Description
血清中では、IgAは主に単量体の形で主に存在し、低いパーセンテージで多量体IgA (pIgA)が存在する。
粘膜分泌物(唾液、涙液、初乳、胃腸液、鼻−気管支分泌物及び尿)には、IgAはJ鎖とよばれるポリペプチドで連結された二量体として産生される。二量体IgAは、膜結合性多量体Ig受容体(pIgR)に結合し、得られる複合体は、粘膜上皮の基底外側から先端/管腔(apical/luminal)側に輸送される。この輸送の間、結合したIgAは、pIgRからのタンパク質分解性切断により解放される。しかし、pIgRの一部分、分泌成分は二量体IgAと結合したままであり、全体で分泌型IgA (SIgA)を形成する。
上記の一価の抗体断片の抗炎症性特性は、FcαRI発現骨髄細胞を伴う病理的炎症性反応のダウンレギュレーションに起因する。
例えば、Fab断片は、FcαRI受容体のEC2ドメインに指向された抗体から、酵素消化の通常の方法により得ることができる。上記の抗体は、通常のハイブリドーマ技術により得られたマウスモノクローナル抗体であり得る。有利には、該抗体はキメラ抗体、ヒト化抗体又は完全ヒト抗体でもあり得る。キメラ抗体は、上記のモノクローナル抗体から、定常領域ドメインをヒトドメインで置き換えることにより得ることができる。ヒト化抗体は、上記のモノクローナル抗体のCDRを、それ自体で公知のCDRグラフティング技術を用いて、ヒト抗体のフレームワーク領域(framework region:FR)に組み込むことにより得ることができる。完全ヒトモノクローナル抗体は、例えばPCT WO 02/064634に記載されるように、免疫にされるマウスがヒト免疫グロブリンレパトアを有するトランスジェニックマウスであること以外は、通常のマウスモノクローナル抗体と同様にして得ることができる。
例えば、該断片は、筋肉内、皮内、静脈内、腹腔内、皮下又は局部注射を含む非経口経路により投与できる。
本発明の一価の抗体断片が気道の粘膜表面への送達に適する形態であれば、呼吸器官での局部投与も用いることができる。例えば、これらを、エアロソル形態又は喘息の治療で現在用いられているものに類似のネブライザ装置により患者に送達するための液体製剤に懸濁できる。
血液単球のIgG-媒介性食作用活性の調節におけるFcαRIの役割を調べた。
ヒト末梢血単核細胞を、健康なボランティアから、フィコール−ハイパック密度勾配遠心法により単離した。濃縮した(70〜80%)単球の集合を、MONTEIROら(1990, 上記)に記載されるようなプラスチックへの接着により得た。
結果を、図1に示す。
FcαRIの阻害機能を、野生型ヒトFcαRIでトランスフェクションした、高親和性受容体IgE (FcεRI)を構成的に発現するラット肥満細胞系統RBL-2H3の脱顆粒応答を試験することによりさらに研究した。
細胞トランスフェクション:
RBL-2H3細胞のトランスフェクションは、LAUNAYら(1999, 上記)に記載されるようにして行った。野生型ヒトFcαRI構築物を、XbaI-BamHI制限部位の間にネオマイシン耐性遺伝子を含有するpSRαNEOベクターにクローニングし、配列はDNA配列決定により制御した。ROAら(J. Immunol. 159: 2815〜2823, 1997)に記載されるようにして維持されるRBL-2H3細胞を、Easyjet+装置(Eurogenetec, Seraing, Belgium)を用いて250 V及び1500μFaでのエレクトロポレーションにより、15μgのDNAでトランスフェクションした。
細胞に含有される顆粒状メディエイタのエキソサイトーシスを、(ROAら, 1997, 上記)に記載されるようにして、異なる試験化合物での感作に対する、FcαRIトランスフェクションされた細胞、又はコントロールとして用いられる非トランスフェクション細胞によるβ−ヘキソサミニダーゼの放出を測定することにより測定した。
細胞を、96ウェルプレート(Becton Dickinson)に、5×104細胞/ウェルで入れた。細胞を、各試薬について、以下に記載されるように、異なる試験化合物で感作した。細胞をあらかじめ温めておいたTyrodeバッファー(135 mM NaCl、5 mM KCl、5.6 mMグルコース、10 mM HEPES、pH 7.3、1.8 mM CaCl2、1 mM MgCl2及び5% BSA)中で洗浄し、0.1μg/ml DNP-HAS (Sigma)で脱顆粒を引き起こした。合計のβ−ヘキソサミニダーゼ放出を、自然発生的な放出を引いた後に合計含量のパーセンテージとして算出した。
ヒトFcαRIトランスフェクタント(クローン15.4)及び非トランスフェクション(NT) RBL細胞を、IgE抗DNP (1:200)又はIgE抗DNPと10μg/ml 無関係なFab 320コントロール又は抗FcαRI Fab A77で、1時間、37℃で感作した。細胞を洗浄し、DNP-HASで脱顆粒を引き起こし、β−ヘキソサミニダーゼ放出を決定した。
結果を図2aに示す。
結果は、IgE-感作トランスフェクタント(クローン15.4)の抗原刺激が、強い脱顆粒応答を誘導したことを示す。抗FcαRI Fab A77とのプレインキュベーションは、無関係のFab 320に比べて、FcαRIにより開始された脱顆粒を著しく阻害した(74%)。他の2つのトランスフェクタントを用いて同様の結果が得られた(示さず)が、非トランスフェクション細胞(NT)を用いては得られなかった。A77 Fabの阻害効果は、より長い期間(2〜12時間)プレインキュベーションしたときにより強かった(示さず)。
注目すべきことに、抗FcαRI Fabは、IgE結合を修飾することに失敗した(示さず)。ゲルろ過により精製した抗FcαRI Fabは、同様の阻害作用を有しており、観察された効果での凝集物の役割を除外する(示さず)。
ヒトFcαRIトランスフェクタント(クローン15.4)を、異なる濃度の抗FcαRI Fab A77又は無関係のFab 320の存在下に、37℃で1時間、IgEで感作した。細胞を洗浄し、脱顆粒をDNP-HASで引き起こし、β−ヘキソサミニダーゼ放出を決定した。
結果を図2bに示す。
結果は、抗FcαRI Fabによる阻害が濃度依存的であり、1〜10μg/mlの間で最大であったことを示す。
ヒトFcαRIトランスフェクタント(クローン15.4)を、異なる抗FcαRI mAb:A3 (EC1とEC2との間の結合部位を認識する;EC2内の結合部位を認識するA59、A62、A77)又は無関係のFab 320からのFab断片10μg/mlの存在下に、37℃で1時間、IgEで感作した。
細胞を洗浄し、脱顆粒をDNP-HASを用いて引き起こし、β−ヘキソサミニダーゼ放出を決定した。
結果を図2cに示す。
試験した4つの抗FcαRI Fabのうち3つが、FcεRI誘発脱顆粒を>50%で阻害した。4つ目の抗FcαRI Fab (A3)は、脱顆粒の阻害に失敗したが、3つの対照と同様に、FcαRIでトランスフェクションされた細胞に容易に結合した(示さず)。
この目的のために、F(ab')2を、抗FcαRI mAb (A77)又は無関係な抗体320から、SILVAINら(J. Immunol. 155: 1606〜1618, 1995)に記載されるようにして、基質に対する比(w/w)が1/50の酵素を用いて、0.1 M酢酸バッファー、pH.4.4中で、37℃で8時間ペプシン消化することにより作製した。完全消化及び純度は、SDS-PAGEにより制御した。
細胞を洗浄し、脱顆粒をDNP-HASを用いて引き起こし、β−ヘキソサミニダーゼ放出を決定した。
結果を、図2dに示す。
結果は、一価の抗FcαRI Fabが、二価のF(ab')2断片よりもより強い阻害効果を有したことを示す。
ヒトFcαRIトランスフェクタント(クローン15.4)を、IgE、又はA77からの10μg/ml Fab若しくはF(ab')2断片、又は320からの無関係のFab若しくはF(ab')2断片で、37℃で1時間感作した。
次いで、細胞をウサギ抗マウスIgG (RAM、40μg/ml)のF(ab')2断片で刺激した(LAUNAYら, J. Leukoc. Biol. 63: 636〜642, 1998)。
細胞を洗浄し、β−ヘキソサミニダーゼ放出を決定した。
結果を図2eに示す。
抗FcαRI F(ab')2をウサギ抗マウスIg (RAM) F(ab')2を用いて架橋した後の、FcαRIの高度に多価の凝集が、脱顆粒をもたらしたことを示す。抗FcαRI FabとRAM F(ab')2とを用いてのより少ない程度に多価の凝集は、より少ない脱顆粒をもたらした。抗FcαRI Fab、F(ab')2又はRAM F(ab')2のみでは脱顆粒は観察されなかった(示さず)。
FcαRI RBL-2H3トランスフェクタント(クローン15.4)に対する生理学的リガンドIgAの影響を、上記の実施例2に記載のようにして、脱顆粒応答を試験することにより試験した。
プロテアーゼを含む多数のメディエイタを含有するIgAが炎症部位で生物学的活性を示すので、FcαRIがトリプシンに耐性であると仮定して、IgA媒介阻害機能に対する細胞のトリプシン処理の影響を試験した(MONTEIROら, 1990, 上記)。
ヒトFcαRIトランスフェクタントを、DMEM中の1 mg/ml トリプシン-TCPK (Sigma)を用いるか又は用いずに、37℃で30分間予備処理し、次いでIgEのみ、又はIgEと0.2 mg/ml血清IgG若しくは精製血清IgA (バッチ番号39328及び02828, ICN Biomedicals Inc, Aurora, Ohio)で一晩感作した。
細胞を洗浄し、脱顆粒をDNP-HASを用いて引き起こし、β−ヘキソサミニダーゼ放出を決定した。
結果を、図3aに示す。
バーの上の数字は、阻害の平均パーセンテージを示す。
結果は、血清IgAとのインキュベーションが、IgE-依存性脱顆粒を著しく阻害した(43%)が、IgGとのインキュベーションは阻害しなかったことを示す。血清IgAの阻害効果は、トリプシン処理した細胞において著しく増大した(〜50%増大)が、IgGの阻害効果は増大せず、IgE媒介脱顆粒応答は影響されなかった。同様の増大が、精製骨髄腫IgAを用いて観察された(示さず)。
ヒトFcαRIトランスフェクタントを、DMEM中の1 mg/mlトリプシン-TCPK (Sigma)で、37℃で30分間予備処理し、IgEのみ、又はIgEと種々の濃度の2つのバッチの精製血清IgA (バッチ番号39328及び番号02828, ICN Biomedicals Inc, Aurora, Ohio)、分泌IgA (SIgA, バッチ番号42K3780, Sigma Aldrich, St-Louis, Missouri)、又はヒトIgGとで感作した。
細胞を洗浄し、脱顆粒をDNP-HASを用いて引き起こし、β−ヘキソサミニダーゼ放出を決定した。
結果を、図3bに示す。
結果は、市販の血清IgAの2つの異なるバッチが、用量依存様式で脱顆粒を阻害し、最大の阻害(66%)が0.5 mg/mlで得られたことを示す。初乳SIgAも、より少ない程度ではあるが、細胞活性化を阻害した。
FcαRIはIgA1及びIgA2の両方に結合するので、この2つのサブクラスの阻害能力を、分泌粘膜の種類に応じてIgA1及びIgA2の両方を種々の量で含有するSIgAの能力に対して比較した。
ヒトFcαRIトランスフェクタントを、DMEM中の1 mg/mlトリプシン-TCPK (Sigma)で、37℃で30分間処理し、IgEと0.2 mg/ml血清IgG、精製された骨髄腫IgA1及びIgA2又はSIgAとで一晩感作した。細胞を洗浄し、脱顆粒をDNP-HASを用いて引き起こし、β−ヘキソサミニダーゼ放出を決定した。
結果を図3cに示す。
結果は、すべての試験された調製物は、ヒトIgG (<5%)に比べて、著しい阻害(30〜40%)を発生したことを示す。FcαRI阻害応答は、IgA1及びIgA2の両方により誘導できる。
FcαRIは、単量体IgAよりも多量体IgAにより効率的に結合するので、二次架橋を有さないIgAの種々の分子形(HPLCにより分離)の阻害可能性を試験した。
ヒトFcαRIトランスフェクタントを、DMEM中の1 mg/mlトリプシン-TCPK (Sigma)で、37℃で30分間予備処理し、IgEと0.1 mg/ml血清IgG (IgG)、全血清IgA (IgA)、多量体血清IgA (pIgA)、二量体血清IgA (dIgA)又は単量体IgA (mIgA)とで一晩感作した。血清IgAは、HPLCによりサイズ分画した。
細胞を洗浄し、脱顆粒をDNP-HASを用いて引き起こし、β−ヘキソサミニダーゼ放出を決定した。
結果を、図3dに示す。
結果は、多量体血清IgAが、単量体血清IgAよりもより阻害的であることを示す。阻害能力は、IgA種のサイズとともに増加する。多量体IgAは、二量体IgA (38%)及び単量体IgA (20%)のどちらよりも効果的であった(60%)。同様のデータが、HPLCにより分離された血清IgAの別のバッチを用いて得られた(示さず)。
阻害シグナルについての構造的要件を探索するために、一連のFcαRI変異体及びキメラ構築物を用いた。
- FcαRI膜貫通ドメイン内の位置209の荷電アルギニンがロイシン(R209L)に置換されたFcαRIR209L;この変異は、FcαRIの、FcRγ鎖との結合を廃止する(LAUNAYら, 1999, 上記; MORTONら, J. Biol. Chem. 270: 29781〜29787, 1995)。
- R209L/γキメラ構築物は、FcαRIR209Lの細胞外及びR209L膜貫通ドメインの、ヒトFcRγ鎖の細胞質内尾部との融合に起因する。
すべての構築物は、実施例2に記載のようにして、pSRαNeoベクターにクローニングし、RBL-2H3細胞にトランスフェクションした。
野生型ヒトFcαRI (クローン15.5)、FcαRI-γ2 (クローン5.26)又はR209L/γキメラ(クローン9.4)構築物でトランスフェクションされた細胞を選択した。
脱顆粒応答は、実施例2の2)に記載されたようにして試験した。
結果を図4cに示す。
結果は、IgEのみで感作された全てのトランスフェクタントは、50%を超えるFcεRI-媒介脱顆粒を示したことを示す。抗FcαRI Fab A77処理は、R209L変異体でトランスフェクションされたRBL-2H3 (クローン5.26)において阻害的でなく、このことはFcαRIの細胞内尾部は、阻害シグナリングを担うモチーフを含有しないことを示唆した。対照的に、トランスフェクションされた細胞(クローン9.4)での抗FcαRI Fab A77のFcαRIR209L/γキメラ受容体への結合は、野生型受容体でトランスフェクションされた細胞(クローン15.5)で観察されたものと同様の程度で、脱顆粒に対する阻害効果を回復させた(それぞれ91%及び72%)。トランスフェクタントのそれぞれの種類について、少なくとも3つのさらなるクローンを用いて同様の結果を得た(示さず)。
FcRγ鎖はいずれの既知の阻害モチーフを有さないので、活性化(activatory)モチーフとして通常は知られるFcRγ ITAMを、これが阻害効果をも媒介できるかを知るために研究した。ヒトFcRγ鎖は、細胞活性化において役割を演じることが知られているITAMモチーフの一部分である2つのカルボキシ末端チロシン(FcαRIR209L/γキメラ受容体内のY268及びY278)を有する(17,24)。点突然変異(Y268F、Y278F及び二重Y268/278F)を、FcαRIR209L/γキメラのITAMモチーフに導入した。
A) チロシンリン酸化アッセイ
ITAM-媒介シグナリングはチロシンキナーゼの活性化を伴うので、FcαRI/γ複合体の単量体ターゲティングを、これがチロシンリン酸化を伴うかを知るために研究した。
示されたRBLトランスフェクタント(FcαRI-γ2、FcαRIR209L/γキメラ野生型、FcαRIR209L/γキメラ Y268F/Y278F)を、15分間、10μg/mlの抗FcαRI Fab A77、無関係のFab 320、40μg/mlのRAM F(ab')2、又は抗FcαRI A77 F(ab')2とRAM F(ab')2との組み合わせで刺激した。
刺激及び氷冷PBS中での2回の洗浄の後に、細胞を溶解バッファー(50 mM HEPES pH 7.4、1% Triton X-100、0.1% SDS、50 mM NaF、50 mM NaCl、1 mM Na3VO4、30 mM Na4P2O7、50 U/mlアプロチニン、10μg/mlロイペプチン)中に溶解し、核後(post-nuclear)上清を調製した。溶解物を、SDS-10% PAGEにより分離し、PVDFメンブレンに移した。4% BSAでブロッキング後、メンブレンを4G10 抗PY Ab (Upstate Biotechnology, Lake Placid, NY)と、室温で1時間インキュベートし、HRP結合ヤギ抗マウスIg (Southern Biotechnology Associates, Birmingham, AL)とインキュベートした。メンブレンをストリップし、抗ラットリン脂質スクランブラーゼ(PLSCR) mAb (PASTORELLIら, 2001, 上記)で再プローブして等しいローディングを評価した。フィルタをECL (Amersham-Pharmacia Biotech)により現像した。
結果を図5に示す。
* は、刺激された細胞内の顕著なチロシンリン酸化されたタンパク質を示す。
結果は、FcαRIトランスフェクタントの、抗FcαRI Fab A77とのインキュベーションは、無関係のコントロールFabに比べて、いくつかのチロシンリン酸化タンパク質の出現を誘導したことを示す。リンタンパク質のパターンは、FcαRIの多量体凝集後に得られるものと、強度は違うものの同じなようである。同様のデータが、FcαRIR209L/γキメラ受容体を用いて得られたが、ITAM内の変異により、単量体及び多量体のターゲティングの後に、この受容体のチロシンリン酸化を開始する能力が廃止された。
活性化工程の調節を、細胞活性化についての鍵となるメッセンジャーであるサイトソルのカルシウム流入([Ca2+]i)に対する抗FcαRI Fab A77の効果に関して調べた。
ヒトFcαRIトランスフェクタント又は非トランスフェクションRBL-2H3細胞の一定量(1.5×106細胞)を、異なる試験化合物(各実験について以下に記載する)で、20 mM HEPES pH 7.6を補充した完全DMEM中に、37℃で1時間感作した。細胞に、4μMの蛍光プローブFURA-2-AM (Molecular probes, Leiden, The Netherlands)を37℃で30分間負荷した。
ヒトFcαRIトランスフェクタント(a、クローン15.4)及び非トランスフェクションRBL-2H3細胞(b)を、IgE単独又はIgEと10μg/ml抗FcαRI Fab A77又は無関係のFab 320とで感作した。
結果を図6a及び6bに示す。
FcεRI刺激後の細胞内最大カルシウムは影響を受けなかった(図6a、□)が、上昇した[Ca2+]iのプラトー段階は、無関係のコントロールFab (図6a、△)又はトランスフェクションされていない細胞(図6b)に比べて、FcαRIトランスフェクタントの抗FcαRI Fab A77とのプレインキュベーション後に著しく阻害された(図6a、○)。
ヒトFcαRIトランスフェクタント(クローン15.4)を、IgE単独、又はIgEと10μg/ml 抗FcαRI Fab A77若しくは無関係のFab 320とで感作した。次いで、細胞にFURA-2-AMを記載したようにして負荷し、細胞外カルシウムを3.5 mM EGTAでキレート化し、その直後に[Ca2+]iを決定して、細胞内貯蔵からのカルシウム放出と細胞外媒質からのカルシウム侵入とを識別した。結果を図6cに示す。
結果は、抗FcαRI Fab A77処理がEGTA処理された細胞に影響を及ぼさないことを示し、細胞内カルシウム貯蔵の放出を阻害しなかったことを示唆する。
カルシウム流入のみが影響されることを確かめるために、外部のCa2+をMn2+で置き換えた。これは、貯蔵感受性カルシウムチャネル(SOC)を通って細胞に侵入し、内部の遊離のカルシウムと競合し、それによりFURA-2-AM蛍光を消す(BERTHONら, 1994, 上記)。
ヒトFcαRIトランスフェクタント(クローン15.4)を、IgE単独(d)、又はIgEと10μg/ml 抗FcαRI Fab A77 (e)若しくは無関係のFab 320 (f)とで感作した。
結果を図6d〜fに示す。

Agは、Mn2+流入によりFURA-2-AM蛍光の消光を誘導した。刺激の前と後のカルシウム侵入に対応するスロープは、それぞれ2.5及び4.1 (d)、1.8及び1.9 (e)、並びに3.5 (f)である。
FcαRI媒介阻害が、内部の貯蔵からのカルシウム放出とカルシウム流入との間の事象を標的したかについて調べるために、タプシガルジンを用いた。これは、イノシトール三リン酸感受性貯蔵を枯渇させ、膜貫通受容体の連動状態(engagement)の不在下でSOC開放をもたらす薬理作用剤である(THASTRUPら, Agents Actions 27: 17〜23, 1989)。
データは、少なくとも3回の別の実験の代表である。
結果を、図6g及び6hに示す。
FcαRIの阻害活性はインビトロで証明されたが、この受容体のインビボターゲティングを、それが炎症性応答を阻害できるかを知るために試験した。
マウスはFcαRIホモログを発現しないので(KABAGAWAら, Proc. Nat. Acad. Sci. 94: 5261〜5266, 1997; HAYAMIら, J. Biol. Chem. 272: 7320〜7327, 1997)、ヒトにおいて観察されるのに類似の骨髄細胞発現を生じる、CD11bプロモータの制御下にヒトFcαRI(CD89、83系統)を発現するBalb/cトランスジェニックマウス(Tg)を用いた(LAUNAYら, J. Exp. Med. 191: 1999〜2009, 2000)。遺伝子型決定は、PCRにより行った(LAUNAYら, 2000, 上記)。マウスは、IFR 02及びBichat Medical Schoolのマウス施設で飼育し、維持した。全ての実験は、国のガイドラインに従って行った。
結果を図7aに示す。
対応するPenh値の曲線下累積面積(AUC)は、群あたり少なくとも8匹のマウスを含む3回の別々の実験の平均±SDであり、図示した。

抗FcαRI Fabで処置した(FcαRI-)同腹子の肺において、効果は観察されなかった(示さず)。
抗FcαRI免疫療法を、間質性腎臓繊維症及び閉塞性腎症の炎症モデルである、マウスにおける片側尿管閉塞(UUO)の後にも試験した(KLAHR及びMORRISSEY, Am. J. Physiol. Renal Physiol. 283(5): F861〜875, 2002)。腎臓は、尿細管拡張、マクロファージのような炎症性細胞の浸潤、及び腎臓の上皮−間葉性推移を特徴とする。簡単に、左腎の尿管の片側閉塞を、2箇所での結紮により、麻酔をかけたTg CD89マウスに対して行った。外科的介入の1日前及びその後毎日、マウスをPBS、100μgの無関係なFab 320又は100μgのFab A77のいずれかで処置した。第6日にマウスを犠牲にし、閉塞した腎臓を、組織病理学的評価に供した(過ヨウ素酸シッフ(PAS)染色及び抗CD11b抗体を用いる免疫組織化学染色)。
結果を図8に示す。

Claims (4)
- FcαRI受容体のEC2ドメインに指向された一価抗体断片の、炎症性疾患の治療用の医薬の製造のための抗炎症性有効成分としての使用。
- 前記炎症性疾患が、狼瘡、リウマチ性関節炎、糖尿病、腎炎、間質性腎臓線維症、閉塞性腎症及び腸管炎症性疾患から選択される請求項1に記載の使用。
- 前記疾患がアレルギー性疾患である請求項1に記載の使用。
- 前記アレルギー性疾患が喘息である請求項3に記載の使用。
