CRACチャネルの分子性質はまだ完全にはわかっていない。最も広く研究されたCRACチャネルについての候補遺伝子は、ショウジョウバエ(Drosophila)光受容体TRP(Transient Receptor Potential(一過性受容体ポテンシャル))遺伝子の>25個の哺乳動物相同体であった。しかし、たいていのTRPタンパク質は、非特異的陽イオンチャネルを形成し、二価陽イオンに対していくらか優先を示すものでさえ、異種性に発現した場合、CRACチャネルの重要な生物物理学的に顕著な特徴のすべてを示すとは限らない(Clapham, 2003. Nature 426:517)。最近まで、TRPV6は、その生物物理学的特徴の一部がCRACのそれと重複するため、最も有望なCRACチャネル候補遺伝子であった。CRACのようにTRPV6はCa2+を選択的に運ぶが、それは、CRACチャネルをよく表している特徴である貯蔵枯渇により活性化されない。RNAiを用いてTRPV6発現を抑制するノックダウン研究、およびTRPV6-/-マウス由来のT細胞を用いる本発明者らの研究は、TRPV6の非存在下においてSOCEまたはICRACにおける欠陥を示さなかった(Kahr, et al. 2004. J Physiol 557:121; Kepplinger, et al. CaT1もTRPC3タンパク質も、Tリンパ球のCRACに寄与しない。原稿準備中)。従って、TRPV6も任意の他の遺伝子も、SOCEまたはCRACチャネル活性に関与するとは確認されていない。
CRACチャネルが活性化される機構もまた不明である。細胞内Ca2+貯蔵の枯渇は、CRAC活性化に必要であるが、どのようにしてERにおける低下したCa2+濃度についての情報がCRAC細孔へ伝達されるかは、知られていない。3つの主要なモデルが提案されているが、合意には至っていない(Parekh and Putney, Jr. 2005, Physiol Rev 85:757)。(i)「立体構造的結合モデル」は、ERの表面における分子の立体構造的変化を前提とし、その後、分子はCRACチャネルに結合する;(ii)「分泌結合モデル」は、(恒常的に活性な)CRACチャネルが、貯蔵が枯渇すると原形質膜に融合する細胞質内小胞に存在することを示唆する;(iii)「カルシウム流入因子(CIF)モデル」は、刺激された細胞の細胞質へCIFが放出された場合に、CRACチャネルを通ってCa2+流入を活性化する可溶性小分子を予想する。
最近の証拠は、STIM1が貯蔵量作動性Ca2+流入およびCRACチャネル機能において重要な役割を果たすことを示唆している。Roosら(2005, J Cell Biol 169:435)、Liouら(2005, Curr Biol 15:1235)、および本発明者らのグループ(後述の実施例2参照)による3つの独立したRNAiスクリーニングは、RNAiによるSTIM発現の抑制が、キイロショウジョウバエ(Drosophila melanogaster)S2細胞および哺乳動物細胞におけるCa2+流入を障害することを見出している(図5)。STIM1は、最初は、プレB細胞の増殖を促進する間質タンパク質として、および推定の腫瘍抑制因子として特徴付けられたI型膜貫通タンパク質である(Oritani, et al. 1996. J Cell Biol 134:771; Sabbioni, et al. 1997. Cancer Res 57:4493)。STIM1についてのヒト遺伝子は、ウィルムス腫瘍を含むいくつかの小児悪性腫瘍に関連した遺伝子を含むと考えられている染色体11p15.5上に位置する(Parker et al. 1996, Genomics 37:253)。STIM1は、そのER/細胞外領域におけるCa2+結合EFハンドモチーフおよび不稔性α-モチーフ(sterile α-motif)(SAM)ドメイン、1回膜貫通ドメイン、ならびに2つの推定細胞質コイルドコイル領域を含む(Manji et al. 2000, Biochim Biophys Acta 1481:147)。ドメイン構造およびゲノム組成は、STIM2と呼ばれる関連遺伝子に保存されており、STIM2は、主にそのC末端においてSTIM1と異なる(Williams et al. 2002, Biochim Biophys Acta 1596:131)。STIM1は、STIM2とホモ二量体化またはヘテロ二量体化することが可能である(Williams et al. 2002、前記)。ERに発現している場合、グリコシル化およびリン酸化研究により判断すると、そのC末端領域は細胞質に位置するが、N末端はERの内腔に存在する(Maji et al. 2000、前記; Williams et al. 2002、前記)。わずかなSTIM1が、原形質膜に位置する。STIM1発現のRNAi媒介性抑制はSOCEおよびCRACチャネルに干渉するが、STIM1がCa2+チャネル自体である可能性は低い。むしろ、STIM1は、ERにおいてそのEFハンドを介してCa2+レベルを感知する可能性がある(Putney, Jr. 2005. J Cell Biol 169:381; Marchant, 2005, Curr Biol 15:R493)。貯蔵量作動性Ca2+流入の立体構造的結合モデルと一致して、STIM1は、ERと原形質膜の間の空間に物理的に橋を架け、それに従って、原形質膜において枯渇Ca2+貯蔵の感知を貯蔵量作動性Ca2+チャネルへ直接連絡する、重要なアダプタータンパク質として働きうる(Putney, Jr. 2005. 前記; Putney, Jr. 1986, Cell Calcium 7:1)。
本明細書に用いられる場合、「NFAT制御因子(NFAT regulator)」という用語は、NFAT活性を制御するタンパク質(NFAT制御タンパク質(NFAT regulator protein))およびコード遺伝子(NFAT制御遺伝子(NFAT regulator gene))を指すように用いられる。本発明の方法は、本明細書に記載されたNFAT制御因子の相同体、類似体、アイソフォーム(例えば、選択的スプライスバリアント)、誘導体、および機能性断片の使用を含むことを意図される。好ましくは、NFAT制御タンパク質の相同体は、本明細書に明確に同定されたものと少なくとも70%、より好ましくは80%、およびより好ましくは90%のアミノ酸同一性を有する。
一つの好ましい態様において、本発明のNFAT制御タンパク質は、ORAI遺伝子によりコードされる。本発明の基礎となる発見以前には、ORAI遺伝子の機能は知られていなかった。ORAI1核酸配列はGenBankアクセッション番号NM_032790に対応し、ORAI2核酸配列はGenBankアクセッション番号BC069270に対応し、かつORAI3核酸配列はGenBankアクセッション番号NM_152288に対応する。本明細書に用いられる場合、ORAIは、ORAI遺伝子、例えば、ORAI1、ORAI2、ORAI3のいずれか1つを指す。
一つの態様において、本発明は、本発明のNFAT制御タンパク質に対する抗体を提供する。本発明のペプチドの1つまたは複数の特定のドメインに結合する抗体を調製することができ、抗体をNFAT制御遺伝子またはタンパク質活性を調節するために用いることができる。
本発明の方法はまた、NFAT制御不全/機能不全および/またはカルシウムシグナル伝達に関連した状態および疾患を処置するために、またはその状態および疾患の処置に有用な作用物質を同定するために利用されうる。そのような疾患には、非限定的に、免疫系の機能亢進または不適当な活性を含む免疫系疾患、例えば、急性免疫疾患、慢性免疫疾患、および自己免疫疾患が挙げられる。そのような疾患の例には、リウマチ様関節炎、炎症性腸疾患、同種または異種移植拒絶(器官、骨髄、幹細胞、他の細胞および組織)、移植片対宿主病、再生不良性貧血、乾癬、紅斑性狼瘡、炎症性疾患、MS、I型糖尿病、喘息、肺線維症、強皮症、皮膚筋炎、シェーグレン症候群、心膜切開後症候群、川崎病、橋本甲状腺炎、グレーブス病、重症筋無力症、尋常性天疱瘡、自己免疫性溶血性貧血、特発性血小板減少症、慢性糸球体腎炎、グッドパスチャー症候群、ヴェグナー肉芽腫症、多発性硬化症、嚢胞性線維症、慢性再発性肝炎、原発性胆汁性肝硬変、ブドウ膜炎、アレルギー性鼻炎、アレルギー性結膜炎、アトピー性皮膚炎、クローン病、潰瘍性結腸炎、結腸炎/炎症性腸症候群、ギラン・バレー症候群、慢性炎症性脱髄性多発根ニューロパチー、湿疹、および自己免疫性甲状腺炎が含まれる。移植片拒絶は、組織または器官移植に起因しうる。移植片対宿主病は、骨髄または幹細胞移植に起因しうる。免疫系の機能亢進を含む免疫系疾患には、例えば、HIV疾患などの後天性免疫不全症、および分類不能型免疫不全症(CVID)を含む免疫不全疾患が挙げられる。
一つのNFAT関連疾患/障害は、重症複合型免疫不全症(SCID)である。SCIDは、Tリンパ球およびBリンパ球の両方が損なわれたもしくは障害された発生ならびに/または機能により引き起こされる一群の先天性免疫障害である。100,000人の個体群あたり1人の推定有病率をもつまれな疾患であるSCIDは、20個より多い異なる遺伝子における突然変異により引き起こされうる。X連鎖SCIDをもたらす、インターロイキン2(IL-2)、IL-4、IL-7、IL-9、およびIL-15受容体のコモンγ鎖(cγ)における突然変異が、全症例の50%を占める。全SCID症例の約10%は、T細胞およびB細胞の発生または機能、特にシグナル伝達(CD3εおよびγ、ZAP-70、p56lck、CD45、JAK3、IL-7Rα鎖)、にとって重要な遺伝子における様々なまれな突然変異による。これらの突然変異が低発生率であり小さなファミリーサイズであるため、古典的な位置クローニングは、通常、これらのSCID疾患の大部分に対して不可能であり、突然変異は、しばしば、T細胞の機能分析、続いて候補遺伝子の配列決定により、公知のシグナル伝達遺伝子に見出された。科学的には、SCID疾患は、T細胞およびB細胞の機能解明において極めて価値があり、免疫系における遺伝子機能不全の帰結を浮き彫りにしている。
一つの態様において、本発明は、NFAT制御因子発現または機能を変化させる作用物質についてスクリーニングするための方法に関する。一つの態様において、本発明は、NFAT制御タンパク質の機能を変化させる作用物質についてスクリーニングするための方法に関する。CRACチャネル活性化の調節に関して、NFAT制御因子の機能が変化しうる。NFATリン酸化の調節に関して、NFAT制御因子の機能が変化しうる。NFAT細胞内局在性の調節に関して、NFAT制御因子の機能が変化しうる。遊離細胞内カルシウムレベルの調節に関して、NFAT制御因子の機能が変化しうる。カルシニューリン活性の調節に関して、NFAT制御因子の機能が変化しうる。一つの態様において、変化させる、または調節するとは、活性の上方制御または増強を指す。一つの態様において、変化させる、または調節するとは、下方制御または阻害を指す。
本発明の試験化合物は、以下を含む、当技術分野において公知のコンビナトリアルライブラリー方法における多数のアプローチのいずれかを用いて得られうる:生物学的ライブラリー;ペプトイドライブラリー(ペプチドの機能性を有するが、酵素分解に対して抵抗性である新規な非ペプチド骨格を有し、それにもかかわらず生理活性が残存する、分子のライブラリー;例えば、Zuckermann, et al., 1994 J. Med. Chem. 37:2678-85参照);空間的にアドレス可能なパラレル固相または液相ライブラリー;逆重畳積分を必要とする合成ライブラリー方法;「1ビーズ1化合物」ライブラリー方法;およびアフィニティークロマトグラフィー選択を用いる合成ライブラリー方法。生物学的ライブラリーおよびペプトイドライブラリーアプローチは、ペプチドライブラリーに限定されるが、他の4つのアプローチは、ペプチド、非ペプチドオリゴマー、または小分子の化合物のライブラリーに適用できる(Lam (1997) Anticancer Drug Des. 12:145)。
本発明のもう一つの局面は、実施例1〜3に記載されているように、遊離細胞内Ca2+レベル、カルシニューリン活性化、および細胞におけるNFAT局在性の制御因子についてスクリーニングするための方法に関する。一つの態様において、全NFAT制御ドメインまたはその機能性断片もしくは誘導体、および機能的に連結したレポータータンパク質(制御ドメインの細胞内局在を測定するための、例えば、GFPまたは抗原性エピトープ)を含む融合タンパク質をコードする組換えベクターが、細胞、すなわち、試験細胞へトランスフェクションされる。ベクターをトランスフェクションした試験細胞を、試験作用物質と接触させる。何時間か後、例えば、48〜72時間後、試験細胞に、NFAT-レポーター融合タンパク質の細胞内局在についてスコアを付ける。スコア付けは、実施例におけるように自動顕微鏡法、例えば、蛍光顕微鏡法、共焦点顕微鏡法を手段として、または手作業の顕微鏡法を手段として、達成されうる。二次試験アッセイは、カルシウム流入検出アッセイを含む。試験作用物質が組換えベクターの発現産物の細胞内局在性に対する効果を有する場合には、これは、その試験作用物質がNFAT制御因子の機能を調節することを示している。
本明細書に提供されたスクリーニング/同定方法のいずれかにおいて試験作用物質の細胞内カルシウムに対する効果をモニタリングする段階において、細胞カルシウム(サイトゾルおよび細胞内のオルガネラまたは区画を含む)および/または細胞、オルガネラ、もしくはそれらの部分(例えば、膜)の内側もしくは外側へのイオンの移動の、直接的または間接的な評価または測定が行われうる。カルシウムレベルおよびイオン移動または流束を評価するための様々な方法は、本明細書に記載されており、および/または当技術分野において公知である。使用される特定の方法および用いられる条件は、細胞内カルシウムの特定の局面をモニタリングするかどうかに依存しうる。例えば、本明細書に記載されているように、試薬および条件は、貯蔵量作動性カルシウム流入、休止サイトゾルカルシウムレベル、ならびに細胞内オルガネラのカルシウムレベルおよび細胞内オルガネラによるカルシウム取り込みまたは細胞内オルガネラからのカルシウム放出を特異的に評価するために用いることができ、公知である。細胞内カルシウムに対する試験作用物質の効果は、例えば、細胞、細胞内オルガネラもしくは貯蔵区画、膜(例えば、剥離した膜パッチまたは脂質二重層を含む)、または無細胞アッセイシステムを用いてモニタリングされうる。
一つの態様において、アッセイは、NFAT制御タンパク質またはその生物活性部分を発現する細胞を試験化合物と接触させ、NFAT制御因子活性を調節する試験化合物の能力を測定する、細胞に基づいたアッセイである。NFAT制御因子活性を調節する試験化合物の能力を測定する段階は、例えば、細胞におけるカルシウム流束の変化をモニタリングすることにより、またはIL-2発現の活性化などのカルシウム流束を調節する下流効果を試験することにより、達成されうる。そのような下流効果を試験する方法は、当技術分野において公知であり、細胞増殖および細胞成長の調節を含む。例えば、細胞におけるNFAT制御因子分子の数を減少させる、またはNFAT制御因子チャネルの機能に影響を及ぼす化合物は、細胞増殖を減少させうる。または、NFATトランス活性化を必要とする遺伝子の転写がモニタリングされうる。
a. カルシウム指示薬により測定される場合、増加した[Ca2+]iの直接的な阻害がある;
b. パッチクランプにより測定される場合、ICRACの直接的な阻害がある;
c. カルシニューリン活性、NFAT細胞内局在、NFATリン酸化、および/もしくはサイトカイン、例えばIL-2の産生などの下流シグナル伝達機能の阻害がある;または
d. 活性化誘導性細胞増殖、分化、および/もしくはアポトーシスシグナル伝達経路における改変がある。
以下の1つまたは複数を調節する作用物質を同定するのに用いるシステムもまた本明細書に提供される:NFATタンパク質、NFAT制御タンパク質、および細胞内または細胞質カルシウム。そのようなシステムは、1つもしくは複数のタンパク質、例えば、本発明のNFAT制御タンパク質、またはその断片もしくは誘導体、例えば、ORAIタンパク質、またはその断片もしくは誘導体を含む細胞またはその部分を含む。一つの態様において、タンパク質は、細胞に対して外因性(異種性または相同性)である。一つの態様において、細胞は、NFAT制御タンパク質またはその断片もしくは誘導体をコードする外因性(例えば、異種性または相同性)核酸を含む。一つの態様において、システムはさらに、細胞の原形質膜を横断する電流をモニタリング、検出、または測定するために用いられるモニタリング剤を含む。多くのそのようなモニタリング剤は当技術分野において公知である。「モニタリング剤」という用語はまた、そのようなモニタリングに用いられる任意の装置を含むと意図される。
本発明の局面はさらに、本明細書で考察された方法に記載されたアッセイに用いられる組換え細胞に関する。一つの局面において、本発明はまた、本明細書に記載された任意の組換え細胞を含む。一つの態様において、組換え細胞は、少なくとも1つの外因性(異種性または相同性)NFAT制御タンパク質またはその断片もしくは誘導体を含む。組換え細胞はまた、さらに、NFAT制御タンパク質またはその断片もしくは誘導体をコードする少なくとも1つの外因性(異種性または相同性)核酸を含む。NFAT制御タンパク質は哺乳動物起源でありうる。組換え細胞は、NFAT制御タンパク質またはその断片もしくは誘導体を過剰発現しうる。この過剰発現は、外因性(異種性または相同性)NFAT制御タンパク質(例えば、外因性核酸由来)の発現に起因しうるか、または天然/内因性NFAT制御タンパク質の過剰発現に起因しうる。
本発明は、NFAT制御因子を過剰発現する、NFAT制御因子を異所的に発現する、低下したレベルのNFAT制御因子を発現する、突然変異体NFAT制御因子を発現する、またはNFAT制御因子の発現についてノックアウトされるように操作されている非ヒトトランスジェニック動物を提供する。そのような動物およびそのような動物由来の細胞株は、NFAT制御タンパク質の機能および/または活性を研究するために、ならびにNFAT制御因子活性の調節因子を同定および/または評価するために、有用である。NFAT制御因子ポリペプチドを過剰発現する動物は、例えば、NFAT制御因子ポリペプチドの活性を調節するための候補化合物の効果を試験する、および化合物の効果をインビボで評価するために有用である。
治療的適用について、本発明のペプチドおよび核酸、NFAT制御因子に対する抗体、または本発明のスクリーニング方法により同定された作用物質、例えば、小分子、siRNA、shRNAは、哺乳動物、特にヒトなどの被験体に、単独で適切に投与されてもよく、または1つもしくは複数の許容される担体および任意で他の治療用成分と共にペプチド、核酸、抗体、もしくは作用物質を含む薬学的組成物の一部として、適切に投与されてもよい。担体は、製剤の他の成分と適合性であり、かつそのレシピエントに対して有害でないという意味で「許容され」なければならない。
材料および方法:
症例報告
この研究において調べられた2人のSCID患者の詳細な症例報告が記載されている(Feske 1996, 2000)。
T細胞株を、2人の患者および21人の親族メンバーの末梢血リンパ球から樹立し、記載されているように48増殖させた。新生児SCID患者2および健康な新生児(Hs27細胞株、ATCC、Manassas, VA)由来の包皮線維芽細胞を、テロメラーゼ発現プラスミド(hTERT、S. Lessnick, DFCI, Boston, MAの親切な贈与)のレトロウイルス形質導入により不死化した。マイクロファージ血球様ショウジョウバエ細胞株S2R+を、10%ウシ胎児血清を含むシュナイダー培地(Invitrogen)で標準プロトコールに従って増殖した。タプシガルジンを、LC Biochemicals(Woburn, MA)から、カルブドトキシン(CTX)および2-アミノエトキシジフェニルボレート(2-APB)をSigma(St. Louis, MO)から購入した。
SCID患者および21人の親族のゲノムDNAを、ゲノムDNA Maxi prepキット(Qiagen)を用いて、末梢血単核細胞から調製した。遺伝子型決定を、SNP Genotyping Center(Broad Institute, Cambridge, MA)およびHarvard Partners Center for Genetics and Genomics(Boston, MA)で、「GeneChip」Human Mapping 10K Arrays(Xba 142 2.0, Affymetrix, Santa Clara, CA)を用いて行い、0.38の平均SNPヘテロ接合性および258kbの平均マーカー間密度であった。このプラットフォームは、ヒトゲノムにおける10,000個より多いSNPの同時的遺伝子決定を可能にした。パラメトリック連鎖解析について、データを、「比較連鎖」49を用いて「連鎖」型式へ変換した。親遺伝子型と矛盾するメンデルの遺伝子型エラーを検出し、不明遺伝子型に設定した。多点パラメトリック連鎖解析を、Allegro50を用いて各SNP位置におけるLODスコアを計算するように行った。連鎖を確認するために、本発明者らは、Genehunter 2.1r651およびMerlin52を用いてSNPデータを再分析し、非常に類似した結果を得た。パラメトリック解析について、0.001の疾患対立遺伝子頻度、0.99の浸透度値、および0.01の表現型模写を全系図について用いた。パラメトリック連鎖解析を、遺伝の劣性および優性モデルのそれぞれを用いて行った。「劣性」モデルについて、両方の患者、彼らの親、罹患していない兄弟、および祖父母(図1Aにおける個体8、11、35、36、37、38、39、63、64)由来のハプロタイプを、SCID疾患についての遺伝の常染色体劣性型を仮定して解析し、両方のSCID患者は、一般的な疾患原因突然変異についてホモ接合性であった。この解析からの予想最大オッズ比のlog10(LOD)スコアは〜1.9(すなわち、-log10[0.25×0.25×0.25×0.75])であった。「優性」モデルについて、貯蔵量作動性Ca2+流入の低下をもつ12人の親族メンバーが「罹患した」、すなわち、優性作用性突然変異の保因者として定義され、それらのSNPハプロタイプを、正常な貯蔵量作動性Ca2+流入をもつ8人の健康な親族メンバーのSNPハプロタイプと比較した。この解析からの予想最大LODスコアは〜3.8(すなわち、-log10[0.512])であった。
2人の患者、21人の親族メンバー、および3人の独立した対照のゲノムDNAを、以下のオリゴヌクレオチドプライマーを用いてOrail1のエクソン1および2における突然変異について配列決定した。
DNAを、AmpliTaq Goldポリメラーゼを用いて増幅し、1%アガロースゲル上で分離した。PCR産物をゲル精製し、以下のプライマーを用いて直接配列決定した。
配列決定は、DF/HCC DNA Resource Core(DFCI)において行い、DNA配列は、Xplorer Lite (dnaTools, Ft. Collins, CO)を用いて分析した。
Orai1(NM_032790)のコード配列における271位におけるC>T点突然変異がSNPである可能性を排除するために、本発明者らは、International HapMapプロジェクト30,31のために構築された多様な地理的起源の270個の個体のパネル由来のDNAを調べた。遺伝子決定は、以前に記載されているように53、ハイスループットプライマー伸長方法を用いて、Sequenomプラットフォーム上での質量分析(MALDI-TOF)による検出と共に行った。この方法の詳細な説明は、「Sequenomプラットフォーム」下のhttp://www.hapmap.org/downloads/genotyping_protocols.htmlに見出されうる。試料の89%は遺伝子決定に成功し、すべてがCCホモ接合体として同定された。
PCR断片(最大600bpまでのサイズ)をインビトロ転写反応のための鋳型として用い、続いて、鋳型DNAを除去するためにDNアーゼI処理を行った。精製後、dsRNA(5μg)を、8チャンバーのスライドにおいて(12ウェルプレートについては10μg)、S2R+細胞へNFAT-GFP発現プラスミドと共に同時トランスフェクションした。インキュベーションの72時間後、局在性アッセイのために、細胞をCa2+流入誘導物質、1μMイオノマイシンまたは1μMタプシガルジンで処理し、[Ca2+]iレベルの測定のためにトリプシン処理した。
RNAiスクリーニングを記載されているように行った(Armknecht S. et al., 2005, Methods Enzymol 392, 55-73; Btros M. et al. 2004 Science 303, 832-835)。マクロファージ血球様ショウジョウバエ細胞株S2R+に、発現プラスミドpAc5.1(Invitrogen)へサブクローニングしたNFAT1(1〜460位)-GFP融合タンパク質についてのコード配列を安定にトランスフェクションした。トランスフェクションは、Effectene(Qiagen)を用いて、比率19:1の発現プラスミド対pCoHygro(Invitrogen)(恒常的活性プロモーターの制御下でハイグロマイシン抵抗性遺伝子をコードする)で達成した。細胞を、300μg/mlハイグロマイシンで3〜4週間選択し、安定なクローンを目視検査によって選択した。NFAT1(1〜460位)-GFPを発現する104個のS2R+細胞を、0.25μgのショウジョウバエmRNAに対するdsRNA(10μlの無血清培地中)を含む384ウェルプレートの各ウェル上に加え、26℃で1時間インキュベートし、RNAiを達成するために26℃で48〜72時間インキュベートした。S2R+細胞を、5mM CaCl2を含むシュナイダー培地において1μMタプシガルジンで、室温、10分間刺激し、固定し、DAPIで染色した。同時GFPおよびDAPI画像を、各ウェルにおいて3つの異なる位置から自動カメラにより取得し、目視検査によりスコアを付けた。個々のdsRNAがアレイされている合計21,884ウェルを含む、合計58個の384-プレートを分析した。この研究について、dStimおよびdOraiの両方についてのdsRNA単位複製配列は、19ヌクレオチドまたはそれ以上の完全な一致をもつ予想されるオフターゲットはなかったことを述べておく。
Orai1についての完全長cDNA(BC015369)を、OpenBiosystems(Huntsville, AL)から購入し、pENTR11(「Gateway」system, Invitrogen, Carlsbad, CA)へmycエピトープをコードするN末端またはC末端の末端配列とインフレームでサブクローニングした。その後、Orai1を、バイシストロン性レトロウイルス発現ベクターpMSCV-CITE-eGFP-PGK-Puro(Masatsugu Oh-horaの親切な寄贈)へ移動させたが、それは、Orai1、GFP、およびピューロマイシン抵抗性遺伝子の同時発現を可能にする。gp293パッケージング細胞株に、orai1、gag-pol、およびenvをコードするプラスミドを同時トランスフェクションし、広宿主性複製無能レトロウイルスを産生した。ウイルス含有上清を24時間収集し、濾過し(0.45マイクロメートル、低いタンパク質結合性)、6000xgで16時間の遠心分離により濃縮した。T細胞および線維芽細胞に、それぞれ、4日間および1日間のウイルス上清の添加により、形質導入した。形質導入効率は、フローサイトメトリーを用いてGFP発現により評価し、myc-Orai1発現は、免疫ブロッティングおよび免疫細胞化学法を用いて評価した。場合によっては、形質導入したT細胞をさらに、1μg/mlピューロマイシンで3日間選択した。
Orai1のヒドロパシープロットを、Kyte-Doolittleアルゴリズム29を用いて作成した。膜トポロジーをさらに、隠れマルコフモデルに基づいたPhobiusアルゴリズム26を用いて評価した。配列アラインメントは、MegAlign(DNAStar, Madison, WI)を用いて行った。
Orai1についての免疫細胞化学法を、記載されているように11行った。簡単には、レトロウイルスで形質導入したT細胞および線維芽細胞を、3%パラホルムアルデヒドで固定し、非透過のままにしておく、または0.5%NP-40を含む水緩衝液で透過処理し、抗myc抗体(9E10)およびCy3標識二次抗体とインキュベートした。免疫蛍光を、63×水浸対物レンズを用いるBX50BWI Olympus顕微鏡上で、Radiance 2000レーザースキャニング共焦点システム(Bio-Rad Laboratories)を用いる共焦点イメージングにより分析した。
T細胞に、負荷培地(RPMI+10%FBS)中の1μM fura-2/AM(Molecular Probes)を1×106細胞/mlで22〜25℃で30分間負荷し、それらを負荷培地に再懸濁し、ポリ-L-リジンコーティング化カバースリップに15分間付着させた。線維芽細胞を、UV滅菌したカバースリップ上で直接増殖させ、それらに3μM fura-2/AMを22〜25℃で45分間負荷した。[Ca2+]i測定について、細胞をRC-20閉鎖槽フローチャンバー(Warner Instrument Corp., Hamden, CT)に載せ、OpenLabイメージングソフトウェア(Improvision)を用いてAxiovert S200エピ蛍光顕微鏡(Zeiss)上で分析した。細胞を、Ca2+を含まないリンガー液に灌流し、Ca2+貯蔵を、1μMタプシガルジンで受動的に枯渇させた。貯蔵の能動的枯渇は、10μg/ml抗CD3抗体(OKT3, eBioscience, San Diego, CA)とのインキュベーションにより10分間22〜25℃で誘導した。Fura-2発光を、340nmおよび380nmで励起して、510nmで検出し、Fura-2発光比(340/380)を、バックグラウンドの引き算後、各5分間隔について計算した。各実験について、約100個の個々の細胞を、Igor Pro(Wavemetrics, Lake Oswego, OR)分析ソフトウェアを用いて340/380比について分析した。[Ca2+]iは、比較式[Ca2+]i=K*(R-Rmin)/(Rmax-R)から推定した。K*、Rmin、およびRmaxは、以前に記載されているように54、インサイチューで対照ヒトT細胞において測定した。Ca2+流入比は、0.2mM細胞外Ca2+の再添加後のCa2+濃度における上昇(d[Ca2+]i/dt)の最大比から計算した。
標準細胞外リンガー液は以下を含んだ(mM単位):155 NaCl、4.5 KCl、20 CaCl2、1 MgCl2、10 D-グルコース、および5 Na-Hepes(pH 7.4)。二価を含まない(DVF)標準リンガー液は以下を含んだ(mM単位):155 Na、10 HEDTA、1 EDTA、および10 Hepes(pH 7.4)。カリブドトキシン(CTX)は、Kv1.3チャネルを遮断して、DVF溶液におけるICRAC記録の混入を防止するために全外液に含めた。標準内部溶液は以下を含んだ(mM単位):150 Cs-アスパラギン酸、8 MgCl2、8 BAPTA、および10 Cs-Hepes(pH 7.2)。
パッチクランプ実験は、ITC-16入力/出力ボード(Instrutech, Port Washington, NY)およびMacintosh G3コンピューターにインターフェイスで接続したAxopatch 2000増幅器(Axon Instruments, Foster City, CA)を用いて22〜25℃で標準ホールセル記録構成で行った。記録電極を100μlピペットから引き抜き、Sylgardでコーティングし、先端を熱加工して2〜5MΩの最終抵抗にした。刺激およびデータ獲得および分析は、Igor Proプラットフォーム(Wavemetrics, Lake Oswego, OR)で開発された施設内の慣例を用いて行った。保持電位は、他に規定がない限り、+30mVであった。電圧刺激は、通常、1.3sごとに加えられる、-100mVまでの100-msステップ、続いて-100mVから+100mVまでの100-ms傾斜からなる。電流は、4極のBesselフィルターを用いて2kHzで濾過し、5kHzでサンプリングした。データは、槽のリンガー液に対するピペット溶液の液間電位について(-10mV)、および槽接地寒天橋のリンガー液に対する槽DVF溶液について(+5mV)、補正する。ノイズ分析について、200-msスイープを-100mVの保持電位において3Hzの速度で得、5kHzにおいてデジタル化し、2kHzにおいてAxopatch 200増幅器の内部Besselフィルターを用いて低域フィルタリングした。平均および分散は、デジタル化データの100-msセグメントから計算した。
特に断りのない限り、全データは、2μM La3+か、またはBAPTAによる受動的透析中のICRAC誘導前に収集されたトレースかのいずれかで、収集した漏洩電流について補正した。透過性比(PCs/PNa)は、以下の方程式を用いて2イオン性逆転電位(biionic reversal potential)から計算した:
R、T、およびFはそれらの通常の意味をもち、Erevは逆転電位である。
Ca2+は、ほとんどすべての細胞型における必須の二次メッセンジャーである。特に、原形質膜を横断する持続性Ca2+流入は、リンパ球活性化および適応免疫応答にとって重大である1。Tリンパ球およびBリンパ球の表面抗原受容体による抗原認識は、複数のチロシンキナーゼの活性化、ならびに多様なアダプターおよびシグナル伝達タンパク質を含む大きな足場複合体の構築を含む、精巧なシグナル伝達カスケードを誘発する。初期の生化学的結果は、PLCγの活性化であり、それは、IP3を生成することにより小胞体(ER)からCa2+を放出する;結果として生じる内腔ER Ca2+における減少は、Ca2+放出活性化Ca2+(CRAC)チャネルと呼ばれている、非常に特異的な電気生理学的性質をもつ「貯蔵量作動性」Ca2+チャネルの類を開く1-3。CRACチャネル開口は、結果として、原形質膜を横断するCa2+イオンの持続性流入を生じ、細胞内遊離Ca2+([Ca2+]i)レベルの持続性上昇を促進し、タンパク質ホスファターゼ、カルシニューリンを含む多様なCa2+/カルモジュリン依存性酵素を活性化する;最終的結果は、増殖およびエフェクター免疫機能に必要とされるCa2+依存性転写経路の活性化である4,5。主要なCa2+制御性転写因子の一つは、休止細胞の細胞質に存在する高度リン酸化タンパク質のファミリーである、NFATである5。持続性Ca2+流入は、結果として、カルシニューリンによるNFATの脱リン酸化を生じ、その核への移行を促進し、そこで、それは、多数の活性化関連遺伝子の発現を作動させる4,6。
ヘテロ接合性疾患保因者の表現型同定
2人のSCID患者は同族の両親の下に生まれ、SCID患者の親もSCID患者の親族のいずれの他のメンバーも免疫不全の臨床症状を示さなかったため、常染色体劣性遺伝様式を示唆した(図1A)。さらに、SCID患者の両親由来のT細胞は、2mM細胞外Ca2+の存在下においてほとんど正常な貯蔵量作動性Ca2+流入を示した10。親のT細胞におけるCa2+流入の潜在的欠陥の仮面をはぐために、本発明者らは、タプシガルジン媒介性貯蔵枯渇後、Ca2+流入の初速度(本明細書では、細胞内遊離Ca2+濃度の変化の初速度として定義される、d[Ca2+]i/dt)を測定したが、2mMから0.2〜0.5mMへCaCl2の細胞外Ca2+濃度を低下させることによりCa2+流入の原動力を減少させた。これらの条件下で、両方の両親由来のT細胞におけるピークCa2+レベルおよびCa2+流入速度は、野生型対照T細胞に観察されたものの〜50%またはそれ未満であった(図1B)。本発明者らは、この所見が、両親がSCID患者における原因突然変異のヘテロ接合性保因者であったという事実に起因する、潜在的遺伝子量効果を反映していると仮定した。
SCIDファミリーの23個のメンバー由来のゲノムDNAを、ゲノムワイドなSNPアレイを用いる遺伝子型決定に用いた。SNPデータは、2つの独立した連鎖解析を用いて評価した。第一の解析は、臨床表現型に基づいて常染色体劣性遺伝様式を仮定し、2人の患者、彼らの両親、彼らの罹患していない兄弟、および彼らの祖父母由来のDNAを解析した(図1Aにおいて灰色斜線部分により示された、系図A)。対照的に、第二の解析は、系図の残りを完全に独立した解析において利用した。ここでは、インビトロでの表現型解析による疾患突然変異のヘテロ接合性保因者を同定する本発明者らの能力に基づいて、常染色体優性遺伝様式を仮定した(図1Aにおいて緑色四角形により示された、系図B)。重要なことには、第一解析は、個体のヘテロ接合性表現型状態を考慮せずに行い、第二(優性遺伝)は、大きな系図において、2つの関連していない半分として(親35および36の親族は独立して処理する)、これらの2つの解析の結果が完全に独立しているように行った。従って、これらの2つの実行の分析を、3つの独立した系図(1つのホモ接合性マッピング実行および2つの非関連優性系図)から出たとみなし、これらからのパレメトリックLODスコアを単に加えるだけで統計学的にロバストな複合LODスコアを得ることができる(材料および方法参照)。
位置クローニングの試みと並行して、本発明者らは、CRACチャネルの構成要素およびCRAC活性化へ導くシグナル伝達経路を同定する独立した方法として、ショウジョウバエにおいてNFAT制御因子についてのゲノムワイドなRNAiスクリーニングを行った。NFAT-GFP融合タンパク質を安定に発現するショウジョウバエS2R+細胞を、遺伝子発現のノックダウンに達するように〜21,000個のショウジョウバエ遺伝子のそれぞれに対するアレイdsRNAと共に3日間インキュベートした。その後、細胞を、Ca2+貯蔵を枯渇させるためにタプシガルジンで10分間刺激し、それに従って、貯蔵量作動性Ca2+流入およびNFAT-GFPの核移行を活性化させた。その後、細胞を固定し、細胞を含むウェルをロボットにより写真撮影し、NFAT-GFPの細胞内分布を目視検査で評価した。枯渇がNFAT核移行に干渉する陽性候補の中に、いくつかの予想されるCa2+/カルシニューリン/NFATシグナル伝達経路の制御因子があり、カルシニューリンB(CanB)、カルシニューリンA(CanA-14F)、およびショウジョウバエStimを含んだ24,27。
本発明者らのデータはdOraiを貯蔵量作動性Ca2+流入の第二の新規な制御因子として関係づけたため(図2)、およびヒトOrai1についての遺伝子がSCID患者においてホモ接合性である12q24領域に位置していたため、本発明者らは、SCID欠陥がヒトOrai1における突然変異と関連しているかどうかを求めた(図3)。図1Aに示された23人の個体(患者および彼らの親族)由来のゲノムDNAを配列決定することにより、本発明者らは、両方のSCID患者がOrai1のエクソン1におけるミスセンスの突然変異についてホモ接合性であることを見出した。Orai1のコード配列の271位(NM_032790の444位)の突然変異、C>T転移は、そのタンパク質(NP_116179、図3B)の91位において、高度に保存されたアルギニン残基の代わりにトリプトファンへの置換(R91W)をもたらす。突然変異した残基は、Kyte-Doolittle方法29を用いてOrai1の疎水性を計算することにより予想される、Orai1における4つの可能性のある膜貫通セグメントの第一の最初に位置している(図3B、3C)。すべての13個の表現型的に予想されたヘテロ接合性疾患保因者(図1)は遺伝子型的には突然変異についてヘテロ接合性であるが(C/T)、健康な対照および非罹患親族メンバーは野生型対立遺伝子についてホモ接合性であった(C/C)。この位置における突然変異は、注釈付きSNPではなく(dbSNP Build 124)、これが単によくある多型であるという可能性はなくなる。この仮説を確認するために、本発明者らは、HapMapパネル(ユタ、イバダン(ナイジェリア)、東京、および北京からの合計270個体)全体においてこの多型をタイピングし、このパネルにおいて推定上の原因の「T」対立遺伝子の1つのコピーも見出さなかった(材料および方法、データ未提示)30,31。これらのデータは、C>T転移が一般的な集団においてよくある配列変異体ではないことを明白に示している;従って、その突然変異は、SCID患者の祖先において自発的に生じた可能性が高く、疾患に強く関連している。
本発明者らは、Orai1が、SCID患者由来のT細胞および線維芽細胞において野生型および突然変異体Orai1のN末端およびC末端エピトープタグ付き型を発現することによりSCID T細胞(および線維芽細胞)におけるCa2+流入欠陥を補完するかどうかを求めた。バイシストロン性IRES-GFPベクターを用いるSCID T細胞または線維芽細胞におけるMyc-Orai1WTのレトロウイルス発現は、GFP陽性細胞においてタプシガルジン処理に応答してCa2+流入を回復させたが、GFP陰性細胞においては回復させず、一方、突然変異体R91>W Orai1(Myc-Orai1R>W)のレトロウイルス発現はCa2+流入を回復させなかった。Myc-Orai1R>Wの、SCID T細胞および線維芽細胞においてCa2+流入を回復させる能力の欠如は、突然変異体および野生型タンパク質が等価レベルで存在し、かつ免疫ブロッティング(データ未提示)および免疫細胞化学により判定される場合、原形質膜に、または近くに類似して位置しているように思われるため、Myc-Orai1WTと比較した、Myc-Orai1R>Wの異常発現のせいではなかった。本発明者らは、N末端およびC末端の両方が細胞質に配向し、そのため、抗体に近づきにくいというトポロジーと一致して、抗myc抗体で非透過性細胞を染色することができなかった(図3C)。
前の実験で見られたCa2+流入の回復は、患者の細胞における活性CRACチャネルの再構成を反映することができた、またはCRACとは異なる、貯蔵量作動性Ca2+透過性イオンチャネルの発現(または活性化)から起こることができた。これらの可能性の間を区別するために、本発明者らは、ホールセルパッチクランプ構造を用いて、野生型または突然変異(R91W)Orai1で再構成されたSCID細胞における貯蔵枯渇から生じる電流を詳細に特徴付けた。SCID T細胞に、バイシストロン性IRES-GFPベクターにおけるOrai1をレトロウイルスで形質導入し、上記のように、Orai1を発現する細胞を、GFP蛍光により同定した。本明細書に示された実験において、貯蔵枯渇は、パッチピペットに8mM BAPTAを含むことによるか、またはタプシガルジンでの処理によるかのいずれかにより達成させた。
本明細書で、本発明者らは、貯蔵量作動性Ca2+流入の進化的に保存された構成要素およびICRACの必須貢献因子であるとOrai1を同定する。本発明者らは、Orai1における点突然変異が、重症複合型免疫不全症(SCID)のまれな型をもつ2人の患者における貯蔵量作動性Ca2+流入およびICRAC機能の遺伝的欠陥の原因であることを示す10,11。欠陥のある遺伝子としてのOrai1の同定は、2つの独立した遺伝分析の相乗的組み合わせを通して達成され、両方の分析は不偏のゲノムワイドなスクリーニングを含んだ。
材料および方法
ゲノムワイドな一次スクリーニング
方法は参考文献12,13から適合させた。104個のS2R+細胞を、10μlの無血清培地中0.25μgのdsRNAを含む各ウェルへ加え、26℃で1時間インキュベートした。その後、細胞に、シュナイダー培地(Invitrogen)(30μl)においてNFAT1(1〜460)-GFP発現プラスミド9,17(10ng)を一過性にトランスフェクションした。26℃で48〜72時間のインキュベーション後、細胞を固定し、DAPIで染色し、同時発生的GFPおよびDAPI画像を自動カメラにより各ウェルにおける3つの異なる位置から得た。個々のdsRNAがアレイされている合計21,884個のウェルを含む合計58個の384プレートを分析した。
一次スクリーニングからの699個の陽性の可能性のある候補における確証的スクリーニングは、NFAT1(1〜460)-GFPを安定にトランスフェクションしたS2R+細胞を用い、かつ候補を、それらの枯渇が休止細胞および刺激されたS2R+細胞の両方におけるNFAT細胞内局在を変化させるかどうかについて試験したこと以外、本質的には、一次スクリーニングについて記載されているように行った。全細胞が細胞質NFAT-GFPを含むウェルは最低スコア(0)を得、細胞の>90%が核NFAT-GFPを示すウェルには最高のスコア(3)が付けられた。すべての3つの実験から総計されたスコアは表Iに提示されている。最高の可能なスコアは9であるが、本発明者らは確証的スクリーニングにおいて控えめにスコアを付けたため、任意の候補により得られる最高の実際のスコアは6であることに留意されたい。全候補はまた、それらが、タプシガルジン(1μM、30分間)で処理された細胞におけるNFAT核局在化を阻止するかどうかについて試験した;ショウジョウバエSTIM(dSTIM)のみがこのアッセイにおいて陽性のスコアを付けた。
スコアを、DRSC(Drosophila RNAi Screening Center at Harvard Medical School)への提出のために整理統合して、書式設定し、その後、アッセイされた遺伝子の独自性(identity)(FlyBase識別子;既知の場合、ショウジョウバエ遺伝子名;いくつかの遺伝子オントロジー(Gene Ontology)(GO)識別子;およびいくつかのヒト相同体)を提供した。遺伝子オントロジー(GO)アノテーションは2つの方向で検索された。第一に、本発明者らは、各遺伝子についてのFlyBase識別子を使用するEnsembl's EnsMartツールを用いてGO記載を得た。第二に、本発明者らは、スクリーニングセンターにより提供されたGO識別子を用いて、遺伝子オントロジーコンソーシアム(Gene Ontology Consortium)から「GO用語およびID」ファイルからの記載を得た。遺伝子の機能カテゴリーは、陽性のキーワード検索、続いて手作業のキュレーション(curation)により構築した。陽性遺伝子はまた、KEGG経路データベースにおけるものなどのツールを用いて一般的な経路への関与について調べた。
PCR断片(最高600bpまでのサイズ)を、インビトロ転写反応のための鋳型として用い、続いて、鋳型DNAを除去するためにDNアーゼI処理を行った。精製後、dsRNA(5μg)を、8チャンバースライドにおいて(12ウェルプレートについては10μg)、NFAT-GFP発現プラスミドと共にS2R+細胞へ同時トランスフェクションした。インキュベーションの72時間後、局在性アッセイのために、細胞を未処理のままにしておくか、またはCa2+流入誘導物質、1μMイオノマイシンもしくは1μMタプシガルジンで処理し、そして、[Ca2+]iレベルの測定のためにトリプシン処理した。
FLAGタグ付きヒトキナーゼを、抗FLAG抗体結合型プロテインGビーズ(Sigma)を用いて一過性にトランスフェクションしたHEK293細胞の細胞全体の可溶化物から免疫沈降させ、免疫沈降物を、細菌細胞に発現させたNFAT1制御ドメイン全体(GST-NFAT1[1-415])か、またはNFAT1のSRR-1(アミノ酸149〜183位)、SP-2(アミノ酸206〜237位)、およびSP-3(アミノ酸264〜295位)モチーフに対応するGST融合ペプチドのいずれかのリン酸化について分析した(野生型、およびインビボでリン酸化されるセリンにおけるSer→Ala突然変異体の両方)10。免疫複合体を、溶解緩衝液(1.0%NP-40、50mM HEPES pH7.4、150mM NaCl、5mM EDTA、5mM EGTA、1mMジチオスレイトール[DTT]、20mM β-グリセロール-リン酸、10mMピロリン酸ナトリウム、0.1mMオルトバナジウム酸ナトリウム、10mM NaF、1mMフッ化フェニルメチルスルホン酸[PMSF]、10μg/mlアプロチニン、10μg/mlロイペプチン)で2回、およびキナーゼ緩衝液(20mM HEPES、pH7.4、20mM MgCl2、1mM DTT、0.1mMオルトバナジウム酸ナトリウム、20mM β-グリセロール-リン酸)で2回洗浄し、20μM ATP、2μCi[γ32P]-ATP、および10μgの野生型または突然変異体GST-ペプチド基質の存在下、最終容量40μlのキナーゼ緩衝液において30℃で20分間インキュベートした。ペプチドは、グルタチオン-セファロース上で単離し、リン酸化はSDSゲル電気泳動およびオートラジオグラフィーにより評価した。
pOZベクター42においてHAタグ付き完全長NFAT1を安定に発現する指数関数的に増加中(107個)のジャーカットT細胞に、250Vおよび960μFにおけるエレクトロポレーションによりトランスフェクションした。ルシフェラーゼ実験について、細胞に、0.5μg pRLTKレポーター(内部対照としてレニラ(Renilla)ルシフェラーゼ)、5.0μg pGL3レポーター(ホタルルシフェラーゼ、実験プロモーター)、および空ベクター、野生型DYRK2、またはキナーゼ不活性型(kinase dead)DYRK2をコードする発現プラスミドをトランスフェクションした。トランスフェクションから24時間後、未処理のままにしておいた細胞、またはPMA(20nM)、イオノマイシン(1μM)、および2mM CaCl2で6時間刺激した細胞を、Dual-Luciferase Reporter Assay(Promega)を用いて製造会社により推奨されるとおり、レポーター遺伝子活性について測定した。細胞内サイトカイン染色について、細胞に、GFPをコードしたプラスミド、および空ベクタープラスミド、野生型DYRK2、またはキナーゼ不活性型DYRK2を同時トランスフェクションした。トランスフェクションから24時間後、未処理のままにしておいた細胞、またはPMA(20nM)、イオノマイシン(1μM)、および2mM CaCl2で6時間、最後の4時間についてはブレフェルジンA(2μg/ml)の存在下で刺激した細胞を、PBS中4%パラホルムアルデヒドで25℃で20分間固定し、PBSで2回洗浄し、サポニン緩衝液(PBS、0.5%サポニン[Sigma]、1%BSA、および0.1%アジ化ナトリウム)において透過処理し、フィコエリトリン結合ラット抗ヒトIL-2(PharMingen)で25℃で30分間染色した。細胞をPBSにおいて2回洗浄し、FACSCaliburフローサイトメトリー(Becton Dickinson)およびFlowJoソフトウェアで分析した。
NFAT1(1〜460)-GFPを安定に発現する0.5×106個のHeLa細胞を、6ウェルプレートに播種し、翌日、リポフェクタミン2000トランスフェクション試薬(Invitrogen, Carlsbad, CA)を用いて製造会社のプロトコールに従い、対照siRNAまたはヒトDYRK1A siRNAに対応するsiRNA(Dharmacon, Inc., Lafayette, CO)をトランスフェクションした。細胞を再播種し、ノックダウンの効率を増加させるためにトランスフェクション手順を24時間後に繰り返した。細胞を、免疫ブロット分析または免疫細胞化学法のためにトランスフェクションから4日後に収集した。DYRK転写産物レベルをリアルタイムRT-PCRにより測定した。DYRK1Aについての閾値サイクル(CT)を、GAPDHハウスキーピング遺伝子発現レベルに対して標準化し(ΔCT)、0.5ΔCT*104(任意の単位)としてプロットした。siRNA配列は、

HEK 293T細胞を、UV滅菌カバースリップ上で直接増殖させ、Ca2+指示薬色素Fura-2 AM(3μM、Molecular Probes, Eugene, OR)を室温で45分間負荷し、洗浄し、負荷培地(RPMI+10%FCS)に再懸濁した。レシオメトリックCa2+ビデオイメージングについて、カバースリップを閉鎖型槽RC-20フローチャンバー(Warner Instrument Corp., Hamden, CT)上に載せ、2mMカルシウムリンガー液(155mM NaCl、4.5mM KCl、10mM D-グルコース、5mM Hepes(pH 7.4)、1mM MgCl2、2mM CaCl2)において灌流した。Ca2+を含まないリンガー液(2mM Ca2+が2mM MgCl2で置換された)への切り換え後、細胞内Ca2+貯蔵を1μMタプシガルジンで枯渇させ、貯蔵量作動性Ca2+流入を、2mM CaCl2を含むリンガー液で細胞を灌流した後、測定した。単一細胞ビデオイメージングを、OpenLabイメージングソフトウェア(Improvision, Lexington, MA)を用いてS200倒立エピ蛍光顕微鏡(Zeiss, Thornwood, NY)で行った。Fura-2発光を、340nmおよび380nmでの励起後、それぞれ、510nmで検出し、340/380の比をバックグラウンド引き算後、5秒間隔ごとに計算した。較正値(Rmin、Rmax、Sf)は、以前に記載されているように43、キュベット測定から導いた。各実験について、約50〜100個の細胞を分析した。[Ca2+]iおよびDYRK2発現の同時測定について、ジャーカットT細胞に、10:1の比率でDYRK2 cDNAおよびeGFPを同時トランスフェクションした。トランスフェクションから48時間後、細胞を、上記のようにCa2+イメージングに用いた。[Ca2+]i、GFP-(すなわち、DYRK2-)、およびGFP+(すなわち、DYRK2+)の単一細胞分析について、細胞をゲーティングし、別々にプロットした。
S2R+細胞をトリプシン(CellGro, Herndon, VA)でディッシュから脱離させ、Ca2+指示薬色素Fluo4-AM(2μM Molecular Probes, Eugene, OR)を室温で45分間負荷し、その後、負荷培地(RPMI+10%FCS)に再懸濁した。フローサイトメトリーのCa2+測定直前に、細胞をCa2+を含まないリンガー液に再懸濁し、FACSCalibur(BD Biosciences, San Jose, CA)で分析した。細胞内Ca2+貯蔵を枯渇させるための、Ca2+を含まないリンガーにおけるタプシガルジン(3μM)の添加から180秒後、2mM Ca2+の最終濃度に達するように4mM Ca2+リンガー液を細胞へ加えた。その後、細胞Ca2+レベルをFloJoソフトウェア(Tree Star, Inc., Ashland, OR)を用いて分析した。
キナーゼ候補のヒトオルソログをコードする完全長cDNAを、Flexgene Kinase Repository(Harvard Institute of Proteomics)36またはMammalian Gene Collection(MGC, Open Biosystems)から得、N末端にFlagタグの挿入を含むpENTRY.11(Invitrogen)ベクターへサブクローニングし、その後、pDEST12.2(Invitrogen)へ再結合させた。キナーゼ不活性型DYRK2は、PCRに基づいた方法(QuikChange Site-Directed Mutagenesis, Stratagene)を用いて活性部位のATP結合ポケットにK251R点突然変異を導入することにより構築し、ポリメラーゼ忠実性を保証するために配列決定した。
NFATの細胞内局在性は、多様なシグナル伝達経路からの入力を含むシグナル積分の複雑な過程により測定される3-5。休止細胞において、NFATタンパク質は、高度にリン酸化され、細胞質に存在する;細胞内遊離Ca2+([Ca2+]i)レベルを上昇させる刺激に曝された細胞において、それらは、カルモジュリン依存性ホスファターゼカルシニューリンにより脱リン酸化され、核へ移行する3,6。カルシニューリンによるNFATの脱リン酸化は、別個のNFATキナーゼ、中でも、CK1、GSK3、およびMAPキナーゼファミリーの様々なメンバーにより相殺される3,7-10。NFATの転写活性は、N末端トランス活性化ドメインのリン酸化、共アクチベーターおよび共リプレッサーの補充、および核におけるパートナータンパク質の選択を含む追加の入力により制御される3,9,11。
における少なくとも2つのセリン残基(太字かつ下線)は、-2位または-3位にアルギニン、および+1位にプロリン(またはバリン)を有するセリン/スレオニン残基に対するDYRKキナーゼの公知の配列選択性に一致し27-29、どちらも細胞においてリン酸化されることが知られている9(図6A参照)。SP-3モチーフにおける2つの他のリン酸化セリン残基(下線)が、インビボでDYRKまたは他のNFATキナーゼについての標的であるかどうかを確立するためにさらなる研究が必要とされると思われる。
における4番目のセリン(太字)をリン酸化することによりGSK3についてプライムし、GSK3による下線の引かれたセリンの発展的なN末端リン酸化を可能にするが、一方で、DYRK2は、別のモチーフであるSP-3モチーフをリン酸化することにより制御ドメインモチーフのGSK3媒介性リン酸化を増強することを示唆している。実際、SP-2モチーフにおけるPKAにより標的とされるセリンは、細胞においてリン酸化されているのを見出されず10、DYRKによるNFATの生理学的制御についてのさらなる証拠を提供している。
本発明者らは、ショウジョウバエにおけるゲノムワイドなRNAiスクリーニングが、シグナル伝達経路の重要なメンバーが進化的に保存され、かつショウジョウバエゲノムにおいて表されているとの条件で、哺乳動物細胞におけるシグナル伝達の新規な局面を探索するための妥当かつ強力なストラテジーであることを示している。本発明者らは、純粋に脊椎動物の転写因子であるNFATの保存された制御因子を同定するための方法を用いている;本発明者らの知る限りでは、これは、このように進化的境界を交差するゲノムワイドなRNAiスクリーニングの最初の例である。ストラテジーは、脊椎動物に出現した時、Ca2+制御性NFATタンパク質により最近用いられた進化的ニッチをショウジョウバエが発展させたため、成功した。このアプローチを用いて、本発明者らは、DYRKをNFATの新規な生理学的制御因子、および最初のSP-3モチーフ指向性キナーゼであると同定している。他の哺乳動物の過程の制御の保存された局面もまた、ショウジョウバエ細胞における発展したアッセイにより首尾よく定義されると考えられる。

(図5)図5A〜5Dは、SCID患者由来の線維芽細胞におけるOrai1の発現が貯蔵量作動性Ca2+流入を回復させることを示す。図5Aは、75μM 2-APBによる、Orai1WT発現SCID線維芽細胞におけるCa2+流入の阻害を示す。図5Bは、3μM 2-APBによる、Orai1WT発現SCID線維芽細胞におけるCa2+流入の増強を示す。図5C〜5Dは、20mM Ca2+の再添加前(図5C)または後(図5D)に、加えられた2μM La3+によるOrai1WT発現SCID線維芽細胞におけるCa2+流入の阻害を示す。各実験について、〜15個から20個までのGFP陽性線維芽細胞を分析した。実験は、各プロトコールについて少なくとも3回、繰り返した。
(図6)図6A〜6Cは、NFAT制御ドメイン、およびショウジョウバエにおけるゲノムワイドなRMAiスクリーニングの結果を示す。図6Aは、刺激で脱リン酸化される保存されたリン酸化セリンモチーフ(丸)を示すNFAT1のN末端制御ドメインの概略図を示す。インビトロキナーゼアッセイに用いられたSRR1、SP2、およびSP3モチーフに対応するペプチドが表されている。下線で示されたセリン残基は、インビボでNFAT1においてリン酸化されていることが同定されており、これらは、突然変異体SP2およびSP3モチーフにおいてアラニンに突然変異している残基である。図6Bは、異種性に発現したNFATがショウジョウバエS2R+細胞においてCa2+およびカルシニューリンにより正しく制御されることを示す。ショウジョウバエS2R+細胞にNFAT1-GFP発現ベクターをトランスフェクションした。48時間後、細胞を未処理のままにしておく(Untr)、またはタプシガルジン(TG、1μM)で30分間処理し、そして、細胞由来の可溶化物を抗NFAT1での免疫ブロッティング(IB)により分析した。PおよびdePは、それぞれ、リン酸化および脱リン酸化のNFAT-GFPの移動位置を指す。図6Cは、一次スクリーニングの結果の作表を示す。
(図7)図7A〜7Cは、NFATリン酸化についてのショウジョウバエS2R+細胞RNAiスクリーニングにおいて同定した候補キナーゼのスクリーニング、およびNFATの負の制御因子としてのDYRKの同定を示す。図7Aは、候補キナーゼの過剰発現した哺乳動物相同体のNFAT制御ドメインを直接的にリン酸化する能力を示す。選択したショウジョウバエキナーゼのFLAGタグ付き哺乳動物の相同体をHEK293細胞において発現させ、免疫精製したキナーゼをGST-NFAT1(1〜415)のリン酸化についてのインビトロのキナーゼアッセイを用いて試験した。リン酸化レベルは、短時間(上部パネル)または長時間(中央パネル)曝露のいずれかでのオートラジオグラフィーにより評価した。各キナーゼの発現は、抗FLAG抗体を用いる免疫ブロッティング(IB)により検証した。試験したキナーゼは以下のとおりである;レーン1、CK1α;レーン2、CK1ε;レーン3、Bub1;レーン4、STK38;レーン5、STK38L;レーン6、CDC42BPA;レーン7、ARAF;レーン8、PRKG1;レーン9、SGK;レーン10および11、CSNKA1およびCSNKA2(CKIIアイソフォーム);レーン12、SRPK1;レーン13、DYRK2;レーン14、ALS2CR7;レーン15、IRAK4。Bub1は、後で、>10個の予想されるオフターゲットのため、本発明者らの候補リストから除外した(実施例3)。図7Bは、DYRK2の過剰発現がNFAT1のカルシニューリン媒介性脱リン酸化を遮断することを示す。各キナーゼを、NFAT-GFPと共にHEK293細胞へ同時トランスフェクションした;18時間後、細胞を、2mM CaCl2の存在下で1μMイオノマイシンで刺激した。可溶化液をNFAT1抗体を用いて免疫ブロットした。キナーゼの相対的発現レベルは、抗FLAG抗体を用いる免疫ブロットにより測定し、図6A(下部パネル)に示したものと同一であった。図7Cは、内因性DYRK1Aの枯渇がNFAT活性化を増強することを示す。Haタグ付きNFAT1-GFPを安定に発現するHeLa細胞に、対照siRNAまたはDYRK1A特異的siRNAをトランスフェクションした。4日後、細胞を、1μMタプシガルジン(TG)、または1μMタプシガルジン(TG)、続いて、示された時点について20nM CsAで刺激した;可溶化液を抗HA抗体を用いてNFAT-GFPについて免疫ブロットした(左)。DYRK1A mRNAレベル(右)を3日および4日後、リアルタイムPCRにより評価した。si対照、スクランブル対照siRNA;siDYRK1A、DYRK1A特異的siRNA。結果は3つの独立した実験の平均および標準偏差を示す。

Claims (48)
- 以下の段階を含む、NFAT制御タンパク質(NFAT regulator protein)を調節する作用物質を同定する方法:
少なくとも1つの試験作用物質を、少なくとも1つのNFAT制御タンパク質またはその断片もしくは誘導体を含む組換え細胞に接触させる段階;
NFAT制御タンパク質またはその断片もしくは誘導体についての活性、相互作用、発現、または結合に対する試験作用物質の効果を評価する段階;および
NFAT制御タンパク質またはその断片もしくは誘導体についての活性、相互作用、発現、または結合に対する効果を有する試験作用物質を同定する段階であって、同定された試験作用物質がNFAT制御タンパク質を調節する作用物質として特徴付けられる、段階。 - NFAT制御タンパク質が、ORAI1(SEQ ID NO:1)、ORAI2(SEQ ID NO:2)、ORAI3(SEQ ID NO:3)、DYRK1A(SEQ ID NO:4)、DYRK1B(SEQ ID NO:5)、DYRK2(SEQ ID NO:6)、DYRK3(SEQ ID NO:7)、DYRK4(SEQ ID NO:8)、およびDYRK6(SEQ ID NO:9)からなる群より選択される少なくとも1つのNFAT制御因子(NFAT regulator)によりコードされる、請求項1記載の方法。
- NFAT制御タンパク質が表Iに列挙された遺伝子の少なくとも1つによりコードされる、請求項1記載の方法。
- 試験作用物質の効果を評価する段階が、ORAI1(SEQ ID NO:1)、ORAI2(SEQ ID NO:2)、ORAI3(SEQ ID NO:3)、DYRK1A(SEQ ID NO:4)、DYRK1B(SEQ ID NO:5)、DYRK2(SEQ ID NO:6)、DYRK3(SEQ ID NO:7)、DYRK4(SEQ ID NO:8)、またはDYRK6(SEQ ID NO:9)によりコードされるNFAT制御タンパク質に特異的に結合する抗体を用いる事を含む、請求項2記載の方法。
- 細胞の原形質膜を横断する電流に対する試験作用物質の効果を評価する段階をさらに含む、請求項1記載の方法。
- 電流が、細胞を横断する一価陽イオンまたは二価陽イオンの流束(flux)によるものである、請求項5記載の方法。
- 細胞内における細胞内カルシウムに対する試験作用物質の効果を評価する段階をさらに含む、請求項1記載の方法。
- 以下の段階をさらに含む、請求項7記載の方法:
細胞内における細胞内カルシウムに対する効果を有する試験作用物質を同定する段階であって、それによって、同定された試験作用物質が、細胞内カルシウムを調節する作用物質およびNFAT制御タンパク質を調節する作用物質として特徴付けられる、段階。 - 細胞が、少なくとも1つの異種性NFAT制御タンパク質またはその断片もしくは誘導体を含む、請求項1記載の方法。
- 細胞が、少なくとも1つのNFAT制御タンパク質またはその断片もしくは誘導体をコードする異種性核酸を含む、請求項1記載の方法。
- 細胞が、少なくとも1つのNFAT制御タンパク質またはその断片もしくは誘導体を過剰発現するかまたは過小発現する、請求項1記載の方法。
- 以下の段階を含む、細胞内カルシウムを調節する作用物質を同定する方法:
少なくとも1つの試験作用物質を、少なくとも1つのNFAT制御タンパク質またはその断片もしくは誘導体を含む組換え細胞に接触させる段階;
細胞内における陽イオンもしくは二価陽イオンの細胞内量もしくは濃度に対する、または細胞へのイオン流入に対する試験作用物質の効果を評価する段階;および
細胞内における陽イオンもしくは二価陽イオンの細胞内量もしくは濃度に対する、または細胞へのイオン流入に対する効果を有する試験作用物質を同定する段階であって、それによって、同定された試験作用物質が細胞内カルシウムを調節する作用物質として特徴付けられる、段階。 - 細胞内陽イオンがカルシウムである、請求項12記載の方法。
- 試験作用物質の効果を評価する段階が、細胞質におけるカルシウムレベルをモニタリングする事、細胞内カルシウム貯蔵におけるカルシウムレベルをモニタリングする事、カルシウム移動をモニタリングする事、またはカルシウム流入媒介性事象をモニタリングする事を含む、請求項13記載の方法。
- NFAT制御タンパク質またはその断片もしくは誘導体についての活性、相互作用、発現、または結合に対する試験作用物質の効果を評価する段階をさらに含む、請求項12記載の方法。
- NFAT制御タンパク質が、ORAI1(SEQ ID NO:1)、ORAI2(SEQ ID NO:2)、またはORAI3(SEQ ID NO:3)、DYRK1A(SEQ ID NO:4)、DYRK1B(SEQ ID NO:5)、DYRK2(SEQ ID NO:6)、DYRK3(SEQ ID NO:7)、DYRK4(SEQ ID NO:8)、またはDYRK6(SEQ ID NO:9)からなる群より選択される少なくとも1つのNFAT制御因子によりコードされる、請求項15記載の方法。
- 細胞内カルシウムを調節する作用物質が、NFAT制御タンパク質を調節する作用物質としてさらに特徴付けられる、請求項15記載の方法。
- 組換え細胞が、少なくとも1つの異種性NFAT制御タンパク質またはその断片もしくは誘導体を含む、請求項12記載の方法。
- 組換え細胞が、少なくとも1つのNFAT制御タンパク質またはその断片もしくは誘導体をコードする異種性核酸を含む、請求項12記載の方法。
- 組換え細胞が、少なくとも1つのNFAT制御タンパク質またはその断片もしくは誘導体を過剰発現する、請求項12記載の方法。
- 組換え細胞がホメオスタシス異常(dyshomeostasis)を示す、請求項12記載の方法。
- 組換え細胞がカルシウムホメオスタシス異常を示す、請求項12記載の方法。
- 以下の段階を含む、NFAT制御因子の機能を調節する作用物質についてスクリーニングするための方法:
レポータータンパク質に機能的に連結している少なくとも1つのNFAT制御ドメインまたはその断片もしくは誘導体、および少なくとも1つのNFAT制御タンパク質またはその断片もしくは誘導体をコードする異種性核酸を含む少なくとも1つのベクターを含む細胞に、少なくとも1つの試験作用物質を投与する段階;
ベクターによりコードされる少なくとも1つの発現産物の細胞内局在をモニタリングする段階であって、それによって、発現産物の細胞内局在に対する効果を有する試験作用物質が、NFAT制御因子の機能を調節する作用物質として特徴付けられる、段階。 - NFAT制御因子の機能を調節する作用物質が、発現産物の細胞質局在または核局在と関連している、請求項23記載の方法。
- 細胞が休止状態下にある、請求項23記載の方法。
- 細胞がカルシウム調節剤で刺激される、請求項23記載の方法。
- 細胞がタプシガルジンまたはイオノマイシンで刺激される、請求項26記載の方法。
- 以下の段階を含む、被験体において未解明の免疫不全を診断するための方法:
ORAI1(SEQ ID NO:1)、ORAI2(SEQ ID NO:2)、またはORAI3(SEQ ID NO:3)、DYRK1A(SEQ ID NO:4)、DYRK1B(SEQ ID NO:5)、DYRK2(SEQ ID NO:6)、DYRK3(SEQ ID NO:7)、DYRK4(SEQ ID NO:8)、DYRK6(SEQ ID NO:9)、または表Iに列挙された遺伝子のいずれかに対応する被験体由来の遺伝子において、少なくとも25個の連続したヌクレオチドを配列決定する段階;および
被験体の遺伝子の配列をその遺伝子の野生型配列と比較する段階であって、その遺伝子の野生型配列との差異が、被験体の遺伝子が免疫不全の原因であることを示す、段階。 - 比較する段階が、被験体由来の生物学的試料を得る事、生物学的試料中のDNAを配列決定する事、および生物学的試料から得られたDNA配列を野生型配列と電子的に整列させる事を含む、請求項28記載の方法。
- 差異が、ORI1(SEQ ID NO:1)のコード配列の271位におけるCからTへのヌクレオチド突然変異を含む、請求項28記載の方法。
- 未解明の免疫不全がNFAT活性の制御における欠陥と関連している、請求項28記載の方法。
- 差異がスプライス部位における突然変異を含む、請求項28記載の方法。
- 差異が非同義(nonsynonymous)変異を含む、請求項28記載の方法。
- NFAT制御タンパク質に関連した疾患または障害を示す生物体に対する試験作用物質の効果を評価する段階;および
疾患または障害に関連した生物体の表現型に対する効果を試験作用物質が有する場合には、該試験作用物質を、NFAT制御タンパク質に関連した疾患または障害を処置または予防するための作用物質であると同定する段階
を含む、NFAT制御タンパク質に関連した疾患または障害を処置または予防するための作用物質を同定するための方法であって、
該試験作用物質が、少なくとも1つのNFAT制御タンパク質またはその断片もしくは誘導体の活性、相互作用、発現、または結合を調節する、方法。 - 生物体が、カルシウムホメオスタシス異常を示す1つまたは複数の細胞を含む、請求項34記載の方法。
- NFAT制御タンパク質に関連した疾患または障害が、リウマチ様関節炎、炎症性腸疾患、同種または異種移植拒絶、移植片対宿主病、再生不良性貧血、乾癬、紅斑性狼瘡、炎症性疾患、MS、I型糖尿病、喘息、肺線維症、強皮症、皮膚筋炎、シェーグレン症候群、心膜切開後症候群、川崎病、橋本甲状腺炎、グレーブス病、重症筋無力症、尋常性天疱瘡、自己免疫性溶血性貧血、特発性血小板減少症、慢性糸球体腎炎、グッドパスチャー症候群、ヴェグナー肉芽腫症、多発性硬化症、嚢胞性線維症、慢性再発性肝炎、原発性胆汁性肝硬変、ブドウ膜炎、アレルギー性鼻炎、アレルギー性結膜炎、アトピー性皮膚炎、クローン病、潰瘍性結腸炎、結腸炎/炎症性腸症候群、ギラン・バレー症候群、慢性炎症性脱髄性多発根ニューロパチー、湿疹、および自己免疫性甲状腺炎、移植片拒絶、後天性免疫不全症、分類不能型免疫不全症、心筋肥大、重症複合型免疫不全症、拡張型心筋症、過剰または病理的骨吸収、過剰脂肪細胞分化、肥満、または潜伏ウイルスの再活性化である、請求項34記載の方法。
- ORAI1(SEQ ID NO:1)、ORAI2(SEQ ID NO:2)、またはORAI3(SEQ ID NO:3)、DYRK1A(SEQ ID NO:4)、DYRK1B(SEQ ID NO:5)、DYRK2(SEQ ID NO:6)、DYRK3(SEQ ID NO:7)、DYRK4(SEQ ID NO:8)、またはDYRK6(SEQ ID NO:9)によりコードされるNFAT制御タンパク質に特異的に結合する抗体。
- NFAT制御因子またはその断片もしくは誘導体をコードする核酸配列を含む組換えベクターを含む宿主細胞を、NFAT制御因子の発現に適した条件下で培養することによって、NFAT制御タンパク質が産生される、請求項37記載の抗体。
- 少なくとも1つの異種性NFAT制御タンパク質またはその断片もしくは誘導体を含む、および/またはNFAT制御タンパク質またはその断片もしくは誘導体をコードする少なくとも1つの異種性核酸を含む、単離された細胞;ならびに
細胞の原形質膜を横断する電流をモニタリング、検出、または測定するために用いられるモニタリング剤
を含むシステム。 - 電流が、細胞を横断する陽イオンまたは二価イオンの流束によるものである、請求項39記載のシステム。
- モニタリング剤が、細胞内における細胞内カルシウムに対する試験作用物質の効果をモニタリングするために用いられる、請求項40記載のシステム。
- モニタリング剤が、カルシウム流入媒介性事象をモニタリング、検出、または測定するために用いられる、請求項40記載のシステム。
- 少なくとも1つの哺乳動物NFAT制御タンパク質またはその断片もしくは誘導体を過剰発現する組換え細胞;および
カルシウム流入媒介性事象をモニタリング、検出、または測定するために用いられるモニタリング剤
を含むシステム。 - NFAT制御因子が、ORAI1(NM_032790)、ORAI2(BC069270)、ORAI3(NM_152288)、DYRK1A(NM_001396)、DYRK1B(NM_004714)、DYRK2(NM_003583)、DYRK3(NM_003582)、DYRK4(NM_003845)、およびDYRK6(NM_005734)からなる群より選択される少なくとも1つの核酸によりコードされる、請求項43記載のシステム。
- 少なくとも1つの異種性NFAT制御タンパク質またはその断片もしくは誘導体を含む、および/またはNFAT制御タンパク質またはその断片をコードする少なくとも1つの異種性核酸を含む、組換え細胞。
- 少なくとも1つの哺乳動物NFAT制御タンパク質またはその断片もしくは誘導体を過剰発現する組換え細胞。
- 以下の段階を含む、カルシウムシグナル伝達に関連した疾患または障害を処置または予防するための作用物質を同定するための方法:
a. 該疾患または障害を示す生物体に対する試験作用物質の効果を評価する段階;および
