Abstract
【解決手段】yessotoxin(YTX)及びその類似物(YTXs)の細胞標的はホスホジエステラーゼ(PDEs)により活性化され、PDEs−YTX結合は測定可能なシグナルを生成する。PDEsを用いた親和性バイオセンサー又は蛍光によって結合を定量することができるので、当該バイオセンサーは生体分子の相互作用を検出し、そのPDEsとの相互作用のためにYTXの存在を検出することができる。

本方法は22〜37℃の温度で実行される。
22〜37℃の温度範囲でマイクロ滴定プレートで本方法を実行し、360ナノメーターの励起波長と460ナノメーターの放射波長で蛍光を測定する。

Claims (7)
- 細胞ホスホジエステラーゼ中に毒素によって生じる活性化に基づいて魚介類のyessotoxins(YTXs)を検出するための定量方法であって、さらに、yessotoxin(YTX)及びその化学的な類似物(YTXs)の活性化作用の、YTXs又は同様の活性の化合物を検出するための定量方法でのホスホジエステラーゼ活性についての使用を特徴とする方法。
- PDEsとのYTX活性化に基づいて請求項1に記載された魚介類のYTXsを検出するための方法であって、20〜37℃の温度範囲でエバネセンス効果によって定量可能な分子間相互作用を生成することができる平坦又はマトリクスの支持体表面に結合するリガンドとしてPDEsを用いる親和性バイオセンサーの使用であり、レセプターとして機能するYTXsが添加され、見かけのYTX−PDE結合率(Rap)が得られる擬似−一次速度式にこの結合を当てはめることを特徴とする方法。
- 請求項2に記載された魚介類のYTXsを検出するための方法であって、既知の濃度のYTXsの見かけの結合率の決定、及び見かけの結合率が既知である魚介類抽出物のテストサンプルでYTX濃度を計算する較正線の準備であることを特徴とする方法。
- 請求項1に記載された魚介類のYTXsを検出するための方法であって、PDE活性蛍光による検出であり、類似のcAMP蛍光基質を用い、既知の濃度のYTXで得られる前記蛍光基質の破壊率で準備された較正線から魚介類サンプルのYTX濃度を計算することを特徴とする方法。
- 請求項4に記載された魚介類のYTXsを検出するための方法であって、前記検出が基質としてアントラニロイル−cAMPを用いるPDE活性蛍光によることを特徴とする方法。
- 請求項4に記載された魚介類のYTXsを検出するための方法であって、YTXがPDEsに結合するときに生じる偏光蛍光、その結果偏光ユニットの強度の変化を決定することを特徴とする方法。
- 2、3又は6に記載された魚介類のYTXsを検出するための方法の、天然又は合成化学化合物でのYTXに等価な活性(PDE活性化)を検出するための使用。
