Abstract
【解決手段】 非感染性レトロウィルス様粒子は、env遺伝子産物、pol遺伝子産物およびgag遺伝子産物のアセンブリを有し、非レトロウィルス性または非HIVレトロウィルス性である抗原マーカーを有する。該マーカーは、抗原的に活性な挿入部位のgag遺伝子産物に挿入されたエピトープを有するアミノ酸配列、あるいは、内因性アンカー形成機能部分に置換するenv遺伝子産物に操作的に連結された抗原性アンカー配列である。さらには、env遺伝子産物の免疫優性領域の修飾により、実質的に免疫優性領域の認識を防止する。抗原マーカーに特異的な抗体の存在は、抗レトロウィルス抗体を有する抗血清が、非感染性レトロウィルス様粒子による免疫感作よって生じたことを証する。

Description
本願は、1994年8月15日出願の米国特許出願08/290105号の一部継続出願である。
ヒト免疫不全ウィルスはヒトレトロウィルスであり、後天性免疫不全症候群(AIDS)の病原体である。1981年に米国でAIDSが最初に報告されて以来、194000名を超える人がAIDSによって死亡し、米国のみで330000例を超えるHIV感染が報告されている。世界的には、1400万人を超える人々がHIVに感染していると推定されている。
本発明は、HIVまたは別のレトロウィルス、特にヒトレトロウィルスによる感染と免疫原性製剤による免疫感作との間を識別する能力に関するものである。
(a)env遺伝子産物、(b)pol遺伝子産物、(c)gag遺伝子産物のアセンブリ、ならびに
(d)非レトロウィルス性または非HIVレトロウィルス性である少なくとも1個の抗原マーカーを含んでなる非感染性レトロウィルス様粒子を提供する。
(a)内因性アンカー形成機能部分が、レトロウィルス様粒子に当該env遺伝子産物をアンカーする機能を果すように、当該env遺伝子産物に対して連結しされている異種のアンカー配列によって置換されてなる、修飾env遺伝子産物、
(b)pol遺伝子産物、
(c)gag遺伝子産物のアセンブリを有してなる非感染性レトロウィルス様粒子が提供される。
アミノ酸配列WILWISFAISCFLLCVVLLGFIMW(配列番号2)またはその配列を認識する抗体を産生する能力を有するその配列の一部、変化体もしくは突然変異体、
アミノ酸配列STVASSLALAIMIAGLSFWMCSNGSLQ(配列番号3)またはその配列を認識する抗体を産生する能力を有するその配列の一部、変化体もしくは突然変異体、あるいは
アミノ酸配列WILWISFAISCFLLCVVCWGSSCGPAKKATLGATFAFDSKEEWCREKKEQWE(配列番号4)またはその配列を認識する抗体を産生する能力を有するその配列の一部、変化体もしくは突然変異体などがあり得る。
(a)検体を本発明で提供される非感染性レトロウィルス様粒子と接触させて、非感染性レトロウィルス様粒子および該粒子との特異的反応性を有する検体中に存在するそのような抗体を有してなる錯体を生成する段階、ならびに
(b)該錯体の生成を確認する段階を有してなる方法が提供される。
(a)宿主を本発明で提供される免疫原性組成物で免疫感作してレトロウィルス性抗原特異的抗体を生成する段階、
(b)検体を前記レトロウィルス性抗原特異的抗体と接触させて、検体中のレトロウィルス抗原と前記レトロウィルス性抗原特異的抗体とを有してなる錯体を生成する段階、および
(c)該錯体の生成を確認する段階を有してなる方法が提供される。
(a)本発明で提供されるそのような少なくとも1個のレトロウィルス性抗原特異的抗体、
(b)検体と該少なくとも1個の抗体とを接触させて検体中のレトロウィルス性抗原とレトロウィルス性抗原特異的抗体とを有してなる錯体を生成する手段、および
(c)該錯体の生成を確認する手段を有してなる、検体中のレトロウィルス性抗原の存在を検出するための診断キットを提供するものである。
−非感染性および非複製性となっている、天然コンホメーションにgag、polおよびenv遺伝子産物を有してなる免疫原性レトロウィルス様粒子、および
−ビルレントレトロウィルスと免疫的に識別可能な免疫原性レトロウィルス様粒子などがある。
図面を参照しながら、以下の説明により、本発明についての理解を深めることができる。
本発明の各種実施態様が予防接種、診断、HIV感染治療、および免疫試薬生成の分野において多くの利用場面を有することは当業者には明らかである。そのような用途についての考察を以下に行うが、本発明はその記載内容に限定されるものではない。
ワクチンの製造および用途
本発明による免疫原性製剤が免疫応答を誘発し得ることは上記で示した。従って、本発明の用途で可能性のあるものとしては、AIDSおよびAIDS関連状態を含めたレトロウィルス疾患に対するワクチンの基礎としてのものである。そこで別の態様において本発明は、本発明による免疫原性組成物を有してなるAIDSおよびAIDS関連状態に対するワクチンを提供するものである。
イムノアッセイ
本発明のレトロウィルス様粒子は、免疫原として、すなわち、酵素結合性免疫吸着剤アッセイ(ELISA)、RIAおよび他の非酵素結合性抗体結合アッセイなどのイムノアッセイ、あるいは抗レトロウィルス(例:HIV)HIV抗体およびレトロウィルス抗原(例:HIV)の検出に関して当業界で公知の方法における抗原として有用である。ELISAアッセイでは、レトロウィルス様粒子を、ポリスチレン微量力価測定プレート孔などの蛋白を結合する能力を有する表面などの所定の表面に固定化する。洗浄によって吸着が不完全なレトロウィルス様粒子を除去した後、ウシ血清アルブミン(BSA)またはカゼインの溶液などの被験検体に関して抗原的に中性であることが知られている非特異的蛋白を所定の表面に結合させることができる。それにより、固定化表面上の非特異的吸着部位の遮断ができ、従ってその表面への抗血清の非特異的結合によって生じるバックグラウンドが低下する。
他の用途
本発明の基礎となるレトロウィルス様粒子に結合する分子、特には抗体、抗体関連分子およびそれらの構造的類縁体も、AIDSおよび関連状態の治療および診断に試薬として有用である可能性がある。
生物寄託
本明細書において記載・言及される本発明の各種態様によるレトロウィルス様粒子をコードするある種のプラスミドは、ブダペスト条約に従い、本願出願に先だって、アメリカン・タイプ・カルチャー・コレクション(American Type Culture Collection(ATCC),12301 Parklawn Drive,Rockville,Maryland,USA,20852)に寄託した。寄託プラスミドのサンプルは、本米国特許出願に基づく特許交付時に公表される。寄託した実施態様は、単に本発明の例とすることを意図したものであることから、本願で説明され特許請求される発明は、寄託したプラスミドに限定されるものではない。本願に記載のものと類似または等価なレトロウィルス様粒子をコードする等価もしくは類似のプラスミドは、本発明の範囲に含まれる。
本開示および以下の実施例に明瞭には記載されていないが使用した分子遺伝学、蛋白生化学および免疫学の方法は、科学文献中に数多く報告されており、当業者の能力の範囲内のものである。
本実施例は、プラスミドpMTHIVBRUの構造を説明するものである。
上流プライマーは5’−GGACTAGTACCCTTCAGGAACAAATAGGATGGATGACAAATAATCCACCTATCCCAGTAGGAG−3’(配列番号15)の配列を持ち、5’末端にSpeI部位を有するHIV−1LAIのヌクレオチド1507〜1567を有し(ヌクレオチドの番号付けは、ウエイン−ホブソンら(Wain−Hobson;1985)による)、
下流のプライマーは5’−CTCGGGCCCTGCAATTTCTGGCTATGTGCCCTTCTTTGCCACTATTGAAACTCTTAACAATC−3’(配列番号16)の配列を持ち、5’末端にApaI部位を持つヌクレオチド2011〜1953の逆補体である。下流側プライマーでは、ヌクレオチド1963および1972(Wain Hobson et al.,1985; Myers et al.,1990)の逆補体を表す2個のアデノシン残基がチミジンに変わって、gag遺伝子産物のアミノ酸位置392および395の2個のシステインが2個のセリンによって置換されている(図1)。これら2つのプライマーを用いて、pMTHIVのSpeI−ApaI DNA断片(ヌクレオチド1507〜2008)(Rovinski et al.,1992)を増幅し、それを鋳型として用いた。PCR増幅SpeI−ApaI断片をアガロースゲル電気泳動によって精製し、制限酵素SpeIおよびApaIで消化させた。この断片を用いて、pMTHIVd25の相当する断片を置換した(Rovinski et al,1992)。得られたプラスミドはpMTHIV−Aと称し、RNAパッケージング配列欠失およびCys−Hisボックス突然変異の両方を有している。
AGCGGCCGCAC−3’(1本鎖のみを示した−配列番号17)と連結し
た。得られたプラスミドは、pMTHIVBRUと名付けた。
本実施例は、TMVからの抗原性エピトープを有するHIV様粒子をコードするプラスミドの構成について説明する。
本実施例は、抗原的に標識したHIV様粒子の免疫原性および免疫反応性を説明するものである。
本実施例は、gp41の免疫優性領域に対する突然変異によって抗体的に標識されたHIV様粒子の形成について説明するものである。
本開示をまとめると、本発明は、例えば予防接種すなわちレトロウィルス特異的抗血清の形成に有用な免疫原性製剤ならびに診断法・診断キットにおける抗原としてのある種の非感染性非複製性レトロウィルス様粒子およびそれをコードする核酸分子を提供する。レトロウィルス様粒子は、polおよび/またはgag遺伝子産物に対する修飾によって非感染性になったと考えられる。特定のレトロウィルス様粒子は、非レトロウィルス性非抗原マーカーを有する。本発明の範囲内での改良は可能である。

- (a)env遺伝子産物、
(b)pol遺伝子産物、
(c)gag遺伝子産物、ならびに
(d)非レトロウィルス性または非HIVレトロウィルス性である少なくとも1個の抗原マーカーのアセンブリを有する非感染性レトロウィルス様粒子。 - 前記少なくとも1個の抗原マーカーが約5〜約100個のアミノ酸残基を有する請求項1記載のレトロウィルス様粒子。
- 前記少なくとも1個の抗原マーカーが約10〜約75個のアミノ酸残基を有する請求項2記載のレトロウィルス様粒子。

- 検体中のレトロウィルス抗原と特異的に反応する抗体の存在を測定する方法であって、 (a)請求項1または14に記載の非感染性レトロウィルス様粒子と検体とを接触させて、非感染性レトロウィルス様粒子および該粒子と特異的に反応する検体中に存在する前記抗体とを有する錯体を形成する段階、ならびに (b)前記錯体の形成を確認する段階を有する方法。
- 検体中のレトロウィルス抗原の存在を確認する方法であって、
(a)請求項70記載の免疫原性組成物で宿主を免疫感作して、レトロウィルス抗原特異的抗体を産生する段階、
(b)レトロウィルス抗原特異的抗体と検体とを接触させて、検体中のレトロウィルス抗原とレトロウィルス抗原特異的抗体を有してなる錯体を形成する段階、ならびに
(c)前記錯体の形成を確認する段階を有してなる方法。 - 検体中のレトロウィルス抗原の存在を検出する診断キットであって、
(a)請求項78で産生されるような、少なくとも1個のレトロウィルス抗原特異的抗体、
(b)前記少なくとも1個の抗体を検体と接触させて、検体中のレトロウィルス抗原および前記レトロウィルス抗原特異的抗体を有してなる錯体を形成する手段、ならびに
(c)前記錯体の形成を確認する手段を有してなるキット。 - 請求項70に記載の免疫原性組成物を用いる免疫感作によって生じる抗血清を確認する方法であって、前記抗血清中の前記抗原マーカーに特異的な抗体を検出する段階を有してなる方法。
