Description
好ましくは、カルボキシル基を有するポリマーはカルボキシメチルデキストランである。
好ましくは、金属は金、銀、銅、白金またはアルミニウムのいずれかである。
好ましくは、本発明のバイオセンサーは、非電気化学的検出に使用される。
好ましくは、本発明のバイオセンサーは、表面プラズモン共鳴分析に使用される。
好ましくは、生理活性物質と相互作用する物質を表面プラズモン共鳴分析により検出または測定する。
核酸としては、測定の対象とする核酸と相補的にハイブリダイズするものを使用することができる。核酸は、DNA(cDNAを含む)、RNAのいずれも使用できる。DNAの種類は特に限定されず、天然由来のDNA、遺伝子組換え技術により調製した組換えDNA、又は化学合成DNAの何れでもよい。
低分子有機化合物としては通常の有機化学合成の方法で合成することができる任意の化合物が挙げられる。
免疫グロブリン結合性蛋白質としては、例えばプロテインAあるいはプロテインG、リウマチ因子(RF)等を使用することができる。
糖結合性蛋白質としては、レクチン等が挙げられる。
脂肪酸あるいは脂肪酸エステルとしては、ステアリン酸、アラキジン酸、ベヘン酸、ステアリン酸エチル、アラキジン酸エチル、ベヘン酸エチル等が挙げられる。
本実施例は、タンパク質を固定するためのセンサーチップの作製に関するものである。
(1)試料1(比較例)の作成
カルボキシメチルデキストランが結合した表面として、Biacore社センサーチップCM-5(research grade)を、そのまま用いた。
2.8mM のHODhbt(3,4-ジヒドロ-3-ヒドロキシ-4-オキソ-1,2,3-ベンゾトリアジン)水溶液と0.4M のEDC水溶液の1:1混合溶液100μlを、Biacore社センサーチップCM-5(research grade)表面に接触させ、室温で10分間反応させた。水洗後、真空乾燥機を用いて室温で10分間乾燥させた。この表面に対し、1,2-ビス(2-アミノエトキシ)エタン100μlを接触させ、室温で10分間反応させた後、水洗することで、目的表面を得た。
本実施例は、1,2-ビス(2-アミノエトキシ)エタンで修飾された表面(試料2)に対する、等電点以上のpHにおけるタンパク質のプレコンセントレーションに関するものである。タンパク質としては、ペプシン(和光純薬社製)、BSA(Bovine Serum Albumin:SIGMA社製)、CA(Carbonic Anhydrase:SIGMA社製)を用いた。
本実施例は、1級アミノ基を有する表面にプレコンセントレートされたタンパク質の固定に関するものである。Biacore3000を用い、ペプシン(1mg/ml、pH7.4)を5分間プレコンセントレートさせた試料2に対しカルボン酸活性化剤であるEDC(0.4M)/NHS(0.1M)水溶液を接触させずに洗浄したサンプル(試料3)、および、EDC(0.4M)/NHS(0.1M)水溶液を5分間接触させた後に洗浄したサンプル(試料4)に対するペプシン固定量を検討した。洗浄は、1%EDTA水溶液(1分×2回)およびグリシン緩衝液(pH1.5、Biacore社製)の注入(各1分×2回)により行った。得られた結果を図4に示す。
本実施例は、1,2-ビス(2-アミノエトキシ)エタン以外のポリアミンで修飾されたセンサーチップの作製に関するものである。1,2-ビス(2-アミノエトキシエタン)を、表1に示すポリアミンに変更した以外は、試料2の作成と同様の操作を行うことで、試料5〜9を作成した。
本実施例は、実施例4で作成されたセンサーチップに対する、タンパク質の荷電濃縮に関するものである。実施例2で示した方法で、試料5〜試料9に対するBSA(pH7.4, 0.1mg/ml)のプレコンセントレーションの程度を検討した。得られた結果を、図5に要約する。
Claims (17)
- 生理活性物質を生理活性物質の等電点以上のpHでプレコンセントレーションさせた後に、該生理活性物質を化学的に固定化することが可能な1級または2級アミノ基を有するポリマーが結合した固体基板からなるバイオセンサー。
- 1級または2級のアミノ基を有するポリマーが、カルボキシル基を有するポリマーとポリアミンとを反応させることにより得られるポリマーである、請求項1に記載のバイオセンサー。
- カルボキシル基を有するポリマーがカルボキシメチルデキストランである、請求項2に記載のバイオセンサー。
- 1級または2級アミノ基を有するポリマーが結合した固体基板が、水溶性高分子が結合している固体基板、疎水性高分子が結合している固体基板、又は自己組織化単分子膜が形成されている固体基板の何れかである、請求項1から3の何れかに記載のバイオセンサー。
- 1級または2級アミノ基を有するポリマー層が金属上に形成されている、請求項1から4の何れかに記載のバイオセンサー。
- 金属が金、銀、銅、白金またはアルミニウムのいずれかである、請求項5に記載のバイオセンサー。
