Abstract

Claims (17)
- (a)「出発表面(starting surface)」と「行き先表面(destination surface)」を有する多孔質膜を提供すること;
(b)前記多孔質膜の出発表面に細胞を付着させ、前記多孔質膜を通る前記出発表面から前記行き先表面への前記細胞の一部の移動を促進すること;
(c)前記出発表面に残存する細胞の部分又は前記行き先表面に移動した細胞の部分のいずれかに光学的分別溶液を加えて、前記処理部分を電気光学的読み取り装置によって大なり小なり検出可能にすること;及び
(d)前記電気光学的読み取り装置を使用して、前記出発表面に残存する又は前記行き先表面に移動した又はその両方の前記細胞の光学特性を読み取ること
の工程をこの順序で含む、細胞の光学特性を検出することができる電気光学的読み取り装置を使用して細胞を分析するための方法。 - 前記多孔質膜の出発表面上の工程(b)にある細胞を、異なる所定暴露バンド幅の放射線への暴露時に所定蛍光バンド幅の放射線を放射する化合物で標識する、請求項1に記載の方法。
- 前記化合物がカルセイン、カルセイン前駆体又はカルセイン誘導体である、請求項2に記載の方法。
- 少なくとも前記所定蛍光バンド幅内の光を吸収することができる着色剤を含み、及び少なくとも工程(c)及び(d)の間前記多孔質膜を通って実質的に流動することができない、前記光学的分別溶液を、前記非移動細胞と前記出発表面に適用することによって、前記出発表面に残存する細胞の部分をより検出しにくくする、請求項2に記載の方法。
- 前記光学的分別溶液がグリセロール中の前記着色剤の懸濁液を含み、及び前記着色剤がカーボンブラックである、請求項4に記載の方法。
- 工程(d)の電気光学的読み取り装置を、前記所定暴露バンド幅の照射による前記細胞の移動部分の暴露後の前記所定蛍光バンド幅内の放射を検出するために利用し、前記暴露が前記細胞の移動部分への入射前に前記多孔質膜を通過しない、請求項5に記載の方法。
- 前記多孔質膜が同多孔質ポリカーボネート膜である、請求項1に記載の方法。
- 前記多孔質膜が入れ子になった頭部液体容器と底部液体容器の間に存在し、前記入れ子式液体容器が前記多孔質膜によって上部区域と下部区域に分けられている、請求項1に記載の方法。
- 前記方法を、前記入れ子式液体容器のアレイを含むマルチウエルアレイを使用して実施する、請求項8に記載の方法。
- 前記マルチウエルアレイが24個の前記入れ子式液体容器を含む、請求項9に記載の方法。
- 前記多孔質膜を通る前記出発表面から前記行き先表面への前記細胞の一部の工程(b)での移動が、化学誘引物質含有溶液を前記下部区域内に入れることによって促進される、請求項8に記載の方法。
- 少なくとも前記所定蛍光バンド幅内の光を吸収することができる着色剤を含み、及び少なくとも工程(c)及び(d)の間前記多孔質膜を通って実質的に流動することができない、前記光学的分別溶液を、前記上部区域に添加することによって、前記出発表面に残存する細胞の部分を電気光学的読み取り装置によってより検出しにくくする、請求項11に記載の方法。
- 実質的に約1%のカーボンブラックと50%グリセロールから成る水性マスキング溶液。
- 共通の包装内に納められた、
各々の対が、入れ子にしたとき、半透明の同多孔質膜によって分けられた上部区域と下部区域を有する、入れ子式又は入れ子可能な液体容器の対を含むマルチウエルアレイ;及び
カーボンブラックとグリセロールを含む水溶液
を含む、細胞を分析するのに適した走化性アッセイキット。 - カーボンブラックの濃度が少なくとも約0.05%(重量/容積)であり、グリセロールの濃度が約5%から約75%(重量/容積)の範囲内である、請求項14に記載の走化性アッセイキット。
- 異なる所定暴露バンド幅の放射線に暴露したとき所定蛍光バンド幅の放射線を放射する化合物を含む細胞標識溶液
をさらに含む、請求項14に記載の走化性アッセイキット。 - 前記化合物がカルセイン、カルセイン前駆体又はカルセイン誘導体である、請求項16に記載の走化性アッセイキット。
