Abstract
Description
この出願は、米国特許法§119(e)の下、2008年4月25日に出願された、米国仮特許出願第61/048,130号(この全体の内容は、参考として本明細書に援用される)の利益を主張する。
本開示は、Abi1/Hssh3bp1遺伝子に条件的欠失を有する、遺伝的に操作したコンディショナルノックアウトマウスおよび関連する方法に関連する。
遺伝的に操作したAbi1/Hssh3bp1マウス
Abi1/Hssh3bp1ヘテロ接合性loxP導入マウス(Abi1/Hssh3bp1loxP+/wt、Abi1/Hssh3bp1(fl/+)とも呼ばれる)の胚を、American Type Culture Collection(Rockville、MD)に、受入番号XXXXXXで寄託し、そしてマウス胚9050671VF−1から9050671VF−10として同定した。これらのマウスは、遺伝子の条件的破壊のために必要な最小限の量の配列を有する(Abi1/Hssh3bp1のエキソン1周辺のloxP部位、下で「エキソン1」)。これらのマウスは、組換えAbi1/Hssh3bp1遺伝子座にネオマイシン遺伝子を有さない;neoカセット由来の少量の配列が残っている(図2Aを参照のこと)。Abi1/Hssh3bp1loxP+/wtを繁殖させて、Abi1/Hssh3bp1loxP+/loxP(Abi1/Hssh3bp1(fl/fl))マウスを得る。
正常および変異Hssh3bp1遺伝子を有する細胞系統
実施例1で記載されたヘテロ接合体マウスから、細胞系統を単離した。マウス胚線維芽細胞のためにMEFと呼ばれるその細胞系統は、1コピーの野生型遺伝子および1コピーの遺伝的に組換えした、エキソン1に隣接するloxP配列を有するAbi1/Hssh3bp1遺伝子を有する(この細胞系統はネオマイシン遺伝子を含んでいた)。初代MEFの単離後、その細胞をPCRによって遺伝子型解析した(図5)。いくつかのMEF細胞系統を、無作為に選択した。これらは、野生型Abi1/Hssh3bp1を発現する細胞系統、および望ましいホモ接合性loxP導入Abi1/Hssh3bp1遺伝子を発現する系統を含んでいた(図5)。
Abi1/Hssh3bp1遺伝子発現を欠く細胞の形態
Abi1/Hssh3bp1は、前駆(fl/fl)細胞においてPDGF(血小板由来増殖因子)刺激末梢および背面ラフリングに局在するが、ノックアウトMEF細胞には存在しない。Abi1/Hssh3bp1は、ラメリポディウムを生じるアクチン再構成、およびアクチンが豊富な末梢および背面ラフリングの形成に関与することが公知である。従って、単離したMEF細胞において、Abi1/Hssh3bp1がこれらの構造に局在するかどうか、および機能的Abi1/Hssh3bp1遺伝子の発現を欠く細胞において、これらの構造におけるいかなる欠損が観察されるかを決定した。
Abi1/Hssh3bp1ノックアウトマウスゲノムの配列決定
下記に示した供給源から全ゲノムDNAを単離し、そして示したプライマー(PCRプライマー)を用いてPCR増幅にかけた。PCR断片を1%アガロースゲルで分離した(図14)。PCR産物バンドの配列決定を、5’loxP部位の上流に位置する、プライマーDL75’(前向き配列決定プライマー;配列番号第7番)、および3’loxP部位の下流に位置する、プライマーFlankneo13’(逆向き配列決定プライマー;配列番号第4番)を用いて行った。MEF細胞系統およびマウスの名前を示す。
「RARA1」を、ヘテロ接合性Abi1/Hssh3bp1(loxP/+)系統の繁殖から生じ、そしてloxP対立遺伝子に関してネガティブであると遺伝子型解析されたマウス胚から得る。配列決定データは、Abi1/Hssh3bp1遺伝子のWT配列において、5’loxP部位の欠如および3’loxP部位の欠如を確認する。
MEF#3(親#3MEF Abi1/Hssh3bp1 loxP導入細胞系統):配列決定は、組換えAbi1/Hssh3bp1遺伝子において、5’および3’loxP部位両方の存在を確認した。
マウス#418前方および後方前立腺:配列決定は、組換えAbi1/Hssh3bp1遺伝子において、5’および3’loxP部位両方の存在を確認した。
マウス#419前方および後方前立腺:配列決定は、組換えAbi1/Hssh3bp1遺伝子において、5’および3’loxP部位両方の存在を確認した。
Abi1/Hssh3bp1ノックアウトマウス
組換えloxP導入Abi1/Hssh3bp1対立遺伝子に関してホモ接合性であるマウスを、プロバシンプロモーター駆動Creリコンビナーゼ(Pb−Cre)を発現する系統:B6.D2−Tg(Pbsn−Cre)4Prb(National Cancer Institute−Frederick;the Mouse Repository of the Mouse Models of Human Cancers Consortium)と繁殖させた。これらのトランスジェニックマウスにおいて、ラットプロバシン遺伝子(Pb)の前立腺特異的プロモーターが、Creリコンビナーゼの発現を制御し、従ってその酵素は、成熟前立腺組織においてのみ発現することが予測される。Pb−Cre系統を、C57BL/6Jメスと繁殖させることによって、ヘミ接合性状態で維持する。Creは、小さい、しかし有意な卵母細胞による組換えを避けるために、オスマウスを通じて伝達しなければならない。繁殖ペアは、ヘミ接合性オスおよびメスC57BL/6Nのつがいとして供給された。Pb−Creマウスの連続的な世代を、C57BL/6Jマウスと繁殖させて、実験のために十分なPb−Creマウスを産生した。尾部から得たDNAを用いて、マウスを遺伝子型解析した。
Abi1/Hssh3bp1を欠く細胞の増殖
Abi1/Hssh3bp1を欠く細胞の増殖速度は、コントロールAbi1/Hssh3bp1(fl/fl)MEF細胞よりも速い。原発性前立腺腫瘍におけるAbi1/Hssh3bp1の喪失、および続くAbi1/Hssh3bp1機能の破壊を引き起こす原発性腫瘍の変異の同定は、Abi1/Hssh3bp1タンパク質の喪失が、細胞増殖の調節不全を引き起こすことを示唆した。これを、Abi1/Hssh3bp1 CKOマウスから単離したMEF細胞を用いて試験した。腫瘍抑制因子仮説と一致して、MEF Abi1/Hssh3bp1(−/−)細胞(クローン#3−11)において、親クローン#3Abi1(+/+)と比較して、ミトコンドリアデヒドロゲナーゼの活性によって決定された増殖速度の増加が観察された(図13)。プライマーDL75’およびFlankneo13’を、遺伝子型解析のために使用した。
前立腺癌におけるAbi1/Hssh3bp1の役割
特定の染色体領域の欠失が、ヒト前立腺腫瘍において報告された。第10染色体に関して、10pおよび10qアームの両方が、しばしば欠失していると報告された。10qの欠失は、多くの場合候補前立腺癌抑制遺伝子、PTENまたはMMAC1にマッピングされた配列を含む、10q23−24領域を含む。第10染色体の短腕、10pにおけるヘテロ接合性の喪失(LOH)も、前立腺腫瘍において観察された。多型マーカーを用いて行われたいくつかの研究は、特に10p11.2領域における高率のLOHを示した。LOHの率は、使用したマーカーおよび調査した癌のステージに依存して、研究の間で変動する。10pにおける遺伝的変化は、多くの場合10qにおける変化と関連して存在する。13ヶ所の高度の多型の遺伝子座におけるヒト前立腺腫瘍の10pの広範囲な欠失マッピングを行った。この研究において、調査した35個の腫瘍の57%が、10p配列の喪失を示した。10pにおける最も高い濃度の対立遺伝子喪失は、4−から7−cM領域にまたがり、そして遺伝子座D10S211、D10S89、およびD10S111を含んでおり、それらはヒト前立腺腫瘍において10pの最少共通欠失領域を規定する。さらに、いくつかの腫瘍は、D10S211またはD10S89−D10S111においてのみ欠失していたので、この研究は、1つまたはそれ以上の欠失ドメインを10pにマッピングし得ることを示唆した。これらの研究は、マーカーD10S111を用いて、局在化(ステージB)前立腺癌における3.2%のLOH、および進行前立腺癌(ステージCおよびD)における27%のLOHという観察によって確認された。合わせると、これらの対立形質解析(allelotyping)研究は、ヒト前立腺腫瘍において1つまたはそれ以上の癌抑制遺伝子が、10pにマッピングされることを示唆する。この仮説を支持する機能的研究が、染色体10pの一部を加えた前立腺細胞系統における研究によって提供された。10pter−q11を含む染色体下断片の、PC3前立腺癌細胞系統への導入は、ヌードマウスにおいてハイブリッドクローン注入後の腫瘍形成能を抑制した。PPC−1細胞系統、PC3の亜系統を用いた別のグループによって、同様の結果が得られ、ここで10p配列の導入後に軟寒天におけるコロニー形成の減少が観察された。
10p最少欠失領域の完全なコンティグを含む、11個のCEPH YAC(965D10、746D9、815C7、747H10、857C9、934E11、796F8、899E10、875B4、746G7、および961C7)のそれぞれ由来のコロニーを取り、そして10μlのリチカーゼ溶液(1.2Mソルビトール、10mMリン酸ナトリウム、pH7.4[1Mのストックから1:4v/v一塩基:二塩基])および2.5mg/mlリチカーゼ(Sigma)と、37℃で5分間インキュベートした。各消化混合物の5マイクロリットルを、続く55℃のアニーリング温度を用いた、それぞれ200μMのdGTP、dATP、dTTP、およびdCTP;1×PCR緩衝液(50mMのKCl、10mMのTris−HCl、pH8.3、2.5mMのMgCl2);それぞれ1μMの前向きおよび逆向きプライマー;および0.6UのTaqポリメラーゼ(Life Technologies)を含むポリメラーゼ連鎖反応(PCR)において使用した。これらのマーカーの連結順序は、pter−D10S211−WI4906−10S553−D10S1789−D10S550−WI4133−D10S582−D10S1673−D10S586−D10S1749−D10S1747−D10S572−D10S89−D10S111−セントロメアと報告された。
10p遺伝子量(dosage)状態に関して以前に特徴付けた、凍結した1対の正常および腫瘍前立腺組織を利用した。標準的な技術を用いて、the Institute for Basic Research in Developmental Disabilities Antibody Facilityにおいて、Hssh3bp1の組換えC−末端部分、プラスミドC5に対して、モノクローナル抗体(mAb)2G8を産生した。1対の凍結正常および前立腺全摘除術標本由来の悪性組織から切断し、そして氷冷アセトンで10分間固定し、次いで4℃で短時間空気乾燥した、5ミクロンの切片において免疫組織化学を行った。そのスライドを、Ventana320ES Automatic Immunohistochemistry/IPOX Staining Stationを用いて、製造会社のプロトコールに従って、1:2000希釈のmAb 2G8で染色した。その抗体染色を病理学者が評価し、そして染色の程度を、0(無し)、1(低度)、2(中程度)、または3(強度)と評価した。
前立腺細胞系統LNCAP.FGC−10(CRL−10995)、LNCaP.FGC(CRL−1740)、およびPC3(CRL1435)をATCCから得て、そしてATCCの指示に従って増殖させた。培養細胞由来のRNAを、Tri−Reagent(Molecular Research Center)を用いて調製した。Hssh3bp1特異的プライマーT7−M(5’−GATTAATACGACTCACTATAGGGACGCGAGAGGAAGCGATGC AGAG−3’、5’プライマー、配列番号第12番)、およびP3(5’−CTTGAATTCAAGCAAATCAGTGAAGGAAAGGAC−3’、3’プライマー、配列番号第13番)を用いて、RT−PCRを行った。ゲル精製PCR産物(200ng)のインビトロにおける翻訳を、T7−h1プライマーおよびT7 TNTシステム(Promega)を用いて行った。SDS−PAGEタンパク質電気泳動およびウェスタンブロッティングを行った。その分析において、Hssh3bp1に対するポリクローナル抗体Ab−2を用いた。
11個のCEPH YACをそれぞれ、10p最少共通欠失領域内の14遺伝子座のマッピングのために増幅した。これらの実験は、YACをこの領域にまたがるコンティグに整列させた(order)(表1)。Hssh3bp1遺伝子はYAC961C7のみに局在し、それはまたマーカーD10S89およびD10S111に特異的な配列を含む。D10S89はまた、よりテロメア側のYAC、875B4にも局在するので、可能性のある配列順は:10pter−D10S89−Hssh3bp1/D10S111−10cenであり、ここで「/」は実際の方向は不明であることを示す(表1)。1.67Mbである、YAC961C7の比較的小さいサイズは、Hssh3bp1遺伝子は、D10S89またはD10S111が欠失した腫瘍において、同時に欠失し得る可能性を示唆した。
Hssp3bp1に対するmAbを用いた前立腺組織の免疫組織化学的分析を行って、前立腺腫瘍においてHssp3bp1タンパク質発現がD10S89またはD10S111量と関連するかどうかを決定した。D10S89およびD10S211の量に関して以前に特徴付けた、17組の正常および悪性前立腺標本を、これらの研究のために利用した。17個の腫瘍組織のうち、6個がD10S89またはD10S111における欠失によって特徴付けられた(表2)。残りの11個の腫瘍は、D10S89、D10S111、または両方の遺伝子座において、正常な二倍体量を保持していた(表2)。上皮細胞質の免疫組織化学的染色を、4つのグループ:なし(0)、低度(1)、中程度(2)、または強度(3)に段階をつけた。Hssp3bp1の中程度または強度の発現を、試験した82%(14/17)の正常組織で検出した(表2)。対照的に、Hssp3bp1の中程度または強度の発現を、試験した41%(7/17)の悪性組織においてのみ検出した。さらに、最少共通欠失領域内で、D10S89またはD10S111における10p配列が欠失した4/6(67%)の腫瘍が、これらの遺伝子座において正常な二倍体量を維持する5/11(46%)の腫瘍と比較して、Hssp3bp1タンパク質を発現できなかった。正常および悪性におけるHssh3bp1染色の例を、図16に示す。
2つの前立腺腫瘍細胞系統、LNCaP(CRL−10995)およびPC3(CRL−1435)において、PTTを行った。そのPTTテストは、Hssh3bp1のC−末端に対する抗体を使用し、そしてPC3細胞系統における2つのポリペプチドと比較して、LnCaP細胞系統における短縮ポリペプチドの存在を示した(図16)。DNA配列決定によって決定されたように、LNCaP細胞系統は、Hssh3bp1cDNAのヌクレオチド660−800(全部で141ヌクレオチド)の欠失を含むが、PC3細胞系統は含まない。これは、Hssh3bp1のアミノ残基194から240までのインフレーム欠失を引き起こし、そしてPTTにおけるより小さい産物の翻訳の観察と一致する。Hssh3bp1配列の同一の欠失が、別の腫瘍細胞系統、CRL−1740において観察された。これらの細胞系統由来のHssh3bp1遺伝子の配列決定は、前のイントロンの3’スプライスジャンクション近くに位置する遺伝子のエキソン6における、ヘテロ接合性の点突然変異の存在を明らかにした:配列
Claims (14)
- 体細胞および生殖細胞が、条件的に破壊されたAbi1/Hssh3bp1遺伝子を含むコンディショナルノックアウトマウスであって、ここで該破壊は、該マウスが検出可能なレベルのAbi1/Hssh3bp1タンパク質を産生できないことを引き起こす、コンディショナルノックアウトマウス。
- 前記条件的破壊は、前記マウスを、フリッパーゼまたはCreリコンビナーゼを発現するマウスと繁殖させることによって誘発される、請求項1に記載のコンディショナルノックアウトマウス。
- 前記Abi1/Hssh3bp1遺伝子の少なくとも一部に隣接する、ネオマイシン遺伝子、frt部位およびloxP部位を含む、組換えAbi1/Hssh3bp1対立遺伝子を含む、請求項1に記載のコンディショナルノックアウトマウス。
- 前記Abi1/Hssh3bp1遺伝子の前記少なくとも一部は、該Abi1/Hssh3bp1遺伝子のエキソン1である、請求項3に記載のコンディショナルノックアウトマウス。
- 前記条件的破壊は、前記Abi1/Hssh3bp1遺伝子のエキソン1において起こる、請求項1に記載のコンディショナルノックアウトマウス。
- 前記Abi1/Hssh3bp1遺伝子は、全ての前記マウスの組織において発現されない、請求項1に記載のコンディショナルノックアウトマウス。
- 前記Abi1/Hssh3bp1遺伝子は、前記マウスの組織の一部のみにおいて発現されない、請求項1に記載のコンディショナルノックアウトマウス。
- 前記Abi1/Hssh3bp1遺伝子は、前記マウスの前立腺組織において発現されない、請求項1に記載のコンディショナルノックアウトマウス。
- 前記マウスは、細胞運動性の破壊、方向持続性の増加、遊走距離の減少、および遊走速度の減少からなる群より選択される、少なくとも1つの表現型を示す、請求項1に記載のコンディショナルノックアウトマウス。
- 請求項1に記載のマウスから単離された細胞。
- 前記細胞は、前記マウスの前立腺組織由来である、請求項10に記載の細胞。
- Abi1/Hssh3bp1遺伝子の一部を含む、Abi1/Hssh3bp1遺伝子コンディショナルノックアウト構築物であって、ここで該Abi1/Hssh3bp1遺伝子のエキソン1は、5’loxP部位および3’選択マーカーカセットに隣接し、ここで該選択マーカーカセットは、frt部位および、3’frt部位の3’側および5’frt部位の3’側のloxP部位に隣接する選択マーカーを含む、Abi1/Hssh3bp1遺伝子コンディショナルノックアウト構築物。

- Abi1/Hssh3bp1遺伝子において標的化条件的破壊を有するマウスを産生する方法であって、該方法は、
請求項12に記載のノックアウト遺伝子構築物を、マウス胚幹(ES)細胞の集団にトランスフェクトする工程;
選択マーカーを発現するトランスフェクトしたES細胞を選択する工程;
該トランスフェクトしたES細胞を、該マウスの祖先の胚に導入する工程;
該胚を満期まで発生させて、生殖系列にコンディショナルノックアウト構築物を有するキメラマウスを産生する工程;
該キメラマウスを繁殖させて、条件的に破壊可能なAbi1/Hssh3bp1遺伝子を有するヘテロ接合性マウスを産生する工程;および
該ヘテロ接合性マウスを、フリッパーゼまたはCreリコンビナーゼを発現するマウスと繁殖させて、該Abi1/Hssh3bp1遺伝子に破壊を有し、そして該選択マーカーを含まないマウスを産生する工程、
を包含する、方法。
