Abstract

本発明は終末糖化産物受容体(RAGE)の調節に関する。特に、本発明は、RAGEポリペプチドを含む融合タンパク質、そのような融合タンパク質の作製方法、及びRAGEベースの障害の処置のためのそのようなタンパク質の使用について説明する。

実施例1A:RAGE融合タンパク質の製造
2つのプラスミドを構築して、RAGE-IgG融合タンパク質を発現させた。両方のプラスミドを、ヒトRAGE由来の異なる長さの5’側のcDNA配列と、ヒトIgG(γ1)からの同じ3’側のcDNA配列とを連結することによって構築した。これらの発現配列(すなわち、連結産物)を、次に、pcDNA3.1発現ベクター(Invitrogen, CA)内に挿入した。RAGE融合タンパク質コード領域をコードする核酸配列を、図2及び3に示す。TTP-4000RAGE融合タンパク質について、第1〜753の核酸配列(ボールド体で強調した)が、RAGEのN末端タンパク質配列をコードするのに対して、第754〜1386の核酸配列は、IgGタンパク質配列をコードする(図2)。TTP-3000について、第1〜408の核酸配列(ボールド体で強調した)が、RAGEのN末端タンパク質配列をコードするのに対して、第409〜1041の核酸配列はIgGタンパク質配列(図3)をコードする。
プラスミドを構築して、RAGE-IgG融合タンパク質を発現させた。プラスミドを、ヒトRAGE由来の5’側のcDNA配列と、ヒトIgG(γ1)からの3'側のcDNA配列とを連結することによって構築した。cDNAを増幅するためにPCR法を使用した。さらに、5’末端において、PCRプライマーが、クローニングからのECO RI制限酵素部位、及びコザック・コンセンサス翻訳開始配列を加えた。3’末端において、PCRプライマーが、終止コドンの直後にXho I制限を加えた。3’末端において、PCRプライマーが、また、終止コドン近くに免疫グロブリン部分内のクリプティックRNAスプライシング部位を取り除く2つのサイレント塩基変化も含んでいた。プロリン(配列番号32のタンパク質配列における番号付けに基づく第409残基)をコードするコドンをCCGからCCCに変更し、そして、グリシン(配列番号32のタンパク質配列における番号付けに基づく第410残基)をコードするコドンをGGTからGGGに変更した。PCRフラグメントを、Eco RI及びXho Iで消化し、次に、(Eco RIとの互換性末端を形成するために)Mfe Iで消化し、且つ、Xho Iで消化したレトロベクター・プラスミド(pCNS-新しいMCS-WPRE(新しいori)、Gala, Inc.から入手可能)内に挿入した。クローン化プラスミド及びクローニング接合部から成る挿入部分を配列決定し、クローニング中に突然変異が起こらなかったことを保証した。
A.インビトロにおけるリガンド結合
既知のRAGEリガンドを、1ウェルあたり5マイクログラムの濃度でMaxisorbプレート表面上にコートした。プレートを4℃で一晩インキューベートした。リガンド・インキュベーションに続いて、プレートを吸引し、そして、50mMのイミダゾール・バッファー(pH7.2)中、1%のBSAから成るブロッキング・バッファーをプレートに加えた(室温で1時間)。プレートを、次に、吸引し、及び/又は洗浄バッファー(20mMのイミダゾール、150mMのNaCl、0.05%のTween-20、5mMのCaCl2、及び5mMのMgCl2、pH7.2)で洗浄した。1.082mg/mLの初期濃度にてTTP-3000の溶液(TT3)、及び370μg/mLの初期濃度にてTTP-4000の溶液(TT4)を調製した。RAGE融合タンパク質を、初期サンプルを漸増する希釈度にて加えた。RAGE融合タンパク質を、固定化リガンドと一緒に37℃で1時間インキューベートし、その後、そのプレートを洗浄し、そしてRAGE融合タンパク質の結合についてアッセイした。結合を、21ng/100μLの最終アッセイ濃度(FAC)まで1:11,000に希釈したモノクローナル・マウス抗ヒトのIgG1、500ng/μLのFACまで1:500に希釈したビオチン化ヤギ抗マウスIgG、及びアビジン連結アルカリホスファターゼを含む免疫検出複合体の添加によって検出した。複合体を、固定化RAGE融合タンパク質と一緒に室温で1時間インキューベートし、その後、プレートを洗浄し、そして、アルカリホスファターゼ基質・パラニトロフェニルホスファート(PNPP)を加えた。固定化RAGE融合タンパク質への複合体の結合を、405nmにて分光光度測定で計測されるPNPPのパラ-ニトロフェノール(PNP)への転換を測定することによって定量化した。
これまでの研究によると、骨髄性THP-1細胞がRAGEリガンドに応答してTNF-αを分泌する可能性があることが示されている。このアッセイでは、THP-1細胞を、ATCCによって提供されたプロトコールを使用して10%のFBSを補ったRPMI-1640培地中で培養した。その細胞を、RAGE融合タンパク質TTP-3000(TT3)又はTTP-4000(TT4)(10μg)、sRAGE(10μg)、ヒトIgG(10μg)(すなわち、ネガティブコントロールとして)の不存在下、及び存在下の両方で、0.1mg/mlのS100bを伴ったRAGEの刺激によってTNF-αを分泌するように誘導した。THP-1細胞によって分泌されたTNF-αの量を、細胞培養へのタンパク質添加の24時間後に、TNF-αのための市販のELISAキット(R&D Systems, Minneapolis, MN)を使用することで計測した。図10の結果は、RAGE融合タンパク質が、これらの細胞におけるTNF-αのS100b/RAGEに誘発された産生を抑制するということを実証している。図10に示されているとおり、10μgのRAGE融合タンパク質TTP-3000又はTTP-4000の添加で、S100b(0.1mg/ml FAC)によるTNF-αの誘導はそれぞれ約45%〜70%まで減少した。TNF-αのS100b誘導を妨げることにおいて、融合タンパク質TTP-4000は、少なくともsRAGEと同じくらい有効でありうる(図10)。TTP-4000及びTTP-3000のRAGE配列に対する阻害の特異性は、S100b刺激細胞にIgG単独で加えられた実験によって示されている。アッセイへのIgG及びS100bの添加は、S100b単独の場合と同じレベルのTNF-αを示している。RAGE融合タンパク質のRAGE配列に対するTTP-4000及びTTP-3000によるTNF-α誘導の阻害の特異性は、S100b刺激細胞にIgGのみを加えた実験によって示される。アッセイへのIgG、すなわち、RAGE配列を持たないヒトIgG(SigmaヒトIgGを10μg/ウェルで加えた)及びS100bの添加が、S100b単独の場合と同じレベルのTNF-αを示すことがわかった。
ヒトsRAGEと比べてTTP-4000が優れた薬理動態学特性を有するか否かを判定するために、ラット及び非ヒト霊長類をTTP-4000の静脈内(IV)注射(5mg/kg)に供し、次に、TTP-4000の存在について血漿を評価した。これらの実験では、2匹の未処置雄サルに、末梢静脈中へのTTP-4000の単回IV大量投与(5mg/ml/kg)と、それに続く約1.0ミリリットル(mL)の生理食塩水洗浄液を与えた。血液サンプル(約1.0mL)を、投薬前(すなわち、TTP-4000注射の前)、又は投薬後0.083、0.25、0.5、2、4、8、12、24、48、72、96、120、168、240、288、及び336時間に、(リチウム・ヘパリン)を含む試験管内に採取した。採取に続いて、その試験管を、冷却(2〜8℃)下、1500×gで15分間の遠心分離にかけるまで湿った氷上に(最大30分)に置いた。それぞれの得られた血漿サンプルを、次に、実施例6に記載されるように、注射に続いて様々な時点でELISAを使用してRAGEポリペプチドについてアッセイするまで冷凍(−70℃±10℃)で保存した。
ヒトIgGと比べた場合のRAGE融合タンパク質TTP-4000によるFc受容体の活性化を測定するために実験を実施した。Fc受容体の活性化を、Fc受容体を発現するTHP-1細胞からのTNF-α分泌を測定することによって計測した。これらの実験では、96ウェル・プレートを10μg/ウェルのTTP-4000又はヒトIgGでコートした。Fc刺激がTNF-α分泌をもたらした。TNF-αの量を、酵素結合免疫吸着検定法(ELISA)によって計測した。
ヒト疾病のいくつかのin vivoモデルにおいて、TTP-4000の活性をsRAGEと比較した。
A.再狭窄の動物モデルにおけるTTP-4000
RAGE融合タンパク質TTP-4000を、血管傷害後21日間の平滑筋増殖及び内膜拡張の計測に関与するラット糖尿病モデルの再狭窄において評価した。これらの実験では、左総頸動脈のバルーン障害を、標準的は手順を使用してザッカー糖尿病、及び非糖尿病のラットにおいて実施した。IgG、TTP-4000、又はリン酸緩衝生理食塩水(PBS)の初回量(3mg/ラット)を、傷害の1日前に腹腔内(IP)に投与した。維持量を、傷害の7日目後まで一日おき(すなわち、傷害後1、3、5、及び7日目)に提供した。維持量は、高=1つの群に1mg/動物、又は低=もう1つの群に0.3mg/動物であった。血管平滑筋細胞(VSMC)増殖を測定するために、動物を、傷害の4日後と21日後に屠殺した。細胞増殖の計測のために、4日目の動物には、安楽死の18、12、及び2時間前にブロモデオキシウリジン(BrDdU)50mg/kgの腹腔内投与を与えた。屠殺後、左及び右の頚動脈の全体を採取した。標本を、包埋前の少なくとも24時間Histochoice中に保存した。VSMC増殖の評価を、マウス抗BrdUモノクローナル抗体を使用することで実施した。蛍光標識ヤギ抗マウス二次抗体を適用した。切片あたりのBrdU陽性の核の数を、処置計画を伏せられた2人の観察者によってカウントした。
TTP-4000がマウスADモデルにおけるアミロイド形成と認知機能障害に影響を及ぼすかどうか評価するために、実験を実施した。実験には、PDGF-B鎖プロモーターの制御下のヒト・スウェーデン型突然変異アミロイド前駆タンパク質(APP)を発現するトランスジェニック・マウスを利用した。時間が経つにつれて、これらのマウスは、高レベルのRAGEリガンドであるアミロイド-β(Aβ)を産生した。これまでは、3カ月間のsRAGE処置が、このモデルにおける脳内のアミロイド斑形成及び炎症性マーカーの関連した増加の両方を減少させることが示された。
組織学的検査のために、動物を、ペントバルビタール・ナトリウム(50mg/kg)の腹腔内投与(IP)で麻酔した。動物に、4℃のリン酸緩衝食塩水(PBS)と、それに続いて4%のパラホルムを経心臓的に潅流した。脳を取り出し、4%のパラホルムアルデヒド中に一晩、置いた。その脳を、パラフィン処理し、そして包埋した。脳を通した10枚の連続した30μm厚の切片を得た。トランスジェニック動物の脳内のアミロイド沈着を検出するために、切片を一次抗体(Aβペプチド抗体)に4℃で一晩、晒した(Guoら、J. Neurosci. 22:5900-5909ページ(2002年))。切片を、トリス緩衝食塩水(TBS)中で洗浄し、二次抗体を加え、そして室温で1時間インキューベートした。洗浄後に、切片を、ベクターABC Eliteキット(Vector Laboratories)に指示されるとおりにインキューベートし、そしてジアミノ安息香酸(DAB)で染色した。反応を水により止めて、キシレンでの処理後にカバースリップを乗せた。各切片内のアミロイド領域を、Quick Captureフレーム取り込みカード、オリンパス顕微鏡に備え付けられたヒタチCCDカメラ、及びカメラ・スタンドを備えたPower Macintosh(登録商標)コンピュータから成るコンピュータを利用した画像解析システムを用いて測定した。NIH画像分析ソフトウェア、v. 1.55を使用した。画像を得、そしてアミロイドの総面積を、10枚の切片にわたって測定した。処置状況を伏せられた1人のオペレーターが全ての測定を実施した。切片のアミロイド量を合計し、そして切片の総数で割ることで、アミロイド量を計算した。
モリス水迷路試験を以下のとおり実施した。全てのマウスを、実験の終わりにモリス水迷路試験で一度試験した。マウスを、1.2mのオープンフィールド水迷路で訓練した。プールを、30cmの深さまで水で満たし、そして25℃に維持した。エスケープ・プラットフォーム(10cm四方)を水面の1cm下に配置した。試験中、プラットフォームをプールから取り出した。手掛かり試験(cued test)を、白いカーテンで囲まれて、あらゆる余分な迷路の手掛りを隠したプールの中で行った。全ての動物に、連続した3日間、空間的な事前訓練を与えなかった(NSP)。これらの試験は、プラットフォームを見つけるための記憶の保持力を測定するための最終的な行動試験のための動物を調製するためにある。これらの試験は、記録されなかったが、訓練目的だけのためのものであった。訓練する、及び研究を学習するために、カーテンは余分な迷路の手掛りを取り除いた(これは水泳障害を伴う動物の識別を可能にする)。1日目に、マウスを、20秒間、隠したプラットフォーム上に置き(試験1)、試験2〜3のために、動物を、手掛かりのあるプラットフォーム又は隠したプラットフォーム(試験4)から10cm離れた水中に放して、プラットフォームまで泳がせた。試験の2日目に、隠したプラットフォームを、プールの中央、又はそれぞれの象限の中央の間で無作為に移動させた。動物を、無作為に壁と向き合わせてプール内に放し、そして60秒でプラットフォームに到達させた(試験3)。3番目の試験において、動物に、隠したプラットフォームによる2つと、手掛かりのあるプラットフォームによる1つの、3つの試験を与えた。NSP後の2日間、動物を、最終的な行動試験(モリス水迷路試験)にかけた。これらの試験(動物1匹あたり3つ)のために、プラットフォームをプールの1つの象限の中央に配置し、そして動物を無作為に壁に向かい合わせて放した。動物に、プラットフォームを見つけさせるか、又は60秒間、泳がせた(反応時間、プラットフォームを見つける時間)。全ての動物を、投薬の4〜6時間以内に試験し、そして試験群が分からないようにしたオペレーターによって試験するために無作為に選択させた。
脳卒中の疾患関連動物モデルにおいて、TTP-4000をsRAGEとも比較した。このモデルでは、マウスの中央頚動脈を1時間結紮し、続いて23時間の再潅流をして、その時点でマウスを屠殺し、そして脳内の梗塞の面積を評価した。再潅流直前に、マウスを、sRAGE、又はTTP-4000、あるいはコントロール免疫グロブリンによって処理した。
最初に、1×PBS pH7.3中、10ug/mLの濃度にて50uLのRAGE特異的モノクローナル抗体1HB1011を、一晩のインキュベーションによってプレートにコートする。レディーフォーユースの場合、プレートを、300uLの1×イミダゾール-Tween洗浄バッファーで3回洗浄し、そして1%のBSAでブロックした。(希釈した)サンプルと、既知のTTP-4000希釈物の標準希釈物を、100uLの最終量で加える。サンプルを、室温で1時間インキューベートする。インキュベーション後に、そのプレートを3回洗浄する。1%のBSAを含む1×PBS中、ヤギ抗ヒトIgG1 1(Sigma A3312)APコンジュゲートを加え、室温で1時間インキューベートする。プレートを3回洗浄する。呈色をパラニトロフェニルホスファートではっきりさせた。
図15は、種々の固定化された既知のRAGEリガンドに対するTTP-4000による飽和結合曲線を示している。リガンドを、マイクロタイタープレート上に固定し、0〜360nMの漸増する濃度のRAGE融合タンパク質の存在下でインキューベートする。RAGE融合タンパク質-リガンド相互作用を、融合キメラのIgG部分に特異的である、アルカリホスファターゼを抱合したポリクローナル抗体を使用して検出する。相対Kdsを、Graphpad Prizmソフトウェアを使用して計算し、そしてRAGE-RAGEリガンド値の立証された文献値と照合した。HMG1B=アンホテリン、CML=カルボキシメチルリジン、Aβ=アミロイドβ1-40。
RAGE遮断剤には、同種間移植拒絶反応をブロックすることを期待することができる。これらの実験は、本発明のRAGE融合タンパク質を使用したリガンド-RAGE相互作用の遮断剤が、移植した動物が目標濃度を下回る血糖値を維持した時間の長さによって計測される、健常ドナーから糖尿病動物に移植された島細胞の拒絶反応を軽減するか否かを調査した。本明細書中で議論したように、島細胞移植を受けた糖尿病動物へのRAGE融合タンパク質(例えば、TTP-4000)の投与が、移植の2つ(同種間と同系間)の動物モデルにおいて、高血糖の再発とそれによる移植した島細胞の拒絶反応をかなり遅らせたことがわかった。
第1の実験セットは、RAGE融合タンパク質(TTP-4000)の投与が移植された島細胞の同種間拒絶反応、及びC57BL/6J(B6)マウス糖尿病モデルにおける糖尿病の再発を調節するか否かを試験した。
糖尿病の動物モデル
C57BL/6J(6〜8週齢)(B6)マウスを、200mg/kgでのストレプトゾトシン(STZ)(Sigma Chemical Co., St Louis, MO)の単回静脈内投与によって糖尿病にした。BALB/cJ(6〜8週齢)(BALB)マウスは、島移植のためのドナーとしての役割を担い、それにより島移植に関する同種不一致をもたらす。
マウス(BALB/c)を塩酸ケタミン/塩酸キシラジン溶液(Sigma, St Louis MO)で麻酔した。1.5mg/mlのコラゲナーゼP(Roche Diagnostics, Branchburg, NJ)を含む冷ハンクス平衡塩類溶液(HBSS, Gibco, Grand Island NY)3mlの導管内注射の後に、膵臓を、外科的に得、そして37℃で20分間、消化した。膵島を、HBSSで洗浄し、4つの異なる密度(26%、23%、20%、及び11%)を持つPolysucrose 400(Cellgro, Hemdon VA)を使用することで不連続勾配遠心分離によって精製した。20%と23%の層の界面の組織片を、回収し、洗浄し、そしてHBSS中に再懸濁した。個々の膵島、遊離の付随する腺房、血管及び管組織を、倒立顕微鏡下で選び出し、移植のための高度に精製された膵島を得た。
ストレプトゾトシン誘発糖尿病C57BL/6(B6)マウスは、糖尿病の診断の2日以内に島移植を受けた。BALB/cJ(6〜8週齢)(BALB)マウスが、同種間島移植のためのドナーの役割を担った。移植のために、ドナー・マウスからの500〜600個の新しく分離した膵島(すなわち、約550個の膵島相当物)を、輸液セットで吸い上げ、そして移植個体の右腎の被膜下空間内に移植した。
膵島を移植してすぐに、試験化合物を投与した。どの程度コントロール動物がうまくいっているかどうかに依存して、投与を約60日間続けた。マウスに、以下の処置計画(表3)に従って、リン酸緩衝生理食塩水(PBS)、PBS中のTTP4000、又はPBS中のIgGのいずれかの0.25mlを注射した。
膵島移植片機能を、膵島移植後の最初の2週間は毎日、続いてその後は一日おきの連続した血糖測定によって観察した。糖尿病の好転は、2回の連続した測定値にて200 mg/dl未満の血糖値と規定した。2回の連続した測定値にて血糖が250mg/dlを超えた場合に、移植片の喪失を確定した。結果を表4中に示す。
第2の実験セットは、同種間NOD移植モデルを使用して、RAGE融合タンパク質(すなわち、TTP-4000又はTTP-3000)の投与がNODマウスにおける糖尿病の再発の経過を調節するか否かを試験した。
糖尿病の動物モデル
突発性自己免疫性非肥満糖尿病マウス(NOD/LtJ)(12〜25週齢)が膵島細胞のための移植個体としての役割を担った一方で、若い前糖尿病性NOD/LtJマウス(6〜7週齢)が同種間島移植のドナーとしての役割を担った。移植のための膵島を、A節において先に記載のとおり分離した(同種間島移植)。
糖尿病NOD/LtJマウスは、糖尿病の診断の2日以内に膵島移植を受けた。ドナー・マウスからの500〜600個の新しく分離された膵島(約550個の膵島相当物)を、輸液セットを用いて吸い上げ、そして右腎の被膜下空間内に移植した。
試験化合物での処置
膵島を移植してすぐに、試験化合物を投与し、そして約8週間続けた。マウスに、以下の処置計画(表6)に従って、PBS、PBS中のTTP4000、又はPBS中のTTP-3000のいずれかの0.25mlを注射した。
膵島移植片機能を、膵島移植後の最初の2週間は毎日、続いてその後は一日おきの連続した血糖測定によって観察した。糖尿病の好転は、2回の連続した測定値にて200 mg/dl未満の血糖と規定した。2回の連続した測定値にて血糖が250mg/dlを超えた場合に、移植片の喪失の割合を確定した。結果を表7中に示す。
