免疫応答のモジュレーションは重要な抗癌の手法となっている。癌ワクチンおよび免疫療法の設計における多大な研究は腫瘍細胞中に選択的に存在する抗原の発見に着目している。独特の腫瘍の免疫原性は腫瘍特異的抗原または腫瘍特異的抗原を発現する遺伝子を含むワクチンを使用した腫瘍特異的免疫応答の誘導を可能にしている。ワクチン接種の方法は抗原提示細胞を修飾して腫瘍関連抗原を提示する適合的細胞方法を含んでいた。癌治療のための別の免疫学的手法はサイトカインおよびケモカインの投与を包含し、これは、アジュバントとして、または免疫エフェクター細胞の増大と収集の能力に基づいた抗癌治療法として治療能力を有する。例えばHomey et al.,(2002)Nat Rev Immunol 2:175−84;Parmiani et al.,(2002)J Natl Cancer Inst 94:805−18;Bronte (2001)Curr Gene Ther 1:53−100;Fehniger et al.,(2002)Cytokine Growth Factor Rev 13:169−83参照。
(A1.非病原性ウイルスの抗腫瘍活性)
がん治療の免疫学的方法は治療剤としてのケモカインの使用を含む。ケモカインはその機能が(a)リンパ球の遊走および活性化を媒介すること;(2)血管形成を調節すること;および(c)免疫のホメオスタシスおよび二次的リンパ様臓器構造を維持すること、を含む相同性蛋白のファミリーである。Baggiolini et al.,(1997)Annu Rev Immunol 15:675−705;Jung&Littman(1999)Curr Opin Immunol 11:319−25;Homey et al.(2002)Nat Rev Immunol 2:175−84を参照できる。
感染性疾患を治療するための現在の方法は有害または望ましくない副作用を誘発する医薬の使用を包含する。更にまたワクチン接種を含む多くの有効な治療法は僅か単一の感染性実体またはそれに緊密に関連する実体に対してのみ特異的である。本発明者等は意外にも非病原性ウイルスが感染性疾患に強力に対抗できることを発見した。実施例17を参照できる。
本発明は更に、医薬組成物およびそれを使用する方法も提供する。本発明の非病原性ウイルスは安全で有効な抗腫瘍および/または抗感染性疾患および/またはアジュバント活性のために、本明細書に記載するとおり製造し、製剤される。
一部の実施形態においては、本発明の方法において使用される非病原性ウイルスは対象への投与の前に不活性化される。非病原性ウイルスは上記において定義したとおり、哺乳類宿主内で複製することができない。ウイルスをそのネイティブの宿主細胞において非複製性とする不活性化は別の安全性対策として実施できる。
本明細書に記載したとおり、非病原性ウイルスは生存ウイルス、不活性化ウイルス、ウイルス粒子、ウイルス閉鎖体またはウイルス成分またはこれらの組み合わせを含むことができる。非病原性ウイルスの対象における癌細胞へのデリバリーを促進するために、対象において抗癌および/または抗感染性疾患応答をもたらすために投与される組成物は(a)有効量の非病原性ウイルス、および(b)薬学的に許容される担体を含む。適宜、2種以上の担体を共に使用できる。
対象への本発明の組成物の投与のための適当な製剤は、抗酸化剤、緩衝剤、殺菌剤、抗細菌抗カビ剤(例えばパラベン、クロロブタノール、フェノール、アスコルビン酸、およびチメロサル)、製剤が意図するレシピエントの体液と等張となるための溶質(例えば糖類、塩類およびポリアルコール)、懸濁剤および濃厚化剤を含有できる水性および非水性の滅菌注射用溶液を包含する。製剤は単位用量または多用量の容器、例えば密封されたアンプルおよびバイアル中に提供することができ、そして、凍結状態で、または、使用直前に滅菌液体担体を添加することを必要とするのみの凍結乾燥状態で保存することができる。
インビボの抗腫瘍活性を予測することは困難で信頼性の低い場合が多い。本発明はインビボの抗腫瘍活性を予測するための方法を提供する。一部の実施形態においては、方法は腫瘍細胞および末梢血単核細胞に化合物を接触させること、および、腫瘍細胞の細胞死を測定することを含む。一部の実施形態においては、化合物は非病原性ウイルスまたは非病原性昆虫特異的ウイルスである。例えばアポトーシス、壊死、細胞生存性等の測定を含む何れかの方法を用いて細胞死を測定できる。使用できる腫瘍細胞は例えば、肺癌細胞(例えばA549細胞、3LL−HM細胞)、乳癌細胞(例えば4T1細胞;MT901細胞;MAT BIII細胞)、前立腺癌細胞、結腸癌細胞、皮膚癌細胞(例えばB16国職種細胞)、膵臓癌細胞、肝癌細胞、脳癌細胞、骨癌細胞(例えばMG−63細胞)、胃癌細胞、または食道癌細胞等である。
医薬品の一般への販売に先立って、政府機関、例えば食品医薬品局(FDA)はその当局または他の政府機関により許可された医薬品中に含有されていたものと同様の成分を医薬品が含むことを確認するためにこのような医薬品に対して厳重な品質管理を行うことを要求する場合が多い。このような品質管理は販売試験、即ち規制ガイドラインに医薬品が合致することを確認する試験を用いて行われることが多い。
ヒトCCL21をコードする完全長配列(ゲンバンクアクセッション番号NM002989)をバキュロウイルス転移ベクターpVL1392(Pharmingen,San Diego,Calfornia)にクローニングし、そして、入手元の推奨する方法を用いてBACULOGOLD(登録商標)WTゲノムDNA(Pharmingen,San Diego,Calfornia)と同時トランスフェクトした。この操作法により得られた組み換えバキュロウイルスをSf9昆虫細胞上のプラーク精製によりサブクローニングし、ヒトCCL21を発現する数種の単離体を得た。クローンを元のウイルスと比較した場合の例外的な発現特性があるものを選択した。このバキュロウイルス単離体の増幅を低多重度の感染(MOI)において実施し、高力価少継代の蛋白生産用保存株を得た。ヒトCCL21を発現するバキュロウイルスはBC422と命名した。追加の組み換えバキュロウイルスも同様に作成した。例えば細胞内Raf蛋白を発現するバキュロウイルスを作成し、BV762と命名した。
9〜11週齢のC57BL/6マウスを最低7日間馴化させた後に腫瘍細胞を接種した。マウスには右脇腹皮下に2x105個の早期継代(10継代未満)の3LL−HM腫瘍細胞を接種した。腫瘍の大きさは週当たり2回計測した。腫瘍が50〜100mm3に達した時点(典型的には腫瘍接種後7日間)で、マウスを無作為に群分けした。バキュロウイルス発現CCL21は腫瘍保有マウスに対し腫瘍内投与した。用量および投与の用法は腫瘍の抑制を旨適化するために変動させた。何れかの群の腫瘍体積が3000mm3(典型的には対照群のマウスにおける接種の33〜35日後)に達した時点で、マウスを屠殺した。
9〜11週齢のBalb/cマウスを最低7日間馴化させた後に腫瘍細胞を接種した。マウスには右脇腹皮下に2x105個の4T1細胞を接種した。腫瘍の大きさは週当たり2回計測した。腫瘍が50〜100mm3に達した時点(典型的には腫瘍接種後7日間)で、マウスを無作為に群分けした。バキュロウイルスは腫瘍保有マウスに対し腫瘍内投与した。用量は旨適有効用量を決定するために変動させた。何れかの群の腫瘍体積が3000mm3(典型的には対照群のマウスにおける接種の33〜35日後)に達した時点で、マウスを屠殺した。図2に示すとおり、CCL21の腫瘍内投与により4T1腫瘍の生育遅延が起こった。
マウス黒色腫細胞系統B16−BL6を用いて6〜8週齢のピンク皮膚雌性BDF−1マウス(Charles River Laboratories,Boston,Massachusetts)において皮下腫瘍を樹立する。皮膚腫瘍を作成するために、培地0.2ml中の106B16−BL6細胞を第0日に6〜8週齢の雌性BDF−1マウスの上背部領域に注射する。細胞の生存性は細胞注射の前後にトリパンブルー排除を用いて試験する。注射前に死滅した細胞の数は典型的には細胞の総数の10%以下である。第6日までに、腫瘍は典型的には直径5〜10mmとなる。
上記した通り腫瘍を保有するマウスを作成してバキュロウイルスを投与した。完全な腫瘍の減衰を示したマウスまたは完全な腫瘍の減衰を示さなかったマウスは、その腫瘍をバキュロウイルス採集投与の2日後に外科的に摘出し、腫瘍再攻撃に付した。マウスを200μlのケタミン/キシラジン混合物(リン酸塩干渉食塩水中10倍希釈した4:1ケタミン:キシラジン)の腹腔内投与により麻酔し、腫瘍を摘出し、創傷をホチキスで閉鎖した。腫瘍摘出の1〜4日後、元の腫瘍の部位以外の部位において2x105個の4T1細胞を皮下投与することによりマウスを再攻撃した。再攻撃腫瘍態勢を週当たり2回計測した。完全な腫瘍減衰を示したマウスでは、マウスを均等に2群に再分割した。1つの群は同じ腫瘍細胞で腹部の逆側をマウス当り1x105個で再攻撃した。もう1つの群は異なる同系の腫瘍細胞(B16F10)で左側の腹部をマウス当り1x105個で攻撃した。再攻撃部位における腫瘍の生育をモニタリングした。バキュロウイルスを投与したマウスは3LL−HM腫瘍細胞による再攻撃に耐性を示すことができたが、異なる同系の腫瘍細胞に対しては耐性ではなかった。
実施例3に記載したとおり腫瘍保有マウスを作成し、バキュロウイルスを投与した。対照群が肺転移による重度の症状を呈した場合にはマウスを屠殺した。典型的な指標は呼吸困難、脂性の体毛および体重減少とした。肺を採取し、Buoin溶液中に保存した。肺の転移の存在を測定した。図3はバキュロウイルス発現CCL21が腫瘍の転移を有意に抑制したことを示している。
実施例2〜3および5〜6において記載したバキュロウイルス発現hCCL21の抗腫瘍活性はCCL21よりはむしろバキュロウイルスに起因している。図6に示すとおり、バキュロウイルス発現CCL21の濾過調製物は3LL腫瘍モデルにおいて腫瘍の減衰を起こすのには不十分であった。何れの所定の試料におけるCCL21の濃度も濾過により変化していなかった。全てではないが一部のバキュロウイルス発現CCL21調製物は同様に非効果的であった。
バキュロウイルスを実施例1に記載の通り調製した。未感染のSf9細胞を対照として使用した。遠心分離後、上澄みおよび細胞ペレットの両方を回収した。細胞毒性試験を実施するために、試料を生育培地中1:11に稀釈し初期ウイルス力価5x106pfu/mlとした。
野生型バキュロウイルスを樹状細胞(DC)の培養物に添加したところ、活性化マーカーの細胞表面発現が増大したことにより明らかな通り、突然変異が誘導された。図11Aおよび11Bに示す通り、バキュロウイルスはマウス骨髄由来DCおよびヒト単球由来DCを活性化する。
バキュロウイルスおよび可溶性蛋白抗原(HIVp24)によるマウスの免疫化により頑健な抗原特異的CTL応答が誘導された。免疫化されたマウスの脾臓を第3回目の免疫化の2週間後に採取した。脾臓5個がかく試料中に含まれるように個々の脾臓をあわせた。免疫化マウスの脾細胞を植えるあたり5x106個の密度で24ウェルのディッシュ中で培養した。これらの細胞のうち、1x106個の細胞を(1)合成p7gペプチド、即ちHIV−1SF2p24gagのアミノ酸199〜208に相当するH−2Kd制限CTLエピトープ;および(2)pGagbペプチド、即ちHIV−1SF2p55gagのアミノ酸390〜398に相当するH−2Db制限CTLエピトープで感作した。ペプチドは37℃で1時間10μMの濃度で使用した。次に脾細胞を洗浄し、残りの4x106個の未投与細胞と共に培養した。脾細胞は100mM L−グルタミン(Giboco,Grand Island,New York)およびα−Mem(最小必須培地Alpha Medium、L−グルタミン、デオキシリボヌクレオシドまたはリボヌクレオシド添加)(1:1)と共に、脾細胞培地:RPMI1640(Sigma−Aldrich,St.Louis,Missouri)2ml中で塊状培養物として刺激し、10%熱不活性化ウシ胎児血清(Hyclone,Logan,Utah)を添加し、100U/mlペニシリン、10μg/mlストレプトマイシン、10ml/mlの100mMピルビン酸ナトリウムおよび50μM2−メルカプトエタノール中30分間56℃水浴中で不活性化した。更に、5%ラットT−StimIL2(ラットT−Stim:Collaborative Biomedical Products,Bedford,Massachusetts)をIL2原料として使用し、細胞を培養する直前に培地に添加した。
Trichoplusia ni(Tn)細胞の懸濁培養物2リットルにバキュロウイルスを感染させる。28℃で3日間インキュベートした後、感染細胞懸濁液を採取し、10分間800xgで遠心分離することにより透明化した。採取した培地中のバキュロウイルスの力価は、例えばKing&Possee(1992)The Baculovirus Expression System:A Laboratory Mannual,Chapman&Hall,Londonに記載の通り、Spodoptera frugiperda(Sf)細胞中のプラーク試験により測定した。
CCL21の種々のロットを共に以下に記載する細胞毒性試験およびインビボマウス腫瘍モデルにおいて試験した。細胞毒性活性係数は以下に記載するとおりである。活性および不活性のロットはインビボ実験の終了時に腫瘍サイズを分析することにより求めた。
誘導PBMC細胞毒性試験は付着標的細胞系統(A549またはMG−63細胞)に対抗するサイトカイン(IL−2またはβ−IFN)またはバキュロウイルス(BV)のような特定の活性化物質により誘導されるPBMCの細胞毒性(細胞増殖抑制)応答を測定するものであり、そして、エフェクター細胞(PBMC)と標的細胞の共同培養法を用いる。インキュベーション期間の後、標的細胞の生存性をアラマーブルー染色により定量する。試験に使用した成分は以下の通りである。

10%アラマーブルー(バイオソース インターナショナル)添加RPMI1640。
A549ヒト肺癌細胞をATCCから76継代で入手した(ATCC CCL185)。細胞を増殖させ、保存株を低継代で凍結保存した。A549細胞のマスター保存株は凍結し、液体窒素中に保存した。
A549細胞は付着単層として成育し、継代のためにはトリプシン溶液で脱着させなければならない。培地は各T−175フラスコから除去した。単層を10〜15mlのPBSで2回洗浄した。3〜5mlのSTVを各フラスコに添加した。次に各フラスコを3〜4分間、または細胞が脱着し始めるまでインキュベートした。フラスコを穏やかにたたいて細胞を脱着させた。生育培地5mlを各フラスコに添加し、細胞を穏やかにピペットで磨砕して単細胞懸濁液を調製した。細胞を新しい生育培地の入った50ml容の遠沈管に移し、穏やかに混合した。細胞混合物の種々の比率(1:5、1:10または1:20、それぞれ2、3および4日培養用)を37℃に加温された新しい生育培地40±/−5mlの入ったT175培養フラスコに分注した。
MG−63ヒト骨肉腫細胞系統はATCCから入手した(ATCC CRL−1427)。MG−63のマスター保存株は液体窒素中に凍結保存した。
作業ストック培養は3または4日おきに分割した。コンフルエントの単層を3日おきに1:6に、4日おきに1:8に分割した。
MG−63細胞は付着単層として成育し、継代のためにはトリプシン溶液で脱着させなければならない。培地は各T−175フラスコから吸引し、単層を10〜15mlのPBSで2回洗浄した。4±0.1mlのSTVを各フラスコに添加し、次に、フラスコを細胞が脱着するまで3〜5分間37℃±2℃、5±1%CO2でインキュベートした。フラスコを穏やかにたたいて細胞を脱着させた。
PBMCは同意を得たドナーから得た。
培地をA549またはMG−63作業培養用のT−175フラスコから採取した。単層を10〜15mlのPBSで2回洗浄した。3〜5mlのSTVを各フラスコに添加し、次に各フラスコを5分間、または細胞が脱着し始めるまでインキュベートした。フラスコを穏やかにたたいて細胞を脱着させた。生育培地5mlを各フラスコに添加し、細胞を穏やかにピペットで磨砕して単細胞懸濁液を調製した。細胞を50ml容の三角試験管に移し、更に生育培地25mlを添加した。試験管を反転させて細胞を混合し、細胞濃縮物を得た。細胞密度は細胞濃縮物として測定した。慨すれば、細胞懸濁液の1:3希釈物をZ1モデルのコールターカウンターを用いて計数した。A549細胞については、カウンターの下の閾値を8ミクロンに設定し、MG−63については8ミクロンとした。
(初期スクリーニング試験)
PBMC細胞毒性の誘導物質を検出する機会を向上させるために、資料をまず比較的高濃度に調製した。試料濃度は試料の活性に応じてその後の試験では低下させた。
CCL21の低蛋白濃度試料(3000μg/ml未満)をPBS(カルシウムおよびマグネシウム非含有)で400〜600μg/mlに希釈した。FBSおよびL−グルタミンを添加することにより試料を10%FBSおよび1%−L−グルタミン含有とした。試料をそのまま96穴プレートの第1のウェルにロードした(240μl容量)。1:2連続希釈を96穴移行プレート中で10%FBSおよび1%L−グルタミン含有PBS中に行った。
BV試料を生育/試験培地(GAM)中で1:2〜1:10に稀釈し、そしてGAM中に上記したとおり連続希釈した。
β−IFNまたはIL−2の最終バイアル試料を適切な希釈剤(食塩水または注射用水)中に希釈再調製した。希釈再調製したβ−IFNは0.25mg/ml=13.9μM、希釈再調製したIL−2は1.1mg/ml=64.7μMであった。各サイトカインの200,000IU/mlの作業保存溶液は更に生育試験培地(GAM)で希釈することにより作成した。β−IFNのロットIFN−01−001をGAMで1:35.75に希釈した。IL−2ロットMLAPM006をGAMで1:91.75に希釈した。その後の試験濃度への希釈はGAMを使用した。IL−2は試験用には2000IU/mlに稀釈し、β−IFNは試験用には2000IU/mlに希釈した。
濃度が3mg/mlより高値のCCL21試料はGAMで200〜600μg/mlの試験濃度まで希釈した。
希釈プレートの内容物を試験(細胞)プレートに移行させた。採集試料濃度は元の希釈プレートの1/2であった。プレートからプレートへの移行のプログラムのプロペットを移行に用いた。試験プレートはPBMCを準備しながらインキュベートした。
2個の同一の移行(希釈)プレートを樹立し、2細胞プレートに培地を移行させた。プレート1にはPBMC調製物50μlを添加して誘導細胞毒性測定を行った。2枚目のプレートには50μlのPBMC調製培地を添加して被験試料の直接細胞毒性を測定した。
漂白剤の1:10希釈物(3リットル)を調製した。血液に接触した全ての試験管およびピペットを一夜漂白した。ビーカーを肺賦形剤、全ての物品を鋭利物品または生物学的有害物質用の容器に廃棄した。50ml試験からキャップを除去した後に血液を取り扱った(血液によるキャップの汚染を防止するため)。血液は250ml容の使い捨てポリカーボネートフラスコに添加し、等量のHBSSで希釈した。50ml容のポリスチレン試験管当りフィコールプラーク溶液約17mlを添加し、30分後に血液を添加した。血液はフィコール層を撹乱することなくフィコールプラーク溶液の上面に重層させた。4mlの血液/HBSSを各3mlのフィコールプラークに対して添加した(即ち50ml容の試験管当り17mlフィコールおよび23ml血液/HBSSとした)。
試験で迅速に使用するために、細胞をPBMC調製培地に再懸濁した。細胞を更にPBMC調製培地中1:2に稀釈し、最終細胞密度1x106個/mlとした。細胞を標的細胞50μl/ウェル(50000PBMC/ウェル)で試験プレートに添加した。PBMCをマルチチャンネルピペットで添加した。試験プレートは3〜4日間37℃および5%CO2でインキュベートした。
凍結のために細胞を90%FBS(熱不活性化)+10%DMSO中に再懸濁し、10x106個/mlの密度で凍結した。細胞を2mlの滅菌クリオチューブに分注し、−70℃の冷凍庫中でクリオ凍結コンテナ中イソプロピルアルコール中に凍結した。最終細胞密度はコールターカウンターで確認した。
PBMCの凍結分を37℃の水浴中で急速に解凍した。細胞を三角遠沈管中13mlのRPMI1640中に再懸濁し、次に15〜20分間900rpm(100xg)で遠心分離した。上澄みを血清学的ピペットで除去し、細胞を13mlのPBMC調製培地中に再懸濁した(0.5%BSA添加RPMI)。細胞をコールターカウンターで計数して細胞密度を求め、PBMC調製培地中の細胞濃縮物の1:5希釈物を使用して計数した。
アラマーブルーを用いた試験プレートの染色
3〜4日間のインキュベーションの後、培地を試験プレートから吸引した。100μl/ウェルのアラマーブルー染色培地(0.5%BSA添加RPMI中10%アラマーブルー)を添加し、プレートを2〜3時間インキュベートした。応答は分光光度計(570nm〜630nmの吸光度)または蛍光分析(530nm励起、590nm発光)の何れかにより測定した。
試験活性はPBMC応答を示す試料ウェルで割った比較対照ウェルの応答として定義される。大部分の場合、比較対照ウェルは、通常は最高試料濃度のPBMCウェル(連続希釈の第1のウェル)と同様の試料濃度の試料に対する無PBMC対照ウェルであった。無PBMC対照プレートを使用するには不十分な試料しかなかった一部の場合においては、比較対照ウェルはPBMCの存在下または非存在下のPBS/生育培地とした。
バキュロウイルスの出発物質(CΔ3)は抗gp64で処理した。慨すれば、1〜500μgの精製マウス抗gp64モノクローナル抗体(クローン1.3A、提供者:Dr.Donald Jarvis,U.of WY)またはIg対照をコントロールされた容中の組み換えバキュロウイルスの種々の感染単位と混合した。ウイルスへの抗体の結合は約15時間暗所で室温で行った。試料は1〜2日間冷蔵庫に保管した後にプラークまたは細胞毒性の試験に付した。試料は上記したとおり細胞毒性試験において試験した。図15を参照できる。
バキュロウイルスの原料(CΔ3)はトリオキサレンおよびUV光による処理により不活性化した。バキュロウイルスをソラレンおよび紫外線により光不活性化した(Weightman,S.A.and Banks,M.J.Virol.Met.81:179−182(1999)。バキュロウイルスを含有する昆虫細胞培地を6穴の細胞培養プレート(Costar)に移行させた(3ml、約6E7pfu)。4,5’,8−トリメチルソラレン(トリオキサレン)の保存溶液をジメチルスルホキシド(DMSO)中2mg/mlに調製し、終濃度100μg/mlとなるように各ウェルに添加した。次にプレート上1.5cmにハンドヘルドのランプ(UVL−56型、UVP、Upland,CA)を置くことにより15分間培養ディッシュを紫外線(365nm)に曝露した。対照試料はソラレン非含有DMSO含有とし、ソラレンを含有する非照射対照も設けた。照射後、培地を10MWCO透析カートリッジ(Slidalyzer,Pierce)に移し、リン酸塩緩衝食塩水に対して透析した。全試料につきSf9細胞上のプラーク形成活性を試験した。試料を上記したとおり細胞毒性試験において試験し、結果を図16に示した。
A549標的細胞の未損傷の単層に3時間バキュロウイルスを投与し、次に生育試験培地で3回洗浄した。同時に、エフェクター細胞(PBMC)もまた3時間バキュロウイルスを投与し、3回洗浄した。バキュロウイルス投与および未投与の標的およびエフェクター細胞を以下の通りの組み合わせ、即ち、1)未投与標的細胞のみ;2)未投与標的細胞およびエフェクター細胞;3)バキュロウイルス投与標的細胞および未投与PBMC;および4)未投与標的細胞およびバキュロウイルス投与PBMCとした。
8〜10週齢のC57BL/6マウスを最低7日間馴化させた後、腫瘍細胞を接種した。マウスには右脇腹皮下に2x105個の早期継代(10継代未満)の3LL−HM腫瘍細胞を接種した。腫瘍の大きさは週当たり2回計測した。腫瘍が50〜100mm3に達した時点(典型的には腫瘍接種後7日間)で、マウスを無作為に群分けし、同日、バキュロウイルスの第1回目の用量を腫瘍内投与した。投与予定はqdx6日間とした。マウスは以下の4群、即ち、1)アルブミン陰性対象25μg/投薬(0.5mg/ml);2)生存ウイルス陽性対照(力価約1x107/ml);3)生存ウイルス陰性対象(力価約800/ml);および4)不活性化ウイルス(力価約800/ml)に割り付けた。腫瘍は4週間まで毎週2回測定した。腫瘍容が2500mm3に達するか、または以下の症状のいずれかが観察された場合には、マウスを屠殺した。症状は体重減少が20%超、腫瘍潰瘍領域が腫瘍面積の30%超または腫瘍面積の30%未満であるが開口部を有する場合、出血または漏出、重度の呼吸困難、または瀕死状態とした。
生存バキュロウイルスはネズミおよびヒトの細胞系統からインターフェロン(IFN)を誘導し、そして脳心筋炎ウイルス感染からのマウスのインビボ保護を誘導することがわかっている。しかしながら、例えばUVによりバキュロウイルスの不活性化は感染性およびIFN誘導活性の両方を排除する(Gronowski et al.,J Virology,Dec.1999,p.9944−9951 Vol.73,No.12)。
腫瘍細胞の抑制が直接的であるのみかどうか(即ち、各細胞が非病原性ウイルスにより特異的にターゲティングされなければならないか)を調べるために、感染腫瘍細胞および非感染腫瘍細胞を種々の比率で混合した。
