Abstract
Description
本発明は、標的遺伝子の発現を阻害するためのショート・インターフィアリングRNA(short interfering RNAs:siRNAs)を発現するためのポリヌクレオチドまたはベクターに関する。また、本発明は、ポリヌクレオチドまたはベクターを含む細胞および非ヒト・トランスジェニック動物、並びに標的薬物バリデーションにおける、およびヒト薬物療法学におけるものを含むこれら種々の使用にも関する。
特定の遺伝子の機能を阻害または破壊させる能力は、遺伝子機能を研究する観点から、更に治療的な展望からも、たいへん望ましい。

本発明の構築物のsiRNAの発現は、RNAポリメラーゼのプロモーターによって駆動される。このようなプロモーターは、典型的には特定の遺伝子の高レベルの発現を生じさせることができ、RNAポリメラーゼIIプロモーターによって達成可能なレベルを上回って優れていることが多い。この高レベルの発現により、標的遺伝子の高レベルの阻害が達成されることを確認するのを助けることができる。
転写終結エレメントは、本発明のポリヌクレオチドに含まれる。転写ターミネーターは、siRNAをコードする領域の下流にあり、および好ましくは、該領域をコードするすぐ下流にあるか、または最小の距離で分離されている。
本発明のポリヌクレオチドは、siRNAをコードする領域を含む。siRNAは、配列が標的遺伝子と同一である二重鎖の領域を含む短い二重鎖RNA分子を意味する。siRNAは、特定の標的遺伝子をサイレンシングまたは阻害することができる。
本発明のポリヌクレオチドは、単一のポリヌクレオチドとして、または代わりにベクターの形態で提供されてもよい。好ましくは、これらは、プラスミドなどのベクターの形態で提供される。このようなベクターは、これらを原核または真核細菌系において大量に再生し、次いで標的細胞に導入することができるように、シャトルベクターであってもよい。
標的遺伝子は、その機能を阻害するか、または調整することが要求される任意の遺伝子であってもよい。阻害の目的は、治療的または特定の遺伝子の機能を研究するためであってもよい。商業的に育てられた動物の形質を改善するなどのために、遺伝子を阻害して、いくつかの所望の方法で細胞の表現型または生物体を変化させてもよい。典型的には、標的遺伝子は、真核細胞遺伝子であるが、あるいは標的遺伝子は、真核生物の宿主細胞に発現されているウイルス遺伝子などの原核生物のものであってもよい。標的遺伝子は、ポリペプチドまたは代わりに構造または酵素RNAをコードしてもよい。しかし、好ましくは、標的遺伝子は、ポリペプチドをコードする。
本発明のポリヌクレオチドは、特定の対立遺伝子の発現を阻害し、一方でその他の対立遺伝子の通常の発現を可能にするために使用されてもよい。このような態様において、遺伝子の2つの対立遺伝子は、配列に一部の相違を有し、これによりsiRNAによって区別することができる。
本発明の系は、種々の細胞および生物体に発現する遺伝子を阻害するために使用されてもよい。また、系は、これらの宿主細胞内の発現またはウイルス遺伝子を阻害するために使用されてもよい。典型的には、系は、真核細胞および生物体、並びに特に哺乳類の細胞または生物体における発現を阻害するために使用される。
本発明の多くの態様において、標的遺伝子の発現をブロッキングする際に、siRNAsの効力を点検することが要求されると考えられる。種々の方法で、典型的にはRNA、タンパク質、または表現型のレベルで、遺伝子の阻害を測定してもよい。
本発明の好ましい態様において、本発明のポリヌクレオチドまたはベクターは、標的細胞のゲノムに組み込まれる。これは、siRNAの発現が、非組み込みベクターに関連した一過性の発現よりも、恒久的であることを確実にするのを助ける。
本発明のポリヌクレオチドは、標的遺伝子の発現が阻害され、または減少した非ヒト・トランスジェニック生物を作製するために使用されてもよい。トランスジェニック動物は、好ましくはゲノムに組み込まれた本発明のポリヌクレオチドまたはベクターを有し、それゆえに、組み込まれたポリヌクレオチドまたはベクターをその子孫に伝えることができる。しかし、本発明は、本発明のポリヌクレオチドまたはベクターを含む細胞が移植され、または伝達された正常な動物を包含する。このような動物は、生体外の治療を含む特定の治療のためのモデルを提供するであろう。
本発明の方法により、種々の疾患症状および障害のモデルの作製することができる。たとえば、これらを使用して、特定の遺伝子が阻害された株化細胞または生物体を作製してもよい。また、これらを使用して、選択した内在性の遺伝子の両方のコピーが阻害され、かつ内在性の遺伝子の変異された対立遺伝子が発現され、そして常染色体優性症状または癌などの症状を非常にモデリングするモデルを作製してもよい。
本発明は、候補薬剤をスクリーニングし、問題の症状を予防し、治療し、または改善することができるものを同定するために使用するための、本発明の細胞または動物モデルの使用を提供するする。また、標的のバリデーションに本モデルを使用して、症状に潜在的に治療的価値を有すると考えられる候補薬剤をさらに特徴づけ、または候補遺伝子が障害に関与することを確認してもよい。
本アッセイ法は、所望の薬剤がリポーター遺伝子の、または選択可能なマーカーの発現を引き起こすものであってもよい。また、これにより、多くの試験薬剤のスクリーニングが促進され、所望のクローンを同定することが容易になるであろう。本明細書で言及した任意の選択可能なマーカーおよび既報告の遺伝子をこのような態様で使用してもよい。
また、本発明のベクターは、siRNAsの大規模なコレクションを作製して、生物学的に関連した経路で作用する遺伝子のゲノムワイドなスクリーンを行うために使用することができる。したがって、本発明を使用して、siRNAsのライブラリーを作製することができる。このようなライブラリーを使用する遺伝子「機能喪失表現型」スクリーンにより、薬剤開発の候補である新規の治療標的が得られる可能性があり、または生物反応に対する限定された数の候補遺伝子の貢献を評価するために使用してもよい。
本発明のスクリーニング法を使用して同定される種々のポリヌクレオチド、ベクター、株化細胞、および薬剤は、治療または診断によるヒトまたは動物体の治療方法に使用してもよい。これらは、特定の疾患症状または感染症状を予防する、治療する、改善する、または診断するために使用してもよい。
哺乳類細胞内に合成の短い干渉RNA(siRNAs)を導入することにより、特定の遺伝子の発現を有意に抑制することができる。しかし、この遺伝子発現の減少は、過渡的である。
本発明の方法のp53を阻害する能力を評価した。腫瘍抑制因子p53は、電離放射(IR)に続いて安定化され、DNA損傷に続く細胞周期のG1停止の維持において重要な役割を演ずる転写制御因子である(Agami & Bernard, supra andPluquet, & Hainaut, Cancer Lett 2001,174, 1-15))。
本発明の方法が特定の対立遺伝子の発現を抑制する能力を評価した。
本発明の方法が、さらなる遺伝子CDC20の発現を阻害する能力を評価した。
p53 mRNAの安定度に対するpSUPER-p53ベクターの効果を検査した。MCF-7細胞をpSUPER-p53またはベクターで電気穿孔し、総RNAを48時間後に抽出した。30μgのRNAをアガロースゲルで分離し、ブロットして、32pラベルされたp53特異的なプローブによってプローブした。充填の対照として、リボソームRNAの対照をブロットを臭化エチジウム染色によって視覚化した。得られたノーザンブロットおよびゲルRNA充填の対照は、図5(A)に示してある。pSUPER-p53ベクターをトランスフェクトした細胞は、空のベクターpSUPERをトランスフェクトした細胞と比較して、実質的に減少したp53 mRNAレベルを有する。
ヒト癌細胞における腫瘍化表現型に対する発癌性RAS発現の阻害の効果を研究するために、内在性の変異体K-RASV12対立遺伝子の発現を、ヒト膵臓株化細胞CAPAN-1のpSUPERベクターでターゲットした(図8A)。特に、変異体K-RASV12対立遺伝子をターゲットするために、変異体K-RAS癌遺伝子のバリン12をコードする領域にわたる19ntのターゲティング配列をpSUPERベクターにクローン化し、pSlJPER-K-RASV12を得た。pS-K-RASV12を作製するために使用した2オリゴは、以下の通りである:

19ntのK-RASV12標的配列は、大文字であり、K-RASのアミノ酸12にグリシン-バリン置換を生じるG-T変異は、太字である。これらのオリゴをアニールさせて、BglIIおよびHindIIIで消化したpSUPERベクターに適合する末端を有するインサートを作製した。図8Bは、pSUPER-K-RASV12を一過性にトランスフェクトしたCAPAN-1ヒト膵癌細胞が内在性のK-RASV12発現の有意な抑制をもたらしたが、対照サイクリンDタンパク質は影響を受けなかったことを示す(図8B)。
本実施例は、哺乳類細胞のゲノムDNAへのノックダウンベクターの遺伝子特異的なインサートの組込み、その後のこのようなベクターのPCR増幅、およびマイクロアレイでの増幅産物のハイブリダイゼーションに関する。
複合プローブ混合物(たとえばcDNA)のハイブリダイゼーションがマイクロアレイ技術を使用して頻繁になされているという事実は、上記の技術を多くのベクターでの実施に拡張することができることを強く示唆する。重要なことに、プローブ並びにマイクロアレイにスポットされたオリゴヌクレオチドの自己相補的な性質では、強い特異的なハイブリダイゼーションシグナルを阻止しない。これはまた、同時にバーコード・タグの混合物の特異的なハイブリダイゼーションを刺激するハイブリダイゼーション条件が存在することを示す。
本実施例は、レンチウイルスベクターの構築、およびRNA干渉を介してp53発現の安定な抑制を媒介するp53特異的な短いヘアピン転写物の合成の誘導におけるその使用に関する。
プラスミドの構築
マウスのp53特異的な短いヘアピン・オリゴヌクレオチドを、まずpRETRO-SUPERにクローン化した(本明細書に記載されているように)。pRETRO-SUPERベクターをBglIIおよびHindIIIで消化して、マウスのp53をターゲットするアニールしたオリゴの
5'gatccccGTACATGTGTAATAGCTCCttcaagagaGGAGCTATTACACATGTACtttttggaaa3'、および、
5'agcttttccaaaaGTACATGTGTAATAGCTCCtctcttgaaGGAGCTATTACACATGTACggg-3'、
をベクターにライゲーションして、pRETRO-SUPER-mp53を得た。オリゴヌクレオチド内の19merのp53ターゲティング配列は、大文字で示してある。レンチウイルス導入ベクターのHIV-CS-CG(Miyoshiら、(1998). J Virol 72,8150-8157)をEcoRIおよびXhoIによって消化し、CMV-GFP配列を除去した。H1プロモーターおよびp53標的配列を含むカセットをpLENTI-SUPER-p53からEcoRIおよびXhoIで切除し、HIV-CSにライゲーションしてpSUPER-mp53を得た。
野生型FVBマウス胚性線維芽細胞(MEFs)、ST.HdhQ111マウス線条体細胞(Trettelら、(2000). Hum Mol Genet 9, 2799-2809)、および293T細胞は、10%のウシ胎児血清を補ったダルベッコ修飾イーグル培地(DMEM)中で培養した。レンチウイルスの産生のためには、293Tの細胞に、10μgの導入ベクターHIV-CS-CGまたはpLENTI-SUPER-mp53、3.5μgのVSVgエンベロープベクターpMD.G、2.5μgのRSV-Rev、および6.5μgのパッケージングベクターpCMV□R8.2(Miyoshiら、(1998). J Virol 72,8150-8157)を使用して、カルシウム-リン酸法によってトランスフェクトした。レンチウイルスをトランスフェクション24時間および48時間後に回収して、0.45μMフィルターを通してろ過した。レンチウイルスの感染の14日前にST.HdhQ111細胞を39℃へ移した。WT MEFsは、レンチウイルスの感染の14日前に、細胞を3〜4日ごとに計数することにより、9〜10継代培養した。また、感染時に、酸性βガラクトシダーゼ染色によって老化の表現型を調査した(Dimriら、(1995). Proc Natl Acad SciUS A92, 9363-9367)。6cmのディッシュ内の1.8×105の老化WT MEFsには、0.8μg/mlポリブレンの存在下で、少なくとも12時間レンチウイルスを感染させ、次いでコロニー形成アッセイ法および増殖曲線のために再びまき直す前に48時間回復させた。コロニー形成アッセイ法のために、0.5×105または1×105個の細胞を10cmのシャーレにまいた。播種の16日後に、細胞を固定してスーバーステイン(50%のメタノール、10%の酢酸、0.1%のクマシーブルー)で染色した。増殖曲線のために、1.5×103個の細胞を3.5cmのシャーレにまいて、3日間隔で細胞を0.5%のホルムアルデヒドで固定し、0.1%のクリスタルバイオレットで染色し、続いて10%の酢酸中に再び可溶化した。細胞数の相対的な測定としてOD590を定量した。
全細胞抽出物を12%のSDS-PAGEゲルに分離して、ポリフッ化ビニリデン膜(Millipore)に転写した。増強化学発光(Amersham Biosciences, Inc.)を使用して可視化した。使用した抗体は、p16INK4aに対するM-156(Santa Cruz)、19ARFに対するab80-50(Abcam)、p21に対するF-5(Santa Cruz)、p53に対するAb-7(Oncogene)、およびP30620(Transduction labs)に対するPAI-1である。
5×104の老化MEFsを3.5cmのシャーレにまいて、レンチウイルスに感染させた。微速度顕微鏡観察は、温度およびCO2制御されたチャンバ内で感染の234時間後に、10×位相差を使用して開始した。フレームは、38時間の期間にわたって20分ごとにとった。
本明細書に記載されたpRETRO-SUPERベクターのレンチウイルス誘導体は、pRETRO-SUPER由来のマウスp53pをターゲティングするH1 RNAショート・ヘアピン遺伝子発現カセットを、自己不活性化するレンチウイルスベクターのpHIV-CSにクローニングすることによって作製した(Miyoshiら、(1998). J Virol 72,8150-8157)。このベクターは、pLENTI-SUPER-p53と名づkeた(図11A)。対照として、GFP(HIVCS-CG)を発現するレンチウイルスベクター(Miyoshiら、(1998). J Virol 72,8150-8157)を使用した。
