Abstract
Description
本願は、2006年5月15日に出願された米国仮出願第60/747, 220号および2006年10月6日に出願された米国仮出願第60/849,968号の優先権を主張する。上記の出願の全教示は、本明細書において参考として援用する。
本発明は、腫瘍細胞の増殖の阻害に関する。より具体的には、本発明は、CDKI経路の阻害を介した腫瘍細胞増殖の阻害に関する。
プログラムされた細胞周期の停止は、損傷に対する応答、成長因子欠乏、接触阻止、有糸分裂後の細胞の最終分化、および老化などの、多様な生理学的状況において起こる。Vidal and Koff, Gene 247: 1−15 (2000)は、これらの状況の全てに、サイクリン依存性キナーゼ阻害剤(CDKI)タンパク質の一時的および/または恒常的な上方調節が関連することを教示する。CDKIは、細胞周期の異なる期の間の遷移を媒介するサイクリン/サイクリン依存性キナーゼ(CDK)複合体を阻害することにより、細胞周期停止を誘導する。CDKIタンパク質は、Cip/KipまたはInk4タンパク質ファミリーに属する。Roninson, Cancer Letters 179: 1−14 (2002)は、これらのタンパク質のうち最も多面発現性のものは、p21Waf1/Cip1/Sdi1であり、これは、異なるサイクリン/CDK複合体を阻害し、損傷により誘導されるチェックポイント停止、老化および最終分化において役割を果たすことを教示する。Sharpless, Experimental Gerontology 39: 1751−1759 (2004)は、他のCDKIの中で、多様な癌においてしばしば不活化される腫瘍抑制因子であるCDK4/6阻害剤p16Ink4aが、老化細胞における細胞周期停止の維持の主要な調節因子であることを教示するが、一方、上記のVidal and Koffは、CDK2阻害剤p27Kip1が、接触阻止の重要なメディエーターであることを教示する。
本発明者らは、以下のCDK関連活性を有するいくつかの化合物を同定した。これらの化合物は、p21Waf1などのCDK阻害タンパク質の発現への応答における複数の遺伝子の転写の誘導として定義されるCDKI経路を阻害する。これらのCDKI経路阻害剤は、SNX9クラス化合物と称され、また、正常細胞に対して優先的に異なる型の腫瘍細胞の増殖を阻害する所望の能力を有する。

CDKI経路阻害剤の同定
同時係属の特許出願PCT/US06/01046において記載するように、本発明者らは、CDKI経路を阻害する化合物のためのハイスループットスクリーニング(HTS)法を開発した。この方法は、生理学的に中性(neutral)のβガラクトシドIPTG(イソプロピル−β−チオガラクトシド)によって誘導されるプロモーターからp21を発現するHT1080線維肉腫細胞の誘導体であるHT1080 p21−9細胞を、CDKI誘導性サイトメガロウイルス(CMV)プロモーターから緑色蛍光タンパク質(GFP)を発現するレンチウイルスベクターで感染させ、その後、GFP陽性細胞をサブクローニングし、IPTGによるGFP発現の誘導をモニタリングすることにより作出した、高感度のレポーター細胞株を利用する。IPTGの添加によりおよそ10倍のGFPの増大を示す細胞株を、96ウェルのフォーマットにおけるHTSのために用いた。
CDKIにより誘導される転写に対する化合物の効果
図1は、IPTG(p21誘導因子)の存在下または不在下における、レポーター細胞株における正規化されたGFP発現に対するSNX9の効果を示す。化合物は、p21により顕著な転写の阻害を示すが、p21が誘導されない場合、プロモーター機能を阻害せず、このことは、その転写効果が、CDKIにより誘導される転写に特異的であることを示す。同様の結果を、SNX9−1ならびに化合物3および4によって得た。図2における実験は、SNX9はまた、p21により誘導される転写を転写を逆転することを示す。この実験において、細胞のNK4遺伝子のCDKI応答性プロモーターからホタルルシフェラーゼを発現するHT1080 p21−9細胞を、IPTGと共に2日間培養した。これは、最大に近いNK4の誘導に十分である(Poole et al.、上記)。
SNX9クラス化合物による腫瘍特異的増殖阻害
驚くべきことに、CDKI経路阻害剤のスクリーニングにおいて同定された62個の化合物の大部分が、HT1080により誘導されるレポーター細胞株の顕著な増殖阻害を示した。SNX9、SNX9−1ならびに化合物3および4の場合、細胞増殖阻害は、細胞死(細胞の剥離により顕微鏡で検出される)および細胞周期停止の両方の誘導と関連した。後者を図6に示し、これは、未処理か、またはSNX9単独、IPTG(p21を誘導する)単独、もしくはSNX9およびIPTGの組合せにより処理されたHT1080 p21−9細胞のDNA含有量のFACS分析を示す。SNX9処理は、G2/M画分の顕著な増大を誘導した。
マイクロアレイ分析はCDKI経路阻害剤のCDKI様活性を示唆する
IPTG誘導性のCDKIであるp21を有するHT1080 p21−9細胞株を、未処理か、または、72時間100μMのIPTG(これはp21を誘導する)で、もしくはCDKI経路阻害剤SNX9−1(20μM、SNX9ファミリー)もしくはSNX14(80μM、SNX9とは関連しない)単独もしくはIPTGと組み合わせて、処理した。各々の処理の後で、RNAを抽出し、本質的に全てのヒト遺伝子に対応する56,000個のプローブセットを含有するAffymetrix U133 2.0 Plusマイクロアレイによるハイブリダイゼーションに用いた。マイクロアレイのデータを、Gene Spring software(Agilent)を用いて分析した。図10は、2群の遺伝子の発現の変化を表す。第1群(p21により誘導された遺伝子、上段のパネル)は、p21誘導性IPTGによる処理の後で少なくとも2倍誘導された遺伝子に対応する1124個のプローブセットを表す。
CDKI経路阻害剤は、直接的なCDK阻害剤活性を有する
上記の仮説を試験するために、本発明者らは、2種のSNX9クラス化合物(SNX9およびSNX9−1)、2種の異なる構造的クラスの化合物(SNX14およびSNX2)ならびに別の関連しない阻害剤(SNX35)を含むいくつかのCDKI経路阻害剤が、精製されたサイクリン/CDK複合体によりインビトロで形成される異なる複合体のキナーゼ活性を阻害するか否かを決定した。この分析は、Upstate Biotechnology, Inc.によるサービスとして行った。
25μlの最終容積において、CDK1/サイクリンB(h)(5〜10mU)を、8mMのMOPS pH7.0、0.2mMのEDTA、0.1mg/mlのヒストンH1、10mMのMgAcetate、および[γ−33P−ATP](特異的活性約500cpm/pmol、45μMの濃度)とともにインキュベートする。反応を、MgATPミックスの添加により開始する。40分間室温にてのインキュベーションの後で、5μlの3%リン酸溶液の添加により反応を停止する。次いで、10μlの反応を、P30フィルターマット(filtermat)の上にスポットし、75mMのリン酸中で5分間3回、メタノール中で1回洗浄し、その後乾燥してシンチレーション計数に供する。

CDK1、CDK2またはCDK4の選択的阻害剤の増殖阻害効果は、SNX9クラス化合物の腫瘍選択性を示さない
本発明者らは、先に、SNX9クラス化合物が、多数の異なる腫瘍細胞株の増殖を、正常細胞(ヒト線維芽細胞および正常乳腺上皮細胞)と比較して優先的に阻害することを実証した。SNX9クラス化合物の腫瘍特異的増殖阻害活性が特定のCDKの阻害に起因し得るかを決定するために、本発明者らは、CDKタンパク質のいくつかについて選択的阻害剤のアベイラビリティの平均を得た。

SNX9に関連しないCDK3の特異的阻害剤を探索するために、本発明者らは、RNA干渉(RNAi)機構を介してCDK3発現を阻害するショートヘアピンRNA(shRNA)配列を発現する組み換えレンチウイルスベクターを用いることを選択した。CDK3を標的とする異なるshRNA配列を含む3種のレンチウイルスベクターを、Open Biosystems、Huntsville、ALから入手した。これらのベクターは、pLKO1骨格からなり(www.openbiosystems.com)、これは、ピューロマイシン耐性についての選択マーカーを有し、ヒトU6プロモーターからshRNAインサートを発現する。CDK3標的化shRNA配列を、表3に列記する。
SNX9およびSNX9−1のインビボでの研究
約6〜8週齢の雄のNCrヌードマウスを、Taconic Biotechnology、Rensselaer、NYによるサービスとして行われた研究のために用いた。動物を、ウイルスフリーのバリア条件下に維持し、継続的に健康状態をモニタリングした。試験材料の投与、全てのデータ収集および研究動物の処分を、全ての適切なTaconic Biotechnology Standard Operating ProceduresならびにThe Guide for the Care and Use of Laboratory Animalsによるコンプライアンスにおいて行った。研究動物を、到着時から研究を通して毎日、全体的な健康状態、行動および罹患率について観察した。試験動物の体重を測定し、試験物の投与量を決定した。
Claims (40)
- 腫瘍細胞の増殖を阻害する方法であって、腫瘍細胞の集団を、構造I:
を有する化合物、または、その遊離の酸、塩、もしくはプロドラッグ;
と接触させることを含む、前記方法。 - R1が水素またはメチルであり、R2およびR3が、独立して、水素、メチルおよびC2−C6アルキルまたはアルケニルから選択される、請求項1に記載の方法。
- 1または2以上の電子吸引性基が、ハロゲン、窒素含有基、または酸素含有基から選択される、請求項1に記載の方法。
- 1または2以上のハロゲンがフッ素でない、請求項3に記載の方法。
- 1または2以上のハロゲンが塩素および臭素から選択される、請求項3に記載の方法。
- 腫瘍細胞の増殖を阻害する方法であって、腫瘍細胞の集団を、構造II:
を有する化合物、または、その遊離の酸、塩、もしくはプロドラッグと接触させることを含む、前記方法。 - R1が水素またはメチルであり、R2およびR3が、独立して、水素、メチルおよびC2−C6アルキルまたはアルケニルから選択される、請求項6に記載の方法。
- 1または2以上の電子吸引性基が、ハロゲン、窒素含有基、または酸素含有基から選択される、請求項6に記載の方法。
- 1または2以上のハロゲンがフッ素でない、請求項8に記載の方法。
- 1または2以上のハロゲンが塩素および臭素から選択される、請求項8に記載の方法。
- 腫瘍を有する患者を処置するための方法であって、該患者に、構造I:
を有する化合物、または、その遊離の酸、塩、もしくはプロドラッグ、および薬学的に受容可能な希釈剤、キャリアもしくは賦形剤を含む医薬製剤を投与することを含む、前記方法。 - R1が水素またはメチルであり、R2およびR3が、独立して、水素、メチルおよびC2−C6アルキルまたはアルケニルから選択される、請求項12に記載の方法。
- 1または2以上の電子吸引性基が、ハロゲン、窒素含有基、または酸素含有基から選択される、請求項12に記載の方法。
- 1または2以上のハロゲンがフッ素でない、請求項14に記載の方法。
- 1または2以上のハロゲンが塩素および臭素から選択される、請求項14に記載の方法。
- 腫瘍を有する患者を処置するための方法であって、該患者に、構造II:

- 細胞におけるCDKI経路により誘導される転写を妨害または低減するための方法であって、該細胞を、構造I:
を有する化合物、または、その遊離の酸、塩、もしくはプロドラッグ、および薬学的に受容可能な希釈剤、キャリアもしくは賦形剤と接触させることを含む、前記方法。 - R1が水素またはメチルであり、R2およびR3が、独立して、水素、メチルおよびC2−C6アルキルまたはアルケニルから選択される、請求項24に記載の方法。
- 1または2以上の電子吸引性基が、ハロゲン、窒素含有基、または酸素含有基から選択される、請求項24に記載の方法。
- 1または2以上のハロゲンがフッ素でない、請求項26に記載の方法。
- 1または2以上のハロゲンが塩素および臭素から選択される、請求項26に記載の方法。
- 腫瘍細胞の増殖を選択的に阻害するための方法であって、腫瘍細胞において、3型サイクリン依存性キナーゼ(CDK3)を選択的に阻害することを含む、前記方法。
- 腫瘍細胞においてCDK3を選択的に阻害することが、腫瘍細胞を、CDK3活性の低分子特異的阻害剤またはCDK3のドミナントネガティブ型変異体と接触させることを含む、請求項29に記載の方法。
- CDK3の低分子特異的阻害剤が、構造Iまたは構造IIを有する、請求項30に記載の方法。
- 腫瘍細胞においてCDK3を選択的に阻害することが、腫瘍細胞を、CDK3遺伝子発現の阻害剤と接触させることを含む、請求項29に記載の方法。
- CDK3遺伝子発現の阻害剤が、ショートヘアピンRNA(shRNA)、低分子干渉RNA(siRNA)、アンチセンス核酸(AS)、およびリボザイムからなる群より選択される、請求項32に記載の方法。
- 腫瘍細胞増殖の特異的阻害剤を同定するための方法であって、CDK3と相互作用する精製されたサイクリンとCDK3とのインビトロ複合体を、CDK3と相互作用する精製されたサイクリンとCDK3との複合体がキナーゼ活性を示すことができる条件下において、かかる活性の候補阻害剤と接触させること、かかる複合体のキナーゼ活性を、かかる候補化合物の存在下または不在下において測定すること、を含み、ここで、候補化合物は、候補阻害剤の存在下におけるサイクリン/CDK3複合体の活性が、候補阻害剤の不在下においてより低い場合、腫瘍細胞増殖の特異的な阻害剤としてみなされる、前記方法。
- 請求項34に記載の方法であって、CDK1、CDK2、CDK4またはCDK6と、かかるCDKと相互作用するサイクリンとの複合体を用いることをさらに含み、ここで、候補阻害剤は、サイクリン/CDK3複合体の活性を、CDK1、CDK2、CDK4またはCDK6とそれらの相互作用するサイクリンとの複合体のキナーゼ活性より高い程度まで阻害する場合、腫瘍細胞増殖の特異的な阻害剤としてみなされる、前記方法。
- 請求項34または35に記載の方法により同定される、腫瘍細胞増殖の特異的阻害剤。
- 細胞においてCDKI経路により誘導される転写を低減または妨害するための方法であって、細胞を、構造II:
を有する化合物、または、その遊離の酸、塩、もしくはプロドラッグ、および薬学的に受容可能な希釈剤、キャリアもしくは賦形剤と接触させることを含む、前記方法。 - R1が水素またはメチルであり、R2およびR3が、独立して、水素、メチルおよびC2−C6アルキルまたはアルケニルから選択される、請求項37に記載の方法。
- 1または2以上の電子吸引性基が、ハロゲン、窒素含有基、または酸素含有基から選択される、請求項37に記載の方法。
- 1または2以上のハロゲンがフッ素でない、請求項46に記載の方法。
- 1または2以上のハロゲンが塩素および臭素から選択される、請求項39に記載の方法。
