Description
本出願は、2006年10月26日に出願された米国仮出願番号60/854,550の優先権を主張し、参照することによってその全体を本明細書に組み込む。
適用なし。
抗体生成に使用される動物:単冠白色レグホーンの産卵性雌鳥を抗体生成に使用した(ペプチド抗原当たり3羽の雌鳥)。抗フィチン酸塩抗体を、標準グルタルアルデヒド法を用いて、ペプチドをウシガンマグロブリンに接合することによって調製した。
表1.1α−ヒドロキシビタミンD3を加えたフィチン態リンベースの食餌を与えた雛の血漿リン濃度1に関して、フィチン酸に対する抗体を与えたときの効果
11日齢のレグホーン雛(n=6)に、1α−ヒドロキシビタミンD3(20μg/kg食餌)のみ、または、1α−ヒドロキシビタミンD3プラス卵抗体(1g/kg乾燥卵黄抗体粉末の食餌)、を加えたフィチン態リンを含むフィチン酸塩態リン(Pi欠乏)食餌を与えた。この食餌を与えて10日後に血漿リンを測定した。
2ビタミンD食餌(1α−ヒドロキシビタミンD3)を与えた雛は、Pi欠乏食餌を与えた雛と比べて、血漿リンが増大した(p=0.0004)
*食餌ビタミンDの存在下で、フィチン酸に対する抗体を与えた雛は、ビタミンDのみの処置と比べて、血漿リン濃度を低下させた(p=0.001)。

この実験例に使用された動物モデルは、アデニン誘導尿毒症ラットモデルである(例えば、Yokazawa et al.,Nephron 1986,44:230−234;Katsumata et al.,Kid Intl 2003,64:441−450;およびLevi R et al.,J Am Soc Nephrol 2006,17:107−112、各々は、ここで参照することでその全体を組み込む)。
5/6腎摘出に関して(例えば、Cozzolino M et al.,Kidney Int.2003,64:1653−61参照)、左側腎動脈のいくつかの分岐を縛り、右側の腎臓を摘出した。5/6腎摘出したラット(例えば、雄のSprague Dawleryラット、約175−250g、1群あたり最大10ラット)に、無機リン酸塩およびフィチン態リン酸塩の両方を含む高リン酸塩食餌(例えば、0.9%リン酸塩)を与える。これらのラットは、手術後数週間で(例えば、4乃至8週間)尿毒症になり、腎不全、高リン血症および活性ビタミンD3(1α−ヒドロキシビタミンD3および1α,25−ジヒドロキシビタミンD3)不足を発症する。抗フィチン酸抗体または抗フィチン酸塩抗体の量を増やしながら、高リン酸塩食餌を与えた上記5/6腎摘出ラットに毎日の経口投与処置することによって、上記ラットにおいて、投与量に相関して血清リン酸塩濃度が低減した。抗体を、手術後最初の数週間内、および、その後に与える場合、これらの抗体は、上記ラットの高リン血症の発症を予防または遅延する。
Claims (15)
- フィチン酸またはフィチン酸塩を含む食餌をとっていて、高リン血症を発症しているか、または、その危険性があるヒト対象またはヒト以外の動物対象のリン酸塩吸収を低減する方法において、当該方法が、
前記対象の血清リン酸塩濃度を低減または維持するのに有効な量の抗フィチン酸抗体または抗フィチン酸塩抗体を前記対象に経口投与するステップを具えることを特徴とする方法。 - 前記対象が腎臓病を患っていることを特徴とする請求項1に記載の方法。
- 前記腎臓病が、末期腎臓病、急性腎不全、慢性腎不全、多嚢胞性腎臓病、慢性腎臓病、急性腎尿細管壊死、腎機能を低下させる感染症、および、尿管閉塞から選択されることを特徴とする請求項2に記載の方法。
- 前記対象がビタミンD化合物を摂取していることを特徴とする請求項1に記載の方法。
- 前記抗体がIgY抗体であることを特徴とする請求項1に記載の方法。
- 前記抗体が鳥類の卵から得られることを特徴とする請求項1に記載の方法。
- 前記抗体がリン酸塩結合剤とともに投与されることを特徴とする請求項1に記載の方法。
- 前記対象がヒト対象であることを特徴とする請求項1に記載の方法。
