【解決手段】GABABレセプターおよびそれをコードするDNAと有意な相同性を有する精製GABABレセプターおよびレセプタータンパク質を提供することにより解決する。

Description
分子生物学およびタンパク質精製法の進歩、かつ薬理学的実験のために精製GABABレセプターを得る明確な要求にもかかわらず、GABABレセプターはこれまでには均質物にまでクローン化または精製されていない。その部分的精製に関する先の報告は(Nakayasu et al., J. Biol. Chem., 268, 8658−8664, 1993)、現在我々が小さすぎることを知っている80Kdaタンパク質に関して不正確であると考えられる。GABABレセプターをクローンできるようにするために、我々は多くのGABABレセプター特異的リガンドを開発している。高度に特異的な放射活性で標識したこのような高度に選択的なGABABレセプターリガンドの一つを使用した発現クローニングにより、我々は本発明によりラットおよびヒト源から異なるGABABレセプターをクローン化し、それらを配列決定し、哺乳類細胞培養において対応する組換えレセプターを発現させた。
本発明は、精製GABABレセプターおよびGABABレセプタータンパク質ならびにこのようなタンパク質をコードする核酸を提供する。本発明のタンパク質および核酸は、本明細書に記載のようにGABABレセプターおよびそれをコードするDNAと有意な相同性を有する。特に、ラットから単離されたGABABの明確な変異体である、GABABR1aおよびGABABR1bと名付けられた二つのGABABレセプタータンパク質を提供する。それぞれのcDNAおよび由来するアミノ酸配列は、配列番号1、2および5、6にそれぞれ記載する。更に、ヒト源から単離されたGABABR1a/b(部分的レセプタークローンを示す)およびGABABR1b(完全な長さのレセプタークローンを示す)と名付けた二つのヒトGABABを提供する。それぞれのcDNAおよび由来するアミノ酸配列は、配列番号3、4および7、8にそれぞれ記載する。
図1aは、COS1細胞での組換えGABABR1aの発現を記載する。ラット皮質膜(レーン1)およびGABABR1aラットcDNAでトランスフェクトしたCOS1細胞(レーン2および3)由来の膜を光親和性リガンド[125I]CGP 71872で標識する。レーン当たり25μgタンパク質を充填した6%SDSゲルのオートラジオグラフィ−を示す。レーン1および2:0.6nM[125I]CGP 71872の特異的結合。レーン3:0.6nM[125I]CGP 71872での特定の結合が、1μMの非標識CGP 54626A(GABABレセプター特異的アンタゴニスト)と競合する、対照実験。天然および組換えGABABレセプターの見かけの分子量は、SDS−PAGE標準(BioRad)と比較して、ゲル移動性で概算した。図1bは更にGABABR1bラットーcDNAでトランスフェクトしたCOS1細胞の結果を示す(レーン3)。
本発明は精製GABABレセプターおよびGABABレセプタータンパク質、それをコードする核酸およびそれらの種々の適用に関する。本発明の前には、GABABレセプターは精製形で入手可能ではなく、粗膜調製物のみであった。本発明は、最初に、組換えDNA法により、実質的に純粋な形で、種々の相互に関連するGABABレセプターの製造を可能にした。一般に、グリコシル化形のこのようなタンパク質は、100から130kDaの間の観察される分子量を有し、一方非グリコシル化形はそれぞれ90から110kDaの間の観察される分子量を有することが予期される。
実施例1
リガンドCGP 64213の合成
COS細胞内で発現したGABABレセプターの可視化に使用する放射性リガンド[125I]CGP 64213は、スキーム1に従って、以下の試薬および条件を使用して合成する:
(1)NaH、THF、室温、3時間;5−ブロモバレロニトリル、室温、16時間;(2)ラネイニッケル、EtOH中4%NH3、45℃、16時間;(3)N−エトキシ−カルボニルフタルイミド、Na2CO3、H2O、CH2Cl2、室温、5時間;(4)Me3SiCl、EtOH、CH2Cl2(1:9)、室温、17時間;(5)Me3SiCl、Et3N、THF、室温、17時間;(R)−エピクロロヒドリン、10mol% ZnCl2 THF、80℃、17時間;HOAC、MeOH、室温、17時間(6)i−Pr2EtN、EtOH、80℃、7日;(7)LiOH、EtOH、H2O(1:1)、100℃、17時間;MeOH、H3PO4;(8)濃HCl、100℃、17時間;(9)i−Pr2EtN、DMF、室温、72時間;(10)Na125I、リン酸緩衝液pH7.4、H2O2、cat.ラクトペルオキシダーゼ、30分、RP−HPLC。
非標識CGP 64213をわずかに異なる方法で製造する:3−(4−ヒドロキシ−5−ヨードフェニル)プロピオン酸13を、3−(4−ヒドロキシ−フェニル)プロピオン酸から、Runeberg, J., Acta Chem. Scand. (1958), 12, 188-91に従って製造する。N−ヒドロキシ−スクシンイミジル−3−(4−ヒドロキシ−5−ヨードフェニル)プロピオネート14(沸点:191−4℃)をスキーム2に従って製造し、収率は73%である。CGP 57604A(スキーム1)と14の、スキーム1の反応9のように行う室温で72時間のDMF中のヒューニッヒ塩基を使用した縮合により、非放射標識CGP 64213(沸点:170−5℃、アセトンから結晶化)を、53%の収率で得る。
新しい物質で2000Ci/mmolの特異的放射活性の[125I]CGP 64213とのインキュベーションを、0.2mlクレブス−ヘンゼライト−トリス緩衝液、pH7.4中で、20℃で90分、基質としての50μg皮質膜タンパク質と共に行う。インキュベーションをGF/Bワットマングラスファイバー濾紙での濾過により終結させる。非特異的結合を、10−6M CGP 54626Aにより定義し、それは2nM濃度で全結合の5%である。増加した濃度の[125I]CGP 64213および非直線最小2乗法フィッティングでの飽和実験により、2.66nMの解離定数KDを測定する。0.1nM[125I]CGP 64213の濃度での阻害実験で、L−バクロフェンは442nMの阻害定数Kiを示し、2.5nMのKiのアンタゴニストCGP 54626Aは、他のGABABレセプターアンタゴニスト放射性リガンドで得られたKiと良好に一致する。GABAA、ベンゾジアゼピン、カイネート、AMPA、NMDAレセプターのアッセイで、ムスカリンコリン作動性、α1−およびα2−アドレナリン作動性、β−アドレナリン作動性、5HT1、5HT2、5HT3、ヒスタミン1、ヒスタミン2、アデノシン1μ−オピエートおよびサブスタンスPレセプターでのストリキニーネ非依存的結合部位で、非標識CGP 64213は、1μMで不活性である。本化合物は、従って、GABABレセプターに選択的である。[125I]CGP 64213の0.1nMの濃度で、結合および解離定数を測定する。結合の半減期は20℃で20分、および解離の半減期は40分である。解離の半減期は、温度を4℃に下げると4時間に延長する。この[125I]CGP 64213の遅い解離速度および高い特異的放射活性が、GABABレセプターの発現系としてのCOS細胞で結合しているレセプターのオートラジオグラフィー実験を可能にする。
光親和性リガンドの製造
ラット皮質膜由来のタグGABABレセプターおよびCOS細胞内で発現した組換えGABABレセプターを使用した光親和性リガンド[125I]CGP 71872をスキーム3に従って製造する:商品として入手可能なN−ヒドロキシ−スクシンイミジル−4−アジド−サリチレート15をCGP 57604Aと縮合し、中間体16を得、それを、クロラミンTを使用してヨウ化ナトリウム125アイソトープでヨウ素化し、5−ヨード誘導体[125I]CGP 71872および3−ヨード−誘導体[125I]CGP 72565の約1:1の混合物を得る。それらを、Vydac 218TP54カラムの逆相HPLC(保持時間:それぞれ16.4分および17.4分)で分離する。試薬および条件は下記の通りである:
(1)CGP 57604A(スキーム1)、i−Pr2EtN、DMF,室温、70時間;(2)Na125I、クロラミンT、0.01N NaOH、室温、1時間;RP−HPLC。
リガンドの結合アッセイおよび特徴付け
[125I]CGP 64213アッセイで記載したようにラット皮質膜を基質として使用する。新鮮な物質で特異的放射活性2000Ci/mmolである[125I]CGP 71872とのインキュベーションを、0.2mlクレブス−ヘンゼライト緩衝液、pH7.4中、20℃で90分、基質としての50μg膜タンパク質と共に行う。インキュベーションをGF/Cワットマングラスファイバー濾紙での濾過により停止させる。非特異的結合を10−6M CGP 54626Aで測定し、2nMの[125I]CGP 71872の濃度で、全結合の5%である。増加した濃度の[125I]CGP 71872および非直線最小2乗法フィッティングでの飽和実験により、3.1nMの解離定数KDと計算される。L−バクロフェンは阻害実験で、340nMのKiおよびアンタゴニストCGP 54626Aは3.1nMのKiを示す。非標識CGP 64213は、[125I]CGP 64213で記載したのと同じレセプターアッセイで1μMの濃度で不活性であり、従って、GABABレセプターにまた特異的であることが判明する。2nMの濃度および20℃で、結合の半減期は5分、解離の半減期は10分である。8℃での解離時間は長い。わずか25%の放射性リガンドが120分後に解離する。
それぞれGABABR1aおよびGABABR1bラット−cDNAで一過性に形質転換したラット大脳皮質およびCOS1細胞由来の膜をクレブス−ヘンゼライト−トリス緩衝液、pH7.3に、4mgタンパク質/mlの濃度で懸濁し、暗中、0.6nM[125I]CGP 71872と1時間室温でインキュベーションする。インキュベーションを20000×g、10分、4℃の遠心により停止させる。この段階は、遊離非結合光親和性標識を除去する。これらの条件下で、使用した全放射活性の約50%がレセプターに結合した。ペレットをポリエチレンバイアルに4mgタンパク質/mlの濃度で再懸濁し、UV光(365nm)で3分(24W)照射する。懸濁液を20000×gで10分遠心し、緩衝液中に8mg/mlタンパク質の濃度で再懸濁する。過剰の非標識GABABレセプターアンタゴニスト(10−6M CGP 54626A)存在下で標識を行った時、放射活性は膜に結合しない。標識膜は−80℃で貯蔵できる。結果は図1aおよび1bに示す。

a試薬および条件:(1)NaH、THF、室温、3時間;3,4−デヒドロブロモ−メチルシクロヘキサン、還流、24時間;(2)Me3SiCl、EtOH、CH2Cl2(1:9)、室温、24時間;(3)Me3SiCl、Et3N、THF、室温、24時間;(R)−エピクロロヒドリン、10mol% ZnCl2 THF、80℃、17時間;HOAc、MeOH、室温、17時間;(4)i−Pr2EtN、EtOH、80℃、4日;(5)LiOH、EtOH、H2O、100℃、17時間;HCl、MeOH、室温、1時間;(6)3H2、5%Pd/C、HCl、MeOH、pH=1、室温、15分、分取TLC。
インビトロ電気生理学的測定による、CGP 64213およびCGP 71872のGABABレセプターアンタゴニストとしての機能的活性の証明
実験は、雌ウィスターCOBラット(3−4週齢)または雄ラットTif:RAI f(SPF)から、標準法を使用して得た400μm厚の海馬薄片で行う。簡単に、ラットを断頭前に頸部脱臼させる。小脳を除いた脳をすぐに取りだし、氷冷人工脳脊髄液(ACSF)に入れる。海馬を注意深く単離し、組織チョッパー(Sorvall(登録商標))または振動スライサー(Campden)を使用して、横方向400μm厚切片を切断する。各切片のCA3領域を外科用メス切断で除去する。この方法を、CA3領域における癲癇様発作のために起こり得るネットワーク機能の変化を除くために行う。得られるCA3−切除切片を暖めた(32℃)インターフェースでナイロンメッシュに置き、ACSFおよび酸素に富む(95%O2、5%CO2)、加湿雰囲気を環流(1−2ml.分−1)する。標準環流培地は:NaCl、124;KCl、3;NaHCO3、26;NaH2PO4、1.25;CaCl2、2;MgSO4、1;D−グルコース、10を含み(mM)、95%O2、5%CO2であわ立たせる。アクソプローブまたはアクソクランプ−2増幅器(Axon Instruments, Foster City, CA, USA)をブリッジモードで使用し、4M NaCl−充電微小電極(2−5MΩ)を使用して、放線状層または多形細胞層から細胞外記録をする。細胞内記録は、2Mメチル硫酸カリウム微小電極(60−100MΩ)を使用して行う。デジタル化記録をオフライン分析のためにIBM−互換性PCのハードディスクに保存する。55μm直径の絶縁ニッケル−クロムワイヤーからなる2極性刺激電極を、放線状層または多形細胞層に置いた記録電極の近くに置き、CA1ニューロンの順方向モノシナプス性活性化を提供する(Davies et al., (1990) Journal of Physiology 424:513)。各実験において、刺激は固定強度で相同シナプス的に送達される方形波パルス(20−200μs;5−30V)を含む。全ての医薬は、環流媒体で投与する。データは平均±平均からの標準誤差(S.E.M.)で示し、統計的有意をスチューデンドのt検定を使用して評価する。n価は特定の実験を行った数であり、それぞれ異なるラットから取った異なる切片である。

(1982) Molecular cloning:A laboratory manual (Cold Spring Harbor Lab., Cold Spring Harbor, NY)に記載のように、オリゴ(dT)カラム(Boehringer Mannheim)を2回通過させて富化する。オリゴ(dT)刺激された二本鎖cDNAを、商品cDNA合成システム(Amersham)を使用して、5μgのポリA(+)RNAから合成する。キットに供給されている逆転写酵素は、RNAseH(-)SuperscriptII逆転写酵素(Gibco BRL)と置きかえる。cDNA溶液をtRNAで予備吸着したCentricon-100装置(Amicon)で濃縮し、最終量100μlとする。小cDNAをChromaspin-1000カラム(Clontech)を通して除去する。BstXIアダプター(Invitrogen)をT4 DNAリガーゼ(Boehringer Mannheim)を使用して添加し、cDNAをアガロースゲルでサイズ分画する。2kbより大きいサイズのcDNAを精製(Qiaex, Qiagen)し、発現ベクターpcDNAI(Invitrogen)のBstXI部位にライゲートする。ライゲーション混合物のアリコートで、エレクトロコンピテントMC1061/P3エシェリキア・コリ細胞を形質転換(BioRad Gene Pulser II)する。ライブラリーの複雑さを2×106の個々のクローンで概算する。48クローンの分析から推測される平均挿入サイズは、2.9kb(2.0kbから6.6kbのサイズの範囲)である。
COS細胞のcDNAでのトランスフェクション
COS1細胞をアメリカン・タイプ・カルチャー・コレクション(ATCC)から得、10%ウシ胎児血清(FCS)および15μg/mlゲンタマイシン(Gibco BRL)を添加したダルベッコ修飾イーグル培地(DMEM)で、5%CO2の加湿雰囲気下で生育させる。
リガンド結合アッセイによるGABABレセプタークローンの同定
DNAのプール(各2000の個々のコロニー)を、COS1細胞の一過性形質転換後に、ヨウ素化CGP 64213(特異的活性2000Ci/mmol)での放射性リガンド結合アッセイを使用して、GABABレセプター発現に関して分析する。
クローン化GABABレセプターのアッセイ
クローン化GABABレセプターを含む膜を単離するために、GABABレセプター−発現COS細胞を含む培養皿を2回クレブス−ヘンゼライト−トリス緩衝液で洗浄する。その後、細胞を皿から掻き出し、ガラス−ガラスホモジナイザーで均質化し、30分、4℃で40,000gで遠心する。均質化および遠心段階をもう一回繰り返す。ペレットを緩衝液に再懸濁し、更に分析するまで液体窒素中に貯蔵する。
関連遺伝子をクローンするためのGABABR1aレセプターの使用
単離したラットGABABR1a−レセプター(配列番号1)は、更なるGABAB−レセプター遺伝子ファミリーのcDNA、遺伝子およびタンパク質の同定および単離のためのプローブとして有用である。例えばヒトのような他の種におけるGABAB−レセプター遺伝子ファミリーのcDNA、遺伝子およびタンパク質の同定および単離にもまた有用である。
HEK293細胞で安定に発現されるGABABレセプターは、アデニレートシクラーゼに陰性に結合する
GABABレセプターは、アデニレートシクラーゼを阻害し、ホスホリパーゼA2を刺激し、K+−チャンネルを活性化し、ボルト数依存的Ca2+−チャンネルを不活性化し、イノシトールリン脂質加水分解を調節すると記載されている。GABABR1aおよび−bは細胞内と予期される全てのドメインで同一の配列を有するため、同じエフェクターシステムに結合すると予期される。ラット皮質切片調製物を使用して、L−バクロフェンはホルスコリン−刺激cAMP蓄積を約40%まで減少させることが示されている。HEK293細胞で安定に発現されているGABABR1aがホルスコリン−刺激cAMP蓄積を減少させる能力を分析する(図5)。我々は、最大効果を製造するホルスコリンおよびL−バクロフェンの濃度を選択した。ホルスコリンはHEL293細胞内でcAMPレベルを基底レベルの10倍以上まで刺激する。組換え発現GABABレセプターの300μM L−バクロフェンとの共投与による刺激は、ホルスコリン刺激cAMP蓄積を約30%まで減少させる。この阻害は、GABABレセプターアンタゴニストであるCGP 54626Aにより拮抗される。アデニレートシクラーゼ活性のGABABR1aによる調節は、百日咳毒素に感受性であり、Goを欠失するKEK293細胞で、GABABR1aがGiに結合することを示す。コントロールとして、L−バクロフェンはホルスコリン−刺激cAMP形成を非トランスフェクトHEK293細胞で阻害しない(図5)。
ラット由来のGABABレセプタークローンGABABR1aは、ブタペスト条約に基づいて、ドイツ、デー−38124ブラウンシュヴァイク、マシェローダーヴェーク1ベー番のドイッチェ・サムルング・フォン・ミクロオルファニズメン・ウント・ゼルクルツレン・ゲゼルシャフト・ミット・ベシュレンクテル・ハフツング(DSMZ)に、1996年5月17日に、受託番号DSM10689で寄託した。
ラット由来のGABABレセプタークローンGABABR1bおよびヒト源由来のGABABR1bは、ブタペスト条約に基づいて、ドイツ、デー−38124ブラウンシュヴァイク、マシェローダーヴェーク1ベー番のドイッチェ・サムルング・フォン・ミクロオルファニズメン・ウント・ゼルクルツレン・ゲゼルシャフト・ミット・ベシュレンクテル・ハフツング(DSMZ)に、1997年2月21日に、それぞれ受託番号DSM11422および11421でで寄託した。
Claims (21)
- (a)配列番号1のヌクレオチド182位から3061位;配列番号3のヌクレオチド1位から2379位;配列番号5のヌクレオチド228位から2759位;または配列番号7のヌクレオチド169位から2700位に記載の核酸配列を含む核酸配列のいずれかにより;
(b)受託番号DSM10689、DMS11421およびDSM11422でDSMZに寄託されたクローンから成る群から選択される核酸クローンにより;または
(c)配列番号1、配列番号3、配列番号5または配列番号7のポリヌクレオチドと中位のストリンジェンシー条件下(ここで、中位のストリンジェンシー条件は、60−62℃で、1×SSC、0.1%SDSでの洗浄段階で終わる)でハイブリダイズする核酸配列のいずれかにより;
コードされる、単離されたGABABレセプターまたはレセプタータンパク質。 - ヒトGABABレセプターまたはレセプタータンパク質である、請求項1記載のGABABレセプターまたはレセプタータンパク質。
- アミノ酸配列が配列番号2、配列番号4、配列番号6または配列番号8と30%を超える相同性を有するアミノ酸配列を含む、請求項1記載のGABABレセプターまたはレセプタータンパク質。
- 配列番号2、配列番号4、配列番号6または配列番号8に記載のアミノ酸配列を含む、請求項1記載のGABABレセプターまたはレセプタータンパク質。
- 配列番号1、配列番号3、配列番号5または配列番号7に記載のアミノ酸配列を含むGABABレセプターまたはレセプタータンパク質をコードする、単離核酸。
- 核酸配列が配列番号1のヌクレオチド182位から3061位;配列番号3のヌクレオチド1位から2379位;配列番号5のヌクレオチド228位から2759位;または配列番号7のヌクレオチド169位から2700位に記載のヌクレオチド配列を含む、請求項6記載の単離核酸。
- 核酸配列が受託番号DSM10689、DMS11421およびDSM11422でDSMZに寄託されたクローンから成る群から選択される核酸クローンによりコードされる、請求項6記載の単離核酸。
- 配列番号1、配列番号3、配列番号5または配列番号7を含む核酸配列のいずれか一つと中位のストリンジェンシーまたは高ストリンジェンシー条件下(ここで、中位のストリンジェンシー条件は、60−62℃で、1×SSC、0.1%SDSでの洗浄で終わり、高ストリンジェンシー条件は65−68℃で、0.2−1×SSC、0.1%SDSでの洗浄で終わる)でハイブリダイズできる、核酸プローブ(ただし、EMBL受託番号X90542の核酸プローブを除く)。
- 核酸プローブが請求項9に記載の核酸配列に関してアンチセンス核酸配列を有する、請求項9記載の核酸プローブ。
- 配列番号2、配列番号4、配列番号6または配列番号8に記載のレセプターまたはレセプタータンパク質に対して発生させた、抗体。
- モノクローナル、ポリクローナルまたはキメラ抗体である、請求項11記載の抗体。
- Fab、F(ab')2、FvまたはScFvフラグメントである、請求項11記載の抗体。
- 制御配列に作動可能に結合した請求項6記載の核酸から成るコード配列を含む、複製可能核酸ベクター。
- 請求項14記載のベクターで形質転換した宿主細胞。
- 請求項1記載のGABABレセプターまたはレセプタータンパク質を含む、トランスジェニック非ヒト動物細胞(ただし、該トランスジェニック非ヒト動物細胞は請求項1記載のGABABレセプターまたはレセプタータンパク質を内因性に発現しない)。
- (a)GABABレセプターまたはレセプタータンパク質をコードする可能性のあるcDNAまたはゲノムDNA分子をコードする発現ライブラリーの製造;
(b)請求項10に記載の核酸プローブとのハイブリダイズによる発現ライブラリーのスクリーニング;および
(c)プローブとハイブリダイズする核酸分子の同定:

の段階を含む、GABABレセプター活性の調節剤の可能性のある化合物または化合物の混合物のスクリーニング法。 - 通常GABABレセプターまたはレセプタータンパク質をコードする遺伝子と結合している制御配列と作動可能に結合した試験遺伝子を含む発現系の提供;
