Abstract
Description
癌治療分野で最も必要とされているものの1つは、癌細胞の死滅を選択的に増強する手段を開発することである。癌患者に最も頻繁に使用されている治療方法は、放射線への曝露および/または化学療法であり、これは効果を得るために多数の正常な細胞を死滅させ、さらなる効果を得るためには、癌細胞よりも多くが死滅する。したがって、これらの治療薬の用量が制限される毒性は深刻な問題である。化学療法の副作用は多数あり、例えば、比較的良性の脱毛から骨髄毒性、認識機能障害、肝臓および腎臓の損傷、心臓の損傷、ならびに複数の他の器官系の損傷にまでわたる。非癌細胞に対する毒性が軽減された抗癌介入および正常な細胞の死滅が増大することなく非癌細胞も害し得る任意の抗癌治療による癌細胞の死滅を増強する試薬が必要である。これらのニーズを扱う組成物および方法を開示する。非癌細胞毒性が増大することなく癌細胞の成長を阻害する組成物および方法を開示する。非癌細胞に有毒な抗癌薬投与量を減少させても類似の量の抗癌活性が認められるので正常な細胞への全細胞毒性が減少する、任意の他の抗癌薬投与計画と組み合わせて作用する組成物および方法も開示する。

開示の組成物および組成物の使用方法には、カスパーゼインヒビターでもない抗癌薬の使用が含まれ得る。本明細書に開示される、カスパーゼインヒビターではなく抗癌薬である組成物を有するカスパーゼインヒビターである組成物の組み合わせは、望ましい抗癌活性を有し得る。開示の組成物中に任意の抗癌薬を含め、開示の方法で使用することができる。用語「非カスパーゼ抗癌薬」は、カスパーゼインヒビターが本明細書に開示の抗癌薬ではないことを示すことを意味せず、カスパーゼインヒビターは抗癌薬としても作用することができる。むしろ、非カスパーゼインヒビター抗癌薬は、カスパーゼインヒビターとして機能しないが抗癌活性を有する組成物をいう。典型的には、非カスパーゼインヒビター抗癌薬は、非癌細胞および癌細胞の死滅に影響を与えるが、本発明の組成物および方法で使用した非カスパーゼインヒビター抗癌薬はこの効果を有する必要はない。したがって、典型的には、非カスパーゼ抗癌薬は、非癌細胞に対して有毒である。
抗酸化剤はまた、抗腫瘍活性を有することが示されており、開示の混合物中で任意の組み合わせで使用することができる。
開示の組成物は、カスパーゼインヒビターを含む。本明細書で開示されるように、カスパーゼインヒビターは、単独で投与した場合、抗癌活性を有するが、カスパーゼインヒビターは他の抗癌治療(化学療法、放射線療法、手術、および他の癌治療との組み合わせが含まれる)と組み合わせて投与される有用な化合物でもある。カスパーゼインヒビターを、非カスパーゼインヒビター抗癌薬および/または抗酸化剤と組み合わせることができると理解される。カスパーゼインヒビター、非カスパーゼインヒビター抗癌薬、および抗酸化剤も任意の組み合わせで使用することができると理解される。例えば、異なる抗酸化剤の組み合わせを、一つまたは複数の異なるカスパーゼインヒビターと組み合わせて使用することができる。同様に、異なる非カスパーゼインヒビター抗癌薬の組み合わせを一つまたは複数の異なるカスパーゼインヒビターと組み合わせて使用することができる。さらに、本明細書に開示の組み合わせはカスパーゼインヒビターの任意の組み合わせを含み得ると理解される。
抗癌治療を必要とする被験体への投与のためのカスパーゼインヒビター処方物を本明細書に開示する。本明細書で考察されるように、任意の薬学的に許容される担体および処方物を使用することができる。本明細書で開示されるように、抗癌治療で使用するために、カスパーゼインヒビターは、カスパーゼ活性を阻害する同一濃度(例えば、アポトーシスを阻害する濃度)で活性を有する。
開示の組成物は、カスパーゼインヒビターと他の非カスパーゼインヒビター抗癌薬との混合物を含む。非カスパーゼインヒビター抗癌薬は、典型的には非癌細胞および癌細胞に対して細胞死を引き起こし得ると当技術分野において理解される。本発明の組成物の混合物は、抗癌活性レベルが得られる濃度の非カスパーゼインヒビター抗癌薬のみを含む処方物と比較して混合物がより低い濃度の非カスパーゼインヒビター抗癌薬を含む場合でさえ非カスパーゼインヒビター抗癌薬のみと類似の抗癌活性レベルを得ることができるので、非カスパーゼインヒビター抗癌薬の毒作用を、本発明の組成物の混合物によって減少させることができる。
抗酸化組成物を含む処方物もまた本明細書で開示される。癌治療で使用するためのカスパーゼインヒビターの処方物およびカスパーゼインヒビターおよび非カスパーゼインヒビター抗癌薬を有する処方物を、抗酸化剤と組み合わせ、抗癌投与計画のために投与することができる。抗酸化剤の添加により、カスパーゼインヒビターまたは非カスパーゼインヒビター抗癌薬のいずれかの量が減少し、これは本明細書で考察するように、有利であり得る。抗酸化剤はまた、死滅する腫瘍細胞量を増加させるために他の化学療法の有効性を増強することができる。抗酸化剤を含む処方物を、カスパーゼインヒビターと非カスパーゼインヒビター抗癌薬との組み合わせ処方物のための本明細書で考察したそれぞれおよび全ての方法で取り組むことができる。言い換えれば、処方物中の抗酸化剤量は、カスパーゼインヒビターを含む処方物ならびにカスパーゼインヒビターおよび非カスパーゼインヒビター抗癌薬を含む処方物の両方について単独で治療するために必要な比率に基づき得る。同様に、抗酸化剤量を、癌細胞死滅率および非癌細胞死滅率(または死滅しない比率)の調査によって取り組むことができる。これに関して任意の抗酸化剤を使用することができるが、現在、抗酸化剤の組み合わせにより有効性が増大することが示されている。
上記のように、薬学的に許容される担体中の組成物を、インビボで投与することもできる。「薬学的に許容される」は、生物学的または望ましくない材料(すなわち、いかなる望ましくない生物学的効果を生じないか、含まれる薬学的組成物のいかなる他の成分と有害な様式で相互作用せずに組成物と共に被験体に材料を投与することができる)を意味する。当業者に周知のように、担体は、本来、有効成分のいかなる分解も最小にし、被験体の任意の副作用も最小にするように選択される。
組成物(抗体が含まれる)を、薬学的に許容される担体と組み合わせて治療で使用することができる。
組成物投与のための投薬量範囲は、障害の症状が影響を受ける所望の効果を得るのに十分な範囲である。投薬量は、副作用(望ましくない交差反応およびアナフィラキシー反応など)を引き起こすほど大量であるべきではない。一般に、投薬量は、患者の年齢、病態、性別、および疾患の程度によって変化し、投薬量を当業者が決定することができる。任意の禁忌(counterindication)事象において各医師が投薬量を調整することができる。投薬量を変化させることができ、毎日1回またはそれ以上を1日または数日間投与することができる。
本明細書に開示の方法の実施において使用することができる試薬に適切なキットを本明細書に開示する。キットは、本明細書で考察したか、開示の方法の実施に必要であるか有利であると理解される任意の試薬または試薬の組み合わせを含み得る。例えば、キットは、カスパーゼインヒビターおよび非カスパーゼインヒビター抗癌薬を含む被験体に送達する準備ができている処方物を含み得る。
他で特記しない限り、特定の試薬または化合物について当業者に公知の任意の方法を使用して、本明細書に開示の組成物および開示の方法の実施に必要な組成物を作製することができる。
a)研究ツールとしての組成物の使用方法
研究ツールとして種々の方法で開示の組成物を使用することができる。例えば、開示の化合物(開示の組み合わせなど)を使用して、アポトーシス経路を研究することができる。
開示の組成物および処方物を使用して、異常な細胞増殖を阻害することができる。例えば、開示の組成物を使用して、癌細胞の細胞成長を阻害することができる。この開示の組成物を使用して、癌細胞増殖を阻害することができる。したがって、組成物を使用して、癌患者を治療することができる。任意の治療効果が有利であり、治療される患者は治癒する必要はないと理解される。組成物を使用して癌細胞を死滅させることができる。癌細胞の死滅は、細胞が分裂しないだけでなく、破壊されることを意味する。癌細胞の成長の阻害および癌細胞の死滅は共に有利であり得る。
本願を通して、種々の刊行物を参照する。本発明が属する分野の状況をより完全に説明するために、そのこれらの刊行物全体の開示が本出願に参考として組み入れられる。参考文献中の文で考察された参考文献に含まれる材料について、開示の参考文献はまた個別且つ詳細に参照として本明細書に組み入れられる。

種々の細胞を、種々のカスパーゼインヒビターの存在下でBCNU(カルムスチンとしても公知)に曝露した。このアルキル化剤は、中枢神経系癌の治療および一定のリンパ腫の治療で頻繁に使用されている。既知組成培地中での腫瘍細胞および正常なヒト脳前駆細胞の成長を、種々のアルキル化剤および/またはカスパーゼインヒビターの条件下でアッセイした。細胞を、BCNUに対する感受性の特徴づけの結果に依存して種々の投薬量のBCNUに曝露した。一般に、このアルキル化剤の細胞傷害効果からの防御を認識し、例えば、カスパーゼインヒビターおよびこの化合物の活性の条件に依存して抗癌活性の増大を認識することができる投薬量を使用した。
Claims (37)
- カスパーゼインヒビターおよび非カスパーゼインヒビター抗癌薬を含む組成物。
- カスパーゼインヒビターが、パン-カスパーゼインヒビターである、請求項1記載の組成物。
- カスパーゼインヒビターが、カスパーゼ9インヒビター、カスパーゼ-3インヒビター、カスパーゼ-8インヒビター、またはパン-カスパーゼインヒビターからなる群より選択される、請求項2記載の組成物。
- カスパーゼインヒビターがカスパーゼ産生を阻害する、請求項1記載の組成物。
- カスパーゼインヒビターがカスパーゼの活性化を阻害する、請求項1記載の組成物。
- カスパーゼインヒビターがカスパーゼのシグナル伝達経路を阻害する、請求項1記載の組成物。
- 非カスパーゼ抗癌薬が、アルキル化剤、DNA鎖破壊薬、代謝拮抗薬、トポイソメラーゼインヒビター、チューブリン相互作用薬、および分裂インヒビターからなる群より選択される、請求項1〜7のいずれか一項記載の組成物。
- 非カスパーゼインヒビター抗癌薬がBCNUである、請求項1〜7のいずれか一項記載の組成物。
