Abstract
【解決手段】過去に多数のエピトープに対し初回刺激されたことがあるか又はそれらを被曝したことがある患者における免疫応答を追加刺激する改良された様式を決定した。その改良された方法は、追加刺激期において、エピトープが個々に投与されること、即ち別々のペプチド構築物上に保持されたエピトープが投与されることを必要とする。さらに、予防接種戦略を改良するための材料及び方法であって、排他的にではないが、免疫系が、初回刺激組成物により誘導され、そして追加刺激組成物の投与により追加刺激される「初回刺激−追加刺激」予防接種プロトコールに関し、新規の四量体可溶性クラスIMHC/ペプチド複合体とする。

材料及び方法
プラスミドDNA構築物
研究の全体にわたり使用されたDNAベクターpSG2は、SV40複製開始点及びネオマイシン耐性マーカーを含有しているBamHI断片を除去し、CMVプロモーターを、イントロンAを含有している同プロモーターのより長いバージョンに交換することにより、pRc/CMV(Invitrogen、Paisley、UK)から派生した。得られたプラスミドは、マルチクローニングサイト及びウシ成長ホルモンポリA配列の前に、真核細胞における発現のためのイントロンAを含むCMVプロモーターを含有している。このプラスミドは、哺乳動物細胞における複製能を有しない。mel3配列(表1)をコードする遺伝子が、標準的な方法を使用してマルチクローニングサイトへ導入された。注射用のプラスミドDNAは、陰イオン交換クロマトグラフィー(Qiagen、Hilden、Germany)を使用して精製され、リン酸緩衝生理食塩水(PBS)で1mg DNA/mlに希釈された。
組換え及び非組換えのMVAは、10%ウシ胎仔血清(FCS)が補足された最小必須培地において増殖したニワトリ胚繊維芽細胞(CEF)において、常法により繁殖させられ滴定された。組換えMVAは、ワクシニアシャトルベクターpSC11へmel3ポリエピトープストリング(表1)をクローニングすることにより、記載されたようにして作成された。1細胞当たり0.05pfuの多重度でMVAにより感染されたCEFが、記載されたようにして23、リポフェクチン(Gibco)及びシャトルプラスミドによりトランスフェクトされた。ワクシニアP7.5プロモーターがポリエピトープの発現を駆動する。組換えMVAは8回プラーク精製された。
mel3ポリエピトープストリングが、トランスファーベクターpSFV4.2-mel3へクローニングされた。このベクターから作製されたRNAが、組換えSFVmel3粒子を構築するために使用された。組換えSFVストックは、以前に記載されたようにして16、作成され精製された。
ヒトCTLクローンは、記載されたようにして25、単離された。簡単に説明すると、5%ヒト血清、100U/ml IL-2が補足されたイスコブ(Iscove)培地中の105個の照射済みB細胞を含有している、PBS中100ng/mlの抗CD3及び抗CD28の両方で予めコーティングされたU底96穴プレートへ、四量体/CD8二重陽性染色CTL培養物が単一細胞として分取された。増殖性クローンを、>107個にまで増幅し、標準的なCr51放出アッセイ法のためのエフェクターとして使用した。本発明者らは、HLA A2拘束性CTLの標的としてMVA mel3により感染されたJY B細胞を使用し、HLA-A1拘束性エピトープに関してはXY B細胞を使用し、Db拘束性インフルエンザ核タンパク質の標的としてH-2b陽性MC57細胞を使用した。本発明者らは、標的に対して3倍希釈でCTLクローンを滴定した。
蛍光四量体HLA-A2.1/ペプチド複合体は、以前に記載されたようにして1、合成された。A2-Kb/ペプチド複合体は、A2.1分子のα1ドメイン及びα2ドメイン並びにH-2Db分子のα3ドメインのキメラ重鎖をヒトβ2-ミクログロブリンと共に使用して、類似の方式で合成された。
赤血球溶解緩衝液(Invitrogen、Paisley、UK)を使用して、尾静脈から採取された血液より、新鮮なPBLが単離された。四量体染色については、3×105個の細胞が、10%FCSが補足されたRPMI1640(Sigma、St Louis、MO)20μlに再懸濁させられた。細胞は、37℃で15分間、四量体と共にインキュベートされた。次いで、PBLは、4℃で15分間、ラット抗マウスCD8α(Pharmingen、San Diego、CA)と共にインキュベートされた。細胞は、PBSで2回洗浄され、FACScan(Becton Dickenson、Mountain View、CA)分析のためPBSに再懸濁させられた。
HHDマウスは、ヒトβ2mのC末端が、キメラ重鎖(HLA-A2.1 α1-α2、H-2Dbα3膜貫通ドメイン及び細胞質内ドメイン)のN末端に共有結合的に連結されているトランスジェニック単鎖組織適合性クラスI分子を発現している20。これらのマウスのH-2Db遺伝子及びマウスβ2m遺伝子は、相同的組み換えにより破壊されており、血清学的に検出可能なマウス組織適合性クラスI分子の細胞表面発現は完全に欠如している。A2-Kbマウスは、マウスβ2mと非共有結合的に会合したキメラ重鎖(HLA-A2.1α1α2、H-2Kbα3膜貫通ドメイン及び細胞質ドメイン)を発現している。それらは、さらに、C57BL/6に由来する(H-2b)クラス1a及び1bマウス組織適合性分子のフルセットを発現している26。使用された全てのA2トランスジェニックマウスが、本発明者らの動物施設において交配された。4〜6週齢の雌C57/BL6マウスは、ハーラン・オーラック(Harlan Orlac)(Shaws Farm、Blackthorn、UK)より得られた。プラスミドDNA(25〜50μg/筋肉)がPBSに溶解させられ、全身麻酔下の各頸骨筋へ注射された。DNA注射後10日目に、PBSで106〜107pfuに希釈され、外側尾静脈へ静脈(i.v.)注射された組換えワクシニアウイルスにより、マウスが追加刺激された。又は、新鮮に単離された脾臓細胞が、37℃で0.1%BSAが補足されたRPMI中で、90分間、mel3ワクシニア、全長チロシナーゼワクシニア、及び全長NY-ESO-1ワクシニアにより別々に感染された(感染多重度5)。細胞が3回洗浄され、6×107個/mlの濃度で無菌PBSに再懸濁させられ、合わせられ、外側尾静脈へ注射された。マウスは6×106個の脾臓細胞を受容した。
HHDマウスから新鮮に単離された脾臓細胞が、2時間、10-6Mの濃度の異なるペプチドと共にRPMI培地中で別々にインキュベートされた。次いで、インビボの異なる集団の同時の追跡を可能にするため、異なる濃度のカルボキシフロオレセインジアセテートスクシンイミジルエステル(carboxyfluorescein diacetate succinimidyl ester)(CFSE、Molecular Probes、Eugene、Oregon)により各細胞プールが標識された27(Hermans,I.F.、Yang,J.及びRonchese,F.の未発表の結果)。標識された細胞がプールされ、107個/匹で尾静脈へ注射された。5-(及び-6-)-(((4-クロロメチル)ベンゾイル)アミノ)テトラメチルローダミン(CellTracker Orange、Molecular Probes、Eugene、Oregon)により標識されたペプチドを含まない対照集団が、未パルス処理細胞と比較された、ペプチドでパルス処理された標的の死滅を査定するために共注射された。マウスは、異なるエピトープに対するCTL頻度を決定するため、蛍光色素標識標的の注射の時点で採血された。注射後5時間目に新鮮に単離されたPBMCのFACS分析を使用して、ペプチド/蛍光色素標識細胞の消失が追跡された。セルトラッカーオレンジ(Cell Tracker Orange)で標識された未パルス処理集団に対する相対死滅率が計算された。100−(100×(パルス処理%/未パルス処理%))。WinMDI 2.8及びセルクエスト(CellQuest) 3.3ソフトウェアが、facsデータを分析するために使用された。
5個のHLA-A2黒色腫エピトープ及び2個のHLA-A1黒色腫エピトープのストリングが、4個の別個のベクター;a)裸プラスミドDNA(mel3.DNA);b)ワクシニアウイルス(mel3-ワクシニア);c)修飾型ワクシニア・アンカラ・ウイルス(mel3.MVA);d)セムリキ森林熱ウイルス(mel3-SFV)へクローニングされた。ヒト及びマウスのクラスI分子により拘束されたCTL応答のモニタリングを保証するため、本発明者らは、H-2DbクラスI分子により拘束されたインフルエンザ核タンパク質(NP)由来の付加的なエピトープを導入した。アミノ末端NP366-374エピトープの提示は、近隣のアミノ酸残基の影響を受ける場合があることが以前に示されているため24、本発明者らは、ポリエピトープ構築物のカルボキシル末端にインフルエンザNP366-374エピトープを発現させることに決定した。ここで使用されたポリエピトープ構築物の配列は、表1に示されている。
最初の実験は、mel.3構築物に含まれる各エピトープのA2結合親和性を比較するために実施された。これらの実験の結果から、mel3ペプチドエピトープが、A2分子に対する広範囲の結合親和性を有することが証明された。異なる濃度におけるインフルエンザマトリックスエピトープ58-66の提示を阻害する能力(データ示さず)により定義されたように、Melan-A 26-35ペプチド類似体28が、最も高い結合親和性を有し、NY-ESO-1 157-165ペプチド及びチロシナーゼ1-9ペプチドは、有意に低い親和性を有していた。最適な隣接残基がクラスI拘束性エピトープの提示を保証するのに重要であることが、本発明者ら及び他の研究者らによって以前に証明されている24、29。mel.3ペプチドエピトープが適切なプロセシングを受けることを確証するため、そしてA2分子との結合に関する競合が、低親和性エピトープのCTL認識を減損させないことを査定するため、mel.3ワクシニアを標的細胞に感染させ、ポリエピトープmel.3カセットに含まれる7個のエピトープの各々が、特異的CTLへと同時に提示されることを証明した(図1)。本発明者らは、プロテアソーム依存性の分解が、MAGE3 A2エピトープ271-279の提示を減損させることを以前に示した30。従って、mel3ワクシニアによる標的細胞の感染が、MAGE3 271-279特異的CTLによる溶解に対してそれらを感作する能力を有していることが観察されたのは、驚くべきことであった。さらなる実験によって、小胞体常在性プロテアーゼによるmel.3構築物のプロセシングと一致して、mel3構築物に含まれるMAGE3 271-279エピトープのプロセシングが、全長MAGE3タンパク質に含まれるもののプロセシングとは異なり、ラクタシステイン(lactacystein)抵抗性であり、非TAP依存性であることが証明された(データ示さず)。
mel3ワクシニア感染細胞によるNP366-374エピトープの効率的な提示のため、本発明者らは、強力なNP366-374特異的CTL応答を誘導する異なる予防接種戦略の能力をC57/B6マウスにおいて査定することにした(図2)。NP特異的CTL応答のエクスビボモニタリングを、Db/インフルエンザNP366-374四量体を使用してPBLにおいて実施した。本発明者らは、同種初回刺激追加刺激予防接種プロトコールを異種のものと比較し(図2)、DNA又はMVAによる初回刺激によるCTL誘導の動力学を分析した(データ示さず)。これらの実験の結果より、異種予防接種戦略が、長期持続性のワクチン駆動CTL応答を誘導する能力を有し、その頻度が、同一抗原送達系の反復注射に基づく戦略により得られた頻度より最大100倍大きいことが確認された(図2)。
インビボのA2拘束性CTL応答を初回刺激するmel.3ポリエピトープ構築物の能力を試験するため、A2トランスジェニックマウスを、DNA-mel.3により初回刺激し、MVA-mel.3、ワクシニアmel.3、又はSFV-mel3により追加刺激した。
DNA.mel3によるHHDマウスの初回刺激によって、予防接種を受けた全てのマウスにおいて、Melan-A特異的CTLが、エクスビボ四量体染色により検出可能な頻度にまで増幅された(データ示さず)。対照的に、NY-ESO-1及びチロシナーゼに特異的なCTLの増幅は、免疫されたマウスのうちの少ない割合においてのみ検出可能であり、チロシナーゼ1-9及びMage3 271-279に対する応答は、血液四量体染色において検出されなかった(データ示さず)。
melan-A特異的CTLを誘導する能力に対する既存のflu記憶CTL応答の阻害効果(図3)が証明されたため、本発明者らは、DNA初回刺激後の免疫応答を支配している多数のmelan-A特異的CTLが、ウイルス追加刺激中のNY-ESO-1及びチロシナーゼに特異的なCTLの増幅に干渉する能力を有するか否かを研究しようとした。
エクスビボアッセイ法における抗原特異的CTL応答のモニタリングの最近の進歩によって、異なる予防接種プロトコールの比較の性能は急速に改良されつつあるため、ワクチン開発には非常に勢いがある。腫瘍及びウイルスの抗原エスケープバリアントの生成を最小限に抑えるためには、ドミナントエピトープ及びサブドミナントエピトープの両方を含むいくつかのエピトープに特異的なワクチン駆動CTLの増幅を保証することが重要である。いくつかの論文が、ウイルス抗原34-36及び組織適合性抗原33、37、38に特異的なCTLの免疫優性を担う原因を検討している。しかしながら、どのようにすれば、最適なワクチン戦略によって、ドミナントエピトープ及びサブドミナントエピトープに特異的なCTLを同時に増幅し得るかは、未だ確立されていない。
本発明者らは、初回刺激−追加刺激計画で予防接種を受けたA2トランスジェニックマウスにおいて増幅されたA2拘束性CTLの頻度を直接モニタリングするための新規の四量体に基づく技術を開発した。マウスCD8のA2分子に対する結合親和性を増加させるため、マウスH-2Kbα3ドメインを含有しているA2分子(A2/Kb分子)を設計し、四量体A2Kb分子が、四量体A2分子と比較して、マウスA2拘束性CTLに対する増加した染色効率を有し、A2トランスジェニックマウスの大きな割合においてA2拘束性CTLを同定し得ることを証明した。
いくつかの臨床試験が、現在、ポリエピトープ構築物を用いた異種初回刺激追加刺激予防接種プロトコールを使用しているため、これらの結果は重要である。本発明者らは、多数のワクチン特異的CTLを誘発する異種初回刺激追加刺激プロトコールの能力を確認し、異種初回刺激−追加刺激プロトコールにおいて、イムノドミナントCTL応答の頻度が、比較的ドミナントでない決定基に特異的なCTL応答の頻度を超えて有意に増幅されることを証明している。多数のメカニズムがワクチンに応答するCTLレパートリーを狭小化する原因となり得るが、本発明者らの結果は、抗原提示細胞(APC)に関するT細胞の競合に基づく免疫優性のモデルと一致している32、33、38。未感作マウスへのvac.mel3の注射によって、リンパ球増加症が誘導され、CD8+の頻度が0.5%から30%へとシフトすることは、ワクシニアウイルスに対する強力なCTL応答を示しており、注目に値する。血中で誘導されたCD8+T細胞のうち、50%もが、Melan-A 26-35に特異的であり(データ示さず)、これは、melan-A 26-35が、ウイルスの構造、転写、及び複製を担う200個を超えるワクシニアタンパク質の中で最もイムノドミナントなウイルス発現エピトープのうちの一つであることを証明している。この観察は、Melan-Aエピトープ26-35をコードするウイルスに基づくワクチンの設計にとって重要な臨床的意義を有する。本発明者らは、SFVが、MVAと組み合わせて初回刺激ベクターとして使用されてもよいし(図2)、DNAと組み合わせて追加刺激に使用されてもよい(図4C)ことを証明したため、初回刺激−追加刺激プロトコールにおける組換えSFVの使用は、極めて魅力的である。これらの結果は、マカークにおける、組換えSFVによる初回刺激及びMVAによる追加刺激により誘導されるサル免疫不全ウイルス特異的免疫応答の増強を証明している最近発表された報告11を支持する。
