体液性免疫応答は抗原が抗体と特異的に結合した時に誘発される。抗体分子と抗原の合体は、小型で相対的に溶解性の高い免疫複合体を形成する。抗原は、ウイルス性もしくは細菌性のポリペプチドなどの外来性物質であってもよく、またはヒトの体内に通常認められるポリペプチドなどの「自己抗原」であってもよい。免疫系は通常、外来性抗原を自己抗原と識別する。しかし、この系が壊れて、免疫系が身体を攻撃して、組織または臓器系をそれらが外来性物質であるかのように破壊するようになると、「自己免疫」性疾患が起こる恐れがある。大型の免疫複合体であるほど、小型で相対的に溶解性の高い可溶性免疫複合体よりも病原性が高い。大型で相対的に溶解性の低い免疫複合体の形成は、抗体分子と抗原との相互作用、および抗体分子同士の相互作用の両方によって起こりうる。また、そのような免疫複合体は、抗原の不在下での抗体間の相互作用によっても起こりうる。

(1997) J. Exp. Med. 186:777-783)。IgG Fcに対するFcRnの唯一の結合部位がIgG Fc CH2-CH3クレフトであり、HIS 435は3D構造およびアラニンスキャンによってFcRnとIgG Fcとの結合に必須であることが示されている(Shields et al. (2001) J. Biol. ポリペプチドは、さまざまな方法によって産生することができ、その多くは当技術分野で周知である。例として、限定的ではないが、ポリペプチドを、天然の供給源から(例えば、単離された細胞、組織または体液から)の抽出によって、ポリペプチドをコードする組換え核酸の発現によって(例えば、以下に述べるように)、または化学合成(例えば、固相合成、またはABIペプチド合成装置;Applied Biosystems, Foster City, CAを用いた合成を含む、当技術分野で周知の他の方法により)によって入手することができる。レトロ-インバーソポリペプチド類似体を合成するための方法(Bonelli et al. (1984) Int. J. Peptide Protein Res. 24:553-556;およびVerdini and Viscomi (1985) J. Chem. Soc. Perkin Trans. I:697-701)および部分的レトロ-インバーソペプチド類似体の固相合成のためのいくつかの工程も記載されている(例えば、欧州特許第EP0097994号を参照)。

本文書は、Fcを介した免疫複合体形成の異常(例えば、Fcを介した免疫複合体の過剰産生)によって起こる病状を治療するための方法において使用され得る組成物および製造品を提供する。本明細書で提供されるポリペプチド、化合物、および組成物は、Fcを介した免疫複合体形成を調節することによって緩和されうる例えばウイルス感染症を有する対象(例えば、ヒトまたは別の哺乳動物)に対して投与されて、mC1q、sC1q、FcγR、およびFcRnに対する免疫複合体を形成したIgG Fcを阻害することができる。典型的には、1つまたは複数のポリペプチドまたは化合物を、免疫複合体形成と関連のある疾患または病状を有する疑いのある対象に対して投与することができる。組成物は概して、1つまたは複数の本明細書に記載のポリペプチドおよび化合物を含みうる。例えば、CH2-CH3結合性ポリペプチドを、薬学的に許容される担体または希釈剤に入れて、特定の疾患の性質、その重症度および対象の全般的状態に応じて変化する量および期間にて投与することができる。典型的には、ポリペプチドは、阻害量(すなわち、ポリペプチドが接触した細胞または組織における免疫複合体の産生を阻害するのに十分な量)で投与される。また、本明細書に記載のポリペプチドおよび方法を、例えば、免疫複合体の異常な産生または過剰産生のリスクのある対象(例えば、移植レシピエント)における免疫反応性を最小化するために、予防的に用いることもできる。
デング熱は、世界で最も流行している蚊媒介性のウイルス性疾病であり、熱帯地域において、25億人を超える人々が感染のリスクにある。デング熱は、主に、都市に生息するアエデス・アエギプチ蚊(Aedes aegypti)(黄熱病ウイルスも伝達するステゴミア・アエギプチ(Stegomyia aegypti)とも呼ばれる)によって、ヒトに伝達される。黄熱病を駆除するための第二次世界大戦期のプログラムは、標的区域においてデング熱を制御するという利点をもたらした。しかしながら、過去30年において、これらのプログラムの中止、および熱帯区域における急速な都市化によって、再びデング熱が出現してきている。
によるICに対するNS1結合の阻害を、下記の実施例6に示す。さらに、実施例2の表3は、免疫複合体を形成したIgGに対する、FcyRIIaまたはFcyRIIbのいずれかの結合を、SEQ ID NO:20が90%を超えて阻害したことを示し、SEQ ID NO:20は、DENV抗体力価の部分中和により引き起こされるDCにおけるADEを抑制する可能性があることを示唆している。
CH2-CH3結合ポリペプチドは、Fc介在性免疫複合体(IC)の形成のインビトロアッセイにおいて使用され得る。そのような方法は、例えば、Fc介在性IC形成を阻止するCH2-CH3クレフト結合ポリペプチドの能力を評価するために、有用である。インビトロの方法は、例えば、本明細書において提供されるポリペプチドの存在下および非存在下において、免疫グロブリン分子(例えば、抗原に結合した免疫グロブリン分子等)をエフェクター分子(例えば、mC1q、sC1q、FcR、およびFcRn、または別の抗体)に接触させる段階、および各試料においてIC形成レベルを決定する段階を包含し得る。IC形成レベルは、例えば、クマシーブルー染色もしくは銀染色を伴うポリアクリルアミドゲル電気泳動によって、または免疫共沈降によって評価され得る。そのような方法は、当業者に公知であり、かつ例えば、感染性ウイルス疾患に関連するIC形成を阻害する候補ポリペプチドまたは候補化合物の能力を試験するために使用され得る。

本明細書に記載されるポリペプチドおよび化合物による、FcR:(FcγRI、FcγIIa、FcγRIIb/c、FcγRIIIa/b)、FcRn、mC1qおよびsC1q等の免疫を調節する因子に対する、免疫複合体を形成したIgG Fcの結合の、(競合的)阻害を実証するため、酵素結合免疫吸着アッセイ(ELISA)を包含するインビトロアッセイを使用した。例えば、本明細書の実施例、ならびに米国特許公報第20070225231号および同第20070276125号、およびPCT公報WO2007/030475号等を参照のこと。標準化された試薬およびELISAキットは、コストを減少させ、かつ実験の再現性を増加させるのに有用である。
1ml蒸留水中で2μlのウサギ抗ペルオキシダーゼ抗体(Sigma Chemicals, 製品P7899)を50μlのペルオキシダーゼ(Sigma-Aldrich, P6782)と混合することによってPAP複合体を形成させた。PAP(100μl)は、100μlのペプチドと共に1時間プレインキュベートし、100μlは100μlのペプチドまたはヒトC1q(Quidel Corp.)と共に1時間プレインキュベートした。C1q/PAPおよびペプチド/PAP混合物(100μl)を、C1qをコーティングしたプレートを用いて30分間インキュベートした。洗浄の後に、プレートをATBS(Quidel Corp.)と共に15分間インキュベートし、405nmで読み取った。結果は表1に示されている。
ひとたび逆ELISAプロトコールをC1qアッセイを用いて確立したところで、FcγIIa、FcγIIbおよびFcγIIIをC1qの代わりに用いるアッセイを再びデザインした。高精製FcγIIa、FcγIIbおよびFcγIII(R&D Systems, Minneapolis, MN)を合成樹脂製マイクロウェル上に免疫吸着させた。FcγR逆ELISAシステムを最適化した後に、精製FcγRに対する免疫複合体の結合を阻害する能力を調べるために、本明細書で提供されるポリペプチドを用いる簡単な競合阻害実験を実施した。
ELISA被覆緩衝剤(Alpha Diagnostic International)を使用して、約1 ngのDENV2(16681株;Microbix Corp.)をCostarマイクロタイタープレート上に、24時間4℃で被覆した。24時間後、ELISA洗浄溶液(Quidel Corp.)でプレートを5回洗浄し、高度に精製されたBSAで1時間ブロッキングした。5回の洗浄後、DENV被覆されたマイクロタイタープレートを用いて、PAP ICを1時間インキュベートした。1時間後、Quidel ELISA洗浄溶液でプレートを再度5回洗浄し、続いてABTS(Immunochem Corp.)と共に20分間インキュベートした。OD405 nmが高いほど、DENV2(16681)ウイルス粒子に対するDENV非関連免疫複合体(PAP-IC)の結合が高くなった。表4に結果を示す。
ELISA被覆緩衝剤(Alpha Diagnostic International)を使用して、高度に精製された組換えヒトFcγIIa(R&D Systems)を、プラスチックのマイクロウェル(Costarマイクロタイタープレート)上に、24時間 4℃で免疫吸着させた。24時間後、ELISA洗浄溶液(Quidel Corp.)でプレートを5回洗浄し、高度に精製されたBSAで1時間ブロッキングし、5回洗浄した。DENV回復期の個体に由来するポリクローナルの、プールされた、親和性精製された中和IgG(Calbiotech Dengue Fever IgG ELISA;Cat. No. DE050G4)と、不活性化されたDENV-2ウイルス粒子(16681株、Microbix Corp.)を組み合わせることによって、ヒト抗DENV2 ICを形成させた。1:250および1:500の段階希釈(ADEは、1:250〜1:500の血清希釈で最も明確であった)のDENV-IgG-ICの結果を、huFcγRIIa被覆されたマイクロタイターウェルに対するDENV-IgG-IC結合の阻害に対して試験した。阻害剤を含まない1:250希釈のDENV-IgG-IC、SEQ ID NO:20を含む1:250希釈のDENV-IgG-IC、阻害剤を含まない1:500希釈のDENV-IgG-IC、およびSEQ ID NO:20を含む1:500希釈のDENV-IgG-ICのいずれかを含む、huFcγRIIa被覆されたウェルを使用した結果を、表5に示す。
ELISA被覆緩衝剤(Alpha Diagnostic International、San Antonio、Texas)を使用して、高度に精製された組換えDENV(100μgのデングウイルス組換えNS1;Prospec Tech)を、プラスチックのマイクロウェル(Costarマイクロタイタープレート)上に、24時間 4℃で免疫吸着させた。24時間後、ELISA洗浄溶液(Quidel Corp)でプレートを5回洗浄し、高度に精製されたBSAで1時間ブロッキングし、再度5回洗浄した。ペプチドなしで、またはSEQ ID NO:20と共に、1時間PAP ICをインキュベートした。1時間後、Quidel ELISA洗浄溶液でプレートを5回洗浄し、続いてABTS(Immunochem Corp.)と共に20分間インキュベートした。OD405 nmが高いほど、DENV NS1タンパク質に対するDENV非関連の免疫複合体(PAP-IC)の結合が高くなったことが観察された。表6に結果を示す。
ELISA被覆緩衝剤(Alpha Diagnostic International)を使用して、高度に精製された組換えTNF-α(Sigma-Aldrich)を、プラスチックのマイクロウェル(Costarマイクロタイタープレート)上に、24時間4℃で免疫吸着させた。24時間後、ELISA洗浄溶液(Quidel Corp)でプレートを5回洗浄し、高度に精製されたBSAで1時間ブロッキングし、5回洗浄した。ペプチドなしで、またはペプチドSEQ ID NO:20と共に、1時間PAP ICをインキュベートした。1時間後、Quidel ELISA洗浄溶液でプレートを5回洗浄し、続いてABTS(Immunochem, Corp)と20分間インキュベートした。OD405 nmが高いほど、TNF-αに対するTNF-α非関連の免疫複合体(PAP-IC)の結合が強くなった。表7に結果を示す。
上に記載された、デングウイルス用のRAG2-/-γc-/-(RAG-hu)マウスモデルは、デングウイルス感染症および免疫性について、多くの重要かつ時宜を得た問題に対処するための他に例をみない研究手段を提供する(Kuruvilla et al. (2007) Virology 369: 143-152)。異種DENV IgG(すなわち、DENV1用の中和IgG;血清は、DENV1 IgGおよびDENV2感染性ウイルス粒子、またはDENV2 IgGおよびDENV1感染性ウイルス粒子等の、任意のDENV異種中和DENV感染症に由来し得る)を用いて、RAG-huマウスを受動的に免疫する。例えば、マウスを抗DENV1 IgGで受動的に免疫し、続いて、致死用量の、例えばDENV2ウイルスを接種する。配列
を有するペプチドを、例えば1 mg/kg、10 mg/kg、100 mg/kg、および1,000 mg/kgの1日用量で、DENV感染時から腹腔内投与する。全体的な臨床徴候、体温等を毎日モニタリングする。DENV NS1もしくはDENVの複製、ならびにDENV NS1 ICおよびDENV-ICの存在に関する、血清ELISAまたはRT-PCRを毎日実施する。DENVに感染した瀕死マウスの部分集団を毎日屠殺し、上に記載されたDENV複製の測定値に加えて、血小板計測値、HCT/PCV値、AST/ALT値を確認する。DENVに感染し、
で処置された同数のマウスを屠殺し、比較目的のため上記のように試験する。研究終了時に、全ての免疫学的プロファイルおよび複数のDENV許容組織の生検を含む剖検を実施する。ADE DENVに感染したマウスの最終的な解析を、ADE DENVに感染し、SEQ ID NO:20で処置されたマウスと比較する。

- 前記ポリペプチドが、DENV感染症のADEの免疫病因をもたらすICの形成を阻害する、請求項1記載の方法。
