Abstract

膜転位ペプチドZElan094、204Nおよび204、ならびにターゲッティングペプチドHAX42、PAX2、P31およびSni34(米国特許出願第09/079,819号、同第09/079,723号および同第09/079,678号明細書)はfmoc合成プロトコル(Anaspec,Inc.、カリフォルニア州サンノゼ)を使用して化学的に合成した。抗ダンシル抗体でのペプチドの検出を可能にするために、各配列のN末端にダンシル基を付加した(表1)。
活性粒子はコアセルベーション法を使用してポリマーから製造した。好ましくは、粒子径は約5nmと750μmとの間、より好ましくは約10nmと500μmとの間、および最も好ましくは約50nmと800nmとの間である。MTLP若しくはターゲッティングペプチドは、当業者に既知の多様な方法を使用して粒子に複合体形成した。
即効型ウシインスリン(28.1IU/mg)をナノ粒子210mgあたり300IUのインスリンの理論的負荷量でポリラクチド−コグリコリド(PLGA、Boehringer Ingelheim、インジアナ州インジアナポリス)に組み込み、そして該ナノ粒子をダンシル化ZElan094(配列番号2)で被覆した。
1.水を沸騰近くに加熱し、PVAを5%wvまで添加し、そして溶液を冷却まで攪拌した(相A)。
2.アセトンおよびエタノールを混合して有機相(相B)を形成した。
3.PLGAをアセトンおよびエタノール(段階2)に添加しそして攪拌により溶解した(相B)。
4.IKATM反応容器を25℃に設定した。相A(段階1)を反応容器に添加しかつ400rpmで攪拌した。
5.ウシインスリンを攪拌している相A(段階4)に添加した。
6.清浄なチューブおよび未使用の(green)針を使用して、相B(段階3)を、40に設定した蠕動ポンプを使用して攪拌している溶液(段階5)にゆっくりと滴下した。
7.IKATM反応容器の口を開放することおよび400rpmで一夜攪拌することにより溶媒を蒸発させて懸濁液(相C)を形成した。
8.懸濁液、相C(段階7)をXL90遠心機中、12,500ないし15,000rpmで4℃で25ないし40分間遠心分離した。
9.上清を廃棄し、粒子「ケーキ」を粉砕し、そして粒子(相D)をXL90遠心機中12,500ないし15,000rpmで4℃で10〜15分間の遠心分離により200mlのH2Oで2回洗浄した。ダンシル化ZElan094(配列番号2)を最終洗浄液に添加した。
10.上清をデカンテーションし、「ケーキ」を粉砕し、そして粒子を真空オーヴン中で乾燥した。乾燥した粒子を粉砕し、セキュリテナー(securitainer)に入れかつ分析した。インスリン負荷は5%すなわち50mgインスリン/g粒子であった。HPLCで測定したインスリン効力は51.4mg/gであった。走査型電子顕微鏡検査は、直径が約300〜400nmの分離した合理的に球状の粒子を示した。
即効型ウシインスリン(28.1IU/mg)を、ナノ粒子210mgあたり300IUのインスリンの理論的負荷量でPLGAナノ粒子中に組み込み、そしてナノ粒子をMTLP ZElan094(配列番号2)で被覆した。

MTLP−インスリン粒子複合体(実施例3)およびターゲッティングペプチド−インスリン粒子複合体(実施例7)からのin vivoでの経口インスリンの生物学的利用性を、開放ループラットモデルで評価した。
領した動物の間で、60分および120分に血糖値の有意の差異は存在しなかった。これらのデータは、ダンシル化ZElan094−インスリン粒子複合体ならびにダンシル化Zelan011、055、091、144、129、101、129、128および104−インスリン粒子複合体から送達されたインスリンが生物活性のままであったことを示す。さらに、これらのデータは、MTLP−インスリン粒子複合体から送達されたインスリンが、血糖値の有意のかつ持続性の減少を可能にしたことを示す。
DNA含有リポソームおよびDNA含有MTLP被覆リポソームは、後に続くとおり製造した:
溶液1 12nmolのリポフェクタミン(Gibco BRL、メリーランド州ロックビル)±0.6mgのプロタミン硫酸を、最終容量75mlのoptiMEM中で調製した。
溶液2 1mgのpHM6lacZ DNA(Boehringer Mannheim)を最終容量75mlのoptiMEM中で調製した。レポータープラスミドpHM6lacZは、細菌のβ−ガラクトシダーゼをコードするlacZ遺伝子を含有する。
溶液3 溶液1および溶液2を合わせ、そしてRTで15分間インキュベートして複合体形成を可能にした。
溶液4 ZElan094、204N若しくは204(配列番号2、23、24)を溶液3に100mMの最終濃度まで添加しそしてRTで5分間インキュベートした。600mlのoptiMEMを添加しそして溶液を穏やかに混合した。
リポソームおよびMTLP被覆リポソームからCaco−2細胞中へのDNAの送達を、細胞上清中の総タンパク質1mgあたりのβ−ガラクトシダーゼ発現として計算した。β−ガラクトシダーゼ発現はBoehringer Mannheimの化学発光キットを使用して測定した。タンパク質はPierceの微量(Micro)ビシンコニン酸(BCA)タンパク質アッセイを使用して測定した。
公開されたPCT出願第PCT/US98/10088号明細書)の効果を測定した。Caco−2単層はトランスウェル−スナップウェル(Transwell−Snapwell)上で増殖させた。各実験の間にCaco−2単層のTEERを測定することにより細胞の生存率を決定した。TEERの有意の下落は測定されなかった。細胞浸透性は各実験の間にCaco−2単層を横切るマンニトール流入を測定することにより決定した。MTLP、ZElan094の存在下でマンニトール流入の増大は測定されなかった。
、fMLPはGIT中の炎症においてある役割を演じていることが既知であるため、Caco−2単層を横切るfMLPの輸送を減少させるMTLPは、GITにおけるfMLPの化学誘引物質効果を減少させることにより局所炎症の予防において治療的役割を有するかもしれない。
MTLP、ZElan094およびZElan207(配列番号2および102)を水に溶解し、そしてブタ由来パンクレアチンを含有する模擬腸液pH6.8(SIF+パンクレアチン)と37℃で混合した。混合物を37℃で60分間までインキュベートし、指定された時間点でアリコートを採取した。SIFとリガンドとの間の反応を停止するために、適切な時間点後にクエンチ溶液で反応をクエンチした。

されないことを示した。また、クエンチ溶液は回収に影響を及ぼさない。上で使用したHPLC法は安定性を示すアッセイとして至適化されていないが、リガンドピークの消失および新たな成分のピークの出現を明らかに見ることができたことを覚えておくべきである。
モデルのD−体のオピオイドペプチド(H−ffir−NH2;κ受容体特異的;分子量581Da)をSIF中での安定性について評価した。
以下の勾配とともにC−18ショートカラム(Betasilカラム、5μm(50×3mm)PN:055−701−3)を用いるRP−HPLC分析:
・ペプチド対照を最初に注入
・SIF中溶液を0、1、3および24時間に注入

κペプチド、H−ffir−NH2は24時間にわたって非常に安定であった(表9を参照されたい)。質量分析は、最初および24時間後に検出可能なただ1種の化合物を伴いこの結果を確認した。κペプチドをターゲッティング試験におけるさらなる評価に適するモデル薬物として選択した。
κペプチド、H−ffir−NH2ならびに多様なMTLP(ELAN094、ELAN207、ELAN208およびELAN178)の複合物を多様な形式で合成して、in vivo評価のためのその後のバッチ合成の至適の複合戦略を決定した。構造的形式は:
i)C末端若しくはN末端オピオイドペプチド、
ii)リシンリンカーと/伴わずに複合物形成
iii)未標識/ビオチン標識
を包含した。
i)ELAN207オピオイドペプチド複合物(P34−154)がビオチン標識後に活性を保持した
ii)C末端オピオイドペプチドおよびリシンリンカーを伴うELAN094(P37−114)、178(P37−116)および208(P37−115)オピオイドペプチド複合物が、ELAN207(P34−154)で観察されたものに同等の活性を表した
ことを示した。
κペプチド、H−ffir−NH2ならびにMTLP、ELAN094およびELAN207(配列番号2、102)の複合物を、標準的ペプチド合成プロトコルを使用して合成した。κペプチド上の2個のフェニルアラニン残基をトリチウム交換により標識し、そして放射ペプチドの純度をRP−HPLCにより評価した。>95%のペプチド純度および42〜54Ci/mmolの比活性が達成された。
κペプチドの腸吸収に対するELAN094およびELAN207 MTLP(配列番号2、102)の可能な増強効果を覚醒ラットモデルで検査した。
試験は非無作為化並行群デザインであった。250〜350gの重量範囲内のWistarラットを使用した。全動物は試験開始前16時間絶食させた。水は全時点で利用可能であった。
群1(n=6)
10μCiの3H−κペプチド−Kaffiralin−1(普通のκペプチド)の静脈内注入(尾静脈注入)。

100μCiの3H−κペプチド−Elan094(分析済)の十二指腸内点滴注入。Elan094リガンドは膜転位配列を含有する。

100μCiの3H−κペプチド−Kaffiralin−1の十二指腸内点滴注入。
血漿(100〜250μl)はBTS−450(有機組織可溶化剤)で1mlまでにし、そして1時間インキュベートした。10mlのシンチレーション液を添加しそして放射活性を測定した。
該試験は、覚醒ラットモデルにおける十二指腸内点滴注入後のトリチウム標識κペプチド複合物の全身の生物学的利用率を評価するようデザインされた非無作為化並行群生物試験であった。各ラットは1処置のみを受領し、そして血漿サンプルをトリチウム含量について分析した。AUClast、Amax、tmax、分布容積(Vd)およびクリアランスの計算は基礎補正済データに基づいた。絶対的生物学的利用率は効力補正済データを使用して計算した。

順位序列でのID処置のAUClastは後に続くとおりであった。すなわち、処置3−100μCiの3H−κペプチド−Elan094(ID)について260642.53±35010.58dpm/ml.h(CV 13.4%)であり、処置4−100μCiの3H−κペプチド Elan207(ID)(反復)について249021.32±37503.78dpm/ml.h(CV 15.1%)であり、処置4−100μCiの3H−κペプチド−Elan207(ID)(元)について162743.70±13549.10dpm/ml.h(CV 8.3%)であり、処置1−10μCiの3H−κペプチド−Kaffiralin−1(IV)について132184.24±13288.32dpm/ml.h(CV 10.1%)であった一方、リガンドの非存在下で、処置10−100μCiの3H−κペプチド Kaffiralin−1(ID)は73098.43±9493.88dpm/ml.h(CV% 13.0)であった。
大きさの順序でのID処置の投与後の最大放射活性は後に続くとおりであった。すなわち、処置1−10μCiの3H−κペプチド−Kaffiralin−1(IV)について903603.03±186855.31dpm/ml(CV 20.7%)であり、処置4−100μCiの3H−κペプチド Elan207(ID)(反復)について201464.40±44854.19dpm/ml(CV 22.3%)であり、処置3−100μCiの3H−κペプチド−Elan094(ID)について158512.67±18907.79dpm/ml(CV% 11.9)であり、処置4−100μCiの3H−κペプチド−Elan207(ID)(元)について100041.17±6274.57dpm/ml(CV% 6.3)であり、κペプチド単独の処置10−100μCiの3H−κペプチド Kaffiralin−1(ID)については70925.33±23631.28dpm/ml(CV% 33.3)であった。
最大放射活性に達する時間は全ID処置について0.08±0.00時間であった。
処置1、10μCiの3H−κペプチド−Kaffiralin−1(IV)の観察された容積は963.87±255.65ml(CV 26.5%)であった。
処置1、10μCiの3H−κペプチド−Kaffiralin−1(IV)の観察されたクリアランスは142.85±17.24ml/h(CV 12.1%)であった。
組織を秤量しそして代表的サンプル(約0.1g)をシンチレーションバイアルに取り出した。組織を1mlのBTS−450(有機組織可溶化剤)で可溶化した。10mlのシンチレーション液を添加しかつ放射活性を測定した。放射活性はdpm/組織初期重量として表した。
結果を表14に要約する。
カテーテル中で回収された、投与した用量の%は、多様な処置について0.06から0.39%の範囲にわたった。このデータは、投与した用量の無視できる量がカテーテル中で失われることを示唆する。
血漿中で回収された、投与した用量の%をt=6hで計算した。投与した用量の%は多様な処置について0.06から0.42%の範囲にわたった。多様な処置の順位序列は後に続くとおりであった。すなわち、処置3−100μCiの3H−κペプチド−Elan094(ID)について0.42±0.08%(17.7%)であり、処置1−10□Ciの3H κペプチド−Kaffiralin−1(IV)について0.16±0.03%(CV 18.9%)であり、処置4−100μCiの3H−κペプチド−Elan207(ID)(元)について0.10±0.01%(CV 9.2%)であり、また、送達増強剤リガンドの非存在下の処置10−100μCiの3H−κペプチド Kaffiralin−1(ID)で、投与した用量の%は0.06±0.01%(CV 11.6%)であった。この順位序列は、多様な処置についてのBiostudy 1000003のデータパックで報告された絶対的生物学的利用率について観察されたものと同一に相関する。
肝で回収された、投与した用量の%は0.26から6.66%の範囲にわたった。IV投与すなわち処置1−10μCiの3H κペプチド−Kaffiralin−1(IV)後の回収%は6.66±1.77%(CV 26.6%)であった。この回収率はいずれかの他の処置についての肝組織中で回収された量をはるかに超えた。
腎で回収された、投与した用量の%は多様な処置について0.12から0.51%の範囲にわたった。このデータは、3H κペプチドの投与した用量の無視できる量がt=6hに腎組織中に蓄積していたことを示唆する。
GI管中で回収された、投与した用量の%は27.10から89.26%の範囲にわたった。GI管はGI組織、GI内容物およびGI洗浄液にさらに細分することができ、それぞれの範囲はそれぞれ1.62〜11.97%、23.50〜65.68%および1.98〜25.84%であった。GI管で回収された、投与した用量の%として提示されるデータはGI組織の6区分の総和を表す。GI管内で、最高レベルの放射活性は後のほうの区分と関連した(生データの表を参照されたい)。GI区分6およびその対応するGI内容物と関連する回収された放射活性は、他の区分について観察されたものより高かった。
前記L−ペプチドは配列番号2〜13および15〜24よりなる群から選択されるアミノ酸配列を有し、
前記d−ペプチドは配列番号2〜24のL−ペプチドのd−体に対応する配列番号102〜124よりなる群から選択されるアミノ酸配列を有し、また、
逆反転ペプチドは、配列番号2〜24のL−ペプチドの逆反転体に対応する配列番号202〜224のペプチドよりなる群から選択されるアミノ酸配列を有する、
転位ペプチドを含んでなる組成物。
前記d−ペプチドが、配列番号2〜4、16、23および24のL−ペプチドのd−体に対応する配列番号102〜104、116、123および124よりなる群から選択されるアミノ酸配列を有し、
逆反転ペプチドが、配列番号2〜4、16、23および24のL−ペプチドの逆反転体に対応する配列番号202〜204、216、223および224のペプチドよりなる群から選択されるアミノ酸配列を有する、
1.に記載の組成物。
前記d−ペプチドが配列番号5〜13のL−ペプチドのd−体に対応する配列番号105〜113よりなる群から選択されるアミノ酸配列を有し、
逆反転ペプチドが、配列番号5〜13のL−ペプチドの逆反転体に対応する配列番号205〜213のペプチドよりなる群から選択されるアミノ酸配列を有する
4.に記載の組成物。
Claims (13)
- 輸送ペプチドおよび前記輸送ペプチドを含んでなる伸長されたペプチドよりなる群から選択され、前記輸送ペプチドはZElan207のアミノ酸配列kkaaavllpvllaapであるd−体アミノ酸配列を含んでなる逆反転ペプチドである、
転位ペプチドを含んでなる、製薬学的有効成分の細胞中への取り込みを高めるための組成物。 - 前記輸送ペプチドが部分的に若しくは完全に環状である、請求項1に記載の組成物。
- 前記転位ペプチドが輸送ペプチドの伸長されたペプチドであり、前記輸送ペプチドはZElan207のアミノ酸配列kkaaavllpvllaapであるd−体アミノ酸配列を含んでなる逆反転ペプチドである、請求項1に記載の組成物。
- 伸長されたペプチドが、長さが100アミノ酸を超えない、請求項3に記載の組成物。
- 伸長されたペプチドが、長さが50アミノ酸を超えない、請求項4に記載の組成物。
- 転位ペプチドが輸送ペプチドであり、前記輸送ペプチドはZElan207のアミノ酸配列kkaaavllpvllaapであるd−体アミノ酸配列を含んでなる逆反転ペプチドである、請求項1に記載の組成物。
- 転位ペプチドのカルボキシル端基がアミド基を創製するように修飾されている、請求項1に記載の組成物。
- 転位ペプチドが有効成分と複合体形成して転位ペプチド−有効成分複合体を形成する、有効成分をさらに含んでなる請求項1に記載の組成物。
- 転位ペプチドが活性粒子と複合体形成して転位ペプチド−活性粒子複合体を形成する、活性粒子をさらに含んでなる請求項1に記載の組成物。
- (A)輸送ペプチドおよび前記輸送ペプチドを含んでなる伸長されたペプチドよりなる群から選択され、前記輸送ペプチドはZElan207のアミノ酸配列kkaaavllpvllaapであるd−体アミノ酸配列を含んでなる逆反転ペプチドである、転位ペプチド、(B)転位可能なペプチド、および(C)転位可能なペプチドに転位ペプチドを直接連結するアミノ酸リンカー配列を含んでなり、前記転位可能なペプチドが3と200アミノ酸との間であり、かつ、前記アミノ酸リンカー配列が1と20アミノ酸との間である、キメラポリペプチド。
- 前記転位可能なペプチドが3と30アミノ酸との間である、請求項10に記載のキメラペプチド。
- 転位可能なペプチドがオピオイドペプチドである、請求項10に記載のキメラペプチド。
- 前記リンカー配列が7アミノ酸を超えない、請求項10に記載のキメラペプチド。
