a)オオアワガエリ花粉プロフィリン Phl p12からの低アレルギー性変異体の生成、発現および精製
Phl p12誘導体を再構成する技術として、オーバーラップPCR法を用いた。pet17b発現ベクターでサブクローニングしたオオアワガエリ花粉 Phl p12をコードするcDNAを、PCRのテンプレートとして用いた。NdeIおよびEcoRIの制限部位とオーバーラップする配列、およびC末端6ヒスチジン残基をコードする配列を含む2つのPCRフラグメントを生成するために、以下のプライマーを用いた。これらの配列は、タンパク質を精製するためのものである。フラグメント1を、MDE−1プライマー:5’CATATGAGGCCCGGCGCGGTCATC3’、およびMDE−2プライマー:5’GTACGTCTGCCACGCCATCATGCCTTGTTCAAC3’を用いて生成した。フラグメント2を、MABC−1プライマー:5’GTTGAACAAGGCATGATGTCGTGGCAGACG3’、およびMABC−2プライマー:5’GAATTCTTAATGGTGATGGTGATGGTGACCCTGGATGACCATGTA3’を用いて生成した。ついで、上述したように得られた両方のPCR産物を、プライマーMDE−1およびMABC−2を用いたオーバーラップPCR反応のテンプレートとして用いた。このオーバーラップPCR反応は、Phl p12誘導体(例えばMP12)をコードするcDNAを生成するために行った(図1に概略図を示した。)。MP−12をコードするDNAを、pBluescriptベクターシステム(Stratagene)によりクローニングし、DNA配列をDouble−strand sequencing(MWG Biothch、Germany)により確認した。
Jasco J−715分光偏光計で、20℃で平衡状態になるパス長0.1cmの細胞を用いて、円偏光二色性(CD)測定を行った。スキャニング速度100nm/min、解像度0・5nmでスペクトルを記録した。結果を3回のスキャニング結果の平均値とした。同様の条件下で得たMilliQスペクトルに対応して減算することによって、最終的なスペクトルを基準調整した。結果を二次構造評価プログラムJ−700に適合させた。
ポリプロリンに対する親和性は、様々な生物由来のプロフィリンに共通する特徴である。低アレルギー性Phl p12誘導体であるMP12は、ポリプロリンに結合せず、それゆえに変化した生化学特性を示すことが明らかにされた。
a)大幅に低下したIgE結合能を示すMP12
24人のプロフィリン感受性患者からの血清を点染色分析することによって、組換えMP12のIgE結合能を、組換えPhl p12の野生型のIgE結合能と比較した(図4)。Phl p12およびMP12は、コントロールとして用いたヒト血清アルブミン(HSA)と同様に、ニトロセルロース上に点染色され、24人のプロフィリン感受性患者の血清で検出された。125I標識抗ヒトIgE抗体を用いて、IgE抗体に対する結合を検出した。全ての患者において、Phl p12野生型のIgE反応性が見られた。一方、24人の患者において、MP12またはコントロールとして用いたタンパク質HSAがIgEに対して反応した患者は24人中1人もいなかった(図4)。
IgE結合抑制を、以下の式により算出した:

ここで、Aは血清をBSAと共にインキュベートした後に得られるOD値を表し、Bは血清をPhl p12またはMP12と共にそれぞれインキュベートした後のOD値を表している。
ついで、プロフィリンアレルギー患者の好塩基性白血球からのヒスタミン放出誘因能を、再構成したPhl p12と野生型Phl p12とにおいて比較した。
再構成Phl p12による免疫によって、野生型Phl p12および他の花粉由来のプロフィリンに反応するIgG抗体が誘導されるかどうかを実験するために、フロイントの完全および不完全アジュバント(200mg/注入)(Charles River、Kisslegg、Germany)を用いて、ウサギをPhl p12またはMP12により3回免疫した。血清サンプルを4週間間隔で得た。得られた血清を、分析するまで−20℃で保存した。
アレルギー患者のIgEがPhl p12に結合するのを抑制する、MP12誘導ウサギIgGの可能性をELISA競合分析によって調査した。ELISAプレート(Nunc Maxisorp、Rosklide、Denmark)をPhl p12(1μg/ml)でコーティングし、それぞれ1:250で希釈した抗MP12抗血清、またはPhl p12抗血清と共にプレインキュベートした。また、コントロールとして、それぞれに対応する免疫前の血清と共にPhl p12をプレインキュベートした。洗浄後、7人のPhl p12感受性芝花粉アレルギー患者からの血清を1:3に希釈したものと共にプレートをインキュベートした。ついで、モノクローナルラット抗ヒトIgE抗体(Pharmingen、San Diego、CA)を1:1000に希釈し、これを用いて処理した後、1:2000に希釈したHRP結合ヒツジ抗ラットIg抗血清(Amersham)で処理することによって、結合IgE抗体を検出した。抗ペプチドまたは抗変異抗血清と共にプレインキュベートすることによるIgE結合抑制率を、次式により算出した:

ここでODIおよびODPは、ウサギ免疫血清および対応する免疫前血清と共に、それぞれプレインキュベートした後の吸光度を表している。表3に示すように、抗Phl p12抗体用いて達したPhl p12に対する患者由来IgE結合抑制は、30.2〜66.7%の間であった(平均抑制率49.8%)。同様に、MP12に対する抗体を用いた場合、10.8〜27.6%(平均抑制率20.8%)の範囲の抑制を示し、抗Phl p12のIgE反応性が大幅に減少したことを観察した(表3)。
ペプチド刺激IgG抗体の生物学上の関連性および有効な保護活性を、アレルゲン特異的IgEを導入したラット好塩基球白血球(RBL)細胞を用いた確立した細胞モデルシステムにおいて調査した。
ハウスダストダニ(HDM)アレルギーは、全てのアレルギー患者の50%以上に影響する、世界中で最も一般的なアレルギーに属する。ヤケヒョウヒダニは、ヨーロッパにおいて、ハウスダスト中のアレルゲンの最も重要な原因として同定された。
ヒスチジン標識したDer p 2、Der p 2フラグメント(1〜53アミノ酸+54〜129アミノ酸)、およびDer p 2ハイブリッド(54〜129+1〜53アミノ酸)をコードするcDNAを、表4に示したプライマー(MWG、Ebersberg、Germany)を用いたPCR増幅により合成した。また、Der p RNAから逆転写することによって、Der p 2cDNAを得た。
レーザ脱離質量スペクトルを、飛行時間Compact MALDI II instrument(Kratos、U.K.;piCHEM、Austria)を用いて線形モードで得た。10%アセトニトリル、0.1%トリフルオロ酢酸、およびα−シアノ−4−ヒドロキシ桂皮酸(60%アセトニトリル、0.1%トリフルオロ酢酸が溶解している)中に溶解したサンプルを、マトリックスとして用いた。サンプルの準備のために、タンパク質およびマトリックス溶液を1:1で含む混合液を標的上に堆積し、空気乾燥した。
JASCO J715分光偏光計を用いて、製造者が推奨した方法にしたがってネオジムガラスを用いて波長を測定し、精製した組換えタンパク質CDスペクトルを記録した。2回蒸留した室温の水に溶解したrDer p 2およびrDer p 2誘導体(c=0.1〜0.5mg/ml)のCD測定を行った。0.1cmのパス長を有する円形の石英キュペットを用いて、解像度0.2nm、スキャン速度50nm/分でスペクトルを記録した。少なくとも3回のスキャンデータを蓄積し、スペクトルを信号平均した。その結果を、一定の波長で残留した楕円率の平均として示した。
精製した組換えDer p 2、フラグメント1(1〜53アミノ酸)およびフラグメント2(54〜129アミノ酸)の2つのrDer p 2 フラグメント、ならびにrDer p 2ハイブリッドについて、非変性点染色分析によりIgE反応性を調べた。精製したタンパク質(0.1mg/ml)を2マイクロリットル、およびコントロールとしてBSAを、ニトロセルロース膜片(Schleicher&Schuell、Germany)上に点在させた。点染色したタンパク質を含むニトロセルロース片を、バッファA(40mM Na2HPO4、0.6mM NaH2PO4、pH 7.5、0.5%[v/v]Tween 20、0.5%[w/v]BSA、0.05%[w/v]NaN3)中に固定し、ダニアレルギー患者の血清、非アレルギー患者の血清(1:10に希釈)、または血清を含まないバッファAと共にインキュベートした。IgE抗体結合を。125I標識抗ヒトIgE抗体を用いて検出し、オートラジオグラフィにより可視化した。
ヘパリンで処理した血液サンプルをアレルギー患者から得た。様々な濃度のrDer p 2、rDer p 2 フラグメント、rDer p 2ハイブリッド、モノクローナル抗IgE抗体(Immunotech、Marseille、France)、またはPBS中において、血液サンプル(100μl)を37℃で15分間インキュベートした。Hauswirth,A.W.らによるJ Allergy Clin Immunol 110:102(2002)に記載の方法で、CD 203c発現を測定した。上記文献には、CD 203cのアップレギュレーションが、アレルゲン刺激好塩基球活性および脱顆粒のための代用マーカーとして記載されている。それゆえに、ハウスダストダニアレルギー患者の好塩基球においてCD203cのアップレギュレーションを測定することによって、組換えrDer p 2、rDer p 2フラグメント、およびrDer p 2ハイブリッドの各アレルギー反応活性を比較した(図13および14)。図13に、代表的な3人の患者の結果を示す。調査した各患者において、10μg/mlの野生型rDer p 2と共にインキュベートした好塩基球は、顕著にCD 203c発現をアップレギュレートした。一方同じ濃度の個々のフラグメント、または2つのフラグメントの等モル混合物とインキュベートした好塩基球は、アップレギュレートしなかった(図13)。加えて異なる濃度(5μg/ml〜0.32ng/ml)のrDer p 2およびrDer p 2ハイブリッドを1:5で希釈し、同じ10人の患者の好塩基球をさらした。図14に、代表的な6人の患者の結果を示す。40ng/mlおよび5000ng/mlの間の濃度において、rDer p 2ハイブリッドにさらした好塩基球がCD 203cをアップレギュレートした。一方、野生型rDer p 2は、8ng/mlおよび200ng/mlの間の濃度ですでにCD 203cのアップレギュレートを引き起こした。10人中8人の患者において、rDer p 2と比較してrDer p 2ハイブリッドの好塩基球活性化能が10倍以上減少した。
8週齢のメスBALB/cマウスの5つのグループに対して、それぞれ精製した5μgのタンパク質(rDer p 2、rDer p 2フラグメント1、rDer p 2フラグメント2、またはrDer p 2ハイブリッド)を用いて免疫し、200μlのAluGel−S(SERVA Electrophoresis、Germany)を、20週間を通じて4週間間隔で首の皮下に吸着させた。血液サンプルを各免疫処理の1日前に採取し、−20で保存した。
ELISAプレート(Greiner、Austria)を、100μlの精製したrDer p 2でコーティングし、PBSで希釈して、濃度を5μg/mlにした後、4℃で一晩おいた。PBSTで2回洗浄し、ブロッキングバッファ(PBST;1% w/v BSA)を用いて室温で3時間固定した後、抗rDer p 2抗血清、抗rDer p 2フラグメント1抗血清、抗rDer p 2フラグメント2抗血清、または抗rDer p 2ハイブリッド抗血清、あるいは対応する免疫前血清と共に、プレートを4℃で一晩インキュベートした。0.5% w/v BSAを含むPBSTでマウス抗血清を1:20で希釈し、ウサギ抗血清を1:100で希釈した。参考文献44および45に記載されているように、プレートを洗浄後、ダニアレルギー患者の血清を1:10で希釈した溶液と共に、4℃で一晩インキュベートし、0.5% w/v BSAを含むPBSTで1:2500に希釈したHRP結合ヤギ抗ヒトIgE抗体(KPL、USA)を用いて、結合したヒトIgEを検出した。IgE結合抑制率を次式にしたがって算出した。

rDer p 2およびrDer p 2誘導体で免疫刺激したマウスIgG1抗体について、ダニアレルギー患者IgEのrDer p 2に対する結合抑制能を、ELISA競合実験により調べた。
細胞培養培地(100ml RPMI 1649、10% FCS、4mM L−グルタミン、2mM ピルビン酸塩ナトリウム、10mM HEPES、100μM 2−メルカプトエタノール、1% Pen/Strep)中のラット好塩基球白血病(RBL)細胞(subline RBL−2H3)を、ELISAプレート(Nunc、Denmark)(100μl:4×104細胞)上に播種し、5%二酸化炭素雰囲気下、37℃で一晩おいた。
MP12で免疫した後誘導された抗体が、食品に派生する植物由来のプロフィリンと同様に、花粉由来プロフィリンを認識するか否かを実験するために、ELISA実験を行った。
MP12で刺激したウサギIgGが、Phl p12、他の花粉由来プロフィリン、および植物食品由来プロフィリンに対するアレルギー患者IgEの結合を抑制するか否かを、ELISA競合実験により調査した。

ここで、ODIおよびODPは、ウサギ免疫血清および対応する免疫前血清とプレインキュベートした後の吸光度をそれぞれ表している(表7)。
Claims (29)
- アレルゲン活性が低下した野生型タンパク質アレルゲン誘導体を生成する方法であって、
アレルゲン活性を有する野生型タンパク質アレルゲンを用意する工程と、
上記野生型タンパク質アレルゲンを、アレルゲン活性が低下した、またはアレルゲン活性が欠如した2つのフラグメントにスプライシングする工程と、
上記2つのフラグメントを逆方向に再結合する工程とを含むことを特徴とする方法。 - 上記誘導体は、宿主によって組換えタンパク質として生成され、当該宿主は特に高い発現能力を有することを特徴とする請求項1に記載の方法。
- 上記野生型タンパク質アレルゲンは、Phl p 12に代表されるプロフィリン、Bet v 4に代表されるカンバアレルゲン、Der p 2に代表されるチリダニアレルゲン、Lep d 2に代表される貯蔵庫ダニアレルゲン、およびPhl p 7に代表されるオオアワガエリアレルゲンからなる群より選択されることを特徴とする請求項1または2に記載の方法。
- 上記野生型アレルゲンと比較したときの上記誘導体によるIgE結合能の抑制低下率が、少なくとも10%、好ましくは20%、さらに好ましくは30%であることによって、上記誘導体のアレルゲン活性の低下を評価することを特徴とする請求項1から3のいずれか1項に記載の方法。
- アレルゲンに感作した患者の血清IgE抗体が、点染色した上記誘導体に対して結合しないことによって、上記誘導体のアレルゲン活性の低下を評価することを特徴とする請求項1から4のいずれか1項に記載の方法。
- 上記誘導体を薬学的に許容される添加剤と組み合わせて、医薬調整物とすることを特徴とする請求項1から5のいずれか1項に記載の方法。
- 上記誘導体を適当なワクチン補助剤と組み合わせて、薬学的に許容されたワクチン調整物とすることを特徴とする請求項1から6のいずれか1項に記載の方法。

上記誘導体は、野生型アレルゲンの2つのフラグメントを、N末端からC末端に向かう方向に隣接して含有し、N末端側の当該フラグメントは、X〜Zのアミノ酸からなるアミノ酸配列により示され、C末端側の当該フラグメントは、1〜Xのアミノ酸からなるアミノ酸配列によって示されるものであり、当該2つのフラグメントは、アレルゲン活性が低下または欠落していることを特徴とするアレルゲン誘導体。 - 上記X〜Zのアミノ酸からなるアミノ酸配列、および上記1〜Xのアミノ酸からなるアミノ酸配列は、少なくとも30アミノ酸、好ましくは少なくとも50アミノ酸、さらに好ましくは少なくとも60アミノ酸からなることを特徴とする請求項11に記載のアレルゲン誘導体。
- 上記X〜Zのアミノ酸からなるアミノ酸配列と、上記1〜Xのアミノ酸からなるアミノ酸配列とは、その全長において、50%未満、好ましくは30%未満、より好ましくは20%未満相違することを特徴とする請求項11または12に記載のアレルゲン誘導体。
- 上記野生型アレルゲンは、I型アレルゲン、好ましくは表1に示すのアレルゲン、より好ましくはPhl p12に代表されるオオアワガエリ(Phelum pratense)花粉アレルゲン、Bet v 4に代表されるカンバ(Betula verrucosa)花粉アレルゲン、スズメバチ(Vespula vulgaris)毒アレルゲン、アシナガバチ(Polistes annularis)毒アレルゲン、カベイラクサ花粉アレルゲン、ライグラス花粉アレルゲン、Der p 2に代表されるチリダニアレルゲン、またはこれらの混合物であることを特徴とする請求項11から13のいずれか1項に記載のアレルゲン誘導体。
- 請求項11から14のいずれか1項に記載のアレルゲン誘導体と、さらなるアレルゲン、好ましくは野生型アレルゲン、より好ましくは野生型アレルゲンの混合物、組換え野生型アレルゲンの混合物、野生型タンパク質アレルゲン誘導体の混合物またはそれらの混合物とを含有していることを特徴とするアレルゲン組成物。
- アレルゲン抽出物をさらに含有していることを特徴とする請求項15に記載のアレルゲン組成物。
- 薬学的に許容される添加剤をさらに含有していることを特徴とする請求項15または16に記載のアレルゲン組成物。
- 請求項11から17のいずれか1項に記載のアレルゲン誘導体またはアレルゲン組成物を、アレルゲン特異的免疫療法の調整医薬として使用する使用方法。
- 請求項11から17のいずれか1項に記載のアレルゲン誘導体またはアレルゲン組成物を、受動免疫のための調整医薬として使用する使用方法。
- 請求項11から17のいずれか1項に記載のアレルゲン誘導体またはアレルゲン組成物を、予防免疫のための調整医薬として使用する使用方法。
- 上記調整医薬は、補助剤、希釈剤、防腐剤またはこれらの混合物をさらに含有することを特徴とする請求項18から20のいずれか1項に記載の使用方法。
- 上記組換えアレルゲン誘導体を10ng〜1g、好ましくは100ng〜10mg、より好ましくは0.5〜200μg含有することを特徴とする請求項18から21のいずれか1項に記載の使用方法。
- 請求項11から17のいずれか1項に記載のアレルゲン誘導体を生成する方法であって、
請求項11から17のいずれか1項に記載のアレルゲン誘導体をコードするDNAを用意する工程と、
上記DNAを用いて宿主細胞を形質転換する工程と、
上記宿主細胞において上記誘導体を発現させ、上記誘導体を単離する工程とを含むことを特徴とする方法。 - 上記宿主細胞は、高い発現能力を有していることを特徴とする請求項23に記載の方法。
- 請求項11から17のいずれか1項に記載のアレルゲン誘導体を、化学合成により生成することを特徴とするアレルゲン誘導体の生成方法。
- 請求項1から10または23から25のいずれか1項に記載の方法によって得られる第1の野生型プロフィリン分子から得られるプロフィリン誘導体、または
請求項11から14のいずれかに記載された第1の野生型プロフィリン分子のアレルゲン誘導体を、
第2の野生型プロフィリン分子によって引き起こされるアレルギー疾患を予防または治療するための調整医薬を生成するために用いる使用方法。 - 上記第1および上記第2の野生型プロフィリン分子は、Phl p 12、Bet v 2、Art v 4、Ana c、Api g 4、Mus xp 1、Cor a 2、およびDau c 4からなる群より選択されることを特徴とする請求項26に記載の使用法。
- 上記第1の野生型プロフィリン分子はPfl p 12であり、かつ上記第2のプロフィリン分子は、Bet v 2、Art v 4、Ana c、Api g 4、Mus xp 1、Cor a 2、およびDau c 4からなる群より選択されることを特徴とする請求項26に記載の使用法。
- プロフィリンアレルギーに寄与する花粉と食品との交差性感作の治療および予防の少なくとも一方のための調整医薬の生成に用いることを特徴とする請求項27または28に記載の使用法。
