトロスピンした(図1A、DおよびFを参照)。イブプロフェンの酸形の存在は、図1に示したように繊維性スカフォールドの外観を有意には変化させなかった。酸形を使用したのは、イブプロフェンのナトリウム塩がポリマーを処置するために適合する溶媒中に不溶性であったためである。後者はスピニング溶液中での薬物の均質な分散を生じさせることが見いだされ、スピニングすると、薬物は本プロセスに含まれる静電力のために繊維の表面に近付く傾向を有した(図1B参照)。
3種のスカフォールドタイプ(各々10%のイブプロフェンを含有)からのイブプロフェンの放出を、UV/Vis分光計を用いてモニターした。各実験のためにスカフォールドの25mgの切片を切削し、3mLの純水中に沈めた。これは1サンプルに付きおよそ2.5mgのイブプロフェンに相当し、これは50/50、75/25およびブレンド各々について4.1mM、3.9mMおよび3.2mMの最大計算濃度と等価である。サンプルからのイブプロフェンの放出を、様々な時点に222nmでUV/Vis分光計を用いてモニターし(イブプロフェンのλmax)検量線値と比較した。3種のスカフォールドについての放出プロファイルを、図3aおよびbに示した。
A.イブプロフェンへの細胞の細胞毒性および抗炎症性応答
最初に、細胞に対する細胞毒性を伴わずに抗炎症性になるイブプロフェンの濃度を決定することが必要であった。したがって、イブプロフェンナトリウム塩およびイブプロフェン異性体混合物(酸形)の両方をそれらが線維芽細胞の生存性に及ぼす効果について試験した(結果を表3に要約した)。両方の形態のイブプロフェンは同様に機能し、細胞は10−3M(p<0.05)までのイブプロフェン濃度を認容したが、10−2Mの濃度で細胞毒性応答を示した。その後の試験のためには、調査したイブプロフェンの濃度は10−3Mを超えなかった。
イブプロフェンへの24時間曝露がヒト線維芽細胞生存性に及ぼす効果は、MTT−ESTAアッセイを使用して評価した。示した結果は、3つずつのウエルの平均値±SEMである。コントロールとは相違する数値は、*p<0.05と表示した。
実験の次の段階は、イブプロフェンが装填されていない、および装填されている両方のPLLA−PGAスカフォールドを3Dスカフォールド内の線維芽細胞の培養の上方に配置することであった。PLGAスカフォールドが細胞生存性に及ぼす、および炎症性刺激に対する細胞の応答に及ぼす効果を次に調査した。装填されたスカフォールドは、2つの方法で試験した。一部は、イブプロフェンの任意の初期バースト放出をこれらの実験で得ることができるように、故意に洗浄せずに放置した。その他は70%EtOH中で強力に洗浄し、次に任意の緩やかに結合したイブプロフェンを除去するためにPBS中で3回洗浄した。
最後に、スカフォールド上の線維芽細胞の接着および増殖の比較は、線維芽細胞が、1、2および5重量%のイブプロフェンが装填されたスカフォールド上ならびに非装填スカフォールド上に接着して増殖したことを証明した(図9a)。しかし、10%イブプロフェンを用いたスカフォールドの装填は初期細胞接着を減少させたが、これは、24時間にわたりスカフォールドを洗浄することによって、(それらのイブプロフェンの一部を放出するスカフォールドと一致して)コントロールレベルへ部分的に(図9a参照)または完全に(図9b参照)回復した。
A.結果
スカフォールドの10mgサンプル(水3mL中)中での総濃度は1.68mMであり、1週間にわたってこのうちの37%しか放出されなかった。イブプロフェン装填スカフォールドと同様に、初期8時間くらいには初期迅速放出が生じ、その後にはより持続性でその後の7日間にわたる段階的放出が続いた。
PLGA繊維性スカフォールドからのアスピリンの放出プロファイルは、初期8時間にわたる初期迅速放出とともに、イブプロフェン装填スカフォールドについて見られた放出プロファイルと極めて類似であった。初期バースト後、繊維が分解し始めるにつれてより徐放性の放出が観察されるが、各場合においてスカフォールドは、図10に例示したように、水中に沈めてから7日後にあるパーセンテージ(イブプロフェンについてはおよそ10%およびアスピリンについては37%)しか放出しなかった。
Claims (50)
- エレクトロスパン・スカフォールドを含む創傷被覆材であって、前記スカフォールドが、生分解性ポリマーもしくはコポリマーと非ステロイド性抗炎症薬とを含み、前記ポリマーもしくはコポリマーと前記非ステロイド性抗炎症薬とが各々共通溶媒中で溶解できることを特徴とする創傷被覆材。
- 前記生分解性ポリマーもしくはコポリマーが、生体適合性である請求項1に記載の創傷被覆材。
- 前記生分解性ポリマーが、コラーゲン、ポリアルファエステル、ポリオルトエステル、ポリ無水物またはそれらのコポリマーである請求項1または2に記載の創傷被覆材。
- 前記ポリマーが、セルロースエーテル、セルロース、セルロース系エステル、フッ素化ポリエチレン、フェノール類、ポリ−4−メチルペンテン、ポリアクリロニトリル、ポリアミド、ポリアミドイミド、ポリアクリレート、ポリベンゾキサゾール、ポリカーボネート、ポリシアノアリールエーテル、ポリエステル、ポリエステルカーボネート、ポリエーテル、ポリエーテルエーテルケトン、ポリエーテルイミド、ポリエーテルケトン、ポリエーテルスルホン、ポリエチレン、ポリフルオロオレフィン、ポリイミド、ポリオレフィン、ポリオキサジアゾール、酸化ポリフェニレン、硫化ポリフェニレン、ポリプロピレン、ポリスチレン、ポリスルフィド、ポリスルホン、ポリテトラフルオロエチレン、ポリチオエーテル、ポリトリアゾール、ポリウレタン、ポリビニル、フッ化ポリビニリデン、再生セルロース、シリコーン、尿素ホルムアルデヒド、またはそれらのコポリマーからなる群から選択される請求項1または2に記載の創傷被覆材。
- 前記ポリマーが、ポリ乳酸である請求項3に記載の創傷被覆材。
- 前記ポリマーが、ポリグリコール酸である請求項3に記載の創傷被覆材。
- 前記コポリマーが、ポリ乳酸とポリグリコール酸のコポリマーである請求項3に記載の創傷被覆材。
- 前記コポリマーが、ポリ(D,L−乳酸−コ−グリコリド)である請求項7に記載の創傷被覆材。
- 前記ポリ(D,L−乳酸−コ−グリコリド)が、75:25のラクチド対グリコリドの比率を有する請求項8に記載の創傷被覆材。
- 前記ポリ(D,L−乳酸−コ−グリコリド)が、50:50のラクチド対グリコリドの比率を有する請求項8に記載の創傷被覆材。
- 前記NSAIDが、プロピオン酸系NSAIDである請求項1〜10のいずれか一項に記載の創傷被覆材。
- 前記NSAIDが、[2−(4−イソブチルフェニル)プロピオン酸]である請求項11に記載の創傷被覆材。
- 前記NSAIDが、アセチルサリチル酸酸である請求項1〜10のいずれか一項記載の創傷被覆材。
- 前記スカフォールドが、約5重量%〜約30重量%のNSAIDを含む請求項1〜13のいずれか一項に記載の創傷被覆材。
- 前記スカフォールドが、約10重量%〜約20重量%のNSAIDを含む請求項14に記載の創傷被覆材。
- 前記スカフォールドが、約10重量%のNSAIDを含む請求項14に記載の創傷被覆材。
- 生分解性ポリマーもしくはコポリマーと非ステロイド性抗炎症薬とを含むエレクトロスパン・スカフォールドであって、前記ポリマーもしくはコポリマーと前記非ステロイド性抗炎症薬とが、各々共通溶媒中で溶解できるエレクトロスパン・スカフォールド。
- 医薬品として使用するための生分解性ポリマーもしくはコポリマーと非ステロイド性抗炎症薬とを含むエレクトロスパン・スカフォールドであって、前記ポリマーもしくはコポリマーと前記非ステロイド性抗炎症薬とが、各々共通溶媒中で溶解できるエレクトロスパン・スカフォールド。
- 創傷の治療に使用するための生分解性ポリマーもしくはコポリマーと非ステロイド性抗炎症薬とを含むエレクトロスパン・スカフォールドであって、前記ポリマーもしくはコポリマーと非ステロイド性抗炎症薬とが、各々共通溶媒中で溶解するできるエレクトロスパン・スカフォールド。
- 前記創傷が慢性創傷であるか、または前記創傷が急性創傷である請求項19に記載のエレクトロスパン・スカフォールド。
- 疼痛または炎症の治療に使用するための生分解性ポリマーもしくはコポリマーと非ステロイド性抗炎症薬とを含むエレクトロスパン・スカフォールドであって、前記ポリマーもしくはコポリマーと前記非ステロイド性抗炎症薬が、各々共通溶媒中で溶解できるエレクトロスパン・スカフォールド。
- 前記生分解性ポリマーもしくはコポリマーが、生体適合性である請求項17〜21のいずれか一項に記載のエレクトロスパン・スカフォールド。

- 前記コポリマーが、ポリ(D,L−乳酸−コ−グリコリド)である請求項27に記載のエレクトロスパン・スカフォールド。
- 前記ポリ(D,L−乳酸−コ−グリコリド)が、75:25のラクチド対グリコリドの比率を有する請求項28に記載のエレクトロスパン・スカフォールド。

- 前記創傷被覆材またはエレクトロスパン・スカフォールドが、創傷に適用される請求項37に記載の方法。
- 創傷治癒において使用するためのエレクトロスパン・スカフォールドを製造する方法であって:
i)ポリマーもしくはコポリマーとNSAIDとを共通溶媒中に溶解させるステップ;および
ii)ステップi)の結果として生じた溶液をエレクトロスピンするステップ
を含む方法。 - 請求項39に記載の方法にしたがって製造されたエレクトロスパン・スカフォールド。
- 医薬品として使用するための、請求項40に記載のエレクトロスパン・スカフォールド。
- 創傷の治療において使用するための、請求項40に記載のエレクトロスパン・スカフォールド。
- 前記創傷が慢性創傷である、または前記創傷が急性創傷である請求項42に記載のエレクトロスパン・スカフォールド。
- 疼痛または炎症の治療において使用するための請求項40に記載のエレクトロスパン・スカフォールド。
- ポリマーもしくはコポリマーを含むエレクトロスパン・スカフォールドの分解を触媒するための非ステロイド性抗炎症薬の使用。
- 前記非ステロイド性抗炎症薬が、前記ポリマーもしくはコポリマーと一緒にエレクトロスピンされる請求項45に記載の使用。
