Abstract
Description
これらの低アレルギー誘発性のアレルゲンの変異体は、草花粉症の患者の特定の免疫療法(低感作)または草花粉症の予防的免疫療法のために用いることができる。
本発明の好ましい態様は、オオアワガエリ(timothy grass)(Phleum pratense)の花粉からの主要アレルゲンPhl p 1の変異体に関する。
I型アレルギーは世界的な重要性を有する。先進工業国の人口の20%までがアレルギー性鼻炎、結膜炎または気管支喘息などの病訴にさいなまれている。これらのアレルギーは、植物の花粉、ダニ、イヌまたはネコなどの、さまざまな発生源により遊離する、空気中に存在するアレルゲン(アエロアレルゲン)により引き起こされる。40%までのこれらのI型アレルギーの罹患者は同様に、草花粉アレルゲンで特異的なIgE反応性を示す(Freidhoff et al., 1986, J. Allergy Clin. Immunol. 78: 1190-2002)。
オオアワガエリ(Phleum pratense)の場合、Phl p 1(Petersen et al., 1993, J. Allergy Clin. Immunol. 92: 789-796)、Phl p 5(Matthiesen and Lowenstein, 1991, Clin. Exp. Allergy 21: 297-307; Petersen et al., 1992, Int. Arch. Allergy Immunol. 98: 105-109)、Phl p 6(Petersen et al., 1995, Int. Arch. Allergy Immunol. 108, 49-54)、Phl p 2/3(Dolecek et al., 1993, FEBS 335 (3), 299-304)、Phl p 4(Haavik et al., 1985, Int. Arch. Allergy Appl. Immunol. 78: 260-268; Valenta et al., 1992, Int. Arch. Allergy Immunol. 97: 287-294, Fischer et al., 1996, J. Allergy Clin. Immunol. 98: 189-198)およびPhl p 13(Suck et al., 2000, Clin. Exp. Allergy 30: 324-332; Suck et al., 2000, Clin. Exp. Allergy 30: 1395-1402)が今までに主要アレルゲンとして同定されてきている。
相同性のために、ドジョウツナギ(イネ科)のI型のアレルゲンは、ヒトIgE抗体との高い交差反応性を有する(Laffer et al., 1996, Mol. Immunology 33: 417-426)。この免疫学的交差反応性は、図1で選択された種からのI型分子と非常に類似した、Phleum pratense(Phleum pratense)のI型のアレルゲンであるPhl p 1との配列の比較により示される、アミノ酸配列を基にしている。
減感作に対する異なる概念は、防御免疫反応は特に、IgG4抗体により作用が阻害されることで誘発されるという事実に基づいている。この仮説に基づき、組換えPhl p 1断片は防御的IgG4反応の誘発に適合すると言われていると記述されてきた(Ball et al., 1999, FASEB J. 13:1277-1290)。この概念は低下したIgE反応性および維持されたT細胞反応性を有する低アレルギー誘発性のアレルゲンの変異体の概念と完全に異なる。
図1は、イネ科種:Poa pratensis (Poa p)、Holcus lanatus (Hol l)、Lolium perenne (Lol p)、Cynodon dactylon (Cyn d)、Oryza sativa (Ory s)およびPhalaris aquatica (Pha a)のPhl p 1相同性アミノ酸配列(cDNA配列から推定した成熟タンパク質の配列)の構造であり、全米バイオテクノロジー情報センター(NCBI, Bethesda, USA)の"GenBank"データベースからのcDNA配列から推定したタンパク質配列であり、番号は成熟タンパク質のアミノ酸部位であり、下線で強調するのはPhl p 1の配列と異なるアミノ酸であり、四角囲みはシステインであることを示す図である。
図3は、低アレルギー誘発性の変異体Phl p 1 NoCys、Phl p 1 NoCys Δ213-220およびPhl p 1 NoCys Δ1-6、115-119、213-220のアミノ酸配列の配置であり、例示の目的で示すものであり、番号はアミノ酸部位であり、下線により強調するのは欠失であることを示す図である。

図7は、4人の草花粉症罹患者(P)からの好塩基球での好塩基球活性化試験によるPhl p 1 NoCysの低アレルギー誘発性の決定を示す図である。
図9は、4人の草花粉症罹患者(P)からの好塩基球での好塩基球活性化試験によるPhl p 1 NoCys Δ1-6、115-119、213-220の低アレルギー誘発性の決定を示す図である。
このようにして、このI型のアレルゲンの配列はいまだに未知ではあるが、上記および下記で得られる結果はSecale cereale由来のSec c 1でもまた適用することができ、または、Sec c 1を用いて得られるであろうこともまた仮定できるに違いない。
配列番号2のアミノ酸配列を、野生型Phl p 1のcDNA配列から推測した。
Phl p 1は240個のアミノ酸からなっていて、天然形態ではグリコシル化されており、−全てのI型のアレルゲンのように(図1参照)−成熟分子中の7つのシステインの存在を特徴とする。Cyn d 1およびOry s 1を除外すると、これらのアミノ酸の位置は全てのI型のアレルゲン中の41、57、69、72、77、83および139番である(Petersen et al., 1995, J. Allergy Clin. Immunol 95: 987-994)。
低アレルギー誘発性のPhl p 1変異体の調製および特徴化
前述のrPhl p 1 wtのcDNAから始まり、遺伝子技術により修飾されたPhl p 1の組換え変異体が調製された。
該組換えタンパク質は、大腸菌においてヒスチジン融合タンパク質として発現した。該免疫学的特徴付けは、この種の融合タンパク質とともに実施された。
低下したIgE抗体の結合は、7つのシステインのうち少なくとも2つが置換されずに除去される、もしくは他のアミノ酸によって置換された場合に得られた。しかしながら、好ましくは、7つ全てのシステインがセリンにより置換される場合である。
Phl p 1 NoCysを基にして調製されたさまざまな欠失変異体を、IgE結合能(ストリップ試験、およびEAST)および機能的作動(塩基性活性化)に関して同様の方法により調査した。驚くことに、欠失変異体は特別に強い低アレルギー誘発性の特性を示した。
ここで特に好ましいのは、I型のアレルゲンPhl p 1、Poa p 1、Hol p 1、Lol p 1およびPha a 1の対応する欠失変異体である。非常に特に好ましいのは、対応するPhl p 1変異体である。
加えて、本発明は薬剤としての特性のある、上記のアレルゲンの変異体、DNA分子および発現ベクターに関する。
医薬組成物がセカンドタイプである場合(少なくとも1つのDNA分子または発現ベクターを含む)、これらの組成物は好ましくはさらに、水酸化アルミニウム、オリゴヌクレオチドを含む免疫刺激的CpGまたは前記の2つの組み合わせを、アジュバントとして含む。
さらに、遅延放出製剤は、本発明によるアレルゲンの変異体の適切な剤により、例えば水酸化アルミニウム上に吸収させることにより得ることができる。
変異体Phl p 1 NoCys、Phl p 1 NoCys Δ213-220およびPhl p 1 NoCys Δ1-6、115-119、213-220(図3)の調製およびその免疫学的特徴化を、上記の遺伝子工学修飾のある低アレルギー誘発性Phl p 1 変異体の例証として、以下に述べる。
組換えタンパク質は、大腸菌(JM109株)でヒスチジン融合タンパク(発現ベクターpProExHT;Invitrogen, Carlsbad, USA)として発現した。rPhl p 1 wtおよび該変異体を、はじめはN末端ヒスチジン残基のNi2+キレート基質(固定化金属イオンアフィニティークロマトグラフィー、IMAC)への特異的な結合により、それに続き調製ゲルろ過(サイズ排除クロマトグラフィー、SEC)により精製した。溶出した精製タンパク質の純度は、SDS−PAGEおよび分析的SECによりモニターした(図4A)。精製したタンパク質の同一性はPhl p 1特異的モノクローナル抗体の結合により実証された(図4B)。
膜結合性試料タンパク質での、アレルギー罹患者からの血清からの特異的なIgE反応性の決定のための単純な試験的方法は、ストリップテストである。
本目的のために、試験物質を同濃度および同量でニトロセルロース膜のストリップ上に非変性状態下で互いに平行に結合させる。一連のかかる膜ストリップは平行にアレルギー罹患者からの異なる血清とともに培養することができる。洗浄段階ののちに、抗ヒトIgE/アルカリフォスファターゼ接合により進められた呈色反応により、特異的に結合したIgE抗体は膜上で可視となる。
天然のPhl p 1に対して強いIgE力価を有するアレルギー罹患者からの血清のみを用いた。これらの患者からのIgE抗体は同様に組換えの同等なrPhl p 1 wtと反応する。
全ての患者の血清のPhl p 1特異的なIgE抗体が、組換え変異体Phl p 1 NoCysに低下した程度で結合するが、野生型アレルゲンPhl p 1にはしないことが明らかとなった。
このようにして、システインの置換ならびに特異的な配列部位の欠失の両方が、Phl p 1分子のIgE結合能を低下させることが明らかになる。
EAST阻害試験は、以下のように実行する。マイクロタイタープレートをアレルゲン、ここではnPhl p 1で被覆する。非結合アレルゲンを洗浄により除去した後に、後の非特異的な結合を防ぐためにウシ血清アルブミンを用いてプレートをブロックする。アレルギー罹患者からのIgE抗体を、個々の血清の提示プール(血清プール)として、または個々の血清として、好適な希釈でアレルゲンコートしたマイクとタイタープレートとともに培養する。アレルゲン結合IgE抗体の量は、色の付いた最終生成物にする基質の反応により共役した抗hIgE/アルカリフォスファターゼを介して光学的に定量化する。
Phl p 1 NoCys、Phl p 1 NoCys Δ213-220およびPhl p 1 NoCys Δ1-6、115-119、213-220の試験結果は、ここで参照nPhl p 1分子と比較して記述する修飾Phl p 1分子の例証として示す。
変異体Phl p 1 NoCys Δ213-220およびPhl p 1 NoCys Δ1-6、115-119、213-220に対する曲線は、これらのPhl p 1変異体のより低いIgE結合能を実証する。個々の種別の阻害作用は検出できないか、非常に小さな範囲(最大阻害作用の0〜20%)であろう。
ストリップ試験の結果に相同して、付加的な特異的欠失の挿入によりPhl p 1のIgE結合能をさらに低下させることを確認することができる。
アレルギー誘発性の機能的な低下は、in vitroにおいて好塩基球活性試験の手段により決定された。好塩基球活性化試験では、草花粉症罹患者からのヘパリン化された全血をさまざまな濃度の試験物質とともに培養する。アレルゲン性物質は特異的に好塩基球のFcεRI結合IgE抗体に結合し、結果としてFcεRI分子の架橋が起こる。
このアレルゲン誘発性、IgE推進性FcεRI架橋は、結果として好塩基球の活性化をもたらす。該活性化は、これらのエフェクター細胞のアレルギー性反応にける第1段階である。引き続くシグナル伝達は、結果としてエフェクター細胞の脱顆粒をもたらし、このようにしてin vivoにおけるアレルギー性反応の引き金をもたらす。
ストリップ試験およびIgE阻害試験からの結果と一致して、変異体Phl p 1 NoCys Δ213-220およびPhl p 1 NoCys Δ1-6、115-119、213-220の試験結果は、図8および9において代表的なカーブとの比較で示されるように、相対的なアレルゲン有効性におけるさらに大幅な減少を示唆する。
参照のnPhl p 1と比較して、A50価のカーブにより計算できる変異体Phl p 1 NoCys Δ213-220のアレルゲン有効性は約100〜1000倍まで低下し、変異体Phl p 1 NoCys Δ1-6、115-119、213-220のアレルゲン有効性は1000倍以上低下した。(A50:活性化好塩基球最大数の50%でのアレルゲン濃度)
ヘルパーTリンパ球はアレルゲンペプチド断片(約12〜25アミノ酸)と反応し、前記断片は抗原提示細胞(antigen-presenting cells)(APCs)において酵素変性により形成され、抗原提示細胞(APCs)表面でのMHCクラスII分子への好適なペプチドの包含ののちにT細胞に提示される。Tヘルパーリンパ球のこのアレルギー特異的な活性化は、増殖および機能的分化(TH1およびTH2)のための必要条件である。減感作中のアレルゲンまたはアレルゲン誘導体での処置によりアレルゲン特異的なTリンパ球の影響は、治療的有効性の鍵とされる。
表1で示す8人の草花粉症罹患者からのT細胞株での結果は、組換えアレルゲンの変異体によりTリンパ球を刺激して増殖させることはが可能であることを示した。Phl p 1 NoCys、Phl p 1 NoCys Δ213-220およびPhl p 1 NoCys Δ1-6、115-119、213-220のT細胞反応性は非修飾の天然および組換え野生型アレルゲンと比較してわずかしか低下しておらず、決定的なT細胞エピトープの保持を実証する。
2 ドナー:臨床的に定められた草花粉症罹患者
例1:Phl p 1 NoCys
変異体Phl p 1 NoCys(配列番号3および4)のために、rPhl p 1 wt ("GenBank" entry Z27090; NCBI, Bethesda, USA)から開始して6段階のPCRを実行した。システインの変わりにセリンをエンコードするコドンを含む特異的なPCRのプライマーを用いて、点変異を導入した(プライマーの配列は表2を参照のこと)。
ステップ1−N末端DNA断片"Phl p 1 [C41S、C57S、C69S] (bp 1-212)"の調製:PCRによる長い重複オリゴヌクレオチド(P 1-63、P 49-111、P 97-158およびP 144-212)の増幅によりC41S、C57S、C69S変異を含むDNA断片を産生した。
ステップ3−"Phl p 1 [C41S、C57S、C69S、C72S、C77S、C83S] (bp 1-720)"をエンコードするDNAの調製:P 1-63およびP 703-720 HindIIIプライマーを伴った重複断片"Phl p 1 [C41S、C57S、C69S] (bp 1-212)"および"Phl p 1 [C69S、C72S、C77S、C83S] (bp 193-720)" のPCR。
ステップ5−C末端DNA断片"Phl p 1 [C139S] (bp 406-720)"の調製:P 406-428sおよびP 703-720 HindIIIプライマーを伴ったrPhl p 1のcDNAのPCR。
Phl p 1 NoCysをエンコードするDNAを制限酵素HindIIIを用いて分解し、制限部位EheIおよびHindIIIを介して発現ベクターpProExHT(Invitrogen, Carlsbad, USA)へと結合させ、それに引き続き完全に配列決定した。
欠失変異体Phl p 1 NoCys Δ213-220(配列番号15および16)をエンコードするDNAを5’プライマーP 1-18および3’プライマーP 613-720 (Δ637-660)を用いて、欠失すべき配列部位によりHindIII特異的に短縮することによりPhl p 1 NoCysのDNAのPCRにより産生した。
cDNAを制限酵素HindIIIを用いて分解し、制限部位EheIおよびHindIIIを介して発現ベクターpProExHT(Invitrogen, Carlsbad, USA)へと結合させ、それに引き続き完全に配列決定した。
欠失変異体Phl p 1 NoCys Δ1-6、115-119、213-220 (配列番号5および6)をエンコードするDNA配列を、欠失すべき配列領域により特異的に短縮したオリゴヌクレオチドを用いてPCRにより3段階で産生した。
ステップ1−N末端DNA断片"Phl p 1 NoCys Δ1-6 (bp 1-300)"の調製:プライマーP22-63 (Δ1-18)およびP 250-318を伴ったPhl p 1 NoCysのcDNAのPCR。
ステップ3−Phl p 1 NoCys Δ1-6、115-119、213-220をエンコードする完全形DNAの調製:プライマーP22-63 (Δ1-18)およびP 703-720 HindIIIを伴った重複断片"Phl p 1 NoCys Δ1-6 (bp 1-300)"および"Phl p 1 NoCys Δ115-119、213-220 (bp 283-663)"のPCR。
変異体Phl p 1 NoCys Δ1-6(配列番号5および6)、Phl p 1 NoCys Δ1-30(配列番号7および8)、Phl p 1 NoCys Δ92-104(配列番号9および10)、Phl p 1 NoCys Δ115-119(配列番号11および12)、Phl p 1 NoCys Δ175-185(配列番号13および14)を、同様に調製し、クローン化し、そして配列決定した。
