Description

プライマーペアは、両領域間の塩基配列を増幅できるように、すなわち、一方がセンスプライマーとなり他方がアンチセンスプライマーとなるように設定される。潜在プラスミドは環状であるため、両領域間の塩基配列は2つあるが、通常には短い方の塩基配列が増幅されるように設定される
本発明キットは、プライマーペアの他に、PCRおよび/または増幅産物の検出を行うのに必要とされる試薬類をさらに含んでいてもよい。このような試薬類の例には、上記のハイブリダイゼーションプローブも含まれる。

【実施例1】PCRによる検出
クラミジア・トラコマティス(GenBankアクセッション番号X06707)の潜在プラスミドの塩基配列(配列番号1)の第1の領域(塩基番号5157〜5201)および第2の領域(塩基番号5245〜5276)において、GC含量に富み(48−55%)、高いTm値を有する(63−65℃(最近接塩基対法により算出))塩基配列を選択した。すなわち、種類の上流プライマー(配列番号:2、3、)および種類の下流プライマー(配列番号:5、)の標的結合配列を選択した。そしてこれらの標的結合配列を有するオリゴヌクレオチドをプライマーとして合成した。
配列番号:3−5のプライマーペアについては、アニーリング温度68℃でクラミジア・トラコマティス菌体ゲノムDNA2コピーを検出可能であった。配列番号:4−6のプライマーペアについては、アニーリング温度65℃でクラミジア・トラコマティス菌体ゲノムDNA2コピーを検出可能であった。
