タンパク質ワクチン:ポリペプチド(又はそれらの少なくとも1つ免疫原性部分)、ペプチド混合物又は融合ポリペプチドを含むワクチン組成物;
組み替え生ワクチン:非病原性微生物又はウイルス中におけるワクチンに関連ある抗原の発現。そのような微生物の周知の例は、ウシ型結核菌BCG、サルモネラ及びシュードモナス(Pseudomonas)であり、ウイルスの例は、ワクシニアウイルス及びアデノウイルスである。
ウシ型結核菌BCGからの無極性及び極性の脂質の抽出
ウシ型結核菌BCG(コペンハーゲン株)をSauton変法培地中で培養した。マイコバクテリアを2〜3週間後に収集してPBS中に懸濁させ、60℃で1.5時間インキュベートすることにより殺菌した。無極性及び極性の脂質を、標準的手順(Dobson et al,1985)(Rosenkrands et al.2005)に従って抽出した。
PDIM及びTAGを石油エーテル/アセトン(98:2)におけるTLC処理を使用して単離した。フチオセロールAに基づくPDIMの主成分のみ回収された。PGL及びMMGは、トルエン/アセトン(95:5)におけるTLC処理により一緒に分離された。PGLとMMGとは、クロロホルム:メタノール:0.880アンモニア(97:3:0.5)で1次元TLCにより分離された。PDIM、TAG、PGL及びMMGを、500MHz 1H及び13C核磁気共鳴(NMR)(Bruker drx500)及びMALDI−TOF質量分析(MS)(Bruker Biflex IV)にかけた。再水和した脂質抽出物(1mg/ml)の試料(10μl)を、SDS−PAGE(Laemmli et al,1970)及び銀染色(Blum et al,1987)により分析し、残留タンパク質含有量を求めた。脂質調製物は、エンドドキシン汚染のないことが認められた(<0.001ngLPS/μg脂質)。
モノミコリルグリセロールを16×100mmの試験管中で5%TBAH水溶液(2.5ml)とともに100℃で終夜加熱した(Minnikin et al,1988)。冷却後、混合物を水(2ml)で希釈し、ジクロロメタン中ヨードメタンの10%溶液(3ml)を加えて、試験管を回転装置上に1時間置いた。上の水層を廃棄して、下層を等量の1MのHCl及び水で洗浄し、蒸発乾固すると残渣が生じて、TLC(石油エーテル:アセトン95:5)により、α−及びケトミコレートメチルエステルを含有することが示された。分取TLC(上記の)により、αメチルミコレート及びケトメチルミコレートが得られた。
3−ヒドロキシ−2−テトラデシル−オクタデカン酸(Datta et al,1991に従って合成された合成C32コリノミコール酸)(100mg、0.20mmol、1eq)及び4−ピロリジノピリジン(100mg、3eq)を50mlの丸底フラスコ中に入れて、モレキュラーシーブ4Aと一緒にジクロロメタン(500μl)中50μlの2,2−ジメチル−4−ヒドロキシメチル−1,3−ジオキソラン(sn−イソプロピリデングリセロール)溶液を加えた。その混合物を、高真空下に室温で完全に乾固させ、N’,N−ジシクロヘキシルカルボジイミダゾール(DCC)(15ml、DCM中0.1MのDCC、5eq)を加えて、反応混合物を室温で終夜攪拌するままにした。モレキュラーシーブを濾過により除去し、反応混合物を真空で濃縮して乾固し、残渣を、フラッシュカラムクロマトグラフィ(Fluka 60741シリカゲル60)を使用して、ヘキサンからヘキサン:酢酸エチル(8:2)を使用して5%増分で溶出させて精製し、純粋なイソプロピリデン保護化合物(3−ヒドロキシ−2−テトラデシル−オクタデカン酸−2,2−ジメチル−[1,3]−ジオキソラン−4−イルメチルエステル)を収率56%で得た(68mg)。1H NMR(CDCl3、300MHz)δH0.90(t、6H、CH3)、1.20(s、54H、CH2)、1.40(s、3H、CH3)、1.45(s、3H、CH3)、2.50(m、1H、CH)、4.05〜4.40(m、5H、CH2、CH);13C NMR(CDCl3、75MHz)δC15.0(CH3)、22.1、28.8、28.9、29.0、31.4(CH2)、29.1(CH3)、52.1(CH(CH2)13CH3)、63.2(CH2OCO)、69.3(CH2O)、73.4(CH(CH2)14CH3)、174.3(C=O);m/z(EI)633.55[M+Na+](100%);HRMS C38H74O5Na[M+Na+]に対する計算値633.5536、実測値633.5527。
ヒトPBMC由来のDCを、Romani et al.,1994から改良した方法に従って得た。末梢血はバフィーコートから得た。簡単に述べると、単球は、Ficoll−Hypaque遠心分離により単離し(Lymphoprep 1077密度媒質、Nycomed、オスロー、ノルウエイ)、続いて抗CD14標識磁性ビーズ(MACS;Miltenyi Biotech、Bergesh Gladbach、ドイツ)を使用してCD14陽性細胞を分離した。単球は、10%FCS、50μMの2−メルカプトエタノール、100U/mlのペニシリン、100μg/mlのストレプトマイシン、2mMのL−グルタミン(全てGibco)(CM)で補完した完全RPMI 1640中で、100ng/mlのヒト組み替えGM−CSF(Prepotech、Rocky Hill、NJ、USA)及び50ng/mlのヒト組み替えIL−4(Becton Dickinson(BD))を存在させて、37℃、5%CO2で7日間培養した。
DCを先ず関連するmAb(BD Pharmingen)と(30分、4℃)、続いて1/20希釈FITC結合ヤギ抗マウスIg(Jackson ImmunoResearch Laboratories、West Grove、PA)と(30分、4℃)インキュベートすることにより、表面マーカーを染色した。非特異的Abの結合は、関連する1次ヒトmAbの添加前に、10%ウシ胎児血清溶液で阻止した(15分、4℃)。染色された細胞は、直ちにFACScanフローサイトメトーター(BD)を使用して、フローサイトメトリーにより検査し、CellQuestソフトウェアで分析した。
DC培養上清を収集して、−20℃で貯蔵した。分泌されたIL−12p70、IL−6及びTNF−αを、ELISA(BD)によりメーカーの説明書に従って測定した。
混合リンパ球反応(MLR)アッセイのためのiDCは、上で概説したようにして、単球から発生させた。生成した細胞(iDC)は、同じ培地中で、又は脂質(10μg又は100μg/ml)を含有する培地中で24時間培養した。0.125×105から2×105まで漸増させたDCを、37℃/5%CO2でPPD陰性の提供者からの同種異系のT細胞(105細胞/ウェル)と平底96ウェルのマイクロタイタープレート中でインキュベートした。T細胞は、Pan−T細胞単離キット(Miltenyi)を使用し、メーカーの説明書に従って単離した。DCと同種異系T細胞との共培養は、6日間インキュベートした。上清を収集して、分泌されたIFN−γがELISA(BD)によりメーカーの説明書に従って測定されるまで、−20℃で貯蔵した。次に、両方のアッセイには、18時間の最終培養の間、1μCi/ウェルの[3H]チミジンを含む培地をパルスで適用した。細胞を収集して、T細胞の増殖を液体シンチレーション計数(Microbeta Systems)により測定した。全てのアッセイは、少なくとも3名の異なる提供者を使用して3重で実施した。
Ag85BとESAT−6との融合タンパク質(以下、Ag85B−ESAT−6と称する)は、前に記載したように(Olsen et al,2001)組み替えタンパク質として産生された。
8ないし12週齢の雌BALB/c又はC57BL/6マウスを、Bomholtgaard(Ry、デンマーク)又はHarlan Scandinavia(デンマーク)から入手した。感染させたマウスは、BL−3層流安全密閉箱内のケージ中で飼育した。
マウスは、尾部の底部で皮下に(s.c.)、各免疫化間2週間の間隔で最高3回免疫した。ワクチン(0.2ml/マウス)は、特に断らない限り、250μgのDDAと10μgの再水和脂質抽出物との中で乳化された2μgの融合タンパク質Ag85B−ESAT−6からなるものであった。幾つかの場合には、11mol%のTDBがDDAリポソーム中に組み込まれた(Davidsen et al、PCT/DK2005/000467)。結核菌感染を含む実験における陽性対照として、1群のマウスが、尾部の底部皮下に、1用量のBCGデンマーク株1331の注射を受けた。全ての又は個々の脂質抽出物は、乾燥ウシ型結核菌脂質材料を1mg/ml又は5mg/mlのMilli Q水で再水和し、続いてプローブ超音波処理することにより調製した。標準的脂質ワクチンは、抗原を食塩水と混合し、続いて再水和した脂質抽出物及びDDAを添加して、ボルテックスで混合することにより調製した。ワクチンを終夜放置して、抗原を吸着させた。
血液試料又は鼠径部リンパ節を、最終免疫の7〜150日後のマウスから採取し、前に記載したようにして(Rosenkrands et al,2005)調製した。細胞培養は、2−メルカプトエタノール、グルタミン、ペニシリン−ストレプトマイシン、Hepes、及び10%ウシ胎児血清で補完された、体積200μlのRPMI中に2×105の細胞を入れた丸底マイクロタイターウェル中で、3重で実施した。再刺激のための抗原は、5μg/mlの濃度で使用した。培地のみ又は5μg/mlのConAを入れたウェルが、全ての実験でそれぞれ陰性及び陽性対照として含まれた。培養上清は、並行する培養液から、抗原を存在させて72時間インキュベートした後収集して、IFN−γの量を酵素結合免疫吸着測定法(Brandt et al,2000)により測定した。
ワクチンの有効性を評価するために、マウスに、最初の免疫の2.5ヵ月後に、約25CFUの有毒結核菌Erdman株を投入するように較正されたGlas−Colの吸入曝露系で、エアロゾル経路により肺にチャレンジした。脾臓及び肺における細菌負荷は、BCGの増殖を選択的に阻害するために1ml当たり2μlの2−チオフェン−カルボン酸ヒドラジドで補完したMiddlebrook 7H11アガー上に、段階希釈液を蒔くことにより、6週間後に測定した。37℃で2〜3週間インキュベーション後に、コロニーを計数した。
感染マウスと対照マウスとの間のコロニー数の差を、変動分析により検定した。有意の効果が示されたときは、平均間の差をダネット検定により評価した。ヒトDCアッセイについては、異なる脂質に対する応答におけるサイトカイン放出の差を、変動分析により検定し、有意の効果が示されたときは、平均間の差をチューキー検定により評価した。
ウシ型結核菌BCGの全脂質を、極性及び無極性分画に分離した。極性分画中で同定することができた脂質は、ホスファチジルイノシトールマンノシド(1〜4)、ホスファチジルイノシトール(PI)、ホスファチジルエタノールアミン(PE)、ジホスファチジルジグリセロール(DPG)及びL−α−ホスファチジル−DL−グリセロール(PG)であった。多数の未知のリン脂質も同定された(7及び8)(図1A)。無極性分画中で同定された主な脂質は、フチオセロールジミコセロセート(PDIM)、トリアシルグリセロール(TAG)、フェノール性糖脂質(PGL)及びモノミコリルグリセロール(MMG)(図1)であった。FFAは遊離脂肪酸である。
免疫刺激性の無極性分画の脂質は、分取TLCを使用して単離され、フチオセロールジミコセロセートA(PDIMA)、TAG、PGL及びMMGの純粋な試料を与えた。少量のフチオセロールB及びフチオジオロンAも検出されたが、利用した分取方法では回収されなかった(図2A)。脂質の構造及び同一性は、1H核磁気共鳴(NMR)及び質量分析法(MS)により確認された(図2B及び不掲載データ)。MMG成分は、1−モノアシルグリセロールに特徴的な1H及び13C NMRスペクトルを示した(Gunstone et al,1991)。全MMG分画のNMRデータと結びつけたMALDI−TOFMS(表1)は、α−ミコレート並びにcis及びtrans型のケト−ミコレートの存在を明らかにした。主成分のおよその比は、それぞれ1.00:0.29:0.24であった。
精製されたMMG、PDIMA、PGL及びTAGを、ヒトiDCを活性化するそれらの能力について評価した。これらのアッセイにおいて、MMGは、CD86、CD40及びHLA−DRの顕著な上方制御に通ずる、DC活性化の最も強力な誘発因子であることが一貫して認められた(図3A)。MMGは、マイコバクテリアの病原性と長らく関連づけられてきた脂質であるPDIMAよりさらに大きくDCを活性化し(Cox et al,1999)、PDIMAは2番目に活性な脂質となったが、一方、PGL及びTAGが誘発する活性化はそれより小さかった。6名の個々の提供者における活性化の順は、MMG>PDIMA>PGL>TAGと認められ、CD86レベルにおいて、未処理iDCに関するレベルを超える平均の上昇倍率は、それぞれ1.91±0.29、1.82±0.43、1.52±0.26及び1.32±0.14であった。サイトカイン誘発も全体的に同じ傾向をたどり(図3B)、MMGは最も強力な免疫刺激剤脂質として卓越している。IL−6は、MMGに曝されたDCにより、PGL又はTAGにより誘発されるレベルを有意に超えるレベルで(P<0.05)放出された。他の脂質によるサイトカイン誘発の間で、有意の差は記録されなかった。したがって、MMGは、ウシ型結核菌BCGの無極性脂質分画中で最も強力な免疫刺激性脂質として分類することができる。
この実施例において、本発明者らは、MMGの免疫刺激特性を精査し、その強力な免疫刺激性能の原因となる有効成分を同定することをさらに求めた。α−MMGとケト−MMGとを、MMGのトリメチルシリルエーテルの調製、分取TLC処理及びそれに続くトリメチルシリルエーテル保護基の加水分解によって分離し、α−MMG及びケト−MMGを得た。α及びケト−ミコレートの構造を図2Bに示す。ヒトiDCを活性化するそれらの能力を評価したとき、α−及びケト−MMGは、活性化マーカーのレベルにおいて2〜3倍の程度で刺激した(図4)。したがって、MMGの2つの下位成分も、ヒトDCを刺激する顕著な能力を示す。
MMGのアジュバント活性を研究するために、単離された脂質を、マウスにおいてIFN−γ産生を誘発するそれらの能力について試験した。C57BL/6マウスに、10μgの全脂質又は個々の脂質を投与した。In vivoにおいて、DDAは、脂質を入れて送達するビヒクルとして役立つ。それゆえ、250μgのDDA中で乳化された2μgの融合タンパク質Ag85B−ESAT−6及び10μgの再水和脂質抽出物を、皮下経路により投与した。DDAリポソーム中に組み込まれた10μgのMMGの用量は、排出するリンパ節から単離されたPBMCの再刺激で、10ng/mlのIFN−γレベルを生じた。このレベルは、DDA中の全脂質に対して記録されたレベルと、等用量で同等である(図5)。PDIM−Aも、より低いレベルとはいえ、IFN−γ産生を誘発したが、TAG又はPGLの組み込まれたDDAリポソームは、IFN−γ放出を殆ど助長しなかった(図5及び不記載データ)。これらの個々の脂質は、全脂質抽出物又は個々の脂質のいずれで再刺激しても記憶応答は観察されなかったので、アジュバントとして作用しているように思われることは注目すべきである(不記載データ)。したがって、MMGは、in vivoにおいて最も活性な無極性脂質でもあるとして確認されて、BCG由来の全脂質のアジュバント活性の大部分を単独で説明することができた。
MMGに基づくアジュバントのTB感染に対する防御提供能力を評価するために、C57BL/6マウスを、MMG(2つの異なる用量)及びDDAに送達されたAg85B−ESAT−6により免疫した。BCGワクチン接種及びアジュバント単独を受けるマウスの群は、それぞれ陽性及び陰性対照として含まれた。最後のワクチン接種の6週間後に、マウスに、生結核菌をエアロゾル経路によりチャレンジした。ワクチンが細菌負荷を減少させる能力を、その6週間後に肺及び脾臓で測定した。これらのデータは、MMG/DDAをアジュバントとして有意の防御レベル、及びBCGのレベルと同等の防御レベルを示した(図6)。予想されたように、抗原なしでアジュバントを接種されたマウスは、細菌の増殖を阻止することができなかったので、この効果は特異的である。
免疫調節剤(MMG)と送達系(DDA)とを組み合わせる効果を評価するために、DDA単独若しくはDDA/MMGの組合せで(実験1、図7A)、又は実験2(図7B)においてはMMG単独若しくはDDA/MMGの組合せで、C57BL/6マウスにワクチンを接種した。これらの実験から、免疫応答は、DDAとMMGとを組み合わせることにより、劇的に増強されることが明らかである。
他の免疫刺激性成分と組み合わせたときの、MMGのアジュバント活性に対する効果を研究するために、C57BL/6マウスにおける免疫応答を、DDAリポソーム中に組み込まれたAg85B−ESAT−6及び10μgのMMG、又は免疫調節剤TDBの組み込まれたDDAリポソーム(Davidsen et al,PCT/DK2005/000467)を使用して皮下で免疫した後、最初のワクチン接種の5ヵ月後に評価した。DDAリポソーム中に組み込まれたMMGという組合せは、血液から単離されたPBMCの再刺激で、約25ng/mlのIFN−γレベルを生じたが、IFN−γの放出は、TDBの組み込まれたDDAリポソームが使用されたとき、劇的に上昇した(図8)。したがって、MMG、DDA及びTDBの間で相乗的効果が観察され、MMGとDDAとの組合せに対する第3成分の添加は、アジュバント活性をさらに増強させ得ることを示した。
合成MMG類似体の免疫学的効果を評価するために、C57BL/6マウスを、天然MMG、16個の炭素を有する合成MMG類似体(図9に示した)及び36個の炭素を有する合成MMG類似体(図9に示した)(全て10μg/DDA/MMG)を加えたDDA中のAg85B−ESAT−6で免疫した。免疫応答は、最後のワクチン接種の1週間後に血液で測定され、3種のMMGに基づくアジュバントで同等のレベルの応答を示したが、DDAは、それだけではより低い効果を再び示した。
鎖長がより短い合成MMG類似体の免疫学的効果を評価するために、単独のDDA中、天然MMGを加えたDDA中、C8ないしC36の範囲の異なる合成類似体を加えたDDA中のAg85B−ESAT−6で(全て1μg/用量)、C57BL/6マウスを免疫した。免疫応答は、最後のワクチン接種の3週間後に血液で測定され、合成MMG類似体は1μgの用量レベルでも活性であることを示した。さらに、これらの結果は、より短い合成MMG類似体(16C以下)は、天然MMGと比較してより有効であることも示す。***P<0.001(図11)

Claims (17)
- アジュバント又は免疫調節剤を調製するための、モノミコリルグリセロール(MMG)又はそれらの合成同族体、類似体及び改変体の使用。
- 脂質尾部を場合により有する、8〜36個の炭素を有するアルキル鎖に基づく合成MMG同族体、類似体又は改変体であって、最も好ましくは8〜16個の炭素を有するアルキル鎖を有し、各脂質尾部に0〜3個の二重結合を場合により有する、請求項1に記載の合成MMG同族体、類似体又は改変体の使用。
- 図9に示されたような、8ないし36個の炭素を有するアルキル鎖に基づく合成MMG同族体、類似体又は改変体の、請求項2に記載の使用。
- 請求項1〜3のいずれか一項に記載に従って調製されたアジュバント又は免疫調節剤。
- 界面活性剤をさらに含む、請求項4に記載のアジュバント又は免疫調節剤。
- 界面活性剤が、DDA−B、DDA−C、DDA−X、DODA−B、DODA−C、DODA−X、DOTAP、DODAP、DOTMA、DXPC、DXPE、DXPG又はそれらの組合せである、請求項5に記載のアジュバント又は免疫調節剤。
- 非TLRリガンド例えばTDB及びMDP、又はTLRリガンド例えばPoly I:C、並びに/又はこれらの任意の類似体である追加の免疫調節剤を含む、請求項4〜6のいずれか一項に記載の、アジュバント又は免疫調節剤。
- 各脂質尾部に0〜3個の二重結合を有する脂質尾部を場合により有する、8〜36個の炭素を有するアルキル鎖に基づく合成MMG同族体、類似体又は改変体。
- 図9に示したような、8〜16個の炭素又は最大で36個の炭素を有する、請求項8に記載の合成MMG類似体。
- 非経口、経口、粘膜投与、舌下、経皮、局所、吸入、鼻腔内、エアロゾル、眼内、気管内、直腸内、膣内のための、又は遺伝子銃、皮膚パッチによる、又は点眼剤若しくはうがい薬の形態における、請求項4〜7のいずれか一項に記載のアジュバント又は免疫調節剤を含むワクチン。
- 抗原性成分が細胞内病原体由来の抗原性エピトープを含む、請求項10に記載のワクチン。
- 抗原性成分が、有毒のマイコバクテリウム、例えば結核菌、ウシ型結核菌又はマイコバクテリウム・アフリカヌム(M.africanum)由来の抗原性エピトープを含む、請求項11に記載のワクチン。
- 抗原性成分が、融合生成物Ag85b_TB10.4、Ag85b_ESAT−6_Rv2660、Ag85b_TB10.4_Rv2660及びAg85a_TB10.4_Rv2660の中から選択される、請求項12に記載のワクチン。
- 抗原性成分が、抗原性エピトープ、即ち、熱帯熱マラリア原虫(Plasmodium falciparum)(例えばMsp1、Msp2、Msp3、Ama1、GLURP、LSA1、LSA3又はCSP)、トラコーマクラミジア(Chlamydia trachomatis)(例えばCT184、CT521、CT443、CT520、CT521、CT375、CT583、CT603、CT610又はCT681)、HIV、インフルエンザ又は肝炎B若しくはC由来の抗原性エピトープを含む、請求項11に記載のワクチン。
- 癌、自己免疫障害、神経障害例えばアルツハイマー病、気道炎症、炎症性障害、感染性疾患、皮膚障害、アレルギー、喘息又は病原体により惹起される疾患を治療するための、請求項10に記載のワクチン。
- 癌、自己免疫障害、神経疾患例えばアルツハイマー病、気道炎症、炎症性障害、感染性疾患、皮膚障害、アレルギー、喘息又は病原体により惹起される疾患を治療するための、請求項4〜7のいずれか一項に記載のアジュバント又は免疫調節剤を含む送達系。
- MMG又はそれらの合成同族体、類似体及び改変体が、1つまたは複数のワクチン、抗原、抗体、細胞傷害性因子、アレルゲン、抗生物質、アンチセンスオリゴヌクレオチド、TLR及び非TLRアゴニスト、TLR及び非TLRアンタゴニスト、ペプチド、タンパク質、遺伝子療法ベクター、DNAワクチン、免疫調節剤又は共刺激性分子との組合せで投与される、請求項14に記載の送達系。
