Description
−本発明の修飾ニューロトロフィンの切断によって、または、天然ニューロトロフィンまたは天然プロニューロトロフィンの断片とニトロ化剤との接触によって、
それによって候補断片の集団を得る、および
−前記候補断片の集団の中から、少なくとも1つの残基、好ましくはTyrおよびTrp残基から選択される少なくとも1つの残基に、少なくとも1つのニトロ基を保持し、且つ、運動ニューロンアポトーシスおよび/または神経突起成長を誘導および/または刺激する能力を有する断片を選択する。
−本発明の修飾ニューロトロフィンまたは本発明の保存的断片の少なくとも1つのアミノ酸の置換および/または欠失および/または付加によって、または、天然ニューロトロフィンまたは天然プロニューロトロフィンの、または天然ニューロトロフィン断片の、または天然プロニューロトロフィン断片の変種とニトロ化剤との接触によって、

本発明の修飾ニューロトロフィン(または保存的断片もしくはその変種)は、運動ニューロンのアポトーシスおよびまたは感覚神経節からの神経突起成長の誘導および/または刺激の能力を有する。
−本発明の非保存的断片および非保存的変種から成る群から選択される少なくとも1つの要素を含む何れかの組成物、または、
−少なくとも1つの前記競合化合物を含む何れかの組成物、または
−本発明の結合剤(とりわけ本発明の特異的な結合剤、さらにとりわけ本発明の抗体、化学的模倣物、および生物学的模倣物)から成る群から選択される少なくとも1つの要素を含む何れかの組成物。
− 神経障害性の痛み(とりわけ、片頭痛、慢性的片頭痛、推定鎮痛薬乱用頭痛(probable analgesic−abuse headache PAAH)、原発性線維筋痛症候群(primary fibromyalgia syndrome PFMS)、神経損傷誘導性神経障害性疼痛(nerve−injury induced neuropathic pain)、坐骨神経傷害(sciatic nerve lesions)、慢性的絞搾損傷(chronic constriction injury))、

本発明は、また、本発明の修飾ニューロトロフィン(または保存的断片もしくはその変種)が運動ニューロンに対して示す可能性のあるアポトーシス活性、および/または、本発明の修飾ニューロトロフィン(または保存的断片もしくはその変種)が感覚神経節に対して示す可能性のある神経突起成長効果を阻害および/または遮断する能力を有する化合物のスクリーニングの方法に関する。
神経成長因子(NGF)の過剰発現およびパーオキシナイトライトの生産の増大は、いくつかの神経変性の疾病において生じる。我々は、NGFが、その生物学的活性を調節する翻訳後の酸化的またはニトロ化的修飾を受け得るのかどうかを調べた。天然NGFと比較して、パーオキシナイトライト処理NGFは、生理的に適切な濃度において、p75NTR依存性運動ニューロンアポトーシスを誘導する特殊な能力を示した。天然NGFが運動ニューロンの死を誘導するために、酸化窒素(NO)の外的供給源を必要とするのに対し、パーオキシナイトライト処理NGFは、外因的なNOがない場合でも、運動ニューロンアポトーシスを誘導した。にもかかわらず、NOはパーオキシナイトライト修飾NGFのアポトーシス活性を増強した。p75NTRに対する阻止抗体またはアンチセンス処理によるp75NTR発現のダウンレギュレーションは、パーオキシナイトライト処理NGFによって誘導される運動ニューロンアポトーシスを阻害した。我々は、どんな酸化的修飾が、この中毒性機能獲得を誘導するかを調べ、パーオキシナイトライトが用量依存的な様式でチロシンニトロ化を誘導することを発見した。そのうえ、3,3’−ジチロシン架橋結合の証拠が認められないにもかかわらず、パーオキシナイトライトは、NGFの安定的な高分子量オリゴマーの形成を誘発した。尿酸塩によるチロシンニトロ化の阻止は、NGFアポトーシス活性に対するパーオキシナイトライトの効果を無効にした。これらの結果は、パーオキシナイトライトによるNGFの酸化は、p75NTRを通したNGFアポトーシス活性を10,000倍に増強することを意味する。我々の知る限りでは、これは、ニューロトロフィンをアポトーシス薬剤へと変換する始めて発見された翻訳後修飾である。
我々は、NGFが、翻訳後の酸化的またはニトロ化的修飾を受け、その機能的活性を変化させ得るかどうかを調べた。我々は、インビトロにおけるパーオキシナイトライトによるNGFの酸化がニトロ化を引き起こし、高分子量のオリゴマーの形成を誘導することを報告する。そのうえ、これらの酸化的修飾は、生理的に適切な濃度において、p75NTR依存的運動ニューロンアポトーシスを誘導する特殊な能力を付与する。我々のデータは、パーオキシナイトライトによる酸化ストレスは、ニューロトロフィン活性を決定的に調整できることを示す。
パーオキシナイトライト処理:パーオキシナイトライトは、特にUpstateから入手可能である(例えばUpstate USA, Charlottesville, VA 22903, USA)。パーオキシナイトライト濃度は、302nm(ε=1,670M−1cm−1)にて分光光度的に測定した。希釈した原液を、0.01MのNaOHにて新鮮に調製した。NGF(Harlan; Indianapolis, USA)と、種々の濃度のパーオキシナイトライト(2mMから0.25mM)との反応が、0.2mg/mLから1.0mg/mLのタンパク質濃度で行われ、同様の結果が得られた。反応は、20mMの炭酸水素ナトリウムを含む50mMのナトリウムリン酸緩衝液(pH7.4)で行った。NGFは、パーオキシナイトライト(各々1μL)の10ボーラス添加(10 bolus addition)に供し、所望の終濃度のパーオキシナイトライトに達成させた。パーオキシナイトライト原液の1ボーラスを試験管の上部に迅速に添加し、3秒間ボルテックスにより混合した。当該方法を10回繰り返した。pH変化または混入に起因するパーオキシナイトライト処理の非特異的な効果の可能性を除外するため、コントロール実験を、希釈したNaOHまたは分解するパーオキシナイトライト(逆順の添加(ROA))を使用して行った。パーオキシナイトライトと、ウシ血清アルブミン(Sigma)、FGF−1(Sigma)およびFGF−2(R&D Systems)との処理を、上述の通り、0.2 mg/mLの濃度で行った。
パーオキシナイトライトによる酸化はNGFアポトーシス活性を増強する
GDNF (1ng/mL)で維持した培養物において、p75NTRを発現している運動ニューロンは、NGFに感受性でない。しかしながら、パーオキシナイトライト処理したNGFは、1ng/mLと同程度に低い濃度で、運動ニューロン死を誘導した(図2A)。分解したパーオキシナイトライト(ROA)で処理したNGFは、運動ニューロンの生存に影響を与えなかった(図2A)。パーオキシナイトライト処理は、用量依存的にNGFアポトーシス活性を増強し、パーオキシナイトライト濃度が0.5mMを超える場合にはプラトーに達した(図2B)。以前に報告されたように[25]、酸化窒素供与体DETA−NONOate(10μM)から作られる、定常状態濃度の酸化窒素(<50 nM)の存在下、NGF−ROAは、10ng/mLを超える濃度で、運動ニューロン減少を有意に誘導した(図2C)。さらに、酸化窒素の存在下、パーオキシナイトライトで修飾されたNGFは、アポトーシス活性の増大を示し、わずか1pg/mLで33%の運動ニューロン減少を誘導した(図2C)。運動ニューロン培養物へのパーオキシナイトライト処理−BSA、−FGF−1または−FGF−2の添加は、運動ニューロン死を誘導せず(図2D)、このことは、パーオキシナイトライト処理NGFの特異的な効果を示している。
我々は、その後、NGFに対するパーオキシナイトライト処理によって誘導される修飾を分析した。パーオキシナイトライトの連続した大量添加にNGFをさらすと、3つの高分子量種の用量依存的な出現が生じることがSDS−PAGEからわかる。天然NGFの染色強度は、次第に、パーオキシナイトライト濃度の増大とともに減少した(図4A)。分解したパーオキシナイトライト(ROA)によるNGFの処理は、この移動シフトを誘導した。NGFオリゴマーの形成は、溶液中で、リアルタイム多角度光散乱検出(MALS)分析と組み合わせたHPLCサイズ排除クロマトグラフィーによって確認した(図4B)。分解したパーオキシナイトライトで処理したNGF(NGF−ROA)は、サイズ排除カラムから、二量体(33.0±1.2KDa)に相当する質量を有した単一のピークとして溶出した。対照的に、パーオキシナイトライト処理NGFは、二量体(33.2±0.6KDa)、四量体(68.5±3.5KDa)および八量体(125.0±10.0KDa)に相当すると思われる3つのピークとして溶出した。驚くべきことに、パーオキシナイトライト処理したNGF二量体は、天然二量体(NGF−ROA)の前に溶出し、タンパク質のニトロ化に起因する構造変化の存在を反映している。
チロシンニトロ化がNGF生物活性の修飾に重大な意味を持つかどうかを確認するために、我々は、テトラニトロメタンでNGFを処理した(TNM; 40倍過剰)。TNMは、タンパク質における3−ニトロチロシンを形成するために、アルカリpHにて一般に使われる。使用される条件において、TNMは、質量分析によって証明されるように、チロシンニトロ化だけを誘導した(下記参照)。TNMで処理したNGFは、37.9および39.9分で溶出する2つの生成物を分離した(図8A)。質量分析によって明らかにされるように、TNM処理は、NGF鎖Aの分子量を90Da増加させ(13,252から13,342Daまで;図8B)、これは、主要な生成物は二重のニトロ化種であることを示す。しかしながら、単一ニトロ化種はまた、13,297Daで認められた(図8B)。同じパターンがNGFの鎖Bについて認められた。消化された生成物のタンデム型質量分析は、ニトロ化残基としてTyr52およびTyr79を同定し(図8C)、全分子量変化を説明しうる(図8B)。トリプトファンニトロ化または3,3’−ジチロシン形成は、TNM処理NGFにて認められなかった。40倍過剰のTNMで処理したNGFの電気泳動パターンは、パーオキシナイトライト処理NGFにて認められる場合に匹敵し、このことはNGFオリゴマーの形成を示唆する(図8D)。
ALSにおける運動ニューロン死およびアストロサイト反性は、活性酸素および窒素種の生産の増大に関与するため[28、50、51]、我々は、分泌されたNGFの酸化またはニトロ化が、運動ニューロンに対するアポトーシス活性を増強するかどうかを調べようと考えた。インビトロにおけるパーオキシナイトライトによるNGFの酸化は、酸化窒素の存在下、運動ニューロンのアポトーシスを誘導する効力を10,000倍まで増大させた。我々の知識の及ぶ限りでは、これは、pg/mLの範囲の生理的に適切な濃度の培養におけるニューロトロフィン誘導細胞死の最初の報告である。NGFレベルの増加は、ALSで生じる運動ニューロンの進行性の死に関係していた[23−25]。我々は、以前に、SOD1G93A ALSマウスからの脊髄抽出物が、酸化窒素の外的供給源の存在下で、培養された運動ニューロンのp75NTR依存的アポトーシスを刺激するのに十分なNGFを含むことを報告した[25]。しかしながら、SOD1G93Aマウスからの変性している脊髄における、ELISAによって測定されるNGFレベルは、mLあたりピコグラムの範囲であり[25]、純粋なNGFが純粋な運動ニューロン培養にてアポトーシスを誘導するために必要な濃度の10,000倍低い濃度であり、パーオキシナイトライト処理NGFの効力と同程度である。

− 関節の痛み(とりわけ、骨関節炎症状、骨関節症、骨関節炎)、
− 炎症性疼痛、

- 痛みの状態または症状または感覚、とりわけ以下のものの治療および/または予防および/または寛解における使用のための、請求項37に記載の組成物:
− 神経障害性の痛み(とりわけ、片頭痛、慢性的片頭痛、推定鎮痛薬乱用頭痛 PAAH、原発性線維筋痛症候群 PFMS、神経損傷誘導性神経障害性疼痛、坐骨神経傷害、慢性的絞搾損傷)、
− 関節の痛み(とりわけ、骨関節炎症状、骨関節症、骨関節炎)、
− 炎症性疼痛、
− 癌疼痛。 - 運動ニューロンアポトーシスの誘導および/または刺激のために、および/または神経突起成長の誘導および/または刺激のために有用な薬剤としての使用のための、請求項1から24の何れか1項に記載の誘導物質および/または刺激物質。
- 神経障害、神経損傷、脳卒中、脊椎損傷および神経変性の疾病から成る群から選択される疾病、症状または外傷の治療および/または予防および/または寛解における使用のための、請求項1から24の何れか1項に記載の誘導物質および/または刺激物質。
- 請求項1から24の何れか1項に記載の少なくとも1つの誘導物質および/または刺激物質を含む組成物。
- 神経障害、神経損傷、脳卒中、脊椎損傷および神経変性の疾病から成る群から選択される疾病、症状または外傷の治療および/または予防および/または寛解における使用のための、請求項46に記載の組成物。
- 以下を含む、神経変性の症状または疾病、および/または、痛みの状態または症状または感覚の診断のためのインビトロ方法:
−生体試料を、請求項28から34の何れか1項に記載の1つの抗体に接触させること、
−前記少なくとも1つの抗体が前記生体試料中に含まれるリガンドに結合したかどうかを決定し、
そのような結合によって、前記疾病、状態、症状または感覚を示すことまたは予想すること。
