Claims (19)

  1. a.処理される細胞の一群を提供する工程;
    b.細胞を試験物質と共にインキュベートする工程;
    c.群内の細胞の画像を取得する工程;
    d.画像を解析して、細胞の機能分類を示す細胞の特徴を測定または検出する工程;
    e.少なくとも6つの細胞の特徴を含む応答プロフィールを作成する工程;
    f.試験物質の応答プロフィールを、既知の生物学的システム効果を伴う物質についての応答プロフィールのデータベースと比較する工程であって試験物質の応答プロフィールと、既知の生物学的システム効果を伴う物質についての応答プロフィールのデータベースとの間の関連性の程度が、試験物質が生きている細胞、組織または生命体内で生物学的システム効果を発揮するであろう可能性を示す工程
    を含む、試験物質の生物学的システム効果を予測するための方法。
  2. 前記細胞の特徴が、細胞増殖、ストレス経路、オルガネラの機能、細胞周期の状態、形態、アポトーシス、DNAの損傷、代謝、シグナル伝達、細胞の分化および細胞−細胞の相互作用からなる群より選択される2つ以上の細胞の能分を示す、請求項1に記載の方法。
  3. 前記細胞の特徴が、細胞増殖、細胞周期、アポトーシス、酸化的ストレス、ストレスキナーゼの活性化、ミトコンドリアの機能、DNAの損傷およびペルオキシソームの増殖からなる群より選択される2つ以上の機能的な応答の分類を示す、請求項1に記載の方法。
  4. 前記細胞の特徴が、6つ以上の細胞の機能分類を示す、請求項1に記載の方法。
  5. 前記細胞の特徴が、ストレス経路の活性化、オルガネラの機能、細胞周期、形態、アポトーシスおよびDNAの損傷を含む6つ以上の機能的な応答の分類を示す、請求項4に記載の方法。
  6. 前記細胞の特徴が、細胞周期、アポトーシス、細胞骨格、神経毒性、DNAの損傷および転写因子の活性化を含む6つ以上の機能的な応答の分類を示す、請求項4に記載の方法。
  7. a.処理される細胞の一群を提供する工程;
    b.細胞を第一の参照物質と共にインキュベートする工程;
    c.群内の細胞の画像を取得する工程;
    d.画像を解析して、細胞の機能分類を示す細胞の特徴を測定または検出する工程;
    e.少なくとも6つの細胞の特徴を含む応答プロフィールを作成する工程;
    f.第一の参照物質についての応答プロフィールをデータベースに追加する工程;および
    g.必要に応じて、第一の参照物質の代わりに第二の参照物質を用いて、工程a〜fを繰り返す工程
    を含む、既知の生物学的システム効果を伴う参照物質についての応答プロフィールのデータベースを構築するための方法。
  8. 画像を取得する前に、細胞が一つ以上の蛍光レポーターまたは発光レポーターを含む、請求項1〜7のいずれかに記載の方法。
  9. 細胞が一つ以上の蛍光レポーターまたは発光レポーターを発現する、請求項に記載の方法。
  10. 一つ以上の蛍光レポーターまたは発光レポーターが細胞内に導入される、請求項に記載の方法。
  11. 細胞の機能分類を示す細胞の特徴を標的とする一つ以上の蛍光レポーターまたは発光レポーターで標識した後に、細胞の画像を取得する、請求項1〜10のいずれかに記載の方法。
  12. レポーター分子が、蛍光標識、蛍光タンパク質、発光標識およびバイオセンサーからなる群より選択される、請求項8〜11のいずれかに記載の方法。
  13. 画像を取得する前に、細胞が一つ以上の操作を含む、請求項1〜12のいずれかに記載の方法。
  14. a.各化合物についての各化合物の濃度における各特徴の測定値について、細胞集団の変化の基準としてKolmogorov-Smirnov値または平均値等のパラメーターを計算し、希釈列についてのパラメーターを生じさせる工程、
    b.4-パラメーターのロジスティックフィットを利用して、かかる希釈列パラメーターを適合させる工程
    c.得られた適合データを解析してEC50値を算出する工程、
    d.化合物活性の基準としてEC50値を対数尺度に変換する工程
    e.クラスター解析を利用して、プロフィールの類似性および細胞システムの応答間の関連性を同定する工程
    を含む特徴の測定からプロフィールが構築される、請求項1〜13のいずれかに記載の方法。
  15. 一つ以上の試薬、ならびに細胞の一群をアッセイするための試薬を用いるためのおよび細胞の機能分類を示す少なくとも6つの細胞の特徴を含む応答プロフィールを作成するための指示書を含むキットであって、一つ以上の試薬が、細胞の機能的な分類を示す少なくとも6つの細胞の特徴を検出するための試薬を含む、キット。
  16. 6つ以上の細胞の機能分類を示す少なくとも6つの細胞の特徴を含む、請求項15に記載のキット。
  17. 請求項に記載の方法に従って構築されたデータベース。
  18. クラスター解析を含む方法を用いて、特徴が応答プロフィールに組み合わされる、請求項1〜14のいずれかに記載の方法。
  19. 処理される細胞の群が、2以上の細胞のタイプを含む、請求項1〜14のいずれかに記載の方法。
