本件出願は、2007年8月30日に英国で出願された出願番号0716900.6に基づく優先権を主張し;その開示は、全体として、参照として本明細書中に援用される。
本発明は、様々な材料、特に感受性材料、および特に、親和性リガンド(タンパク質性リガンド等)を結合させたクロマトグラフィー媒体、の滅菌方法に関する。
一態様において、クロマトグラフィー分離媒体は、30〜35バールの範囲の圧力下、より好ましくは34〜35バールの範囲の圧力下、加圧蒸気に対して曝露される。
特定の態様において、タンパク質性リガンドを結合させた固体支持体マトリクスを含むクロマトグラフィー分離媒体を、121℃の温度および34バールで、12分間、媒体を加圧蒸気に対して曝露することにより、滅菌する。
さらに別の態様において、固体支持体は、表面、チップ、キャピラリー、またはフィルターなどの別の形態である。
この滅菌方法は、幅広く適用が可能であり、そして抗体(特にモノクローナル抗体)、核酸(例えば、ゲノムDNA、RNA)を含む生物活性分子の単離等のいずれかの目的のため、そして生物学的サンプルから細胞を単離および分離するために意図された、クロマトグラフィー媒体を滅菌するために使用することができる。
本発明は、以下の実施例および図面を参照することによりさらに説明される。
MABSELECT媒体およびMABSELECT SURE媒体を、34.4バールの圧力および121℃の温度で、12分間、湿式加熱滅菌(オートクレーブ)に供した。この目的のため、33 mlのゲルを、Accelerated Solvent Extractor(ASE 200)(Dionex, USA)のカラム中に、0.1M NaCl溶液を用いて充填し、そして121℃および34.4バールの条件に、12分間曝露した。ASE 200は、プログラム可能な圧力および34.4バールでの最低圧力での加熱を提供する高圧装置である。
Claims (13)
- 約121℃〜約135℃の温度でクロマトグラフィー分離媒体を加圧蒸気に対して曝露することを含む、固体支持体マトリクスに対して結合させたタンパク質性リガンドを含むクロマトグラフィー分離媒体の滅菌方法。
- クロマトグラフィー分離媒体が、2バール〜35バールの範囲の圧力下、加圧蒸気に対して曝露される、請求項1に記載の方法。
- クロマトグラフィー分離媒体が、30〜35バールの範囲の圧力下、加圧蒸気に対して曝露される、請求項1に記載の方法。
- クロマトグラフィー分離媒体が、34〜35バールの範囲の圧力下、加圧蒸気に対して曝露される、請求項1に記載の方法。
- クロマトグラフィー分離媒体が、10〜60分の範囲の期間、加圧蒸気に対して曝露される、請求項1に記載の方法。
- クロマトグラフィー媒体が、34.4バールの圧力および約121℃の温度の下、蒸気に対して曝露される、請求項1〜3のいずれか1項に記載の方法。
- 滅菌すべきクロマトグラフィー分離媒体が、クロマトグラフィーカラム中に含有されるか、またはフィルター上に支持化される、請求項1〜6のいずれか1項に記載の方法。
- タンパク質性リガンドのアミノ酸配列が、親タンパク質リガンドと比較した場合、少なくとも1つのアスパラギン残基をグルタミン以外のアミノ酸により置換することにより変異させたものである、請求項1に記載の方法。
- タンパク質性リガンドが、Fc-フラグメント-結合タンパク質である、請求項1に記載の方法。
- 滅菌すべきクロマトグラフィー分離媒体が、支持体マトリクスに結合させたアルカリ-安定化プロテインA-誘導性リガンドである、請求項1〜7のいずれか1項に記載の方法。
- クロマトグラフィー分離媒体が、MABSELECT SUREである、請求項10に記載の方法。
- 請求項1〜6のいずれか1項に記載の方法により滅菌された、クロマトグラフィー分離媒体。
- 請求項1〜6のいずれか1項に記載の方法により滅菌された、MABSELECT SURE。
