本発明は、一般に生体分子を単層を有する商品上に不動化することに関する。本発明は、商品、この商品の製造法、不動化された生体分子の密度の制御法、及びこの密度を特徴決定する方法にも関する。
ポリペプチド、オリゴヌクレオチド、炭水化物、脂質及び細胞のような生物学的物質を、表面上に不動化することには、このような表面結合した物質の可能性のある適用のために非常に興味深い。例えばこれらの物質は、医学及び薬学研究のための診断用装置及び機器において有用であることができる。従ってタンパク質のような物質を表面上に不動化する様々な技術が研究されている。Blawas, A.S.;Reichert「Protein Patterning」、Biomaterials、19:595-609 (1998)の表1及び引用された参考文献を参照のこと。
国際公開公報第98/30575号は、巨大分子が他の分子実体に複合する方法として、ディールスアルダー反応を開示している。
本発明は、貨幣金属表面及び貨幣金属表面の少なくとも一部を被覆する混合型自己組織化単層表面を有する商品を提供し、この混合型自己組織化単層表面は、第一の単層部分及び第二の単層部分を含み、第一の単層部分は、共有結合形成する反応基を生じるチオラートを含み、並びに第二の単層部分は不活性基を生じるチオラートを含む。
第二の単層部分の不活性基は、ポリエチレングリコールのような、生体分子の非-特異的吸着に抵抗する。
別の態様において、R2及びR3は各々、線状であり、並びに硫黄原子に結合した第一のアルキルセグメント、並びにこのアルキルセグメントに結合したポリアルコキシ、ポリペルフルオロアルキル、ポリ(ビニルアルコール)及びポリプロピレンスルホキシドからなる群より選択される第二のセグメントで形成される。R2は、式:-(CH2)m-(O(CH2)n)o-NHC(O)-(CH2)pであって良く、ここでmは10〜24の数であり、nは2であり、oは1〜10の数であり、及びpは1〜16の数である。R3は、式:-(CH2)i-((CH2)j-O)k-であって良く、ここでiは10〜24の数であり、jは2であり、及びkは1〜10の数である。Wは、ヒドロキシル、スルホネート、ヒドロキシ置換されたC1-C4アルキル又はメチルであることができる。

好ましい混合型単層表面は、第一の単層部分の第二の単層部分に対する予め定められた比を有する。
本開示において使用したいくつかの用語は、以下に示した意味を有する。
「アグリコン」は、糖質でないグリコシドの構成要素である。
「非対称」ジスルフィドは、2個の異なるチイル(thiyl)ラジカルのイオウ原子の共有結合により形成されたと見なすことができるジスルフィドである。
「貨幣金属」は、金、銀、白金及び銅である。
「誘導体」は、単純な化学的方法により他の化合物から得ることができる化合物である。Grant及びHackh、「Chemical Dictionary」第5版(1987)。
「リガンド」は、別の分子に特異的に結合する分子である。
「マイケル受容体」は、マイケル付加(Michael addition)に参画することができる電子求引基に共役炭素-炭素二重結合を有する求電子化合物である。マイケル受容体は、キノン、マレイミド、α-β不飽和ケトン、α-β不飽和アミド、及びα-β不飽和エステルを含むが、これらに限定されるものではない。
本明細書において使用される「小分子」は、分子量1,000ダルトン又はそれ未満を有する小さい有機又は非-有機構造を意味する。
用語「チイル断片」は、ジスルフィド基のイオウ原子のひとつ及びそのイオウ原子に結合した置換基を含むジスルフィドの断片(同義語、部分、ラジカル)を意味するように使用される。
この溶媒は、貨幣金属表面と接触された場合に、ジスルフィドが単層を形成することを可能にするものである。エタノール及びメタノールが、ふたつのこのような溶媒である。当業者は、作業することができる他の溶媒を認識しかつ理解するであろう。例えば、DMSOのよう溶媒は、単層を形成することができないので適していない。
不純物のために、ある比を有する溶液の異なるバッチは、表面上に常に同じ密度を生じるわけではないことに注意することは重要である。従って表面の特徴決定に関連している本発明の方法は、溶液の新たなバッチが作成される度に表面の密度を特徴決定するために使用されなければならない。
好ましい方法において、炭水化物は、単層表面上の共有結合形成する反応基(例えばマレイミド)と反応するであろうチオール化学タグを有するように誘導体化される。この方法において、連結化学は選択的であり、従って炭水化物上の又はその内部の官能基との非-特異的化学反応が発生しないことを確実にすることを助ける。このような選択的連結化学は、炭水化物の配向した不動化を生じる。これは、例えば、不動化した炭水化物に対する特異的タンパク質結合を試験する場合に利点であり、その理由は炭水化物との全ての結合相互作用は、炭水化物との特異的タンパク質結合の結果であり、非特異的結合に起因しないからである。
このチオール-炭水化物は、マレイミドを提示しているSAMの単層へ共有的に結合することができる。図11参照。好ましくは、マレイミド提示しているSAMは、不活性トリ(エチレングリコール)マトリックス内に約0.1〜約50%の密度でマレイミドを含む。より好ましくは、マレイミド密度は約1%〜約10%である。
この誘導体化反応は、メタノール又は水のような溶媒中において行われ、並びに好ましくはpH範囲が約pH4〜約pH10で行われる。使用される溶媒は、好ましくはメタノール又は水である。より好ましくは、pHは約5〜約8である。最も好ましくは、pHは約6.0である。最適な温度範囲は、約0℃〜約60℃である。より好ましくは、最適温度範囲は、約10℃〜約40℃である。誘導体化反応後、この表面は、好ましくは水及びエタノールで洗浄され、並びに窒素流れ下で乾燥される。
炭水化物の不動化を提供することにより、本発明は、炭水化物-ベースのアッセイ表面を提供する。例えば、グリコシルトランスフェラーゼアッセイのような、様々な酵素アッセイにおいて、不動化されたチオール-炭水化物を使用することができる。図12及び実施例4を参照のこと。
放射標識した供与体糖質は、好ましくは約0.1uM〜約1000uMである。好ましくは、この濃度は、約1uM〜約10uMである。グリコシルトランスフェラーゼアッセイのための好ましい温度は、約0℃〜約60℃である。より好ましくは、この温度範囲は、約20℃〜約40℃である。
ベースプレート上に不動化された炭水化物基質のグリコシル化の検出は、放射能測定により行われた。14Cの取込みは、貯蔵リン光体スクリーン(storage phosphor screen)をプレートに露光することにより定量し、その後これをVariable Mode Imager (Typhoon 8600;Molecular Dynamics社、サニーベール、CA)上に造影した。
更に融合タンパク質の反応基は、抗体であることができる。この態様において、二官能性アフィニティータグの第二の反応基は、この抗体の抗原標的である。この抗体及び標的抗原は、抗体-抗原会合において会合し、融合タンパク質を不動化するであろう。あるいは、融合タンパク質の反応基は、標的抗原であっても良く、及び二官能性アフィニティータグの第二の反応基は、標的抗原を認識する抗体であっても良い。本明細書において使用される抗体は、抗体全体である必要はない。Fab、Fab2、Fv、又はScFvなどのような、抗体断片であってもよい。
前述の方法において、好ましい態様での、融合タンパク質の共有結合形成する基はクテナーゼであり、及び二官能性アフィニティータグの第一の反応基はチオールである。二官能性アフィニティータグの第二の反応基は、p-ニトロフェノールリン酸("PNPP")である。この態様においてPNPPは、クテナーゼに共有的に結合し、融合タンパク質を不動化するであろう。
ベクターは、様々な生物及び/又は細胞へ導入することができる。あるいは、ベクターは、例えば、T7プロモーター調節配列及びT7ポリメラーゼを用い、in vitroにおいて転写及び翻訳することができる。
従ってひとつの態様において、二官能性アフィニティータグの第二の反応基は、p-ニトロフェノールリン酸基であり、及びこの融合タンパク質の共有結合形成する基は、クテナーゼタンパク質である。
図17は、本発明の実施態様に従う、生物学的物質のアレイ形成のための装置を示す。図17(a)は未組立図を示す一方、図17(b)は組立図を示す。
装置100の要素は、追加的に多くの変形及び態様をとるこができ、そのいくつかは本明細書において説明されており、他のものは本明細書に提供された指針が与えられることで当業者には明らかであろう。これらの図面を通して、類似の要素には、異なる態様に含まれる場合であっても同じ番号を付けた。
表面111及び/又は表面131のための好ましい材料は、生物学的に不活性であるか及び/又は非-特異的反応によりタンパク質のような生体分子の吸着に抵抗可能なものである。非-特異的吸着は、関心のある特異的生体分子を不動化する能力及び/又は特異的位置で生体分子を不動化する能力を妨害するので、避けるべきである。
別の例として、位置合わせピンに関連して一方の軸上で伸長している位置合わせオリフィスを、取り外し可能な部材内に配置することができ、及びそのようには伸長していない対応する位置合わせオリフィスは、ベースプレート及びその上のいずれかの層内に配置することができる。
本発明の取り外し可能な部材のシール可能で、取り外し可能で、及び再シール可能な特性は、本発明の装置を用いて行われたアッセイ並びにアッセイ結果の読取り、観察及び測定の両方の性能を促進する。
バルクプロセス工程として行うことができる他のプロセス工程の例は、洗浄液、試薬溶液、生体分子溶液中の浴技術による含浸;物質の抗体溶液とのインキュベーション;未結合の抗体又は他の物質を除去するための洗浄工程;不動化された物質を固定するための固定工程;シグナル分子の添加、又はシグナル分子の、それらに例えば非-呈色から呈色もしくは非-蛍光から蛍光へのような変化を生じるような物質(例えば「発色剤」)への曝露のような発色工程;光が補助する化学などを含むが、これらに限定されるものではない。
プロセス工程及び/又は検出工程の後、取り外し可能な部材140は、表面111又は表面131上に再シールし、ウェル145を再形成することができる。ウェル145が再形成される場合は、追加のプロセス工程を、ウェル底147上に不動化された物質に個別に行うことができる(及び、検出工程を行うことができることは理解されるであろう)。取り外し可能な部材140を表面111又は131にシールし、及び物質を不動化し、及び/又は個別のウェル底(複数)147上に不動化された物質に工程を行うこと;取り外し可能な部材140を取り外し、及び個別のウェル底(複数)147に不動化された物質にプロセス工程又は検出工程を行うこと;並びに、取り外し可能な部材140を、表面111又は113に再シールし、及び個別のウェル底(複数)147に不動化された物質に工程を行うサイクルを、望ましい限り複数回反復してもよい。
液密シールの使用は、プレートの表面全体が個別の領域595へと再生可能に修飾されることを可能にし、これは剛性プレートにおいては、各ウェルに液体をピペッティングすることを伴わずに達成することはできず、従ってこれらのウェル内で夾雑する可能性がある。
図23(c)に示された取り外し可能な部材740は、取り外し可能な部材140毎に実質的に同じ構造、組成及びシール特性を有し、並びに同様の方法及び物質を使用し製造することができる。
図24に認められるように、ウェルオリフィス843は、表面111、131、141、及び841により規定された平面中のウェルオリフィスよりも大きく、及びウェルオリフィス843は、再シール可能な部材141及び841が互いに接触するように配置された場合に、1個よりも多いウェルオリフィス143を包含している。
例えば、図24に示されたように、取り外し可能な部材140は、表面131にシールされ、並びに取り外し可能な部材840は、表面141にシールされる。ウェル底147を有する複数のウェル145は、ウェルオリフィス143及び表面141により形成される。複数のウェル845も形成される。ウェル845の壁は、ウェルオリフィス843により形成され、並びにウェル845の底は、表面141及び表面131により形成される。ウェル845は、1個よりも多いウェル145を包含している。
多くの好ましい態様において、取り外し可能な部材140及び取り外し可能な部材840は、アッセイ又は手法の経過の途中で1回又は複数回、交互にシールされ、取り外され、及び表面131へ再シールされる。
再度、取り外し可能な部材140及び/もしくは第二の取り外し可能な部材840のシール、取り外し、及び再シール、物質の不動化、並びに/又は個別のウェル底(複数)147及び/もしくは847上に不動化された物質に対するプロセス工程及び/もしくは検出工程を行うことは、望ましいならば複数回繰り返すことができる。そこでオリフィスの異なる配列を有する追加の取り外し可能な部材が、本発明に従い集成され及び使用され得ることは理解されるであろう。
図25に認められるように、ウェルオリフィス843及びウェルオリフィス143の寸法及び形状は、第二の取り外し可能な部材840が表面141にシールされる場合に、ウェルの壁144及びウェルの壁844は、実質的に滑らかかつ連続した表面を形成し、その結果表面141の一部が実質的に露出されないように選択することができる。
装置1000の上側集成体1090は、特徴、材料、及び製造が、ベースプレート110に類似したベースプレート1010;特徴、材料、及び製造が、層120に類似した層1020;並びに、特徴、材料、及び製造が、ベースプレート110に類似した層1030を具備している。取り外し可能な部材141は、取り外し可能な部材140が表面131に結合することが可能であるのと同じ方法で、表面1031にセルフシールすることが可能である。
図27に見られるように、集成体1180及び1190は、互いに同じフットプリントを有することが好ましい。取り外し可能な部材1140の表面1141が取り外し可能な部材140の表面141にシールされる場合、ウェル145(取り外し可能な部材140及び表面131により形成)及びウェル1145(取り外し可能な部材1140及び表面1131により形成)は、複数の液密流路1170を形成し、並びに互いに流動的に連結され、及び外側周囲から流動的にシールされている。
同様に、本発明の装置は、ベースプレートと同じフットプリントを有さないが、むしろベースプレートよりもより小さいフットプリントを有するような取り外し可能な部材を具備することもできる。このような装置の例は、図30に示されている。
装置2500を用い、本明細書により詳細に説明されているもののような、アッセイ及び方法を行うことができる。
図43に見られるように、オリフィス2743の空間的配列及び形状は、表面131及び111により規定されたウェル底133の配列及び位置に対応しており、その結果部材2740が表面131にシールされている場合、各ウェルオリフィス2743はひとつのウェル底133に包含される。
非-平面のベースプレートは、いずれか望ましい幾何を有することができる。例えば図45(a)に示されたように、非-平面のベースプレート100は凹であるか、又は図45(b)に示されたように、非-平面のベースプレート100は凸であることができる。
自己組織化単層(SAM)の使用は、物質、特に生体分子の不動化及び/又はパターン化にとって好ましい方法を提供する。SAMは、最も広範に試験され、最も良く開発された非生物学的自己集合システムの例である。これらは、適当な基板の表面に、官能基化された長-鎖有機分子の化学吸着及び自己組織化により自発的に形成する。SAMは、基板を表面と反応するリガンドを含有する溶液中に含浸することにより、又は基板を反応性種の蒸気に曝すことにより、通常調製される。SAMを製造するための多くのシステムが当該技術分野において公知である。
特定の好ましい態様において、表面物質としての金、並びにチオール、スルフィド、又はジスルフィドのような少なくとも1個のイオウ-含有官能基を有するSAM-形成する化合物の組合せが選択される。
本発明は更に下記の限定的でない実施例により例証される。
実施例1:一般的実験法
NMR
1H NMRスペクトルは、CDCl3、CD3OD又はD2O中で、Varian 400MHzスペクトロメーターで、各溶媒の残存ピークに関連して報告された化学シフトと共に記録した。反応は、アルゴン大気下で行った。試薬は、特に記さない限りは、受け取ったまま使用した。
フラッシュクロマトグラフィーを、Merckシリカゲル60(230-400)メッシュを用い行った。薄層クロマトグラフィー(TLC)を、EM Scienceシリカゲル60プレート(厚さ0.25mm)上で行った。全ての化合物を、短波長紫外線、ニンヒドリン染色液、又は硫酸セリウム/七モリブデン酸アンモニウム四水和物染色液のいずれかにより、視覚化した。全ての試薬は、Aldrich社又はVWR社から購入した。
金基体は、顕微鏡用カバーガラス(VWR 24x50mm #2)又はカバースリップ(VWR 25x75mm)上の、チタン接着層(1.5nm)、それに続く金層(45nm)の蒸着(evaporation)により調製した。蒸着前に基板を、ヘキサン(20分間)及び95%EtOH(20分間)中の音波処理により洗浄し、その後窒素流れ下で乾燥した。蒸着は、サーマルエバポレーター(Edwards Auto 306)を圧力6x10-6 Torr (8x10-4Pa)及び速度0.3nm/秒で用いて行った。この金で被覆した基板を、約1x1cmの小片に切断し、無水エタノールで洗浄し、及び窒素流れ下で乾燥した。単層を、様々な比のジスルフィド1及び2の混合物(総ジスルフィド濃度0.2mM)のエタノール性溶液への清潔な金基板の含浸により形成した。16時間後、単層を無水エタノールですすぎ、窒素ガス流れ下で乾燥した。
EG3バックグラウンド中に1.5%マレイミド基を示す金チップを、SPR機械(Biacore 3000)に装填した。このチップ上のツーフローチャネルは、1.5%ビオチンがその表面に存在するために、マレイミドと反応するシステイン-ビオチンに同時に曝露した。引き続き、ひとつのチャネルは、ストレプトアビジン70μg/mLに曝露し、次にひとつのチャネルは、フィブリノーゲン500ug/mLに曝露した。ストレプトアビジンの吸着により引き起こされた大きいシグナルの変化(2100RU)は、ビオチンの大きい特異的結合能を示し;フィブリノーゲンの吸着により引き起こされた小さいシグナルの変化(90RU)は、その表面が、低い非-特異的結合(NSB)を保持したことを示している。この実験は、このマレイミド/EG3表面のバッチが、タンパク質不動化(下記参照)に適していることを示した。
SPRは、Biacore 3000装置で行った。分析されるSAMを提示している金で被覆したガラスの顕微鏡用カバーガラスを、SPRカートリッジに実装した。全ての実験は、流量10μL/分を用いた。
電気化学的試験は、BAS Epsilon定電位装置を用いて行った。SAMに関する電気化学は、作動電極として金基板を、対電極として白金線を、及びAg/AgCl/KCl参照電極を用い、電解質として0.5M KNO3を含有する水中で行った。全ての実験は、サイクリックボルタンメトリー様式で、走査速度200mV/秒で行った。

レッドクス波下面積から、総電荷(Q)を決定し(式1)、これは表面上のレドックス-活性分子のモル数(n)に比例している。密度は、レドックス活性分子のモル数を表面上の分子の総モル数で除算する(これらふたつの数値は、SAM表面積に対して標準化された後) (式2)ことにより決定した。本発明者らは、表面上のマレイミド基は全てフェロセン-チオール分子と反応したと想定し、そのような表面上のフェロセン密度から、本発明者らは、これのマレイミド密度を推察した。
SAM調製のためのジスルフィド合成
2-{2-[2-(2-{2-[2-(11-トリチルスルファニル-ウンデシルオキシ)-エトキシ]-エトキシ}-エトキシ)-エトキシ]-エトキシ}-エタノール(4)
THF 10mLに溶解した、Pale-GrosdemangeらHの方法に従い調製した2-{2-[2-(2-{2-[2-(11-メルカプト-ウンデシルオキシ)-エトキシ]-エトキシ}-エトキシ)-エトキシ]-エトキシ}-エタノール(3)(1.22g, 2.6mmol)の溶液に、トリフェニルメチルクロリド(1.46g, 5.2mmol)を添加した。室温で48時間撹拌後、反応混合物を濃縮した。粗残留物を、フラッシュクロマトグラフィー(勾配溶離、EtOAcから20:1 EtOAc/MeOHへ)により精製し、化合物4を910mg(49%)得た。
p-トルエンスルホニルクロリド(900mg, 4.7mmol)を、CH2Cl2 12mL及びピリジン2mLの0℃の溶液中に溶解したアルコール4(910mg, 1.28mmol)の溶液に添加した。この溶液を室温に温め、16時間攪拌した。反応混合物を、ブライン(2x30mL)及びH2O(2x30mL)で洗浄し、その後有機物をMgSO4上で乾燥した。溶媒を蒸発させた後、粗残留物を、フラッシュクロマトグラフィー(勾配、1:1ヘキサン/EtOAcからEtOAcへ)で精製し、純粋な化合物5を1.01g(91%)得た。
DMF 10mL中のイミノジカルボン酸ジ-tert-ブチル(136mg, 0.63mmol)の0℃の溶液に、水素化ナトリウム(60%, 25mg, 0.63mmol)を添加した。室温で40分間攪拌した後、3mLのDMF中の化合物5(452mg, 0.52mmol)の溶液を滴下した。この溶液を45時間攪拌し、次に溶媒を真空蒸発させた。粗残留物を、CH2Cl2 30mLに溶解し、H2O(2x10mL)で洗浄した。有機層をMgSO4上で乾燥し及び溶媒を蒸発させた後、この残留物を、カラムクロマトグラフィー(ヘキサン/EtOAc, 1:1(v/v))で精製し、純粋な化合物6 343mg(72%)を得た。
トリフルオロ酢酸10mLに溶解したエタンジチオール(0.5mL)、H2O(0.5mL)、フェノール(1g)、及びチオアニソール(0.5mL)の溶液に、化合物6(247mg, 0.27mmol)を添加した。室温で6時間攪拌した後、この反応混合物を濃縮し、カラムクロマトグラフィー(勾配溶離、CH2Cl2/MeOH 20:1(v/v)から10:1から5:1へ)により精製し、化合物7(約75%)及びトリチル保護されたチオール(約25%)の混合物147mgを得た。更に精製することなく次工程を続けた。
アルドリチオール-2(145mg, 0.66mmol)を、MeOH 5mL中の2-{2-[2-(11-メルカプト-ウンデシルオキシ)-エトキシ]-エトキシ}-エタノールref(201mg, 0.66mmol)の溶液に添加した。この溶液を18時間室温で攪拌した後、溶媒を蒸発させ、残留物を、フラッシュクロマトグラフィー(勾配溶離、ヘキサン/EtOAc 1:1(v/v)からEtOAc)で精製し、化合物9を148mg(56%)得た。
MeOH 5mL中の化合物7(120mg, 0.2mmol)の溶液に、化合物9(101mg, 0.22mmol)を添加した。この溶液を室温で33時間攪拌した。濃縮後、この反応混合物を、フラッシュクロマトグラフィー(勾配:CH2Cl2/MeOH、20:1から10:1から5:1(v/v))で精製し、不純物8を137mg得た。
無水DMF 4mL中の粗化合物8(137mg, 0.2mmol)の溶液に、γ-マレイミド酪酸N-ヒドロキシスクシンイミドエステル(10)(58mg, 0.21mmol)及びEt3N (48μL, 0.34mmol)を添加した。この溶液を、室温で24時間攪拌した。濃縮後、この反応混合物を、フラッシュクロマトグラフィー(EtOAc/MeOH、10:1(v/v))により精製し、化合物1を22mg得た。
THF 5mLに溶解した2-{2-[2-(11-メルカプト-ウンデシルオキシ)-エトキシ]-エトキシ}-エタノール(145mg, 0.43mmol)の溶液に、NaOH溶液(0.1M)1mL、その後ヨウ素(5結晶)を溶解した。この溶液を室温で24時間攪拌し、CH2Cl2を20mL添加した。この溶液をH2O(2x5mL)で洗浄した後、有機物をMgSO4上で乾燥し、濃縮した。粗残留物を、フラッシュクロマトグラフィー(CH2Cl2/MeOH、20:1(v/v))で精製し、化合物2を78mg(54%)得た。
フェロセン-2-カルボン酸[2-(ピリジン-2-イルジスルファニル)-エチル]-アミド(12)
EDC(213mg, 1.1mmol)を、CH2Cl2 20mLに溶解したフェロセンカルボン酸(232mg, 1.0mmol)及びN-ヒドロキシスクシンイミド(128mg, 1.1mmol)の溶液に添加した。この溶液を室温で5時間攪拌し、濃縮した。粗残留物を、DMF 8mL中に溶解し、及び2-(ピリジン-2-イルジスルファニル)-エチル-塩化アンモニウム(222mg, 1.0mmol)を、引き続きEt3N (0.14mL, 1.0mmol)を添加した。48時間攪拌した後、溶媒を真空蒸発させた粗残留物を、フラッシュクロマトグラフィー(勾配:ヘキサン/EtOAc、1:1(v/v)からEtOAcへ)で精製し、化合物12を113mg(2工程で28%)得た。
MeOH 6mL中に溶解した化合物12(113mg, 0.28mmol)の溶液に、DTT(438mg, 2.8mmol)及びEt3N(79μL, 0.57mmol)を添加した。この溶液を室温で18時間攪拌し、その後溶媒を蒸発させた。この残留物を、EtOAc 15mLに溶解し、H2O(8x15mL)で洗浄し、過剰なDTTを除去した。MgSO4上で乾燥し濃縮した後、残留物を、フラッシュクロマトグラフィー(ヘキサン/EtOAc、1:1(v/v))により精製し、化合物13を46mg(57%)得た。
8-ブロモ-1-オクテン(14)(5g, 26.16mmol)及び亜リン酸トリエチル(8.69g, 52.32mmol)の混合物を、ゆっくり加熱し156℃とした。この反応混合物を、この温度で、アルゴン下で一晩攪拌した。発生した臭化エチルは、冷却器及び氷浴中の受けフラスコで捕集した。過剰な亜リン酸トリエチルを真空除去し、残留油分を高真空下で蒸留し(〜160℃)、無色の油状物(15)を得た(6.22g, 96%)。
化合物(15)(1.00g, 4.03mmol)を、CH2Cl2 (30mL)に溶解した。混合物をアルゴン下で0℃に冷却した後、塩化オキサリル(1.28g, 10.07mmol)を滴下した。この混合物をゆっくり室温とし、16時間攪拌した。過剰な塩化オキサリル及び溶媒を真空除去した。中間体モノクロロリン酸エステル及び4-ニトロフェノール(561mg, 4.03mmol)を、無水CH2Cl2 (30mL)に溶解した。トリエチルアミン(816mg, 8.06mmol)を滴下し、この混合物を室温で5時間攪拌した。これを真空で濃縮し、黄色油状物を得、これをフラッシュクロマトグラフィー(シリカゲル、Hex:EtOAc(1:1))で精製し、純粋な化合物(3)を油状物として得た(1.06g, 77%)。
チオール酢酸(359mg, 4.72mmol)及びAIBN(15.4mg, 0.094mmol)を含有する無水1,4-ジオキサン(5mL)中の化合物(16)(161mg, 0.47mmol)の溶液を、アルゴン下で、還流温度で2.5時間攪拌した。この混合物を濃縮し、フラッシュクロマトグラフィー(シリカゲル、CH2Cl2:EtOAc (4:1))で精製し、化合物(4)を黄色油状物として得た(149mg, 76%)。
濃塩酸(545mL, 13.6mmol)を含有するMeOH (30mL)中の化合物(17)(1.14g, 2.72mmol)の溶液を、40℃で16時間攪拌した。この混合物を真空で濃縮し、CH2Cl2に再溶解し、飽和した炭酸水素ナトリウム(2x20mL)及びブライン(1x20mL)で洗浄した。有機層をNa2SO4上で乾燥し、濃縮し、純粋な化合物(18)を黄色油状物として得た。
ペプチド不動化
C-末端システインを伴うsrcキナーゼ基質(IYGEFKKKC)を、1mMペプチド溶液(pH6)と1時間インキュベーションし、引き続き水で洗浄し及び窒素流れで乾燥することにより、マレイミドを提示しているSAMへ密度2%で共有的に不動化した。
スタウロスポリン(Calbiochem社)を、DMSO中に様々な濃度で調製した。活性p60c-src は、Upstate Biotechnology社から購入し、最終アッセイ濃度20pM p60c-src となるよう、キナーゼアッセイ緩衝液(KAB)(50mM HEPES pH7.4、0.1mM EDTA、0.015% Brij35)及びキナーゼ希釈緩衝液(KDB)(KABに0.1mg/ml BSA及び0.2% β-メルカプトエタノールを添加)で希釈した。キナーゼ反応を、スタウロスポリン希釈液0.6μlを、4μlのp60c-src 希釈液と予め混合させ、最終濃度50μM ATP及び10mM Mg2+となるような、ATP/Mg2+カクテル(KAB中に作成)を添加することにより開始した。反応混合液6μlを、直ぐに各ウェルに分配し、この試料を37℃で30分間インキュベーションした。インキュベーション後、試料を、8mM SDSで1分間、TBST (TBS(10mM Tris pH7.4、150mM NaCl)と0.05% Tween-20)で3分間3回、並びにTBSで3分間2回洗浄した。
炭水化物タグ付け
全ての炭水化物は、還元末端にn-ペンテニル基を有する過アセチル化された糖質のチオールアセテート誘導体への転換により誘導体化した。次にこれらの糖質は、酸素非存在条件下でケン化し、中和後、還元末端にチオール基を含む十分に脱保護された炭水化物を得た。図10参照。
炭水化物は、還元末端ペンタンチオール基を含む完全に脱保護された炭水化物の溶液(pH6のリン酸緩衝液中の1mM炭水化物)と共に1時間インキュベーションすることにより、密度2%のマレイミドを提示する単層(顕微鏡スライド上)に共有的に不動化した。水及びエタノールで洗浄後、これらの単層を窒素流れ下で乾燥した。図11参照。
グリコシルトランスフェラーゼアッセイのための3種の異なる基質を提示している表面を、誘導体化されたマレイミド表面を、3種の異なる炭水化物-チオール溶液と接触することにより形成した。これらの炭水化物は、剥離可能及び再シール可能な装置を用い、溶液を制限することにより、スライド上の個別の位置に不動化された。図12参照。
Cutenaseの調製
乾腐病菌(Fusarium solani pisi)クテナーゼ遺伝子は、50bpイントロンにより分離されたふたつのエキソンを含む。イントロンを取り除くために、各エキソンを、制限エンドヌクレアーゼ部位を含むプライマーセットを用いて増幅した。PCR増幅及びPCR産物の制限消化後、これらふたつのエキソンを連結し、イントロンを含まないクチナーゼ遺伝子を得た。次にこの遺伝子を、組換え法を用いてプラスミドへ挿入した。
ホスホン酸結合パートナー部分を末端とする自己組織化単層(SAM)を調製した。このリガンドは、非-特異的タンパク質吸着に抵抗するトリ(エチレングリコール)基と混合され、低密度で存在した。クチナーゼのこの単層への不動化は、SPR分析により特徴決定した。リン酸緩衝した生理食塩水(pH7.4)を、単層上に2分間流し、ベースラインを確立し、その後同じ緩衝液中のタンパク質溶液を10分間流し、結合を観察した。最後に、このタンパク質溶液を、緩衝液と6分間交換し、不可逆的に不動化されたタンパク質量を定量した。クチナーゼ(25M)が表面に不可逆的に結合した。この単層のドデシル硫酸ナトリウム(SDS)(0.5mg/mL)による処理は、表面からのクチナーゼの取り外しを生じず、これはこの不動化は共有的であることを確認した。SDSは、非-共有的に不動化された分子を表面から取り除くために利用する界面活性剤である。最初に20で妨害されたクチナーゼは、表面への結合を示さず、これはこの不動化が特異的であることを明らかにしている。
EG3バックグラウンド中に1.5%マレイミド基を提示している金チップを、(8-メルカプト-オクチル)-ホスホン酸エチルエステル4-ニトロ-フェニルエステル−二官能性分子(化合物5;HS-PNPP)−の5mMメタノール溶液に1時間曝した。このプロセスの間に、HS-PNPPのチオール基は、表面のマレイミド基と反応し、表面に提示されたPNPPリガンドの1.5%単層を生じた。
前述のように作成されたチップは、次に異なる溶液を、最大4フローチャネルで表面に送達することを可能にする微小流体マニホールドを金チップへ接触するSPR装置へと搭載した。下記の流体プロトコールをこの表面に適用し、SPRシグナル−これは表面に吸着するタンパク質質量を測定する−を、時間の関数として測定した:
t=0〜300秒:フローチャネル1-3(Fc=1〜3)は全て、20mMトリス(pH7.4)緩衝液に曝露した。
t=300〜1500秒:その後Fc=1〜3を、3種の異なる溶液に曝した。
Fc=1:20mMトリス(pH7.4)緩衝液中の500ug/mLフィブリノーゲン。
Fc=2:20mMトリス緩衝液中の40μMヘキサヒスチジンに融合したクチナーゼ(クチナーゼ-his6)
