Abstract
Description
1.これまで、全ての有効な炭疽ワクチンはPA(すなわち、83kDaプロ型またはその活性化された63kDa誘導体のいずれか)を含むかまたは生産する。実際、現行の定説は、有効な炭疽ワクチンの製造のためにPAが必要であり、それのみで十分であるというものであり、このようなワクチンを開発する試みが進行中である[例えば、Baillie, L.(2001), 91, 609-613頁を参照のこと];
2.非莢膜形成毒素産生性生胞子ワクチンは、認可された無細胞ワクチンよりも、これまで試験された全ての炭疽菌株に対してより高い程度の防御を引き起こす[Little, S. F. (1986) Inf. and Immunol. 52巻, 2号, 509-512頁を参照のこと];
3.現行の無細胞ワクチンは、一般に定義が不十分であり、バッチ毎で有効性が大きく変動し得る。したがって、各バッチが、ヒトへの使用の前に動物モデルにおける効力について個々に試験されなければならない;
4.現行の無細胞炭疽ワクチン製造プロセスは、製造プロセスおよび最終製品の梱包の完了時にのみ評価される。したがって、ワクチン材料のいずれか1つのバッチが検証試験基準に合わない場合、寄与因子は容易に同定できない。このような因子は、製造バッチ間で異なり得、そしてこの理解に欠けることにより、製造プロセスで遭遇する困難性が悪化される;
5.現行の無細胞ワクチンのあまり十分に定義されていない性質の結果として、これらのワクチンは、LFおよび/またはEFと共に多量のPAを含有し得る。このことにより、63kDa型へのPAのインビボ(もしくはインビトロ)での活性化に際して、LTおよびETを形成してワクチンの受容者に副作用を生じさせ得る。もちろん、このようなワクチンはまた、他の炭疽菌タンパク質(分泌産物および溶解産物ともに)、ペプチドグリカン、核酸および炭水化物を含み得、これらは、防御効力を損ない得る;
6.現行の無細胞ワクチン組成物は、LF、PA、およびEF濃度が非常に変動し得、そのためいくつかの調製物からEFがなくなり得る;および
7.現行の無細胞組成物は、総タンパク質含有量が非常に変動し得る。したがって、所定の組成物中に存在する毒素成分の濃度は、顕著に変動し得る。したがって、これにより、ヒトでの効力および潜在毒性に影響を及ぼし得る。

以下のいずれかの発現を指向するpMTL発現ベクター構築物を生成した:野生型PA遺伝子配列または改変型rPA遺伝子配列(エルウィニア・カロトボーラ(Erwinia carotovora)pelBまたはシュードモナスカルボキシペプチダーゼG2(cpg2またはcpg)リーダー配列のいずれかに融合させた)。後者の配列は、欧州特許0 121 352に記載されており、大腸菌で効率的にプロセシングされて、可溶性タンパク質をペリプラズム腔に方向付けることが示されている。これらのrPA発現構築物を、評価のために、Dynport Vaccine Company(DVC)のpET26bおよびInvitrogenのpTrkベクターベースの構築物と比較した。
1.ともにpelB、ompA、およびcpg2リーダー配列に融合させた2つのPAをコードするヌクレオチド配列(野生型および改変型)をPCR増幅する。このことにより、6つのrPA配列選択肢が生じた;
2.PCR産物クローニングベクター系(例えば、Invitrogen TAクローニング)で一次クローンを構築した;
3.一次クローンをPA/リーダー配列複合体のDNA配列分析によって確認した;
4.6つのrPA配列をpMTL1015発現ベクターにサブクローニングし、そして組換えプラスミドを確認した;
5.4つの確認したクローンに由来するプラスミドDNAを用いて、プロテアーゼ欠損発現株である大腸菌RV308(ATCC31608)を形質転換した。
pMTL発現プラスミドに基づく6つのクローンを、現行の生産培地での振盪フラスコ培養で評価した。既存のDVC生産生物をコントロールとして用いた。増殖条件および誘導開始/持続期間(適用可能な場合)は、所与の実験条件下での発現レベルの真の比較をすることが可能になるように、可能な限りで標準化した。例えば、接種のために標準細胞密度を用いた。生産レベルを、増殖の全期間にわたって培養物をサンプリングし、そして回収した細胞のBugBusterTM(Novagen)を用いる化学的細胞溶解に供することにより、SDS−PAGEのデンシトメトリー分析により、比較した。ウェスタンブロッティングを用いて、rPAタンパク質バンドの同一性を確認した。
遺伝子操作によりrPAの発現レベルを増大させることに加え、より高い栄養素レベルを含む培地を用いて培養物をより高い細胞密度にまで増殖させることにより、産物の最終収量を上昇させた。
(2.1 フィトンペプトンベースのTerrific Broth)
振盪フラスコ培養を用いて、フィトンペプトンベースのTerrific Brothでの6つのpMTL1015クローンによるrPAの発現を比較する実験を行った。CAMRでのpMTL1015発現系を用いる以前の研究は、低い酸素添加速度が産物の発現に好ましいものであり得ることを示したので、培養をバッフル付フラスコ(高酸素添加)およびバッフルなしのフラスコ(低酸素添加)の両方で設定した。
上記の実験(実施例2.1)を、バッフル付フラスコのみでHy−soyベースの半限定培地を用いて繰り返した。増殖曲線(図6、7および8)は、この培地で、Terrific Brothに比較して低い増殖速度および最終細胞密度が得られたこと、および合成遺伝子を発現しているクローンについて8時間までの停滞期があったことを示している。rPA発現レベルは、一般に、フィトンペプトンベースのTerrific Brothで観察されるよりも低かった。しかし、野生型遺伝子と比較して合成遺伝子を発現するクローンによる優れた発現レベルの同様のパターンが、SDS−PAGE(図9)およびELISA(表3)によって観察された。
発酵培養における以下の4つのさらに選択したクローンについて評価を続けた:
大腸菌RV308 pMTL1015−cpg−PA−synt
大腸菌RV308 pMTL1015−ompA−PA−synt
大腸菌RV308 pMTL1015−ompA−PA−wt
大腸菌DH5 pTrcK−pelB−PA−synt。
前に記載したような条件下で各培地において、DOTおよびpHを調節しながら、8L発酵を実施した。
大腸菌RV308 pMTL1015−cpg−宿主/ベクター系を組み込んでいる前の開発プログラムは、産物の発現が25〜30℃の間の温度で最も効率的であることを示していた。
抗生物質減少による選択圧下でのプラスミドの安定性の確認として、大腸菌RV308 pMTL1015−cpg−PA−syntを、培地中のテトラサイクリン濃度レベルを変更させながら、上記条件下、生産培地で培養した(表4のPRECRV0042〜0044を参照のこと)。テトラサイクリン濃度は、15μg/mL(100%)、1.5μg/mL(10%)および0であった。第二の種培養物はそれぞれ15、1.5、および15μg/mLを含んだ。したがって、種増殖の間のテトラサイクリンの分解がないと仮定すると、抗生物質を添加していない発酵槽は、選択圧を供給するために第二の種からのキャリーオーバーに頼った。発酵槽に移した種の容量は124mLであり、接種時の発酵培地に名目上0.23μg/mLのテトラサイクリンを与えた。
初期のダウン選択後に調べたクローンについてこれまで得られた結果は、4つのうち、大腸菌RV308 pMTL1015−cpg−PA−syntが、等しい条件下で他のpMTL1015クローンと比較した場合、ほとんどの場合において最も高い収量を示したことを示していた。
(4.1 大腸菌RV308(ATCC 31608)に形質転換した目的のクローン[クローンpMTL1015−cpg−PA−synt]のシードバンク)
配列確認後、研究用シードバンクを、ダイズペプトンベースのL−ブロス(フィトンペプトン15g/L、バクト酵母エキス5g/L、NaCl 5g/L、pH6.8〜7.0)でのテトラサイクリン(15mg/L)の選択圧下での増殖によって調製した。テトラサイクリンを含む栄養寒天プレートからの1つのコロニーを、500mLバッフル付振盪フラスコ中の100mL培地に接種し、そしてOD600が1.5になるまで振盪インキュベーター中で30℃にて150rpmでインキュベートした。次いで、この培養物を増殖培地(上記を参照のこと)中で滅菌50%グリセロールと混合して10%の最終グリセロール濃度とし、そして1.8mL凍結バイアル中に1mLアリコートとして−80℃で凍結保存した。
1バイアルのWRCBを溶解し、そして100μLを、25mLユニバーサルボトル中の15mg/Lでテトラサイクリンを含む10mLのダイズペプトンベースのL−ブロス(上記を参照のこと)に接種し、150rpmで振盪しながら30℃にて7〜9時間インキュベートした。これは、生物が生存していることを確実にし、そしてより一貫した種菌生産プロセスを得る回収工程であった。この工程の最終OD600は0.7〜1.0であった。
この工程は、発酵用の接種材料を生産し、そして合理的な大きさの振盪インキュベーターを用いて、振盪フラスコ中での5〜250Lの培養に十分な接種材料を製造し得る。50Lスケールでは、200μLの一次種菌を、5×1000mLバッフル付振盪フラスコの各々の中の200mLの生産培地に接種した。この培養物を、150rpmで振盪しながら30℃にて11〜12時間インキュベートし、13〜16の最終OD600を得た。
次いで、種菌培養物をひとまとめにして、発酵槽中で出発OD600が0.2となるのに十分な容量を、72LのApplikon攪拌発酵タンク中の50L生産培地(上記を参照のこと)に接種した。複合培地成分を、40Lバルクとして、121〜123℃にて30分間インサイチュで滅菌し、25℃を下回るまで冷却し、次いで残りの成分を補充して総容量を50Lとした。
(5.1 細胞破壊)
約4.5kgの凍結細胞ペースト回収物を、まず最小容量の20mMトリス/1mM EDTA pH8.5によって懸濁し、滑らかなペーストとした。さらなる緩衝液を添加して16Lの総懸濁液容量を得た。
遠心分離したホモジネートを、2つの「Pall」OS030F07 0.5m2「Centrasette 2 Omega」懸架式スクリーンチャンネル30kDa膜が取り付けられた「Millipore Pellicon」濃縮器を用いて、その容量の3倍の精製水でダイアフィルトレートした。この濃縮器を、1.6バールの膜透過圧で17L/分の流速で操作した。pHを8.0に調整し、そして伝導度を2mS/cmに調整した。
25cm直径のクロマトグラフィーカラムに5Lの「Amersham」「Q−セファロースファストフロー(Q-Sepharose Fast Flow)」アニオン交換体を充填して10cmのベッド高とした。次いで、工業用UVモニターを流出ラインに接続した。カラムを全体にわたって330mL/分の流速で操作した。充填カラムを10Lの水、次いで5Lの0.5M水酸化ナトリウム、引き続き精製水で洗浄した。10Lの0.5Mトリス、pH8.0をポンプで注入し、次いでカラムを開始緩衝液(20mMトリス、pH8.0)で平衡化した。
UVモニターに接続した30cm直径のカラムに、7L/分の流速で20Lの「Ciphergen」「MEPハイパーセル(HyperCel)」を充填した。充填すると、さらに続く全ての工程を800mL/分で実施した。カラムを、40分以下の接触時間で5Lの1M水酸化ナトリウムで洗浄した。次いで、カラムを水で洗浄し、次いで20Lのローディング緩衝液(50mMトリス、pH8.0)で平衡化した。Qプール(すなわち、前のQクロマトグラフィー工程からのプール)を流し、カラムをローディング緩衝液でベースラインまで洗浄し、次いで結合したrPAを精製水で溶出させた。収集した産物をSDS−PAGEおよびSEC−HPLCによってアッセイした。MEPプール(すなわち、MEPハイパーセルカラムからのプール)を0.22μmの2000cm2のPall「ポジダイン(Posidyne)」フィルターを通して濾過した。カラムを10Lの1M水酸化ナトリウムで再生し、精製水で洗浄し、次いで0.2μm濾過した50mM水酸化ナトリウム中に保存した。
精製したrPAを、「Pall Centramate」媒体スクリーン「Omega」50kDaカートリッジ(部品番号OS0350C12、0.093m2)を用いてダイアフィルトレートした。800mL/分の流速および1.6バールの膜透過圧を用いた。ダイアフィルトレーションを、5Lの処方緩衝液;25mMリン酸ナトリウム、150mM塩化ナトリウム、pH8.0に対して実施した。5000cm2面積の0.22μm Pall「ポジダイン」フィルターを用いてさらなる濾過を実施し、次いで最終産物を適切なバイアル中に分配した。

Claims (60)
- 配列番号1に対して少なくとも75%の同一性を有する核酸配列であって、組換え炭疽菌防御抗原(rPA)をコードする核酸配列;または
該核酸配列のフラグメントであって、組換え炭疽菌防御抗原(rPA)のフラグメントをコードするフラグメント
を含む、ポリヌクレオチド配列。 - 前記核酸配列が、配列番号1に対して少なくとも80%の同一性を有する、請求項1に記載のポリヌクレオチド配列。
- 前記核酸配列が、配列番号1に対して少なくとも90%の同一性を有する、請求項2に記載のポリヌクレオチド配列。
- 前記組換え炭疽菌防御抗原(rPA)のフラグメントが、配列番号5の少なくとも200、好ましくは少なくとも300、および最も好ましくは少なくとも400のアミノ酸残基を含む、請求項1に記載のポリヌクレオチド配列。
- 前記核酸配列のフラグメントが、請求項1に記載の核酸配列の少なくとも600、好ましくは少なくとも900、および最も好ましくは少なくとも1200のヌクレオチドを含む、請求項1に記載のポリヌクレオチド配列。
- 分泌配列をコードするポリヌクレオチドをさらに含む、請求項1から5のいずれかに記載のポリヌクレオチド配列。
- 前記分泌配列が、ompA、ompT、ompF、pelB、phoA、lamB、βラクタマーゼ、黄色ブドウ球菌プロテインA、枯草菌エンドグルカナーゼ、マウスRNAse、ヒト成長ホルモン、エンテロトキシンST−II、LT−AもしくはLT−B、およびcpg2(cpg)から選択される、請求項6に記載のポリヌクレオチド配列。
- 前記分泌配列が、発現されたポリペプチドの細菌宿主細胞の細胞質からのペリプラズム転位のためである、請求項6または請求項7に記載のポリヌクレオチド配列。
- 前記分泌配列が、発現されたポリペプチドの細菌宿主細胞の細胞質からの細胞外転位のためである、請求項6または請求項7に記載のポリヌクレオチド配列。
- 前記分泌配列が、ペリプラズム転位の間に前記発現されたポリペプチドから切り離され得る、請求項8に記載のポリヌクレオチド配列。
- 前記分泌配列が、細胞外転位の間に前記発現されたポリペプチドから切り離され得る、請求項9に記載のポリヌクレオチド配列。
- 前記分泌配列が、配列番号3によりコードされるcpg2(cpg)リーダー配列である、請求項6から11のいずれかに記載のポリヌクレオチド配列。
- 前記核酸またはそのフラグメントが3’末端および5’末端を有し、そして該核酸またはそのフラグメントが該5’末端に対してクローニングされたメチオニン残基をコードするコドンを有する、請求項1から12のいずれかに記載のポリヌクレオチド配列。
- 前記核酸が配列番号7またはそのフラグメントであり、該フラグメントが配列番号7の5’末端コドンを含む、請求項13に記載のポリヌクレオチド配列。
- 請求項1から14のいずれかに記載のポリヌクレオチドを含む発現ベクター。
- rPAポリペプチドまたはそのフラグメントが高度に発現されることを確実にするように選択されるプロモーターを含む、請求項15に記載の発現ベクター。
- 選択マーカーをさらに含む、請求項15または請求項16に記載の発現ベクター。
- 前記ベクターが、pETベクター、pTrKHisベクター、およびpMTLベクターから選択される、請求項15から17のいずれかに記載の発現ベクター。

- 前記ベクターが、配列番号4によりコードされるプラスミドpMTL1015である、請求項16から21のいずれかに記載の発現ベクター。
- 前記ベクターが、ECACC番号04061401で寄託されている、請求項16から21のいずれかに記載の発現ベクター。
- 請求項15から23のいずれかに記載の発現ベクターを含む宿主細胞。
- 前記宿主が大腸菌細胞である、請求項24に記載の宿主細胞。
- 前記大腸菌細胞が大腸菌RV308(ATCC番号31608)である、請求項25に記載の宿主細胞。
- rPAまたはそのフラグメントを製造する方法であって、請求項1から14のいずれかに記載のポリヌクレオチドを発現させる工程を含む、方法。
- 前記ポリヌクレオチドが宿主細胞で発現される、請求項27に記載の方法。
- 前記ポリヌクレオチドがDNAであり、そして必要に応じてインビトロにてRNAにまず転写され、次いで該RNAが宿主細胞で翻訳される、請求項27または請求項28に記載の方法。
- 前記宿主細胞が大腸菌宿主細胞である、請求項28または請求項29に記載の方法。
- 前記大腸菌宿主細胞が大腸菌RV308(ATCC番号31608)である、請求項30に記載の方法。
- 請求項15から23のいずれかに記載の発現ベクターからrPAを発現させる工程を含む、請求項27から31のいずれかに記載の方法。

- 前記発現ベクターを宿主細胞に形質転換する工程、および増殖培地で該形質転換した宿主細胞を培養する工程の初期工程をさらに含む、請求項28から35のいずれかに記載の方法。
- 前記増殖培地が、動物製品を実質的に含まない、請求項36に記載の方法。
- 前記形質転換した宿主細胞を低温で培養する工程を含む、請求項36または請求項37に記載の方法。
- 前記宿主細胞を回収する工程をさらに含む、請求項36から38のいずれかに記載の方法。
- 前記宿主細胞からrPAを抽出する工程をさらに含む、請求項39に記載の方法。
- 分離工程をさらに含む、請求項27から40のいずれかに記載の方法。
- 前記分離工程が、1つ以上のクロマトグラフィー工程および1つ以上の濾過工程から選択される、請求項41に記載の方法。

- 前記クロマトグラフィー工程の少なくとも1つがイオン交換クロマトグラフィー工程および疎水性電荷クロマトグラフィー工程から選択される、請求項42または請求項43に記載の方法。
- 請求項28から44のいずれかに記載の方法であって、
(a)請求項1から14のいずれかに記載のポリヌクレオチドまたは請求項15から23のいずれかに記載の発現ベクターを発現する宿主細胞を得る工程;
(b)該宿主細胞から発現されたrPAを抽出する工程;
(a)該抽出されたrPAをダイアフィルトレーション工程に供する工程;
(b)該ダイアフィルトレートされたrPAを、アニオン交換クロマトグラフィーおよび疎水性電荷クロマトグラフィーから選択される少なくとも1つのクロマトグラフィー工程に供する工程;および
(c)さらなるダイアフィルトレーション工程を実施する工程
を含む、方法。 - 請求項27から45のいずれかに記載の方法により製造されたポリペプチドまたはそのフラグメント。
- 請求項13または請求項14に記載のポリヌクレオチドによりコードされたポリペプチドまたはそのフラグメント。
- 請求項1から14のいずれかに記載のポリヌクレオチド;請求項15から23のいずれかに記載の発現ベクター;請求項24から26のいずれかに記載の宿主細胞;および請求項46または請求項47に記載のポリペプチド;の1つ以上を含むキット。
- 請求項46または請求項47に記載のポリペプチドを含む抗原性組成物。
- 炭疽菌による感染に対して免疫応答を誘発する方法であって、請求項46または請求項47に記載のポリペプチドまたは請求項49に記載の抗原性組成物を投与する工程を含む、方法。
- 炭疽菌による感染に対して防御するための、請求項50に記載の方法。
- 炭疽菌による感染に対して免疫応答を誘発するための医薬の製造のための、請求項46または請求項47に記載のポリペプチドの使用。
- 炭疽菌による感染に対して防御するための、請求項52に記載の使用。
- 配列番号4の配列を有するベクター。
