Abstract
Description

酵素は、それらが触媒する反応の一般的な種類によってグループに分類されている。トランスフェラーゼは、1つの基質から別の基質への基の転移を触媒し、キナーゼおよびホスファターゼを含む。タンパク質キナーゼは、アデノシン三リン酸(ATP)またはグアノシン三リン酸(GTP)などのドナーからペプチドまたはタンパク質などのアクセプタへホスホ部分を転移し、それぞれ、リン酸化ペプチドまたはタンパク質、およびアデノシン二リン酸(ADP)またはグアノシン二リン酸(GDP)をもたらす。タンパク質ホスファターゼは、リン酸基をホスホペプチドまたはリンタンパク質ドナーから水などのアクセプタへ転移する酵素である。
本開示の末尾に提示された請求の範囲によって規定されている本発明は、上記の問題の少なくとも一部を解決することを目的としている。例えば、本発明の1つの態様では、試料のトランスフェラーゼ活性を検出するための方法が提供される。この方法の好ましい実施形態では、試料は、基質およびリン酸基ドナーとリン酸基アクセプタの少なくとも1つと接触される。基質はレポーター化合物およびアミノ酸を含む。非リン酸化ペプチド基質を第1の速度で切断し、リン酸化ペプチド基質を第2の速度で切断するペプチダーゼが添加される。2つの速度の差は、トランスフェラーゼ活性の尺度である。次いで、レポーター化合物のアウトプットが検出される。

好ましい実施形態では、キナーゼ反応物は、バッファー、金属または二価陽イオン源、ヌクレオチド三リン酸(NTP)を含み、これらはリン酸ドナー、ペプチド基質、および、場合によっては、キナーゼの活性剤として作用しうる。バッファー、陽イオン、NTP、およびペプチド基質は、以下で説明されるように、検討中のタンパク質キナーゼに基づき選択される。必要に応じて、キナーゼの活性剤も添加することができる。試料は反応物に添加される。
ペプチド基質のアミド結合に作用する加水分解酵素であるペプチダーゼがペプチド基質に添加される。本発明において特に有用であるペプチダーゼは、エンドペプチダーゼ活性がない、または実質的にないものを含む。また、非リン酸化ペプチド基質を第1の速度で切断し、リン酸化ペプチド基質を第2の速度で切断するペプチダーゼを有することが好ましい。例えば、好ましいペプチダーゼは、リン酸化反応によって修飾されていないアミノ酸を結合するアミド結合を切断する場合、リン酸化反応によって修飾されているアミノ酸を結合するアミド結合を切断するよりも比較的高い活性を示す。同じ濃度のプロテアーゼで処理されたリン酸化ペプチドのものと比べた、プロテアーゼで処理された非リン酸化ペプチドから生成される蛍光の比のこの差異を、キナーゼの指標におけるように使用することができ、ペプチド基質がリン酸化されているかどうの判定が可能である。好ましいペプチダーゼは、ペプチド基質の非リン酸化アミノ酸を連続的に加水分解し、次いでリン酸化アミノ酸に達すると加水分解を劇的に減速させるものである。この加水分解の減速により、リン酸化アミノ酸の分子の大部分において放出されるレポーター化合物の不足が生じる。これによりバックグラウンド蛍光、または非リン酸化アミノ酸が存在する場合よりも相当に低い蛍光が生じる。リン酸化アミノ酸の部分的加水分解は以下で概略的に示されている。
使用されるレポーター化合物のアウトプットは、ペプチド基質のペプチダーゼ処理の後に検出される。蛍光発生レポーター化合物が使用される場合、蛍光はアウトプットとして使用されうる。蛍光光度計を用いて蛍光を検出することができる。単一チューブ機器である蛍光光度計またはマルチウェルプレート蛍光リーダーである蛍光光度計を用いて蛍光を検出することができる。例えば、クオーツキュベットを装備したフルオロログ−2スペクトロ蛍光光度計(SPEXインダストリーズ社(SPEX Industries, Inc.)、ニュージャージー州、エジソン)を単一チューブアッセイ用に用いることができる。Cytofluor(登録商標)IIマルチウェル蛍光プレートリーダー(パーセプティブバイオシステムズ社(PerSeptive Biosystems, Inc.)、マサチューセッツ州、フラミンガム)およびフルオロスキャンアセントCF(Fluoroscan Ascent CF)(ラブシステムズ(LabSystems)OY、フィンランド、ヘルシンキ)は、いずれも適切なフィルタが装備されているが、これらを用いて蛍光を検出することができる。蛍光単位または測定値を記録することができる。ローダミン110がレポーター化合物として使用される場合、ペプチダーゼ処理後にキナーゼ反応は好ましくは485nmでの励起および520〜530nmでの発光を読むことによって測定される。AMCが使用される場合、反応は好ましくは360nmで励起し、420nmでの発光を読むことによって測定される。
本発明の別の好ましい実施形態は、ホスファターゼ活性をスクリーニングするアッセイである。一般に、ホスファターゼ活性のスクリーニングは、キナーゼ活性のスクリーニングと同様に達成されるが、主な例外は、ホスファターゼの基質、通常はホスホペプチド基質をキナーゼにおけるペプチド基質の代わりに使用することである。キナーゼ活性アッセイとホスファターゼ活性アッセイとの間の他の差異は以下およびその後の実施例において述べられている。

本発明の別の実施形態は、キナーゼ反応における、またはこれに対する変化についてスクリーニングするアッセイである。変化として、キナーゼ反応の活性化または阻害が挙げられるが、これらに限定されない。このために、キナーゼの有望な活性剤または阻害剤である被験物質が、キナーゼといっしょにアッセイに添加される。アッセイは通常、バッファー、陽イオン、NTP、ペプチド基質、および0.05単位以上の問題のキナーゼを含む。
本発明のさらなる実施形態は、ホスファターゼ反応における、またはこれに対する変化についてスクリーニングするアッセイである。変化としては、ホスファターゼ反応の活性化または阻害が挙げられるが、これらに限定されない。このために、ホスファターゼの有望な阻害剤である被験物質が、ホスファターゼといっしょにアッセイに添加される。アッセイは通常、バッファー、陽イオン、ホスホペプチド基質、および0.1単位以上の問題のホスファターゼを含む。
本発明は、上述された方法を実施するためのキットにも関する。好ましい実施形態では、キットは、レポーター化合物を含む基質、トランスフェラーゼの酵素活性を維持するバッファー、リン酸ドナーとリン酸アクセプタの少なくとも1つ、および基質と適合するペプチダーゼを含む。検討中のトランスフェラーゼをキットに含めることができ、または使用者によって供給されうる。トランスフェラーゼは、キナーゼ、ホスファターゼ、または検討中の別のトランスフェラーゼでありうる。トランスフェラーゼがキナーゼである場合、基質は好ましくはリン酸基アクセプタとして作用するペプチド基質であり、リン酸ドナーは好ましくはキナーゼが使用可能であるNTPである。トランスフェラーゼがホスファターゼである場合、基質は好ましくはリン酸基ドナーとして作用するホスホペプチド基質である。他の成分、例えば、そのすべてが既述されている、検討中のトランスフェラーゼの活性剤、トランスフェラーゼのターミネーター、ペプチダーゼのターミネーター等なども含めることができる。好ましい実施形態では、トランスフェラーゼ活性のスクリーニングのためのキットも、制御反応のために使用されうるトランスフェラーゼを任意に含む。トランスフェラーゼを含むキットは、被験物質がトランスフェラーゼの活性を変化させるかどうかを判定するためにも使用されうる。例えば、キットは、被験物質が試験中のトランスフェラーゼを増強または阻害するかどうを判定するために使用されうる。
以下の実施例は、例示目的のみに提供されている。これらの実施例は、本明細書では、現在記述された発明をより完全に理解するのに単に役立てるために含まれている。これらの実施例は、任意の方法で本明細書で記述され請求された発明の範囲を限定することはない。
LRRASLG−(R110)−GLSARRLによるPKAアッセイ。プロメガ社(Promega Corp.)(ウィスコンシン州、マディソン)製のcAMP依存性タンパク質キナーゼ(PKA)の触媒サブユニットのキナーゼ活性について以下の反応成分を用いて96穴プレートにおいてトリプリケートで試験した。すなわち、40mM トリス−HCl、pH7.5、20mM MgCl2、0.lmg/ml ウシ血清アルブミン(BSA)、50μM ATP、および5μMのLRRASLG(配列番号:1)−R110−GLSARRL、「ビス−ケムプチド(kemptide)」としても知られるビス−ローダミンペプチドキナーゼ基質。最終反応体積は50μLであった。各反応に添加されるPKAの量は、0.001単位から1単位の範囲で、2倍の単位増分で滴定された。0単位による対照反応も実行された。すべてのキナーゼ反応物が20分間、室温でインキュベートされた。
阻害剤によるPKAアッセイ。PKAキナーゼの種々の阻害剤の効果を試験した。PKAの既知かつ特異的阻害剤(PKI−「タンパク質キナーゼ阻害剤」)、PKAの一般かつ非特異的阻害剤(スタウロスポリン((9S−(9a,10β,11β,13a)−2,3,10,11,12,13−ヘキサヒドロ−10−メトキシ−9−メチル−11−(メチルアミノ)−エポキシ−1H,9H−ジインドロ[1,2,3−gh:3’,2’,1’−1m]ピロロ{3,4−j]][1,7]ベンゾジアゾニン−1−オン)))、PKAの弱い不十分(poor)阻害剤(H7)(1−(5−イソキノリンスルホニル)−2−メチルピペラジン)、およびPKAを阻害しない化合物(U0126)(1,4−ジアミノ−2,3−ジシアノ−1,4−ビス−(2−アミノフェニルチオ)ブタジエン)をPKAのキナーゼ活性に対するその効果について試験した。キナーゼ反応およびアミノペプチダーゼ反応を、阻害剤を上昇濃度で含み、かつ0.5単位のPKAを使用したことを除き、実施例1に記載されたものと同様の条件下に実行した。いかなる阻害剤も含有しない対照を含めた。
LRRASLG−AMCによるcAMP依存性タンパク質キナーゼ(PKA)アッセイ。他の蛍光発生レポーター、例えば、7−アミノ−4−メチルクマリン(AMC)をPKAと使用するためにその安定性について試験した。AMCは遊離アミノ基によってアミド結合でペプチドLRRASLGに結合され、ペプチド基質ケムプチド−AMCを産生した。この基質をローダミン110修飾ペプチド基質とともに使用したものと同一であるアッセイ条件下に使用したが、例外として、40μMの濃度で基質を添加した。反応物を30分間、室温でインキュベートし、アミノペプチダーゼMを50mU/ウェルの最終濃度で添加し、60分間、室温でインキュベートした。反応は50μM ATPの存在下および非存在下に行い、PKAのホスホトランスフェラーゼ活性がATPを必要とすることを示した。蛍光データは、アクトニンの添加なしに60分で得られた。蛍光は、360nmの励起および420nmでの蛍光放出で検出された。
チロシンキナーゼアッセイ。チロシンキナーゼのキナーゼ活性は、リン酸化可能アミノ酸残基としてチロシンを含有するペプチド基質を用いて示された。タンパク質チロシンキナーゼのSrcファミリー、例えば、Fyn、Lyn A、Lyk、Src、Src N1などの一部の酵素のキナーゼ活性、および成長因子受容体チロシンキナーゼ(インスリン受容体)のキナーゼ活性について試験した。
STP−R−110によるPP2A活性。ホスファターゼ2A(PP2A)のホスファターゼ活性は、5μM ホスホペプチド基質ビス−STP−R110(RRAT(PO3)VA(配列番号:5)−R110−AV(PO3)TARR)、40mM トリス−HCl、pH7.5、および0.1mg/ml BSAを含有する50μl体積中で実行された。ホスファターゼ反応は、プロメガ(Promega)(ウィスコンシン州、マディソン)製の酵素セリン/スレオニンホスファターゼPP2Aを添加することによって開始された。各反応物に添加されるPP2Aの量は、0.0075ng(0.015m単位)から7.5ナノグラム(15m単位)の範囲で、1/2増分、および酵素を含有しない対照反応物で滴定した。ホスファターゼ反応は、96穴プレート中で室温に10分間、実行された。
STP−R110によるPP2A活性。PP2Aのホスファターゼ活性を、上記の実施例5において述べられた同じ条件を用いて試験したが、例外として、PP2Aを0.000001mUから0.01mUの範囲の濃度で試験し、酵素を含有しない対照反応をプラスした。反応は、10分間、96穴プレート中で室温で行い、オカダ酸(9,10−デエピチオ(Deepithio)−9,10−ジデヒドロアカンティホリシン)で終了させた。アミノペプチダーゼMの代わりに、25mUのアミノペプチダーゼIIを添加し、反応物を90分間、室温でインキュベートした。蛍光アウトプットを既述したように測定した。図8は、ホスファターゼ活性の増大により蛍光の増大が生じたことを示す。これらのデータは、任意のアミノペプチダーゼを本発明において使用できることを示す。
PTK5−R110によるCD45およびPTP−1Bアッセイ。ホスファターゼ反応は、組換えヒト受容体タンパク質チロシンホスファターゼであるCD45、または可溶性チロシンホスファターゼであるPTP−1Bのいずれかを用いて行われた。反応は、1μMのホスホペプチド基質ビス−PTK5p−R110(YIY(PO3)GAFKRRG(配列番号:6)−R110−GRRKFAG(PO3)YIY)、40mM トリス−HCl、pH7.5、0.1mg/ml BSAを含有する50μl体積中で行われた。反応は、96穴プレート中、室温で10分間、増大させた濃度のホスファターゼ(0〜2単位のCD45、または0〜0.025単位のPTP−1B)の存在下に行われた。
PTK5−R110によるPTP 1B活性。酵素チロシンホスファターゼPTP−1Bによるビス−PTK5p−R110の脱リン酸化反応に対する種々の阻害剤の効果を試験した。バナジン酸ナトリウム、(Na3VO4、PTP−1Bの特異的阻害剤)、およびスタウロスポリン(PTP−1BではなくPKAの既知の阻害剤)を試験した。阻害剤は、阻害剤ゼロから50μMまでの範囲の濃度で反応において含めた。ホスファターゼ反応は、PTB−1Bを添加することによって開始されたが、これは各反応物に25mU/ウェルで添加された。酵素を含有しない対照反応も実行した。ホスファターゼ反応は一般にPTP−1Bについて実施例7で述べられているように行われた。
LRRASLG−(ルシフェリン)によるPKAアッセイ。タンパク質キナーゼアッセイは、室温、96穴プレート中の50μl体積中、50μMでのペプチド基質LRRASLG−ルシフェリン、および実施例1において述べられたように反応バッファーにおけるタンパク質キナーゼAの異なる酵素濃度(0.001から1単位)で行われる。反応は、20分後に5分間、70℃で加熱不活性化によって終了される。反応混合物は室温に冷却され、40mM トリスHCl、pH7.5、および0.1mg/ml BSA中50mU/μlのアミノペプチダーゼMを含有する25μlの検出バッファーである。反応は、2.5μMの最終濃度でアクチノニンを添加することによって最適な終了の前にさらに60分間室温に維持される。25μlの安定グロー(steady glow)バッファー(プロメガ社(Promega Corp.))中100μg/mlでルシフェラーゼ(プロメガ社(Promega Corporation))が添加され、次いでルミネセンスが30分時点でオリオンプレート照度計、ベルトホールドディテクションシステムズ(Berthold Detection Systems)(ドイツ、フォルツハイム(Pforzheim))で測定される。酵素活性の発現は、実施例1に示されている蛍光標識基質で述べられているものと同様であること、すなわち、酵素濃度または活性の増大に応じてルミネセンスアウトプットの減少が予想される。
Claims (54)
- 試料のトランスフェラーゼ活性を検出するための方法であって、前記方法は、
(A)前記試料を、基質およびリン酸基ドナーとリン酸基アクセプタの少なくとも1つと接触させるものであって、前記基質がレポーター化合物およびアミノ酸を含み、
(B)非リン酸化ペプチド基質を第1の速度で切断し、リン酸化ペプチド基質を第2の速度で切断するペプチダーゼを添加し、および、
(C)前記レポーター化合物のアウトプットを検出すること、
を含む方法。 - 前記レポーター化合物が、前記ぺプチド基質のアミノ酸に結合されていない場合に比較すると前記ペプチド基質の少なくとも1つのアミノ酸に結合している場合に、異なるアウトプット特性を示す、請求項1に記載の方法。
- 前記レポーター化合物が蛍光発生化合物を含む、請求項1に記載の方法。
- 前記蛍光発生化合物が、ローダミン110およびアミノメチルクマリンの少なくとも1つから選択される、請求項2に記載の方法。
- 前記レポーター化合物が光発生化合物を含む、請求項1に記載の方法。
- 前記ペプチダーゼの添加が、前記基質の末端から前記基質を加水分解するペプチダーゼの添加を含む、請求項1に記載の方法。
- 前記ペプチダーゼの添加が、前記ペプチド基質のアミノ末端から前記基質を加水分解するペプチダーゼの添加を含む、請求項6に記載の方法。
- 前記ペプチダーゼの添加が、前記基質のカルボキシ末端から前記ペプチド基質を加水分解するペプチダーゼの添加を含む、請求項6に記載の方法。
- 検出される前記トランスフェラーゼがキナーゼを含む、請求項1に記載の方法。
- 前記基質がリン酸基アクセプタである、請求項9に記載の方法。
- 添加される前記基質がリン酸基ドナーである、請求項8に記載の方法。
- 検出される前記トランスフェラーゼがホスファターゼを含む、請求項1に記載の方法。
- 添加される前記基質がリン酸基ドナーである、請求項12に記載の方法。
- 添加される前記基質がリン酸基アクセプタである、請求項12に記載の方法。
- 前記基質を固体担体に結合させることをさらに含む、請求項1に記載の方法。

- 試料のトランスフェラーゼ活性を検出するための方法であって、前記方法は、
(A)前記試料を、基質およびリン酸ドナーとリン酸アクセプタの少なくとも1つと接触させるものであって、前記基質が蛍光発生レポーター化合物およびアミノ酸を含み、
(B)非リン酸化ペプチド基質を第1の速度で切断し、リン酸化ペプチド基質を第2の速度で切断するアミノペプチダーゼMを添加し、および、
(C)前記レポーター化合物のアウトプットを検出するスこと、
を含む方法。 - 検出される前記トランスフェラーゼがキナーゼを含む、請求項17に記載の方法。
- 検出される前記トランスフェラーゼがホスファターゼを含む、請求項17に記載の方法。
- トランスフェラーゼ反応における変化を検出するための方法であって、前記方法は、
(A)前記トランスフェラーゼが活性である条件下にレポーター化合物およびアミノ酸を含む基質に被験物質を接触させ、
(B)非リン酸化ペプチド基質を第1の速度で切断し、リン酸化ペプチド基質を第2の速度で切断するペプチダーゼで前記基質を切断し、および、
(C)前記レポーター化合物のアウトプットを検出すること、
を含む方法。 - 前記被験物質と接触されていない対照試料のアウトプットと比較されたアウトプットの変化が、前記トランスフェラーゼ反応の変化の尺度である、請求項20に記載の方法。
- 前記レポーター化合物が蛍光発生化合物を含む、請求項20に記載の方法。
- 前記レポーター化合物が光発生化合物を含む、請求項20に記載の方法。
- ペプチダーゼの添加が、前記基質の末端から前記基質を加水分解するペプチダーゼの添加を含む、請求項20に記載の方法。
- ペプチダーゼの添加が、前記基質のアミノ末端から前記基質を加水分解するペプチダーゼの添加を含む、請求項24に記載の方法。
- ペプチダーゼの添加が、前記ペプチド基質のカルボキシ末端から前記基質を加水分解するペプチダーゼの添加を含む、請求項24に記載の方法。
- トランスフェラーゼ反応の変化を検出するための方法であって、前記方法は、
(A)前記トランスフェラーゼが活性である条件下に蛍光発生レポーター化合物およびアミノ酸を含む基質に被験物質を接触させ、
(B)非リン酸化ペプチド基質を第1の速度で切断し、リン酸化ペプチド基質を第2の速度で切断するアミノペプチダーゼMで前記基質を切断し、および、
(C)前記レポーター化合物のアウトプットを検出すること、
を含む方法。 - トランスフェラーゼ反応の前記変化がキナーゼ反応の変化である、請求項27に記載の方法。
- トランスフェラーゼ反応の前記変化がホスファターゼ反応の変化である、請求項27に記載の方法。
- 試料のトランスフェラーゼ活性を検出する方法であって、
(A)それに共役したレポーター化合物を有する基質を提供し、
(B)前記試料を含有する溶液に多量の前記基質を添加し、
(C)トランスフェラーゼ活性が起こるのに十分な時間、前記試料が活性である条件下に、前記基質と前記試料をインキュベートし、
(D)前記試料を含有する前記溶液にペプチダーゼを添加し、
(E)前記レポーター化合物のアウトプットを検出すること、
を含む前記方法。 - ステップ(A)において、それに共役した蛍光分子を有する基質が提供される、請求項30に記載の方法。

- 前記レポーター化合物の第2の側面上で前記レポーター化合物に結合された第2のトランスフェラーゼ基質をさらに含むものであって、レポーター化合物がローダミン110であり、かつ前記第1および第2のトランスフェラーゼ基質がGKLSARRLAKKである、請求項44に記載のペプチド基質。
- 前記レポーター化合物の第2の側面上で前記レポーター化合物に結合された第2のトランスフェラーゼ基質をさらに含むものであって、レポーター化合物がローダミン110であり、かつ前記第1および第2のトランスフェラーゼ基質がGRVSAKRLAKKである、請求項44に記載のペプチド基質。
- 前記レポーター化合物の第2の側面上で前記レポーター化合物に結合された第2のトランスフェラーゼ基質をさらに含むものであって、レポーター化合物がローダミン110であり、かつ前記第1および第2のトランスフェラーゼ基質がGRRKFAGYIYである、請求項44に記載のペプチド基質。
- 前記レポーター化合物の第2の側面上で前記レポーター化合物に結合された第2のトランスフェラーゼ基質をさらに含むものであって、レポーター化合物がローダミン110であり、かつ前記第1および第2のトランスフェラーゼ基質がAV(PO3)TARRである、請求項44に記載のペプチド基質。
- 前記レポーター化合物の第2の側面上で前記レポーター化合物に結合された第2のトランスフェラーゼ基質をさらに含むものであって、レポーター化合物がローダミン110であり、かつ前記第1および第2のトランスフェラーゼ基質がGRRKFAG(PO3)YIYである、請求項44に記載のペプチド基質。
- 前記レポーター化合物が7−アミノ−4−メチルクマリンであり、かつ前記第1トランスフェラーゼ基質がLRRASLGである、請求項44に記載のペプチド基質。
