Abstract
Description
SP3D Solanum esculentum遺伝子(アクセッション番号AY186735)は、Solanum pennelii la716の遺伝子をクローニングするために使用されてきた。S.pennelliiのBACライブラリーは、プライマーSP3D−f2/SP3D−rを用いて、すなわち92℃で30秒、60℃で30秒および72℃で60秒での40サイクルによりスクリーニングされ、SP3Dpenを有するBAC52、1c06ellが得られた(図1)。
ナス種の決定的重要点の1つは、仮軸指数である。果房間に2枚の葉を備える(spi=2)種は、Solanum neorickii、Solanum chmielewskii、Solanum chilense、Solanum peruvianum、およびSolanum pennelliiである。果房間に3枚の葉を備える(spi=3)種は、S.habrochaites、S.cheesmaniae、S.pimpinellifolium、およびS.lycopersicumである。
S.pennellii la716由来のSP3Dpenを有する遺伝子移入系49015−2は、Enza Zaden社のS.lycopersicum系のOT1464およびOT1690と異種交雑させられている。結果として生じたF1雑種は、OT1464と戻し交雑されているか、OT1690交雑の場合には自家交雑されている。SP3Dpenを備える植物は、dCAPSマーカーであるdSP3D−1fr、プライマーのdSP3D−f/dSP3D−rを用いて、92℃で30秒、55℃で60秒および72℃で60秒での40サイクルのPCR条件で、HpyCH4Vによる消化、3%ms−8アガロース(Hispanagar社)での分離を用いて選択された。dSP3D−1frマーカーの他に、当業者には公知であるように、最高の反復親を同定するためにマーカーアシスト戻し交雑が使用され、OT1464バックグラウンドについては植物NT05−96c11が得られ、OT1690バックグラウンドについてはNT05−108h10およびNT05−108e12が得られた。これらの個々の植物を2回自家交雑させ、dSP3Dマーカーを用いて、およびマーカーアシスト戻し交雑によって再び選択し、OT1464バックグラウンドについては植物111B6が得られ、OT1690バックグラウンドについては植物117F1および117G1が得られた。その後、111B6×111F1を交雑させると15751(ホモ接合型SP3Dpen)が得られ、111B6×117G1を交雑させると15753(ヘテロ接合型)が得られ、OT1690×OT1464を交雑させるとSP3Descホモ接合型コントロールとしての15769がF1雑種として得られた。図6を参照されたい。
Claims (28)
- SP3Dプロモーター配列であって、前記プロモーター配列に作動可能に連結している下流SP3D遺伝子の転写を指示することができ、仮軸指数3以上を有する植物における仮軸指数を低下させるために、仮軸指数2を有するナス科の1種に由来するSP3Dプロモーター配列。
- 前記プロモーター配列が、前記SP3D遺伝子の開始コドンの62−61ヌクレオチド上流の位置にCAモチーフを含む、請求項1に記載のプロモーター配列。
- 前記プロモーター配列が、Solanum pennellii、Solanum neorickii、Solanum chmielewskii、Solanum chilense、およびSolanum peruvianumからなる群から選択されるナス科の1種に由来する、請求項1または2に記載のプロモーター配列。
- 前記プロモーター配列が、配列番号1(図2)のヌクレオチド1〜624と少なくとも75%同一性を有するヌクレオチド配列を含む、請求項1、2または3に記載のプロモーター配列。
- 配列番号1のヌクレオチド1〜624と少なくとも85%同一性を有するヌクレオチド配列を含む、請求項4に記載のプロモーター配列。
- 配列番号1のヌクレオチド1〜624と少なくとも90%同一性を有するヌクレオチド配列を含む、請求項5に記載のプロモーター配列。
- 配列番号1のヌクレオチド1〜624と少なくとも95%同一性を有するヌクレオチド配列を含む、請求項6に記載のプロモーター配列。
- 配列番号1のヌクレオチド1〜624と少なくとも99%同一性を有するヌクレオチド配列を含む、請求項7に記載のプロモーター配列。
- 前記プロモーター配列が、S.Pennelliiに由来し、配列番号1のヌクレオチド1〜624のヌクレオチド配列からなる、請求項1〜8のいずれかに記載のプロモーター配列。
- 請求項1〜9のいずれかに記載のプロモーター配列および植物SP3D遺伝子由来のcDNA配列を含む遺伝子構築物。
- 前記cDNA配列が、図3に示される前記SP3DpenのcDNA配列と少なくとも75%同一性を有するヌクレオチド配列を含む、請求項10に記載の遺伝子構築物。
- 前記cDNA配列が、図3に示される前記SP3DpenのcDNA配列と少なくとも85%同一性を有するヌクレオチド配列を含む、請求項11に記載の遺伝子構築物。
- 前記cDNA配列が、図3に示される前記SP3DpenのcDNA配列と少なくとも90%同一性を有するヌクレオチド配列を含む、請求項12に記載の遺伝子構築物。
- 前記cDNA配列が、図3に示される前記SP3DpenのcDNA配列と少なくとも95%同一性を有するヌクレオチド配列を含む、請求項13に記載の遺伝子構築物。
- 前記cDNA配列が、図3に示される前記SP3DpenのcDNA配列と少なくとも99%同一性を有するヌクレオチド配列を含む、請求項14に記載の遺伝子構築物。
- 前記cDNA配列が、図3に示される前記ヌクレオチド配列からなる、請求項10〜15のいずれかに記載の遺伝子構築物。
- 減少した仮軸指数を有するナス科の植物を提供するための方法であって、前記植物のゲノムに、下流SP3D遺伝子と作動可能に連結している請求項1〜8のいずれかに記載のプロモーター配列を導入する工程、または前記植物に請求項9〜15のいずれかに記載の遺伝子構築物を導入する工程を含み、前記プロモーター配列もしくは前記遺伝子構築物を備える前記植物の前記仮軸指数が、前記プロモーター配列もしくは前記遺伝子構築物を含まない植物と比較して減少する方法。
- 前記仮軸指数が、仮軸指数2に減少する、請求項17に記載の方法。
- 仮軸指数3以上を有するナス科の植物の収穫量を増加させるための方法であって、前記植物のゲノムに、下流SP3D遺伝子と作動可能に連結している上述したプロモーター配列を導入する工程、または前記植物に請求項9〜15のいずれかに記載の遺伝子構築物を導入する工程を含む方法。
- 前記植物が、S.habrochaites、S.cheesmaniae、S.pimpinellifoliumおよびS.lycopersicum、ならびに/またはそれらに由来する植物からなる群から選択される、請求項17、18または19に記載の方法。
- 前記植物が、S.lycopersicumである、請求項21に記載の方法。
- 請求項17〜21のいずれかに記載の方法によって得られる植物。
- 請求項22に記載の植物に由来する種子および/または他の植物部分。
