Abstract
Description
「によってコードされる」は、ポリペプチド配列をコードする核酸配列を表わし、但しそのポリペプチド配列又はその部分は、該核酸配列によってコードされるポリペプチドからの少なくとも3個から5個のアミノ酸、より好ましくは少なくとも8個から10個のアミノ酸、さらに一層好ましくは少なくとも5個から20個のアミノ酸を含む。また、その配列によってコードされるポリペプチドで免疫学的に特定しうるポリペプチド配列も包含される。従って、「ポリペプチド」、「タンパク質」又は「アミノ酸」配列は、マンマグロビン遺伝子(配列番号1)、BU101遺伝子(配列番号2)、α’又はβ’遺伝子(すなわち、そのコードされるポリペプチドが多量体ポリペプチド複合体を形成する遺伝子)によってコードされるアミノ酸配列と少なくとも約50%の同一性、好ましくは約60%の同一性、より好ましくは約75%から85%の同一性、最も好ましくは約90%から95%又はそれ以上の同一性を有する。さらに、マンマグロビン遺伝子(配列番号1)、BU101遺伝子(配列番号2)、α’遺伝子又はβ’遺伝子(すなわち、そのコードされるポリペプチドが多量体ポリペプチド複合体を形成する遺伝子)によってコードされる「ポリペプチド」、「タンパク質」又は「アミノ酸」配列は、マンマグロビン遺伝子(配列番号1)、BU101遺伝子(配列番号2)、α’遺伝子、又はβ’遺伝子のポリペプチド又はアミノ酸配列と少なくとも約60%の類似性、好ましくは約75%の類似性、より好ましくは約85%の類似性、最も好ましくは約95%又はそれ以上の類似性を有していてもよい。
本発明は、少なくとも1コピーのBU101ポリペプチド(配列番号6)及び少なくとも1コピーのマンマグロビンポリペプチド(配列番号5)を含むが、1又はそれ以上の未知のポリペプチドも同時に含みうる、多量体ポリペプチド複合体又は抗原、及びこの多量体ポリペプチド複合体に特異的な抗体などの試薬を提供する。本発明のポリペプチド配列又は抗体は、乳癌のような、乳房の疾患及び状態を検出する、診断する、病期分類する、監視する、予後を判定する、インビボで画像化する、予防する又は治療する、若しくは素因を判定することを導く情報を提供するために使用しうる。
本発明は、多量体ポリペプチド複合体に特異的に結合する少なくとも1つの化合物を特定するために、その多量体ポリペプチド複合体又はそのフラグメントへの特異的結合に関して多数の化合物をスクリーニングする方法を提供する。そのような方法は、少なくとも1つの化合物を提供すること;多量体ポリペプチド複合体を、結合のために適切な条件下で十分な時間、各々の化合物と一緒にすること;及び各々の化合物への多量体ポリペプチド複合体の結合を検出すること、の段階を含む。
本発明は、BU101ポリペプチド(配列番号6)、マンマグロビンポリペプチド(配列番号5)、及び未知のα’及び/又はβ’ポリペプチドの同時発現のための宿主細胞及び発現ベクター、及びそれらがコードする多量体ポリペプチド複合体の生産のための方法を提供する。そのような方法は、BU101ポリペプチド(配列番号6)、マンマグロビンポリペプチド(配列番号5)、及び未知のα’及び/又はβ’ポリペプチドの発現に適した条件下で宿主細胞を培養すること、及びその細胞培養から多量体ポリペプチド及び/又は多量体ポリペプチド複合体を回収することを含む。
そのフラグメント、誘導体及び類似体を含む、ポリペプチド、多量体ポリペプチド複合体、又はそのようなポリペプチド又は多量体ポリペプチド複合体を発現する細胞は、乳房組織に対する抗体を検出するための様々なアッセイにおいて利用でき、その多くをここで述べる。それらはまた、抗体を産生するための免疫原としても使用できる。これらの抗体は、例えば、ポリクローナル又はモノクローナル抗体、キメラ、単鎖及びヒト化抗体、ならびにFabフラグメント、又はFab発現ライブラリーの産物でありうる。そのような抗体及びフラグメントの生産のためにこの技術分野で既知の様々な手順が使用しうる。
本発明の抗体はインビボで使用することができる。すなわち、それらは、診断又は治療用途のために乳房の疾患を有する又は有することが疑われる患者に注入することができる。インビボ診断のための抗体の使用はこの技術分野において周知である。Sumerdonら、Nucl.Med.Biol 17:247−254(1990)は、インジウム−111を標識として使用する、腫瘍を発現する癌胎児性抗原(CEA)の放射線免疫シンチフォトグラフィー画像化のための最適化抗体−キレート化剤を記述している。Griffinら、J Clin Onc 9:631−640(1991)は、再発性結腸直腸癌を有することが疑われる患者において癌を検出する場合のこの物質の使用を記述している。磁気共鳴画像のための、常磁性イオンを標識とする同様の物質の使用は、この技術分野において既知である(R.B.Lauffer,Magnetic Resonance in Medicine 22:339−342(1991))。多量体ポリペプチド抗原に対して産生される抗体は、乳癌などの乳房の疾患を有することが疑われる患者に、患者の疾患の状態を診断する又は病期分類することを目的として注入できることが想定される。使用する標識は、選択する画像化様式に依存する。インジウム−111、テクネチウム−99m又はヨウ素−131などの放射性標識は、プレーナースキャン(planar scan)又はシングルフォトンエミッション断層撮影法(SPECT)のために使用できる。フッ素−19などの陽電子放射標識も、陽電子放射断層撮影法(PET)のために使用できる。MRIのためには、ガドリニウム(III)又はマンガン(II)などの常磁性イオンが使用できる。乳房内又は乳房の外部における標識の局在は、疾患の拡大の測定を可能にしうる。乳房内の標識の量は、乳癌の存在又は不在の判定を可能にしうる。
(実施例)
発現配列タグ(EST)又は転写産物像のライブラリー比較。cDNAクローンインサートの部分配列、いわゆる「発現配列タグ」(EST)を、乳癌組織、乳房非腫瘍組織、及び腫瘍及び非腫瘍の多数の他の組織から作製したcDNAから誘導し、遺伝子転写産物像としてデータベース(LIFESEQ(商標)データベース、Incyte Pharmaceuticals,Palo,Alto,CAより入手可能)に登録した。国際特許公開公報第WO95/20681号参照。(転写産物像は、所与の組織ライブラリーの中に表示される遺伝子の各々についてのESTの数のリストである。ESTの相互配列重複の共有領域はクラスターに分類される。クラスターには、代表的5’ESTからのクローン番号が割り当てられる。しばしば、対象クラスターは、そのコンセンサス配列を自動クラスター化の判定基準に合致しなかった他のESTの配列と比較することによって延長できる。すべての使用可能なクラスターと1個のESTの整列は、そこからコンセンサス配列が導かれるコンティグを表わす。)次に転写産物像を、主として乳房組織ライブラリーを代表するEST配列を特定するために評価した。これらの標的クローンを、標的ライブラリーにおけるそれらの存在率(発生率)及びバックグラウンドライブラリーにおけるそれらに不在率に従って等級付けた。低いバックグラウンド発生率とより高い存在率を有するクローンには、より高い試験優先権が与えられた。
A.ポリクローナル抗血清の生産
1.多量体ポリペプチド複合体を免疫原として使用する動物免疫。体重2kg以上の雌性白色ニュージーランドウサギを、ポリクローナル抗血清を惹起するために使用した。初回免疫の1週間前に動物から血液5mlから10mlを採取して、非免疫採血前試料として使用した。
1.多量体ポリペプチド複合体を免疫原として使用する免疫プロトコール。マウスにおけるモノクローナル抗体産生のための多量体ポリペプチド複合体の量が、ウサギにおいてポリクローナル抗血清を生産するために使用する量の10分の1であることを除いて、実施例7で述べるように調製した免疫原を用いてマウスを免疫する。従って、一次免疫原は、CFA乳液0.1ml中の多量体ポリペプチド複合体100μgから成る。ブースター免疫に使用する免疫原は、IFA0.1ml中の多量体ポリペプチド複合体50μgから成る。モノクローナル抗体の作製のためのハイブリドーマを生成し、標準手法を用いてスクリーニングする。モノクローナル抗体産生のために使用する方法は、KohlerとMilstein,Nature 256:494(1975)において詳述され、J.G.R.Hurrel編集、Monoclonal Hybridoma Antibodies:Techniques and Applications,CRC Press,Inc.,Boca Raton,FL(1982)に総説されているような、この技術分野で既知の手順に従う。KohlerとMilsteinの方法に基づくモノクローナル抗体産生のもう1つの方法は、参照してここに組み込まれる、L.T.Mimmsら、Virology 176:604−619(1990)の方法である。
A.マイクロタイタープレート直接検出EIA
組換えポリペプチド複合体(本出願の実施例7又は1998年12月18日出願の米国特許願通し番号第09/215,818号(参照してここに組み込まれる)の実施例2に従って生成される)ポリクローナル及び/又はモノクローナル抗血清(BU101又はマンマグロビンのいずれかに対する)の免疫反応性をマイクロタイタープレートEIAによって測定した。
に基づいて見掛け解離定数[Kd(app)]を導いた。曲線への適合の間、[Ag−Ab]を、所与の濃度のAbでOD405nmのバックグラウンド控除値に置き換えた。Kd(app)に対応するKd、及び[Ag−Ab]maxに対応する[OD405nm]maxの両方を、適合パラメータとして処理した。ソフトウエアプログラムOriginを曲線への適合のために使用した。図1は、多量体ポリペプチド複合体を認識したモノクローナル抗体の3つの結合曲線を示す。Kd(app)値は、この複合体に対するモノクローナル抗体の相対的親和性の測定である。表2においてKd(app)値をモノクローナル抗体の各々について列記する。
簡単に述べると、多量体ポリペプチド抗原を含むことが疑われる試料を、抗原/抗体複合体を形成するために、抗BU101、抗マンマグロビン、抗α’ポリペプチド、抗β’ポリペプチド、又は抗多量体ポリペプチドの抗体で被覆したマイクロタイターウエルの何らかの組合せの存在下でインキュベートする。次にマイクロタイターウエルを洗い、シグナル生成化合物(すなわちアルカリホスファターゼ又はホースラディッシュペルオキシダーゼなどの酵素)に複合した抗体を含む指示試薬をマイクロタイターウエル上の抗原/抗体複合体に加えて、インキュベートする。マイクロタイターウエルを洗い、シグナル生成化合物と反応して測定可能なシグナルを生じる基質(例えば、それぞれ4−メチルウンベリフェリルリン酸塩(MUP)又はOPD/ペプチドルオキシド)を加えることによって結合抗体/抗原/抗体複合体を検出する。陰性対照によって生じるシグナルと比較して、被験試料中の高いシグナルは、多量体ポリペプチド抗原の存在を検出する。被験試料における多量体ポリペプチド抗原の存在は、乳癌などの乳房の疾患又は状態の診断の指標である。
競合結合アッセイは、標識ポリペプチド又はタンパク質複合体を抗ポリペプチド抗体で被覆したマイクロタイターウエルと接触させたとき測定可能なシグナルを生じる、標識ポリペプチド又はタンパク質複合体を使用する。その標識ポリペプチドを、多量体ポリペプチド抗原を含むことが疑われる被験試料の存在下に、多量体ポリペプチド抗体で被覆したマイクロタイターウエルに加えて、標識ペプチド(又は標識タンパク質)結合抗体複合体及び/又は患者抗原結合抗体複合体を形成するのに十分な時間及び条件下でインキュベートする。被験試料中の多量体ポリペプチド抗原は、マイクロタイターウエル上の結合部位に関して標識ポリペプチド(又はタンパク質)と競合する。被験試料中の抗原とペプチド又はタンパク質が抗体結合部位に関して競合するので、アッセイにおいて、被験試料中の多量体ポリペプチド抗原は、抗体被覆したマイクロタイターウエルへの標識ペプチドの結合低下を生じさせる。低いシグナル(対照と比較して)は、被験試料における多量体ポリペプチド抗原の存在を指示する。多量体ポリペプチド抗原の存在は、乳癌などの乳房の疾患又は状態の診断を示唆する。
実施例2で上述したようにして得られる免疫血清を使用して、組織、血液、血清又は他の体液から調製した溶液から多量体ポリペプチド複合体を免疫沈降させる。組織標本については、組織試料を0.1M Tris−HCl(pH7.5)、15%(w/v)グリセロール、0.2mM EDTA、10μg/mlロイペプチン及び1.0mMフェニルメチルスルホニルフルオリド中で均質化することによってタンパク質抽出物を調製する[Kainら、Biotechniques,17:982(1994)]。均質化後、ホモジネートを4℃、2000×gで5分間遠心分離して、デブリから上清を分離する。上記と同様であるが、0.1Mトリシン及び0.1%SDSも同時に含む緩衝液で均質化することによってデブリを再抽出する。血清標本は直接使用することができる。他の体液は、免疫沈降法の前に調製が必要であると考えられる。
実施例2で上述したようにして得た免疫血清を、実施例7に従って調製する固定化組換えポリペプチド複合体を用いてアフィニティー精製する。希釈した粗抗血清をプロテインAカラム(Affi−GelプロテインA、Bio−Rad,Hercules,CA)に通すことによって抗血清のIgG分画を得る。緩衝液(Binding Buffer,製造者から供給される)で溶出して、免疫グロブリンでない実質的にすべてのタンパク質を除去する。0.1M緩衝グリシン(pH3)で溶出して、アルブミン及び他の血清タンパク質を実質的に含まない免疫グロブリン調整物を得る。
実施例2で上述し、表3に列挙したモノクローナル抗体を使用して、ホルマリンで固定し、パラフィンに包埋した細胞系統(下記の実施例7で述べるような、MB8)ならびに悪性及び正常乳房組織を、標準的な手順を用いて免疫組織化学的に標識した。D.L.Spectorら、Cells:A Laboratory Manual,Plainview,NY:Cold Spring Harbor Laboratory Press 1998。簡単に述べると、5μm切片を切断し、正に荷電したスライド上に置いて、それらを60℃のスライド加温器上で30分間加熱した。切片をザイレン中で各々5分間ずつ2回、100%エタノール中で各々1分間ずつ2回、95%エタノール中で各々1分間ずつ3回、及び蒸留水で3分間、再水和した。切片を、10mMクエン酸緩衝液pH6.0中、Black and Decker Vegetable Steamerで30分間加熱した。切片を蒸留水で5分間洗い、その後Tris緩衝食塩水[0.05M Tris−HCl pH7.6、0.15M NaCl](TBS)に希釈した1Xカゼイン(Dako Corp.,Carpinteria,CA)で15分間遮断した。ハイブリドーマ培養上清をTBSに1:1希釈した。この希釈上清を切片に加え、室温で60分間インキュベートして、その後TBS中で各々5分間ずつ2回洗った。検出のために、LSAB+キット(Dako Corp.,Carpinteria,CA)を使用した。切片をリンク抗体と共に室温で30分間インキュベートし、その後TBS中で各々5分間ずつ2回洗った。次に切片をストレプトアビジンと共に室温で30分間インキュベートし、TBS中で各々5分間ずつ2回洗った。切片をBCIP/NBT/INT基質(Dako Corp.,Carpinteria,CA)で15分間視覚化し、蒸留水に入れて、水性封入媒質で封入した。切片を10X対物レンズのNikon Optiphot II光学顕微鏡で観察し、Photometrics CoolSnap CCDカメラ及びMetamorphバージョン4.0ソフトウエアで記録した。図2及び3は、それぞれモノクローナル抗体H9C65及びH95C30による2つの悪性乳房切片、1つの正常乳房切片、及びMB8細胞系統の免疫組織化学的染色の結果を示す。
A.安定な細胞系統、HEK293−MB8の生産
クローン603148又はクローン2083578を使用したBU101 myc/his(M/H)発現プラスミドの作製、及びクローン899895を使用したマンマグロビンM/H発現プラスミドの作製を述べている、1997年8月15日出願の米国特許願通し番号第08/912,276号のために放棄された、1996年9月19日出願の米国特許願通し番号第08/697,105号、及び1997年8月15日出願の米国特許願通し番号第08/912,149号のために放棄された、1996年8月19日出願の米国特許願通し番号第08/697,106号は、参照してここに組み込まれる。これらの発現プラスミドを、DH5α細胞に再形質転換し、LB/アンピシリン寒天に塗布して、LB/アンピシリン肉汁10ml中で増殖させた。QIAfilter(商標)Maxi Kit(Qiagen,Chatsworth,CA)を用いてプラスミドを精製し、HEK293細胞にトランスフェクトした[F.L.Grahamら、J.Gen.Vir.36:59−72(1977)]。
MB8細胞からの上清のアリコートを、マンマグロビンM/H及びBU101 M/H組換えタンパク質の両方の存在に関して分析した。還元試料緩衝液(50mM Tris pH6.8、10%グリセロール(v/v)、2%ドデシル硫酸ナトリウム(w/v)、2%β−メルカプトエタノール(v/v)、0.01%ブロモフェノールブルー(w/v)の最終濃度)中でアリコートを調製し、この技術分野で既知の標準的な方法及び試薬を使用してSDS−ポリアクリルアミドゲル(SDS−PAGE)で電気泳動した(J.Sambrookら、前出)。詳細には、試料40μlを試料緩衝液(5X)10μlに加え、その混合物を5分から10分間煮沸した。その調製した試料15μlを厚さ1mmの10%から20%Tricineゲル(Novex,San Diego,CA)に充填し、110Vで約90分間電気泳動した。次にこれらのゲルを20Vでニトロセルロースなどの固体媒体に一晩ブロットし、mycエピトープを認識するモノクローナル抗体(Invitrogen,Carlsbad,CA)若しくはBU101又はマンマグロビンのいずれかを認識するポリクローナル抗血清によるウエスタンブロット手法を用いてマンマグロビンM/H及びBU101 M/Hタンパク質バンドを視覚化した。詳細には、ニトロセルロースブロットを取り出し、0.2%I−block(登録商標)(Tropix,Bedford,MA)により室温で60分間遮断した。次に適切な量の一次抗体を加えた。例えば、前記ポリクローナル抗血清を1:5000の希釈で使用し、抗mycエピトープモノクローナル抗体を1:5000の希釈で使用した。一次抗体溶液を、振とうしながら室温で60分間ブロットに暴露した。その後ブロットをI−block(登録商標)溶液で3回洗った。ビオチニル化ヤギ抗ウサギIgG又はビオチニル化ヤギ抗マウスIgGのいずれかを含む二次抗体を、適切な希釈(1:5000)でI−block(登録商標)溶液中で調製し、振とうしながら室温で60分間ブロットに暴露した。その後ブロットをI−block(登録商標)溶液で3回洗った。次に、コンジュゲートであるアルカリホスファターゼ標識ストレプトアビジンを、適切な希釈(1:10,000)でI−block(登録商標)溶液中で調製し、振とうしながら室温で30分間ブロットに暴露した。その後ブロットをI−block(登録商標)溶液で3回、次いでアッセイ緩衝液(20mM Tris(pH9.8)/1mM塩化マグネシウム)で2回洗った。5−ブロモ−4−クロロ−3−インドリルホスフェート、BCIP 25mgをジメチルホルムアミド0.5mLに溶解した。次にこのBCIP溶液100μlをアッセイ緩衝液30mLと混合し、このBCIP基質溶液を、バンドが可視となるまでブロットに暴露した。その後ブロットを基質溶液から取り出し、空気中で乾燥した。乾燥ブロットを走査してブロットの電子コピーを得た。
上述したようなMB8細胞増殖からの上清を、マンマグロビンM/H及びBU101 M/Hの精製のためにニッケルを充填したChelating Sepharose Fast Flow(Pharmacia)に適用した。詳細には、Chelating Sepharose Fast Flow 40mLを16mm×10cmカラムに注入した。水中の硫酸ニッケル(0.1M)40mLをカラムに通してカラムにニッケルを充填した。カラムを洗い、リン酸ナトリウム10mM、塩化ナトリウム500mM、pH7.4で平衡させた。MB8細胞の増殖からの上清220mLを平衡カラムに適用し、イミダゾールの直線勾配を用いてヒスチジン標識タンパク質を溶出した。流速は2mL/分であった;勾配は緩衝液ミリリットル当り6.25Mイミダゾールであった;及び溶出時間は80分間で、0から500mMイミダゾールへの溶出プロフィールを創造した。各々4mLの分画を採取した。各々の分画100μlをドットブロット装置のウエルに適用し、その容量をニトロセルロース片を通して吸引した。次に、mycエピトープを認識するモノクローナル抗体を使用して、実施例7Bで上述したウエスタンブロットを視覚化するための同じ手順でニトロセルロースフィルターを視覚化した。図5(上のブロット)は視覚化したドットブロットを例示し、抗mycモノクローナル抗体による分画20−35中の物質の免疫認識を示している。これらの分画は250mMから438mMイミダゾールの溶出条件に対応し、ヒスチジン標識タンパク質のニッケルカラムへの結合の成功及びヒスチジン類似体によるそれらの溶出を示唆する。分画20−35をプールし、Slide−a−Lysers(3500 MWCO)を用いて、リン酸緩衝食塩水(PBS、50mMリン酸塩、150mM塩化ナトリウム、pH7.4)2×4Lに対して各々少なくとも4時間透析した。プールして透析した半精製MB8上清を、ウエスタンブロットによってマンマグロビンM/H及びBU101 M/H組換えタンパク質の両方の存在に関して分析した。
対象タンパク質の等電点を、Wisconsin Sequence Analysis Package(Genetics Computer Group)の等電関数を用いて測定した。等電点はすべてのタンパク質の性質であり、タンパク質がゼロの正味電荷を有するpHである。Thomas E.Creighton編集、Proteins;Structures and Molecular Properties,第2版、W H Freeman and Company,NY(1993)。表4は対象タンパク質pIを列挙している:
ニッケルキレート化クロマトグラフィー後、MB8細胞の増殖からの半精製上清を、陰イオン交換クロマトグラフィーを用いてさらに精製した。詳細には、実施例7Cで上述したように、ニッケル精製MB8上清10mLを20mMピペラジン、pH6.0 2Lに対して透析した。この物質を、20mMピペラジン、pH6.0で平衡させたMono Q 5/5カラム(Pharmacia)に適用した。塩化ナトリウムの直線勾配を用いてタンパク質を溶出した。流速は1mL/分であった;勾配は緩衝液ミリリットル当り20mM塩化ナトリウムであった;及び溶出時間は50分間で、0から1000mM塩化ナトリウムへの溶出プロフィールを創造した。試料充填時間は、溶出勾配の適用の30分前であった。各々1mLの分画を採取した。各々の分画100μlを0.1%β−メルカプトエタノールの存在下で煮沸し、冷却して、その後ドットブロット装置のウエルに適用し、その容量をニトロセルロース片を通して吸引した。次に、mycエピトープを認識するモノクローナル抗体を使用して、実施例7Bで上述したウエスタンブロットを視覚化するための同じ手順でニトロセルロースフィルターを視覚化した。図8(上のブロット)は視覚化したドットブロットを例示し、抗mycモノクローナル抗体による分画36−47中の物質の免疫認識を示している。免疫反応性物質は120mMから340mM塩化ナトリウムの間で溶出し、ピークの中心は180mM塩化ナトリウムで溶出する。これらの結果は、前記物質がpH6.0の陰イオン交換カラムに弱く結合したという点で、マンマグロビンM/HとBU101 M/Hを合わせた場合の5.9という算定等電点に合致する。BU101 M/H単独の等電点は7.6と算定され、正味正電荷を持たないので、このタンパク質はpH6.0の陰イオン交換カラムに結合しないと予想される。BU101 M/Hは、試料の充填中(最初の30分画)カラムを通って流出すると予想される。しかし、これらの30流出分画中に抗myc免疫反応性物質は認められなかった。これらの結果は、BU101 M/HがマンマグロビンM/Hとの共有結合のために変化した等電特性を有することと一致する。
ニッケルキレート化クロマトグラフィー後、MB8細胞の増殖からの半精製上清を、ゲルろ過クロマトグラフィーを用いてさらに精製した。詳細には、上述したようなニッケル精製MB8上清4.65mLを650μLに濃縮し、濃縮物200μLをSuperose 12(Pharmacia)の10mm×30cmカラムに適用した。カラムに0.4mL/分の流速でPBS(50mMリン酸塩、150mM塩化ナトリウム、pH7.4)の単一緩衝液を通過させた。Pharmaciaから入手可能な分子量標準品でカラムを検定した。生じた分子量測定に関する標準曲線が図9に示されており、その図は分子量と溶出量の関係を明らかにしている。
A.組織抽出物の調製
−70℃で急速凍結して保存しておいた乳癌組織1グラムの10分の2をウエスタンブロット分析のために調製した。組織試料を0.1M Tris−HCl(pH7.5)、15%(w/v)グリセロール、0.2mM EDTA、10μg/mlロイペプチン及び1.0mMフェニルメチルスルホニルフルオリド中で均質化することによってタンパク質抽出物を調製した。S.R.Kainら、Biotechniques,17:982(1994)。均質化後、ホモジネートを4℃で5分間遠心分離して、デブリから上清を分離した。タンパク質の定量のために、上清2μLから5μLをクマシータンパク質試薬(Pierce Chemical Co.,Rockford,IL)1.5mLに加え、吸光度を595nmで読み取った。
上述したように、乳癌組織抽出物をウエスタンブロットで分析した。他の試料は、組換えマンマグロビンM/HとBU101 M/H及び下記で述べるようなもう1つの乳癌組織抽出物を含んだ。還元試料緩衝液(50mM Tris pH6.8、10%グリセロール(v/v)、2%ドデシル硫酸ナトリウム(w/v)、2%β−メルカプトエタノール(v/v)、0.01%ブロモフェノールブルー(w/v)の最終濃度)及び非還元試料緩衝液(50mM Tris pH6.8、10%グリセロール(v/v)、2%ドデシル硫酸ナトリウム(w/v)、及び0.01%ブロモフェノールブルー(w/v)の最終濃度)の両方において試料を調製し、次にこの技術分野で既知の標準的な方法及び試薬を使用してSDS−ポリアクリルアミドゲル(SDS−PAGE)で電気泳動した(J.Sambrookら、前出)。詳細には、試料40μlを試料緩衝液(5X)10μlに加え、その混合物を5分から10分間煮沸した。その調製した試料15μlを厚さ1mmの10%から20%Tricineゲル(Novex,San Diego,CA)に充填し、110Vで約90分間電気泳動した。次にこれらのゲルを20Vでニトロセルロースなどの固体媒体に一晩ブロットし、BU101又はマンマグロビンのいずれかを認識するモノクローナル又はポリクローナル抗血清によるウエスタンブロット手法を用いてマンマグロビン及びBU101タンパク質バンドを視覚化した。詳細には、ニトロセルロースブロットを取り出し、0.2%I−block(登録商標)(Tropix,Bedford,MA)により室温で60分間遮断した。次に適切な量の一次抗体を加えた。例えば、前記ポリクローナル抗血清を1:5000の希釈で使用し、モノクローナル培養上清を1:50の希釈で使用した。一次抗体溶液を、振とうしながら室温で60分間ブロットに暴露した。その後ブロットをI−block(登録商標)溶液で3回洗った。ビオチニル化ヤギ抗ウサギIgG又はビオチニル化ヤギ抗マウスIgGのいずれかを含む二次抗体を、適切な希釈(1:5000)でI−block(登録商標)溶液中で調製し、振とうしながら室温で60分間ブロットに暴露した。その後ブロットをI−block(登録商標)溶液で3回洗った。次に、コンジュゲートであるアルカリホスファターゼ標識ストレプトアビジンを、適切な希釈(1:10,000)でI−block(登録商標)溶液中で調製し、振とうしながら室温で30分間ブロットに暴露した。その後ブロットをI−block(登録商標)溶液で3回、次いでアッセイ緩衝液(20mM Tris(pH9.8)/1mM塩化マグネシウム)で2回洗った。5−ブロモ−4−クロロ−3−インドリルホスフェート、BCIP 25mgをジメチルホルムアミド0.5mLに溶解した。次にこのBCIP溶液100μlをアッセイ緩衝液30mLと混合し、このBCIP基質溶液を、バンドが可視となるまでブロットに暴露した。その後ブロットを基質溶液から取り出し、空気中で乾燥した。乾燥ブロットを走査してブロットの電子コピーを得た。
侵襲性のほとんど分化していない腺癌(乳房)を有する患者から誘導した乳癌組織抽出物(図11、レーン4と10)を、陰イオン交換クロマトグラフィーを用いて精製した。詳細には、前記抽出物800μLを、20mMピペラジン、pH6.0で平衡させたMono Q 5/5カラム(Pharmacia)に適用した。塩化ナトリウムの直線勾配を用いてタンパク質を溶出した。流速は1mL/分であった;勾配は緩衝液ミリリットル当り20mM塩化ナトリウムであった;及び溶出時間は50分間で、0から1000mM塩化ナトリウムへの溶出プロフィールを創造した。試料充填時間は、溶出勾配の適用の30分前であった。各々1mLの分画を採取した。各々の分画100μlを0.1%β−メルカプトエタノールの存在下で煮沸し、その後ドットブロット装置のウエルに適用して、その容量をニトロセルロース片を通して吸引した。次に、BU101又はマンマグロビンのいずれかを認識するポリクローナル抗血清を使用して、実施例7Bで上述したウエスタンブロットを視覚化するための同じ手順でニトロセルロースフィルターを視覚化した。図12は視覚化したドットブロットを例示し、抗マンマグロビンポリクローナル抗血清及び抗BU101ポリクローナル抗血清による分画41−48中の物質の免疫認識を示している。分画41−48は220mMから360mM塩化ナトリウムの塩濃度に対応する。これらの結果は4.6未満の等電点に合致する。表5は、陰イオン交換カラムからの溶出条件を、溶出したタンパク質又はタンパク質複合体の対応するpIと共に要約している。
上述したような半精製乳癌組織抽出物を、ゲルろ過クロマトグラフィーを用いてさらに精製した。詳細には、上述したようなイオン交換クロマトグラフィーからの分画41−48を、3000×gで25分間回転させたCentriplus 30濃縮器を用いて100μLに濃縮した。残留物をSuperose 12(Pharmacia)の10mm×30cmカラムに適用した。カラムに0.4mL/分の流速でPBS(50mMリン酸塩、150mM塩化ナトリウム、pH7.4)の単一緩衝液を通過させた。Pharmaciaから入手可能な分子量標準品でカラムを検定した。生じた分子量測定に関する標準曲線が図9に示されており、その図は分子量と溶出量の関係を明らかにしている。
HEK293−MB8細胞の増殖及びマンマグロビンM/Hの一過性トランスフェクションからの半精製上清(実施例7Cで述べたようなニッケルキレート化クロマトグラフィー後)を、加熱を伴う又は伴わない、非イオン性界面活性剤、陰イオン界面活性剤、又は還元剤を含む種々の前処理プロトコールに供した。詳細には、Tween 20、ドデシル硫酸ナトリウム(SDS)、又はβ−メルカプトエタノール(β−ME)を、添加物の最終濃度を0.1%、0.25%、0.5%、1.0%、2.0%及び4.0%として、上述した半精製上清100μLに加えた。2組の試料を5分間加熱処理して、冷却した。次に、処理した試料をドットブロット装置に充填し、その容量をニトロセルロース片を通して吸引した。その後、抗mycモノクローナル抗体を使用して、実施例7Bで上述したウエスタンブロットを視覚化するための同じ手順でニトロセルロースフィルターを視覚化した。
A.背景
高度に組織又は疾患特異的に発現される遺伝子は、治療、癌の早期検出、及び治療中と治療後の疾患症状の監視のための可能性のある標的を提供する。さらに、分泌又は流出タンパク質をコードするこの種の遺伝子は、我々がすべての状況で癌を特異的に検出する能力を助ける、その産物の血清中での検出を可能にしうる。
本発明者らは、定量的RT−PCRを使用して、101の原発性乳癌及び乳癌を有していない患者からの17のリンパ節のシリーズにおいて3つの乳房特異的マーカー(BU101、マンマグロビン及びBS106)の発現を測定した。すべての癌が、この同じアッセイによりサイトケラチン19に関して陽性であった。
各々の癌からの全RNAの等量を単離し、ランダムプライミングによってcDNAに変換した。各々のマーカーに特異的なプライマーをABI 7700サイクラーでのPCR増幅において使用して、各々の反応にSybr Greenを添加した。発現値は、内蔵の交点解析ソフトウエアを用いて誘導し、乳癌細胞系、SKBR−3における各マーカーの発現レベルに基準化した。
Claims (5)
- 次の乳癌マーカーであるマンマグロビン、BU101及びBS106の少なくとも2つを含む、乳癌を検出するためのアッセイ。
- (a)患者から被験試料を得る段階、
(b)前記被験試料を、マンマグロビン、BU101及びBS106から成る群より選択される少なくとも2つのポリペプチドと接触させる段、及び
(c)段階(b)のポリペプチドの1つ又はそれ以上の存在を乳癌に相関させる段階、
を含む、乳癌を検出するための方法。 - (a)患者から試料を得る段階、
(b)前記試料を、BS106、マンマグロビン、BU101及び多量体抗原(MPA)に特異的な少なくとも2つの抗体と接触させる段階、
但し、前記多量体抗原は、少なくとも1つのBU101ポリペプチド及び少なくとも1つのマンマグロビンポリペプチドを含み、
前記接触は、抗原/抗体複合体を形成するのに十分な時間及び条件下で行われ、及び
(c)前記複合体の存在が前記患者における癌の存在を指示する、前記複合体を検出する段階、
を含む、乳癌の存在を検出する方法。 - (a)患者から切除した腫瘍から誘導される組織切片又は細胞培養を調製する段階、
(b)前記組織切片又は細胞培養を、次のポリペプチド:BS106、マンマグロビン及びBU101の少なくとも2つに特異的な抗体に、抗原/抗体複合体を形成するのに十分な時間及び条件下で接触させる段階、及び
(c)前記複合体の存在が前記患者における癌の存在を指示する、前記組織切片又は細胞培養における前記複合体の存在の場所を特定する段階、
を含む、患者において乳癌を診断する方法。 - (a)患者から被験試料を得る段階、
(b)前記被験試料を、マンマグロビン、BU101、BS106及びMPAから成る群より選択される少なくとも2つのポリペプチドと接触させる段階、及び
(c)段階(b)のポリペプチドの1又はそれ以上の存在を乳癌に相関させる段階、
を含む、乳癌を検出するための方法。
