1.セレンテラミド又はその類縁体の製造方法
本発明では、セレンテラミド又はその類縁体を、O−メチルセレンテラミン又はその類縁体からジ−O−メチルセレンテラミド又はその類縁体を経由して製造する。本発明のセレンテラミド又はその類縁体の製造方法のいくつかの態様では、O−メチルセレンテラミン又はその類縁体からのセレンテラミド又はその類縁体の精製前の収率は、従来の製造方法よりも高く、例えば、55%以上、60%、65%以上、又は70%以上である。
本発明は、ジ−O−メチルセレンテラミド又はその類縁体(一般式(3)で表わされる化合物)を提供する。すなわち、本発明は、下記一般式(3)

下記式
図6に示すように、緑色蛍光蛋白質(gFP)は、EDTA等のカルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンを除去するためのキレート剤の存在下、セレンテラミド又はその類縁体(一般式(4)で表わされる化合物)に、アポイクオリン等のアポ蛋白質を反応させることにより製造することができる。また、gFPにカルシウムイオンを加えることにより、青色蛍光蛋白質(BFP)を製造することができる。
2.1.1.緑色蛍光蛋白質(gFP)の製造方法
本発明の緑色蛍光蛋白質(gFP)の製造方法において製造する緑色蛍光蛋白質(gFP)は、カルシウム結合型発光蛋白質のアポ蛋白質に、セレンテラミド又はその類縁体が配位した複合体である。gFPは、光の励起を受けて蛍光を発生することができる。
(b)天然型アポ蛋白質のアミノ酸配列に対して90%以上の同一性を有するアミノ酸配列からなり、かつ、カルシウム結合型発光蛋白質のアポ蛋白質活性若しくは機能を有する蛋白質、及び
(c)天然型アポ蛋白質の塩基配列に相補的な塩基配列からなるポリヌクレオチドとストリンジェントな条件下でハイブリダイズするポリヌクレオチドによってコードされるアミノ酸配列からなり、かつ、カルシウム結合型発光蛋白質のアポ蛋白質活性若しくは機能を有する蛋白質。
(1)レポーター蛋白質としての利用
gFPは、レポーター蛋白質としてプロモーターなどの転写活性の測定に利用することができる。例えば、アポ蛋白質をコードするポリヌクレオチドを、目的のプロモーター又は他の発現制御配列(例えば、エンハンサーなど)に融合したベクターを構築する。前記ベクターを宿主細胞に導入し、さらに、これに、セレンテラミド又はその類縁体を、カルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンを除去するためのキレート剤の存在下で接触させることで、gFPを生成させ、gFPに由来する蛍光を検出することにより、目的のプロモーター又は他の発現制御配列の活性を測定することができる。
gFPは、蛍光による検出マーカとして利用することができる。検出マーカは、例えば、イムノアッセイ又はハイブリダイゼーションアッセイなどにおける目的物質の検出に利用することができる。gFPを化学修飾法など通常用いられる方法により目的物質(蛋白質或いは核酸など)と結合させて使用することができる。このような検出マーカを用いた検出方法は、通常の方法によって行うことができる。また、本発明の検出マーカは、例えば、アポ蛋白質と目的物質との融合蛋白質として発現させ、マイクロインジェクション法などの手法により細胞内に導入し、さらに、これに、セレンテラミド又はその類縁体を、カルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンを除去するためのキレート剤の存在下で接触させることでgFPを生成させること等によって、前記目的物質の分布を測定するために利用することもできる。このような目的物質などの分布の測定は、蛍光イメージング等の検出法などを利用して行うこともできる。なお、アポ蛋白質は、マイクロインジェクション法などの手法により細胞内に導入する以外に、細胞内で発現させて用いることもできる。
gFPは、アミューズメント用品の材料の蛍光材として好適に使用することができる。アミューズメント用品としては、たとえば、蛍光シャボン玉、蛍光アイス、蛍光飴、蛍光絵の具等があげられる。アミューズメント用品は、通常の方法によって製造することができる。
2.2.1.青色蛍光蛋白質(BFP)の製造方法
前記本発明の製造方法により製造したgFPに、カルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンを加えることにより、青色蛍光蛋白質(BFP)を製造することができる。或いは、青色蛍光蛋白質(BFP)は、カルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンの存在下、前記本発明の方法により製造したセレンテラミド又はその類縁体(一般式(4)で表わされる化合物)に、カルシウム結合型アポ蛋白質を反応させることによっても製造することができる。本発明において製造する青色蛍光蛋白質(BFP)は、カルシウム結合型発光蛋白質のアポ蛋白質に、セレンテラミド又はその類縁体が配位した複合体である。BFPは、光の励起を受けて蛍光を発生することができるとともに、カルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンと反応して発光することもできる。
BFP再生に影響しなければ、BFPの製造のために用いる還元剤の量は、特に制限されないが、アポイクオリンには、3カ所のシステイン残基が存在することより、S-S 結合形成を防ぐ濃度であるのが好ましい。例えば、最終濃度が、1 mMジチオトレイトール や0.1%メルカプエタノールである。
(1)発光触媒としての利用
本発明のBFPは、発光基質に作用しそれを発光させるので、発光触媒として利用できる。そこで、本発明は、BFPに、セレンテラジン又はその類縁体を接触させることを含む、発光方法を提供する。ここで、「接触」とは、BFPとセレンテラジン又はその類縁体とを同一の反応系に存在させることを意味し、例えば、セレンテラジン又はその類縁体を収容した容器にBFPを添加すること、BFPを収容した容器にセレンテラジン又はその類縁体を添加すること、又はBFPとセレンテラジン又はその類縁体とを混合すること、などが含まれる。
BFPは、レポーター蛋白質としてプロモーターなどの転写活性の測定に利用することもできる。アポ蛋白質をコードするポリヌクレオチドを、目的のプロモーター又は他の発現制御配列(例えば、エンハンサーなど)に融合したベクターを構築する。前記ベクターを宿主細胞に導入し、さらに、これに、本発明の方法で製造したセレンテラミド又はその類縁体及びカルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンを接触させ、本発明の蛍光蛋白質に由来する蛍光を検出することにより、目的のプロモーター又は他の発現制御配列の活性を測定することができる。ここで、「接触」とは、宿主細胞とセレンテラミド又はその類縁体とカルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンとを同一の培養系・反応系に存在させることを意味し、例えば、宿主細胞の培養容器にセレンテラミド又はその類縁体及びカルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンを添加すること、宿主細胞とセレンテラミド又はその類縁体とカルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンとを混合すること、宿主細胞をセレンテラミド又はその類縁体及びカルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンの存在下で培養することなどが含まれる。
BFPは、蛍光による検出マーカとして利用することができる。本発明の検出マーカは、例えば、イムノアッセイ又はハイブリダイゼーションアッセイなどにおける目的物質の検出に利用することができる。BFPを化学修飾法など通常用いられる方法により目的物質(蛋白質或いは核酸など)と結合させて使用することができる。このような検出マーカを用いた検出方法は、通常の方法によって行うことができる。
BFPは、光の励起をうけて蛍光を生じる。よって、BFPは、アミューズメント用品の材料の蛍光基材として好適に使用することができる。アミューズメント用品としては、たとえば、蛍光シャボン玉、蛍光アイス、蛍光飴、蛍光絵の具等があげられる。本発明のアミューズメント用品は、通常の方法によって製造することができる。
図6に示すように、イクオリン等のカルシウム結合型発光蛋白質は、EDTA等のカルシウムイオン又はカルシウムイオンと置換可能な2価若しくは3価のイオンを除去するためのキレート剤の存在下、gFPに、セレンテラジン又はその類縁体を反応させることで、製造することが出来る。
本発明のカルシウム結合型発光蛋白質は、本発明のgFPから製造することができる。すなわち、本発明のカルシウム結合型発光蛋白質は、gFPに、発光基質であるセレンテラジン又はその類縁体を反応させることによって、得ることができる。
(1)カルシウムイオンの検出又は定量
本発明のカルシウム結合型発光蛋白質は、カルシウムイオンの作用によって発光する発光蛋白質(ホロ蛋白質)である。よって、本発明の発光蛋白質は、カルシウムイオンの検出又は定量に使用することができる。
本発明のカルシウム結合型発光蛋白質は、生物発光共鳴エネルギー移動(BRET)法による分子間相互作用の原理を利用した生理機能の解析や酵素活性の測定等の分析方法に利用することができる。
セレンテラミドの合成
分析用薄層クロマトグラフィー(TLC)には、あらかじめシリカゲルが塗布された(0.25mm)シリカゲルプレート(MERCK社、シリカゲル60 F254、カタログ番号1.05715.0009)を用いた。
分取カラムクロマトグラフィーには、シリカゲル(関東化学社、シリカゲル60N、球状、中性、カタログ番号37563-84)を使用した。
融点(Mp.)は、YANACO MP-J3を使用して測定した(未補正)。1H(300 MHz)核磁気共鳴スペクトル(NMRスペクトル)、および13C(75.5 MHz)NMRスペクトルは、Varian社製Gemini-300を用いてDMSO-d6(CIL社)中で測定した。1H NMRの化学シフトの基準には、測定溶媒であるDMSO-d 6中の残存非重水素化ジメチルスルホキシドのピークをδ2.49とした。13C NMRの化学シフトの基準には、測定溶媒であるDMSO-d6のピークをδ 39.7とし、それぞれ単位ppmで示した。また、結合定数(J)は単位Hzで示した。略号s、m及びbrはそれぞれ、singlet、multiplet及びbroadを表す。赤外分光(IR)スペクトルは、DRS-8000Aを備えた分光計SHIMADZU IRPrestige-21を用い、拡散反射法により測定し、吸収帯は単位cm-1で示した。高分解能質量分析(HRMS)は、電子衝撃イオン化(EI)法の条件下で、質量分析計JEOL JMS-700を用いて行った。
アルゴン雰囲気下、ピリジン中の2-アミノ-3-ベンジル-5-(4-メトキシフェニル)ピラジン(II)(セレンテラミンメチルエーテルともいう)(Kishi et al., Tetrahedron Lett., 13, 2747-2748(1972);Adamczyk et al., Org. Prep. Proced. Int., 33, 477-485(2001))(502 mg、1.72 mmol)の溶液(5 mL)に対し、4-(ジメチルアミノ)ピリジン(DMAP)(21.1 mg、172 μmol)及び4-メトキシフェニルアセチルクロリド(III)(527 μL、3.45 mmol)を続けて室温で加え、混合物を同じ温度で22.5時間攪拌した。これに対し炭酸水素ナトリウム飽和水溶液(100 mL)を加え、ジクロロメタン(50 mL×3)を用いて混合物の抽出を行った。有機抽出物を無水硫酸ナトリウムで乾燥し、ろ過し、減圧下で濃縮した。トルエン(20 mL×3)を用い、残留ピリジンを共沸して除去した。残渣をシリカゲルカラムクロマトグラフィー(85 g、ジクロロメタン/酢酸エチル=9/1)により精製し、淡黄色固体であるとしてセレンテラミドジメチルエーテル(IV)(617 mg、81.5%)を得た。酢酸エチルからの再結晶によって分析的に純粋なサンプルとして無色の固体を得た(全部で2回の再結晶により、458 mg、60.5%を得た)。
アルゴン雰囲気下、無水ジクロロメタン中、セレンテラミドジメチルエーテル(IV)(660 mg、1.50 mmol)の溶液(20 mL)を、0 ℃で10分かけて三臭化ホウ素(6.01 mL、6.01 mmol)の1.0 Mジクロロメタン溶液に加え、同じ温度で15分間、混合物を攪拌した。混合物を室温になるまで暖め、21時間攪拌し続けた。これに対し炭酸水素ナトリウム飽和水溶液(100 mL)を加え、混合物を減圧下で濃縮してジクロロメタンを除去した。残留懸濁水溶液をろ過し、回収した固体を真空で乾燥させ、淡黄色固体としてセレンテラミド(I)(570 mg、92.3))%)を得た。この純度のサンプルは、gFP及びBFPなどの製造において十分に利用することができる。
エタノールからの再結晶によって分析的に純粋なサンプルとして無色の固体を得た(103 mg、16.7%)。
細菌細胞由来の組換えヒスチジン標識アポイクオリンの発現及び精製
組換えヒスチジン標識アポイクオリンは、大腸菌のペリプラズムに発現させた。使用した宿主株は、大腸菌WA802株(CGSC 5610)である。大腸菌WA802株に導入した組換えヒスチジン標識アポイクオリン発現ベクターpiP-His-HEを、piP-HEΔ2Eから構築した(Inouye & Sahara, Protein Express. Purifi. (2007) 53: 384-389 参照)。
蛋白質分析
市販のキット(Bio-Rad社、カリフォルニア州リッチモンド)及び基準としてのウシ血清アルブミン(Pierce社、イリノイ州ロックフォード)を用い、Bradfordの色素結合法(Anal. Biochem. (1976)72, 248-254)によって、蛋白質の濃度を決定した。
また、Laemmli(Nature (1970) 227, 680-685)の記載の通りに、12%分離ゲル(テフコ株式会社、東京)を用い、還元性条件下でSDS-PAGE分析を行った。その結果、図1に示すように、本精製法によって得られた精製アポイクオリンの純度は95%以上であることが明らかとなった。
セレンテラミド及びアポイクオリンからの合成BFPの調製
10mM CaCl2及び1mM DTTを含む1mlの50mM Tris-HCl(pH 7.6)中、アポイクオリン(0.5mg、22nmol)及び10μlのセレンテラミド(無水メタノール中1.2μg/μl、29nmol)を混合し、この混合物を4℃で16時間静置することにより、合成BFP(syn-BFP)を調製した。続いて、遠心濃縮器Vivaspin 2(10,000 MWCO、Sartorius AG、ドイツ)を用いて4℃、5,000 x gで20分間処理し、混合物を0.1 mlまで濃縮した。濃縮された溶液は、長波UVランプ(366nm)の下で強い青色発光を示した。
アポイクオリン及びセレンテラミドからの合成gFP (syn-gFP)の調製に関しては、10mM EDTA及び 1mM DTTを含む1mlの50mM Tris-HCl(pH 7.6)中、アポイクオリン(0.5mg、22nmol)及び10μlのセレンテラミド(無水メタノール中1.2μg/μl、29nmol)を混合し、この混合物を4℃で16時間静置することにより、合成gFP(syn-gFP)を調製した。335nmで励起することにより、図2に示す蛍光発光スペクトルを得た。アポイクオリンとセレンテラミドにより、蛍光能を有するsyn-gFP の生成が確認された。
一方、syn-gFP及びsyn-BFP 調製溶液に、還元剤であるDTTが添加することにより、表2にまとめたように、syn-gFP及びsyn-BFPを短時間に、効率良く調製することができることが明らかとなった。
吸収スペクトル及び蛍光スペクトルの測定
Jasco(日本分光株式会社、東京)のV-560分光光度計(帯域幅:0.5nm、レスポンス:中程度、スキャン速度:100 nm/分)により、石英セル(光路長10mm)を用いて25℃で吸収スペクトルを測定した。また、JascoのFP-6500蛍光分光光度計(発光/励起帯域幅: 3 nm、レスポンス:0.5 sec、スキャン速度:1000nm/分)を用いて蛍光スペクトルを測定した。
得られたsyn-gFPの蛍光スペクトルは、図3に示すように、イクオリンより調製したgFPのスペクトルと一致した。また、表3に示すように、調製したsyn-BFPの吸収スペクトルにおける280 nmと330 nm の比率は、イクオリンより調製したBFPとほぼ同じかそれ以上の比を示すことから、同等物が作ることが可能となった。
発光活性のアッセイ
反応混合物(100μl)は、50mM Tris-HCl(pH7.6)-10mM CaCl2中にセレンテラジン(0.5μg、1μlのエタノール中に溶解)を含有していた。蛋白質溶液を加えることにより反応を開始させ(1μgのBFP及びsyn-BFP)、浜松ホトニクス社製のR4220P光電子増倍管を備えたアトー株式会社(東京)製のAB2200照度計を用いて、発光強度を1分間記録した。1ngの精製組換えイクオリンの最大強度(Imax)は、8.8×105 rlu(relative light units:相対発光量)を示した。
合成セレンテラミドを用いて調製したsyn-BFPは、図4に示すように、イクオリンより調製したBFPと同等の発光活性を有していた。

- 下記一般式(1)

R2は、アミノであり、
R3は、脂肪族環式基によって置換されていてもよい炭素数1〜7のアルキル、脂肪族環式基、又は炭素数7〜10のアリールアルキルである。)
を、
下記一般式(2)

Xは、脱離基であり、
R4は、炭素数1〜3のアルキル、又は炭素数7〜10のアリールアルキルである。)
に反応させることを含む、
下記一般式(3)
