1971年に、Folkman J.(「Tumor angiogenesis:Therapeutic implications」、New Engl. Jour. Med. 285:1182〜1186)は、腫瘍細胞集団ではあらゆる増加が起こる前に、腫瘍に集まる新しい毛細血管の増加が起こるに違いないという仮定を立てた。それ以降、固形腫瘍の増殖、維持、および転移にとって、既存の微小血管床からの新しい血管の増殖が必要であることを示す多くの証拠が蓄積された。

- IL-3の存在下で培養したIL-3依存的細胞の死を促進することができない、以下からなる群より選択される非毒性で強力かつ選択的なc-kit阻害剤を、そのような治療を必要とする哺乳類に投与する段階を含む、腫瘍の血管新生を治療する方法:
‐ピリミジン誘導体、より詳しくはN-フェニル-2-ピリミジン-アミン誘導体、
‐インドリノン誘導体、より詳しくはピロール置換インドリノン、
‐単環式、二環式アリール、およびヘテロアリール化合物、
‐ならびにキナゾリン誘導体。 - R1が複素環基、特にピリジル基であり、R2およびR3はH、R4はC1〜C3アルキル、特にメチル基であり、R5およびR6がH、ならびにR7が少なくとも一つの置換基を有するフェニル基であって、アミノ基のように少なくとも一つの塩基性部位を有する、請求項5記載の方法。
- 阻害剤が、4-(4-メチルピペラジン-1-イルメチル)-N-[4-メチル-3-(4-ピリジン-3-イル)ピリミジン-2-イルアミノ)フェニル]ベンズアミドである、請求項4記載の方法。
- 阻害剤が、構成的に活性化された変異体c-kitおよび/またはSCF活性化c-kitから選択される活性化c-kitの阻害剤である、請求項4記載の方法。
- 活性化変異体c-kitが、Y823に対して近位の変異、より詳しくはc-kit自己リン酸化に関与する配列番号:1のアミノ酸800位〜850位の間の変異、特にD816V、D816Y、D816F、およびD820G変異体、ならびにc-kitの膜近傍ドメイン、好ましくはコドン573位〜579位の間における欠失から選択される、少なくとも一つの変異を有する、請求項9記載の方法。
- 以下を含むスクリーニング法によって得ることができる、選択的で強力かつ非毒性の活性化c-kit阻害剤である化合物を、そのような治療を必要とする哺乳類に投与する段階を含む、腫瘍の血管新生を治療する方法:
a)(i)活性化c-kitおよび(ii)試験する少なくとも一つの化合物を、成分(i)と成分(ii)とが複合体を形成することができる条件下で接触させる段階、
b)活性化c-kitを阻害する化合物を選択する段階、
c)IL-3の存在下で培養したIL-3依存的細胞の死を促進することができない、段階b)において同定された化合物のサブセットを試験して選択する段階。 - スクリーニング法が、SCF活性化野生型c-kitを阻害することもできる、変異体活性化c-kitの阻害剤である、段階b)において同定された化合物のサブセットを試験して選択することからなる段階をさらに含む、請求項11記載の方法。
- 活性化c-kitが、SCF活性化野生型c-kitである、請求項11記載の方法。
- 推定の阻害剤が、段階a)において10μMより高い濃度で試験される、請求項11から13のいずれか一項に記載の方法。
- IL-3が、IL-3依存的細胞の培養培地において0.5 ng/mlから10 ng/ml、好ましくは1 ng/mlから5 ng/mlの濃度で存在する、請求項11から14のいずれか一項に記載の方法。
- 成分(ii)が活性化c-kitを阻害する程度を、インビトロまたはインビボで測定できる、請求項11から15のいずれか一項に記載の方法。
- スクリーニング法が、1μM未満の濃度で野生型c-kitを阻害することができる化合物を、インビトロまたはインビボで試験して選択することからなる段階をさらに含む、請求項11から16のいずれか一項に記載の方法。
- 試験が、肥満細胞、トランスフェクトした肥満細胞、BaF3、およびIC-2からなる群より選択される細胞株を用いて行われる、請求項17記載の方法。
- 試験が、c-kit自己リン酸化量の決定を含む、請求項17記載の方法。
- スクリーニングが以下を含む、請求項11から18のいずれか一項に記載の腫瘍の血管新生を治療する方法:
a)永続的に活性化されたc-kitである変異体c-kit(例えば、トランスホスホリラーゼドメインにおける変異)を発現する細胞を用いて、細胞死の程度を測定することにより、それぞれがIC50<10 μMを有する、活性化c-kitを標的とする候補化合物のサブセットを同定するために、複数の被験化合物について増殖アッセイを行う段階、
b)c-kitを特異的に標的にする候補化合物のサブセットを同定するために、IL-3の存在下で培養したIL-3依存的細胞である野生型c-kitを発現する細胞を用いて、段階(a)において同定された候補化合物のサブセットについて増殖アッセイを行う段階、
c)c-kitを発現する細胞を用いて、段階b)において同定された化合物のサブセットについて増殖アッセイを行い、細胞死の程度を測定することにより、それぞれがIC50<10 μMを有する、好ましくはIC50<1 μMを有する、野生型c-kitを標的とする候補化合物のサブセットを選択する段階。 - ヒトにおける腫瘍の血管新生を治療するための、請求項1から20のいずれか一項に記載の方法。
- 阻害剤が経口投与される、請求項21記載の方法。
- 阻害剤が局所投与される、請求項21記載の方法。
