Abstract
【解決手段】本発明の油脂生産菌の培養方法は、(1) 糖類濃度10%〜30%の培地で油脂生産菌を培養すること、又は(2) 糖類を含有し、かつ糖類及び塩類の合計モル濃度が0.60 mol/L〜1.16 mol/Lである培地で油脂生産菌を培養することを含む。
【効果】本発明によれば、培地に所定量の糖類ないしは糖類及び塩類を添加して培養するという簡便な方法により、油脂生産菌による油脂の生産量を高めることができる。

Description
(1) 培地の糖類濃度を10%以上に高めると、油脂生産菌の油脂生産効率が向上する。
(2) 培地の糖濃度が低くても、塩類を追加して培地のモル濃度を高め、浸透圧を高めると、油脂生産菌の油脂生産効率が向上する。
油脂生産菌として、Lipomyces属酵母を使用し油脂生産を試みた。培地としては、酵母エキス及びポリペプトンから構成されるYEPD培地(1% Yeast Extract, 2% polypeptone)と、酵母エキス及び無機成分から構成されるS培地を使用した。
油脂蓄積の向上に関わる更なる因子の解明を目指し、培養液の浸透圧に注目し研究を行った。
前培養したLipomyces属酵母を、15%又は20%キシロースを含むS培地(100ml)に植菌し、25℃にて振とう培養を行った。経時的に培養液からサンプルを採取し、消費した糖量および油脂生産量を調べた。さらに、培養液あたりの乾燥菌体重量も測定し、菌体内油脂含量を算出した。その結果、いずれの培地においてもにおいても、10日間の培養で、油脂含量は60%以上の数値を示し、対糖収率についても14%以上、1mlの培養液あたり約10mg以上の油脂の生産が可能であった。これらの数値は、実施例1、2に示すグルコースを用いた場合の油脂生産とほぼ同等の結果であった。

- 糖類濃度10%〜30%の培地で油脂生産菌を培養することを含む油脂生産菌の培養方法。
- 前記糖類濃度が15%〜25%である請求項1記載の方法。
- 糖類を含有し、かつ糖類及び塩類の合計モル濃度が0.60 mol/L〜1.16 mol/Lである培地で油脂生産菌を培養することを含む油脂生産菌の培養方法。
- 前記合計モル濃度が0.88 mol/L〜1.16 mol/Lである請求項3記載の方法。
- 前記培地の糖類濃度が3%〜20%である請求項3又は4記載の方法。
- 前記培地の糖類濃度が3%〜10%である請求項5記載の方法。
- 前記塩類は、硫酸アンモニウム、リン酸二水素カリウム、硫酸マグネシウム及び塩化ナトリウムからなる群より選択される少なくとも1種である請求項3ないし6のいずれか1項に記載の方法。
