Abstract

Description
本発明は、一般に、分子生物学の分野に関し、詳細には、αvβ6インテグリンに対する抗体に関する。
インテグリンは、細胞間接着および細胞−マトリクス接着を媒介する、細胞表面レセプターのスーパーファミリーである。これらのタンパク質は、足場、ならびに発生および組織修復の間の細胞の増殖、移動および分化のためのシグナルを提供することが公知である。インテグリンはまた、細胞の分化および侵入(特に、細胞がその特化された形態を失い、転移癌細胞となる場合)に関与している。

β6でトランスフェクトしたNIH 3T3細胞およびFDC−P1細胞を、親細胞株に、全長マウスβ6 cDNAおよびネオマイシン選択マーカーを含むDNA構築物をエレクトロポレーションすることによって作製した。G418を含む培養培地中で14日間細胞を継代し、続いて蛍光活性化細胞選別(FACS)によって最大レベルの表面β6を発現する細胞を単離することにより、安定にトランスフェクトされた細胞を選択した。トランスフェクトされたFDC−P1細胞を、1.5g/L重炭酸ナトリウム、4.5g/lグルコース、および1.0mMピルビン酸ナトリウム、10% FBS、2.5% マウスIL−3培養上清、および1.5mg/mlの活性なG418を含むように調整した、4mM L−グルタミンを補充したDMEM中で培養した。トランスフェクトされたNIH 3T3細胞を、10% FBS、2mM L−グルタミン、ペニシリン/ストレプトマイシン、および1mg/mlの活性なG418を補充したDMEM中で培養した。
αvβ6タンパク質を、Weinacker(上記)に本質的に記載される通りに精製した。hsαvβ6を発現するCHO細胞株を培養し、そして得られる上清を遠心分離により収集した。このインテグリンを、抗αv抗体L230を用いたアフィニティークロマトグラフィーにより精製した。精製したL230を、CNBr活性化Sepharose
4B(Sigma)に、4.8mg抗体/ml樹脂の割合で架橋した。αvβ6上清を0.5mg抗体/ml樹脂にてL230アフィニティーカラムにローディングし、そしてこのカラムを、各々以下である10カラム容量を用いて洗浄した:(1)50mM Tris−Cl(pH7.5)、1M NaCl、1mM MgCl2;(2)50mM Tris−Cl(pH7.5)、50mM NaCl、1mM MgCl2;および(3)10mM Na3PO4(pH7.0)。hsαvβ6を、100mMグリシン(pH2.5)を用いて1:10容量の1M Na3PO4(pH8.0)中に溶出させた。このタンパク質を、数回交換しながらリン酸緩衝化生理食塩水(PBS)に対して透析し、そして−20℃にて保存した。
β6−/−マウスを、完全フロイントアジュバント(CFA)中に1:1の容量比で合計200μlにて乳化した25μgの精製組換えヒトαvβ6を用いた腹腔内(IP)注射によって免疫した。あるいは、β6−/−マウスを、1mg/mlのCaCl2および1mg/ml MgCl2を含む100μlのPBS中に再懸濁したβ6でトランスフェクトしたNIH 3T3細胞(4×106個)を用いてIP注射によって免疫し、そして同じマウスに、100μlのCFAを隣接部位に注射した。最初の免疫の2週間後および4週間後、これらのマウスに、不完全フロイントアジュバントをCFAの代わりに用いたこと以外は同じ試薬を同様に追加免疫した。これらのマウスから、最後の追加免疫の7日後に採血し、抗β6力価を、精製した組換えヒトαvβ6への血清の結合またはβ6でトランスフェクトした細胞への結成の結合によって決定した。精製組換えヒトαvβ6で免疫したマウスの場合、マウスを3ヶ月間休養させ、そしてImmunEasy(Qiagen)と混合した同じ抗原を用いて再度免疫した。ハイブリドーマ融合のために脾臓を単離する3日前に、これらのマウスに、12.5μgの精製組換えヒトαvβ6タンパク質を、IP注射および静脈内注射の両方によって免疫した。融合日に、これらの動物を屠殺し、そしてそれらの脾臓を取り出し、そして梳いて単細胞懸濁物にした。これらの脾細胞を、薬物選択可能な細胞融合パートナーへの融合によって不死化した。
2群の抗体を、β6−/−マウスの免疫によって作製した。1セットの抗体(融合体番号6)を、可溶性ヒト短縮型αvβ6を用いた免疫によって作製した。他のセットの抗体(融合体番号7)を、マウスβ6でトランスフェクトしたNIH 3T3細胞を用いた免疫によって作製した。抗αvβ6抗体についてのスクリーニングを、以下に記載の通りに、細胞ベースおよび無細胞の両方の結合アッセイおよび機能アッセイを用いて実施した。ポジティブクローンの最初の選択は、精製hsαvβ6、ならびにβ6でトランスフェクトされたヒト細胞およびマウス細胞(コントロールとしての、トランスフェクトしていない細胞)への結合に基づいた。選択されたクローンを増大させ、そして最終的な培養物を、β6でトランスフェクトされた細胞およびトランスフェクトされていない細胞の両方に対する結合について、細胞捕捉アッセイ(以下の実施例5b)において再評価した(代表例は図1Aおよび図1Bに示され、ここで、融合体6および融合体7をそれぞれ示すmAbの名称の接頭語「6.」または「7.」は、省略される;以下の表2もまた参照のこと)。いくつかの抗体は、β6でトランスフェクトした細胞に優先的に結合し、一方、他の抗体は、トランスフェクトした細胞およびトランスフェクトしていない細胞の両方に結合した。このことは、これらの抗体のサブセットのみが、β6についての選択性を有することを示す(図1Aおよび図1B)。さらなる選択は、これらの抗体が、LAPへのビオチン化hsαvβ6およびβ6でトランスフェクトされたマウス細胞の両方の結合をブロックする能力に基づいた。選択クローンをFACSを用いてサブクローニングし、そして使用するまで凍結保存した。
(a.αvβ6 ELISA)
96ウェルマイクロタイタープレート(Corning COSTAR EASY−WASH)を、50μl/ウェルの5μg/ml hsαvβ6を用いて4℃で一晩コーティングした。このプレートを、自動プレート洗浄器中で洗浄緩衝液(PBS中0.1% TWEEN−20)を用いて4回洗浄した。次いで、TBS中3% BSA(180μl/ウェル)を添加し、そして25℃にて1時間インキュベートして、非特異的結合をブロックした。このプレートを上記の通りに洗浄し、そしてハイブリドーマ上清(スクリーニングアッセイに関して)または精製抗体(特徴付けに関して)のいずれかのTBS(1mg/ml BSA、1mM CaCl2、および1mM MgCl2を含有)中希釈物を添加した(50μl/ウェル)。このプレートを25℃にて1時間インキュベートし、洗浄し、次いで50μl/ウェルのペルオキシド結合体化ヤギ抗マウスIgG+A+M抗体(Cappel)を用いて1時間インキュベートした。結合した抗体を、3,3’,5,5’−テトラメチルベンジジン(TMB)を用いて検出した。結合は、450nmにて測定される吸光度によって示された。
96ウェルマイクロタイタープレートを、50μl/ウェルの二次抗体(ロバ抗マウスIgG(Jackson Immunoresearch);50mM重炭酸ナトリウム(pH 9.2)中に希釈した5μg/ml)を用いて4℃にて一晩コーティングした。プレートを、100μl/ウェルのアッセイ緩衝液(RPMI+2% BSA)を用いて2回洗浄し、次いで100μl/ウェルのアッセイ緩衝液を用いて37℃にて1時間ブロッキングした。FDC−P1細胞およびβ6でトランスフェクトしたFDC−P1細胞に関して、プレートを、抗マウスIg(Jackson ImmunoResearch;20μg/ml)を用いて室温で10分間ブロッキングして、二次抗体による非特異的Fcレセプター結合を減少させた(他の細胞型については省略した)。これらのプレートをブロッキングしている間に、細胞を、5×106細胞/mlにて、アッセイ緩衝液中の2
μM蛍光色素(Calcein−AM,Molecular Probes)を用いて標識した。これらの細胞を、穏やかに攪拌しながら37℃の水浴中で15分間、アッセイ緩衝液中のこの色素と共にインキュベートし、遠心分離によって収集し、そして5×106細胞/mlになるようにアッセイ緩衝液中に再懸濁させた。ブロッキング工程の後、このプレーとを軽く叩くことによってこの緩衝液を捨て、そして25μl/ウェルの上清または精製抗体をこのプレートに添加した。37℃での15分間のインキュベーション後、25μl/ウェルの標識細胞を添加し、そしてこのプレートを37℃で1時間インキュベートした。このプレートをアッセイ緩衝液(100μl/ウェル)を用いて3〜5回洗浄し、そしてプレート上の捕捉細胞によって発光される蛍光を記録した。結合パーセントを、最終洗浄工程の前(すなわち、添加した総細胞)の蛍光を、洗浄後(すなわち、結合細胞)の蛍光に対して比較することにより決定した。
細胞を、トリプシン処理により収集し、PBS中で1回洗浄し、次いでFACS緩衝液(1×PBS、2% FBS、0.1% NaN3、1mM CaCl2、および1mM
MgCl2)中に再懸濁し、次いで、0.2×105細胞を、氷上で1時間、ハイブリドーマ上清を含むFACS緩衝液(100μlの総容積)中でインキュベートした。インキュベーション後、これらの細胞を氷冷FACS緩衝液を用いて2回洗浄し、5μg/mlロバ抗マウスIgG PE(Jackson ImmunoResearch)を含む100μlのFACS緩衝液中に再懸濁し、そして氷上で30分間インキュベートした。次いで、細胞を、氷冷FACS緩衝液を用いて2回洗浄し、そして200μlのFACS緩衝液中に再懸濁した。PE標識二次抗体の結合を、フローサイトメトリーによってモニタリングした。
96ウェルマイクロタイタープレート(Corning COSTAR EASY−WASH)を、PBS(50μl/ウェル)中に希釈した0.3μg/mlの組換えヒトLAP(R & D Systems,カタログ番号246−LP)を用いて4℃にて一晩コーティングした。コーティング溶液を除去した後、これらのプレートを、180μl/ウェルの3% BSA/TBSを用いて25℃にて1時間ブロッキングした。別個の96ウェル丸底プレートにおいて、60μl/ウェルの2×ストック(1mg/ml BSAを含むTBS中、0.5μg/ml(1.25nM)のビオチン−αvβ6、2mM CaCl2、および2mM MgC12)を、ハイブリドーマ上清(スクリーニングに関して)または精製抗体(これもまた、1mg/ml BSAを含むTBS中)のいずれかの60μl/ウェルの2×ストックと合わせ、そして25℃にて1時間インキュベートした。LAPでコーティングしたプレートを、自動プレート洗浄器において洗浄緩衝液(PBS中0.1% TWEEN−20)を用いて4回洗浄した後、100μlの抗体−αvβ6混合物をこのプレートに移し、そして25℃にて1時間インキュベートした。このプレートを上記の通りに洗浄し、そしてエクストラアビジン(extravidin)−西洋ワサビペルオキシダーゼ結合体(Sigma)のTBS(1 mg/ml BSA)中1:1000希釈物(50μl/ウェル)を用いて25℃にて1時間インキュベートした。結合したタンパク質を、TMB基質を用いて検出した。
96ウェルマイクロタイタープレートを、50mM重炭酸ナトリウム(pH9.2)中に希釈した0.5μg/ml組換えヒトLAP(R & D Systems)(50μl/ウェル)を用いて4℃にて一晩コーティングした。このプレートを、PBS(100μl/ウェル)を用いて2回洗浄し、そしてPBS(100μl/ウェル)中の1% BSAを用いて25℃にて1時間ブロッキングした。このプレートを、100μl/ウェルのアッセイ緩衝液(TBS完全+1mM CaCl2および1mM MgCl2)を用い
て2回洗浄した。次に、このプレートの個々のウェルに対して、25μlのハイブリドーマ上清(または精製抗体)および25μlのβ6−FDC−P1細胞(5×106細胞/ml、上記の通りにCalcein AMで標識した)を添加した。このプレートを25℃で1時間インキュベートし、次いでこのアッセイ緩衝液(100μl/ウェル)を用いて4〜6回洗浄した。このプレート上に捕捉された細胞から発光された蛍光を記録した。結合百分率を、最終洗浄工程の前(すなわち、添加した総細胞)の蛍光シグナルを、洗浄後(すなわち、結合した細胞)の蛍光シグナルに対して比較することにより決定した。
本明細書中で用いたTGF−βバイオアッセイは、Abeら,Anal.Biochem.216:276−284(1994)において記載されるミンク肺上皮細胞(MLEC)PAI−1ルシフェラーゼ共存培養アッセイの変法であり、ここでは、β6でトランスフェクトした細胞をレポーター細胞と共に共存培養して、αvβ6によるTGF−βの活性化(Munger,前出)をモニタリングした。これは、プラスミノゲンアクチベーターインヒビター−1(PAI−1)の発現を誘導する能力に基づく、TGF−βに関する定量的バイオアッセイである。このアッセイにおいて、MLEC細胞は、ホタルルシフェラーゼレポーター遺伝子に融合された短縮型PAI−1プロモーターを含む発現構築物を用いて安定にトランスフェクトされている。トランスフェクトされたMLEC細胞を活性なTGF−β(0.2〜>30pM)に曝露することにより、細胞溶解産物におけるルシフェラーゼ活性の用量依存性増加がもたらされる。
融合体番号6由来の8個のハイブリドーマクローン(接頭語「6.」により示す)および融合体番号7由来の14個のハイブリドーマクローン(接頭語「7.」により示す)を、さらなる大規模化および特徴付けのために選択した(表2)。
A Sepharose 4 Fast Flow(Amersham Pharmacia Biotech,AB,Uppsala,Sweden)(1ml沈降床容積)
に直接ローディングした。このカラムを、PBSを用いて洗浄し、そしてIgG画分を、25mMリン酸、100mM NaCl(pH2.8)を用いて、1:20容積の0.5M Na3PO4(pH8.6)中に溶出させた。マウスIgG1抗体に関して、この上清を、1.5Mグリシン、3M NaCl(pH8.9)に調整した後にローディングし、そしてこのカラムを、25mM Na3PO4、3M NaCl(pH8.6)を用いて洗浄した後に溶出させた。これらの調製物を、本明細書中に記載されるインビトロでの生化学的特徴付けに用いた。
Na3PO4、25mM NaCl(pH8.6)中で平衡化したQ Sepharoseカラム(20mgタンパク質/ml樹脂)にローディングした。このカラムを、5カラム容積の平衡化緩衝液を用いて洗浄し、そして結合したタンパク質を、25mM Na3PO4、150mM NaCl(pH7.2)を用いて溶出させた。溶出させたタンパク質を濾過滅菌(0.45μm)し、そして使用するまで−70℃にて保存した。
精製抗体(表2、前出)を、以下の能力に関して定量的に特徴付けした:(1)hsαvβ6を結合する能力、(2)β6でトランスフェクトされたSW480細胞およびFDC−P1細胞を結合する能力、(3)LAPに対するビオチン−αvβ6の結合を阻害する能力、(4)LAPへの、β6でトランスフェクトされたFDC−P1細胞の結合を阻害する能力、ならびに(5)MLECアッセイ(前出)においてTGF−βのαvβ6媒介活性化をブロックする能力。これらのアッセイの各々における相対的効力を、公知のαvβ6抗体10D5(Huang et al,J.Cell Sci.111:2189(1998))の相対的効力およびいくつかの場合には抗αv抗体L230の相対的効力に対して比較した。融合体番号7の抗体の特徴付けに関して、融合体番号6の抗体6.8G6をまた、ポジティブコントロールとして用いた。

精製したモノクローナル抗体をまた、ELISA形式において、ビオチン化αvβ6への結合に関して6.8G6と競合する能力について試験した。このアッセイにおいて、6.8G6をELISAプレートにコーティングし、そして競合抗体とビオチン化αvβ6との混合物を、各々1mMのCa2+およびMg2+を含む緩衝液に添加した。結合したインテグリンを、エクストラアビジン−HRP結合体を用いて検出し、そして競合抗体を、結合をブロックする能力に関してスコア付けした。6.2B10(弱いブロッカー)以外の全てのコンセンサスブロッカー(表2)は、6.8G6と種々の程度に競合し得ることが示された(表4)。これらのデータは、これらのコンセンサスブロッカーが、6.8G6と同じかまたは重複するエピトープに結合することを確認する。
精製モノクローナル抗体のいくつかについてのcDNAを、Coliganら(編),Current Protocols in Immunology,Wiley,Media,PA(2001)に記載される通りの標準的な技術を用いて、単離し、そして配列決定した。推定アミノ酸配列を、図7Aおよび図7Bに示す。
パラフィン包埋組織切片または他の組織サンプル中のαvβ6発現を検出し得る抗体は、診断物として有用であり得る。これらの診断ツールを用いて、例えば、癌または線維症のような適応症に関して組織切片中のアップレギュレートされたαvβ6を検出し得る。

αvβ6は、通常、健常成体組織において、無視し得るレベルから低いレベルで発現される。しかし、αvβ6発現は、傷害、線維症、および癌においてアップレギュレートされる (例えば、ThomasらJ.Invest.Dermatology 117:67−73(2001);Bruntonら,Neoplasia 3:215−226(2001);Agrezら,Int.J.Cancer 81:90−97(1999);Breuss,J.Cell Science 108:2241−2251(1995)を参照のこと)。従って、パラフィン包埋組織上に発現される、αvβ6に特異的に結合する抗体を標準的免疫組織化学技術において用いて、線維症、癌およびαvβ6がアップレギュレートされる任意の他の疾患の診断のためにαvβ6発現を検出し得る。
コラーゲン4A3(COL4A3)ノックアウト(Alport)マウスは、腎臓線維症に関するインビボモデルとして確立されており、そして薬理学的薬剤の治療効果を試験するために用いられている(前出)。本発明者らは、AlportマウスにおいてmAb
6.8G6(カチオン依存性)および6.3G9(カチオン非依存性)を試験して、これらが、7週齢のAlportマウスにおいて通常観察される線維症を阻害するか否かを決定した。上記に示すように、これらの2つの抗体は、バイオアッセイにおいて、LAPに対するαvβ6結合を阻害することおよびTGF−βの活性化を阻害することが見出された。抗体1E6を、ネガティブコントロールとして用いた。
本発明者らは、腎線維症進行に関する別のマウスモデルを用いて、6.8G6および6.3G9の抗線維症効力を試験した。このマウスモデルでは、この動物の尿管を結紮し、片側性尿管閉塞(UUO)をもたらした。UUOは、マウスにおいて近い将来の腎不全を引き起こさずに進行性腎硬化症を引き起こす。なぜなら、閉塞していない腎臓は、比較的正常な腎機能を維持し得るからである。閉塞した腎臓は迅速な全般的線維症を受ける一方
で、閉塞していない腎臓は、順応性肥大を受ける。
を用いて捕捉し、バックグラウンドに関して補正し、そして距離の標準に対して較正した。左腎臓切片全体を含む連続視野の画像を、定量のために各動物から撮影した。
マウスにおけるブレオマイシン誘発性肺線維症は、肺線維症に関するインビボモデルとして確立されており、そして薬理学的薬剤の治療効果を試験するために用いられている。炎症が、ブレオマイシン処置の5〜15日後に通常明らかである。129系統マウスでは、肺線維症の程度は、ブレオマイシン処置60日後まで徐々に増大する。マトリクス蓄積は、通常、15日目あたりで検出可能になる。この例では、mAb 6.3G9を、0日目または15日目に開始して、1週間に3回、ブレオマイシン誘発性肺線維症を有するマウス中に4mg/kg/用量の濃度で腹腔内注射した。肺線維症を、50μlの無菌生理食塩水中0.03単位/kgの濃度での1回の気管内用量のブレオマイシンの投与によって0日目に誘発した。これらの動物を30日目に屠殺し、そして肺線維症の程度を評価した。抗体1E6を、ネガティブコントロールとして用いた。
αvβ6が乾癬に関与するか否かを決定するために、5人の乾癬患者および4人の正常個体由来のαvβ6発現を損傷皮膚生検および損傷していない皮膚生検について調べた。mAb 6.2A1免疫染色を用いて、本発明者らは、乾癬患者および正常コントロール由来の損傷していない皮膚と比較して、乾癬損傷におけるαvβ6発現の顕著な増大を見出した。従って、乾癬損傷におけるαvβ6のアップレギュレーションは、抗αvβ6抗
体の使用に関する診断的関連および治療的関連の両方を示唆する。
以前に考察した通り、αvβ6発現は、組織損傷に関連している。本研究において、αvβ6の発現を、胆管疾患によって損傷を受けたマウスおよびヒトの肝臓において調査した。
インテグリンαvβ6は、通常、無視し得るレベルから低いレベルで、健常な成体組織にいて発現される。種々のヒト腫瘍組織を、抗体6.2A1および本明細書中に一般的に記載される方法を用いて、αvβ6発現に関して評価した。これらの結果は、αvβ6インテグリン発現が、いくつかのヒト上皮癌において顕著にアップレギュレートされることを示した。顕著なことに、免疫組織学は、αvβ6が、多くの上皮癌において腫瘍島の縁部に特に顕著に発現されることを示した。上皮癌細胞におけるαvβ6の発現をさらに研究するために、Detroit 562細胞(咽頭癌)およびSCC−14細胞(舌扁平上皮癌)ならびにSW480β6細胞(前出)を、6.3G9および6.4B4で染色し、そしてフローサイトメトリーにより分析した。
ヒト腫瘍異種移植片を移植した免疫不全動物(例えば、ヌードマウスおよびSCIDマウス)は、抗癌剤の治療効果を試験するための有用なインビボモデル系として確立されている(例えば、van Weerdenら,Prostate 43(4):263−71(2000);Bankertら,Front Biosci 7:c44−62(2002)を参照のこと)。従って、本発明のブロッキング抗αvβ6モノクローナル抗体は、異種移植片モデルにおいて、腫瘍増殖を阻害する能力に関してインビボで試験され得る。この実験において、本発明者らは、新たなαvβ6抗体のうちのいくつかを、癌性ヒト咽頭(Detroit 562細胞株)異種移植片を移植した無胸腺ヌード雌性マウスにおいて、腫瘍増殖を阻害する能力に関して試験した。
細胞によってインターナリゼーションされる抗体は、特定の臨床的適応症(例えば、癌)に関して利点を提供する。なぜなら、これらの抗体は、次いで、癌細胞を選択的に標的かして癌細胞の増殖を選択的に阻害するために、毒素、放射性化合物または他の抗癌剤と結合体化され得るからである。抗αvβ6抗体がインターナリゼーションされる能力を、SW480β6(前出)およびSCC−14細胞において試験した。
