Abstract
【解決手段】低分子量キトサンオリゴマーは、自己集合してsiRNAをナノサイズ粒子にすることができ、酵素分解に対して保護を提供し、インビトロで長時間にわたって安定な遺伝子サイレンシングに関与することができた。siRNAの低分子量キトサンとの複合体を処方する際の構造変数の制御は、インビトロおよびインビボのsiRNAの有効な代替デリバリーシステムを提供する。

2.1.物質
サイトメガロウイルスプロモーターおよびホタルルシフェラーゼ(pLuc)または緑色蛍光タンパク質(pGFP)を含むGMPグレードのプラスミド(gWiz(商標))をAldevron、Fargo、ND、USAから購入した。リポフェクタミン2000(LF2000)をInvitrogenから購入した。直鎖PEI;ExGen 500(22kDaの分子量)をFerementas、Germanyから購入した。分岐PEI(25kDaの分子量)をAldrich Sweden、Stockholm、Swedenから購入した。

配列番号2:アンチセンス、5’−UCGAAGUACUCAGCGUAAGdTdT−3’)は、ルシフェラーゼ遺伝子(siSNA−Luc)を標的とする未修飾siRNA二本鎖であり、MedProbe(Lund、Sweden)から注文した[非特許文献28]。
配列番号4:アンチセンス、5’−UUGAUGUGUUUAGUCGCUAUU−3’)をDharmacon Research、Inc.(Lafayette、CO)から注文した。
様々な鎖長の完全脱N−アセチル化(脱アセチル化度>99.8%;FA<0.001)直鎖および7%三糖類置換低分子量キトサン(分岐キトサン)を記載されているようにして調製し、特性化した[非特許文献25、29]。34、50および85モノマー単位の数平均重合度(DPn)を有する低分子量キトサンを全体にわたって使用した。多角レーザー光散乱(SEC−MALLS)を用いたサイズ排除クロマトグラフィーにより鎖長分布を分析した。
ヒト胎児腎臓細胞系HEK293(293細胞)をATCC、Rockville、MD、USAから入手した。ホタルルシフェラーゼを発現する、安定してルシフェラーゼを発現するHEK293(293−Luc細胞)はフィンランドのクオピオ大学薬学部のPaavo Honkakoski博士から贈られた[30]。安定してルシフェラーゼを発現する卵巣癌細胞系(SKOV−3−Luc)もドイツ、マールブルグのフィリップス大学薬理学および毒物学部のAchim Aigner博士から贈られた[17]。全ての細胞を供給者の推奨にしたがって維持した。
キトサンを滅菌MilliQ水中にpH6.2で溶解させ、続いて滅菌濾過することにより、キトサンストック溶液(0.2mg/ml)を調製した。キトサン、次いでsiRNAストック溶液またはsiRNA/pDNA(同時トランスフェクションの場合)を、記載されているように、ボルテックスミキサー(Heidolph REAX 2000、レベル4、Kebo Lab、Spanga、Sweden)上で強力に撹拌する間に滅菌MilliQ水に添加することにより、キトサン複合体を処方した[25]。1μgのpDNAまたはsiRNAあたり、次の量の様々なキトサンを使用して、1:1(+/−)の電荷比でキトサン複合体を調製した:0.58μgの直鎖キトサン、7%のA−A−Mで置換された0.69μgのキトサン(分岐キトサン)[25、27]。記載されているように、ボルテックスミキサー上で強力に撹拌する間に、PEI溶液をsiRNAストック溶液またはsiRNA/pDNA(トランスフェクションの場合)に添加することにより、PEIのsiRNA複合体を調製した[非特許文献31]。処方をトランスフェクション前に、約10分間室温で放置した。LF2000複合体を形成するために、siRNAストック溶液またはsiRNA/pDNA(同時トランスフェクションの場合)をボルテックスミキサー上で強力に撹拌する間、キトサン溶液に添加した。siRNAおよびLF2000溶液をどちらもトランスフェクション培地OptiMEM Iで希釈した。LF2000複合体をトランスフェクション前に約30分間室温で放置した。予備実験に基づいて、PEI複合体については15:1(+/−)の電荷比およびLF2000複合体については2:1(+/−)の重量比を選択し、全体にわたって使用した(データは不図示)。
アガロースゲル遅延度アッセイを用いてsiRNA複合体の物理的安定性を調べた。40mMのTAE緩衝液中4%アガロース(MetaPhor(登録商標)Agarose、Cambrex Bio Science Rockland、Inc.、Rockland、ME、USA)を記載されているようにして用いた[非特許文献25]。複合体化siRNAの酵素分解に対する保護を、この複合体を1.5UのRNase A(Ambion、UK)とともに30〜90分間記載されているようにしてインキュベートした後に調べた[非特許文献18]。インキュベーション後、複合体をヘパリン(5mg/ml)で解離させ、アガロース遅延度アッセイを用いてsiRNAの完全性を調べた。ストック溶液から得られたsiRNAを対照として使用した。
記載されているようにしてNanosizer ZS(Malvern Instruments、Malvern、UK)を用いた光子相関分光法により複合体のサイズを測定した[非特許文献25]。複合体をMilliQ水中5μg/mlのsiRNA濃度で調製した。すべての測定を25℃で行った。
トランスフェクション実験の24時間前に、HEK293(293、293−Luc)細胞およびSKOV−3−Luc細胞を96ウェル組織培養プレート(Costar、Cambridge、UK)中に播種して、トランスフェクション当日に80〜90%の細胞密集度を得た。トランスフェクションをpH7.4で無血清培地(OptiMEM I Reduced Serum Media、Gibco/BRL Life Technologies AB、 Taby、Sweden)中、または10%血清(FBS)の存在下で行った。マンニトールの添加により等張性(300mOsm/kg)を得た。細胞を予熱したOptiMEMで洗浄し、50μlのsiRNA複合体処方を各ウェルに添加した。同時トランスフェクション実験において、各ウェルあたり0.33μgのpDNA(pLucまたはpGFP)を使用した。ミスマッチ(対照)siRNAをすべてのインビトロ実験に含めた。5時間のインキュベーション後、処方を取り出し、0.2mlの新鮮な培地を添加した。2日を超える実験については1日おきに培地を交換した。トランスフェクション後24〜120時間の範囲の事前に特定された時点で、細胞を予熱したPBS(pH7.4)で洗浄し、ルシフェラーゼ溶解緩衝液(Promega、Madison、WI)で溶解させた。ルシフェラーゼ遺伝子発現を次に照度計(Mediators PhL、Vienna、Austria)を用いて測定した。発現されたルシフェラーゼの量を、ホタルルシフェラーゼ(Sigma、St Louis、MO)を用いて作成した標準曲線から決定した。
様々なsiRNA処方の293−Luc細胞における細胞内デヒドロゲナーゼ活性(細胞毒性の尺度)に対する影響を、記載されたようなMTT法により評価した[32]。簡単に言うと、293−Luc細胞を前述のようにトランスフェクトした。5時間のトランスフェクション後、MTT(3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミド)(Sigma、Deisenhofen、Germany)のリン酸緩衝塩溶液(PBS)中溶液を添加した。4時間後、100μlの酸−イソプロパノール(0.04Mのイソプロパノール中HCL)を添加することにより、ホルマザン結晶を溶解させた。プレートリーダー(TECAN Safire2、Tecan Austria GmbH、Grodig、Austria)を用いて690でバックグラウンド補正をして、570nmで吸光度を測定した。同じようにして処理した培地に関して装置を0吸光度に設定した。処理細胞の細胞内デヒドロゲナーゼ活性を対照(未処理)細胞の細胞内デヒドロゲナーゼ活性と関連させ、次式から計算した:

式中、A(試験)およびA(対照)はそれぞれ処理細胞および対照細胞の吸光値である。別のセットアップにおいては、細胞を培地中でトランスフェクション後24時間成長させた。次に、これらをMTT試薬で処理して、様々な処方の遅延毒性を試験した。
四連試料を用いて最低2回実験を行った。全てのデータを平均値±標準偏差として表す。平均値間の統計的差を、ANOVAを用いて調べた。グループ平均間の差は、p<0.05で有意であると見なした。
本発明者らは、様々な鎖長の直鎖および分岐キトサンオリゴマーがsiRNAと安定な複合体を形成できる能力をゲル遅延度アッセイにおいてまず調べた。より高い電荷比で、より長い直鎖オリゴヌクレオチドと配合された複合体のみがpH8.0(電気泳動緩衝液に通常用いられるpH)でsiRNAを保持した(図1A)。85モノマー単位(DPπ85)の数平均重合度(DPn)を有する直鎖低分子量キトサンは、試験した全ての電荷比で最高の物理的安定性をもたらした。対照的に、ゲル緩衝液のpHが7.4に低下した場合、試験したすべての低分子量キトサンはsiRNAと安定な複合体を形成することができた。高いpH値での分岐および短鎖キトサンオリゴマーのsiRNA処方の減少した物理的安定性は、ポリカチオンのより高い電荷密度を示唆し、物理的に安定な複合体を形成するために低分子量キトサンと配合され、あらかじめ最適化されたpDNA複合体と比べて、より強力なsiRNAとの相互作用が必要とされる[非特許文献25、27]。
siRNA処方の粒子サイズはこれらの組織分布および細胞取り込みに大きく影響を及ぼし得るので、したがって、本発明者らは低分子量キトサンを配合したsiRNA複合体の粒子サイズを調べた。表1は低分子量キトサンがsiRNAと自己集合してナノサイズ粒子(34〜86nm)になることを示す。得られた粒子のサイズは成分の+/−電荷比に依存していた。小粒子サイズ(34〜46nm)は最低の電荷比10:1(+/−)で得られたが、より高い電荷比を使用した結果、比較的大きな粒子サイズ(61〜86nm)が得られた。粒子サイズを光子相関分光法により決定した。
ほとんどのインビトロ実験において、siRNA分子はそれらのpDNA標的と同時に送達(同時トランスフェクション)されるので、本発明者らは、同じレポーター配列(siRNA−Luc)を標的とするsiRNAと合わせたホタルルシフェラーゼレポーター(pLuc)をコードするpDNAのパッケージを送達する低分子量キトサンの有効性を、対照としてミスマッチ(非サイレンシング)siRNAを用いてまず試験した。トランスフェクションを293細胞(ルシフェラーゼレポーターを発現しない)において、pDNA用量、最も有効な低分子量キトサンおよび電荷比の最適化トランスフェクション(本発明者らの実験室でpDNAデリバリーについて最適化した)下で行った[非特許文献27]。分岐DPn34キトサンオリゴマーまたはLF2000により送達されるsiRNA−Lucは、対照処方(pLucのみ)と比較してルシフェラーゼ発現の著しいノックダウン(85〜90%)をもたらした(図2A)。ルシフェラーゼ発現における非有意阻害が両デリバリーシステムのミスマッチsiRNAに関して観察された。
a)鎖長、主鎖分岐および電荷比
キトサン構造および処方パラメータの低分子量キトサンによるsiRNAデリバリーに対する影響を調べるために、本発明者らは、293−Luc細胞においてsiRNA複合体が関与するルシフェラーゼ発現のインビトロサイレンシングに対する鎖長、分岐および電荷比の影響をまず調べた。直鎖キトサンに関して、34モノマー単位より長い鎖長を有するキトサンは電荷比と独立して顕著なルシフェラーゼサイレンシングに関与した(図3A)。分岐キトサンオリゴマーを配合した複合体が関与するルシフェラーゼサイレンシングは電荷比および鎖長の両方に依存していた(図3B)。より長い分岐キトサンオリゴマーに関して、より高い電荷比はキトサン主鎖の分岐(置換)の負の効果を相殺し得る。これらの結果は、低分子量キトサンの高い電荷密度が物理的に安定な複合体をもたらすだけでなく、インビトロで有効なsiRNA処方ももたらすことを強調する。
次のステップにおいて、様々な直鎖低分子量キトサン(一定の電荷比)を配合した複合体のルシフェラーゼサイレンシング活性に対するsiRNA濃度の影響を293−Luc細胞において調査した。最低のsiRNA濃度(15〜50ng/ウェル、22〜73nM/ウェルに等しい)で、直鎖DPn85複合体は、ルシフェラーゼ発現を対照未処理細胞の72〜95%にノックダウンすることによって最高の効力を示した(図4)。DPn18を除いて、試験した直鎖キトサンの複合体は、siRNA濃度が70から300ng/ウェル(103〜440nM/ウェル)に増加した場合に、匹敵するルシフェラーゼサイレンシング特性を示した。直鎖の、より長鎖の低分子量キトサンを配合した複合体の効力がより高いことは、最高の遺伝子サイレンシング活性の達成におけるsiRNA処方の物理的安定性の役割を裏付ける。
本発明者らは、様々なsiRNA処方の有効性に対するトランスフェクション培地中の血清の効果も調査した。34〜85モノマー単位の範囲のDP値を有する直鎖キトサンおよび分岐DPn85キトサンオリゴマーを配合したsiRNA複合体は、293−Luc細胞中、10%血清の存在下で遺伝子サイレンシング活性を保持していた(図5)。対照的に、PEIおよびLF2000を配合したsiRNA複合体の遺伝子サイレンシング活性は損なわれた。有効性が損なわれる一つの可能な理由は、トランスフェクション中の粒子凝集である。
より高いルシフェラーゼサイレンシング効率が増加した細胞毒性の結果でないことを保証するために、本発明者らは、比較的高濃度のsiRNA(150nM)を配合した様々なポリカチオン複合体の、細胞形態および細胞内デヒドロゲナーゼ活性に対する影響を、MTT法を用いて調査した。5時間のトランスフェクション後、直鎖DPn85を配合したsiRNA複合体において、細胞形態および細胞内デヒドロゲナーゼ活性(細胞毒性の尺度)に対する影響は観察されなかった(図6)。対照的に、PEIおよびLF2000に関して著しい毒性効果が観察され、直鎖PEIについて毒性は最高であった。LF2000複合体で処理された細胞は24時間後にその細胞内デヒドロゲナーゼ活性を回復したが、PEIで処理された細胞は24時間後にデヒドロゲナーゼ活性においてさらなる減少を示した。これらの結果は、直鎖DPn85低分子量キトサンの急性細胞毒性は、比較的高濃度のsiRNAが使用された場合でさえも、PEIおよびLF2000よりも低いことを示す。
最後に、siRNA−Lucの293−Luc細胞へのデリバリー後のルシフェラーゼサイレンシング時間経過を調査した。34〜85モノマー単位の範囲のDP値を有する低分子量キトサン、PEIおよびLF2000を試験した。DPn18キトサンは、さらに長いキトサンよりも有効でないため(図4)、含めなかった。直鎖DPn85を配合したsiRNA複合体はルシフェラーゼサイレンシングの早発性に関与し、この場合、1日後に著しい効果が検出され(70%)、2日後に最大(92%)に達した(図7A)。遺伝子サイレンシング活性は5日間持続し、このことはインタクトなsiRNAが細胞内で安定して放出されることを示唆する。分岐キトサンオリゴマーはこれらの直鎖カウンターパートと比較して有効性が低いルシフェラーゼサイレンシング動態を示した(図7B)。LF2000およびPEIは、低分子量キトサンが関与するものよりも低い遺伝子サイレンシング効果を示した(図7C)。別の細胞系(SKOV−3−Luc)におけるルシフェラーゼサイレンシング動態は、293−Luc細胞におけるものと類似した結果をもたらした(図7D)。
Claims (19)
- (a)30から300の範囲の重合度(DPn)を有する低分子量キトサン(ここで、前記低分子量キトサンの脱アセチル化度は90%を超える)と、
(b)オリゴヌクレオチド
の複合体を含む組成物。 - 化学的または酵素的方法を用いて高分子量キトサンから前記低分子量キトサンを得る、請求項1に記載の組成物。
- 前記低分子量キトサンの脱アセチル化度が95%を超える、請求項1に記載の組成物。
- 前記低分子量キトサンの脱アセチル化度が99%を超える、請求項3に記載の組成物。
- 前記組成物が本質的に正味の正電荷比を有する、請求項1に記載の組成物。
- ターゲティングリガンドおよび安定剤を用いて前記低分子量キトサンを誘導体化する、請求項1に記載の組成物。
- 前記オリゴヌクレオチドが、宿主細胞中に導入されるとその機能を発現するサイレンシング配列を含む、請求項1に記載の組成物。
- 前記オリゴヌクレオチドが、RNA分子、アンチセンス分子、リボザイム、およびミクロRNAからなる群から選択される、請求項7に記載の組成物。
- 前記組成物が3.5から8.0の範囲のpHを有する、請求項8に記載の組成物。
- 前記組成物が7.1から7.6の範囲のpHを有する、請求項9に記載の組成物。
- (a)低分子量キトサンを水性溶媒と接触させるステップと、
(b)ステップ(a)の水溶液をオリゴヌクレオチドと水性溶媒中で混合するステップと、
(c)前記組成物のpHを3.5〜8.0の範囲に維持するステップと、
を含む、請求項1に記載の組成物の調製法。 - ステップ(b)で製造された生成物溶液の体積を減じて、所望の濃度の組成物を得ることをさらに含む、請求項11に記載の方法。
- 核酸を哺乳動物に投与する方法であって、前記哺乳動物中に請求項1に記載の組成物を導入することを含む方法。
- 前記組成物を哺乳動物に、肺、鼻、口腔、眼、頬、舌下、局所、直腸、または膣経路によって粘膜組織に投与することにより導入する、請求項13に記載の方法。
- 前記組成物を、非経口経路(静脈内、筋肉内、皮内、頭蓋内、髄腔内、皮下、もしくは心臓内投与)による粘膜下組織への投与、あるいは内臓器官、血管、または手術中に露出する他の体表面もしくは体腔への投与により哺乳動物中に導入する、請求項13に記載の方法。
- 前記組成物が、哺乳動物の少なくとも1つの細胞の内部でその機能を発現できるオリゴヌクレオチドである、請求項13に記載の方法。
- 哺乳動物を予防または治療するための医薬を製造することを含む請求項1に記載の組成物を使用する方法であって、前記医薬が請求項1に記載の組成物を含む方法。
- インビボまたはインビトロ診断法において使用することを含む、診断薬として請求項1に記載の組成物を使用する方法。
- 悪性腫瘍、自己免疫疾患、遺伝性疾患、病原性感染症および他の疾患を予防または治療するための遺伝子療法、アンチセンス療法、または遺伝子ワクチン接種において使用される医薬の製造における請求項17に記載の組成物の使用であって、
a)肺、鼻、口腔、眼、頬、舌下、直腸、もしくは膣経路により粘膜組織へ、
b)静脈内、筋肉内、皮内、頭蓋内、髄腔内、皮下、もしくは心臓内投与である非経口経路により粘膜下組織へ、あるいは
c)内臓器官、血管、または手術中に露出する他の体表面もしくは体腔へ、の投与によって、哺乳動物中に前記組成物を導入することを含む使用。
