Abstract
Description
発酵後、加圧された空気の添加を遮断し、超過圧を除去し、培養物及びプロセス液より上の上部空間への窒素の噴き付けによって、培養液を無酸素状態にする。上部空間への窒素の噴き付けは、その後の全ての処理工程(細胞の収集及び洗浄、細胞溶解並びに封入体の追加処理が含まれる。)の間、継続される。窒素の噴き付けは、容器、貯蔵タンクなどの処理装置並びに遠心管、ホモゲナイザー及びろ過ユニットなどの補助装置に対しても行われる。窒素の噴き付けは、固定された配管を用いて実施される。
Claims (8)
- 本来の高次構造中にジスルフィド結合を有するタンパク質を実質的に無酸素の条件下で単離することを備える、前記タンパク質を単離する方法。
- 前記実質的に無酸素の条件が実質的に無酸素の雰囲気を備える、請求項1に記載の方法。
- 前記タンパク質が、組換え細胞の封入体として存在する組換えハイブリッドタンパク質である、請求項1又は2に記載の方法。
- 前記単離が、前記細胞の収集及び破壊、前記封入体の単離及び/又は単離された産物の安定化を備える、請求項3に記載の方法。
- 前記タンパク質が前駆体タンパク質である、請求項1から4の何れか1項に記載の方法。
- 前記タンパク質がインシュリンである、請求項1から5の何れか1項に記載の方法。
- 前記実質的に無酸素の雰囲気が窒素雰囲気である、請求項1から6の何れか1項に記載の方法。
- 前記組換え細胞が、プラスミドpDBAST−RAT−N−7−1を担持し、該プラスミドを発現するE.コリ株Sφ733の細胞である、請求項1から7の何れか1項に記載の方法。
