Abstract
【解決手段】ガラクトサミン又はその誘導体を利用することを特徴とする。ガラクトサミン又はその誘導体は、トリプシンによる、トリプシノーゲンからトリプシンへの分解(トリプシノーゲンの活性化)を抑制する効果を有しているため、トリプシノーゲン活性化抑制剤や膵炎の予防・治療剤として用いることができる。

Description
(1)材料
本実験には以下の材料を用いた。なお、カッコ内にはコード番号を示す。
ウシ膵臓トリプシノーゲン(BPTG)は、MP Biomedicals, Inc.製のもの(101195)を用い、ウシ膵臓トリプシン(BPT)は、和光純薬株式会社製のもの(204-13951)を用い、Phenylmethylsulfonyl Fluoride(PMSF)は、Nacalai Tesque, INC.製のもの(27327-52)を用い、Methyl-α-D-galactopyranoside(Me−α−Gal)は、Sigma-Aldrich Corporation製のもの(M1379-25MG)を用い、1-O-Methyl-β-D-galactopyranoside(Me−β−Gal)は、Nacalai Tesque, INC.製のもの(22526-34)を用い、Methyl-α-D-Mannopyranoside(Me−α−Man)は、和光純薬株式会社製のもの(131-08021)を用い、Methyl-N-acetyl-2-deoxy-α-D-glucosaminide(Me−α−GlcNAc)は、Sigma-Aldrich Corporation製のもの(M0257-100MG)を用い、Methyl-N-acetyl-2-deoxy-α-D-galactosaminide(Me−α−GalNAc)は、Toronto Research Chemicals, Inc.製のもの(M275310)を用い、D−ガラクトサミン塩酸塩(GalN・HCl)は、生化学工業製のもの(Lot.S6701)を用い、N−アセチル−D−ガラクトサミン(GalNAc)は、Toronto Research Chemicals Inc.製のもの(A17700)を用い、L−フコース(L−Fuc)は、Fluka AG, Chem. Fabrik製のものを用い、D−フコース(D−Fuc)は、和光純薬株式会社製のものを用いた。
トリプシンによるトリプシノーゲンからトリプシンへの分解(トリプシノーゲンの活性化)に対して、糖がどのような影響を与えるかを調べるために、BPTGをBPT及び糖存在下でアッセイした場合の、BPTG量やBPT量の変化を、SDS−PAGEによる分子量の変化によって確認した。具体的には、以下の方法で行なった。
BPTG、BPT、及び上記1.(1)記載の9種類の各糖を、それぞれ別のエッペンチューブに分取し、さらにそれぞれのエッペンチューブにTBS溶液[10mM Tris−HCl(pH7.5),150mM NaCl]を添加して、5.0mg/mlのBPTG溶液、3.7mg/mlのBPT溶液、及び0.3Mの各糖溶液を調製した。
(1)材料
本実験には以下の材料を用いた。
BPTG、BPT及びPMSFは、実施例1で用いたものと同様のものを用い、糖-biotinylated polyacrylamide probes(糖−BP プローブ)は、GlycoTech Co.(Gaithersburg, Maryland 20879)製のもの(ポリアクリルアミド骨格の−CONH2基に、短いスペーサー(−OCH2CH2CH2−)を介して糖をグリコシド結合で共有結合させたプローブ;例えばα−マンノースBPプローブについて図4参照)を用い、avidin-biotin-horseradish peroxidase complex(ABC−HRP)は、SIGMA社製のもの(S5512)を用い、o-phenylene diamine(OPD)は、和光純薬株式会社製のもの(161-11851)を用いた。
実施例1で示された、トリプシノーゲンからトリプシンへの分解(トリプシノーゲンの活性化)に対する糖の抑制効果が、BPTGやBPTの糖結合性と関連があるかどうかを調べるために、以下のようなELISA法による糖結合性分析を行なった。
BPTG、BPTをそれぞれ3mg計り取り、2.94mlの10mM TBS(pH7.5)と60μlの0.2M PMSF溶液を加えて溶解し、4℃で1時間、プレインキュベーションして、1mg/mlのBPTG溶液、及び、BPT溶液を調製した。これらの溶液を10mM TBS(pH7.5)で0.1〜15μg/mlに希釈し、マイクロタイタープレート(Immulon 1B,Thermo Scientific社製)の各ウエルにそれぞれ100μlずつ添加した。4℃、2時間インキュベーションすることによりBPTG及びBPTをプレートに固定化した。各300μlの10mM TBS(pH7.5)で各ウエルを3回洗浄した後、各300μlの3% BSA(10mM TBS,pH7.5)を添加して、4℃で一晩ブロッキングした。10mM TBS(pH7.5)に、各糖(α−Man、α−Man−6−リン酸、α−Gal、α−GalNAc、α−NeuAc、β−Gal又はLac)−BP プローブ又は糖を持たないPAAプローブを10μg/mlとなるように加えた溶液を100μlずつ各ウエルに添加して、1時間室温で反応させた。各300μlの10mM TBS(pH7.5)で各ウエルを3回洗浄した後、各100μlの0.7μg/ml ABC−HRPを添加して、室温で1時間結合反応させた後、各300μlの10mM TBS(pH7.5)で各ウエルを3回洗浄した。次に、各200μlの発色液[OPD 8mg,50mMクエン酸リン酸バッファー(pH5.0)20ml,30%H2O2 8μl]を各ウエルに添加して、室温で25分間発色させた後、各50μlの2.5N H2SO4を各ウエルに添加して発色を停止させた。マイクロプレートリーダー(MPR−80,Bio-Rad社製)を用いて、490nmの吸光度を測定した。以上の結合実験を、TBSの代わりに10mM 酢酸−酢酸ナトリウム緩衝液(pH5.5)を用いても、全く同様の手順で行った。
図4からわかるように、BPTGとBPTは、α‐Manを持つ糖BP−プローブには糖を持たないPAAプローブより高い結合性を示し、α−GalNAcを持つ糖−BPプローブには、顕著に高い結合性を示した。また、BPTGについて別ロットの製品を用いて同様の実験を行なったが、同様の結果であった。
SPRを用いて、BPTGと糖との相互作用解析を行なった。操作は全てBIACORE2000(GE Healthcare Bio-Sciences KK)を用い、付属のマニュアルに従って操作を行った。リガンドの固定化はアミンカップリング法を用いて、10μl/minで各ステップを14分間ずつ行った。ランニング緩衝液には、HBS−EP bufferに終濃度0.2mMになるように0.2M PMSF−エタノール溶液を加えて使用した。CM5センサーチップをランニング緩衝液で平衡化した後、アミンカップリングキットにより、チップ表面をEDC/NHSで活性化した。リガンドに用いたBPTGやBPTは、あらかじめ2mg秤りとった後、960μlの10mM CH3COOH−CH3COONa buffer(pH6.0)に溶解し、0.2M PMSF−エタノール溶液を40μl加え(終濃度8mM)、4℃、1時間振盪した(プレインキュベーション)。これを10mM CH3COOH−CH3COONa buffer(pH6.0)に、保護糖としてMe−α−Man、Me−α−Gal又はD−GalNAcを終濃度が各0.2 Mになるように加えた緩衝液を用いて10μg/mlに希釈した後、センサーチップにインジェクトして固定化を行った。残っているN-hydroxysuccinimide esterを1.0M エタノールアミン・HCl(pH 8.0)でブロックした後、10mM HClで1分間洗浄してから測定に使用した。固定化されたBPTGは3743.6RU、BPTは8014.9RUであった。リファレンスセル(対照セル)には、タンパク質溶液を固定化せずに、同様のEDC/NHS活性化と1.0M エタノールアミン・HCl(pH8.0)によるブロッキングを行ったフローセルを用いた。アナライトには、Me−α−Man,Lac,Me−α−GalNAc,Me−β−Galを、それぞれ0.05,0.1,0.15,0.2,0.25,0.3Mの濃度になるように、ランニング緩衝液を用いて調製した。アナライトの糖類を含むランニング緩衝液をBPTG固定化センサーチップ上に20μL/minの流速で90秒間インジェクトし、次にTBSで溶出を行った。この実験の結果を図7に示す。

BPT(Bovine pancreatic trypsin)は、Sigma-Aldorich社製のT1426を用い、BAPA(N-α-Benzoyl-DL-arginine-p-nitroanilide hydrochloride)は、株式会社ペプチド研究所製のものを用い、Me−α−Man(Methyl-α-mannoside)は、Sigma-Aldorich社製のものを用い、GalN・HCl(D-Galactosamine hydrochloride)は、生化学工業株式会社製のものを用い、GalNAc(N-Acethyl-D-galactosamine)は、和光純薬工業株式会社製の013-12821を用い、Me−α−GalNAc(o-Methyl-N-Acetyl-2-deoxy-D-galactosamine)は、Tronto Research Chemicals Inc.社製のM275310を用いた。
各種の糖について、基質濃度を変化させてBPTの酵素反応速度を測定し、Lineweaver-Burk plotにより解析して、各種の糖のトリプシン酵素活性に与える影響について調べた。
トリプシンの酵素活性測定には、基質としてBAPAを用いた。BAPAを14.2mg秤量し、1065μlのDMSOに完全に溶解した後、0.1M Tris−HCl buffer(pH7.9)−20mM CaCl2−30mM NaCl(TBS−Ca)を2200μl加えて、あらかじめ10mM BAPA溶液を調製した。次いで、このBAPA溶液をさらに希釈して0mMから3mMの各濃度の基質BAPA溶液を調製した。BPTは2.4mgを秤量し、酸性で安定であるため、1mM HCl−20mM CaCl2溶液0.5mlに溶解し、200μMのBPT溶液を調製した。このBPT溶液からさらに希釈して6μMのトリプシン酵素溶液を調製した。Me−α−Man、GalN・HCl、GalNAc、Me−α−GalNAc についてTBS−Caを加えて、1mM から0.2Mの各糖溶液を数段階調製した。糖溶液140μlと6μM トリプシン溶液10μlをあらかじめ混合して15分間37℃でプレインキュベートした。マイクロプレートに、37℃でプレインキュベートした各濃度の基質BAPA溶液を1ウエルあたり150μl加え、次に、37℃でプレインキュベートした糖−トリプシン混合溶液を1ウエルあたり150μlずつ加えた。素早く撹拌後、マイクロプレートリーダーVient X(DS Pharma Biomedical)を用いて、30秒おきに5分間410nmで吸光度を測定した。基質濃度の逆数1/[S]を横軸に、反応速度の逆数1/[v]を縦軸にとり、Lineweaver Burk Plotにより解析して、各種の糖のBPT酵素活性に与える影響について考察した。その結果を図8に示す。
(1)材料
本実験には以下の材料を用いた。
BPTG、BPT、糖及びPMSFは、実施例1と同様の物を用い、アプロチニンはウシ膵臓由来の物をSIGMA社から購入して用い、NHS−ビオチン(PIERCE社製)を用いてマニュアル通りにビオチン標識して使用した。ELISAに必要な試薬は実施例2と同様の物を用いた。
膵臓由来の内在性トリプシン阻害剤として知られているアプロチニン(BPTI)との結合が、BPTGやBPTの糖結合性に影響があるかどうかを調べるために、ELISA法による結合性分析を行なった。
実施例2と同様にして、PMSF処理を行ったBPTG溶液、及び、BPT溶液を調製し、これらの溶液を10mM TBS(pH7.5)で希釈し、マイクロタイタープレート(Immulon 1B,Thermo Scientific社製)の各ウエルに添加して、BPTG及びBPTをプレートに固定化した。実施例2と同様にしてTBS(pH7.5)を緩衝液に用いて洗浄した後、BSAによるブロッキングを行った。0.1Mの各糖(Me−α−Man、Me−β−Gal又はGalN・HCl)のTBS(pH7.5)溶液を100μlずつ各ウエルに添加して、1時間室温で反応させた。各300μlの緩衝液で各ウエルを3回洗浄した後、各100μlのビオチン化アプロチニン10μg/mlを添加して、室温で1時間結合反応させた。その後、各300μlの緩衝液で3回洗浄した後、各100μlの0.7μg/mlのABC−HRPを添加して、室温で1時間結合反応させた後、各300μlの緩衝液で3回洗浄した。次に、各200μlの発色液[OPD 8mg,50mMクエン酸リン酸緩衝液(pH5.0)20ml,30%H2O2 8μl]を各ウエルに添加して、室温で25分間発色させた後、50μlの2.5N H2SO4を添加して発色を停止させた。各ウエル中の液体について、マイクロプレートリーダー(MPR-80,Bio-Rad社製)を用いて、490nmの吸光度を測定した。その結果を図9に示す。
