実施例3.試験化合物の活性についての結果
[00215]細胞成長阻害アッセイ。ヒト腫瘍細胞株MCF−7(乳腺腺癌)、NCI−H460(肺大細胞)及びSF−268(中枢神経系膠芽腫)を国立癌研究所(National Cancer Institute)から得た。すべての細胞株を、100%湿度の5%CO2雰囲気中、37℃で10%ウシ胎仔血清及び2mMのL−グルタミンで補充したRPMI−1640培地中で培養した。各ビスホスホネートによる成長阻害についてのIC50値を判定するために培養液微量希釈法を用いた。100μLの最終容積になるように半対数連続により予め希釈した(0.316mM〜0.1pM)、10μLの試験化合物を含む96ウェル平底培養プレート中に、5,000細胞/ウェルの密度で細胞を播種した。NBPは通常最初にH2O(0.01M)に溶解し、NNBPは通常DMSO(0.01M)に溶解した。次いでプレートを、100%湿度の5%CO2雰囲気中、37℃で4日間インキュベートし、続いて、MTT((3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド)細胞増殖アッセイ(ATCC、Manassas、VA)を用いて、用量応答曲線を得た。DMSOキャリヤーは細胞増殖に対して効果がなかった。

[00247]腫瘍細胞侵襲性及びインビボでの結果。本発明者らは、親油性ビスホスホネートが、腫瘍細胞侵襲性に対して顕著な効果を有するかどうかを検討した。マトリゲル(Matrigel)侵襲性アッセイにおいて、MDA−MB−231細胞、侵襲性ヒト乳癌の腺癌細胞株を、ビスホスホネートと培養すると、親油性ビスホスホネート、BPH−716は、ゾレドロネート(BPH−716、約30nM IC50;対ゾレドロネート、約40μM IC50)より、約1000倍阻害性が高かった。そうした化合物がインビボで活性を有しているかどうかを判断するために、本発明者らは、マウス異種移植片系(xenograft system)のSK−ES−1肉腫細胞(Kubo2007)を使用した。ゾレドロネートは、対照に対して癌細胞増殖の有意の(p<0.01)低減を引き起こしたが、親油性ビスホスホネート(BPH−715)の効果はより顕著であり(p=0.032対ゾレドロネート)、体重減少又は他の副作用は認められなかった。マウスγδT細胞には、IPPによる活性化に必要なVγ2Vδ2T細胞受容体が欠如しているので、このマウスモデルにおける活性は、癌細胞の増殖及び侵襲性への直接活性に起因すると考えることができる。これらの結果は、より親油性のビスホスホネートは、インビトロとインビボの両方で、腫瘍細胞増殖/侵襲性に対して強力な直接的活性を有することを実証している。これらは、ヒトT細胞活性化において高い効能も有することができ、これはIPP蓄積に起因すると考えられる。

- 前記化合物が請求項1に記載の化合物である、請求項68〜81のいずれか一項に記載の方法。
- 前記化合物が化合物715である、請求項76、77又は80に記載の方法。
- 前記化合物が化合物754である、請求項78、79又は81に記載の方法。
- 1つ又は複数の特性のための有機ビスホスホネート試験化合物をスクリーニングする方法であって、
前記試験化合物を提供するステップと、
FPPS酵素アッセイ;
GGPPS酵素アッセイ;
DPPS酵素アッセイ
からなる群から選択される少なくとも2つの酵素アッセイで前記試験化合物の性能特質を測定するステップと、
癌細胞又は腫瘍成長阻害アッセイ;
T細胞活性化アッセイ;
骨吸収アッセイ;
骨結合アッセイ
からなる群から選択される少なくとも2つの活性アッセイで前記試験化合物の活性レベルを測定するステップと、
前記性能特質及び前記活性レベルを分析するステップと、
前記特質及び活性レベルに基づいて前記試験化合物を選択するステップと、
それによって、前記1つ又は複数の特性について前記試験化合物をスクリーニングするステップと
を含む方法。 - 基準化合物を提供するステップと、前記基準化合物の性能特質を、前記試験化合物の前記性能特質と比較するステップとをさらに含む、請求項85に記載の方法。
- Zが、Z6、Z7、Z8、Z9、Z10、Z11及びZ12のいずれか1つである、請求項1に記載の化合物。
