Abstract
Description
本発明の実施例では、上記核酸アプタマはデオキシリボ核酸アプタマから成る。
本発明はさらに、下記(a)〜(c)の工程から成る凝固タンパクの精製方法にも関するものである:
(a) 凝固タンパクを含むサンプルを上記定義の親和性基質と接触させて (i) 親和性基質に固定された核酸アプタマと(ii)上記凝固タンパクとの間に錯体を形成させ、
(b) 段階(a)で形成させた錯体からタンパクを遊離させ、
(c) 凝固タンパクを精製させた形で回収する。
(1)本発明の親和性基質を用いることで非常に高い純度を有する凝固タンパクの精製方法を実行できる。
(2)本発明の親和性基質は望ましくない物質、特に核酸アプタマ分子を精製された凝固タンパクを含む溶出溶液中に放出しない。
(3)核酸アプタマ分子の離脱はヒトの健康に対する欠点にならない。
(4)親和性基質での当該凝固タンパクの固定とその後の溶出によって、酵素活性を有する凝固タンパクの場合、凝固タンパクの活性形を検出可能な量形成することはない。
(5)親和性基質は比較的安価に製造できる、特に、核酸アプタマの生産コストが安い。
(6)凝固タンパクの精製に親和性基質を使用すること自体が相対的に安価である。すなわち、基質を何度でも再生でき、長期間使用できる。

以下、本発明の実施例を示す。
親和性基質の製造
親和性基質はストレプトアビジン(streptavidin-agarose−Novagen、登録商標)がグラフトされたマトリックスから成る固体の基質材料から製造した。
1mlのカラムから成る容器に1ml容積(すなわち11mm)のゲルを入れた。ゲルを精製水で洗浄して貯蔵用溶剤を除去した。
ゲルの特徴は以下の通り:
(1)ビオチン吸着能:≧85ナノモル/1mlゲル
(2)機能テスト:ATで30分間のコースでビオチン標識化したトロンビンのキャプチャー>99%、
(3)その他のテスト:プロテアーゼ無し、エンド/エクソヌクレアーゼ無し、RNase無し
(4)防腐剤:100mMリン酸ナトリウムpH7.5+NaN3 0.02
配列SEQID番号86のビオチン標識化した抗ヒトFVII核酸アプタマを精製水中に0.5mg/0.187mlの最終濃度(すなわち最終モル濃度=0.1mM)で可溶化した。核酸アプタマ溶液を標準サイクルに従って95℃で活性化して固体基質材料上にアプタマを固定化した。
核酸アプタマ溶液を4.8mlの精製水で、次に1.5mlのMe++緩衝液(5×濃縮)で予備希釈した。
吸光度検出器を1AUFS(吸光度単位フルスケール)に合わせ、254nmでこの溶液のODを0.575U254で記録した。
ビオチン標識化した核酸アプタマ溶液は予めパックしたストレプトアビジン−アダロースゲル上へ注入し、ペリスタル型ポンプで2.5ml/分の流速で再循環させた(すなわちゲル(入口/出口 I/O)上での接触時間=24秒)。上記条件での254nmでのODは直ちに0.05AU254(すなわち理論的なカップリング値の91%)に到達した。すなわち1ミリリットルのゲル当たり0.455mgの核酸アプタマで安定する。
10mM−CaCl2+4mM−MgCl2緩衝液で洗浄し、次いで2M−NaClで洗浄して固体基質材料上にグラフトしたストレプトアビジン分子に特異的に結合していない核酸アプタマを除去した。
組換え型ヒト因子VIIの精製方法
上記のアプタマ親和性基質を下記文献に記載の方法で調製した精製されたFVll/FVllaを使用してテストした。
出発材料の生物物質は組換え型ヒトFVIIを含むトランスジェニクウサギ乳汁である。発現カセットはβ−カゼイン遺伝子プロモータの制御下にあるヒトFVIIトランスジーンから成る。簡単に言うと、140ミリリットルの乳汁を出生後の日4〜日12の間の初乳汁分泌を2匹のウサギから集め、集めた乳汁のアミド分解(amidolytic)FVII(生物学的に活性化されたFVII)の平均タイター値は928IU/mlである。乳汁は−80℃の温度で保存した。
テスト時にはウサギ乳汁を水浴中で37℃の温度で解凍し、クエン酸ナトリウム溶液で希釈してpH7.5での最終クエン酸塩濃度を62g/lにした。このクエン酸ナトリウム処理で燐酸カルシウム(phosphocalcic)カゼインミセルを不安定できる。乳汁のリピドリッチなタンパク溶液は一連の濾過器(それぞれ(i)15〜0.5μm気孔率閾値の深層濾過器と(i)0.2μmメンブランフィルタ)で精製する。
上記段階の終わりに得られた17.5mgの低純度FVII(〜5%)は容積20mlのQ-sepharoseXL(登録商標)(GE Healthcare)を使用したイオン交換クロマトグラフィで精製する。ヒトFVIIは、78ml容積の5mM−塩化カルシウムを含む緩衝液で溶出する。アミド分解(amidolytic)FVII濃度は337IU/mlすなわち0.17mgのFVII/mlで、280nm、ε1%=13でODで測定した全タンパクは0.18mg/ml、と見積もられる。従って、FVII純度は94%。
しかし、人間で繰り返して注射する場合には、遺伝子組換えタンパクで許容される外来タンパクに対する負荷は50ppmを超えてはならない。すなわち純度>99.995%でなければならない。この純度をアフィニティーマトリックスでの精製だけで達成できるように見える。
上記段階の終わりに得られた精製されたヒトFVII溶液の6mlの容積(FVIIの1.1mg)を用いて本発明の親和性基質で高純度の組換え型ヒトFVIIに精製する段階を行う。上記段階で得られたFVII溶液を4mM−MgCl2、10mM−CaCl2でpH7.5に予備調節し、アプタマ−アガロースゲル(親和性基質)上にペリスタル型ポンプで0.1ml/分の流速で注入する(すなわち、親和性基質との接触時間10分)(I/O)。
注射後、ゲルを50mMトリス+50mM−NaCl+4mMMgCl2+10mMCaCl2、pH7.5緩衝液で洗浄する。10mlの非吸着溶液を回収する。FVIIは50mMトリス+10mM−EDTA、pH7.5緩衝液で溶出させる。溶出ピークの収集はODプロフィルに従って実行する。モル計算から親和性基質に固定された核酸アプタマの量は17ナノモルで、これはFVII分子とのモル−モル相互作用で、FVIIの0.9mgの親和性基質の絶対キャパシティに対応する。
この[図1]で注射(1)後の吸収曲線の屈曲(2)は組換え型ヒトFVIIと親和性基質の飽和開始を示す。時間(3)で組換え型ヒトFVIIの注射を停止する。[図1]の直線状の時間のスケールを示すと、注射開始時間(1)と注射終了時間(2)との間の持続時間は10分である。親和性基質は上記凝固タンパクで飽和し続け、(i)親和性基質の抗FVII核酸アプタマと(ii)被精製組成物中に最初に含まれていた組換え型ヒトFVII分子との間に錯体が形成される。被精製組成物をカラムに送り、カラムを緩衝液で洗浄する(洗浄段階6)。その後、時間(4)で10mMの最終EDTA濃度から成る緩衝液を注入して溶出段階を実行する。吸収ピークは核酸アプタマ/組換え型FVII錯体から組換え型ヒトFVIIが開放されたことを示している。組換え型ヒトFVIIの分子は迅速にリリースされ、従って、少容積である点に注意されたい。従って、本発明の親和性基質によって組換え型ヒトFVIIタンパクの高濃度の溶出溶液が得られる。時間(5)で50mMトリス緩衝液を用いて親和性基質を再生させる段階を実行する。(7)で見られる吸光度ピークは再生段階で親和性基質から開放された物質に対応する。
下記の[表1]に示すテスト結果は0.45〜0.49mg/mlの親和性マトリックスすなわちバイオアベイラブル(bioavailable)なリガンドの50〜55%の動的結合能を示す。EDTAで約75%の動的溶出が計算される。
親和性基質の特異性をウサギ乳タンパクに特異的なELISAアッセイで評価した。結果は[表2]に示した。
溶出前に2M−NaClおよび/またはプロピレングリコールまたは50%エチレングリコール溶液で中間洗浄すると結果はより良くなる(これらの条件ではFVIIは溶出されない)。
ヒト血漿IX因子の精製方法
A.材料および方法
A.1 アフィニティークロマトグラフィ基質
「Mapt−1」親和性ゲル材料、スペーサ鎖無し、理論リガンド濃度:0.46mg/ml:容積1ml
アプタマはクロマトグラフィ基質材料に化学共役反応で直接結合した。使用したアプタマは配列SEQ ID番号101のアプタマである。
出発材料はヒト血漿由来のIX因子がリッチな組成物から成り、これはLFB社から「Betafact」の名称で市販されている。出発材料を注入した:Betafact MPVP(血漿FIX 、60%純度)、ゲルml当たりIX因子を200IU負荷(すなわち800μg)、接触時間10分。
ゲル平衡化: 0.050 M トリス-HCl、0.010のM CaCl2、pH 7.5、
溶出: 0.020 M トリス-HCl、0.010のM EDTA、pH 7.5、
再生: 0.020 M トリス-HCl、1 M NaCl、50%プロピレングリコール、pH 7.5
タンパクピークは280nmの波長で吸光度値を測定して検出。
10-ウエルNOVEXゲル(Invitrogen)、4-12%、Bis-トリス;MESランニング緩衝液、200Vで5分間マイグレーション、CBB(G250)またはAgNO3(GEキット)で染色。
結果は[図2]および[図3]に示した。
[図2]は時間を関数とする280nmで測定した吸光度曲線。[図3]のピーク番号1はカラムに保持されなかった出発生成物の分画に対応する。ピーク番号2は溶出分画に対応する。ピーク番号3は再生段階を実行することによって基質から脱着された画分に対応。
[図3]はSDSポリアクリルアミドゲル電気泳動ゲルの画像。[図3]のゲルの左から右に向かって各レーンは下記の出発材料のマイグレーション結果を表す:
レーン「MD」:「Betafact(登録商標)」出発組成物、
レーン「NR」:[図2]のクロマトグラフプロフィルのピークNo.1に対応する保持されない画分、
レーン「EI」:[図2]のクロマトグラフ・プロフィルのピークNo.2に対応する溶出分画。
上記溶出液は優れた電気泳動純度を示し、FIXの官能が維持されることで特徴付けられると結論付けることができる。この実験はクロマトグラフ基質に直接結合した配列SEQ ID番号101のアプタマの能力を示し、従って、スペーサ無しで、血漿不純物を含む錯体培地中のFIXに結合し、精製するという能力を示している。
トランスジェニック雌豚の乳汁抽出物に含まれる組換え型IX因子の精製法
A.材料および方法
A.1 アフィニティークロマトグラフィ基質
スペーサ鎖無しに、Mapt-1アプタマが直接結合で固定された親和性ゲル材料。理論的なリガンド濃度0.46mg/ml:容積1ml。
Mapt-1アプタマは化学的カップリング反応でクロマトグラフィ基質材料に直接結合される。
使用したアプタマは配列SEQ ID番号101のMapt Iアプタマである。
出発材料は清澄し、MEP HyperCel:1.8%純度で精製したトランスジェニック雌豚の乳汁から成る。クエン酸ナトリウムを除去するためにこのサンプルを樹脂の吸着/平衡用緩衝液に対して透析した。1ml当たり302IU(すなわち1200μg)のIX因子を負荷。
ゲル平衡化: 0.050 M トリス-HCl、0.010のM CaCl2、pH 7.5、
溶出: 0.020 M トリス-HCl、0.010のM EDTA、pH 7.5、
再生: 0.020 M トリス-HCl、1M NaCl、50%プロピレングリコール、pH 7.5。
サンプルを0.1ml/mmの流速で10分間注入し、それからゲルを5分間、0.5ml/mmで洗浄する。溶出および再生は2mlの緩衝液を0.5ml/mmの流速で注入して実行する。タンパクピークは280nmの波長での吸光度値を測定して検出する。
10-ウエルNOVEXゲル(Invitrogen)、4〜12%、Bis-トリス;MESランニング緩衝液、200Vで50分間移行。CBB(G250)またはAgNO3(GEキット)で染色。
FIXの量(抗原量)の測定はるSerachrom 0943 FIX Agキット(Stago)を用い、供給者の推薦条件に従って実行した。FIXの酵素活性の測定はBiophen FIXキット 参照番号221802(ハイフンBioMed)で供給元の推薦に従って比色テストで実行した。比活性は下記の比に従って計算される:FIXの酵素活性/FIXの量。
結果は[図4]および[図5]に示す。
[図4]は時間を関数とした280nmでの吸光度の測定値の曲線を表す。[図4]でピークNo.1はカラムに保持されなかった出発材料の画分に対応し、ピークNo.2は溶出画分に対応し、ピークNo.3は再生段階を実行時に基質から脱着した画分に対応する。[図4]の結果は溶出ピークが非常に幅が狭いことを示し、それはアプタマが固定されたクロマトグラフィ基質がヒト・IX因子に対して非常に高い特異性を有することを示す。
[図5]はSDSポリアクリルアミドゲル電気泳動ゲルの像である。[図5]のゲルの左から右に向かって各レーンは下記の出発材料の移行結果を表す:
レーン「E6」:[図4]のクロマトグラフプロフィルのピークNo.1に対応する保持されない画分
レーン「E7」:[図4]のクロマトグラフプロフィルのピークNo.2に対応する溶出画分
レーン「E8」:ピークNo.2の顆粒で、[図4]のクロマトグラフプロフィルのピークNo. 3の上流が集められた溶出画分。
レーン「T FIX」:精製されたIX因子対照(コントロール)。
この溶出液は出発材料と比較して電気泳動純度を大幅な純度ゲイン(>26倍)で示すと結論できる。溶出液中で同定された第2バンドはFIXの他の形に対応する。
ヒト血漿第VII因子の精製方法
A.材料および方法
A.1 親和性クロマトグラフ基質
ビオチンとアプタマとの間にスペーサ鎖なしでビオチンに直接結合した「Mapt-2コア」アプタマを固定した親和性ゲル材料。アプタマは5'−末端ビオチンによってストレプトアビジン・ゲル(供給元Novagen)に固定されている。0.4mg/mlの理論的リガンド濃度:容積1ml。
使用したアプタマは配列SEQ ID番号20のMapt-2コア・アプタマである。
出発材料は98%まで精製したヒト血漿第VII因子組成物から成る。
ゲル平衡化:0.050 M トリス-HCl、0.010M CaCl2、0.05mM MgCl2、pH 7.5、
溶出:0.020 Mトリス-HCl、0.010M EDTA、pH 7.5。
平衡緩衝液中の98%まで精製したヒト血漿第VII因子の240μgを0.5ml/mmの流速で注入。保持されない材料のピークを検出後、溶出緩衝液の2カラム容積を注入。タンパクのピークは280nmの波長での吸光度値を測定して検出。
結果は[図6]および[図7]に示す。[図6]は時間を関数とする280nmでの吸光度の測定値の曲線を表す。[図6]のピークNo.1はカラムに保持されなかった出発材料の画分に対応し、ピークNo.2は溶出分画に対応する。
不純物の除去適度を視覚化するために出発材料および溶出物を硝酸銀染色してSDSポリアクリルアミドゲル電気泳動によって分析した。[図7]はこのゲルを表し、レーンNo.1は出発材料の画分、レーンNo.2は溶出画分に対応する。
出発材料はかなり純度が高いのにかかわらず、溶出画分は不純物を含まず、劣化もない点に注意。
[図6]および[図7]の結果は配列SEQ ID番号20のアプタマはヒトFVIIに結合でき、EDTAの存在下で特異的に溶出できることを示す。
ウサギFVIIへのアプタマの結合非存在
A. 材料および方法
A.1 アフィニティークロマトグラフィ基質
配列SEQ ID番号86のアプタマが5'−末端ビオチンを介してスペーサ鎖(供給元Novagen)により固定された、ストレプトアビジンに結合された親和性ゲル。0.35mg/mlの理論的リガンド濃度:容積1ml。
使用したアプタマは配列SEQ ID番号86のアプタマである
ウサギ血漿から精製して得たウサギFVIIリッチな濃縮ヒドロキシアパタイトの溶出液、接触時間10分、流速0.5ml/mm。
ゲル平衡化:0.050 Mトリス-HCl、0.010M CaCl2、0.05mM MgSO3、pH 7.5、
溶出:0.050 Mトリス-HCl、0.010M EDTA、pH 7.5、
36μgを10分の接触時間でゲルに注入する。溶出は2mlの溶出緩衝液を注入して実行。タンパクのピークは280nmの波長での吸光度値を測定して検出。
アミド分解(amldolytic)活性のために各画分をStagoキットを使用してクロマトアッセイによって分析。供給元の推薦条件に従う(因子Vlla StatClotキット)。アミド分解活性を各画分に含まれるFVIIのμgに変換。
Biacoreでのアプタマとヒト因子VIIとの相互作用のための特定プロトコルの実施例(NaCl耐性)
A. 材料および方法
本発明の配列SEQ ID番号86の核酸アプタマ分子(「Mapt2」という)が固定された固体の基質を製造した。
固体基質に結合する前にMapt2アプタマの5’末端を化学的に変成してPEG(C18)の4つの分子から成るスペーサ鎖に連結した。それから、アプタマに連結した末端とは反対側のスペーサ鎖の遊離末端にビオチン分子を連結した。
固定したストレプトアビジン分子を含む固体基質は提供された(Series S sensor ChipSA.GE)。
次いで、上記アプタマ化合物と接触させて、基質のストレプトアビジン分子とアプタマ化合物のビオチン分子との間の非共役会合によって上記固体基質を配列SEQID番号86の核酸を固定する。
Mapt2アプタマは3772RU(1RUは単位mm2当たり固定した生成物が約1pgに対応)の固定化度で固定された。
サンプルはビオチン−ストレプトアビジン相互作用によって固定されたMapt2アプタマを含むチップ(固体基質)上に順次注入した。次に、NaClの濃度を上げた緩衝液を固体基質上へ注入された。(1M NaClから3M NaClまでの3シリーロを注入)。全ての注入は30μl/分の流速で60秒かけて注入した。NaClを含む上記の3シリーズの緩衝液の注入後、溶出緩衝液(10mM EDTA)を30μl/分の流速で75秒間かけて注入してアプタマからFVII HPTGを分離した。
解析はRPS Biacore T100装置(GE)で実行した。記録された相互作用のモデリングはBiaevaluationソフトウェア(GE)を使用して実行した。トランスジェニック・ヒトFVIIに対するMapt2固定アプタマの結合曲線はBiacore(登録商標)制御ソフトウェア(バージョン1.2)の専用モジュールで計算した。
このヒトFVIIに対するMapt2アプタマの結合結果から、ヒトFVIIへのMapt2アプタマの結合はNaClを含む緩衝液の注入によって有害な状態で変化しないことということが確認できる。
結果は[図8]に示した。
Biacore上のアプタマとヒト因子VIIとの相互作用のためのプロトコルの特定実施例(プロピレングリコール耐性)
A.材料および方法
本発明の配列SEQ ID番号86の核酸アプタマ分子(「Mapt2」という)が固定された固体の基質を製造した。
固体基質に結合する前にMapt2アプタマの5’末端を化学的に変成してPEG(C18)の4つの分子から成るスペーサ鎖に連結した。それから、アプタマに連結した末端とは反対側のスペーサ鎖の遊離末端にビオチン分子を連結した。
固定したストレプトアビジン分子を含む固体基質は提供された(Series S sensor ChipSA.GE)。
次いで、上記アプタマ化合物と接触させて、基質のストレプトアビジン分子とアプタマ化合物のビオチン分子との間の非共役会合によって上記固体基質を配列SEQID番号86の核酸を固定する。
Mapt2アプタマは5319RU(1RUは単位mm2当たり固定した生成物が約1pgに対応)の固定化度で固定された。
サンプルはビオチン−ストレプトアビジン相互作用によって固定されたMapt2アプタマを含むチップ(固体基質)上に順次注入した。次に、50%プロピレングリコールを含む緩衝液を固体基質上へ注入した。全ての注入は30μl/分の流速で60秒かけて注入した。50%プロピレングリコールを含む緩衝液を注入後、溶出緩衝液(10mM EDTA)を30μl/分の流速で75秒間かけて注入してアプタマからFVII HPTGを分離した。
解析はRPS Biacore T100装置(GE)で実行した。記録された相互作用のモデリングはBiaevaluationソフトウェア(GE)を使用して実行した。トランスジェニック・ヒトFVIIに対するMapt2固定アプタマの結合曲線はBiacore(登録商標)制御ソフトウェア(バージョン1.2)の専用モジュールで計算した。
このヒトFVIIに対するMapt2アプタマの結合結果から、ヒトFVIIへのMapt2アプタマの結合はプロピレングリコールを含む緩衝液の注入によって有害な状態で変化しないことということが確認できる。
結果は[図9]に示した。

- 固体の基質材料を含む、凝固タンパク(coagulation protein)を選択的に結合させるための親和性基質(affinity substrate)であって、上記の固体の基質材料上に凝固タンパクに特異的に結合する核酸アプタマ(nucleic adaptamer)が固定されている親和性基質。
- 核酸アプタマがデオキシリボ核酸アプタマ(deoxyribonucleic adaptamer)から成る請求項1に記載の親和性基質。
- 核酸アプタマが式(I)の化合物の構造中に含まれる請求項1または2に記載の親和性基質:

- 下記の(a)〜(c)の工程を有する凝固タンパクの精製方法:
(a) 請求項1〜6のいずれか一項に記載の親和性基質と凝固タンパクを含むサンプルとを接触させて、(i) 上記親和性基質上に固定した核酸アプタマと(ii) 凝固タンパクとの間に錯体を形成し、
(b) 段階(a)で形成された錯体からタンパクをフリーにし、
(c)凝固タンパクを精製された形で回収する。 - 前記サンプルが人の血漿またはその画分、上記凝固タンパクに対してトランスジェニックな哺乳類の乳汁またはその画分の中から選択される請求項8に記載の方法。
- 上記血漿タンパクがヒトの血液である請求項8または9に記載の方法。
- サンプルが少なくとも一種のヒト以外の血液の血漿タンパクを含む請求項10に記載の方法。
- 人の血液の血漿タンパクがヒト以外の血漿タンパクのホモローグ(相同物)である請求項11に記載の方法。
- 人の血液の血漿タンパクがヒト以外の血漿タンパクのホモローグ(相同物)である請求項12に記載の方法。
- 親和性基質を二価-イオンキレート剤、好ましくはEDTAを含む溶出緩衝液と接触させて段階(b)を実行する請求項8〜13のいずれか一項に記載の方法。
- 少なくとも99.9重量%がヒト以外のタンパクを実質的に含まない組換え型ヒトタンパク殻成る組換え型ヒト凝固タンパクの精製された組成物。
- 請求項15に記載の組換え型ヒト凝固タンパクの精製された組成物を含む医薬組成物。
