DCは、当技術分野で公知であるプロトコルを用いて、哺乳動物の骨髄培養、末梢血、脾臓または他の任意の適当な組織から得ることができる。骨髄はDC前駆体を含み、それは、顆粒球マクロファージコロニー刺激因子(「GM−CSF」)およびインターロイキン4(「IL−4」)などのサイトカインによる処理の後に増殖して、DCに分化する。腫瘍壊死細胞因子(TNF)は、DCの成熟を促進するために、任意選択で、単独で用いられるか、またはGM−CSFおよび/またはIL−4と併用される。骨髄から得られるDCは、(例えば脾臓DCと比較して)比較的未熟である。GM−CSF/IL−4によって刺激されたDCは、MHCクラスIおよびクラスIIの分子、B7−1、B7−2、ICAM、CD40および様々なレベルのCD83を発現する。これらの未熟なDCは、脾臓で見られるより成熟したDCよりも融合(または抗原取込み)に適合するが、より成熟したDCは比較的より有効な抗原提示細胞である。末梢血も比較的未熟なDCまたはDC前駆体を含み、それらは、GM−CSFなどの適当なサイトカインの存在下で増殖および分化することができ、融合で用いることもできる。
DCと非樹状細胞との間の融合は、ポリエチレングリコール(「PEG」)、センダイウイルスまたは電気融合を用いるものなどの、周知の方法で実行することができる。DCは自己由来であるか、同種異系である。(例えば、参照によりその全体が本明細書に組み込まれている、米国特許第6,653,848号を参照)。融合の非樹状細胞に対するDCの比率は、1:100〜1000:1と異なることができ、非樹状細胞が培養で激しく増殖する1:1より高い比率が好ましい。最も好ましくは、比率は1:1、5:1または10:1である。融合の後、未融合のDCは培養中で通常2、3日中に死滅するので、融合細胞は下記2つの方法によって未融合の親の非樹状細胞から分離することができ、両方法は、約50%以上の純度の融合細胞を生成し、すなわち、融合細胞調製物は50%未満、しばしば30%未満の未融合細胞を含有する。
本発明の融合細胞は、疾患の治療または予防のために、哺乳動物の免疫系を刺激するために用いることができる。例えば、ヒトで原発性または転移性の腫瘍を治療するために、患者自身のDCおよび腫瘍細胞によって形成される融合細胞を含む組成物を患者に、例えばリンパ系組織の近くの部位に投与することができる。組成物は、適当な間隔(例えば、2〜3週間ごと)および投薬量(例えば、1投与につき約105〜108、例えば約0.5×106〜1×106個の融合細胞)で複数回(例えば、3〜5回)投与されてもよい。癌に対する予防(すなわち、ワクチン接種)のために、同系DCおよび同種異系もしくは異種の癌細胞、または同種異系DCおよび癌細胞によって形成されるものなどの、非同系融合細胞を投与することができる。ワクチン接種の効果を監視するために、治療される個体から得られる細胞傷害性Tリンパ球を、癌細胞に対するそれらの効力について細胞傷害性アッセイで試験することができる。細胞傷害性Tリンパ球の効力を高めるために、複数のブーストが必要かもしれない。
本発明は、ハイブリッド細胞を代償にして培養で増殖する、教育された、抗原特異的な免疫エフェクター細胞の集団も提供し、そこで、ハイブリッド細胞は、1つまたは複数の抗原を発現する細胞に融合している、抗原提示細胞(APC)を含む。一実施形態では、APCは樹状細胞(DC)であり、ハイブリッド細胞は培養で増殖する。別の実施形態では、抗原(複数可)を発現する細胞は腫瘍細胞であり、免疫エフェクター細胞は細胞傷害性Tリンパ球(CTL)である。DCは、血液、皮膚、脾臓、骨髄または腫瘍などの源から単離することができる。細胞集団を調製する方法も、本発明によって提供される。
本発明は、抗原特異的(すなわち、「教育された」)免疫エフェクター細胞の濃縮集団の生成を刺激するために、これらのハイブリッド細胞を利用する。抗原特異的な免疫エフェクター細胞は、培養中に死に至るハイブリッド細胞を代償にして増殖する。ナイーブな免疫エフェクター細胞が他の細胞によって教育される工程は、基本的にCoulie、Molec. Med Today261〜268頁(1997年)に記載されている。
本明細書で記載されるように、本明細書で記載される細胞の有効な量を、養子免疫療法を提供するために対象に投与することができる。サイトカインまたは他の同時刺激分子の有効な量を、対象に共投与することもできる。
本発明の方法は、ハイブリッド細胞、または、刺激因子としてハイブリッド細胞を用いて誘導される細胞の抗原特異的集団に対する、遺伝子導入の任意の方法を包含するものとする。遺伝子改変の例には、ウイルス媒介遺伝子導入、リポソーム媒介導入、形質転換、トランスフェクションおよび形質導入、例えば、アデノウイルス、アデノ随伴ウイルスおよびヘルペスウイルスなどのDNAウイルスに基づくベクター、ならびにレトロウイルスに基づくベクターの使用などのウイルス媒介遺伝子導入が含まれるが、これらに限定されない。本方法は、核局在化要素もしくは配列を欠くので、核酸が細胞質にとどまるベクターまたは構築物に含まれる核酸の組込みに特に適している。これらの例では、核酸または治療的遺伝子は、核膜が破壊され、核酸または治療的遺伝子が宿主細胞染色体に接近することができるM(有糸分裂)相の間に、核に入ることができる。遺伝子改変はエキソビボで実施され、その後、組換え(すなわち、形質導入)細胞はレシピエントに投与される。したがって、本発明は、本明細書で開示される方法によってエキソビボまたはインビボで遺伝子を投与することによる、抗原特異的細胞への遺伝子導入に適合する疾患の治療を包含する。
上記のCTLおよびHTL(「エフェクター細胞」)は、本発明のエフェクター細胞を生成するために用いられる融合細胞の非樹状細胞パートナーによって発現される抗原を、当技術分野で用いられるいくつかの方法によって同定するために用いることができる。手短に言えば、エフェクター細胞を含有する細胞集団を候補ペプチドまたはポリペプチド、および、適当な標的細胞(細胞毒性をアッセイする場合)または抗原提示細胞(APC)(細胞増殖またはサイトカイン生成をアッセイする場合)と一緒に培養し、関連する活性を測定する。エフェクター活性を誘導するペプチドは抗原性のペプチドであり、それは、エフェクター細胞によって認識される。エフェクター活性を誘導するポリペプチドは抗原性のポリペプチドであり、そのペプチド断片はエフェクター細胞によって認識される。
本明細書で記載される教育され、増殖したT細胞集団および方法は、細胞ベースのワクチンを開発するために用いることもできる。本発明による抗原特異的な免疫エフェクター細胞を含むワクチンが、本発明によってさらに提供される。本明細書に記載の抗原特異的な免疫エフェクター細胞を利用する、エピトープまたは配列モチーフなどの抗原またはその断片を含むワクチンが、本発明によってさらに提供される。ワクチンの投与方法は当技術分野で公知であり、ワクチンは、許容される薬用担体と組み合わせることができる。サイトカインおよび/または同時刺激分子の有効な量も、ワクチンに加えて投与することができる。
DCは、正常な供与体から得られたロイコパックコレクションから単離された、接着性単核細胞から生成された。末梢血単核細胞(PBMC)を、Histopaque(登録商標)−1077密度勾配遠心法によって、正常な供与体からのロイコパックから単離した。PBMCを、RPMI 1640完全培地に1×106/mlで懸濁させ、6ウェル組織培養プレートに5mlの一定量で平板培養し、5%CO2の加湿インキュベータ内において37℃で2時間インキュベートした。単球濃縮接着性分画を、GM−CSF(1000U/ml)およびIL−4(1000U/ml)を含有するRPMI 1640完全培地で5日間培養して、未熟なDCを生成した。TNFα(25ηg/ml)の存在下で細胞をさらに48時間培養することによって、DC調製物を成熟させた。
DC、腫瘍および融合細胞調製物は、腫瘍関連抗原ならびにDC関連同時刺激および成熟マーカーの存在について評価するために、免疫細胞化学分析にかけた。RCC細胞を、MUC1(PharMingen、San Diego、CA)、サイトケラチン(Boehringer Mannheim、Indianapolis、IN)およびCAM(Becton Dickson、San Jose、CA)に対する、マウスの一次モノクローナル抗体(mAb)による染色にかけた。骨髄性白血病細胞を、CD34、CD117およびMUC1について染色した。下で概説するDCマーカーの非存在が、確認された。(図1Bを参照)。DC調製物を、HLA−DR、CD80、CD83またはCD86(PharMingen)およびアイソタイプをマッチさせた陰性対照のための染色に60分間かけた。(図1Aを参照)。細胞をウマ抗マウスIgG(Vector Laboratories)のビオチン化F(ab’)2断片と45分間インキュベートし、PBSで2度洗浄し、ABC(アビジン−ビオチン複合体)試薬液、および続くAEC(3アミノ−9−エチルカルバゾール)溶液(Vector Laboratories)と30分間インキュベートした。融合細胞調製物では、ABC試薬による腫瘍関連抗原の検出に続いて、ABC−AP(アルカリホスファターゼ)キット(Vector Laboratories)によるDC関連マーカーのための染色を行った。スライドを洗浄し、2%パラホルムアルデヒドで固定し、オリンパスAX70顕微鏡を用いて分析した。融合は、特異なDCおよび腫瘍抗原を同時発現する細胞のパーセントを測定することによって定量した。(図1Cを参照)。
上で概説した抗原の発現について評価するために、DC、腫瘍および融合細胞調製物をフローサイトメトリー分析にもかけた。細胞を、指示された一次mAbまたはマッチさせたアイソタイプ対照と、4℃で30分間インキュベートした。結合した一次mAbは、二次親和性精製FITC結合ヤギ抗マウスIgG(Chemicon Intl、Temecula、CA)で検出し、続いて2%パラホルムアルデヒドで固定した。二次元フローサイトメトリーのために、細胞を、腫瘍関連抗原(RCC−MUC1、CAMまたはサイトケラチン、AML−CD34、CD117またはMUC1)に対する抗体、FITC結合二次抗体、およびPEとコンジュゲートさせたDRまたはCD86に対する抗体とインキュベートした。分析は、CellQuestソフトウェア(Becton Dickinson)を用いて、FACS Caliburフローサイトメータ(Becton Dickinson)で実施した。
DC生成のために用いたロイコパックコレクションから非接着性PBMCを単離し、10U/mlのIL−2の存在下のRPMI完全培地において1×106/mlの密度で培養した。T細胞は、ナイロンウール分離によって単離した。T細胞を、固定化モノクローナル抗体、抗CD3(クローン−UCHT1;Pharmingen)および抗CD28(クローン−CD28.2;Pharmingen;CD3i/CD28i)に曝露させた。24ウェル非組織培養処理プレート(Falcon、Fisher)を抗体(PBSに1μg/ml)の各々でコーティングし、37℃で一晩放置した。T細胞を:1)抗CD3/CD28でコーティングしたプレート上で48時間培養した;2)1:10の融合対T細胞の比率で、融合細胞と5日間共培養した;3)融合細胞、およびその後抗CD3/CD28でコーティングしたプレートと48時間共培養した;または4)抗CD3/CD28と48時間培養し、その後、融合で5日間刺激した。刺激の後、下で概説するように、T細胞を表現型分析にかけた。
刺激の後、T細胞を収集し、培養期間の終わりの18時間前に各ウェルに加えた[3H]−チミジン(1μCi/ウェル;37kBq;NEN−DuPont、Boston、MA)の取込みで増殖を測定した。その後、自動TOMTECハーベスター(Mach II、Hamden CT)を用いてガラス繊維ろ紙(Wallac Oy、Turku、Finland)の上に細胞を収集し、乾燥させ、10mlのScintiVerse(登録商標)(Fisher Scientific、Fair Lawn、NJ)を含むBetaPlate試料バッグ(Wallac)中に入れて密封した。細胞に結合した放射活性を、液体シンチレーション計数器(Wallac、1205Betaplate(商標))で計数した。(図2を参照)。データを、刺激指数(「SI」)で表す。SIは、未刺激T細胞集団のバックグラウンド[3H]−チミジン取込み(トリプリケートの平均値)に対する[3H]−チミジン取込み(トリプリケートの平均値)の比率を計算することによって測定した。T細胞は、DC/RCC融合または抗CD3/CD28単独への曝露の後に有意な増殖を示さず、SIはそれぞれ0.9および1.0であった(N=9)。対照的に、DC/RCC融合による刺激とそれに続くCD3/CD28への曝露は、T細胞増殖の劇的で相乗的増加をもたらし、SIは13.2であった(融合単独による刺激と比較してp=0.03)。(図2Aを参照)。留意すべきは、融合細胞による刺激の前の抗CD3/CD28への曝露は、T細胞増殖を誘導しなかった(SI 1.0)。
DC/RCC融合および抗CD3/CD28による複合刺激が、調節T細胞と比較して活性化T細胞の増殖をもたらしたかの判定もした。これらの2つのT細胞集団は、CD4およびCD25を同時発現する。活性化T細胞は高レベルのCD69を特徴的に発現するが、Foxp3は、調節T細胞の比較的特異的なマーカーであることが分かった。DC/RCC融合、抗CD3/CD28、またはこれらの剤による逐次的な刺激によって刺激されたT細胞の表現型特性を調べた。(図3を参照)。11回の実験で、DC/RCC融合単独による刺激の後に、CD4+/CD25+の控え目な増加が観察された。総T細胞集団のCD4+/CD25+細胞の平均パーセントは、2.8%から6.4%に増加した。同様に、抗CD3/CD28単独によるT細胞の刺激の後に、CD4+細胞の7.8%は、CD4およびCD25の同時発現を示した。
抗原特異的MUC1+CD8+T細胞を、それぞれMUC1特異的エピトープM1.2(MUC112〜20)LLLLTVLTV(配列番号1)(Beckman Coulter、Fullerton、CA)に結合している4つのHLA MHCクラスI分子で構成される、フィコエリトリン(PE)標識HLA−A*0201+iTAg(商標)MHCクラスIヒト四量体複合体を用いて同定した。対照PE標識四量体を、平行して用いた。抗CD3/CD28、融合、または抗CD3/CD28および融合への逐次的な曝露によって刺激したT細胞を、MUC1または対照四量体とインキュベートし、次に、FITC結合CD8抗体で染色した。細胞を洗浄し、二次元FACS分析によって分析した。刺激されたT細胞集団の細胞溶解能力を、FITC結合CD8およびPE結合グランザイムBによる染色によって評価した。合計3×105個の事象を、最終分析のために収集した。同様に、非接着性未刺激細胞を、平行して分析した。
T細胞集団の機能的特性をさらに特徴づけるために、融合細胞、抗CD3/CD28またはそれらの組合せによって刺激されたT細胞による、Th−1およびTh−2サイトカインの細胞内発現を特定した。8回の連続した検査で、IFNγの細胞内発現が、未刺激CD4+T細胞集団の0.5%で観察された。抗CD3/CD28またはDC/RCC融合単独による刺激の後、IFNγ発現T細胞の平均パーセントは、それぞれ1.7%および1.8%に上昇した。対照的に、DC/RCC融合および抗CD3/CD28による逐次的な刺激は、IFNγ発現細胞の平均レベルの統計的に有意な増加をもたらし(それぞれ抗CD3/CD28または融合による刺激と比較して4.7%、p=0.05)、それは、未刺激T細胞と比較して10.5倍の増加に相当する(p=0.008)(図16)。抗CD3/CD28単独によるT細胞の刺激は、IL−4の細胞内発現を示すCD4+T細胞のパーセントの、1.0%から2.4%への増加をもたらした。対照的に、DC/RCC融合単独への曝露またはDC/RCC融合および抗CD3/CD28による逐次的な刺激は、IL−4発現の増加をもたらさなかった(それぞれ0.9%および0.6%)。IL−10の平均細胞内発現は、DC/RCC融合および抗CD3/CD28による刺激の後、0.9から3.4%に増加した。比較では、DC/RCC融合単独による刺激の後、IL−10発現のいかなる増加も観察されなかった。これらのデータは、DC/RCC融合による逐次的な刺激が、IFNγを発現する活性化エフェクター細胞の増殖を誘導し、IL−10を発現するT細胞の増加は比較的より控え目であることを示唆する。
DC/RCC融合および抗CD3/CD28による逐次的な刺激が、腫瘍反応性リンパ球の選択的な増殖をもたらしたかどうかを判断するために、腫瘍関連抗原、MUC1に特異的なT細胞が増殖後に増加したかどうかを調べた(図17)。この分析のために、DCおよびT細胞を、HLA−A2.1供与体から単離した。抗CD3/CD28単独による刺激の後に、CD8集団のわずか0.93%がMUC1四量体に結合した。対照的に、DC/RCC融合との共培養は、MUC1四量体+細胞の増加(2.3%)をもたらした。留意すべきは、DC/RCC融合とそれに続く抗CD3/CD28による逐次的な刺激は、MUC1四量体+細胞の劇的な増加をもたらした(17.3%、融合または抗CD3/CD28単独による刺激と比較してそれぞれp=0.02および0.004)。対照的に、抗CD3/CD28による非特異的刺激とそれに続く融合との共培養は、MUC1四量体+細胞の増殖を誘導しなかった(0.19%)。これらのデータは、DC/RCCによる抗原特異的刺激剤への最初の曝露が、抗CD3/CD28を用いる腫瘍反応性T細胞の以降の増殖のために重要であったことを示唆する。
その後、患者由来の急性骨髄性白血病試料を用いたDC/腫瘍融合、および抗CD3/CD28による逐次的な刺激を受けたT細胞の表現型特性を調べた。高レベルの循環疾患を有する患者の末梢血または骨髄から骨髄性白血病細胞を得、正常なロイコパックコレクションから生成されたDCと融合させた。DC/AML融合は、DC(CD86)および骨髄性白血病に特異な抗原(CD117−ckitリガンド、CD34および/またはMUC1)を同時発現した細胞のパーセントの測定に従って定量化した(図19)。平均融合効率は、総細胞集団の28%であった。DC/AML融合は自己由来T細胞の控え目な増殖を誘導し、SIは3.3で、記憶エフェクター細胞(CD45RO+)は総T細胞集団の10%を構成した。DC/AML融合とそれに続く抗CD3/CD28による逐次的な刺激は、T細胞増殖の統計的に有意な上昇(SI8.2)をもたらし、その39%はCD45ROを発現した(図20)。同様に、DC/AML融合とそれに続く抗CD3/CD28による逐次的な刺激の後に、CD4+/CD25+細胞の上昇が観察された(9.3%に対してDC/AML融合単独による刺激の後に2.7%)。さらに、融合細胞との共培養に続いて抗CD3/CD28に曝露させたとき、増加したパーセントのCD4+/CD25+細胞がIFNγを発現した。Foxp3+細胞のパーセントの上昇も観察されたが、これは統計的有意性を満たさなかった。DC/AML融合および抗CD3/CD28による逐次的な刺激は、CD8+集団の13%でグランザイムB発現を誘導した。対照的に、融合細胞単独、または抗CD3/CD28とそれに続く融合による刺激は、CD8+細胞の2.5%および2.7%でグランザイムB発現をもたらした。RCCモデルで観察された結果に類似して、これらのデータは、DC/AML融合による刺激とそれに続く抗CD3/CD28への曝露が、細胞溶解能力を有する活性化T細胞の有意な増加をもたらしたことを実証する。
単球由来のDCの生成
施設によって承認されたプロトコルに従って、正常な供与体からのロイコパック、および乳癌患者から収集された末梢静脈血から、末梢血単核細胞(PBMC)を単離した。試料をHistopaque(登録商標)−1077(Sigma)密度勾配遠心法にかけ、5%CO2加湿インキュベータ内の組織培養フラスコ(Becton Dickinson、Franklin Lakes、NJ)中の、2mM L−グルタミン(Mediatech、Herndon、VA)を含有し、熱不活性化10%ヒトAB雄血清(Sigma、St.Louis、MO)、100U/mlペニシリンおよび100μg/mlストレプトマイシン(Mediatech)を添加したRPMI 1640培地(完全培地)に、37℃で2時間平板培養した。単球濃縮接着性分画を、GM−CSF(1000U/ml)(Berlex、Wayne/Montville、NJ)およびIL−4(1000U/ml)(R&D Systems、Minneapolis、MN)を含有する完全培地で5日間培養して、未熟なDCを生成した。DC調製物の分画を、TNFα(25ηg/ml)(R&D Systems)、またはTNFα(25ηg/ml)、IL−1β(10ηg/ml)、IL−6(1000U/ml)(R&D Systems)およびPGE2(1μg/ml)(Calbiochem−San Diego、CA)からなるサイトカインの組合せの存在下でさらなる48時間細胞を培養することによって、さらに成熟させた。成熟は、48〜96時間のTNFαへの曝露によって効果的に誘導され、CD80およびCD83の発現の増加がもたらされた。(図4Aを参照)。15回の連続した実験で、未熟のおよび成熟したDC調製物は、同時刺激分子、CD86を強く発現し(それぞれ75%および84%)、CD14の低いレベルの発現を示した。(図4Bを参照)。しかし、成熟したDCは、CD80(20%対9%、p=0.05)およびCD83(31%対7%、p=0.0003)の平均発現の統計的に有意な増加を示した。抗原提示細胞としてのそれらの機能的能力の尺度として、DC調製物を、同種異系T細胞増殖を刺激するそれらの能力について調べた。連続した研究で、未熟なDCとの比較で、成熟DCは同種異系T細胞の高いレベルの増殖を刺激した。
T細胞濃縮カラム(R&D Systems)またはナイロンウールカラム(Polysciences、Warrington、PA)を用いて、非接着性PBMC分画からT細胞を単離した。両方の方法によるT細胞の純度は、CD3表面発現のFACS分析による測定で、90%を超えた。T細胞は、第三者供与体に由来する場合は同種異系に、DC融合パートナーが由来するのと同じ供与体に由来する場合は自己由来に分類された。
施設によって承認されたプロトコルに従って、一次乳癌細胞を悪性の滲出液または切除された腫瘍病巣から得た。ヒト乳癌細胞系MCF−7およびZR75−1を、ATCC(Manassas、VA)から購入した。すべての腫瘍細胞系を、DMEM(高グルコース)、または2mMのL−グルタミン、100U/mlペニシリン、100μg/mlストレプトマイシンおよび10%熱不活性化ウシ胎児血清(HyClone、Logan、UT)を添加したRPMI 1640で維持した。
腫瘍細胞を1:3〜1:10(細胞収量に依存する)の比で未熟もしくは成熟DC調製物と混合し、前もって加温した無血清RPMI 1640培地で3回洗浄した。細胞ペレットを、50%ポリエチレングリコール(PEG)溶液(分子量:1450)/DMSO溶液(Sigma−Aldrich、St.Louis、MO)に再懸濁した。室温で3分後に、前もって加温した無血清RPMI培地でPEG溶液を段々と希釈し、無血清培地で2回洗浄した。融合調製物を、GM−CSF(500IU/ml)を含む完全培地で、5%CO2において37℃で5〜7日間培養した。
未熟および成熟DCで生成された融合細胞集団の表現型特性を、調べた。未熟および成熟DC集団を、PEGとの共培養によって、患者由来の一次乳癌細胞またはMCF−7ヒト乳癌細胞系と融合させた。DCおよび乳癌細胞を、HLA−DR、CD11c、CD14、CD80、CD86、CD83、CD40、CD54、MUC−1、サイトケラチン、およびマッチさせたアイソタイプ対照(Pharmingen−San Diego、CA)に対する一次マウス抗ヒトモノクローナル抗体とインキュベートし、洗浄し、FITC結合ヤギ抗マウスIgG1(Chemicon International−Temecula、CA)と培養した。細胞を2%パラホルムアルデヒド(Sigma)で固定し、FACScan(Becton Dickinson、San Jose、CA)およびCellQuest Proソフトウェア(登録商標)(Becton Dickinson)を用いて、フローサイトメトリー分析にかけた。DC/乳癌融合調製物を、特異なDC(CD11c−Cychrome)および腫瘍抗原(MUC−1またはサイトケラチン−FITC)を同時発現した細胞のパーセントを定量化するために、二重染色にかけた。融合細胞は、特異な腫瘍(MUC−1および/またはサイトケラチン)およびDC(CD11c)抗原を同時発現した細胞のパーセントを測定することによって定量化した。
抗原提示細胞としてのそれらの効力およびTH1応答を刺激するそれらの能力の尺度として、融合細胞集団によるIL−12およびIL−10の発現を調べた。(図6Aおよび6Bを参照)。融合細胞は、DCおよび腫瘍由来の抗原を同時発現した細胞のFACSゲーティングによって単離した。成熟および未熟DCならびに乳癌で生成された融合細胞を、12回の別々の実験で比較した。IL−12およびIL−10を発現する融合細胞の平均パーセントは、融合細胞集団の間で異ならなかった。IL−12は、未熟および成熟DC/乳癌融合のそれぞれ約40%(±6.7SEM)および49%(±6.3SEM)(p=0.35、NS)で、IL−10は、それぞれ約36.3%(±6.4SEM)および40%(±6.4SEM;n=11)(p=NS)で発現された。
ケモカイン受容体CCR7は、流入領域リンパ節内のT細胞通行部位に細胞移動を誘導し、成熟および活性化を受けるDCによって特徴的に発現される。それらの移動能力の尺度として、未熟および成熟DCで生成された融合について、CCR7の発現を測定した。(図6Cを参照)。CCR7は未熟および成熟融合集団の上で顕著に発現され、腫瘍−DC融合が、成熟し、活性化された表現型の発現をもたらしたことが示唆される。12回の実験では、平均CCR7発現が、未熟および成熟DC/乳癌融合のそれぞれ33%(±9SEM)および38%(±7.3SEM;n=11)で観察された。
同種異系T細胞の増殖を刺激するそれらの能力を評価するために、未熟および成熟DCならびにDC/乳癌融合細胞調製物を、96ウェルU底培養プレート(Costar、Cambridge、MA)内で、同種異系の正常な供与体由来のT細胞と1:10、1:30、1:100、1:300および1:1000の比率で、37℃および5%CO2で5日間共培養した。T細胞の増殖を、培養期間の終わりの18時間前に各ウェルに加えた[3H]−チミジン(1μCi/ウェル;37kBq;NEN−DuPont、Boston、MA)の取込みで測定した。その後、自動TOMTECハーベスター(Mach II、Hamden CT)を用いてガラス繊維ろ紙(Wallac Oy、Turku、Finland)の上に細胞を収集し、乾燥させ、10mlのScintiVerse(登録商標)(Fisher Scientific、Fair Lawn、NJ)を含むBetaPlate試料バッグ(Wallac)中に入れて密封した。細胞に結合した放射活性を、液体シンチレーション計数器(Wallac、1205Betaplate(商標))で計数した。データを、刺激指数(SI)で表す。SIは、未刺激T細胞集団のバックグラウンド[3H]−チミジン取込み(トリプリケートの平均値)に対する[3H]−チミジン取込み(トリプリケートの平均値)の比率を計算することによって測定した。
未熟および成熟DC/乳癌融合と培養されたT細胞により分泌されたサイトカインのプロフィールを、サイトメトリックビーズアレイ(CBA)キット(Becton Dickinson)を用いて測定した。未刺激T細胞、または未融合のDCおよび乳癌に曝露させた細胞からの上清は、対照の役目を果たした。上清は細胞回収の前に収集し、−80℃で凍結させた。IL−2、IL−4、IL−5、IL−10、IFNγ、TNFα、IL−12、IL−6、IL−1βおよびIL−8の濃度は、標準のプロトコルに従って炎症性CBAキットを用いて定量した。簡潔に、キットは各サイトカインに特異的な捕捉抗体でプレコートされている、異なる蛍光強度(FL−3)を有する6つのミクロビーズ集団の混合物を提供した。培養上清または提供された標準化サイトカイン調製物を、予備混合されたミクロビーズに加え、次に、二次PE結合抗体と培養した。個々のサイトカイン濃度をそれらの蛍光強度(FL−2)によって示し、次に、CellquestおよびCBAソフトウェア(BD Pharmingen)の標準参照曲線を用いて計算した。アッセイ間の再現性は、3回の別々の実験で3つの異なるレベルのヒト標準の2つの反復試料を用いて評価した。
未熟および成熟DCで生成したDC/乳癌融合細胞調製物を、1:10の比率で自己由来T細胞と7〜10日間共培養した。標準の5時間51Cr放出アッセイで、T細胞エフェクターに自己由来であるDCで生成したDC/乳癌融合を標的細胞として用いた。標的細胞(2×104個の細胞/ウェル)を、51クロム(NEN−DuPont)と37℃で1時間インキュベートし、続いて反復洗浄をした。51Cr放出は、エフェクターおよび標的細胞集団の5時間の共培養の後に定量した。細胞毒性パーセントは、以下の通りに標準のアッセイによるトリプリケートの平均値を使用して計算した:比細胞毒性百分率=[(試料カウント−自然発生カウント)/(最大カウント−自然発生カウント)]×100。自然発生放出は、最大51Cr取込みの25%未満であった。
抗原特異的MUC1+CD8+T細胞は、それぞれMUC1特異的エピトープM1.2(MUC12〜20)LLLLTVLTV(配列番号1)(Beckman Coulter、Fullerton、CA)に結合されている4つのHLA MHCクラスI分子で構成される、フィコエリトリン(PE)標識HLA−A*0201+iTAg(商標)MHCクラスIヒト四量体複合体を用いて同定された。対照PE標識四量体を、平行して用いた。非接着性細胞をDC/乳癌融合細胞と5日間共培養し、回収し、MUC1または対照四量体とインキュベートし、次に、FITC結合CD8抗体で染色した。細胞を洗浄し、二次元FACS分析によって分析した。合計3×105個の事象を、最終分析のために収集した。同様に、非接着性未刺激細胞を、平行して分析した。
自己由来および同種異系のT細胞調製物を、成熟DC/乳癌融合と10:1の比率で5日間共培養した。細胞調製物を、FITC結合抗CD4、シトクロム結合抗CD25、およびPE結合抗CD69、抗GITRまたは抗CTLA−4とインキュベートした。あるいは、細胞を透過性にし、IFNγ、IL−10、IL−4またはFOXP3に対するPE結合抗体と培養した。次に、細胞を多チャネルフローサイトメトリーによって分析した。一部の検査では、CD4+T細胞を磁気ミクロビーズ単離(Miltenyi Biotec)によって単離し、生じた集団をCD25抗体および指示されたマーカーによる2つの染色方法にかけた。
二次刺激分子の添加の後に活性化T細胞の発生率の上昇があるかどうか判断するために、DC/乳癌融合および自己由来T細胞の共培養への、IL−12、IL−18およびTLRアゴニスト、イミダゾキノロン(TLR7/8)およびCPG−ODN(TLR9)の添加を調べた。DC/乳癌融合を、IL−12(10ηg/ml;R&D Systems)、IL−18(10ηg/ml)またはCPG ODN(10μg/ml、Coley Pharmaceutical Group、Ottawa、Canada)の存在下または非存在下で、自己由来のT細胞と5〜7日間共培養した。CpG ODN2395は、TスペーサーによってGCに富む回文配列5’−CGGCGCGCGCCG−3’(配列番号3)に連結された、六量体CpGモチーフ、5’−TCGTCGTTTT−3’(配列番号2)からなった。各実験で、刺激配列のない対照CpG ODNを同時に試験した。調節T細胞および活性化T細胞の集団を、上で概説される通りに定量した。
T細胞応答を活性化表現型に傾かせ、調節T細胞の影響を制限しようとして、ワクチン応答に及ぼすTLR9アゴニスト、CPG ODNの影響、先天性免疫応答の要素を活性化し、ワクチン効力を増強することが示されたTLRアゴニストを試験した。具体的には、CD4/CD25+細胞でIL−10およびFoxp3と比較したIFNγの発現を定量化することによって、活性化T細胞および阻害性T細胞の集団の融合媒介刺激を調節するCPG ODNの能力を調べた。さらに、DC/乳癌融合と共培養したT細胞の表現型プロフィールに及ぼす刺激性サイトカインIL−12およびIL−18の添加の影響も評価した。CpG ODNおよびIL−18の存在下で、融合刺激CD4+CD25+T細胞においてそれぞれ2.5倍の増加が見られた(p=0.0004およびp=0.006)。対照的に、IL−12をT細胞およびDC/乳癌融合の共培養に加えた場合、CD4/CD25+細胞の有意な増加は見られなかった。(図11Aを参照)。
ワクチン応答を免疫活性化の方に傾かせる別の戦略として、DC/乳癌融合ワクチンに対する応答に及ぼす、CD3およびCD28の抗体媒介連結の影響を調べた。抗CD3/CD28は、周囲の免疫環境の性質によって活性化T細胞または阻害性T細胞の増殖をもたらす、抗原非依存性刺激を提供する。したがって、DC/乳癌融合とそれに続く抗CD3/CD28による逐次的な刺激が、融合ワクチンによって最初に活性化されたT細胞の応答を増幅するであろうと仮定された。
樹状細胞(DC)/乳癌融合によるワクチン接種の安全性、免疫応答および臨床効果を検査するために、転移性乳癌を有する患者に、融合をIL−12と併用投与する。DC/乳癌融合細胞は、DC媒介同時刺激との関連で、広範囲の腫瘍関連抗原を提示する。融合細胞は、自己由来の腫瘍細胞を溶解する能力を有する、腫瘍特異的免疫を刺激する。臨床試験では、融合細胞によるワクチン接種は、良好な耐容性を示し、患者の大多数で免疫応答を誘導し、患者のサブセットで疾患退行をもたらす。IL−12と併用したワクチンの投与は、T細胞活性化を促進することによってさらにワクチン応答を高めると仮定された。
抗腫瘍免疫を刺激し、調節T細胞の増殖を制限するDC/乳癌融合の能力を高めるために、いくつかの戦略を調べた。DC/腫瘍融合による抗原特異的刺激およびT細胞同時刺激複合体(CD3/CD28)の非特異的連結の組合せが、腫瘍特異性リンパ球の活性化をもたらすであろうと仮定された。DC/乳癌融合および抗CD3/CD28による複合刺激は、活性化表現型が優先する腫瘍反応性T細胞の増殖をもたらすことが実証された。
多発性骨髄腫(MM)の患者に由来する自己由来の融合およびT細胞を用いる実験で、類似した知見が観察された。DCを接着性単核細胞から生成し、本明細書に記載の方法を用いて自己由来の骨髄腫細胞と融合させた。自己由来のT細胞は、T細胞分離カラムを用いて単離した。多発性骨髄腫を有する患者に由来するT細胞を、1:10の融合対T細胞の比率で融合細胞と7日間共培養するか、融合細胞と5日間共培養した後、抗CD3/CD28でコーティングしたプレートと48時間共培養した。刺激の後、一晩の適用の後のトリチウムチミジンの取込みによって、T細胞増殖を測定した。DC/骨髄腫融合とそれに続く抗CD3/CD28による逐次的な刺激は、融合細胞単独によって刺激したT細胞と比較して、T細胞増殖のレベルを著しく増加させた(図13)。
本発明を、その好ましい実施形態を参照して詳細に示し、記載したが、添付の請求項によって規定される本発明の精神および範囲から逸脱することなく、形態および詳細の様々な変更をそこに加えることができることは、当業者によって理解されよう。当業者は、常用の実験しか用いずに、本明細書で具体的に記載した本発明の具体的な実施形態の多くの同等物を認識するか、確認することができよう。そのような同等物は、特許請求の範囲に包含されるものとする。
Claims (61)
- 実質的に純粋な、教育され、増殖した抗原特異的な免疫エフェクター細胞集団を生成する方法であって、前記免疫エフェクター細胞はTリンパ球であり、前記集団はCD4+免疫エフェクター細胞および細胞傷害性CD8+免疫エフェクター細胞を含み、
a)複数のハイブリッド細胞を提供する工程であり、前記ハイブリッド細胞のそれぞれは少なくとも1つの樹状細胞と、細胞表面抗原を発現する少なくとも1つの腫瘍細胞または癌細胞との間の融合によって生成され、前記樹状細胞および前記腫瘍細胞または癌細胞は同じ種に由来し、前記樹状細胞は抗原をプロセシングおよび提示することができ、前記ハイブリッド細胞の少なくとも半分は免疫系を刺激するのに有効な量で(a)MHCクラスII分子、(b)B7、および(c)前記細胞表面抗原を発現する、工程と、
b)免疫エフェクター細胞集団を前記複数のハイブリッド細胞と接触させ、それによって、教育され、抗原特異的な免疫エフェクター細胞集団を生成する工程と、
c)前記教育され、抗原特異的な免疫エフェクター細胞集団を抗CD3/CD28抗体と接触させ、それによって前記実質的に純粋な、増殖し、教育され、抗原特異的な免疫エフェクター細胞集団を生成する工程であり、前記接触は、前記ハイブリッド細胞単独または前記抗CD3/CD28抗体単独への曝露と比較して、T細胞増殖、T細胞活性または腫瘍反応性T細胞の増加をもたらす、工程とを含む、方法。 - 前記集団を、増殖の後に調節T細胞の活性を除去または低減する化合物と接触させる工程をさらに含む、請求項1に記載の方法。
- 前記化合物がサイトカインである、請求項2に記載の方法。
- 選択方法の使用、またはsiRNAを用いる重要な遺伝子のサイレンシングによって調節T細胞の活性を除去または低減する工程をさらに含む、請求項1に記載の方法。

- 増殖し、教育され、抗原特異的な免疫エフェクター細胞を含む実質的に純粋な集団であって、前記集団は、教育され、抗原特異的な免疫エフェクター細胞を含み、前記免疫エフェクター細胞はハイブリッド細胞によって教育され、前記ハイブリッド細胞は、1つまたは複数の抗原を発現する腫瘍もしくは癌細胞に融合している樹状細胞を含み、前記樹状細胞および前記腫瘍または癌細胞は同じ種に由来し、前記樹状細胞は抗原をプロセシングおよび提示することができ、前記融合細胞の少なくとも半分は、免疫系を刺激するのに有効な量で(a)MHCクラスII分子、(b)B7、および(c)前記細胞表面抗原を発現し、前記教育された免疫エフェクター細胞は抗CD3/CD28抗体の存在下の培養で増殖し、抗CD3/CD28抗体の存在下の培養での増殖の後に、前記集団におけるT細胞の増殖が、前記ハイブリッド細胞単独に曝露させた免疫エフェクター細胞と比較して少なくとも2倍増加しているか、前記集団におけるT細胞の活性化が、前記ハイブリッド細胞単独に曝露させた免疫エフェクター細胞と比較して少なくとも2倍増加しているか、前記集団における腫瘍反応性T細胞が、前記ハイブリッド細胞単独に曝露させた免疫エフェクター細胞と比較して少なくとも2倍増加しているか、またはそれらの任意の組合せである集団。
- 前記樹状細胞および前記腫瘍または癌細胞が同じ個体から得られる、請求項30に記載の集団。

- 前記腫瘍または癌細胞が腎癌細胞である場合、抗CD3/CD28抗体の存在下の培養での増殖の後に、前記集団におけるT細胞の活性化が、前記ハイブリッド細胞単独に曝露させた免疫エフェクター細胞と比較して少なくとも約2倍増加している、請求項30に記載の集団。
- 前記腫瘍または癌細胞が腎癌細胞である場合、抗CD3/CD28抗体の存在下の培養での増殖の後に、前記集団におけるIFNγおよびグランザイムBを発現する細胞の存在が、前記ハイブリッド細胞単独に曝露させた免疫エフェクター細胞と比較して増加している、請求項30に記載の集団。
- 抗CD3/CD28抗体の存在下の培養での増殖の後に、前記集団におけるIFNγを発現する細胞の存在が、前記ハイブリッド細胞単独に曝露させた免疫エフェクター細胞と比較して少なくとも約2倍増加している、請求項38に記載の集団。
