Abstract
本発明は核酸の検出方法を提供するものであって、当該方法は、サンプルがマイクロ流体の流路内を往復し、核酸の量が核酸のUV吸収に基づいて決定されるマイクロ流体デバイスにおいてPCR反応を実施することを含む。
Description
核酸及び核酸の改変の検出は多くの用途を有しており、疾病の診断において特に重要なツールである。ほとんどの技術が、非常に少量の複雑な遺伝物質の増幅を可能にするポリメラーゼ連鎖反応(PCR)を利用する。
PCRの出現は多様な生物の遺伝的構造に関する研究の進歩を大きく加速させてきた。PCRは、あらゆる核酸配列をインビトロでしかも大量に生成することを可能にする、酵素触媒性の反応である。1986年に最初に報告されて以来、PCRは基礎的な分子生物学、ゲノム配列決定、臨床研究、及び進化の研究において必須のツールとなっている。PCRの成功の理由はその簡易性にある。高温(およそ95℃)では、二本鎖の標的DNAは変性し、すなわち2本の一本鎖にほどける。プライマーとして知られている一本鎖DNAの合成配列を使用して鎖の増幅対象領域を挟み、一方のプライマーは一方のDNA鎖に相補的であり(標的領域の開始部分にある)、第二のプライマーはもう一方のDNA鎖に相補的である(標的領域の最終部分にある)。プライマーは、その場の温度が50℃と65℃の間に低下すると一本鎖にアニーリングする。この後にわずかに高い温度(約72℃)での「伸長」が続き、そこでは、遊離デオキシヌクレオチド三リン酸の存在下で、DNAポリメラーゼ酵素の触媒作用により相補鎖が各プライマーから生じる。この3工程のプロセスが1つのPCRサイクルを構成しており、n回反復すると、元のDNA鎖の2n個のコピーが得られることになる。
PCRを実施するための従来の機器(サーマルサイクラー)は概念的には簡易であるが、増幅の速さ及び効率を制限する多くの技術的弱点を有する。効率的な増幅のための基本的な必要条件は迅速な熱伝達である。加熱時には熱を素早くサンプルに伝達し、冷却時には素早く取り除くことのできる、低熱容量のシステムを有することが望ましい。ほとんどの従来のサーマルサイクラーは大きな熱質量を有しており、その結果、必要とする電力が大きく、加熱及び冷却の速度が遅い。その結果、全反応時間が典型的には90分を超える。変性工程及びアニーリング工程は正確な温度に達するとすぐに生じるため、また伸長はポリメラーゼ酵素の処理能力(1秒当たり50塩基と500塩基の間)にのみ制限されるため、機器の熱質量が減少すれば全反応時間を大幅に短縮することができる。近年、この発想を考慮して、微細加工されたPCRシステムが開発されてきている。当該システムは、構造は様々であるが、全てが、熱質量を減少させることで迅速な加熱及び冷却を容易にしており、反応時間を数分間と短いものにしている。機器の小型化のための標準的なアプローチは、熱質量を減少させるためにシステムの寸法を直接縮小することを伴う。初期の研究はこの概念を用いて、モノリシック基板の外側に搭載された、または内部に一体化された抵抗加熱器を用いて平衡反応チャンバ(100pLと50μLの間の容積を有する)に熱サイクルを実施する、微細加工されたデバイスを作出した。この通常のアプローチを用いることで、反応の速さ、分析のスループット、及び反応効率の著しい向上が実証されている。
従来のPCR法はアガロースゲル電気泳動により産物を分析する。不都合な点は、この方法は時間のかかるものであり、且つ低い感度を示すことである。基礎的なPCR法はさらに開発され、現在は、配列特異的PCRの産物をリアルタイムで検出するために利用することができる方法がある。このような方法の1つは、PCR産物の検出がレポーターの蛍光の検出に基づく、TaqMan(登録商標)アッセイ(Holland. P.ら、1991年、PNAS、88、7276〜7280頁、「Detection of specific polymerase chain reaction product by utilizing the 5'→3' exonuclease activity of Thermus aquaticus DNA polymerase.」)である。しかし、その利点にもかかわらず、TaqMan(登録商標)アッセイもまたいくつかの不都合な点を有する。例えば、プローブの構築のための配列データが利用できなくてはならない。従って、異なる配列の検出のために異なるプローブを合成する必要がある場合、アッセイのコストは特に高い。一般に用いられるリアルタイムPCRのための別の方法は、二本鎖DNAに特異的に結合するが一本鎖DNAには特異的に結合しない染料(SYBR Green(登録商標)、Lipsky, Rら、2001年、Clinical Chemistry 47、635〜644頁、「DNA Melting Analysis for Detection of Single Nucleotide Polymorphisms」)を使用する。しかし、この方法は、染料が非特異的であり偽陽性シグナルを生成することがあるという不都合な点を有する。他の方法はモレキュラービーコン(molecular beacons)またはスコルピオン(scorpions)を用いるが、TaqMan(登録商標)アッセイと同様に、これらの方法は複雑で高価である。
WO03/102238は、PCR混合物のUV吸光度を測定することによるリアルタイムPCR法に関する。WO03/102238の内容は参照することにより本明細書に組み込まれる。WO03/036302は、タンパク質のフォールディング及びアンフォールディングをモニターする方法、並びにタンパク質の温度依存性の立体配置を分析するための装置を開示している。WO03/036302の内容もまた参照することにより本明細書に組み込まれる。
遺伝的変異のため、90%超の薬剤が、所与の個体群の30%〜50%においてしか効果的な治療を示さない。この深刻な問題を低減するため、医療産業はより個別の治療管理体制に関心を向けている。現在、この個別化医療の時代は、ポイントオブケア(PoC)で利用できる診断技術がないことにより遅滞している。核酸の改変には短い縦列反復(STR)及び一塩基多型(SNP)が含まれる。SNPは、ゲノム配列における1つのヌクレオチド(A、T、CまたはG)が変わると生じるDNA配列の変異である。変異がSNPと考えられるためには、変異は個体群の少なくとも1%において生じなければならない。対立遺伝子頻度は、異なる個体群間でも大きく変化する。ヒトの全ての遺伝的変異の約90%を構成するSNPは、30億塩基のヒトゲノムにわたって100から300塩基ごとに生じる。SNPは通常、2つの形態で存在するのみなので、より稀な対立遺伝子は突然変異またはマイナー対立遺伝子と呼ばれ、最も共通の対立遺伝子は野生型対立遺伝子と呼ばれる。SNPは主として2対立遺伝子である(すなわち、1つの遺伝子座に2つの考えられる対立遺伝子がある)が、3対立遺伝子(すなわち、2つの独立した突然変異事象が同時に生じている)でもあり得る。3つに2つのSNPが、シトシン(C)のチミン(T)による置換を伴う。SNPはゲノムのコード領域(遺伝子)及び非コード領域の両方で生じ得る(エクソン性またはイントロン性)。
99%超のヒトDNA配列が個体群を通して同一であるが、DNA配列における変異は、どのようにヒトが疾病、病原体、及び治療に応答するかについて重要な影響を有し得る。このことにより、SNPは、生物医学的な調査及び医薬品または医療診断の開発にとって非常に価値のあるものとなっている。従って、SNPの検出のための効率的な、精密な、安価な、且つ利用しやすい方法の提供は、非常に価値のあるものとなり得る。SNPを分析するために用いられる現在の方法は、PCRと、その後の配列決定、マイクロアレイ、及び質量分析とを含む。しかし、特にマイクロアレイ及び質量分析は複雑で高価である。3'ミスマッチSNPスコアリングに基づく1つのPCR法は、GALIOSシステム(Weber, S.ら、2002年、British Journal of Haematology 116、839〜843頁:「Genotyping of human platelet antigen-1 by gene amplification and labeling in one system and automated fluorescence correlation spectroscopy」)により例示される。このシステムでは、2つのプライマーが推定産物の5'末端で用いられ、そのうちの1つが3'のミスマッチを有している。上側の野生型の鋳型は、ポリメラーゼにより3'方向に伸長され得る相補的プライマーを有している。SNPを含む下側の鋳型はプライマーを完全にはアニーリングできず、3'のミスマッチを生じさせる。これは、実際に存在する場合、産物を著しく減少させるであろう。GALIOS及び他の関連するシステムでは、産物は付着した蛍光染料により検出される。Taqman(登録商標)及びSYBRグリーンシステムのような他の標識されたシステムも存在し、それらの不都合な点については上述している。従って、SNPを分析するための代替的な、且つ改善された方法が必要とされている。
ほとんどのDNA分子は変性すると260nmでの吸光度において1.4の相対的な増加を示し、これは濃色効果として知られている。この効果は一本鎖DNA(ssDNA)と二本鎖DNA(dsDNA)との異なった構造に起因する。ssDNAはdsDNAよりも260nmで約30%多くの光子を吸収する。
吸光度の増加は、吸収の大きいプリン環及びピリミジン環の露出により生じるが、これらはdsDNAにおいては、水素結合部分を形成して、内部でらせん状に積み重なっている。
マイクロ流体デバイスは様々なところで記載されている(Koppら、Science 280、1998年)。例えば、Munchowらは、PCR産物が蛍光またはゲル電気泳動により検出される、マイクロ流体PCR法を開示している(Munchow Gら、2005年、Expert Rev. Mol. Diagn. 5、613〜620頁、「Automated chip-based device for simple and fast nucleic acid amplification」)。

本発明者は、半連続的フローPCRを用い、且つ外因性の標識をPCR産物内に組み込む必要がないため標識を利用しない固有のイメージングに基づく、核酸検出のための代替的な方法を見出した。
本発明はまた、対立遺伝子特異的プライマー及びPCRに基づいた、SNPの検証のための方法も提供する。このような方法の作出により、医療従事者が患者から血液サンプルを採取して、患者の遺伝情報に基づいた薬物治療のインフォームドチョイスを迅速に(数分内に)行うことが可能になるであろう。
第一の態様において、本発明は核酸の検出方法に関し、当該方法は、
a)核酸サンプルを含む溶液を、温度制御し、且つUV照射した流路に沿って移動させ、溶液の流れ方向を複数回変化させる、核酸サンプルを増幅する工程と、
b)核酸のUV吸収を測定する工程
とを含む。

別の態様では、本発明は、少なくとも1つのマイクロ流路、磁性流体の作動手段、加熱要素、UV光源、及び検出器を含む、核酸の存在を決定するための装置に関する。

従って、本発明によると、核酸サンプルを含む溶液は流路内を後方及び前方へ移動する。従って、溶液は非直線状及び非連続的に移動する。サンプルは流路の異なる部分の間を往復する。
増幅は溶液が往復している間に実施される。これは流路の異なる部分に異なる温度を適用することによって達成される。
本方法により、サンプル内に存在する核酸の量を決定することが可能になる。
本発明によると、核酸サンプルという用語は、DNAまたはRNAを含むサンプルをいう。DNAはcDNAを含み得、RNAはmRNAまたはsiRNAを含み得る。好ましくは、核酸サンプルは、一本鎖のDNAもしくはRNA、または二本鎖のDNAもしくはRNAを含む。一実施形態において、核酸は天然の二次構造要素を含むか、または変性した形態である。一実施形態において、核酸サンプルは、微生物、動物または植物から単離された核酸を含み得る。別の実施形態において、核酸は合成配列、例えばベクターの一部またはオリゴヌクレオチドである。さらなる実施形態において、核酸サンプルは動物もしくは植物の細胞または微生物細胞を含む。
一実施形態において、増幅はPCRによる。本発明によると、PCRとは、DNAの増幅のためのポリメラーゼ連鎖反応をいう。典型的には、PCR反応は、異なる温度で実施される、変性工程、アニーリング工程、及び伸長工程を含む。変性、すなわち2つの相補鎖の分離は、約95℃の標準温度を典型的には必要とする。アニーリングに必要とされる温度は、用いる特定のプライマーに依存するものの典型的には約54℃で実施されるが、しかしプライマーの塩基組成に依存して変化する。その温度は40℃と60℃の間であり得る。プライマー分子の伸長は典型的には約72℃で実施される。当業者には、実施するPCRのタイプ及び用いるプライマーに従って用いる温度が変化することが理解されよう。PCRには、反応を触媒するためのポリメラーゼ酵素の存在が必要である。典型的には、DNA PolI、Taqポリメラーゼ、またはあらゆる他の熱安定ポリメラーゼ酵素を用い得る。
PCRはリアルタイムで実施し得る。
プライマーは、標的核酸内にある相補的配列にアニーリングする短いオリゴヌクレオチドである。典型的には、プライマーは15から30ヌクレオチド長である。反応で用いるオリゴヌクレオチドプライマーの配列は目的の配列に依存する。必要ならば、縮重プライマーを本発明の方法で用いてもよい。流路の精密な温度制御により、用いる特異的プライマーに必要な温度を正確に調節することが可能になる。
本発明の一実施形態による方法では、プライマーはまた、さらなるレベルの検出を提供するために標識することもできる。このような標識は当業者に既知であり、蛍光染料または放射性標識が含まれる。
従って、一実施形態において、溶液はさらにPCR混合物を含む。本発明によるPCR混合物またはPCR試薬という用語は、PCR反応を実施するために典型的に必要な構成要素を含む混合物をいう。
このような混合物は、緩衝液、少なくとも1つのオリゴヌクレオチドプライマーのセット、dNTP(4つのDNA塩基であるアデニン(dATP)、チミン(dTTP)、グアニン(dGTP)及びシトシン(dCTP)から成るヌクレオチド)、並びにポリメラーゼを典型的には含む。PCR混合物の構成要素並びにそれらの濃度及び用いる条件が、実施するPCR反応のタイプに従って変化し得るということは、当業者には自明であろう。
当業者には、本発明による方法が、ホットスタートPCR、インバースPCR、RT-PCR、RACE、ネステッドPCR、非対称PCR、及び他のPCR法といった、異なるタイプのPCR反応に関し得るということが承知されよう。従って、本発明はまた、ホットスタートPCR、インバースPCR、RT-PCR、RACE、ネステッドPCR、非対称PCRを実施する、本発明による方法にも関する。
本発明による温度制御という用語は、流路の全長にわたって温度を制御すること、すなわち、温度プロファイルを流路の全長に、ひいては流路内の溶液に適用することをいう。本発明によると、温度を制御することで、様々な温度を流路の全長にわたって溶液に適用することができる。従って、流路の全長にわたるあらゆる所定のポイントで、特定の温度を流路(ひいては溶液)に適用することができる。従って、流路の全長にわたる、考えられる各位置で、特定の温度を溶液に適用することが可能である。流路は温度制御要素としてPeltierセルを含み得る。温度分解能は実験条件に適合するよう調節することができるが、1mm当たり1℃以下の分解能を用いてもよい。一実施形態において、流路はマイクロ流路であり、例えば、Koppら、Science 280、1998年に記載されているような、チップ上のマイクロ流路である。
一実施形態において、流路の温度は40℃から110℃の範囲内である。
別の実施形態において、異なる温度区域を流路の別の部分に適用する。好ましくは、第一の温度区域における温度は40℃から60℃の範囲内であり、第二の温度区域においては約72℃であり、第三の温度区域においては約95℃である。
本発明によると、図2に示されるように例えば左右交互の圧力を適用することによって、マイクロ流体の流路において静的な加熱区域上で、DNAサンプル及びPCR試薬を含む溶液(「サンプルプラグ」)を前後に分流させることにより、増幅は達成される(Munchowら)。本発明は、静的なまたはウェルベースの反応器のサイクリングの柔軟性と、連続的フローPCRのマイクロ構造にともなう迅速な温度推移(及び非常に低い熱質量)とを組み合わせる。当技術分野で既知のマイクロ流体法を用いる連続的フローPCRとは異なり、本発明の方法は、従って、核酸のUV吸収の測定に基づく核酸の検出と組み合わせた半連続的フローPCRを提供する。
本発明の方法によると、PCRの効率についての情報は濃色効果に関するデータから得られる(図4)。従って、核酸サンプルのUV吸収を約260nmで測定し、増幅の間の核酸の産生をモニターすることができる。
95℃での変性段階の間の産物プラグのイメージングはこの特徴により増強され得る。dsDNA産物の増加は滑らかな指数曲線によって表されないと想定され、むしろ、それが濃色効果に関する特徴を有し、その結果DNAが変性するにつれ追加の吸光度が生じることが予想される。図5は産物の曲線がどのようになるかを表したものを示す。
一連の工程の特徴は、産物の量が増加するにつれ生じるであろう。72℃の区域におけるこの新たなdsDNA産物は、生成されるにつれて260nmでの吸光度を増大させるであろう。反応プラグが95℃の区域に移動すると、それはssDNAに融解し、産物の量及び濃色効果の両方に起因したシグナルの増大を示す。反応プラグが72℃の区域まで戻ると、鎖の復元に起因して吸光度が最初わずかに低下するが、しかしこれはdsDNAの増加によりすぐに補われる。これは多くの考えられる結果の1つである。工程の特徴が生じないか、または95℃での増大により産物曲線の角度が非常に急になることがある。得られた情報がデータサンプリングの速さに依存しているであろうと考えることも重要である。イメージングシステムは、512ピクセルのフォトダイオードアレイまたは電荷結合素子(CCD)の要素にわたって20ヘルツ(Hz)以上の周波数で作動し得る。しかし、用いるクロック速度が非常に速く、その結果超高速(数10マイクロ秒)であるため、イメージングもまた可能であり、イメージングはシステムの化学動力学に適合するよう行うことができる。多重検出器を用いることもでき、システムが同調可能となり、且つ変性区域内で考えられる最良のデータを得ることができるよう、往復するプラグの速さを非常に正確に制御した。
PCR産物の濃度または量は、UV吸収の測定に基づいてモニターする。光源と光検出器との間に核酸を通すことで濃度を検出する。例えば、検出器としてUV増感性のフォトダイオードアレイ(PDA)または電荷結合素子(CCD)を用いることで、DNAバンドが流路を移動する速さを決定することができ、これにより、サンプルのプラグ長の測定が可能になる。
一実施形態において、本方法はサンプル内の核酸の速度を測定することを含む。分子は512ピクセル以上の1つまたは複数のフォトダイオードアレイを横断するため、分子の速度は多重検出を用いて確証することができる。このことにより、時空相関の確証が可能になる。流路内の分子の位置は、核酸のUV吸収に基づいて検出される。260nmで分子が最大に吸収する、重水素ランプまたはUVダイオードレーザーからの紫外線光を分子に照射し、それにより、それらが横断するPDA検出器のピクセルでのシグナルを低下させる。従って、核酸は、存在量の測定を得るために用いることができるシグナルにより同定され得る。サンプルが所定の位置に達するために必要な速度を計算することも可能であり、当該サンプルの速度に基づいて、存在量を決定することができる。速度に基づいたシグナル処理のための、本発明で用いられる方法は、WO96/35946、WO02/12876、及びWO02/12877に記載されている。
マルチピクセル検出もまた、適切な時空平均化が実施されるとシグナル対ノイズ比を増大させる。PDAを流路の両端の近傍及び温度区域の近傍に置くことで、DNAが示す温度依存性のあらゆる特徴をモニターすることが可能になる。検出器の厳密な配置、すなわちそれらの数及びそれらの位置は、PCRプラグの速さ及びサイズにより生じるイメージングの制約に応じたものとなろう。サンプルプラグは、移動し始める前はt=0に位置する。このことにより、速度に基づいたシグナル処理の適用が可能になる。提案するシステムにおいて、プラグはそれが存在する温度区域に応じてシステム内において静止しているため、t=0は確立することが比較的容易である。1つの配置において、検出器は加熱要素に隣接して位置するか、または直角に位置する。
従って、一実施形態において、本発明の方法は速度に基づいたシグナル処理を含む。
提案する方法の利点の1つは、それがリアルタイムの測定の実施を可能にすることである。従来のPCR技術を用いて十分な増幅をするために必要なPCRサイクルの数は少なくとも30である。本発明の方法は、検出可能な量の産物を産生するために必要なサイクルの数を著しく減少させることができる。典型的な300bpのPCR産物の、10回という少ないサイクルが、LFII技術を用いてイメージングされている(図7)。本発明によると、PCRは10から40回、好ましくは20回のサイクルを用いて実施される。サンプル及びPCR混合物を含む溶液、すなわちPCRサンプルプラグの流れ方向が変化する回数は、従って、用いるPCRのサイクルに依存する。
さらに、通常使用される最良のPCR機を用いても、PCR反応は少なくとも1時間を要するであろう。より高価な機械であればより少ない時間で反応を行うことができるが、それらは通常実験室で利用されるものではない。本発明の方法によれば反応時間を著しく短縮することができる。
さらに、本方法は典型的に少ない容量のサンプル及び試薬を必要とするが、しかしnLの範囲に限定されるわけではない。少ない瞬間容量及び関連する熱質量により、サンプルプラグは、異なる温度区域に運ばれると10msと100msの間のいくらかの時間規模で熱平衡する。その結果、システムのサイクルの速さに対する熱限界は、マクロ流体及びマイクロ流体のバッチのサイクラーの両方と比較して大きく減少する。さらに、PCR試薬は比較的短い(連続的フローシステムと比較して)マイクロ流路内に含まれるため、DNAポリメラーゼのような重要な反応構成要素の、流路の表面への吸着はより少ない。
本発明の方法で用いられるマイクロ流体の流路は、SU-8フォトリソグラフィー、及びポリジメチルシロキサン(PDMS)、TOPAS、または他のUV透過性プラスチックのチップの微細加工などの、最先端のマイクロ流体技術、レーザーもしくは機械ツール、または複数のUV透過性のガラスもしくは水晶材のウェットエッチングを用いて作製し得る。
本発明は、流路内での溶液の流れ方向を操作するために圧力または磁性流体の作動のいずれかを用い得る。磁性流体の作動の場合、溶液はさらに油及び磁性ナノ粒子を含む。100mbarの圧力を適用することができる。圧力作動装置の実装及び一体化は容易であるが、磁性流体プラグの磁性操作は、図2に示されるような環状のマイクロ流体システムにおいて実施される場合、サンプルプラグの動き及び試薬の拡散の最適な制御を提供すると考えられる。いずれかのアプローチを用いることで、5分間以内の20回と60回の間の熱サイクルを通して、サンプルプラグを駆動し得ることが見込まれる。
本発明による、標識を利用しない固有のイメージングによって、追加の標識を含める必要なく、産物のリアルタイムの検出が可能になる。さらに、本発明によれば、検出器を用いることで産物の量を定量することもできる。
好ましくは、本発明の方法は、複数のマイクロ流路を有するデバイスを用いるように実施することができる。
別の態様において、本発明は、ヌクレオチドの改変の存在を決定するための本方法の使用を提供する。ヌクレオチドの改変は、ヌクレオチドまたは塩基対の置換、欠失、または付加であり得る。本発明によると、1つまたは複数のヌクレオチドの改変を検出することができる。特に、改変はSTR、SNP、標的化した遺伝子改変(GM)工程であり得る。好ましくは、ヌクレオチドの改変はSNPである。このシステムにおいては、推定産物の5'末端で2つのプライマーを用い、そのうちの1つは図4に示されるように3'のミスマッチを有している。上側の野生型の鋳型は相補的プライマーを有しており、ポリメラーゼがそれを3'方向に伸長することができる。下側のSNPを含む鋳型はプライマーを完全にはアニーリングできず、3'のミスマッチを生じさせる。これは、実際にあった場合、産物を著しく減少させるであろう。産物は、本明細書に記載される、標識を利用しない固有のイメージングにより検出される。
本発明を実施するための、機器の考えられるレイアウトを図1に示す。光源(重水素ランプ-HEREAUS Noblelamp DS 225/05J)、光学部品(UVレンズ-Newport、UVフィルター-Andover Optic)、分離段階(キャピラリー-Composite Metal Services Ltd)、及び検出器(HAMAMATSU PDA 3904 S3904-512Q)を、共通のレールに配列させる。低ノイズの重水素ランプからの光またはフィルターホイールの通過により検出波長を選択することが可能になる。その後、光は、典型的には50〜100マイクロメートル(μm)の内径を有する、溶融シリカのキャピラリーに集中する。生体分子が光線を通過すると、生体分子はそのスペクトル特性に依存してエネルギーを吸収する。光線はその後、生体分子により吸収されたエネルギーによるシグナルの低下を測定する検出器に集中する。
従って、さらなる実施形態において、本発明は、少なくとも1つのマイクロ流路、磁性流体の作動手段、加熱要素、UV光源、及び検出器を含む、核酸の存在を決定するための装置を提供する。
検出器は、好ましくはフォトダイオードアレイまたは電荷結合素子である。
一実施形態において、装置は、CCDのアレイを用いて同時にイメージングされる複数の平行な流路を含む。
マイクロ流体デバイスを、DNA抽出及び産物のサイズ決定のような、上流及び下流の処理構成要素の両方と一体化することができること、また一体化されるであろうことも注意されたい。従って、(患者からの)サンプル抽出から最後のSNP検証までの間の全ての処理タスクを実施するためにマイクロ流体のプラットフォームが使用されるであろう。
Claims (23)
- a)核酸サンプルを含む溶液を、温度制御し且つUV照射した流路に沿って移動させ、溶液の流れ方向を複数回変化させる、核酸サンプルを増幅する工程と、
b)核酸のUV吸収を測定する工程
とを含む、核酸の検出方法。 - 核酸の速度を測定する工程をさらに含む、請求項1に記載の方法。
- 速度に基づいたシグナル処理を用いる、請求項2に記載の方法。
- 存在する核酸の量を決定する工程を含む、請求項1から3のいずれか一項に記載の方法。
- 核酸がDNAまたはRNAである、請求項1から4のいずれか一項に記載の方法。
- 増幅がPCRによる、請求項1から5のいずれか一項に記載の方法。
- 溶液がPCR混合物をさらに含む、請求項1から6のいずれか一項に記載の方法。
- PCR混合物が少なくとも1つの相補的プライマー及びポリメラーゼを含む、請求項7に記載の方法。
- プライマーが標識されていない、請求項8に記載の方法。
- プライマーが標識をさらに含む、請求項8に記載の方法。
- 標識が蛍光である、請求項10に記載の方法。
- 流路の温度が50℃から100℃の範囲内である、請求項1から11のいずれか一項に記載の方法。
- 3つの異なる温度区域が流路の別々の部分に適用される、請求項1から12のいずれか一項に記載の方法。
- 第一の温度区域における温度が50℃から60℃の範囲内であり、第二の温度区域における温度が約72℃であり、且つ第三の温度区域においては約95℃である、請求項13に記載の方法。
- PCRサイクルの数が10から40である、請求項1から14のいずれか一項に記載の方法。
- PCRサイクルの数が20である、請求項15に記載の方法。
- 核酸の量が、検出された、核酸分子によるUV光の吸収によって表される、請求項1から16のいずれか一項に記載の方法。
- PCRサイクルにおいて産生される核酸の量が、検出された、核酸分子によるUV光の吸収によって表される、請求項17に記載の方法。
- UV吸収がフォトダイオードアレイまたは電荷結合素子を用いて検出される、請求項1から18のいずれか一項に記載の方法。
- 溶液を圧力または磁性流体の作動を用いて移動させる、請求項1から19のいずれか一項に記載の方法。
- 核酸の改変の検出のための、請求項1から20のいずれか一項に記載の方法の使用。
- 核酸の改変がSNPである、請求項21に記載の使用。
- 少なくとも1つのマイクロ流路、磁性流体の作動手段、加熱要素、UV光源、及び検出器を含む、核酸の存在を決定するための装置。
