Description
本発明は、輸送流体とは異なる性質を持つばかりでなく輸送流体中で安定な懸濁液を形成しえる粒子を記載する。更に、本発明は、該粒子との相互作用によって1種又は数種の試料成分を排除し得る粒子を記載する。本発明はまた、異なる分離及び分析用途におけるこれら粒子の使用を記載する。
全てのクロマトグラフィー技術は、固定相と試料成分との間の相互作用容易にするために、移動相(輸送流体)と固定相との間の特性の相異に依存する。しかし、移動固定相を用いる技術の場合、それにより、その相異が輸送流体中において固定相の安定性を損ねるといった問題を引起すことがある。例えば、移動固定相が粒子の場合、この不安定性は、該移動固定相を液中凝集又は一般凝集(粒子間の特性が同一である)及び沈降させることが出来る。固定相の安定な懸濁液をつくる為には、粒子が輸送流体と同様の特性を持つ必要がある。これは、クロマトグラフィーのための移動固定相の持つ伝統的な逆説であった:クロマトグラフィーを行うためには、固定相と輸送流体との間に特性の相異があるべきである、しかし、同時に、安定な懸濁液を形成するためには、固定相は輸送流体と同様の特性を持つ必要がある。この逆説は、我々の発明により解決する。我々の粒子は、輸送流体とは特性の異なる心部(例えば、心部が疎水性で、一方輸送流体は水性である)を持つが、しかし、この心部は輸送流体と同様の特性を持つ被覆を有する(例えば、親水性の被覆と水性の輸送流体)。これにより、粒子は輸送流体中で安定な懸濁液を形成し、しかし、なお、被覆に浸透する試料成分と相互作用する。
心部の性質は、検出の前の、試料成分からの粒子の最終的な分離にとって重要であることがある。
MilliQ water(MQ)が、MilliQ システム, Millipore, MA, USAにより精製された。アセトン、酢酸アンモニウム、蟻酸アンモニウム及び酢酸はMerck, Darmstadt, Germanyから入手した。過硫酸ナトリウム、スチレン及びDVBは、Polymer Chemistry, Lund Universityから受領した。フタル酸ジメチル及びフタル酸ジエチルは、SigmaAldrich, St. Louise, MO, USAから入手した。フタル酸ジプロピル及び炭酸アンモニウムは、Aldrich, Gillingham, UKから入手した。アセトニトリルは、Merck, Hohenbrunn, Germanyから、ラウリルメタクリレート(LMA)はFluka, Buchs, Germanyから、窒素ガスはAGA, Sundbyberg, Swedenから、そして蟻酸はRiedel-de Haeen, Seelze, Switzerlandから入手した。
CFF-CEC 実験は、データ処理のためのChem Station software(V.B01.03)を持つHP3DCE system(Agilent Technologies, Waldbronn, Germany)により行われた。Polymicro Technologies(Phoenix, AZ, USA)から入手した溶融シリカ毛細管(87 cm, 内径50 μm , 外径 375 μm)が全ての実験に用いられた。電解液は、50 mmol/L 炭酸アンモニウムpH 8.2と、0%超40%未満(容量/容量)のアセトニトリルから調製された。CFF-CEC実験における電解液懸濁液中の粒子は、50 mmol/L炭酸アンモニウムpH 8.2及び0から40%(容量/容量)アセトニトリル中で3.8 mg/mlの粒子濃度を持っていた。分離は室温で、20 KV(230 V/cm)で行われた。その日の最初の分析の前に、全ての溶液と粒子懸濁液は、約10分間の超音波処理によって脱ガスされた。各々の分析の前に、毛細管は、0.1 mol/L 水酸化アンモニウム(1バールで1分)、水(1バールで1分)、及び電解液(1バールで2分)で濯がれた。最後に、毛細管は粒子懸濁液で充填され調整された(1バールで2分)。
検出は、陽イオン化モードで操作される直交ESI接合部を備えたAgilent Technologies LC/MSD イオントラップ trap SL質量分析器により行われた。電子吹付け電圧は4KVであった(毛細管の出口は地表電位近傍に保持された)。水中0.5%(容量/容量)の蟻酸とメタノール(1/1 容量/容量)からなる鞘状液体は、Agilent Technologies series 1100 4つ口(quaternary) ポンプにより0.120 ml/minで送液され、固定分割器により1:20に分割された。該CE計器は、Agilent Technologies 三重管同軸霧吹き器を用いたESI接合部に結合された。
石鹸不存在下の乳化重合は、伝統的な乳化重合の1変形である。乳化液の安定化は親水性開始剤、イオン性開始剤、親水性の共重合モノマー又はイオン性の共重合モノマーの共存により支配される。この検討においては、イオン性で且つ水溶性である開始剤過硫酸ナトリウムが疎水性のモノマーであるスチレン及びLMAと共に用いられた。これらの成分が水と混合されると、1つの開始剤に富んだ水性相と1つのモノマー相を持つ2相系が産生される。水性相中での開始剤とモノマーとの反応は、小さな界面活性ラジカルオリゴマーを産生する。撹拌が、効果的に該界面活性オリゴマーラジカルを捕捉する微小モノマー液滴を産生するために用いられる。かくて、産生されたオリゴマーラジカルは水性及びモノマー相の界面に蓄積され、系を乳化する。

メタクリル酸(MMA) 0.109 モル、トリメチロールプロパントリメタクリレート(TRIM) 0.109 モル、AIBN 8 mg及びアセトニトリル4mlがネジ蓋付き硼珪酸ガラス試験管に添加された、10分間の超音波処理で脱ガスされ、−26℃の冷凍器に入れられそこで重合が350 nmの紫外線照射により4時間開始された。使用された化学物質の由来は前の実施例と同じであった。その後、粒子は3000 rpmで10分間の遠心分離により抽出され、次いで、メタノール:酢酸(9:1, 容量/容量)により2回及びメタノールにより1回の、超音波槽を用いた一回につき20分間の再懸濁を行った。
CEC実験は、HP3DCEシステム(Agilent Technologies, Waldbronn, Germany)により行われ、そしてChem Stationソフトウエアが、データ処理に用いられた。Porymicro Technologies(Phoenix, AZ)から入手した溶融シリカ毛細管(長さ 70cm, 内径50μm, 及び外径375μm)が、全ての実験に使用された。試料溶液は、フタル酸ジメチル、フタル酸ジエチル、フタル酸ジプロピル及びフタル酸ジブチルを電解液に溶解して濃度を0.10 mg/Lとし、そして5-ヒドロキシドーパミン、エピネフリン、メタプロテレノール、ターブタリンを水に溶解して濃度を夫々50 μg/mL、30 μg/mL、10 μg/mL、及び4 μg/mLとして調製された。試料は、流体力学的に毛細管中に注入され(5秒(s), 50ミリバール)、分離電圧は20 kV(286 V/cm)であった。全ての分離は室温で行われた。分析の前に、全ての溶液と粒子懸濁液は超音波処理により脱ガスされ、毛細管は0.1 モル/LのNaOH(5分、1バール)、水(5分、1バール)及び電解液(5分、1バール)で洗浄された。
検出は、陽イオン化モードで操作される直交ESI接合部を備えたAgilent Technologies LC/MSD イオントラップSL質量分析器を用いて行われ、最大イオン蓄積時間50分(ms)超、75分(ms)未満及び全イオン流(TIC)ターゲット75000超、100000未満でm/z 50超、300未満を走査した。鞘状液体流は、蟻酸の0.1%水溶液とメタノールから成り(1:1 容量/容量)、Agilent Technologies シリーズ 1100 4つ口ポンプにより、固定分割器で1:30に分割して、0.180 ml/分で送液された。該CEは、Agilent Technologies 三重管同軸霧吹き器を用いたESL接合部に結合された。該直交接合部は、図8で説明されるように、連続全充填分析の際粒子が質量分析器に進入するのを防ぐ。
疎水性効果は、低濃度の有機改質剤、即ちアセトニトリル濃度20-40%、を持つ電解液中で検討された。使用された資料成分は、同族列の種々のフタル酸アルキルであった。これらの分子は、電荷を持たず、従って負に荷電した移動固体相との静電気的な相互作用を持たない。異なる分析間の内部浸透(endoosmotic)流れにおける相異を最小限にするため、保持時間はEOFマーカーであるDMSO D6の保持時間の方に(toward)標準化(normalize)された。標準化された保持時間は、以後本項を通じて使用されるであろう。再構成(reconstructed)イオンクロマトグラム(RIC)が、どのピークがどの試料分子に対応するかを同定するのに用いられた。試料分子の保持は毛細管に注入された粒子の量に比例することは、先に示されていた。図7から分かるように、これはまた、非荷電試料成分によるこの逆相検討における事実である。図8から分かるように、移動固体相を通過する場合試料分子のシグナル強度が影響を受けないということは、興味ある観察結果である。
硫酸化ジビニルベンゼン粒子
ジビニルベンゼンをモノマーとして用いた(0.109 モル/L)以外は前記した沈殿重合プロトコールを用い(実施例2参照)、ネジ蓋付き硼珪酸ガラス試験管中で粒子が調製された。粒子の沈殿は、分子量の増加により粒子がそれらの溶解限界に達したときに始まる。粒子は、重合後、3000 rpmで10分間の遠心分離により抽出され、次いでメタノールと酢酸の溶液(9:1, 容量/容量)で2回及びメタノールで1回再懸濁された。
CEC実験は、HP3DCE システム(Agilent Technologies, Waldbronn, Germany)により行われ、Chem Stationソフトウエアがデータ処理に使用された。Polymicro Technologies(Phoenix, AZ)から入手した溶融シリカ毛細管(長さ 70cm, 内径50μm, 外径375μm)が、全ての実験に使用された。試料溶液は、フタル酸ジメチル、フタル酸ジエチル、フタル酸ジプロピル、及びフタル酸ジブチルを電解液に溶解して0.10 mg/Lの濃度に調製された。試料は流体力学的に(5秒 50ミリバール)毛細管中に注入され、分離電圧は20kV(286 V/cm)であった。全ての分離は室温で行われた。分析の前に、全ての溶液及び粒子懸濁液は超音波処理により脱ガスされ、毛細管は、0.1モル/L水酸化ナトリウム(5分 1バール)、水(5分 1バール)、及び電解液(5分 1バール)で洗浄された。逆相実験での電解液は、10ミリモル酢酸アンモニウム緩衝液 pH 5.4 及び20%超、40%未満で濃度を変化させたアセトニトリルから成った。
検出は、陽イオン化モードで操作される直交ESI接合部を備えたAgilent Technologies LC/MSD イオントラップSL質量分析器を用いて行われ、最大イオン蓄積時間50分(ms)超、75分(ms)未満及び全イオン流(TIC)ターゲット75000超、100000未満でm/z 50超、300未満を走査した。鞘状液体流は、蟻酸の0.1%水溶液とメタノールから成り(1:1 容量/容量)、Agilent Technologies シリーズ 1100 4つ口ポンプにより、固定分割器で1:30に分割して、0.180 ml/分で送液された。該CEは、Agilent Technologies 三重管同軸霧吹き器を用いたESL接合部に結合された。該直交接合部は、連続全充填分析の際粒子が質量分析器に進入するのを防ぐ。
沈殿重合プロトコールは、安定化する界面活性剤を使用しない球状粒子の合成を容易にする。これは、界面活性剤はESI-MSにおけるシグナル強度を低下させ、それにより検出の可能性を妨げるので、非常に重要である。この技術はまた、界面活性剤は粒子の誘導体化に影響を与える場合があり、そしてまた、懸濁安定性の増加が誘導体化工程によるものか否かは疑問なので、好ましいものである。高度に希釈された過マンガン酸カリウムを用いた硫酸化粒子の滴定により、反応性の炭素-炭素二重結合の転化率は定量的に測定された。
汚れた及び複雑な試料母液
伝統的な分離系にもつ重大な不利は、固定相の汚染、殊に汚れた試料又は複雑な試料母液中の試料によるものである。移動固体相の使用は、これらの問題を上手く回避する。本実施例において、食物に含まれていた発がん物質が尿試料から定量される。尿の分析は、伝統的に、固定相を保護するための、試料母液成分を除去する面倒な試料調製を必要とする。この実施例では、尿は直接分離管に注入される。
緩衝液中に懸濁された、親水性被覆を持つ疎水性粒子が、移動固定相として用いられた。50μmの内径を持つ、長さ75cmの毛細管が、分離に用いられた。該移動固定相は、部分充填及び連続的全充填技術の両方を用いて導入された。濃度が100マイクロモル/LとなるようにPhIPを加えた50 nLの尿が試料として用いられ、移動固定相を充填した毛細管中に注入された。
実験は、実施例4に記載のように行われたが、尿の代わりに血漿が分析された(試料母液は血漿であった)。血漿は、汚染分子に関しては、より以上に複雑な母液である。血漿の伝統的な分析は汚染分子を除去するための固体相抽出を必要とするが、本実施例では、血漿は直接管に注入され、極めてよい結果が得られた。
分離系と検出器(質量分析器)との間のFAIMS接合部の使用による改善された検出感度
ここまで、連続的全充填技術は、電子吹付け質量分析検出と共に用いられ、その場合電子吹付けは、粒子からの試料成分の分離を助ける、鞘状流れと霧化気体の使用により行われた。鞘状流れの不利は、それが、管から溶離する試料成分を希釈することである。電子吹付けイオン化と質量分析との組合せは濃度に敏感な検出技術[参考文献(REF)]なので、この試料成分の希釈(希釈は濃度の減少に等しい)は、検出感度の減少を来す。この問題を解決するため、質量分析器の使用者は、数年間[参考文献 フィンランド(Finn)]、しばしば低速流れ毛細管使用分離技術と組合せて鞘状流れを使用しない電子吹付けを使用して来た。一方、全充填技術は鞘状流れの使用を拠所とする(上記のように)。しかし、分離系と質量分析器との間のFAIMS電子吹付け接合部を使用すれば、鞘状流れは不要であり、それ故高い検出感度が得られる。これは、感度だけでなく適用と試料のタイプの増加に関して連続的全充填技術を大きく改善する。簡単に言えば、FAIMS接合部において、試料成分は、調節可能な(adjustable)電場中の経路を通って移動固定相から分離される。
シクロデキストリンで被覆された不活性粒子を用いる対掌性分離
伝統的に、シクロデキストリンは、毛細管電気泳動分離系の電解液中にシクロデキストリンを溶解することにより、対掌性分離に用いられる。分析試料(例えば鏡像対A及び B)は、毛細管を通って輸送されるにつれて、AとBは対掌性デキストリンとの相互作用が相異するので分離される。しかし、検出は、シクロデキストリン分子がバックグランドノイズ増加させるので悪影響を受ける。本実施例は、本発明がこの問題を解決することを示す。我々の発明のこの実施例において、シクロデキストリン分子は、不活性粒子上の被覆として固定される。この移動固体相の不活性心部は急速な物質移動を可能にするが、それは、AとBはシクロデキストリン被覆とのみ相互作用し、粒子心部とは作用しないからである。増加したバックグランドノイズは、粒子が検出に先立って分析試料(AとB)から分離されるので、この系では問題にならないであろう。これは、伝統的な技術に比べ、極めて大きな利点である。
粒子上のアルキレジオール(alkylediole)被覆の使用による受容制限物質(restricted access material) (RAM)として用いられる粒子
疎水性粒子をアルキレジオール(alkylediol)と反応させることにより、得られる粒子は蛋白質を多量に含む母液中に存在する分子の分析に適したものとなる。該蛋白質はこの被覆の通過から排除され、相互作用のため粒子心部と反応する。
Claims (26)
- 試料成分のクロマトグラフィーによる分離における移動固体相に用いる粒子であって、該粒子が心部と被覆部を含み、該心部は少なくとも1種の分析試料と相互作用し、且つ、該被覆は本質的に液体中での凝集又は一般凝集を妨げる、ことを特徴とする粒子。
- 該心部が実質的に疎水性であり、且つ、該被覆部が実質的に親水性である、請求項1に記載の粒子。
- 該被覆部が実質的に疎水性であり、且つ、該心部が実質的に親水性である、請求項1に記載の粒子。
- 該粒子が多孔質である、請求項1に記載の粒子。
- 該粒子が実質的に非多孔質である、請求項1に記載の粒子。
- 該粒子が中性、陽イオン性、陰イオン性、又は両性イオン性である、請求項1に記載の粒子。
- 該粒子がモノクローナル又はポリクローナルである、請求項1に記載の粒子。
- 該被覆部が不均質又は均質である、請求項1に記載の粒子。
- 該粒子が単分散又は多分散である、請求項1に記載の粒子。
- 該粒子が対掌性又は非対掌性である、請求項1に記載の粒子。
- 該心部が、黒鉛、酸化チタン、アガロース、アルミナ、重合体であってメタクリル酸エステル、ビニルピリジン、アクリル酸エステル、スチレン、ジビニルベンゼン、及びシランの群から選ばれた1種又は数種のモノマーから重合されたもの、を含む、請求項1に記載の粒子。
- 該心部の表面が、燐酸残基、ホスホン酸類、カルボン酸、アルデヒド、エポキシド、1級アミン、2級アミン、3級アミン、4級アミン、エステル、エーテル、及び/又は水酸基、を含む、請求項1に記載の粒子。
- 該被覆部が、直鎖重合体、分岐鎖重合体、交互共重合体、及び/又はブロック共重合体を含む、請求項1に記載の粒子。
- 該重合体及び/又は共重合体が、ポリエチレングリコール、ポリプロピレングリコール、ポリビニルアルコール、及び/又はポリエチレンイミンからなる群から選ばれる、請求項13に記載の粒子。
- 該重合体及び/又は共重合体が、エーテル、エステル、燐酸残基、ホスホン酸類、1級アミン、2級アミン、3級アミン、4級アミン、エポキシド、カルボン酸、スルホン酸、及び水酸基からなる群から選ばれる1種又は数種の官能基を含む、請求項13に記載の粒子。
- 該被覆部が、エステル、エーテル、1級アミン、2級アミン、3級アミン、4級アミン、エポキシド、硫酸塩、スルホン酸、カルボン酸、水酸基、燐酸残基、ホスホン酸、及び/又はアミドからなる群から選ばれる1種又は数種の官能性基を含む小分子で構成されている、請求項1に記載の粒子。
- 該小分子が10g/mol〜10,000g/molの範囲の分子量を持つ、請求項16に記載の粒子。
- 該小粒子が炭化水素又はフッ素化炭化水素である、請求項17に記載の粒子。
- 該被覆部が、蛋白質、DNA、RNA、セルロース、デンプン、再生セルロース、改質デンプン、及び/又は改質セルロースからなる群の1種又は数種からなる、請求項1に記載の粒子。
- 該心部が帯電し、該被覆部が非帯電である、請求項1に記載の粒子。
- 該心部が非帯電であり、該被覆部が帯電している、請求項1に記載の粒子。
- 該心部又は該被覆部が対掌性である、請求項1に記載の粒子。
- 該粒子が架橋されている、請求項1に記載の粒子。
- 該粒子が、少なくとも1種の分析試料より大きな質量対電荷比(m/z)を有する、請求項1に記載の粒子。
- 心部が重合体である請求項1に記載の粒子の製造法であって、1つ又は数個の下記工程を含む方法:
モノマーを重合して心部を形成する;
重合体の置換を改質する;
心部の表面を改質する;
該心部を誘導体化する;及び
該心部を他の重合体で被覆する。 - 少なくとも1種の分析試料をクロマトグラフィーにより他の試料成分から分離し、且つ少なくとも1種の分析試料を検出し、その際該検出を角度付きイオン化源を用いる質量分析によって行う、請求項1から24のいずれか1項に記載の粒子の使用。
