c)形質導入が十分に起こる条件下で上記細胞を(b)のベクター粒子と接触させるステップとを含む方法である。別の一実施態様では、この方法は、形質導入を増大させるのに十分な量のレトロネクチンを提供するステップをさらに含んでいる。この方法の1つの特徴によれば、試験管内で細胞への形質導入がなされる。別の特徴によれば、生体内で細胞への形質導入がなされる。さらに別の実施態様によれば、細胞は、脊椎生物の細胞、霊長類の細胞、ヒトの細胞のいずれかである。細胞は、CD34+細胞またはPBL細胞にすることもできる。上記方法の別の実施態様として、その方法によって形質導入された細胞が含まれる。
(a)(i)少なくとも1つのベクター構造体と;
(ii)少なくとも1つのパッケージング構造体と;
(iii)請求項1のキメラ糖タンパク質をコードしている発現構造体とを細胞にトランスフェクトして産生細胞を作り出すステップと;
(c)その産生細胞を培地内で培養するステップと;
(d)その産生細胞を培地から分離することにより、その培地から組み換えウイルス・ベクター粒子を回収するステップを含む方法である。
タイプCとタイプDに属する哺乳動物レトロウイルスのGPでは、細胞質尾部がウイルスのアセンブリーと融合生成性の両方を調節するプロセスにおいて非常に重要である。糖タンパク質の局在、細胞表面の密度、細胞-細胞融合、異種または同種のウイルス・コアとの相互作用、異種または同種のウイルス・コアの組み込み、ビリオンの感染力に対して細胞質尾部が影響を有することを示すいくつかの証拠がある。GPの細胞質尾部は、Rと呼ばれるカルボキシ末端ペプチドを含む構造モチーフである。このRはチロシン・エンドサイトーシス・シグナル(YXXL)(5)を備えており、そのRがウイルスのプロテアーゼによって開裂するとGPの性質が大きく変化する。GPの元々の形態でRペプチドが開裂前に相互作用すると考えられている相手は、i)細胞エンドサイトーシス機構のアダプチン複合体、ii)ビリオン内に見いだされる成熟GPのカルボキシ末端(図1Aに示した細胞質尾部のTドメイン)、iii)ビリオンの内部タンパク質である(1、8、13、14、17、19、43)。RD114 GP突然変異体の特性を調べた結果(この明細書)は、細胞質尾部によって運ばれる関連シグナルが存在していることを示している。したがって、YXXLモチーフが点突然変異誘発によって破壊されたRDΔYXXL突然変異体GPにより、おそらくは細胞表面での発現が増えることを通じて細胞-細胞融合が増加する。これは、がんレトロウイルスとレンチウイルスの糖タンパク質を用いた別の人による研究が示唆していることである(1、8、13)。しかし細胞-細胞融合生成性および/または細胞表面の発現を増大させた突然変異は、必ずしもウイルスの組み込みおよび/またはウイルスの感染力を増大させない可能性がある。そのことは、RDRなし、RDPrMLV、RDPrSIVRQAG、RDPrHIV、RDΔYXXLという突然変異体GPの性質から推測される。さらに、われわれのデータは、類型化されたベクターの感染力が、RD114 GPの修飾された細胞質尾部とウイルス・コアのタイプの間の適合性に依存することを示している。
RD114 GPの細胞質尾部を修飾しても、レンチウイルス・コアとは異なり、MLVコアへのRD114 GPキメラの組み込みにはほとんど影響がなかった(図4)。これは、GPの組み込みに関してがんレトロウイルス・コアがレンチウイルス・コアよりも許容度が大きいこと、あるいはRD114 GPの持つ不適合性決定基がレンチウイルス・コアに限定されていることを示している。しかし類型化されたMLVベクター粒子の感染力は、RD114 GPキメラの細胞質尾部に導入された開裂部位のタイプによって大きく影響された(図3)。RD114 GPの開裂部位をMLVの開裂部位で置換しても感染力に影響はなかったが、レンチウイルスの開裂部位を挿入すると感染が劇的に低下した。これは、MLV突然変異体がMLVコア粒子に組み込まれたときにそのTMタンパク質の細胞質尾部が十分に処理されていないことと整合性がある(図5A)。これらの結果は、ベクター類型の感染力が低いのは、組み込まれたGPキメラの融合能力が十分に活性化されていないためであることを示唆している。MLV GPでは、開裂部位そのものまたはウイルスのプロテアーゼが突然変異しているために細胞質尾部が開裂しない結果、正常レベルのGPが組み込まれているが感染力のない突然変異体が生成した(33)。
多種類のウイルスがベクターのベースとなっている。ウイルスを感染させるには、ウイルスのゲノムを宿主細胞に導入する。このゲノムを、ウイルスをベースとしたベクター内の遺伝子デリバリー・ビヒクルとして利用する。使用するウイルスは、細胞にトランスフェクトするのに必要な特徴と能力の多くをすでに有する病原性ウイルス種に由来するものであることがしばしばある。しかしすべてのウイルスがあらゆるタイプの細胞の細胞周期のすべての段階でうまくトランスフェクションを起こさせうるわけではない。したがってウイルス・ベクターを開発する際にウイルスのゲノムを修飾することにより、外来性遺伝子構造体(導入遺伝子)または他の核酸を導入する効果と効率を増大させる。そのとき、そのベクターが疾患を引き起こす能力を小さくする修飾、あるいはなくす修飾を行なう。
レトロウイルスに組み込まれるタンパク質は、同種のウイルス糖タンパク質に限られない。宿主細胞に由来する40種類を超えるタンパク質が、HIV-1ウイルス粒子の外側で同定されている。例えば、腫瘍組織適合遺伝子複合体のクラスI(MHC-I)とMHC-IIの分子、接着分子、共刺激分子、相補性制御タンパク質などが挙げられる48。さらに、多数の異種ウイルス糖タンパク質をレトロウイルス粒子に組み込んで感染力を持たせることができる49。類型化として知られるこのプロセスにより、レトロウイルス・ベクターを用いて広い範囲の細胞と組織を形質転換することが可能になる。VSV-G糖タンパク質を用いたレンチウイルス・ベクターの改変は、異種糖タンパク質がベクターの親和性を拡張する能力を有することの具体例である2。しかし所定の糖タンパク質(GP)を異種ウイルス・コアと同時発現させても、必ずしも感染力の大きなウイルス粒子は得られないであろう8、14、15、50。
VSV Gで類型化したレンチウイルス・ベクターは、いくつかのタイプの細胞を生体内と試験管内で形質転換させるのに有効であることが証明されている3-7。しかしヒト24とヒト以外の霊長類の補体に対する感受性が大きい(図4)ため、生体内への全身投与は不可能であろう。VSV-G類型とは異なり、レトロウイルスの糖タンパク質を用いて作ったベクターはヒトとマカクの血清中で安定であり、RD114/TRで類型化したSIVベクターは、ヒト血清に対して常に抵抗性を示した。これは、後者のベクターが全身への遺伝子デリバリーに特に適していることを示唆している(図4)。いくつかの因子が補体に対する感度決定に寄与しており、その因子が依存するのは、i)いろいろな人からの血清;ii)産生細胞のタイプ34、36;iii)糖タンパク質中のα(1-3)ガラクトース糖エピトープの存在;iv)類型化用GPのタイプ34、36、62のいずれかである。ヒト細胞が産生するレトロウイルスは、通常はヒト血清中で抵抗性だが34、36、VSV-Gで類型化したベクターは例外である24。しかし最近のある研究によると、ヒト細胞が産生する、MLV GPでコーティングされたがんレトロウイルス・ベクターは、血清中での補体不活性化に対する感度が、ヒトからの進化距離の増大と相関した形で非ヒト旧世界霊長類とは異なっていることが見いだされた63。マカク血清に対する感受性があるため、99%を超えるベクターが分解した63。ところがわれわれは、レトロウイルスのGPで類型化したレンチウイルス・ベクターはマカク血清中で比較的安定であることを見いだした(図4)のであるから、がんレトロウイルス・ベクターで得られた上記の結果とは明らかに一致しない。血清に対する応答を変化させ、しかもがんレトロウイルス粒子とレンチウイルス粒子の違いを説明できる可能性のある因子は、レンチウイルス粒子へのCD46、CD55、CD59補体抑制分子の組み込みである。これについては、HIVとSIVに関して報告されている48、64。
VSV-Gで類型化されたレンチウイルス・ベクターの幅広い親和性は、特定の用途での遺伝子導入には適していない可能性がある。それは、細胞のタイプに特異的な遺伝子デリバリーが必要とされるような場合である。より選択的な親和性は、膜で覆われたいくつかのウイルスに由来する糖タンパク質が持つ自然の親和性を利用することによって、あるいは組み込み能力のあるGP(例えばMLV、GALV/TR、FPV-HA)の宿主の範囲を変化させることによって実現できよう65、66。例えば肺感染を引き起こし、気道上皮を通じて感染するウイルス(エボラ・ウイルス、インフルエンザ・ウイルス)に由来する表面糖タンパク質を用いると、ヒト気道の遺伝子治療に有効であることがわかる可能性がある10。しかしレンチウイルス・ベクター類型は、親ウイルスの宿主の範囲と必ずしも同じでない可能性があることに注意せねばならない。例えば向神経性狂犬病ウイルスであるモコラ・ウイルスの糖タンパク質は効率的にHIV-1ベクターを類型化する12とはいえ、類型化されたベクターは親ウイルスの特異的向神経性を再現しない。
この明細書では、“細胞”、“細胞系”、“細胞培養物”という用語は、同じ意味で用いることができる。これらのどの用語にも、その子孫が含まれる。子孫とは、その後の世代すべてを意味する。意図的または非意図的な突然変異があるため、すべての子孫が同じとは限らないことがわかる。異種核酸配列の発現という文脈では、“宿主細胞”は、原核細胞または真核細胞を意味し、その中には、形質転換が可能で、ベクターの複製、および/または本発明のベクターによってコードされている異種核酸の発現が可能なあらゆる生物が含まれる。宿主細胞は、ベクターのレシピエントとして用いることができ、実際にレシピエントとして用いられてきた。宿主細胞は、“トランスフェクト”または“形質転換”することができる。これは、外来性核酸を宿主細胞に移植または導入するプロセスを意味する。形質転換された細胞としては、一次対象細胞やその子孫が挙げられる。この明細書では、“操作した”細胞または宿主細胞、“組み換えた”細胞または宿主細胞という用語は、外来性核酸配列(例えば治療用遺伝子構造体を有する本発明のレンチベクター)を導入された細胞を意味する。したがって組み換え細胞は、組み換えによって導入された核酸を含まない自然に発生する細胞から区別することができる。
当業者であれば、本発明が特定の1つのタイプの細胞に限定されることはなく、導入遺伝子を発現させるために本発明のレンチウイルス・ベクターと本発明の方法を多くのタイプの細胞で使用できることが理解できよう。考えられる細胞のタイプの具体例として、ニューロン、肺細胞、筋細胞、肝細胞、膵臓細胞、内皮細胞、心筋細胞、皮膚細胞、骨髄ストロマ細胞、耳細胞、目細胞など、最終分化した細胞が挙げられる。さらに、幹細胞や前駆細胞(例えば膵臓管細胞、神経前躯体、中胚葉幹細胞)も考えられる。しかし注目すべきことに、本発明のより好ましいレンチベクターは、ヒト前駆細胞の中で導入遺伝子の発現レベルが高い一方、ヒトの身体に対する安全性の条件に合致しているという非常に望ましい特徴を持っている。
いくつかの実施態様では、宿主の細胞または組織が少なくとも1つの生物に含まれていてよい。いくつかの実施態様では、その生物として、ヒト、霊長類、ネズミ類のいずれかが可能である。別の実施態様では、当業者であればわかるように、その生物として任意の真核生物が可能であり、原核生物(例えば、真正細菌、古細菌)さえ可能である。本発明によるいくつかのレンチベクターは制御配列を含むことができるため、そのレンチベクターを原核細胞と真核細胞の両方において複製すること、および/または発現させることができる。当業者であればさらに、上記のあらゆる宿主細胞をインキュベートして維持し、ベクターを複製できるようにする条件がどのようなものであるかが理解できよう。本発明のレンチベクターを大規模に製造する方法と条件は公知であり、そのレンチベクターによってコードされている核酸、そのコグネイト・ポリペプチド、タンパク質、ペプチドを製造する方法と条件も公知である。こうやって製造されたもののうちのいくつかは、遺伝子治療に用いられることになる治療用の遺伝子またはタンパク質である。
本発明のレンチウイルス・ベクター組成物を用いて遺伝子治療を実現するためには、一般に、その治療を必要としている細胞を、治療用遺伝子を含むレンチウイルス・ベクターと接触させることになろう。その細胞は、その後、遺伝子治療を必要としているヒトなどの生物に入れられることになる。投与経路は、当然のことながら、疾患の場所と性質に応じてさまざまになろう。例えば、静脈内、動脈内、皮膚内、経皮、筋肉内、鼻腔内、皮下、気管内、腹腔内、腫瘍内、灌流、洗浄などの経路がある。細胞を生物から単離して生体外でレンチベクターに曝露し、あとで再び移植することも時に行なわれる。
A.導入遺伝子と疾患の治療
本発明の一実施態様は、治療用遺伝子をコードしている核酸、中でも造血異常またはリンパ造血異常(例えばすでに記載した先天性または後天性の疾患)を治療するための遺伝子をコードしている核酸を導入するというものである。一実施態様では、核酸が、そのような遺伝子の全長、あるいは実質的に全長、あるいは機能的な等価物をコードしている。このような遺伝子は、導入遺伝子として知られている。
n〜n+y
ここにnは1から配列の最後の数までの範囲の整数であり、yは核酸セグメントの長さから1を引いた値であり、n+yは配列の最後の数を超えない。したがって10量体では、核酸セグメントが塩基1〜10、2〜11、3〜12などに対応する。15量体では、核酸セグメントが塩基1〜15、2〜16、3〜17などに対応する。20量体では、核酸セグメントが塩基1〜20、2〜21、3〜22などに対応する。
本発明のベクターには、多重クローニング部位(MCS)が含まれる。MCSは多重制限酵素部位を含む核酸領域であり、そのうちの任意の制限酵素部位を標準的な組み換え技術と組み合わせることによってベクターを消化する(例えばCarbonelli他、1999年、Levenson他、1998年、Cocea他、1997年を参照のこと)。“制限酵素消化”は、核酸分子の特定の位置だけで機能する酵素を用いて核酸分子を触媒的に開裂させることを意味する。このような制限酵素の多くは市販されている。このような制限酵素を使用することは、当業者に広く知られている。MCS内で切断を行なう制限酵素を用いてベクターを直線化または断片化し、外来性配列がそのベクターに連結できるようにすることがしばしば行なわれる。“連結”は、2つの核酸断片間にホスホジエステル結合を形成するプロセスを意味する。その核酸断片は、互いに連続していてもいなくてもよい。制限酵素と連結反応を含む方法は、組み換え技術の当業者には周知である。
転写されたほとんどの真核生物RNA分子は、RNAのスプライシングを受けて一次転写産物からイントロンが除去される。真核生物のゲノム配列を含むベクターは、タンパク質を発現させる転写産物の適切なプロセシングを保証するドナーおよび/またはアクセプター・スプライシング部位を必要とする可能性がある(例えばChandler他、1997年を参照のこと)。
本発明のベクターまたは構造体は、一般に、少なくとも1つの終結シグナルを含んでいる。“終結シグナル”または“ターミネータ”は、RNAポリメラーゼによるRNA転写産物を具体的に終結させることに関与するDNA配列からなる。したがっていくつかの実施態様では、RNA転写産物の産生を終わらせる終結シグナルが考えられる。望むメッセージ・レベルを実現するのに生体内にターミネータが必要とされる可能性がある。
真核生物の遺伝子発現では、転写産物を適切にポリアデニル化するためのポリアデニル化シグナルが一般に用いられる。ポリアデニル化シグナルの性質は、本発明を成功させるのに不可欠であるとは考えられず、同様の任意の配列を用いることができる。いくつかの実施態様には、便利でさまざまな標的細胞の中でうまく機能することが知られているSV40ポリアデニル化シグナルまたはウシ成長ホルモン・ポリアデニル化シグナルが含まれる。ポリアデニル化により、転写産物の安定性が増大する可能性、あるいは細胞質における輸送が容易になる可能性がある。
本発明のベクターを宿主細胞内で伝播させるため、そのベクターは、複製が開始される特別な核酸配列である複製起点部位(“オリ”と呼ばれることがしばしばある)を1つ以上含んでいるとよい。あるいは宿主細胞が酵母の場合には、自律複製配列(ARS)を用いることもできる。
本発明のいくつかの実施態様では、本発明のレンチベクターを用いて形質導入された細胞は、発現ベクター内にマーカーを含めることによって試験管内または生体内で同定することができる。このようなマーカーは、形質導入された細胞に同定可能な変化を与えるため、その発現ベクターを含む細胞を容易に同定できるようになろう。一般に、選択マーカーは、選択を可能にする性質を与えるマーカーである。正の選択マーカーは、そのマーカーが存在することによってそのマーカーを選択できるようにするマーカーであるのに対し、負の選択マーカーは、そのマーカーが存在することによってそのマーカーの選択が阻止されるマーカーである。正の選択マーカーの一例は、薬剤耐性マーカーである。
“プロモータ”は、核酸配列のうちで転写開始と転写速度を制御する領域となっている制御配列である。プロモータは、調節タンパク質と調節分子が結合して核酸配列の特異的な転写を開始させる遺伝子エレメント(例えばRNAポリメラーゼや他の転写因子)を含むことができる。“機能する位置にある”、“機能上関連した”、“制御下”、“転写制御下”といった表現は、核酸配列に対してプロモータが正しく機能する位置および/または向きにあって、その配列の転写開始および/または発現を制御していることを意味する。

以下の実施例は、本発明の好ましい実施態様を示すためにこの明細書に含めてある。以下の実施例に開示した技術は、発明者が本発明を実施する際にうまくいくことを発見した技術であるため、本発明を実施するための好ましい態様を構成すると考えられることに当業者は注意されたい。しかし当業者であれば、この明細書の開示内容に照らし、開示してある具体的な実施態様において本発明の精神と範囲を逸脱することなく多くの変更をなしても同様の結果が得られることがわかるはずである。
1.細胞
293Tヒト胚性腎細胞系(ACTT CRL-1573)とTE671ヒト横紋筋肉腫細胞系(ACTT CRL-8805)を、10%ウシ胎仔血清(FCS)を追加したDMEM(ライフ・テクノロジーズ社、フランス)の中で増殖させた。
抗RD114 GP(ヴァイロメド・バイオセイフティ・ラブズ社、アメリカ合衆国)は、RD114 gp70エンベロープ糖タンパク質(SU)に対するヤギの抗血清であり、それを1/5,000に希釈してウエスタン・ブロットに使用した。抗SIV CA(NIHエイズ研究・基準試薬プログラム、アメリカ合衆国)は、SIVmac251 p27キャプシド・タンパク質(CA)に対するマウスのモノクローナル抗体(2F12)であり、それを1/500に希釈してウエスタン・ブロットに使用した。抗MLV CA(ヴァイロメド・バイオセイフティ・ラブズ社、アメリカ合衆国)は、ラッシャー白血病ウイルス(RLV)p30キャプシド・タンパク質(CA)に対するヤギの抗血清であり、それを1/10,000に希釈してウエスタン・ブロットに使用した。
pSIV-12パッケージング・プラスミド(図1)はpSIV825の誘導体であり、hCMVプロモータとHIV-1 rev遺伝子発現ユニットの制御下でSIVmac251 gag-pol遺伝子を発現する。HIV-1 rev遺伝子発現ユニットにはrevの2つのエキソンが融合しており、その2つのエキソンは、3-ヒドロキシ-3-メチルグルタリル補酵素Aレダクターゼ(HMG)プロモータと、HMGイントロンIと、SV40ポリアデニル化配列の制御下にある。pSIV-T1+プラスミド30は、パッケージング能力を有するSIVmac251をベースとしたベクターをコードしており、CMVプロモータの制御下で増大したグリーン蛍光タンパク質(GFP)マーカー遺伝子を発現する(図1)。
phCMV-G31、EboV-GP(V. Volchkovからの好意ある提供)、phCMV-HA32、phCMV-10A133、phCMV-GALV33というプラスミドは、水疱性口内炎ウイルス(VSV)のGP、エボラ・ウイルス(EboV)のザイール株のGP、トリ・インフルエンザ・ウイルス(FPV)のH7-HAヘマグルチニン、MLV-10A1、テナガザル白血病ウイルス(GALV)のエンベロープ糖タンパク質をそれぞれコードしている。糖タンパク質はすべて、同じシス作用性シグナルの制御下で発現した:CMVプロモータ、ウサギのβ-グロビンのイントロンII、ポリアデニル化配列(図1)。
融合アッセイで使用するHeLa細胞は、以前に報告されているように(9)、HIV-1の長い末端反復配列(LTR)の制御下にあるβ-ガラクトシダーゼ遺伝子(LacZ)(この遺伝子の発現はTatに依存する)の安定なトランスフェクタントになっている(HeLaCD4LTRLacZ細胞)か、あるいはHIV-1のTatタンパク質を構成的に発現していた(HeLa-Tat細胞)。エンベロープを通じた融合は、ほぼ以前に報告されているようにして定量化した(9、16)。このアッセイでは、エンベロープを発現する細胞が受容体を有する指示細胞と融合したとき、HIV-1 LTRによって引き起こされるβ-ガラクトシダーゼの発現がTatタンパク質によってトランス活性化される。トランスフェクションを行なう24時間前に、5×104個のHeLaCD4LTRLacZ細胞を24ウエルのプレートに植えた。ウイルス糖タンパク質発現構造体を、リン酸カルシウム・トランスフェクション・プロトコル(クロンテック社、フランス)を利用して上記のHeLa細胞にトランスフェクトした。そのとき、製造者の勧めに従ってプラスミドを1μgを用いた。トランスフェクションを行なってから24時間後、105個のHeLa-Tat指示細胞を、ウイルス糖タンパク質を提示する細胞とともに36〜48時間にわたって培養した。以前に報告されているように(9、16)、PBS(リン酸緩衝溶液)の中で0.5%(重量/容積)のグルタルアルデヒドを用いて固定し、PBSで洗浄し、5-ブロモ-4-クロロ-3-インドリル-β-D-ガラクトピラノシド(X-Gal)溶液の中でインキュベートすることによって染色した後に細胞-細胞融合を測定した。エンベロープを提示する細胞とTatを含有する指示細胞が互いに融合していることを示す青いシンシチウムを、シンシチウム1つ当たりの核の数に関係なくカウントした。
類型化されたSIV由来のベクターを、以前に報告されている25ようにして、293T細胞の一過性トランスフェクションによって作った。pSIV-T1+ベクター構造体(8.1μg)と、pSIV-12パッケージング構造体(8.1μg)と、ウイルス糖タンパク質発現構造体(2.7μg)を同時に、前日に10cmのプレートに植えた293T細胞にトランスフェクトした。トランスフェクションを行なった16時間後に培地(プレート1つにつき12ml)を交換し、上清を24時間後に回収した。26mlの超遠心分離管の中でベクター粒子を濃縮し、ビリオンをペレット化した。なおこの超遠心分離管は、70Tiベックマン・ローターの中で4℃にて1時間にわたって32,000rpmで回転させた。ウイルスのペレットは、ウイルスの上清を最初の1/100の体積採取してその中に1%ウシ血清アルブミン(BSA)を追加した無血清DMEMを入れたものの中に再び分散させ、等分し、-80℃で保管した。
ウイルス産生細胞を、1%のトリトン-X100と、0.05%のSDS(ドデシル硫酸ナトリウム)と、5mg/mlのデオキシコール酸ナトリウムと、150mMのNaClと、1mMのPMSFとを含む20mMのトリス-HCl(pH6.5)の中で溶解させた。ライセートを4℃にて10分間にわたってインキュベートし、13,000×gで5分間にわたって遠心分離し、核をペレットにした。次に上清を-80℃で凍結させ、さらに分析を行なうときまで保存した。精製したウイルス・サンプルは、SW41ベックマン・ロータ(25,000rpm、2.5時間、4℃)の中に入れた1.5mlの20%ショ糖クッションの上でウイルスの上清(8ml)を超遠心分離することによって得られた。ウイルスのペレットを100μlのPBSに分散させ、-80℃で凍結させた。
さまざまながんレトロウイルスまたはレンチウイルスのベクター成分をトランスフェクトそた12時間後、ウイルス産生細胞をシステインとメチオニンを含まない培地の中に1時間入れて飢餓状態にし、システインとメチオニンは含まないが、1mlにつき100μCiの35S-システインおよび35S-メチオニン(ICN)と、透析した2%ウシ胎仔血清とを含む3mlのDMEMの中で、37℃にて16時間にわたって標識した。細胞を溶解させ、以前に報告されている(9)ようにしてヤギの抗RD114 SU血清を用いて免疫沈降させた。TM GPサブユニットの細胞質尾部のプロセシングを分析するため、ウイルス産生細胞の上清を回収し、孔のサイズが0.45μmの膜で濾過した。SW41ロータ(ベックマン社)の中に入れた2mlの20%ショ糖クッション上で2時間にわたって30,000rpmで上清を超遠心分離した。溶解用緩衝液(50mMのトリスHCl(pH7.5)、15mMのNaCl、5mMのMgCl2、5mMのKCl、1%のトリトンX-100、0.5%のデオキシコール酸ナトリウム)を添加してペレットを溶解させた。ライセートの1/5を取ってそのまま電気泳動にかけ、ウエスタン・ブロット法でウイルスのタンパク質をおおまかに分析し、残りのライセートに対しては抗RD114 SU抗体を用いて免疫沈降を実施した。還元条件下にてドデシル硫酸ナトリウム(SDS)-12%ポリアクリルアミド・ゲルの中で免疫沈降物を電気泳動にかけ、SUから同時に免疫沈降したTM GPサブユニットを分離した。
以前に報告されている25ようにして、TE671を標的細胞として用いて形質導入の効率と感染力価を調べた。以前に報告されている36ようにして、ウイルス調製物と新鮮な血清が1:1(容積:容積)の混合物の中で37℃にて1時間にわたってインキュベートした後に生き残ったウイルス粒子を滴定することにより、ヒトとマカクの血清におけるベクター類型の安定性を調べた。点1つにつき、類型化されたベクター粒子を約5×104GFP感染単位使用した。文献36に記載されているようにして血清を健康な血液ドナーから採取し、調整した。ビリオンの安定性を、ウシ胎仔血清で処理したウイルスの感染力に対する霊長類の血清で処理したウイルスの感染力の割合として調べた。熱で不活化した血清(56℃、1時間)を対照として用いた。
抗体(ステムセル・テクノロジーズ社、メラン、フランス)と10ng/mlのトロンボポエチン(TPO;ペプロテック社、ロンドン、イギリス)を追加した2mlのステムスパンSFEM培地の中に細胞をウエル1つにつき2×106個植えた12ウエルのプレートの中で、精製したCD34+細胞を一晩にわたってインキュベートした。次に、あらかじめ活性化させたCD34+細胞を96ウエルのプレートに植え(104細胞/ウエル)、TPOと6μg/mlのポリブレンを含む合計体積が200μlのステムスパン培地の中で、類型化されたベクターを用いて形質導入した。TE671標的細胞を用いて決定したさまざまな感染多重度(MOI)を標的細胞に適用したところ、標的細胞1個当たりの感染粒子は0.5〜60個の範囲であった。レトロネクチンでコーティングしたウエル(CH-296;宝酒造、日本)での形質導入を、ウエル1つにつき8μgのレトロネクチンを2時間にわたってあらかじめコーティングした96ウエルのプレートの中で、同じプロトコルを利用して行なった。16時間後、CD34+細胞を洗浄し、10%ウシ胎仔血清(ライフ・テクノロジーズ社、フランス)と、抗生物質と、10ng/mlのFlt3-L、TPO、幹細胞因子(SCF)とを追加した400μlのステムセル培地の中に3日間分散させておいた。感染の5日後、GFPの発現をFACSによって分析した。
実施例1:さまざまなウイルス糖タンパク質がSIVベクターを類型化する能力
われわれは、一群のウイルス糖タンパク質(GP)がサル免疫不全ウイルス(SIVmac251)に由来するレンチウイルス・ベクターを類型化する能力を調べた。糖タンパク質は、タイプCの哺乳動物レトロウイルスに由来するもの(例えばネコ内在性レトロウイルスRD114、両種指向性ネズミ白血病ウイルス(MLV-A)、MLV-10A1、テナガザル白血病ウイルス(GALV)のEnv GP)、あるいは膜で覆われたウイルスに由来するもの(例えばトリ・インフルエンザ・ウイルス(FPVヘマグルチニン、FPV-HA)、リンパ球性脈絡髄膜炎ウイルス(LCMV)、エボラウイルス(EboV)、水疱性口内炎ウイルス(VSV)のGP)であった。類型化されたSIVベクターは、SIVウイルスのコア・タンパク質をコードしている3種類のプラスミド(図1)と、SIVをベースとしていてGFPマーカー遺伝子を含む輸送ベクターと、さまざまなGPとをトランスフェクトした293T細胞の中で一過性に発現させることによって作った。TEI671ヒト横紋筋肉腫細胞に関する感染アッセイにより、VSV、LCMV、MLV-A、MLV-10A1のGPを用いて作ったベクターで105i.u./mlを超える力価が得られることがわかった(図2A)。それとは対照的に、EboV、FPVのGPを用いて作ったベクターは力価が小さく、5×103i.u./ml未満であった。GALV、RD114のGPを用いて作ったSIVベクターでは力価が中間的な値であり、104〜5×104i.u./mlであった。類型化されたベクターの感染力のこうした相対的な違いは、他の標的細胞(例えば293T細胞)でも再現された(データは示さない)。これは、TEI6細胞に関する感染力価が、さまざまなGPがSIVコアを類型化する能力を反映していることを示唆している。
われわれは、修飾されたウイルス糖タンパク質または修飾されていないウイルス糖タンパク質でコーティングされたベクターのうちで、SIVベクター粒子を効率的に類型化したものの性質を明らかにすることを試みた。SIVベクター類型を超遠心分離によって濃縮し、1%のBSAを含む貯蔵用緩衝液の中に再び分散させ、等分し、-80℃で保管した後、感染アッセイを行なった。MLV-10A1 GPでコーティングされたベクターは濃縮前に正常な力価であったが、効率的に濃縮できなかったため(データは示さない)、その後の分析では使用しなかった。それとは対照的に、FPV-HA、VSV-Gの糖タンパク質で類型化したベクター、あるいはGALV/TR、RD114/TR、MLV-A、LCMVの糖タンパク質で類型化したベクターは、非常に効率的に濃縮されたため、物理的粒子を100倍に濃縮した後の感染性粒子の回収率は平均して80%を超えた(データは示さない)。FPV-HA、LCMVの糖タンパク質で類型化したベクターは、ここでテストした一次造血細胞(すなわちPBLとCD34+細胞;データは示さない)に形質導入することができなかったため、それ以上は分析しなかった。残った糖タンパク質(すなわちMLV-A、GALV/TR、RD114/TR、VSV-G)で類型化したベクターの濃縮ストックの感染力価は、TE671標的細胞を用いて測定した。その値は、感染力が小さい類型の5×106i.u./ml(GALV/TR GPで得られた)から、最も感染力の大きい類型の1×108i.u./ml(VSV-Gで得られた)までの範囲であった(図3A)。ベクター類型間での力価に関する同様の違いが、他のヒト接着細胞系で検出された(データは示さない)。これは、許容性の非常に大きなTE671細胞を用いた力価測定が、類型化されたベクターの特異的感染力を反映していることを示している。重要なことだが、感染性粒子の数は、物理的粒子の存在と相関していた。図3Bからわかるように、類型化されたベクターの調製物を相互に比較すると、似た量のビリオン関連キャプシド・タンパク質が、力価が最大であったベクター類型(VSV-G、MLV-A、RD114/TRのGP)において検出された。GALV/TR GPで類型化したビリオンでは、より少ない量の物理的粒子が繰り返して検出された。これは、その力価がより小さいことと整合性がある(図3A)。しかしビリオン関連キャプシド・タンパク質の絶対量における重要な違いは、感染力価がほぼ同じである2つの独立な調製物を比較するときに明らかになった(データは示さない)。そこでベクターのストックの品質差による誤りを最少にするため、同時に作った類型化されたベクターを用いて後で評価実験を行なった。さらに、ビリオン関連キャプシド・タンパク質の検出結果は感染性粒子の有効な指標であるようには見えず、結果の比較から除外したため、TE671細胞に関して測定した力価を用いて類型化されたベクターのストックを規格化した。
生体内の遺伝子デリバリーに適したベクターは、37℃で安定でなくてはならず、霊長類の血清中で大きな感染力を維持していなくてはならない。そこで37℃にて1時間にわたってインキュベートしたウイルス粒子の力価を4℃にてインキュベートした場合と比較することにより、ベクター類型の安定性を調べた。RD114/TR GPまたはVSV-Gで類型化したレンチウイルス・ベクターは37℃で安定であり、85%を超えるベクター粒子が37℃でのインキュベーション後に感染力を保持していた(データは示さない)。それに対してMLV-A GPとGALV/TR GPで類型化したベクターは、37℃でインキュベートした後は75%以上が感染力を失った(データは示さない)。これは、レンチウイルス・コア粒子に組み込まれた後者のGPが温度感受性を持っていたことを示唆している。
われわれは、次に、一次造血細胞(例えばCD34+細胞やPBL)に対するさまざまなベクター類型の形質導入能力を比較した。動員血に由来するヒトCD34+細胞を、TPOを追加した無血清培地の中で一晩にわたってあらかじめ活性化させ、MLV-A、GALV/TR、RD114/TR 、VSV-Gのいずれかの糖タンパク質で類型化したSIVベクターを用いて単一ヒットで16時間にわたって形質導入した。TE671細胞に関して評価した感染力価を用いて決まるさまざまな感染多重度(MOI)を利用し、CD34+細胞に形質導入した。CH-296レトロネクチン断片の存在下または不在下で、密接な関連のある複数の形質導入実験を行なった42-44。感染後、低濃度のTPO、SCF、Flt3-Lの存在下で細胞を5日間にわたって増殖させた。GFPの発現は、フローサイトメトリーにより、形質導入された細胞において容易に検出された。そのためMOIと類型化用GPが形質導入の効率に及ぼす影響を評価することができた(図5A)。レトロネクチンの不在下での形質導入に関しては、GFP+細胞の割合は最初のうちはMOIに比例して増加し、MOIが2と20の間では曲線が平坦になり、GFP+細胞の最大値25%に達した。形質導入効率がこのように中程度であったのは、感染プロトコルが最適ではなく、特に標的細胞をビリオンとともに1回だけ短時間しかインキュベートしなかったためであろう。このような実験条件では、最も効果的なベクターは、VSV-G GPで類型化したベクターであった(平均GFP+細胞:24.75%±3.23%;n=5)が、MOIが2未満では、GALV/TR GP、MLV-A GPで類型化したSIVベクターのほうが、VSV-Gで類型化したベクターよりも形質導入効率が大きかった(図5Aの挿入図を参照のこと)。しかしテストした最も効果的なMOIでは、MLV-A、GALV/TR、RD114/TRの糖タンパク質を用いたベクターの形質導入効率は、VSV-G類型と比べて1/5〜1/12であった(図5A)。GALV/TR GPを用いて作ったベクターの力価は比較的小さいため(図3A)、大きなMOIで形質導入効率を評価することはできなかった。レトロネクチンでコーティングしたプレート上でCD34+細胞に感染させたときには、異なる結果が得られた(図5A)。この条件下において、VSV-Gで類型化したベクターは、レトロネクチンの不在下と同じ最大形質導入効率(GFP+細胞が24.56%±3.27%;n=5)を保持していた。これは、別の結果とも一致している45。それとは対照的に、RD114/TRで類型化したベクターは、形質導入効率が10倍になり、GFP+細胞が65%に達した(平均:51.30%±8.74%;n=5)。これは、RD114/TR GPとレトロネクチンを組み合わせて使用すると感染が相乗的に増大することを示している。レトロネクチンは、GALV/TR GP、MLV-A GPで類型化したベクターの形質導入効率も増大させたが、RD114/TR GPで類型化したベクターと比べてその大きさははるかに小さかった(図5A)。
ネコ内在性ウイルスRD114は、その細胞内コア粒子の形状からわかるように、タイプCの哺乳動物レトロウイルスである(32)。しかしそのRD114のGPは、タイプDのサル・レトロウイルスに典型的なGPであり(28)、そのレトロウイルスと同じ細胞表面受容体RDR(31、38)を備えており、MPMV(メーソン-ファイザーサル・ウイルス)のGPと特にTMサブユニットにかなりの相同部を有する(図7)。われわれは、以前の報告においてRD114ネコ内在性ウイルスの糖タンパク質がレンチウイルス・コアと効率的に類型を形成できないことを見いだした。そこでわれわれは、この明細書で、SIV(サル免疫不全ウイルス)に由来するレンチウイルス・ベクターとの類型化を制限するRD114 GPの決定基を調べることにした。
GP発現細胞の培養物は、シンシチウムを形成した。この現象は、細胞質尾部に導入した突然変異のタイプに依存することがわかった(図8)。野生型MLV-A GPと比べると、修飾されていないRD114 GPそのものの発現によってかなりのシンシチウムの形成が誘導された。この効果は、RD114 CTによるGPの融合生成性が十分に制御されていないことによって引き起こされるようであった。というのも、RD114 CTをMLV-A CT(突然変異体RD/TR、RD/MTR、RD/eMTR)で置換するとトランスフェクトされた細胞内のシンシチウムの数が顕著に低下し、野生型MLV-A GPで検出されたレベルになったからである(図8)。他のRD114キメラGPは、さまざまなレベルのシンシチウムを誘導した。最大の細胞変性効果は、RDPrHIVとRDRのない突然変異体で見いだされた。修飾されていないRD114 GPと同じ程度の融合生成性を示す突然変異体RDPrSIVARLMは例外だが、他のキメラ(RDΔYXXL、RDPrMLV、RDPrSIVRQAG)は、野生型RD114 GPで得られたよりも高レベルのシンシチウム形成を誘導した(図8)。変化した細胞-細胞融合生成性は、元々RD114 GPの修飾と結び付いており、トランスフェクトされた細胞に存在する他のウイルス成分との相互作用の影響は受けなかった。実際、細胞が発現しているかいないかや、がんレトロウイルスまたはレンチウイルスのコア・タンパク質に関係なく、同じレベルのシンシチウムが検出された(データは示さない)。さまざまなRD114 GPキメラでGPの発現に変化が見られなかったため、その結果から、細胞-細胞融合生成性の制御に関してRD114 GPの細胞質尾部が果たす役割が明らかになった。CTを通じたこの融合制御は、タイプCとタイプDの他の哺乳動物レトロウイルス(例えばMLVやMPMV)のGPに関する融合制御を思い起こさせた(2、30、33)。RD114 GPの細胞質尾部の配列を修飾すると、MLV-A GPと同様に(14、17、43)、その融合抑制特性が変化するようであった。そこでウイルス粒子を形成する上で望ましくない可能性のあるRD114 GPの修飾によって誘導される細胞変性効果を最少にするため、産生細胞の上清に含まれるビリオンをトランスフェクション後しばらくしてから(すなわち36時間後に)回収した。
RD114 GP突然変異体がSIVmac251またはMLVをベースとしたベクターを類型化する能力を、野生型RD114 GP、MLV-A GP、VSV-Gと比較した。TE671標的細胞を用い、さまざまなGPの存在下で作ったベクターの感染力を評価した(図9)。修飾されていないMLVをベースとしたベクターはうまく類型化することができた(6)が、レンチウイルスをベースとしたベクターは効率的に類型化できなかった(35)。これは、われわれの以前の結果と一致していた。RDΔYXXL GP突然変異体を用いて作ったSIVベクターとMLVベクターの両方とも、修飾されていないRD114 GPで類型化したベクターと比べて力価が1/3〜1/4になっていた(図9)。RDRのない超融合生成性突然変異体を用いて作ったベクターの力価も、野生型RD114 GPを用いて作ったベクターの力価より低かった。しかしわれわれは、この突然変異体によって及ぼされる特に強い細胞変性効果(図8)がベクター粒子の最適な形成を妨げないことを否定はできなかった。重要なことだが、RD/TRキメラGPとRD/MTRキメラGPは、SIVをベースとしたベクターを効率的に類型化した。その結果、RD/TR突然変異体の場合には、感染力価が106i.u./mlよりも大きくなった。すなわち、野生型RD114 GPで得られた値の25倍に達し、MLV-A GPまたはVSV-Gで類型化したSIVベクターで得られた感染力価よりも大きくはないにしても同程度の値になった(図9B)。それとは対照的に、同じキメラGPを用いて作ったMLV由来のベクターは、すでに力価が大きい野生型RD114 GPで類型化したベクターと比べて感染力がほんの2倍にしかならなかった(図9A)。細胞質尾部は、これらRD114 GPキメラ中で類型化されたレンチウイルス・ベクターの感染力を大きくする修飾された最少ドメインであるため、これらのデータから、MLV-AのTMDではなくCTが、SIVベクターを用いた類型形成を容易にする要素を備えていることが示唆された。RD114 GPキメラをさらに調べることにより(図7B)、この要素が、そのキメラの細胞質尾部内の想定開裂部位を含む領域に位置していることが明確になった。
われわれは、次に、RD114 GPの修飾が、ウイルス粒子へのそのRD114 GPの組み込みに影響を与えるかどうかを調べることを試みた。この実験は、修飾されていないRD114 GPを、シンシチウムの形成を極端に多くは誘導しなかったGP突然変異体であるRDPrMLV、RDPrSIVARLM、RDPrSIVRQAG、RD/TR(図8)のサブセットと比較するために実施した。実際、他の突然変異体(RDPrHIV、RDRなし)では細胞-細胞融合生成活性が大きいために産生細胞の上清に細胞のごみが放出されることにより、精製されたビリオン調製物の品質が損なわれていた(データは示さない)。RD114 GPキメラを用いてSIVコアまたはMLVコアを有するウイルス粒子を作り、20%ショ糖クッションを通じて超遠心分離することによってそのウイルス粒子を精製した。ウイルスのコア・タンパク質と糖タンパク質の両方を検出し定量する操作を、抗CA抗体または抗RD114 GP抗体を用い、精製したビリオンの免疫ブロッティングを行なうことによって実施し、ウイルス粒子上のRD114 GPキメラの密度を測定した(図4)。ウイルス・コアの不在下で糖タンパク質だけを発現した産生細胞からの上清を超遠心分離して得たペレットの中では、GPを検出することができなかった(図4)。したがって、精製したビリオンで得られたシグナルに特異性があることが証明された。野生型RD114 GPと比較すると、RDPrSIVARLMGP突然変異体も、このGPによって感染力が大きくなるにもかかわらず、SIVコア粒子にうまく組み込まれた(図9B)。それとは対照的に、SIVベクター粒子へのRDPrMLVキメラGP、RDPrSIVRQAGキメラGP、RD/TRキメラGPの組み込みは3〜10倍になった(図4)。これは、これら突然変異体GPで非常に大きな力価が得られたことと整合性がある(図9B)。したがって、これらの結果は、SIVコア粒子へのRD114 GPキメラの組み込みと、類型化されたベクターの感染力価とが相関していることを示している。ただしRDPrSIVARLMGP突然変異体の場合は、絶対的相関からはずれていた。
われわれは、次に、RD114 GPキメラの細胞質尾部が類型化されたMLVウイルス・コア粒子またはSIVウイルス・コア粒子において開裂されるかどうかを調べた。類型化されたこれら2つのビリオンのうちのどちらかを産生するトランスフェクトされた細胞に、35S-メチオニンと35S-システインで放射性標識した。ビリオン産生細胞のライセート、または20%ショ糖クッション上で精製したビリオンのライセートを、抗RD114 SU抗体とともにインキュベートした。糖タンパク質を免疫沈降させた後、サンプルを還元し、変性させ、免疫沈降したGP複合体のTMタンパク質を分解させた後、SDS-PAGEで分析した(図5)。ビリオン産生細胞のライセートには、予想されるように、SU糖タンパク質とTM糖タンパク質の両方がほぼ同じ量含まれていることがわかった。すべての突然変異体のTM糖タンパク質は、ゲル上で電気泳動の移動度が同じであった(図5A、図5B)。これは、異なる種類のTMのCTの大部分が、産生細胞の内部で開裂しなかったことを示している。これらの結果から、RD114 GP突然変異体の細胞-細胞融合生成性の変化はそのTMタンパク質のプロセシングに依存しておらず(図8)、本質的にその細胞質尾部の構造または配置の変化と結び付いていることも確認された。
最近の報告によれば、レンチウイルスは、浮遊脂質から選択的に集合して出芽することが明らかになった(18、25、26)。したがって、RD114 GPの細胞質尾部が修飾された結果、ウイルス・アセンブリー部位において、ウイルスのさまざまなコア成分と糖タンパク質の同時局在状態が変化したというのが、類型化されたSIV粒子にいろいろなRD114 GP突然変異体が異なった組み込まれ方をすることを説明するための1つの可能性である。そこでわれわれは、RD114 GPが浮遊脂質に局在する可能性があるかどうかと、そのGPの細胞質尾部が修飾されることによって細胞の局在が変化する可能性があるかどうかを調べた。浮遊脂質は、低温で非イオン洗剤に対して抵抗力があり、ショ糖勾配遠心分離によって可溶性である膜の塊から物理的に分離することができる。さまざまなGP突然変異体をトランスフェクトした細胞の膜をこのような条件下で分画し、膜分画のGP含量をウエスタン・ブロットにより分析した(図6)。野生型RD114 GPと細胞質尾部突然変異体が、トランスフェクトした細胞のライセートの洗剤抵抗性分画の中に現われた。これは、すべてのGPが浮遊脂質に向かったことを示している。

(c)その産生細胞を培地内で培養するステップと;
(d)その産生細胞を培地から分離することにより、その培地から組み換えウイルス・ベクター粒子を回収するステップを含む方法。 - 細胞に形質導入する方法であって、その細胞の形質導入が組み換えベクターによってなされる条件下で、請求項27に従って製造したベクター粒子とその細胞を接触させるステップを含む方法。
- 上記細胞がヒトの細胞である、請求項28に記載の方法。
- 上記細胞が造血幹細胞である、請求項29に記載の方法。
- 上記細胞がヒトのCD34+細胞である、請求項30に記載の方法。
- 細胞に対し、幹細胞の多能性が実質的に失われることなく細胞増殖が促進されるような処理を行なう、請求項30に記載の方法。
- 上記細胞に生体内で形質導入を行なう、請求項28に記載の方法。
- 上記細胞に試験管内で形質導入を行なう、請求項28に記載の方法。
- 形質導入された上記細胞を、対象とする動物に導入する、請求項34に記載の方法。
- 上記対象がヒトである、請求項35に記載の方法。
- 造血疾患またはリンパ造血疾患の治療を目的とした医薬を製造における、請求項15〜17のいずれか1項によるレンチウイルス・ベクターの使用。
