特定の実施形態では、イメージングレポーターがフルオロフォア分子である。本明細書で使用する場合、「フルオロフォア」という用語は、蛍光色素、蛍光色素消光分子、有機または無機染料、金属キレートまたは蛍光酵素基質(プロテアーゼ活性化可能な酵素基質を含む)を意味するものと理解される。

「非蛍光レポーター」という用語は、本明細書で使用する場合、蛍光ではないがイメージングにおいてコントラストまたはシグナルを得る目的で使用可能であり、なおかつ非蛍光イメージング技術で検出可能な化学部分を示す。特定の実施形態では、他の非蛍光レポーターを、イメージング部分と化学結合可能であり、あるいは本発明の造影剤と同時またはこれと連続して被検体に投与可能である。このようなレポーターとしては、フォトルミネセンスナノ粒子、放射性同位元素、超常磁性作用剤、X線コントラスト剤、および超音波作用剤があげられる。また、レポーターには、ポルフィリン、Photofrin(登録商標)、Lutrin(登録商標)、Antrin(登録商標)、アミノレブリン酸、ヒペリシン、光力学療法で用いられているベンゾポルフィリン誘導体、放射線療法用の放射性核種などの治療用レポーターを含むことがある。
作用剤は、1種類以上の放射性標識を含み得る。銅、ガリウム、インジウム、テクネチウム、イットリウム、およびルテチウムなどの金属の放射性同位元素形態を、金属造影剤に化学結合することが可能であり、核イメージングまたは治療用途に使用可能である。代表的な放射性標識としては、99mTc、111In、64Cu、67Ga、186Re、188Re、153Sm、177Lu、および67Cuがあげられるが、これに限定されるものではない。
他の代表的なレポーターは、磁気共鳴造影剤用のキレート剤を含み得る。このようなキレーターは、たとえば、作用剤に対して化学結合可能なポリアミン−ポリカルボキシレートキレーターまたはイミノ酢酸キレーターを含み得る。
超音波イメージングの場合、イメージングレポーターは、Levovist、AlbunexまたはEchovistなどの気体で満たした気泡あるいは、金属イオンの原子番号が21〜29、42、44または57〜83の粒子または金属キレートを含み得る。このような化合物の例が、Tylerら、ULTRASONIC IMAGING、3、pp.323〜29(1981)およびSwanson、「Enhancement Agents for Ultrasound: Fundamentals」 PHARMACEUTICALS IN MEDICAL IMAGING、pp.682〜87(1990)に記載されている。
代表的なレポーターは、原子番号57〜83の重金属イオンのヨウ素化有機分子またはキレートを含み得る。このような化合物の例が、Sovak編、「Radiocontrast Agents」 SPRINGER−VERLAG、pp.23〜125(1984)および米国特許第4,647,447号明細書に記載されている。
リンカーまたはスペーサー部分を使用して、1種類以上のイメージングレポーターおよび/またはインテグリン標的化部分および/または生物学的修飾物質を化学結合し、本発明のインテグリン作用剤を生成することが可能である。有用なリンカー部分としては、天然と非天然の両方のアミノ酸および核酸、ペプチドならびに、アミノエチルマレイミドなどの合成リンカー分子があげられる。リンカーがペプチドである場合、このペプチドは、任意に、酵素などの多岐にわたる作用剤で切断可能なタンパク質分解性切断部位を含むものであってもよい。
意図した用途に応じて、インテグリン作用剤は、インテグリン作用剤の生物学的特性を変えることのできる1種類以上の生物学的修飾物質を含み得る。たとえば、生物学的修飾物質は、未修飾のインテグリンリガンドと比較して、インテグリン作用剤を、投与用の媒質に対する水溶性および分散性がさらに高く、結合特異性が高く、免疫原性が低く、毒性が低く、非特異的結合が少ないものとし、体内分布および薬物動態を変化させることが可能である。
従来技術において周知の標準的な化学を用いて、上述したインテグリン結合部分、イメージングレポーター、生物学的修飾物質およびリンカーのうちの1種類以上を用いて、有用なインテグリン標的化剤を創製可能である。特定の用途に応じて、インテグリン作用剤は水溶性または水分散性(すなわち、水性媒体または生理学的媒体の溶液に対して十分に可溶または懸濁可能)でなければならない。インテグリン作用剤のin vivoでの半減期は、少なくとも約10分、一層好ましくは30分から数時間になるように設計可能なものである。この作用剤のin vivoでの半減期は、好ましくは、良好な組織曝露および結合を達成できるだけの十分な時間(たとえば、少なくとも約30分間)である。一実施形態では、インテグリン作用剤が、水性媒体に対して水溶性または分散性であり、生体適合性すなわち、たとえばLD50が体重1kgあたり約50mgを超えるなど、無毒である。インテグリン作用剤は、好ましくは望ましくない光毒性を持たないおよび/または低血清タンパク質結合親和性を示さないものである。
インテグリン標的化剤を多岐にわたるイメージングおよび治療用途で使用可能である旨は、理解できよう。
本発明は、本明細書に開示の造影剤を用いるin vitroおよびin vivoでのイメージング方法を提供するものである。光学イメージング技術の概要については、たとえば、Alfanoら、ANN. NY ACAD. SCI. 820:248〜270(1997);Weissleder、NATURE BIOTECHNOLOGY 19、316〜317(2001);Ntziachristosら、EUR. RADIOL. 13:195〜208(2003);Gravesら、CURR. MOL. MED. 4:419〜430(2004);Citrinら、EXPERT REV. ANTICANCER THER. 4:857〜864(2004);Ntziachristos、ANN. REV. BIOMED. ENG. 8:1〜33(2006);Kooら、CELL ONCOL. 28:127〜139(2006);Raoら、CURR. OPIN. BIOTECHNOL. 18:17〜25(2007)を参照のこと。
光学in vivoイメージングに関して、このような方法は、(a)本明細書に記載のインテグリン作用剤1種類以上を被検体に投与し、(b)作用剤が被検体で分散できるだけの十分な時間を取り、(c)インテグリン作用剤によって発光するシグナルを検出することを含む。作用剤によって放射されるシグナルを使用して、断層画像などの画像を構成することが可能である。上記のステップをあらかじめ定められた時間間隔で繰り返すことで、被検体においてインテグリン作用剤の放射シグナルを経時的に評価できるようにすることも可能である。
in vitroイメージングに関しては、多岐にわたるin vitroアッセイに造影剤を使用することが可能である。たとえば、代表的なin vitroイメージング方法は、(a)生物学的試料などの試料を本明細書に記載のインテグリン作用剤のうちの1種類以上と接触させ、(b)作用剤を試料中の生物学的標的と相互作用させ、(c)任意に、未結合の作用剤を除去し、(d)作用剤から放射されるシグナルを検出することで、生物学的標識によって作用剤が活性化されているか生物学的標識に結合したかを判断することを含む。インテグリン作用剤が蛍光色素を含む場合、ステップ(d)は、蛍光色素によって吸収可能な波長の光を試料に照射して、放射シグナルを生成することをさらに含む。
放射標識および/または薬物分子を含有する作用剤など、本明細書に記載のインテグリン標的化剤のいくつかを使用して、症候を寛解または特定の疾患または機能障害を治療することが可能である。この方法は、(a)被検体で治療効果を発揮できるだけの量の本明細書に記載の作用剤を1種類以上、被検体に投与し、(b)作用剤が被検体で分散あるいは、そうでなければ治療対象となる被検体の領域で局在できるだけの時間を与えた後、(c)治療用作用剤に応じて、任意に作用剤を活性化して治療効果を発揮させることを含む。たとえば、治療用作用剤が放射標識である場合、以後の活性化は必要ない。しかしながら、治療用作用剤が光力学療法で用いられる染料などの光反応性作用剤である場合、作用剤を活性化させる波長の光に作用剤を曝露することで、作用剤を活性化すればよい。結果として、作用剤を用いて、癌、免疫不全、炎症性疾患、血管障害などの対象症状を治療することができる。さらに、作用剤を用いて、被検体の対象領域での血管新生を防止、寛解または逆行させることも可能である。
以下の非限定的な実施例は、代表的なインテグリン標的化剤の合成について示すものである。本発明の材料の調製に使用できる主な材料および方法に関しては、下記にてさらに説明する。化学物質および溶媒(試薬グレード)はいずれも、市販されているものをそれ以上精製することなく使用した。HPLC分析:Waters 2695システムにて、Phenomenexフェニル−ヘキシルカラム(3μ、100×4.6mm)またはC18カラム(5μ、250×4.6mm)のいずれかを用いて、流量1mL/分で分析用RP HPLCを実施した。A(10mM酢酸トリエチルアミン、pH=7、+5%アセトニトリル)とB(アセトニトリル)の線形勾配を使用した。一般に、勾配は10〜40%のBで開始され、10〜20分でBが50〜90%まで変化する。254nmおよび675nmでクロマトグラムを監視した。VarianシステムでPhenomenex C18カラム(250×10mm)を使用し、4.7mL/分で同様の勾配を分析ランとして用いて分取HPLCを実施した。
化合物1(690mg、1.57mmol)を含有する20mLのエタノール溶液に塩化チオニル(1.3g、10.9mmol)を加えた。添加後、この溶液を40〜45℃で攪拌し、反応をHPLCで監視した。反応終了(約45分)後、溶液を油になるまで濃縮し、酢酸エチル(30mL)と水(20mL)を加えた。混合物をpH約8になるまで固体の重炭酸ナトリウムで中和した。相分離後、水性層を2×15mLの酢酸エチルで抽出した。混合酢酸エチル溶液を2×10mLの飽和ブラインで洗浄し、無水硫酸ナトリウム上で乾燥させた。固体を濾別し、濾液を濃縮して、820mg(111%質量収支)のエステル2を油として得た。
エステル2(820mg、1.57mmol理論)を4mLの酢酸に溶解させた後、臭素(296mg、1.85mmol)を加えた。HPLC分析によって、約10分後に反応が完了したことが明らかになった。この溶液を酢酸エチル(30mL)と水(20mL)で希釈した。混合物をpH約7になるまで重炭酸ナトリウムで中和した。相分離後、水性層を20mLの酢酸エチルで抽出した。混合酢酸エチルを2×20mLの飽和ブラインで洗浄し、硫酸ナトリウム上で乾燥させた。溶媒を蒸発させ、化合物9をオフホワイトの固体(908mg、118%質量収支)として得た。
化合物9(908mg、1.57mmol理論)をジメチルアセトアミド(4mL)中にてシアン化亜鉛(380mg、3.2mmol)およびテトラキス(トリフェニルホスフィン)パラジウム(0)(365mg、0.31mmol)と混合し、得られた混合物を真空/窒素で4回パージした。この混合物を約120℃で20時間加熱し、HPLCで>97%転化したことが示された。約20℃まで冷却後、30mLの酢酸エチルを加えた。固体を濾過し、2×25mLの酢酸エチルで洗浄した。混合酢酸エチル溶液を2×20mLの水と20mLの飽和ブラインで洗浄した。硫酸ナトリウム上で乾燥させた後、溶液を油になるまで濃縮した。残渣を10mLの酢酸エチルに溶解させ、酢酸エチル(v/v)中8%メタノールで溶出されるシリカゲルカラム(70mL総容積)で単離した。適切な画分を組み合わせ、乾燥するまで濃縮して、シアノ−エステル4をワックス状の固体(510mg、66%全体)として得た。
シアノエステル4(170mg、0.345mmol)をメタノール(3mL)に溶解させ、1Mの水酸化ナトリウム水溶液(2mL)を加えた。この溶液を、HPLCで4の加水分解終了が示されるまで20℃で熟成させた(同じくシアノ酸中間体に加水分解されるメチルエステルの形成も観察した)。この溶液を乾燥するまで濃縮し、残渣をアンモニア/メタノール溶液(3.5M、10mL)に溶解させた。得られた溶液を圧力管にてラネーニッケル(約100mg、添加前にメタノールで3回洗浄)に加えた。ギ酸アンモニウム(120mg、1.9mmol)を混合物に加え、続いてこれをHPLCでシアノ酸中間体の転化完了が示される(約20時間)まで20℃で強く攪拌(攪拌子)した。上清を除去し、触媒を4×10mLのメタノールで洗浄した。混合メタノール溶液を乾燥するまで濃縮した。固体を10mLの30/70メタノール/水(v/v)に溶解させ、イオン交換カラム(Dowex(登録商標)50WX8−200、総容積5mL)に仕込んで、ニッケルイオンを除去した。pHが約6になるまで樹脂を水洗し、30/70メタノール/水にて1Mのアンモニアで溶出させた。適切な画分を組み合わせ、蒸発させて、部分アンモニウム塩として存在し得るアミノ酸5(110mg)を得た。C24H32N6O4とした場合のMALDI−MS計算値468.6、実測値469.7。
粗アミノ酸5(90mg、0.185mmol、アンモニウム塩として)を10mLの1:1メタノール/水に溶解させた後、重炭酸ナトリウム(16mg、0.185mmol)を加えた。得られた溶液を乾燥するまで蒸発させ、ナトリウム塩6(97mg、0.20mmol)をオフホワイトの固体として得た。
化合物7(764mg、2.0mmol)の20mLのエタノール溶液に塩化チオニル(1.2g、10mmol)をゆっくりと加えた。添加後、この溶液を40〜45℃で攪拌し、反応をHPLCで監視した。反応終了(約3時間)後、溶液を油になるまで濃縮し、酢酸エチル20mL)と水(10mL)を加えた。混合物をpH約8になるまで固体の重炭酸ナトリウムで中和した。相分離後、水性層を20mLの酢酸エチルで抽出した。混合酢酸エチル溶液を2×10mLの飽和ブラインで洗浄し、無水硫酸ナトリウム上で乾燥させた。固体を濾別し、濾液を濃縮して、762mg(1.86mmol、93%)の化合物8を油として得た。
エステル8(762mg、1.86mmol)を3.5mLの酢酸に溶解させた後、臭素(340mg、2.1mmol)を加えた。HPLC分析によって、20分後に反応が完了したことが明らかになった。この溶液を酢酸エチル(30mL)と水(25mL)で希釈した。混合物をpH約7になるまで重炭酸ナトリウムで中和した。相分離後、水性層を20mLの酢酸エチルで抽出した。混合酢酸エチルを2×15mLの飽和ブラインで洗浄し、硫酸ナトリウムで乾燥させた。溶媒を蒸発させ、化合物9を油(854mg、94%)として得た。これは、放置すると固化した。C24H33BrN4O2とした場合のMALDI−MS計算値489.5、実測値491.8。
化合物9(854mg、1.7mmol)をジメチルアセトアミド(5mL)中にてシアン化亜鉛(590mg、5.0mmol)およびテトラキス(トリフェニルホスフィン)パラジウム(0)(570mg、0.49mmol)と混合し、得られた混合物を真空/窒素で4回パージした。この混合物を約110℃で18時間加熱し、HPLCで>99%転化したことが示された。約20℃まで冷却後、酢酸エチル(30mL)を加えた。固体を濾過により収集し、2×25mLの酢酸エチルで洗浄した。混合酢酸エチル溶液を2×20mLの水と20mLのブラインで洗浄した。硫酸ナトリウム上で乾燥させた後、溶液を油になるまで濃縮した。残渣を10mLの酢酸エチルに溶解させ、酢酸エチル(v/v)中3%メタノールで溶出されるシリカゲルカラム(70mL総容積)で単離した。適切な画分を組み合わせ、乾燥するまで濃縮して、シアノエステル10を固体(710mg、93%)として得た。C25H33N5O2とした場合のMALDI−MS計算値435.6、実測値438.9。
シアノエステル10(200mg、0.46mmol)をメタノール(7mL)に溶解させ、1Mの水酸化ナトリウム水溶液(3mL)を加えた。この溶液を、HPLCでエステルの加水分解終了が示されるまで20℃で熟成させた(同じくシアノ酸中間体に加水分解されるメチルエステルの形成も観察した)。この溶液を乾燥するまで濃縮し、残渣をアンモニア/メタノール溶液(3.5M、15mL)に溶解させた。得られた溶液を圧力管にてラネーニッケル(約150mg、添加前にメタノールで3回洗浄)に加えた。この混合物に、ギ酸アンモニウム(200mg、3.2mmol)を加えた。この混合物をHPLCでシアノ酸中間体の転化完了が示される(約20時間)まで20℃で強く攪拌(攪拌子)した。上清を除去し、触媒を4×10mLのメタノールで洗浄した。混合メタノール溶液を乾燥するまで濃縮した。固体を10mLの30/70メタノール/水(v/v)に溶解させ、イオン交換カラム(Dowex(登録商標)50WX8−200、総容積5mL)に仕込んで、ニッケルイオンを除去した。pHが約6になるまで樹脂を水洗し、30/70メタノール/水にて1Mのアンモニアで溶出させた。適切な画分を組み合わせ、蒸発させて、部分アンモニウム塩として存在し得るアミノ酸11(143mg)を得た。C23H33N5O2とした場合のLC/MS計算値411.5、実測値410(負のモード)。
粗アミノ酸11(130mg、0.30mmol、アンモニウム塩として)を20mLの1:1メタノール/水に溶解させた後、重炭酸ナトリウム(25mg、0.3mmol)を加えた。得られた溶液を乾燥するまで蒸発させ、ナトリウム塩12(103mg、0.23mmol)をオフホワイトの固体として得た。
実施例8:造影剤E−8の調製
化合物12(約3mg)とN−エチルモルホリン(10μL)とをDMSO(0.7mL)に入れた溶液に、AlexaFluor750 NHSエステル(1〜1.5μmol)を加えた。この溶液を20℃で1〜2時間熟成させた。1:1酢酸エチル/ヘキサン(約10mL)を添加して生成物を沈殿させ、MTBEで洗浄した(2×5mL)。固体を水に溶解し、分取HPLCで精製し、分取HPLC画分の蒸発後に化合物E−8を青緑色の固体として単離した。MALDI−MS計算値1278、実測値1279。
パートC。造影剤化合物E−11の調製
トリエチルアミンの存在下でのフルオロフォアC1(表2参照)と化合物15との反応によって、分取HPLC精製後にジカルボン酸C1−15を青色の固体として生成した。
化合物12(約3mg)とN−エチルモルホリン(10μL)とをDMSO(0.7mL)に入れた溶液に、125I Bolton−Hunter試薬(Perkin Elmer/NEN)(1〜1.5μmol)を加える。この溶液を20℃で1〜2時間熟成させる。生成物を分取HPLCで精製し、E−18をHPLC画分として単離する。
化合物6(約3mg)とN−エチルモルホリン(10μL)とをDMSO(0.7mL)に入れた溶液に、125I Bolton−Hunter試薬(Perkin Elmer/NEN)(1〜1.5μmol)を加える。この溶液を20℃で1〜2時間熟成させる。生成物を分取HPLCで精製し、化合物E−19をHPLC画分として単離する。
実施例12のコンジュゲートのガドリニウム複合体を以下の手順で調製する。3〜3.5mgのコンジュゲートを2mLの1M 酢酸アンモニウム緩衝剤(pH7.0)に溶解させ、1当量のGd(NO3)3溶液(0.02M水溶液)を加える。反応混合物を100℃で30分間加熱し、生成物を分取HPLCで単離する。複合体を含む画分を凍結乾燥させる。複合体のアイデンティティを質量分光測定で確認する。
この実施例は、本発明の化合物を使用して腫瘍をin vivoにて画像化できることを示すものである。この実験では、6〜8週齢のNU/NUマウス(Charles River Laboratory、Wilmington、MA)に2×106 4T−1細胞を、それぞれの乳房脂肪体の両側にて皮下(s.c.)注射した。腫瘍がおおむね3×3mmのサイズに達したら(細胞注射後7日前後)、マウスに各インテグリン作用剤4nmoleを100μL容量で尾静脈から静脈内(i.v.)注射した(5マウス/プローブ+2マウス/対照としてプローブなし)。注射の24時間後に蛍光分子断層撮影システム(VisEn Medical、Bedford、MA)を用いてイメージングを実施した。E−6およびE−7を用いる画像の例を、腫瘍を効果的に画像化可能であることを示す図1および図2に示す。
この実施例は、本発明の化合物が、in vitroにてαvβ5、α5β1およびαIIbβ3よりもインテグリンαvβ3に対して選択性であることを示すものである。この実験では、αvβ3(HEK293−αvβ3細胞)を安定的にトランスフェクトした人間の胚腎臓(HEK293)細胞をトリプシン−EDTAで剥離し、無血清MEMで4回洗浄した。ビトロネクチンをコーティングしたマイクロタイターウェルに細胞(25,000個/ウェル)を加え、加湿したインキュベータにて濃度を高めた化合物E−6または未標識の親化合物(E−6の阻害剤として)の存在下または非存在下で、37℃で2時間付着させた。非付着細胞を静かに洗浄して取り除いた。同様の実験を、αvβ5、αIIbβ3またはα5β1を安定的にトランスフェクトしたHEK293細胞でも実施した。Vmaxプレートリーダー(Molecular Devices、Menlo Park、CA)でヘキソサミニダーゼ酵素活性の比色検出によって付着細胞を定量化した。アッセイした細胞株ごとに標準曲線を用いて付着細胞数を定量化し、3重の試料の平均値として表した。結果を表4にまとめておく。
この実施例は、αvβ3に対するE−6の親和性を示すものである。ヒトαvβ3を安定的に発現しているHEK293細胞を用いる細胞結合アッセイを実施した。
平衡結合および解離測定のために、HEK293−αvβ3細胞(1×105)を、「フローサイトメトリー緩衝剤」(145nM NaCl、5mM KCl、1mM MgCl2,および10mM HEPES、pH7.4)にて0、3.12、6.25、12.5、20、25、40、50、80、100および200nMの化合物E−6と一緒に4℃で30分間インキュベートした。HEK293−αvβ3細胞のインテグリンに結合したプローブの量をフローサイトメトリー(FACSCalibur、BD Biosciences、CA)で求めた。FlowJoソフトウェアを用いてデータを分析し、SigmaPLot 10ソフトウェアを用いてKd値を算出した。koffを求めるために、HEK293−αvβ3細胞(1×106)を100nMのE−6と一緒に4℃で30分間インキュベートした後、10mMの親化合物を含有するPBSに標識細胞を移した。未標識の化合物との混合前ならびに、混合1時間後、1.5時間後、2.5時間後、3.5時間後、4.5時間後、5.5時間後、24時間後に、HEK293−αvβ3細胞のインテグリンに結合したプローブの量をフローサイトメトリー(FACSCalibur、BD Biosciences、CA)で求めた。FlowJoソフトウェアを用いてデータを分析し、SigmaPLot 10ソフトウェアを用いてkoff値およびt1/2値を算出した。結果を図4Aおよび図4Bにまとめておく。
この実施例は、マウスにおける化合物E−6の薬物動態と体内分布を示すものである。
この実施例は、化合物E6が内皮細胞およびA673腫瘍細胞と結合することを示すものである。
この実施例は、担腫瘍マウスにおけるE−6シグナルのイメージングおよび定量化を示すものである。6〜8週齢のメスのNU/NUマウス(Charles River Laboratory、Wilmington、MA)にA673細胞(5×106)を第1の乳房脂肪体で皮下(s.c.)注射した。腫瘍が所望の体積に達したら(式 体積mm3=(長さ×幅2)/2を用いてノギスで測定)、過剰な競合親化合物(100nmole)の存在下または非存在下で、マウスに化合物E−6(2nmole)をi.v.注射した。対照マウスには遊離染料を注射した。in vivoイメージング用に、マウスをガス麻酔(イソフルラン/酸素混合物)で麻酔し、VisEnのFMT2500システムイメージングチャンバに1度に1匹ずつ載せて画像化した。
この実施例は、A673担腫瘍マウスで抗血管新生治療がインテグリンおよびカテプシンBシグナルに対しておよぼす効果を示すものである。

- 生物学的修飾物質(BM)が、PEG、リン脂質、アミノ酸、ペプチド、リン脂質PEG、炭水化物、スルホネート、ポリスルホネート、グルタミン酸、システイン酸、ナフチルアラニン、フェニルアラニン、ジフェニルプロピルアミン、4,4−ジフェニルシクロヘキサノール、グルコサミン、マンノサミン、ガラクトサミン、アルギニン、リジン、ホモリジン、ロイシンおよびこれらの組み合わせからなる群から選択される、請求項7〜40のいずれか1項に記載の作用剤。

- (a)請求項1〜43のいずれか1項に記載の作用剤を被検体に投与し、
(b)作用剤を被検体で分散させ、
(c)作用剤によって放出されるシグナルを検出することを含む、in vivoイメージング方法。 - (a)蛍光色素を含む請求項1〜43のいずれか1項に記載の作用剤を被検体に投与し、
(b)作用剤を被検体で分散させ、
(c)蛍光色素が吸収可能な波長の光に被検体を曝露し、
(d)作用剤によって放出されるシグナルを検出することを含む、in vivo光学イメージング方法。 - 作用剤によって放出されるシグナルを使用して画像を構成する、請求項44または45に記載の方法。
- 画像が断層画像である、請求項46に記載の方法。
- ステップ(a)〜(c)をあらかじめ定められた時間間隔で繰り返すことで、被検体においてインテグリン作用剤の放射シグナルを経時的に評価できるようにする、請求項44に記載の方法。

- ステップ(a)において、シグナル特性を互いに区別できる2つ以上のイメージングプローブを被検体に投与し、イメージングプローブの少なくとも一方がインテグリン結合作用剤である、請求項44または45に記載の方法。
- 内視鏡、カテーテル、断層システム、手持ち式光学イメージングシステムまたは術中顕微鏡を用いて照射ステップおよび検出ステップを実施する、請求項45に記載の方法。
- 放射シグナルの有無またはレベルが疾患状態を示す、請求項44または45に記載の方法。
- この方法を疾患の検出および/または監視に使用する、請求項44または45に記載の方法。
- 疾患が、骨疾患、癌、心血管疾患、アテローム性動脈硬化症、再狭窄、心虚血、心筋再灌流傷害、環境疾患、皮膚疾患、免疫疾患、遺伝性疾患、感染症、炎症疾患、代謝疾患、神経変性疾患、眼疾患および呼吸器疾患からなる群から選択される、請求項54に記載の方法。
- ステップ(a)において、インテグリン結合作用剤で標識した細胞を被検体に投与する、請求項44または45に記載の方法。
- インテグリン結合作用剤によって放射されるシグナルを用いて、細胞の輸送と局在化を監視する、請求項56に記載の方法。
- 被検体における血管新生をイメージングする方法であって、
(a)請求項1〜43のいずれか1項に記載の作用剤を被検体に投与するステップと、
(b)作用剤の存在を検出することで、被検体における新たな血管の形成を表す画像を生成するステップと、
を含む方法。 - 請求項1〜43のいずれか1項に記載の作用剤を全身または局所的に被検体に投与することを含む、被検体における疾患を治療する方法であって、作用剤が、疾患部分において局在化し、実効線量の放射線を送達する放射標識を含む、方法。
- (a)請求項1〜43のいずれか1項に記載の作用剤を試料と接触させ、
(b)試料中に生物学的標的が存在する場合、作用剤がこれと結合できるようにし、
(c)任意に、未結合の作用剤を除去し、
(d)作用剤から放射されるシグナルを検出することで、作用剤が活性化されているか生物学的標識に結合したかを判断することを含む、in vitroイメージング方法。 - 試料が生物学的試料である、請求項60に記載の方法。
