Abstract
【解決手段】本発明は、バイオマーカ又はこの成分の存在又は欠乏に基づいて、生物試料の酸化ストレスを検出する方法、酸化損傷の累積記録を決定する方法、及び心疾患など老齢疾患の診断方法に関する。本発明は、また、生物試料の酸化ストレスを検出するキットに関するものであり、安定性反応物質及び抗体から構成されている。

Claims (41)
- 生物試料を使用して酸化ストレスを検出する方法において:
(a)測定可能な成分を有する酸化ストレスのバイオマーカを具える生物試料を取得するステップと;
(b)前記試料中の前記バイオマーカを化学的に安定させるステップと;
(c)前記測定可能な成分を分離するステップと;
(d)前記測定可能な成分を抽出するステップと;
(e)前記測定可能な成分の量を測定するステップ;
とを具えることを特徴とする方法。 - 請求項1に記載の方法において、前記酸化ストレスのバイオマーカは、アルデヒドタンパク付加体及びアルデヒド代謝産物タンパク付加体から選択されることを特徴とする方法。
- 請求項2に記載の方法において、前記アルデヒドは、4−ヒドロキシ−2,3−ノネナール(HNE)であることを特徴とする方法。
- 請求項2に記載の方法において、前記アルデヒドは、1,4−ジヒドロキシノネン(DHN)であることを特徴とする方法。

- 請求項6に記載の方法において、前記アルデヒドをアルコールに還元する前記ステップが、前記生物試料へNaBH4及びNaB2H4の一方を加えるステップを具えることを特徴とする方法。
- 請求項3に記載の方法において、前記アルデヒドをアルコールに還元する前記ステップが、HNEをDHNに還元するステップ、および/または、HNEを重水素アルコール[2H]DHNに還元するステップを具えることを特徴とする方法。
- 請求項1に記載の方法において、前記生物試料が、HNE、HNEタンパク付加体、DHN、DHNタンパク付加体、HNEの代謝産物およびこれらの組み合わせから選択された分子を含むことを特徴とする方法。
- 請求項1に記載の方法が、タンパク質結合を開裂することによって、前記測定可能な成分を分離するステップを具えることを特徴とする方法。
- 請求項10に記載の方法において、前記タンパク質結合を開裂するステップが、ラネーニッケル触媒反応を使用して、タンパク質チオエーテル結合を開裂するステップを具えることを特徴とする方法。
- 請求項11に記載の方法において、前記ラネーニッケル触媒反応が、約5乃至約20時間、約45℃乃至約60℃で行われることを特徴とする方法。
- 請求項1に記載の方法において、前記酸化ストレスのバイオマーカの前記測定可能な成分が、脂肪酸の過酸化によって生産される代謝産物であることを特徴とする方法。
- 請求項1に記載の方法において、前記酸化ストレスのバイオマーカの前記測定可能な成分が:4−ヒドロキシノネナール、4−オキソノネナール、4−ヒドロキシヘキセナール及び4−オキソヘキセナールから選択されることを特徴とする方法。
- 請求項1に記載の方法において、前記生物試料が、全血、血液派生物及びその組み合わせから選択されることを特徴とする方法。
- 請求項15に記載の方法において、前記血液派生物が、血漿、アルブミン及び酸化リポタンパク質から選択されることを特徴とする方法。
- 請求項1に記載の方法において、前記測定可能な成分の前記量は、質量分析に連結されたガスクロマトグラフィを使用して測定されることを特徴とする方法。
- 生物試料を使用して酸化ストレスを検出する方法において:
(a)酸化ストレスのバイオマーカを具える生物試料を取得するステップと;
(b)前記試料中の前記バイオマーカを化学的に安定させるステップと;
(c)前記安定化したバイオマーカに結合する抗体と前記試料を接触させるステップと;
(d)前記試料中の前記結合抗体の存在を検出するステップと;
を具える方法。 - 哺乳動物の酸化損傷の累積記録を経時的に決定する方法において:
(a)第1時点で、アルデヒド代謝産物タンパク付加体を具える血液試料である第1血液試料を哺乳動物から取得するステップと;
(b)前記第1血液試料中の前記アルデヒド代謝産物の量に基づいて、前記第1血液試料中の酸化ストレスレベルを検出するステップと;
(c)第2時点で、アルデヒド代謝産物タンパク質チオエーテル付加体を具える血液試料である第2血液試料を哺乳動物から取得するステップと;
(d)前記第2血液試料中の前記アルデヒド代謝産物の量に基づいて、前記第2血液試料中の酸化ストレスレベルを検出するステップと;
(e)前記第1及び第2血液試料中の測定されたアルデヒド代謝産物の量を使用して、前記哺乳動物の酸化損傷の累積記録を決定するステップと;
と具える方法。 - 請求項19に記載の方法において、前記アルデヒド代謝産物は、4−ヒドロキシノネナール、4−オキソノネナール、4−ヒドロキシヘキセナール、4−オキソヘキセナール、4−ヒドロキシ−2,3−ノネナール(HNE)及び1,4−ジヒドロキシノネン(DHN)から選択されることを特徴とする方法。
- 哺乳動物の心疾患のリスクを評価する方法において:
(a)HNEタンパク付加体とDHNタンパク付加体を具える生物試料を哺乳動物から取得するステップと;
(b)前記試料中のHNEタンパク付加体とDHNタンパク付加体の量を測定するステップと;
(c)前記HNEタンパク付加体と前記DHNタンパク付加体との間の所定の関係を決定するステップと;
(d)前記所定の関係に基づいて前記哺乳動物の心疾患のリスクを評価するステップと;
を具えることを特徴とする方法。 - 請求項21に記載の方法が、前記HNEタンパク付加体とDHNタンパク付加体の量の割合及び請求項19に記載の方法に従って測定された前記哺乳動物の酸化損傷の累積記録に基づいて、前記哺乳動物の心疾患の前記リスクを評価するステップとを具えることを特徴とする方法。
- 哺乳動物の心疾患又はそのリスクを診断する方法において:
(a)アルデヒド代謝産物タンパク付加体を具える生物試料を哺乳動物から取得するステップと;
(b)前記試料中の前記アルデヒド代謝産物付加体の量を測定するステップと;
(c)アルデヒド代謝産物タンパク質チオエーテル付加体の量が所定の閾値より大きい場合に、心疾患又はそのリスクを有する哺乳動物を診断するステップと;
を具えることを特徴とする方法。 - 請求項23に記載の方法において、前記心疾患又はそのリスク因子が、高血圧、インスリン耐性、高血糖症、脂質異常症、拡張機能障害、心筋線維症、及び不整脈、心臓肥大、頻拍、糖尿病、肥満及びメタボリック・シンドロームから選択されることを特徴とする方法。
- 請求項23に記載の方法において、前記アルデヒド代謝産物タンパク付加体が、アルデヒド代謝産物タンパク質チオエーテル付加体であることを特徴とする方法。
- 請求項23に記載の方法において、前記アルデヒド代謝産物が、4−ヒドロキシノネナール、4−オキソノネナール、4−ヒドロキシヘキセナール、4−オキソヘキセナール、4−ヒドロキシ−2,3−ノネナール(HNE)及び1,4−ジヒドロキシノネン(DHN)から選択されることを特徴とする方法。
- 請求項23に記載の方法において、前記哺乳動物がヒトであることを特徴とする方法。
- 哺乳動物の心疾患又はそのリスクを診断する方法において、
(a)HNEタンパク質チオエーテル付加体とDHNタンパク質チオエーテル付加体を具える生物試料を哺乳動物から取得するステップと;
(b)前記生物試料中のHNEタンパク質チオエーテル付加体とDHNタンパク質チオエーテル付加体の量を測定するステップと;
(c)HNEタンパク質チオエーテル付加体とDHNタンパク質チオエーテル付加体の存在量の割合が所定の割合より大きい場合に、心疾患又はそのリスクを有する前記哺乳動物を診断するステップと;
を具えることを特徴とする方法。 - 請求項28に記載の方法において、前記心疾患又はそのリスク因子が、高血圧、インスリン耐性、高血糖症、脂質異常症、拡張機能障害、心筋線維症、及び不整脈、心臓肥大、頻拍、糖尿病、肥満及びメタボリック・シンドロームから選択されることを特徴とする方法。
- 請求項28に記載の方法において、前記哺乳動物がヒトであることを特徴とする方法。
- 生物試料を使用して酸化ストレスに関する事象を検出するキットにおいて、前記生物試料は、全血及び血液派生物から選択され、アルデヒドを具えるものであり、当該キットが:
(a)前記アルデヒドをアルコールに安定させるための安定反応剤と;
(b)前記安定化したアルコールと特に結合する抗体と;
を具えることを特徴とするキット。 - 請求項31に記載のキットにおいて、前記安定反応剤が、前記アルデヒドをアルコールに変換するのに適していることを特徴とするキット。
- 請求項32に記載のキットにおいて、前記安定反応剤が、NaB2H4及びNaB2H4の一方を含むことを特徴とするキット。
- ヒト対象の心疾患の進行を遅らせる又は止める方法において:
(a)請求項28に記載の方法に従って、ヒト対象の心疾患又はそのリスクを診断するステップと;
(b)心疾患の進行を遅らせる又は止めるために知られている所定の処置を施すステップと;
を具えることを特徴とする方法。 - 請求項34に記載の方法において、前記所定の処置が、治療的に有効量のプロブコールを投与するステップを具えることを特徴とする方法。
- 請求項34に記載の方法において、前記所定の処置が、心疾患の進行を遅らせる又は止めるために有効な運動プログラムを推奨するステップを具えることを特徴とする方法。
- 請求項34に記載の方法において、前記所定の処置を施すステップが、心疾患の進行を遅らせる又は止めるために有効な治療食を与えるステップとを具えることを特徴とする方法。
- ヒト対象の所定の処置に対する心疾患の進行の反応を評価する方法において:
(a)アルデヒド代謝産物タンパク付加体を具える生物試料を哺乳動物から取得するステップと;
(b)前記試料中の前記アルデヒド代謝産物タンパク付加体の量を測定するステップと;
(c)前記試料中の前記アルデヒド代謝産物タンパク付加体の前記測定量に基づいて、所定の処置に対する心疾患の進行の反応を評価するステップとを具える;
ことを特徴とする方法。 - 請求項38に記載の方法において、前記所定の処置に対する前記心疾患の進行の前記反応を評価するステップが、提案された処置の起こりうる結果を評価するステップを具えることを特徴とする方法。
- 請求項38に記載の方法において、前記所定の処置に対する前記心疾患の進行の前記反応を評価するステップが、前記所定の処置に応じて前記心疾患の前記進行を評価するステップを具えることを特徴とする方法。
- 哺乳動物の酸化損傷の累積記録を決定する方法において:
(a)血液試料がアルデヒド代謝産物タンパク付加体を具える血液試料を哺乳動物から取得するステップと;
(b)請求項1に記載の方法に従って測定された前記アルデヒド代謝産物の量に基づいて、前記血液試料を使用して酸化ストレスレベルを検出するステップと;
(c)前記血液試料中で測定されたアルデヒド代謝産物の量及び前記アルデヒド代謝産物の所定の基準量を使用して、前記哺乳動物の酸化損傷の累積記録を決定するステップと;
を具えることを特徴とする方法。
