Abstract

Description
本発明は、インビトロまたはインビボにて、神経系細胞(neural cell)(例えば、ニューロンまたはグリア細胞)を選択的に産生する方法に関する。また、神経系細胞を産生することによって神経変性疾患または病状を処置または緩和させる方法を提供する。

本発明は、インビトロまたはインビボで神経系細胞を産生する2工程の方法に関する。本発明者らは、多能性神経幹細胞(NSC)による神経新生およびグリア新生が、増殖および方向付けられた分化に関わることを発見した。図1に示されるように、EGF(すなわち、EGFの成体ホモログのTGFα)は、NSC集団の自己複製/増殖(expansion)を誘導する。NSCは、グリア前駆細胞(GPC)になる規定の経路において自然発生的な分化を起こす。この自然発生的な分化は、毛様体神経栄養因子(CNTF)によって減弱され得る。GPCは、グリア細胞に分化する(この分化は、EGFによって促進される)。あるいは、NSCは、ニューロンだけを産生する神経前駆細胞(NPC)に分化するように、EPOおよび/またはPACAP/cAMPによって指示を受け得る。

(i)例えば、EGFを提供することによって、神経幹細胞の増殖を増大させる工程;
(ii)例えば、CNTFを提供することによって、神経幹細胞の自然発生的な分化を阻害する工程;または
(iii)例えば、エストロゲンを提供することによって、神経幹細胞の生存を促進する工程。
本発明は、インビトロまたはインビボで神経系細胞(ニューロンまたはグリア細胞を含む)を選択的に産生する方法に関する。また、神経系細胞を産生することによって神経変性疾患または病状を処置または緩和させる方法を提供する。従って、因子の組み合わせが、以下の2つの工程を達成するための使用される:神経幹細胞の数を増加させる工程およびニューロンまたはグリア細胞に選択的になるように、神経幹細胞に指示する工程。
「神経幹細胞」は、神経系の細胞系統における幹細胞である。幹細胞は、自己再生し得る細胞である。言い換えると、幹細胞の分割から生じた娘細胞は幹細胞を含む。神経幹細胞は、神経系の細胞系統(ニューロン、アストロサイトおよびオリゴデンドロサイト(アストロサイロトおよびオリゴデンドロサイトは、グリアまたはグリア細胞の総称である)を含む)にける全ての細胞型に最終的に分化し得る。従って、本明細書中で言及される神経幹細胞は、多能性の神経幹細胞である。
神経幹細胞(NSC)(例えば、成体前脳において見出される神経幹細胞)は、限定された神経系前駆体およびグリア前駆体の供給源である可能性が高く、これらは、それぞれ例えば、嗅球および脳梁といった構造中に存在する。NSCが限定された前駆体を生じる機構は、本発明以前には不明である。
(a)プロラクチン、および神経分化またはグリア分化を増強する少なくとも1つの因子(例えば、EPO、PACAP、サイクリックAMPおよび/またはBMP);

(c)プロラクチンと共に、神経幹細胞数を増加させる少なくとも1つの因子;
(d)PACAPと共に、神経幹細胞数を増加させる少なくとも1つの因子;
(e)EPOと共に、神経幹細胞数を増加させる少なくとも1つの因子;ならびに
(f)上記のものの組合せ。
本発明は、神経幹細胞数を増加し得る少なくとも1つの因子、および神経幹細胞の分化を増強し得る少なくとも1つの因子を含む組成物を提供する。いくつかの因子(例えば、プロラクチン)は、両方の機能が可能であることに留意すべきである。PACAPはまた、神経分化を増強することに加えて、別のマイトジェンの存在下で神経幹細胞の増殖を増強する。

(神経幹細胞培養)
神経幹細胞培養のためのプロトコルは、米国特許第5,750,376号またはShingoら,2001に詳細に示される。簡単にいうと、E14内側および側方の神経節隆起から胚性神経幹細胞を調製した。成熟した神経幹細胞を、成体マウスの脳室下帯から調製した。組織を、20ng/ml EGF、または各々のケースに示されるような他の成長因子を含む標準培地内で培養し、ニューロスフェア(neurosphere)を形成した。標準培地の組成物は、以下のようである:DMEM/F12(1:1);グルコース(0.6%);グルタミン(2mM);重炭酸ナトリウム(3mM);HEPES(5mM);インスリン(25μg/ml);トランスフェリン(100μg/ml);プロゲステロン(20nM);プトレシン(60μM);および塩化セレン(30nM)。
2月齢のCD−1マウス(Charles−River,Laval,Quebec,Canada)を、ペントバルビタールナトリウム(120mg/kg,i.p.)で麻酔し、浸透圧ポンプ(Alzet 1007D;Alza Corporation,Palo Alto,CA)で移植した。カニューラを右側脳室(ブレグマに対して、前後に+0.2mm、側方に+0.8mm、および背腹方向に硬膜下−2.5mm、ラムダとブレグマとの間の頭蓋レベルで)に配置した。ヒト組換えEPO(1000IU/ml)、ウサギ抗EPO中和抗体(100μg/ml)、ウサギIgG(100μg/ml)、ラット組換えCNTF(33μg/ml)、またはヒト組換えEGF(33μg/ml)を、0.9%生理食塩水(1mg/ml マウス血清アルブミン(Sigma)を含む)中に溶解した。各動物に、0.5μl/時間の流速で、6日間継続的に注入し、1日あたり約25IUのEPO、3μgの抗体、あるいは400ngのCNTFまたはEGFを送達した。
雄成体ロングエバンスラット(250〜350g)を、Charls River Breeding Farmsから入手し、任意の処理前の2週間、コロニーに適応させた。手術の1週間前に、このラットで、行動試験におけるベースライン試験を行った。
発作研究のための動物に、感覚運動皮質の片側の脈管遮断を施した。イソフルラン麻酔を用いて、皮膚を切開し、収縮させ、そして表面の筋膜を頭蓋から除去した。頭蓋の開口部を、硬膜を傷つけないように注意して、以下の座標に形成した:AP +4.0〜−2.0;L 1.5〜4(傍矢状の隆線;Kolbら,1997)。この硬膜を切り、頭蓋の開口部から収縮させた。生理食塩水中に浸漬した綿棒で、露出した軟膜越しに穏やかに擦り、血管を除いた。次いで、反対側の半球に穴を開け、浸透圧ミニポンプ(AP −0.5;L 1.5)を備えたカニューラのための穴を提供した。浸透圧ミニポンプを、肩甲骨の間の皮膚の下に配置し、チューブを、速乾性セメントで頭蓋に取り付けたカニューラに皮膚の下で接続した。一旦止血がなされると、頭皮を5−O 滅菌縫合糸を用いて縫合し、閉じた。この動物に、Banamine(鎮痛薬(analgesic))の注射を1回与え、ホームケージに戻した。骨に穴を開けた偽手術動物には麻酔のみを与え、皮膚を切開し、そして縫合した。
この試験は、新皮質前方領域の機能的な保全性の、感度の高い測定を構成することが示されている。通常のラットにおいて、泳ぎは、後肢からの推進力によってなされる。前肢は、通常、任意の動作が抑止され、動かないままであり、動物の頚部の下部と一緒にされる。前肢の抑止は、泳いでいる間、動物を撮影することによって評価した。動物を、室温の水(約25℃)で深さ25cmまで満たした水槽(30w×90l×43h cm)の一方の端に導入し、これらの動物が泳ぐのを、9.5cm四方の可視的プラットフォームに撮影した。このプラットフォームを、水面の2cm上に投影し、そして水槽の反対側の端に配置する。水槽の長さに沿った3回の泳ぎにおいて、各前肢によってなされる場合、動作回数の計数によって抑止をスコアリングした。泳ぎは、動物が、泳ぎを試行している間に水槽の側面に接触しなかった場合にのみ、スコアリング可能であると考えられる。
動物の前足の非対称性を、それらの動物(直径25cmのアクリルシリンダー中、アクリルプラットフォーム上)を下から撮影することによって決定した。動物が、5分間に左前足または右前足を上げ下げした回数をそれぞれ計数することによって、姿勢の安定性の挙動における優先度を決定した。この試験は、前肢の使用における非対称性の測定を可能にし、測定結果は、外傷に対する同側の肢の使用に対する信頼できるバイアスを示す。
6週の終りに、動物に過剰量のエタノール(Euthanol)を与え、ピクリン酸中の0.9%生理食塩水および4%パラホルムアルデヒドで心臓内を灌流した。この脳を細胞保護し、そしてビブラトームで、40ミクロンに切った。5セットの切片を、400ミクロンごとに保持した。2セットを染色した(一方をクレシルバイオレットで、そしてもう一方をダブルコルチンで)。残りのセットは、保存した。クレシルバイオレット染色を、スライド上で実施し、一方ダブルコルチン染色は、自由なフロートの切片における免疫組織化学手順で実施した。クレシルバイオレット染色は、損傷領域の評価を可能にし、一方、ダブルコルチン染色は、移動性の神経前駆細胞(neuronal progenitor cell)に存在するタンパク質と会合した。
(インビボでのCNTFおよびEPOの効果)
インビボでのCNTFおよびEPOの有効性を決定するために、CNTFまたはEPOを、材料および方法に記載されるように、6日間にわたり成体マウスに注入した。次いで、その脳組織を収集し、注入後の脳における神経幹細胞の数の指標として神経幹細胞の増殖に用いた。あるいは、チロシンヒドロキシラーゼまたはMash1について脳組織を染色し、神経発生領域を決定した。
(動作モデルにおける因子組合せの効果)
脳に損傷を受けた動物における因子の種々の組合せの効果を決定するために、動作のモデルとして、ラットの脳に局所的な虚血性損傷を誘導した。脳損傷の結果、この動物は、運動皮質に損傷を有し、2つの行動試験において異常な行動をした。1つは前肢抑止試験(新皮質前部の領域の機能保全性の感覚測定)である。通常のラットは、泳ぐとき、前肢の使用を抑止するが、本実験において、運動皮質の一方を損傷させた場合、運動皮質は、体の反対側を制御するので、そのラットは、反対側の前肢の使用を抑止しなかった。他の試験(前肢非対称性試験)において、正常なラットは、それらのラットが自身のバランスをとることを試行する場合と同様に、両前肢を使用する。しかし、損傷したマウスは、同側の前肢を使用することを好む。何故ならば、おそらくそれらのマウスは、自身の反対側の前肢をコントロールし得ないからである。
脳損傷前、全てのラットは、前肢抑止試験において両前足を抑止した。損傷後、全ての虚血性群(群2〜群6)は、反対側の前足を抑止しなかったが、同側の前足は抑止し続けた。試験因子の注入により、2つのプロラクチン群(群3および4群)は、より高い前足の抑止を示した。実際、最終週(注入完了の4週間後)の終りまでには、プロラクチン+EPO群(群4)は、コントロールと区別不可能であった。従って、プロラクチン、および特に、プロラクチンとEPOとの組み合せは、行動の代表的な症状からの回復をもたらした。
前肢非対称性試験の結果は、全ての試験群において、6週目の終りには非対称の程度は下がったが、成長ホルモンを与えた群(群3および群4)は、初めの4週間で、より速いかつ広範囲の回復が見られたことを示す。これらの結果は、解剖学的分析からの結果と一致し、成長ホルモン単独(群3)は、ダブルコルチン陽性細胞の増加をもたらし、そして成長ホルモンとEPOとの組合せ(群4)は、側脳室の周囲でのダブルコルチン陽性細胞の移動をもたらした。
Claims (27)
- 神経前駆細胞および/またはグリア前駆細胞を増殖させるために、そのような処置を必要とする疾患または状態を有する哺乳動物に投与される医薬組成物であって、
(a)神経幹細胞の数を増加させるのに十分な量の、プロラクチン、成長ホルモン、毛様体神経栄養因子(CNTF)および上皮成長因子(EGF)からなる群から選択される因子、および
(b)該神経幹細胞から神経前駆細胞またはグリア前駆細胞の生産を促進させるのに十分な量の、エリスロポエチン(EPO)、下垂体アデニレートシクラーゼ活性化ポリペプチド(PACAP)およびcAMPからなる群から選択される因子、
を有効成分として含む、因子(a)および(b)の同時投与または段階的投与のための組成物(但し、因子(a)がEGFである場合、因子(b)はPACAPまたはcAMPである。)。 - 因子(a)および(b)が、同時投与により投与される、請求項1に記載の医薬組成物。
- 因子(a)および(b)が、段階的投与により投与される、請求項1に記載の医薬組成物。
- 因子(a)および(b)の段階的投与が、初めに因子(a)を投与し、次いで因子(b)を投与することにより行なわれる、請求項3に記載の医薬組成物。
- 因子(a)がプロラクチンである、請求項1−4の何れかに記載の医薬組成物。
- 因子(a)が成長ホルモンである、請求項1−4の何れかに記載の医薬組成物。
- 因子(a)が毛様体神経栄養因子(CNTF)である、請求項1−4の何れかに記載の医薬組成物。
- 因子(a)が上皮成長因子(EGF)であり、因子(b)が下垂体アデニレートシクラーゼ活性化ポリペプチド(PACAP)またはcAMPである、請求項1−4の何れかに記載の医薬組成物。
- 因子(b)がエリスロポエチン(EPO)である、請求項1−7の何れかに記載の医薬組成物。
- 因子(b)が下垂体アデニレートシクラーゼ活性化ポリペプチド(PACAP)である、請求項1−8の何れかに記載の医薬組成物。
- 因子(b)がcAMPである、請求項1−8の何れかに記載の医薬組成物。
- 脳内投与用である、請求項1−11の何れかに記載の医薬組成物。
- 該脳内投与が、哺乳動物の前脳の脳室下帯に対して行なわれる、請求項12に記載の医薬組成物。
- 該疾患または状態が脳損傷である、請求項1−13の何れかに記載の医薬組成物。
- 該脳損傷が発作である、請求項14に記載の医薬組成物。
- 該疾患または状態が、アルツハイマー病、多発性硬化症(MS)、ハンティングトン病、筋萎縮性側索硬化症、およびパーキンソン病からなる群より選択される、請求項1−13の何れかに記載の医薬組成物。
- 神経前駆細胞またはグリア前駆細胞を産生する方法であって、以下:
(a)少なくとも1つの神経幹細胞を提供する工程、
(b)該神経幹細胞を、プロラクチン、成長ホルモン、毛様体神経栄養因子(CNTF)および上皮成長因子(EGF)からなる群より選択される因子の、該神経幹細胞の数を増加させるのに十分な量と接触させる工程、および
(c)工程(b)からの該神経幹細胞を、エリスロポエチン(EPO)、下垂体アデニレートシクラーゼ活性化ポリペプチド(PACAP)およびcAMPからなる群より選択される因子の、該神経幹細胞から神経前駆細胞またはグリア前駆細胞の産生を促進させるのに十分な量と接触させる工程、
を包含する方法(但し、工程(b)の因子がEGFである場合、工程(c)の因子はPACAPまたはcAMPである。)。 - 工程(b)が工程(c)の前に実施される、請求項17に記載の方法。
- 工程(b)および(c)が同時に実施される、請求項17に記載の方法。
- 該神経幹細胞が胚性細胞ではない、請求項17に記載の方法。
- 該神経幹細胞が成体神経幹細胞である、請求項17に記載の方法。
- 該神経幹細胞が非ヒト哺乳動物細胞である、請求項17に記載の方法。
