【解決手段】抽出物の乾燥残分に対してプロシアニジンC1当量として表わして、少なくともプロアントシアニジン15重量%を含む、スノキ属の植物から得られる抽出物で、スノキ属の植物がオオミツルコケモモ又はアメリカンクランベリーである。当該抽出物の調製法として、オオミツルコケモモ又はアメリカンクランベリーを水溶液に入れ、固液分離を行い、有機溶媒を加え、そののち生成物を乾燥する方法を開発した。

Description
水5部当たりベリー1部の量である水相中の溶液にアメリカンクランベリーを入れて置く。擂り潰し、25℃にて傾斜法で固液分離をしたのち、水相中にクランベリー1部当たり酢酸エステル5部の量の酢酸エチルを、温度20℃、pH4.5,圧力1気圧にて添加し、そののち得られた二層を分離する。その後、0.1から0.5気圧の減圧、30から40℃の温度にて脱水し、その後二回目の脱水を圧力0.9から1.5気圧、温度145℃で実施した。
プロアントシアニジン分析用のPorter改良のBate−Smith法を使用した(Bate−Smith E.C.Phythochemistry,1973,12,pp907〜912及びPorter.L.J.等、Phytochemistry,1986,25,223〜230)。
チオアシドリシスは、プロアントシアニジン含量及びプロアントシアニジンの平均重合度を測定できるようにする他の方法である(Guyot等、J.Agric.Food Chem.,1998,1698〜1705)。
本試験では付着の陰性対照として繊毛を持たないDH5α細菌株を使用した。細菌株5及び6は臨床的単離で得た尿路感染症大腸菌株である。
全体で250mgのゼラチンカプセル一つを製造するには、以下のものを混合する:
−当該抽出物の乾燥残分に対してプロシアニジンC1当量で表して15%のプロアントシアニジンの力価を有する本発明による抽出物240mg、
−マイクロクリスタリンセルロース10mg。
各ゼラチンカプセルはプロアントシアニジン36mgを含有する。
全体で300mgのゼラチンカプセル一個を製造するには、以下のものを混合する:
−AFCANN9030抽出物200mg(当該抽出物の乾燥残分に対してプロシアニジンC1当量として表して30.5%のプロアントシアニジンの力価を有する)、
−マイクロクリスタリンセルロース100mg。
各ゼラチンカプセルはプロアントシアニジン61mgを含有する。
Claims (18)
- 抽出物の乾燥残分に対してプロシアニジンC1当量として少なくともプロアントシアニジン15重量%を含む、スノキ属の植物から得られる抽出物。
- スノキ属の植物がオオミツルコケモモ又はアメリカンクランベリーである、請求項1に記載の抽出物。
- アメリカンクランベリーのベリーを使用する、請求項1及び2のいずれかに記載の抽出物。
- 当該プロアントシアニジンの平均重合度が5より大きい、請求項1から3のいずれかに記載の抽出物。
- 当該プロアントシアニジンの平均重合度が6より大きい、請求項4に記載の抽出物。
- 当該プロアントシアニジンの平均重合度が8より大きい、請求項5に記載の抽出物。
- 当該アメリカンクランベリーを水溶液に入れ、固液分離を行い、有機溶媒を加え、そののち当該抽出物を得るために生成物を乾燥する、請求項1から6のいずれかに記載の抽出物の調製方法。
- 二回目の乾燥を行う、請求項7に記載の抽出物を調製する方法。
- 当該有機溶媒がアルコールである、請求項7及び8のいずれかに記載の抽出物の調製方法。
- 当該アルコールがエタノールである、請求項9に記載の抽出物の調製方法。
- 当該有機溶媒がアルデヒドである、請求項7及び8のいずれかに記載の抽出物の調製方法。
- 当該アルデヒドがアセトアルデヒドである、請求項11に記載の抽出物の調製方法。
- 当該有機溶媒がエステルである、請求項7及び8のいずれかに記載の抽出物の調製方法。
- 当該エステルが酢酸エチルである、請求項13に記載の抽出物の調製方法。
- 当該溶媒が水溶液のクランベリー量に対して容量で0から15の比率である、請求項7から14のいずれかに記載の方法。
- 請求項7から14に記載の方法で得られるべき、請求項1から6のいずれかに記載の抽出物。
- 請求項1から6及び16のいずれかに記載の抽出物を含有する、規定食又は栄養補助食品。
- 尿路の壁へある種の大腸菌の付着の低減に寄与するための、請求項1から6及び16のいずれかに記載の抽出物の使用。
