Abstract
Description
下記の実施例は、本発明の実施形態のいくつかに関してさらに詳述するものである。
本発明のヒアルロン酸は任意のタイプのタンパク質により被覆することができる。例えば、コラーゲンおよび/またはアルブミンを、ヒアルロン酸の粒子を被覆するために、または、ヒアルロン酸マトリックスを作製するために使用することができる。好ましくは、ヒアルロン酸を被覆するために使用されるタンパク質は、被覆されたヒアルロン酸の改善されたインビボ寿命を可能にしながら、一般に容易に生体吸収性であることがこの技術分野において知られているタンパク質でなければならない。
本発明のヒアルロン酸は任意のタイプの多糖により被覆することができる。例えば、デンプン、セルロースおよびその誘導体(アルキルセルロース、ヒドロキシアルキルセルロース、セルロースエーテル、セルロースエステル、ニトロセルロース、メチルセルロース、エチルセルロース、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、ヒドロキシブチルメチルセルロース、酢酸セルロース、プロピオン酸セルロース、酢酸酪酸セルロース、酢酸フタル酸セルロース、カルボキシエチルセルロースおよび三酢酸セルロースを含む)、および/または、アルギネートを、ヒアルロン酸粒子を被覆するために、または、ヒアルロン酸マトリックスを作製するために使用することができる。好ましくは、ヒアルロン酸を被覆するために使用される多糖は、被覆されたヒアルロン酸の改善されたインビボ寿命を可能にしながら、一般に容易に生体吸収性であることがこの技術分野において知られている多糖でなければならない。
本発明のヒアルロン酸は、任意のタイプの生体吸収性または生分解性ポリマーあるいはある種の非分解性ポリマーにより被覆することができる。例えば、ポリ(ヒドロキシ酸)(例えば、ポリ(乳酸)、ポリ(グリコール酸)およびポリ(乳酸-コ-グリコール酸))、ポリ(ラクチド)、ポリ(グリコリド)、ポリ(ラクチド-コ-グリコリド)、ポリ無水物、ポリオルトエステル、ポリアミド、ポリアルキレングリコール(例えば、ポリ(エチレングリコール))、ポリアルキレンオキシド(例えば、ポリ(エチレンオキシド))、ポリアルキレンテレフタレート(例えば、ポリ(エチレンテレフタレート))、ポリビニルアルコール、ポリビニルエーテル、ポリビニルエステル、ポリビニルハライド(例えば、ポリ(塩化ビニル))、ポリビニルピロリドン、ポリシロキサン、ポリ(ビニルアルコール)、ポリ(酢酸ビニル)、ポリウレタンおよびそれらのコポリマー、アクリル酸ポリマー、メタクリル酸ポリマーまたはそれらのコポリマーもしくは誘導体、例えばエステル、ポリ(メタクリル酸メチル)、ポリ(メタクリル酸エチル)、ポリ(メタクリル酸ブチル)、ポリ(メタクリル酸イソブチル)、ポリ(メタクリル酸ヘキシル)、ポリ(メタクリル酸イソデシル)、ポリ(メタクリル酸ラウリル)、ポリ(メタクリル酸フェニル)、ポリ(アクリル酸メチル)、ポリ(アクリル酸イソプロピル)、ポリ(アクリル酸イソブチル)およびポリ(アクリル酸オクタデシル)(これらはまとめて本書では「ポリアクリル酸」と称される)、ポリ(酪酸)、ポリ(吉草酸)、ならびに、ポリ(ラクチド-コ-カプロラクトン)、それらのコポリマーおよびブレンドをはじめとする様々なポリマーを、ヒアルロン酸の粒子を被覆するために、または、ヒアルロン酸マトリックスを作製するために使用することができる。そのようなポリマーを、交互積層、化学的コンジュゲート化、乳化、または、この技術分野において知られている様々な被覆方法のいずれかにより、ヒアルロン酸上に被覆することができる。被覆の厚さは、ヒアルロン酸の大きい架橋粒子が使用でき、かつ、注入のために好適な被覆粒子が得られるように、ほんの数ナノメートルの非常に薄い被覆を作製するために改変することができる。あるいは、被覆は、インビボでのヒアルロン酸の寿命を改善するために、より厚くすることができる。
Claims (15)
- 軟組織増強のための組成物であって、
被覆されたヒアルロン酸を形成するように被覆で被覆されたヒアルロン酸を含み、被覆は、生分解性ポリマー、非分解性ポリマー、タンパク質、多糖またはこれらの組合せを含み、
哺乳動物における皮下注入のために好適である組成物。 - ヒアルロン酸が、架橋されたヒアルロン酸の粒子である、請求項1に記載の組成物。
- 被覆が、ポリ乳酸-コ-グリコール酸、アルブミンまたはアルギネートである、請求項1に記載の組成物。
- 被覆されたヒアルロン酸が、通例球形である、ミクロスフェアの形状である、請求項1に記載の組成物。
- 持続型ミクロスフェアは、平均して、直径が約10μm〜約2000μmである、請求項4に記載の組成物。
- ヒアルロン酸粒子を含む組成物であって、
ヒアルロン酸粒子が、封入されたヒアルロン酸粒子を形成するように、ポリマー、タンパク質、多糖またはこれらの組合せに封入され、
封入されたヒアルロン酸粒子は通例球形である組成物。 - 哺乳動物における皮下注入のために好適である、請求項6に記載の組成物。
- ヒアルロン酸粒子が、水性環境におけるヒアルロン酸の持続放出を可能にするポリマー、タンパク質または多糖に封入される、請求項6に記載の組成物。
- 少なくとも1つの生体適合性ポリマーと架橋された封入ヒアルロン酸粒子のヒドロゲルを含む、請求項6に記載の組成物。
- 生体適合性ポリマーがポリビニルアルコールである、請求項9に記載の組成物。
- 被覆されたヒアルロン酸粒子を含む、皮膚増強のための皮膚フィラーであって、被覆されたヒアルロン酸粒子は、水性環境における該ヒアルロン酸の分解速度を低下させる被覆を含む皮膚フィラー。
- 被覆されたヒアルロン酸粒子は、通例球状であり、平均して、直径が約10μm〜約2000μmである、請求項11に記載の皮膚フィラー。
- 被覆されたヒアルロン酸粒子のヒアルロン酸が、架橋されたヒアルロン酸である、請求項12に記載の皮膚フィラー。
- 被覆が約10nm〜50000nmの厚さである、請求項12に記載の皮膚フィラー。
- 哺乳動物の軟組織を修復または増強するための方法であって、修復または増強する哺乳動物軟組織を選択するステップ、および、ポリマー、タンパク質または多糖で被覆されたヒアルロン酸粒子を含む注入可能な生体吸収性組成物を該哺乳動物軟組織の中に配置するステップを含む方法。
