Abstract
【解決手段】 IκB−ζをコードする非ヒト動物の内在性遺伝子の全部又は一部が破壊・欠損・置換等の遺伝子変異により不活性化され、IκB−ζを発現する機能を喪失し、TLRリガンド及びIL−1に応答するIL−6の産生が障害されている、TLRリガンド/IL−1不応答性モデル非ヒト動物を作出する。また、かかる非ヒト動物と被検物質を用いて、TLRリガンド/IL−1に対する応答の促進物質又は抑制物質や、アトピー性皮膚炎様の炎症性皮膚病変の予防・治療薬をスクリーニングする。
Description
IL−1又はLPSによる刺激は、マクロファージ細胞株及び線維芽細胞においてIκB−ζを誘導し、IL−1RとLPSの受容体であるTLR4は、細胞内シグナル伝達経路を共有していることから、他のTLRリガンドにも応答してIκB−ζが誘導されるかどうかを調べた。IL−1及びLPSに加え、ペプチドグリカン(PGN)(TLR2リガンド)、細菌性リポタンパク質(BLP)(TLR1/TLR2リガンド)、フラゲリン(flagellin)(TLR5リガンド)、MALP−2(TLR6/TLR2リガンド)、R−848(TLR7リガンド)、及びCpG DNA(TLR9リガンド)による刺激を受けてもIκB−ζのmRNAは腹腔マクロファージ又は肺由来正常細胞で著しく誘導されたが、TNF−αによる刺激では誘導は認められなかった(図1a)。他方、IκBファミリーの別のメンバーであるIκB−α及びBcl−3は、TLRリガンドやIL−1同様にTNF−αにも応答して誘導された。これらを総合するとIκB−α及びBcl−3と異なり、IκB−ζの誘導はTLR/IL−1Rによる刺激に対して特異的に認められる。
TLR/IL−1R応答におけるIκB−ζのインビボでの役割を解明するために、標的遺伝子破壊法によりIκB−ζ欠損マウスを作製した。IκB−ζの中心部分をコードするエキソン2つをネオマイシン耐性(neoR)遺伝子に置き換えたターゲッティングベクターを構築した(図2a)。胚幹(ES)細胞で相同的組換えが生じたことがサザンブロット分析により確認され、正しくターゲットされたES細胞を用いて変異対立遺伝子をもつマウスを作製した(図2b)。ホモ接合型マウスは予想されるメンデルの法則に従って生まれた。変異マウスにおけるIκB−ζmRNAの発現をノーザンブロット分析により調べた。LPSは、野生型細胞でIκB−ζ誘導を刺激したが、IκB−ζ欠損細胞ではIκB−ζmRNAは全く認められなかった(図2c)。免疫ブロット分析の結果からも、IκB−ζ遺伝子が破壊されることによりIκB−ζタンパク質の発現が消失することがさらに確認された(図2d)。IκB−ζ欠損マウスのBリンパ球及びTリンパ球は正常な構成を示していた。しかし、IκB−ζ欠損脾臓細胞ではLPS応答性の増殖が損なわれていたが、α−CD40、IL−4及びα−IgMに対する応答性は損なわれず(図2e及び2f)、IκB−ζ欠損細胞でTLR4応答が損なわれることを示唆している。
次に、腹腔マクロファージにおけるLPSに誘導される炎症関連分子の産生を分析した。IFN−γ存在下で野生型マクロファージはLPSに応答してTNF−α及び硝酸オキシド(NO)を産生した(図3a)。IκB−ζ欠損マウスにおいてLPS誘導性のTNF−α及びNOの産生は正常であっても、IL−6の産生は著しく減少していた。さらに、CpG DNA、R−848、PGN、MALP−2、BLP等、別のTLRリガンドに刺激されるIL−6の産生もまたIκB−ζ欠損細胞では顕著に阻害されていた(図3b、3c、及び3d)。また、IκB−ζ欠損細胞では、IL−1応答性のIL−6産生の欠如が認められた。しかしTNF−α誘導性IL−6産生は、野生型及びIκB−ζ欠損細胞の両方で同程度に観察された(図3e)。次いで、LPSに応答するIL−6mRNAの発現を調べた。LPSは、野生型細胞においてIL−6mRNA誘導を刺激した。これとは全く対照的に、IκB−ζ欠損細胞におけるIL−6mRNAレベルは劇的に減少しており(図3f)、IκB−ζ欠損細胞におけるIL−6産生量の減少が、mRNA誘導欠損を反映している可能性を示している。
IκB−ζが、リガンドに誘導されるIL−6プロモーターの活性化を促進するかどうかを調べるため、IL−6又はELAM1遺伝子のいずれかのプロモーターを有するルシフェラーゼレポータープラスミドを、対照又はIκB−ζ発現ベクターと共にRAW264.7細胞に導入した。LPSは、IL−6及びELAM1プロモーター両方の活性を投与量依存で刺激した。IκB−ζが過剰発現する場合、RAW264.7細胞においてIL−6のプロモーター活性がさらに亢進されるが、ELAM1においてはその傾向が認められなかったことを見い出した(図4a)。かかる亢進傾向が非刺激細胞で観察されたことから、IκB−ζの異所性発現が、未刺激RAW264.7細胞においてIL−6プロモーターの活性に正の効果を示すかどうかを調べた。IκB−ζの導入量を増加することにより、IL−6プロモーターの基礎活性が未刺激細胞においてもアップレギュレートされることが観察された(図4b)。
遺伝子学的手法により上記仮説を確認するため、NF−κB1/p50欠損マウスから腹腔マクロファージ及びMEFsを得て、LPS誘導性のサイトカイン産生を分析した。既に報告されているように、IFN−γ存在下においてNF−κB1欠損マクロファージにおけるLPS誘導性TNF−αの産生は、野生型細胞の場合と変化がなかった。他方、NF−κB1欠損細胞では、LPSに応答するIL−6産生が損なわれていた(Cell. 80, 321-330 (1995), Proc Natl Acad Sci U S A. 97, 12705-12710 (2000)、及び図5a)。また、IL−1や他のTLRリガンドに誘導されるIL−6産生は、NF−κB1欠損細胞で著しく損なわれていた(図5b)。結果的に、NF−κB1欠損マウスにおけるTLR/IL−1Rを介した応答は、IκB−ζ欠損マウスにおける応答を想起させるものであった。
(1)IκB−ζ欠損マウスの作製
129/SVマウスゲノムライブラリーからIκB−ζ遺伝子を有するゲノムDNAを単離し、制限酵素マッピング及び配列分析により特徴付けを行った。IκB−ζの中心部分をコードする2.0kb断片をneoRカセットに置き換えることにより、ターゲッティングベクターを構築し、負の選択を行うために上記ゲノム断片中のPGKプロモーターによって転写される単純ヘルペスウイルスチミジンキナーゼを挿入した(図2a)。このターゲッティングベクターを胚性幹細胞(E14.1)にトランスフェクトした。G418とガンシクロビアに二重に耐性をもつコロニーを選択し、PCR及びサザンブロッティングによるスクリーニングを行った。2つの異なる相同組換え体をC57BL/6雌マウスにマイクロインジェクションし、ヘテロ接合体F1世代を交配させてIκB−ζ欠損マウスを得た。これら異なるクローンから得られたマウスは同じ表現型を示した。6〜8週齢のIκB−ζ欠損マウス及びそれらの同腹野生型を本発明の実験に用いた。全ての動物実験は、微生物病研究所付属実験動物研究委員会(大阪大学、大阪、日本)の承認を得て実施した。
サルモネラ菌(Salmonella Minnesota Re 595)由来のLPSと黄色ブドウ球菌(S.aureus)由来のPGNは、それぞれSigma社とFluka社から購入した。MALP−2及びCpGオリゴデオキシヌクレオチドは文献(J Immunol. 164, 554-557 (2000), Nature. 408, 740-745 (2000))記載の方法に従って調製した。フラゲリン及びR−848はそれぞれA. Adrem博士及びH. Tomizawa博士から提供を受けた。IκB−ζに対するポリクローナル抗体は、細菌で発現させたマウスIκB−ζタンパク質のC末端部分を用いてウサギを免疫して得た。リン酸化ERK、JNK及びp38に対する各抗体はCell Signaling社から購入した。ERK、JNK、p38、RelA、p50、c−Rel及びC/EBP−βのそれぞれに対する抗体はSanta Cruz社から入手した。組換えTNF−α及びIL−1βはGenzyme社から入手した。NF−κB1/p50欠損マウス又はMyD88欠損マウスは文献(Cell. 80, 321-330 (1995), Proc Natl Acad Sci U S A. 89, 1473-1476 (1992))記載の方法に従って調製した。
チオグリコール酸に誘導される腹腔マクロファージ又はMEFsを96ウエルプレート(マクロファージは1ウエルあたり5×104細胞、MEFsは1×104細胞)において所定濃度の所定リガンド共存下で培養した。製造者マニュアルに従ってELISAを行い、培養上清のTNF−α及びIL−6の濃度を測定した(TNF−αにはGenzyme社製、IL−6にはR&D社製)。NO濃度は製造者(DOJINDO)の指示に従い、Griess法により測定した。
10ng ml-1のIL−1βでMEFs(1×106)を所定期間刺激した。文献(Nature. 408, 740-745 (2000))記載の方法に従い、細胞から核抽出物を精製し、NF−κBのDNA結合部位に対する特異的プローブの共存下でインキュベートし、電気泳動にかけ、オートラジオグラフィーで視覚化した。
レポータープラスミドは、マウスIL−6遺伝子の5'フランキング領域(−1240/+40)で構成され、図4a及び4bで使用した。図4cで用いたレポータープラスミドは文献収載のものである(Proc Natl Acad Sci U S A. 90, 10193-10197(1993), Proc Natl Acad Sci U S A. 89, 1473-1476 (1992))。ELAM1プロモーター由来ルシフェラーゼレポータープラスミドは、D. T. Golenbockから譲られたものである。完全長IκB−ζ(Full)又はC末端部位を欠く欠失変異型IκB−ζ(ΔC、J Biol Chem. 276, 27657-27662 (2001))を文献収載の方法どおりにしてpMRXレトロウイルスベクターにクローニングした(J Biol Chem. 278, 36005-36012 (2003))。Plat−Eパッケージング細胞は、T. Kitamura博士の厚意で提供を受けた(Gene Ther. 7, 1063-1066 (2000))。要点を述べると、レトロウイルスを作出するために、リポフェクタミン2000試薬(Invitrogen社製)を用いてPlat−E細胞にトランスフェクトし、トランスフェクションの60時間後にレトロウイルスを含むPlat−E細胞の培養上清を回収した。4μg/mlのポリブレン存在下でMEFsを6時間感染させ、感染48時間後に分析した。
SUPERFECTトランスフェクション試薬(QIAGEN社製)を用い、レポータープラスミドを対照又はIκB−ζ発現ベクターのいずれかと共にRAW264.7及びP19細胞に一過性に共トランスフェクトした。文献記載(28,29)の方法に従い、全細胞溶解物のルシフェラーゼ活性をデュアルルシフェラーゼレポーターアッセイシステム(Promega社製、Maiden, W1)を用いて測定した(Nat Immunol. 3, 392-398 (2002), Nat Immunol. 2, 835-841 (2001))。
ChIPアッセイは基本的に説明書にあるプロトコール(Upstate Biotechnology社製)に則って行った。概要を述べると、LPS(1μg ml-1、3時間)、IL−1β(10ng ml-1、3時間)又はTNF−α(10ng ml-1、3時間)で、2×106個のRAW264.7細胞又は5×105個のMEFsをそれぞれ刺激し、ホルムアルデヒドを用い、37℃で60分間固定した。これらの細胞を溶解し、超音波処理にかけて分断(sheared)し、特異的抗体共存下で一晩インキュベーションし、続いて鮭精子DNA(Upstate Biotechnology社製)で飽和したプロテインAアガロースの共存下でインキュベーションした。沈殿DNAを以下のプライマーを用いて定量PCR(35〜40サイクル)で分析した。
IL−6プロモーターのκB部位には、

及び 5'-CTCCAGAGCAGAATGAGCTACAGACAT-3'(配列番号2)
IL−6プロモーターの3´遺伝子セグメントには、
5'- GCAGATGGACTTAGCTCGTCTCATTCA -3'(配列番号3)
及び 5'- CCACTCCTTCTGTGACTCCAGCTTATC -3'(配列番号4)
ELAM1プロモーターのκB部位には、
5'-GATGCAGTTGAGAATTTCCTCTTAGCC-3'(配列番号5)
及び 5'-TGGAATAGTTGTTCTGGCGTTGGATCC-3'(配列番号6)
腹腔マクロファージ及びMEFsを所定リガンドで所定期間刺激した。1.0%Nonidet−P40、150mM NaCl、20mM Tris−Cl(pH7.5)、5mM EDTA及び蛋白質阻害物質(Roche社製)を含む溶解緩衝液に上記細胞を溶解した。この細胞溶解物をSDS−PAGEにより溶解し、PVDF膜上に移した。免疫沈降を行うため、細胞溶解物をプロテインGセファロースTM(Amersham Pharmacia Biotech社製)で2時間前処理(preclear)し、所定抗体1.0μgを含むプロテインGセファロースTM共存下において4℃で12時間攪拌下でインキュベーションした。免疫沈降物を溶解緩衝液で4回洗浄し、Laemmliサンプル緩衝液を用いて沸騰溶出し、α−IκB−ζを用い、上述のとおりにしてウエスタンブロッティングに供した。
野生型腹腔マクロファージ及びIκB−ζ欠損腹腔マクロファージを用い、LPSに応答するマイクロアレイ分析(Affimetrix社製)を文献収載の方法(Blood. 102, 4123-4129 (2003))に従って行った。GeneSpring6.0(Silicon Genetics社製)ソフトウエアを使用して遺伝子発現のカラー画像を作成した。
Claims (7)
- IκB−ζをコードする非ヒト動物の内在性遺伝子の全部又は一部が破壊・欠損・置換等の遺伝子変異により不活性化され、IκB−ζを発現する機能を喪失し、TLRリガンド及びIL−1に応答するIL−6の産生が障害されていることを特徴とするTLRリガンド及びIL−1不応答性モデル非ヒト動物。
- TLRリガンドが、LPS、BLP、PGN、MALP−2、R−848又はCpG DNAであることを特徴とする請求項1記載のTLRリガンド及びIL−1不応答性モデル非ヒト動物。
- アトピー性皮膚炎様の炎症性皮膚病変を呈することを特徴とする請求項1又は2記載のTLRリガンド及びIL−1不応答性モデル非ヒト動物。
- 非ヒト動物がマウスであることを特徴とする請求項1〜3のいずれか記載のTLRリガンド及びIL−1不応答性モデル非ヒト動物。
- 請求項1〜4のいずれかに記載の非ヒト動物と、被検物質と、TLRリガンド若しくはIL−1又はこれらの含有物とを用いて、前記非ヒト動物におけるTLRリガンド若しくはIL−1又はこれらの含有物に応答するIL−6の産生の程度を測定・評価することを特徴とするTLRリガンド若しくはIL−1又はこれらの含有物に対する応答の促進物質又は抑制物質のスクリーニング方法。
- 請求項1〜4のいずれか記載の非ヒト動物に由来する免疫細胞と、被検物質と、TLRリガンド若しくはIL−1又はこれらの含有物とを用いて、前記免疫細胞におけるTLRリガンド若しくはIL−1又はこれらの含有物に応答するIL−6の産生の程度を測定・評価することを特徴とするTLRリガンド若しくはIL−1又はこれらの含有物に対する応答の促進物質又は抑制物質のスクリーニング方法。
- 請求項1〜4のいずれかに記載の非ヒト動物に被検物質を投与し、前記非ヒト動物におけるアトピー性皮膚炎様の炎症性皮膚病変の程度を測定・評価することを特徴とするアトピー性皮膚炎様の炎症性皮膚病変の予防・治療薬のスクリーニング方法。
