ユビキチンシステムは、抗原処理、アポトーシス、細胞周期及び分裂、DNA修復、分化及び発達、エンドサイトーシス及びエキソサイトーシス、免疫応答及び炎症、筋肉変性、神経変性、神経発達、細胞小器官発達、タンパク質分解、細胞表面の受容体及びチャネルからの信号伝達、ストレス反応、及び転写のような生物学的プロセスの多数に関与している。
「モジュレーター」という用語は、ユビキチンリガーゼ活性を増加(「活性剤」)または減少(「阻害剤」)できる化合物を意味する。「候補」、「候補薬剤」、「候補モジュレーター」及び「候補ユビキチンリガーゼ活性モジュレーター」は、例えばタンパク質(ここでは、タンパク質、ポリぺプチド、及びペプチドを含む)、小さな有機または無機分子、多糖類、ポリヌクレオチドなどの任意の分子を参照する。これらの分子は、ユビキチンリガーゼ活性モジュレーター活性についてテストされる。候補薬剤は、多くの化学的分類を包含する。好ましい実施形態においては、候補薬剤は、有機分子、特に小さな有機分子であって、タンパク質と構造的に相互作用するのに必要な官能基、特に水素結合、及び典型的には、少なくともアミン、カルボニル、ヒドロキシル基またはカルボキシル基、好ましくは少なくとも二つの化学官能基から構成される。候補薬剤はしばしば環状炭素または複素環構造及び/または芳香族または一つまたは一つ以上の化学官能基で置換された芳香族構造から構成される。
特定の実施形態は、ユビキチン化システムの化合物を組み合わせる方法を提供する。「組み合わせる」とは、ユビキチンリガーゼ活性が行える条件下のレセプタクル内で様々な化合物の追加を意味する。好適な実施形態では、前記レセプタクルは、96ウェルプレートまたは他の市販のマルチウェルプレートである。他の好適な実施形態では、前記レセプタクルはFACSの機械の反応槽である。他のレセプタクルは、限定されるわけではないが、384ウェルプレート及び1536ウェルプレートである。まだ他の適したレセプタクルは当業者に明らかであろう。
さらなる実施形態では、ユビチキンリガーゼアッセイの一つまたはそれ以上の成分は標識を含む。「標識」とは、付着成分の同定または単離に有用な付着単一分子または複数の分子を意味する。標識を有する成分は「標識−X」と呼び、前記Xは、前記成分である。例えば、標識を含むユビキチンは、ここでは「標識−ユビキチン」と呼ばれる。好適には、標識は付着成分と共有結合している。結合成分の一つ以上が標識を有する場合、前記標識は識別のために、例えば「標識1−ユビキチン」というように番号付けされる。好適な標識は、これに限定はされないが、ラベル、結合対のパートナー、及び分子と結合した表面基質を含む。当業者にとって明らかであるように、多くの分子は、標識が使用される方法に応じて、標識の複数のタイプとして使用されているのが見出される。
アミノ末端6xHiS−SUMO−UBA2は以下のように構成されている。PCR産物は、ゲノムDNA(S.セレビシエ)からテンプレートとして酵母RAD23遺伝子(酵母Rad23タンパク質の351−398残基)を用いる5’−GATCGGTCTCAAGGTGTTGACTATACCCCCGAAGA−3’(配列ID番号:4)及び5’−GATCGGATCCTCAGTCGGCATGATCGCTGA−3’(配列ID番号:5)プライマーを使用して生成される。PCR断片は、BsaI及びBamHIで消化され、及びp6xHiS−SUMOプラスミド内(Life Sensors)に結合される。前記プラスミドは、正しい配列(Genewiz)の存在を確認するよう配列される。図IAは下線が引かれたUBA2領域と6xHiS−SUMO−UBA2のアミノ酸配列を示している。
6xHiS−SUMO−UBA2は50mMのMOPSと4℃で一晩かけて透析される(透析チュービングMWCO12〜14,000)。6xHiS−SUMO−UBA2の2mlはアフィゲル15の4ml内に移され、撹拌機で、4℃で4時間撹拌される。1Mエタノールアミン塩酸塩(pH8.0)が前記アフィゲル上の任意の活性エステルをブロックするために添加され、撹拌機上で室温にて1時間保温される。これを水で洗浄し、続いて2M塩化ナトリウムで洗浄し、及び、4℃で20%エタノール内に貯蔵する。
E3リガーゼアッセイは、500nMの6xHis−SUMO−CARP2、50nMのE1(BIOMOL)、150mMのUbcH5c(E2)(BIOMOL)、500んgのユビキチン、2mMのATP、50mMのpH8.0のトリス塩酸緩衝溶液、5mMの塩化マグネシウム、2mMのDTTからなる。ほとんどの場合において、ユビキチンは6xHis標識を含むことがある。CARP2をコード化したPCR産物は、5’−GATCCGTCTCAAGGTATGTGGGCAACCTGCTGCAA−3’(配列ID番号:10)及び5’−GATCGGATCCTCAGGACCGGAAGACATGCA−3’(配列ID番号:11)プライマー及びテンプレートとしてヒトCARP2 cDNAを用いて生成する。前記PCR断片はBsmBI及びBamHIで消化され、及び、その後、6xHis−SUMO−CARP2を生じるようp6xHis−SUMOプラスミド(LifeSensors)内に結合する。前記プラスミドは、正しい配列の存在を確認するよう配列される。反応混合物の30μlは、37℃で90分間保温され、及び、6xHis−SUMO−UBA2に結合されたアフィゲルの総容積50μlを含む3mlカラムに移される。前記カラムは洗浄バッファー(Ixリン酸緩衝生理食塩水(PBS))の5カラムボリュームで洗浄され、及び、溶出バッファー同様に変性バッファー(それぞれ、50mMトリス塩酸緩衝溶液、pH8.0、1M塩化ナトリウム及びトリス塩酸緩衝溶液、pH8.0、1M塩化ナトリウム、6MUREA)の5カラムボリュームと溶出される。洗浄、溶出及び変性バッファーの通過画分からの一定分量は、4〜20%SDS−PAGE勾配ゲル上に積まれ、ウェスタンブロット法により、アンチユビキチン抗体(Sigma)及び抗体と共役したアンチラビットHRP(Jackson Immuno Research)と探査される(図2)。
高結合モジュラープレートは、6xHis−SUMO−UBA2の溶液0.1mg/mlの100μlと4℃で一晩かけてコーティングされる。前記プレートはその後、1×PBS中3%BSAと室温にて3時間保温され、及び、1×PBSと3回洗浄される。0.5μgユビキチンの30μl及びK48ポリユビキチン鎖(BIOMOL)の0.5μgは、プレートコーティング及び2時間保温された6xHis−SUMO−UBA2に移される。前記ウェルは、1×PBSと3回洗浄される。アンチユビキチン抗体の100μl(1×PBS中0.3%BSAの1:10希釈)が前記ウェルに添加され、1時間保温される。プレートは、1×PBSと3回洗浄される。FITC標識アンチラビット抗体溶液の100μl(1×PBS中3%BSAの1:50希釈)が添加される。前記プレートは室温にて1時間保温され、前記ウェルは1×PBSと7回洗浄される。蛍光プレートリーダー(Perkin Elmer Envision)を使用すると、それぞれ485nm及び535nmの励起及び発光波長を使用することで蛍光が検知される。図3Aのデータは、プレートにコーティングされたUBA2がポリユビキチンとモノユビキチンとを識別することを示している。
高結合モジュラープレートは、6xHis−SUMO−UBA2の0.1mg/ml溶液の100μlと4℃で一晩かけてコーティングされる。前記プレートは、1×PBS中3%BSAと室温で3時間保温され、1×PBSと3回洗浄される。K48結合型及びK63結合型ユビキチン鎖(BIOMOL)は、30μl内で5μgから1ngの範囲の濃度で添加される。2時間の保温後、前記ウェルは1×PBSと3回洗浄される。アンチユビキチン抗体の100μl(1×PBS中0.3%BSAの1:10希釈)がウェルに添加され、1時間保温される。プレートは1×PBSと3回洗浄される。FITC標識アンチラビット抗体溶液の100μl(1×PBS中3%BSAの1:50希釈)が添加される。前記プレートは室温で1時間保温され、前記ウェルは1×PBSと7回洗浄される。蛍光プレートリーダー(Perkin Elmer Envision)を使用すると、それぞれ485nm及び535nmの励起及び発光波長で蛍光が検知される。図3Bのデータは、プレートにコーティングされたUBA2がK48及びK63ポリユビキチン鎖と親和性があることを示している。
ユビキチン化反応は、500nMの6xHis−SUMO−MuRF1、50nMのE1、150nMのUbcH5c(E2)、500ngのユビキチン、2mMのATP、50mMのトリス塩酸緩衝溶液 pH8.0、5mMの塩化マグネシウム、2mMのDTTを含む。いくつかの反応は、反応の重要成分(−E3、−E2、−E1及び−Ub)なしで行われる。MuRFIをコード化するPCR産物は、5’−GATCGGTCTCAAGGTATGGATTATAAGTCGAGCCTG−3’(配列ID番号:12)及び5"−GATCGGTCTCAGATCCATGGATTATAAGTCGAGCCT−3’(配列ID番号:13)プライマー及びテンプレートとしてヒトMuRF1 cDNAを使用して生じさせる。前記PCR断片はBsaI及びBamHIで消化され、及び、6xHis−SUMO−MuRF1を生じるp6xHis−SUMOプラスミド(LifeSensors)内に結合される。前記プラスミドは、正しい配列の存在を確認するよう配列される。反応混合物30μlは37℃で30、60、または90分間保温され、及び、6xHis−SUMO−UBA2コーティングされた96ウェルプレートに移される。2時間の保温後、前記ウェルは、1×PBSと3回洗浄される。アンチユビキチン抗体の100μl(1×PBS中0.3%BSAの1:10希釈)がウェルに添加され、1時間かけて保温される。前記プレートは、1×PBSと3回洗浄される。FITC標識アンチラビット抗体溶液の100μl(1×PBS中3%BSAの1:50希釈)が添加される。前記プレートは室温で1時間保温され、前記ウェルは1×PBSと7回洗浄される。蛍光プレートリーダー(Perkin Elmer Envision)を使用すると、それぞれ485nm及び535nmの励起及び発光波長で蛍光が検知される。図4のデータはSUMO−UBA2がMuRF1オートユビキチン化を検知することを示している。
MuRF1、Hrd1、Parkin、CARP2及びAtrogin1は、異なる型のユビキチンとのユビキチン化実験において6xHis−SUMO−UBA2を用いて分析される。発現及び精製の目的のため、切断HrdIタンパク質が使用される。切断HrdIタンパク質では、N末端膜トランス領域(アミノ酸1〜234によりコード化されている)が除かれている。切断タンパク質6xHis−SUMO−Hrd1Δ235をコード化するプラスミドは以下のように構成されている。PCR産物は5’−GATCGGTCTCTAGGTAAGGTGCACACCTTCCCACT−3’(配列ID番号:14)及び5’−GATCGGATCCTCAGTGGGCAACAGGAGACT−3’(配列ID番号:15)プライマー及びテンプレートとしてヒトHrd1 cDNAを使用して生成される。前記PCR断片はBsa1及びBamHIで消化され、その後、p6xHis−SUMOプラスミド(LifeSensors)内で結合される。前記プラスミドは、正確な配列の存在を確認するよう配列される。Parkinをコード化するPCR産物は、5’−GATCCGTCTCAAGGTATGATAGTGTTTGTCAGGTTC−3’(配列ID番号:16)及び5’−GATCGGATCCCTACACGTCGAACCAGTGGTCC−3’(配列ID番号:17)プライマー及びテンプレートとしてヒトParkin cDNAを使用して生成される。前記PCR断片はBsmBI及びBamHIで消化され、その後、6xHis−SUMO−Parkinを生成するp6xHis−SUMOプラスミド(LifeSensors)内で結合される。前記プラスミドは、正しい配列が存在するのを確認するよう配列される。Atrogin1をコード化するPCR産物は、5’−GATCGGTCTCAAGGTATGCCATTCCTCGGGCAGGACTG−3’(配列ID番号:18)及び5’−GATCGGATCCTCAGAACTTGAACAAGTTGATAA−3’(配列ID番号:19)及びPCR内としてヒトAtrogin1 cDNAを使用することで生成される。前記PCR断片はBsal及びBamHIで消化され、6xHis−SUMO−Atrogin1を生じるp6xHis−SUMOプラスミド(LifeSensors)内で結合される。前記プラスミドは正しい配列を確認するよう配列される。
SUMO−CARP2の濃度範囲(0〜5μM)は50μlユビキチン化反応内に含まれる。ユビキチン化反応は、6xHis−SUMO−CARP2、10nMのE1、100nMのUbcH5c(E2)、500ngユビキチン、2mMのATP、50mMのトリス塩酸緩衝溶液pH8.0、5mM塩化マグネシウム、0.1mMのDTTから構成される。50μlの反応は、6xHis−SUMO−UBA2コーティングした96ウェルプレート内にて37℃で90分間保温される。ウェルは1×PBSと3回洗浄される。アンチユビキチン抗体の50μl(1×PBS中0.3%BSAの1:10希釈)が前記ウェルに添加され、1時間保温される。前記プレートは1×PBSと3回洗浄される。FITC標識アンチラビット抗体溶液の50μl(1×PBS中3%BSAの1:100希釈)が添加される。前記プレートは室温にて1時間保温され、及び、前記ウェルは1×PBSと4回洗浄される。蛍光プレートリーダー(Perkin Elmer Envision)を使用すると、それぞれ485nm及び535nmの励起及び発光波長を使用して蛍光が検知される。図5Aのデータは、SUMO−CARP2オートユビキチン化が濃度依存性であることを示している。
GST−Prajalの濃度範囲(0〜100nM)は50μlユビキチン化反応内に含まれる。Prajalをコード化するPCR産物は、5’−GATCGGATCCCCATGGGTCAGGAATCTAGCAAG−3’(配列ID番号:22)及び5’−GATCGAATTCAGAGTGGGGGAGGGAACATGC−3’(配列ID番号:23)プライマー及びテンプレートとしてヒトPrajal(Open Biosystems)を使用して生成される。前記PCR断片はBamHI及びEcoRIで消化され、その後、GST−Prajalを生じるpGEX3Xプラスミド(Pharmacia Biotech)内で結合される。前記プラスミドは、正確な配列の存在を確認するよう配列される。ユビキチン化反応はGST−Prajal、10nMのE1、100nMのUbcH5c(E2)、500ngユビキチン、2mMのATP、50mMトリス塩酸緩衝溶液pH8.0、5mM塩化マグネシウム、0.1mMのDTTから構成される。前記50μl反応は6xHis−SUMO−UBA2コーティングされた96ウェルプレート内にて37℃で90分間保温される。前記ウェルはIX PBSと3回洗浄される。アンチユビキチン抗体の100μl(1×PBS中0.3%BSA1:10希釈)は前記ウェルに添加され、1時間保温される。プレートは1×PBSと3回洗浄される。FITC標識アンチラビット抗体溶液の100μl(1×PBS中3%BSA1:50希釈)が添加され、及び、プレートは室温にて1時間保温され、続いて1×PBSと7回洗浄される。蛍光プレートリーダー(Perkin Elmer Envision)を使用すると、それぞれ485nm及び535nmの励起及び発光波長を用いて蛍光は検知される。図5Bのデータは、GST−Prajalオートユビキチン化が濃度依存性であることを示している。
プレートと結合した培地(Costar、USA)は、6xHis−SUMO−UBA2の0.1mg/mlの100μlと4℃で一晩かけてコーティングされる。前記プレートは、1×リン酸緩衝生理食塩水(1×PBS、pH7.4)内3%BSAの200μlで、4℃で3時間かけてブロックされ、及び、1×PBSと3回洗浄される。E3反応は、以下のように行われる。IC50実験のため、NEM、ヨードアセトアミド及びユビスタチンAの様々な濃度がトリプリケートの個々のウェルに添加された。制御は10%DMSOの5μlまたは50mMのNEMが含まれる。前記化合物に、アッセイバッファー(50mMトリス塩酸緩衝溶液pH8.0、2mM塩化マグネシウム、0.1mMのDTT)内に調製されたGST−E1(10nM)/His6−UbcH5c(100nM)、6xHis−SUMO−CARP2(500nM)を含む酵素混合物の20μlが加えられ、室温で30分間保温される。pFastBac−GST−E1ベクターは、精製方法に関連する昆虫細胞から発現され、精製される。Beaudenon and Huibregtse,Methods Enzymol,398:3−89(2005)。UbcH5cをコード化するPCR産物は、5’−GATCTCTAGAATGGCGCTGAAACGGATTAA−3’(配列ID番号:24)及び5’−GATCCTCGAGTCACATGGCATACTTTCTGAGTC−3’(配列ID番号:25)プライマー及びテンプレートとしてヒトUbcH5c cDNAを使用して生成される。前記PCR断片はBsa1及びBamHIで消化され、その後6xHis−UbcH5cを生じるpET24D(Novagen)内に結合される。前記プラスミドは、正確な配列の存在を確認するよう配列される。E3反応はアッセイバッファー内に準備され、37℃で60分間保温されたユビキチン(500ng)/ATP(2mM)混合物の添加によって開始される。プレートは、1×PBSと3回洗浄され、1×PBS中0.3%BSAに調製されたアンチユビキチンプライマリー抗体(SIGMA5USA)の1:10希釈液と室温で1時間保温される。プレートは1×PBSと3回洗浄され、続いてアンチラビットIgG−FITC複合体(Jakson Immuno Research、USA)の1:100希釈液100μlと室温で1時間保温される。プレートは、1×PBSと6回洗浄され、及び、それぞれ485nm及び535nmの励起及び発光波長の蛍光プレートリーダーを使用してリーディングされる。図6Aに示したように、NEM、ヨードアセトアミド、及びユビスタチンAは、それぞれのIC50値である3.42×10−5M、1.1×10−4M及び4.21×10−7Mで酵素反応と結合されたE1、E2及びE3を阻害する。
96ウェルプレートのA1−H6ウェル内で酵素混合物が2%DMSO内の10mMのNEMとプレインキュベートされる場合及び制御剤として2%DMSOを含むA7−H12ウェル内で行われる場合を除いて、上述したように行われる。データはエクセルに出力され、ZhangなどがJ.Biomol.Screen.,1999.4(2):p.67-73に記述したようにZスコアが計算される。図6Bのデータは、UBA2コーティングプレートを使用すると、6xHis−SUMO−CARP2 E3アッセイは0.72のZスコアとなることを示している。
6xHis−SUMOG2C−UBA1は5−ヨードアセトアミド蛍光剤(「5’−IAF」)で標識される。標識は、ジメチルホルムアミド内に調製された10mMの5’−IAF溶液の900μlを、15mlチューブ内の20mMトリス塩酸緩衝溶液pH7.6及び150mM塩化ナトリウム内に調製された6xHis−SUMOG2C−UBA1タンパク質の8.4mgの2mlに添加し、4℃で15時間弱攪拌下、暗室にて保温することで行われる。6xHis−SUMO−CARP2は上述したように標識されるが、前記反応は4℃で1時間のみ進行させることができる.未反応の遊離5’−IAFは20mMトリス塩酸緩衝溶液pH7.46及び150mM塩化ナトリウムで平衡にされ、及び、3mlバッファーと溶出されたPD−10脱塩カラムを介した試料に通すことで除去される。溶出断片はSDS−PAGEにより分析され、蛍光ゲルイメージャーで可視化され、続いて、クマシーブリリアントブルーによって染色される。図7C及び7Dのデータはそれぞれ、5’−IAFで標識したUBA1及び精製が示されている。
高結合プレートは、4℃で一晩かけて6xHis−SUMOG2C−UBA1の0.1mg/ml溶液の100μlでコーティングされる。前記プレートは、1×リン酸緩衝生理食塩水(1×PBS、pH7.4)中3%BSAの200μlと室温で3時間保温され、及び、1×PBSと3回洗浄される。前記E3リガーゼアッセイ反応は500nMまたは1μMの6xHis−SUMO−GFP−CARP2(E3)、50nMのE1、150nMのUbcH5c(E2)、500ngユビキチン、2mMのATP、50mMトリス塩酸緩衝溶液pH8.0、5mM塩化マグネシウム、0.1mMのDTTを含むチューブ内で集められる。CARP2をコード化するPCR産物は、5’−GACGAGCTGTACAAGATGTGGGCAACCTGCTGCAACTGG−3’(配列ID番号:26)及び5’−GTGGTGCTCGAGTCAGGACCGGAAGACATGCACAGCTCG−3’(配列ID番号:27)及びテンプレートとしてpSUMO−CARP2を用いて生成される。前記PCR断片は、BsrGI及びXho1で消化され、その後、SUMO−GFP−CARP2ベクターを生じるpET−6xHis−SUMO−GFP内に結合される。前記プラスミドは、正確な配列の存在を確認するよう配列される。トリプリケート内の反応混合物の30μlはUBA1コーティングされたウェルを含むプレートに移され、37℃で90分間保温される。前記ウェルは、IX PBStp洗浄され、それぞれ485nm及び535nmの励起及び発光波長の蛍光プレートリーダーを使用してリーディングされる。図7Bのデータは、UBA1が6xHis−GFP−CARP2のポリユビキチン化を検知するのに使用できることが示されいている。
E3リガーゼ反応は、10nMのGST−Prajal、500nMの6xHis−SUMO−CARP2またはE3として500nMの6xHis−SUMO−MuRF1、50nMのE1、150nMのUbcH5c(E2)、500ngユビキチン、2mMのATP、50mMトリス塩酸緩衝溶液pH8.0、5mM塩化マグネシウム、0.1mMのDTTを含む。SCFAtrogin−1E3リガーゼ活性を測定するため、E3リガーゼが6xHis−SUMO−Atrogin−1、Cullin−1(「Cull」)、Roc−1(「Rbx−1」)及び6xHis−Skpl(それぞれ250nM)を含む場合を除いて、前記反応は基本的に上述したように構築される。SCF複合体のためのCull−Rbx1複合体を大量に得るために、E.coli内にCull−Rbx1の過剰発現が用いられる。Zhengなど、Nature,416(6882):703−9(2002)。Cull遺伝子は二つに(1−410残基及び411−776残基)に分けられ、及び、三つの異なったポリペプチド鎖としてRbx1に標識されたGSTと共発現される。Skplをコード化するPCR産物は、5’−GATCGGTCTCAAGGTATGCCTTCAATTAAGTTGCACAGTTCTGAT−3’(配列ID番号:28)及び5’−GATCGGATCCTCACTTCTCTTCACACCA−3’(配列ID番号:29)プライマー及びテンプレートとしてヒトSkpl cDNAを使用して生成される。PCR断片は、Bsa1及びBamHIで消化され、その後、pET24D(Novagen)内に結合される。プラスミドは正確な配列の存在を確認するよう配列される。
E3アッセイは5’−IAF標識6xHis−SUMO−CARP2との反応が6xHis−SUMOG2C−UBA1コーティングプレートと行われるときを除いて、上述したように行われる。図10のデータはUBA1が5’−IAF標識6xHis−SUMO−CARP2または6xHis−SUMO−GFP−CARP2のポリユビキチン化を検知するのに使用できることを示している。

(c)前記候補モジュレーターの存在下における前記基質に結合しているUb/Ublの量と、前記候補モジュレーターの非存在下における前記基質に結合しているUb/Ublの量とを比較する工程であって、これによって前記基質に結合しているUb/Ublの量の差分が、前記候補モジュレーターがユビキチンリガーゼモジュレーターであることを示すものである、前記比較する工程と
を有するものである方法。 - 請求項56記載の方法において、この方法は、さらに、
(d)工程(a)〜(c)を繰り返す工程であって、前記ウェルが、工程(c)において結合しているユビキチンの量に差を有する1つのユビキチンリガーゼを含むまで、前記複数のユビキチンリガーゼは、工程(c)における結合しているユビキチンの量に差を有するウェル由来のユビキチンリガーゼの一部によって置換されるものである、前記繰り返す工程を有するものである。 - ユビキチンリガーゼモジュレーターを選択する方法であって、この方法は、
(a)ウェルの配列を提供する工程であって、これによってそれぞれのウェルが、候補モジュレーターの存在下で、ユビキチン接着モチーフ含有タンパク質、ユビキチン活性化酵素、ユビキチン結合酵素、Ub/Ubl、ATP、複数のユビキチンリガーゼ含むものである、前記提供する工程と、
(b)前記ユビキチンリガーゼに結合しているUb/Ublの量を計測する工程と、
(c)前記候補モジュレーターの存在下における前記ユビキチンリガーゼに結合しているUb/Ublの量と、前記候補モジュレーターの非存在下における前記ユビキチンリガーゼに結合しているUb/Ublの量とを比較する工程であって、これによって前記ユビキチンリガーゼに結合しているUb/Ublの量の差分が、前記候補モジュレーターがユビキチンリガーゼモジュレーターであることを示すものである、前記比較する工程と
を有するものである方法。 - 請求項58記載の方法において、この方法は、さらに、
(d)工程(a)〜(c)を繰り返す工程であって、前記ウェルが、工程(c)において結合しているユビキチンの量に差を有する1つのユビキチンリガーゼを含むまで、前記複数のユビキチンリガーゼは、工程(c)における結合しているユビキチンの量に差を有するウェル由来のユビキチンリガーゼの一部によって置換されるものである、前記繰り返す工程を有するものである。
